The biochemical defense mechanisms of amphibians involve cutaneous secretions of bioactive molecules with antimicrobial activity. This study evaluated the in vitro activity of methanolic extracts from cutaneous secretions of two amphibian species of the Bufonidae family, Rhaebo guttatus and Rhinella marina, in the control of the phytopathogens Fusarium udum, Fusarium solani, Colletotrichum truncatum, Aspergillus flavus, Rhizoctonia solani, Macrophomina phaseolina, and Calonectria pseudometrosideri. The R. guttatus extract decreased the mycelial growth of F. udum, F. solani, A. flavus, and M. phaseolina at some tested concentrations. The R. marina extract decreased the mycelial growth of C. truncatum at the concentration of 0.5 mg mL-¹, and inhibited the mycelial growth of A. flavus at concentrations of 0.1 and 0.5 mg mL-¹, which was similar to the inhibition by the positive control. The R. marina extract also decreased the microsclerotia production by R. solani at concentrations of 0.2 and 0.3 mg mL-¹. In addition, the extracts inhibited conidial sporulation and germination at varying degrees. The inhibition of appressoria formation in C. truncatum by the R. guttatus and R. marina extracts was 85-99% and 63-100%, respectively. Our results demonstrated that treatment with extracts from R. guttatus and R. marina cutaneous secretions showed antifungal activity against the studied phytopathogens. RESUMO Os mecanismos de defesa bioquímica dos anfíbios envolvem secreções cutâneas de moléculas bioativas com atividade antimicrobiana. Este estudo avaliou a atividade in vitro de extratos metanólicos da secreção cutânea de duas espécies de anfíbios da família Bufonidae, Rhaebo guttatus e Rhinella marina, no controle dos patógenos Fusarium udum, Fusarium solani, Colletotrichum truncatum, Aspergillus flavus, Rhizoctonia solani, Macrophomina phaseolina e Calonectria pseudometrosideri. O extrato de R. guttatus inibiu o crescimento micelial de F. udum, F. solani, A. flavus e M. phaseolina em algumas concentrações testadas. O extrato de R. marina inibiu o crescimento micelial de C. truncatum na concentração de 0,5 mg mL-¹, e inibiu o crescimento micelial de A. flavus nas concentrações de 0,1 e 0,5 mg mL-¹, que foi semelhante à inibição pelo controle positivo. O extrato de R. marina também diminuiu a produção de microescleródios de R. solani nas concentrações de 0,2 e 0,3 mg mL-¹. Além disso, os extratos inibiram a esporulação e germinação de conídios em graus variados. A inibição da formação de apressórios em C. truncatum pelos extratos de R. guttatus e R. marina foi de 85%-99% e 63%-100%, respectivamente. Nossos resultados demonstraram que o tratamento com extratos da secreção cutânea de R. guttatus e R. marina apresentou atividade antifúngica sobre os fitopatógenos estudados.

As arboviroses causadas pelo vírus Dengue (DENV) e Zika (ZIKV), são atualmente consideradas doenças que representam um potencial desafio para a saúde pública, resultando em intensas e limitantes manifestações clínicas, podendo culminar em microcefalia (ZIKV) e óbito. Ainda não existem vacinas na rede pública ou qualquer outra droga preventiva, o diagnóstico é impreciso devido as reações cruzadas entre os anticorpos de DENV, ZIKV e de outros flavivírus. Diante desse contexto, objetivamos desenvolver e também testar novas moléculas baseadas em pequenos peptídeos (fagos) ligantes ao vírus ZIKV e DENV-2 utilizando a metodologia Phage-Dispay. Assim, o objetivo do presente estudo foi selecionar, caracterizar e validar ligantes de ZIKV e DENV-2, bem como testar fagos miméticos a um inibidor de fosfolipase A2 avaliando a capacidade dessas moléculas em inibir e também diferenciar ZIKV e DENV. O imunoensaio ELISA, ensaio de inibição da entrada de ZIKV e DENV-2 in vitro por citometria de fluxo utilizando células Vero e anticorpo 4G2 e análises in sílico foram utilizados para a validação dos fagos selecionados. Também foi realizado ELISA utilizando amostras de pacientes infectados com DENV e ZIKV. Notavelmente, os fagos testados foram capaz de diferenciar DENV de ZIKV e de indivíduos saudáveis. A acurácia no diagnóstico foi determinada pela curva ROC. O fago D4, selecionado para ZIKV, foi capaz de inibir a entrada de ZIKV e DENV-2. The brushwood is a technique of environmental complexation, which consists in the use of plant residues. This technique, when well established technically, can exert influence on seedbed quality, fostering native plant establishment and ecosystem restoration. In this way, we aimed to evaluate the efficiency of brushwood for the induction of natural regeneration of native species in an area covered by exotic inhibitory grasses of the genus Urochloa, as well as check the minimum size required to prevent the re-invasion of the forage. As a hypothesis, we adopted the minimum width of 4 m as sufficient for the establishment of natural regeneration prior to the reoccupation of grasses. The experiment was conducted between May 2014 and May 2016 in Morretes-PR in a lowland evergreen rain forest area. Seven different sizes of brushwood were compared: 6 x 1, 6 x 2, 6 x 3, 6 x 4, 6 x 5, 6 x 6 m and control treatment. Woody species were identified and counted and herbaceous cover percentage estimated after 4, 8, 12, 18 and 24 months. There was no resumption of natural succession of native species. At any size, brushwood alone was ineffective to prevent the growth by grasses from the edges of the plots, and, since it constitutes a residue pile, it further complicates the establishment of woody species. For the creation of safe sites and consequent restoration by natural regeneration, there is a need for local elimination of Urochloa forages, without which native species are unlikely to survive. RESUMO A galharia é um método de complexação ambiental, que consiste no aproveitamento de resíduos vegetais. Esse método, quando bem estabelecido tecnicamente, pode exercer influência sobre a qualidade da cama de sementes, favorecendo o estabelecimento de plantas nativas e a restauração do ecossistema. Dessa forma, objetivou-se avaliar a eficiência de galharia para a indução da regeneração natural de espécies nativas em área coberta por gramíneas exóticas inibidoras do gênero Urochloa, bem como verificar o tamanho mínimo necessário para conter a reinvasão das forrageiras. Como hipótese, adotou-se a largura mínima de 4 m como suficiente para o estabelecimento da regeneração natural, antes da reocupação das forrageiras. O experimento foi conduzido entre maio 2014 e maio 2016 em Morretes-PR, na Floresta Ombrófila Densa de Terras Baixas. Foram implantados sete tamanhos de galharia: 6 x 1, 6 x 2, 6 x 3, 6 x 4, 6 x 5, 6 x 6 m e testemunha. As espécies lenhosas foram identificadas e contadas e a porcentagem de cobertura herbácea estimada após 4, 8, 12, 18 e 24 meses. Não se verificou retomada da sucessão natural por espécies nativas. Independentemente do tamanho, a galharia foi ineficiente para conter a reinvasão pelas gramíneas a partir das bordas das parcelas e, como pilha de resíduos, dificultou o estabelecimento de espécies lenhosas. Para a criação de safe sites e consequente restauração via regeneração natural, faz-se necessária a eliminação local das forrageiras Urochloa, sem a qual espécies nativas terão pouca probabilidade de sobreviver.

The recognized allelopathic activity of oats as a soil-cover plant induces the study of the potential of their germplasm. The objective of this study is to verify if there is genotype variability in the Avena spp. with regard to allelopathic activity. Extracts of A. strigosa ‘Agro Quaraí’, ‘Agro Esteio’, ‘Embrapa139’, ‘AF 12104’, ‘AF 12109’, ‘AF 12202’, ‘AF 12209’, A. sativa ‘UPFPS Farroupilha’ and ‘AF 1345 Ucraniana,’ were prepared in three concentrations w/v (5 g, 10 g, and 20 g of dry mass 100 mL-1). The experiment was conducted in a germination chamber for 15 days, with lettuce (Lactuca sativa L.) as the recipient plant. There was complete inhibition of germination under 20% w/v extracts and partial inhibition at 10% w/v. Only 5% w/v observed a difference between the genotypes, in which the highest efficiency was obtained from the extracts of white oats and black oats ‘AF 12104’ and ‘Agro Quaraí’, which reduced germination between 92% and 97%. When the concentration increased above 5% w/v the harmful effect on germination was significantly greater. Only in extracts at 5% w/v there was a higher genotypic effect on germination, with variation in the germination rate index, root, and hypocotyl stretching. The genotype that had the least allelopathic effect was AF 12202, with an inhibitory mean in the evaluated attributes of 35%. The other genotypes inhibited the attributes of germination and seedling growth between 66% and 86%. With regard to the variability in the Avenaspp., the allelopathic activity could be applied to obtain cultivars with greater weed suppressive power. RESUMO: A reconhecida atividade alelopática de aveias como plantas de cobertura do solo leva ao estudo do potencial de seus germoplasmas. O objetivo deste trabalho foi verificar se há variabilidade genotípica em Avena spp. quanto à atividade alelopática. Os extratos de A. strigosa Agro Quaraí, Agro Esteio, Embrapa 139, AF 12104, AF 12109, AF 12202, AF 12209 e de A. sativa UPFPS Farroupilha e AF 1345 Ucraniana foram preparados em três concentrações p/v (5, 10 e 20 g de massa seca 100 mL-1). O ensaio foi conduzido em câmara de germinação durante 15 dias, tendo alface (Lactuca sativa L.) como planta receptora. Houve total inibição da germinação sob extratos a 20% p/v einibição parcial a 10% p/v. Somente a 5% p/v observou-se diferença entre os genótipos, em que a maior eficiência foi dos extratos das aveias-brancas e das aveias-pretas AF 12104 e Agro Quaraí, que reduziram a germinação entre 92% e 97%. O efeito deletério sobre a germinação foi significativamente maior à medida que a concentração aumentou acima de 5% p/v, com variação sobre o índice de velocidade de germinação, alongamento radicial e de hipocótilo. O genótipo com menor efeito alelopático foi AF 12202, com inibição média de 35% nos atributos avaliados. Os demais genótipos inibiram a germinação e o crescimento de plântula entre 66% e 86%. A variabilidade em Avena spp. quanto à atividade alelopática pode ser aplicada para obtenção de cultivares com maior poder supressivo das plantas daninhas.

Endophytic fungi are a promising source for discovery of compounds with biotechnological potential. The aim of this study was to select and identify endophytic fungi from Coffea arabica that produce volatile organic compounds (VOCs), evaluate the effect of the VOCs produced by endophytic fungi on the growth of Rhizoctonia solani, Fusarium oxysporum, Phoma sp., Botrytis cinerea, Fusarium solani, Fusarium verticillioides, Cercospora coffeicola and Pestalotia longisetula, and select endophytic fungi with potential for biological control of Aspergillus ochraceus inoculated in coffee beans and F. verticillioides inoculated in corn seeds. An isolate of Muscodor albus was used as selection tool for VOC producing fungi. Among the 400 endophytic fungi isolates, 11 were able to grow in the presence of VOCs produced by M. albus. These fungi were identified as Muscodor spp. (9) and Simplicillium sp. according to searches in UNITE database using DNA sequences of internal transcribed spacer (ITS). The VOC's produced by endophytic fungi inhibited the growth the phytopathogenic fungi with different efficacies, compared to the control. The VOCs produced by Muscodor coffeanum (COAD 1842) showed fungicidal effect against A. ochraceus on coffee beans. Six endophytic fungi completely inhibited growth of F. verticillioides inoculated in corn seeds. This study demonstrates that the volatile-compound producing endophytic fungi, isolated from Coffea arabica, are promising sources of bioactive compounds. Fungos endofíticos são uma fonte promissora para a descoberta de compostos com potencial biotecnológico. O objetivo deste estudo foi selecionar e identificar fungos endofíticos de Coffea arabica que produzem compostos orgânicos voláteis (COVs), avaliar o efeito dos compostos orgânicos voláteis produzido por fungos endofíticos sobre o crescimento de Rhizoctonia solani, Fusarium oxysporum, Phoma sp., Botrytis cinerea, Fusarium solani, Fusarium verticillioides, Cercospora coffeicola e Pestalotia longisetula e selecionar fungos endofíticos com potencial para controle biológico de Aspergillus ochraceus inoculado em grãos de café e F. verticillioides inoculado em sementes de milho. Um isolado de Muscodor albus foi utilizado como ferramenta de seleção para fungos endofíticos produtores de COVs. Dentre os 400 fungos endofíticos isolados, 11 foram capazes de crescer na presença de COVs produzidos por M. albus. Estes fungos foram identificados como Muscodor spp. (9) e Simplicillium sp. de acordo com pesquisas na base de dados UNITE usando sequências de DNA do espaçador transcrito interno (ITS). Os COVs produzidos por fungos endofíticos inibiram o crescimento dos fungos fitopatogênicos em comparação com o controle com diferentes eficácias. Os COVs produzidos por Muscodor coffeanum (COAD 1842) apresentou efeito fungicida contra A. ochraceus em grãos de café. Seis fungos endofíticos inibiram completamente o crescimento de F. verticillioides inoculado em sementes de milho. Este estudo demonstra que os fungos endofíticos produtores de compostos voláteis isolados de Coffea arabica são fontes promissoras de compostos bioativos.

The aim of this work was to assess the inhibitory effect of essential oils on the growth and aflatoxin production of Aspergillus parasiticus, as well as to correlate it with the chemical composition of the essential oils. Essential oils from six aromatic species (Cymbopogon citratus, Eucalyptus globulus, Origanum vulgare, Ruta graveolens, Salvia officinalis, Satureja montana) were characterised by gas chromatography and tested for their inhibitory effect against A. parasiticus strain MUM 92.02. Furthermore, the in vitro inhibitory effects of these essential oils on the production of aflatoxins were evaluated by HPLC. Results showed that all essential oils retarded the time for visible growth. Growth rate was affected differently depending on the essential oil. S. montana essential oil prevented growth in all cases. The essential oil of R. graveolens inhibited most of the aflatoxin production even though growth inhibition was low, while with C. citratus essential oil trace levels of aflatoxins were detected. Essential oils containing carvacrol and/or thymol (S. montana and O. vulgare) have the highest activity against fungal growth, while an essential oil (R. graveolens) containing 2-undecanone and 8-phenyl-2-octanone inhibited the synthesis of aflatoxins. Although the main component of this essential oil was 2-undecanone (91%), when pure 2-undecanone was tested, it did not inhibit aflatoxin production. Inhibition activity is probably due to the recently identified minor compound or to a synergistic effect. Essential oils seem to be a good alternative to fungicides not only because of environmental issues but also because they do not seem to enhance mycotoxin production as it has been reported for some fungicides.

Allelopathy is analyzed as an alternative to herbicides due to the inhibitory or beneficent activities of its compounds with other organisms. Current paper discusses the effects of Aeschynomene fluminensis Vell. fractions on cultivated plant species, Lactuca sativa L. and Glycine max (L.) Merril, and on weeds, Ipomoea grandifolia (Dammer) O’Donnel and Digitaria insularis (L.) Fedde. Aqueous fractions at 0.80; 0.40; 0.20 and 0.10 mg mL-1 concentrations were employed for initial growth tests. Seeds were pre-germinated in distilled water and transferred to petri plates with separate fractions at different concentrations. Plates were maintained for 48 h in a germination chamber at 25°C for L. sativa and I. grandifolia and at 30°C for G.max and D. insularis. The length of hypocotyl (LH) and root (LR) was measured and LR and foliar length (LF) were taken for D. insularis seedlings. Parameters were employed to calculated inhibition percentage. Plants with morphological changes were fixed and analyzed anatomically. Results revealed LH and LR inhibition of lettuce seedlings in fractions with highest concentration rates. The same has been reported in the case of I. grandifolia. Butanolic, methanolic and chloroform fractions did not affect negatively soybean seedlings but they inhibited D. insularis seedlings´ LR. A. fluminensis fractions, especially at higher concentrations, inhibited seedlings´ growth and confirmed their phytotoxic capacity. La alelopatía es estudiada como una alternativa al uso de los herbicidas por la acción inhibidora o beneficiosa que los compuestos tienen sobre otros organismos. En el trabajo se analizaron los efectos de las fracciones de Aeschynomene fluminensis Vell. sobre especies cultivadas, Lactuca sativa L. y Glycine max (L.) Merril, y especies invasoras, Ipomoea grandifolia (Dammer) O’Donnel y Digitaria insularis (L.) Fedde. En las pruebas iniciales de crecimiento se utilizaron fracciones acuosas con concentraciones de 0,80; 0,40; 0,20 y 0,10 mg mL-1, pre-germinando las semillas en agua destilada, que luego fueron transferidas a placas Petri que contenían las fracciones, por separado, en las diferentes concentraciones. Las placas se mantuvieron durante 48 horas en una cámara de germinación a 25 ° C para L. sativa e I. grandifolia y 30 ° C para G. max y D. insularis. Luego de este período, se obtuvo la longitud del hipocótilo (CH) y raíz (CR) y para plántulas de D. insularis, la CR y la longitud de la hoja (CF). Estos parámetros se utilizaron para calcular el porcentaje de inhibición y las plantas que presentaron cambios morfológicos fueron fijadas y analizadas anatómicamente. Los resultados indicaron que hubo inhibición de CH y CR de plántulas de lechuga mantenidas en las fracciones de mayor concentración, observándose lo mismo para I. grandifolia. Las fracciones butanólica, metanólica y clorofórmica no afectaron negativamente a las plántulas de soja, sin embargo, estas mismas fracciones inhibieron la RC de las plántulas de D. insularis. Así, las fracciones de A. fluminensis, principalmente las de mayor concentración, inhibieron el crecimiento de las plántulas, confirmando su potencial fitotóxico. A alelopatia vem sendo estudada como uma forma alternativa ao uso de herbicidas por seus compostos terem ação inibitória ou benéfica a outros organismos. No trabalho foram analisados os efeitos das frações de Aeschynomene fluminensis Vell. sobre espécies cultivadas, Lactuca sativa L. e Glycine max (L.) Merril, e espécies invasoras, Ipomoea grandifolia (Dammer) O’Donnel e Digitaria insularis (L.) Fedde. As frações aquosas nas concentrações de 0,80; 0,40; 0,20 e 0,10 mg mL-1 foram utilizadas nos testes de crescimento inicial, realizando-se a pré-germinação das sementes em água destilada, que posteriormente foram transferidas para placas de Petri contendo as frações, separadamente, nas diferentes concentrações. As placas foram mantidas por 48 horas em câmara de germinação a 25°C para L. sativa e I. grandifolia e 30°C para G. max e D. insularis. Após este período foi obtido o comprimento do hipocótilo (CH) e da raiz (CR) e para plântulas de D. insularis, o CR e o comprimento foliar (CF). Esses parâmetros foram utilizados para o cálculo da porcentagem de inibição e as plantas que apresentaram alterações morfológicas, foram fixadas e analisadas anatomicamente. Os resultados indicaram que ocorreu inibição do CH e do CR das plântulas de alface mantidas nas frações de maior concentração, sendo o mesmo observado para I. grandifolia. As frações butanólica, metanólica e clorofórmica não afetaram negativamente as plântulas de soja, no entanto, essas mesmas frações inibiram o CR de plântulas de D. insularis. Assim, as frações de A. fluminensis, principalmente de maior concentração, inibiram o crescimento das plântulas, confirmando o seu potencial fitotóxico.

This study evaluated the antagonistic potential of 32 wild yeast isolates from coffee and cocoa bean fermentation. These yeasts were inoculated in co-cultivation with Aspergillus carbonarius (CCDCA 10608 and CCDCA 10408) and Aspergillus ochraceus (CCDCA 10612) isolated from grapes and coffee beans. The mycelial growth and ochratoxin A (OTA) production were evaluated, and the spores were counted after cultivation at 28°C for seven days. The yeasts presented higher inhibitory effects (53% in relation to the control) over the mycelial growth of the isolated A. ochraceus (CCDCA10612). Pichia anomala CCMA0148 and Saccharomyces cerevisiae CCMA0159 provided the greatest inhibition of the growth of all fungal strains. All Pichia species presented the highest inhibitory effects on the production of spores, and S. cerevisiae CCMA 0159 at concentrations of both 104 and 107 mL-1 cells inhibited the production of spores by 100%. Rhodotorula mucilaginosa was effective at inhibiting OTA production by the three isolates of Aspergillus. S. cerevisiae CCMA0159 and Pichia anomala CCMA0148 showed high potential as biocontrol agents in the conditions tested. RESUMO. Este estudo avaliou o potencial antagônico de 32 isolados de leveduras selvagens de fermentação de café e cacau. Estas leveduras foram inoculadas em co-cultivo com Aspergillus carbonarius (CCDCA 10608 e CCDCA 10408) e Aspergillus ochraceus (CCDCA 10612) isolado de uvas e grãos de café. O crescimento micelial e a produção de ocratoxina A (OTA) foram avaliados e os esporos foram contados após o cultivo a 28 ° C por sete dias. As leveduras apresentaram maiores efeitos inibitórios (53% em relação ao controle) sobre o crescimento micelial do isolado A. ochraceus (CCDCA10612). Pichia anomala CCMA0148 e Saccharomyces cerevisiae CCMA0159 proporcionaram a maior inibição do crescimento de todas as estirpes fúngicas. Todas as espécies de Pichia apresentaram os maiores efeitos inibitórios na produção de esporos e S. Cerevisiae CCMA 0159 em concentrações de 104 e 107 células mL-1 inibiram a produção de esporos em 100%. Rhodotorula mucilaginosa foi eficaz na inibição da produção de OTA pelos três isolados de Aspergillus. S. cerevisiae CCMA0159 e Pichia anomala CCMA0148 apresentaram alto potencial como agentes de biocontrole nas condições testadas.

The goal of this study was to restore an area dominated by P. aquilinum in the National Park of Caparaó, by nucleation techniques. In an area covered by P. aquilinum, 60 nuclei or gaps of 2 x 2 m were cleared, i.e., 10 nuclei per treatment. In these open areas, all P. aquilinum cover was removed down to the soil. In the center of each gap was delimited one parcel of 1 x 1 m, in which the treatments were applied: T1-liming, T2-seed bank and litter transposition, T3-direct-seeding, T4-liming + seed bank and litter transposition, T5-liming + direct-seeding, T6-control. The experiment was arranged in a completely randomized design with 10 replications. In each nucleation plot, after one year, all arbustive-arboreous species were recorded, by measuring plant height and diameter at ground level. The plants were classified according to the succession class and dispersion type. One year after the installation of the nucleation experiment, 331 individuals of 19 species, 12 genera and 10 botanical families were recorded. Pioneer plants were the predominant ecological group, and the prevailing dispersion type was zoochoric. The richest families were Solanaceae and Euphorbiaceae. Solanaceae was represented by the highest number of individuals (56.2%), followed by Fabaceae (10%). The genus Solanum had the highest richness and abundance. The applied nucleation techniques triggered the succession process in the area invaded by P. aquilinum. Treatments with seed banks were the most appropriate to facilitate forest regeneration in this area. RESUMO O estudo objetivou aplicar técnicas de nucleação para restauração de uma área dominada por P. aquilinum no Parque do Caparaó. Na área ocupada por P. aquilinum, abriram-se sessenta núcleos de 2 x 2 m, sendo dez núcleos para cada tratamento avaliado. Nestas áreas abertas, toda a cobertura de P. aquilinum foi removida até atingir o solo. No centro de cada clareira foi delimitado uma parcela de 1 x 1 m, em que foram aplicados os tratamentos: T1-calagem, T2-Transposição do banco de sementes e serapilheira, T3-semeadura direta, T4-calagem + transposição do banco de sementes e serapilheira, T5-calagem + semeadura direta, T6-Testemunha. Adotou-se o delineamento inteiramente casualizado com 10 repetições por tratamento. Em cada parcela da nucleação, após um ano, registraram-se todos os indivíduos arbustivo-arbóreos, medidos em altura e diâmetro ao nível do solo. As plantas foram classificadas quanto à classe sucessional e a síndrome de dispersão. Após um ano da implantação do experimento de nucleação, foram registrados 331 indivíduos arbustivo-arbóreos pertencentes a 19 espécies, 12 gêneros e dez famílias botânicas. Predominou o grupo ecológico das pioneiras e a síndrome de dispersão zoocórica. As famílias com maior riqueza foram Solanaceae e Euphorbiaceae. Solanaceae também se destacou com maior número de indivíduos (56,2%), seguida de Fabaceae (10%). O gênero Solanum apresentou maior riqueza e abundância. As técnicas de nucleação adotadas desencadearam o processo de sucessão na área invadida por P. aquilinum. Os tratamentos com banco de sementes foram os mais indicados para facilitar o processo de regeneração florestal na área.

The objective was to evaluate the germination and initial development of three cultivars of Brachiaria brizantha and Brachiaria decumbens in the presence of trace elements of Cd, Pb and Cu and quantify the Cd and Cu contents in plant tissue of these species. First, seed germination occurred in towel paper containing cadmium, lead, copper and the control. We calculated the percentage of germination, germination speed index, vigor index and seedling length. Later, the seeds were germinated in soil pots contaminated with Cd and Cu, where the Soil-Plant Analysis Development index (SPAD index) was evaluated, as well as dry matter of shoot and root, and the Cd and Cu metal content in plant tissue was quantified. B. decumbens showed sensitivity to the elements studied, with decreases in all parameters. Cu was the element that caused more toxic effects on germination and early seedling development of the species studied. In contaminated soil, the species studied showed no differences in dry matter production of shoots, but the exposure to Cd and Cu caused reductions in SPAD index in all species. The highest Cd and Cu contents were found in the roots. Resumo: O objetivo no trabalho foi avaliar a germinação e o desenvolvimento inicial de três cultivares de Brachiaria brizantha e a Brachiaria decumbens em presença dos elementos-traço Cd, Pb e Cu e quantificar os teores de Cd e Cu no tecido vegetal dessas espécies. A germinação das sementes ocorreu em papel germitest(r), contendo cádmio, chumbo, cobre além da testemunha. Foram calculadas a porcentagem de germinação, índice de velocidade de germinação, índice de vigor e comprimento da plântula. Posteriormente, as sementes foram germinadas em vasos de solo contaminado com Cd e Cu, onde se avaliou o índice Soil Plant Analysis Development (SPAD), matéria seca da parte aérea e da raiz e quantificado o teor de metais Cd e Cu no tecido vegetal. A B. decumbens apresentou sensibilidade aos elementos estudados, apresentando decréscimos de todos os parâmetros avaliados. O Cu foi o elemento que mais causou efeitos tóxicos à germinação e desenvolvimento inicial das plântulas das espécies estudadas. Em solo contaminado, as espécies estudadas não apresentaram diferenças na produção de massa seca de parte aérea, porém a exposição de Cd e Cu ocasionou reduções do índice SPAD em todas as espécies. Os maiores teores de Cd e Cu foram encontrados nas raízes.

Estudos de germinação após o fracionamento de sementes monoembriônicas de diversas espécies de Eugenia demonstraram seu potencial regenerativo. Entretanto, a formação de novas plântulas não ocorre de maneira espontânea em sementes intactas, sugerindo a presença de inibidores de germinação na semente germinante. Para investigar a presença de possíveis inibidores, seis concentrações de extratos de sementes de pitangueira (Eugenia uniflora) foram utilizados em testes de germinação, além do controle (água pura). O possível efeito osmótico desses extratos foram aferidos com soluções de PEG 6000 e de NaCl, de -0,05 a -0,40 MPa. Os extratos de pitanga não apresentaram inibição de germinação para a própria espécie. Contudo, para aquênios de alface e para sementes de feijão os resultados apresentaram diferenças significativas, revelando potencial inibidor da germinação e do desenvolvimento inicial das plântulas. Simulando-se os potenciais hídricos dos extratos de pitanga que causaram inibição da germinação, quando as soluções foram preparadas com PEG 6000 houve semelhante inibição, mas não quando as soluções foram preparadas com NaCl. Esta diferença de comportamento apresentada pelos diferentes solutos sugere que soluções de PEG 6000 resultem em alterações não apenas do potencial hídrico. Germination studies of sectioned seeds of Eugenia species demonstrated their potential for regeneration. However, new seedlings are not spontaneously produced by intact seeds, suggesting the presence of germination inhibitors in the seed. Germination tests were made with six extracts prepared from Surinam cherry seeds (Eugenia uniflora) with pure water as the control. The osmotic effects of these extracts were also investigated using solutions of both PEG 6000 and NaCl, varying from -0.05 to -0.4 MPa. Surinam cherry seed extracts did not inhibit germination of the species itself but they showed a potential for inhibiting germination and normal seedling development in lettuce and dry beans. Considering osmotic potentials similar to those of the inhibiting extracts, PEG solutions produced similar effects, but this was not observed for the NaCl solutions. This difference in behavior by the different solutes suggests effects other than osmotic ones for PEG solutions.

We investigated the inhibitory potential of aqueous extracts of bark and leaves of Esenbeckia leiocarpa Engl. on lettuce germination and early seedling growth. We compared the effects of four concentrations (100, 75, 50 and 25%) of each extract to water and polyethylene glycol (PEG 6000) solution controls for four replicates of 50 seeds for germination and four replicates of ten seedlings for seedling growth. The inhibitory effects of E. leiocarpa extracts on the percentage of germination and on the germination speed seemed to be more than simply an osmotic effect, except for the percentage of seeds germinated in bark extracts. When compared to water control, both bark and leaf extracts delayed germination, and leaf extracts also affected the percentage of germinated seeds. Leaf extracts of all concentrations strongly inhibited the development of seedlings and caused them some degree of abnormality; bark extracts also caused abnormalities and reduced seedling growth. Root development was more sensitive to the extracts than hypocotyl growth. The negative effects of leaf extracts on germination and seedling growth were more pronounced than those of bark extracts, and the overall effects of both extracts were positively correlated with extract concentrations. Neste trabalho, nós investigamos o potencial inibitório de extratos aquosos de folhas e cascas de Esenbeckia leiocarpa Engl. na germinação e no crescimento inicial de plântulas de alface. Nós comparamos os efeitos de quarto concentrações (100, 75, 50 e 25%) de cada extrato a um controle em água e outro em uma solução de polietilenoglicol (PEG 6000), com quatro repetições de 50 sementes para o experimento de germinação e quatro repetições de 10 plântulas para o experimento de crescimento. Os efeitos inibitórios dos extratos de E. leiocarpa na porcentagem e na velocidade de germinação foram mais do que um efeito do potencial osmótico das soluções, exceto para a porcentagem de sementes germinadas nos extratos de casca. Ambos os extratos causaram atrasos na germinação, sendo que os extratos de folha afetaram também a porcentagem de sementes germinadas. Os extratos de folha, em todas as concentrações, inibiram fortemente o desenvolvimento das plântulas e causaram a todas elas algum grau de anormalidade; os extratos de casca também causaram anormalidades e reduziram o crescimento das plântulas. O desenvolvimento da radícula foi mais sensível à ação dos extratos do que o crescimento do hipocótilo. Os efeitos negativos dos extratos de folhas foram mais pronunciados do que os causados pelos extratos de casca e os efeitos de ambos os extratos foram sempre positivamente correlacionados com sua concentração.

Human parasitic diseases are the foremost threat to human health and welfare around the world. Trypanosomiasis is a very serious infectious disease against which the currently available drugs are limited and not effective. Therefore, there is an urgent need for new chemotherapeutic agents. One attractive drug target is the major cysteine protease from Trypanosoma cruzi, cruzain. In the present work, comparative molecular field analysis (CoMFA) and comparative molecular similarity indices analysis (CoMSIA) studies were conducted on a series of thiosemicarbazone and semicarbazone derivatives as inhibitors of cruzain. Molecular modeling studies were performed in order to identify the preferred binding mode of the inhibitors into the enzyme active site, and to generate structural alignments for the three-dimensional quantitative structure-activity relationship (3D QSAR) investigations. Statistically significant models were obtained (CoMFA, r2=0.96 and q2=0.78; CoMSIA, r2=0.91 and q2=0.73), indicating their predictive ability for untested compounds. The models were externally validated employing a test set, and the predicted values were in good agreement with the experimental results. The final QSAR models and the information gathered from the 3D CoMFA and CoMSIA contour maps provided important insights into the chemical and structural basis involved in the molecular recognition process of this family of cruzain inhibitors, and should be useful for the design of new structurally related analogs with improved potency.

Medulloblastoma (MB) is the most common malignant childhood brain tumour. Aurora kinases are essential for cell division and are primarily active during mitosis. Recently, the combination of aurora kinases inhibitors (iAURK) and histone deacetylase inhibitors (iHDAC) has shown potential antitumour effects and had significant biological effects in preclinical cancer models. In this study, we analysed the effects of the pan-aurora kinases inhibitor AMG 900 alone or in combination with the iHDAC SaHa (Vorinostat) on paediatric MB cell lines (UW402, UW473 and ONS-76).Cell proliferation was measured by XTT assay, apoptosis was determined by flow cytometry and clonogenic capacity was studied. qRT-PCR assays were used to determine the mRNA expression in MB cell lines after treatment. Drug combination analyses were made based on Chou-Talalay method.AMG 900 caused the inhibition of cell proliferation, diminution of clonogenic capacity and increased the apoptosis rate in cell lines (P0.05). A synergistic effect in the AMG900-SaHa combination was evidenced on the inhibition of cell proliferation in all cell lines, especially in sequential drug treatment. Moreover, the combination of these drugs reached 100% of the inhibition in colony formation (synergistic effect). The treatment with AMG 900 increased the p21 and GDF15 expression, but did not alter the TP53 in one of the cell lines.These results indicate that AMG 900 may be a promising drug for the adjuvant treatment of MB, mainly when combined with iHDAC.

Processos convencionais de tratamento de efluentes utilizam microrganismos vivos, o que sugere limitações relativas À toxicidade de metais para os microrganismos. O experimento consistiu em adicionar soluções monoelementares de Cr (VI) e Zn(II) em diferentes concentrações (0, 20, 50, 100, 200, 300, 400, 500 mg.L-1) ao meio de crescimento e observar a influência dos metais no crescimento micelial e germinativo do fungo Aspergillus Níger por verificação visual da expansão radial do micélio e da germinação de esporos, seguida de registro fotográfico. Os resultados mostraram que o metabolismo do fungo foi completamente inibido em concentrações acima de 500 mg Zn (II).L-1 e 150 mg Cr (VI).L-1. O ED50 (concentração de ingrediente ativo capaz de inibir 50% do crescimento micelial do fungo) para os dois íons metálicos, nas condições estudadas, está na faixa entre 100 e 150 mg.L-1. Many standard processes of wastewater treatment use live microorganisms, which suggests limitations on a metal toxicity to the microorganism. The experiment consisted in adding mono elementary solutions of Cr (VI) and Zn (II) at different concentrations (0, 20, 50, 100, 200, 300, 400, 500 mg.L-1) to the growth mean, and to observe the influence of metals on mycelial and germinative growth of the Aspergillus niger fungus, by means of visual observation of the radial expansion of the mycelius and the germination of spores, followed by photograph registration. The results showed that the metabolism of the fungus was completely inhibited at concentrations above 500 mg Zn (II).L-1 and 150 mg Cr (VI).L-1. The ED50 (concentration of active ingredient capable of inhibiting 50% of mycelial growth of the fungus) for both metal ions, under the studied conditions, is in the range between 100 and 150 mg.L-1.

Background Cancer shows a great diversity in its clinical behavior which cannot be easily predicted using the currently available clinical or pathological markers. The identification of pathways associated with lymph node metastasis (N+) and recurrent head and neck squamous cell carcinoma (HNSCC) may increase our understanding of the complex biology of this disease. Methods Tumor samples were obtained from untreated HNSCC patients undergoing surgery. Patients were classified according to pathologic lymph node status (positive or negative) or tumor recurrence (recurrent or non-recurrent tumor) after treatment (surgery with neck dissection followed by radiotherapy). Using microarray gene expression, we screened tumor samples according to modules comprised by genes in the same pathway or functional category. Results The most frequent alterations were the repression of modules in negative lymph node (N0) and in non-recurrent tumors rather than induction of modules in N+ or in recurrent tumors. N0 tumors showed repression of modules that contain cell survival genes and in non-recurrent tumors cell-cell signaling and extracellular region modules were repressed. Conclusions The repression of modules that contain cell survival genes in N0 tumors reinforces the important role that apoptosis plays in the regulation of metastasis. In addition, because tumor samples used here were not microdissected, tumor gene expression data are represented together with the stroma, which may reveal signaling between the microenvironment and tumor cells. For instance, in non-recurrent tumors, extracellular region module was repressed, indicating that the stroma and tumor cells may have fewer interactions, which disable metastasis development. Finally, the genes highlighted in our analysis can be implicated in more than one pathway or characteristic, suggesting that therapeutic approaches to prevent tumor progression should target more than one gene or pathway, specially apoptosis and interactions between tumor cells and the stroma.

As inúmeras espécies de plantas que compõem a floresta amazônica podem representar excelente alternativa para fazer frente ao desafio de desenvolver a agricultura conforme as exigências da sociedade. Neste trabalho, procurou-se determinar e caracterizar o padrão de atividade alelopática em espécies da família Leguminosae, em função de variações de espécies, fonte de extratos e sensibilidade da planta receptora. Bioensaios de germinação de sementes e alongamento da radícula e do hipocótilo foram desenvolvidos em condições controladas. Os resultados indicam que as espécies estudadas não apresentaram padrão semelhante no tocante aos efeitos potenciais alelopáticos, havendo, entretanto, hierarquização no tocante à intensidade dos efeitos globais, sendo o potencial alelopático inibitório mais amplo e efetivo nas espécies Bauhinia guianensis, Bowdichia virgiloides, Parkia pendula e Platimenia reticulata. O potencial alelopático foi efetivo e mais restrito em Bauhinia macrostachya. O fator fração das plantas revelou diferenciação no padrão de atividade: para a maioria das espécies, as folhas foram a principal fonte de aleloquímicos, e para Bauhinia macrostachya e Inga edulis, a raiz. Em termos de padrão de respostas das espécies receptoras, o alongamento da radícula é mais sensível aos efeitos dos extratos, ficando o alongamento do hipocótilo como o de menor sensibilidade. Os efeitos dos extratos foram mais intensos sobre Mimosa pudica. Esses resultados também atribuem à floresta amazônica importância como fonte de compostos químicos de interesse para o homem, o que, em si, justifica sua preservação. Innumerous plant species in the Amazon forest may be an excellent alternative to meet society's demand for natural products. This work aimed to determine and characterize the pattern of allelopathic activity in species of the Leguminosae family, according to species variation, extract source, and receptor plant sensitivity. Bio assays of seed germination and radicle and hypocotyl elongation were developed under controlled conditions. The results indicated that the species studied do not present a similar pattern regarding the potential allelopathic effects; however, there is a hierarchy concerning the intensity of the global effects, with the allelopathic inhibitory potential being more extensive and effective for the species Bauhinia guianensis, Bowdichia virgiloides, Parkia pendula and Platimenia reticulate, and more restricted and effective for Bauhinia macrostachya. The plant fraction factor showed differences in the activity pattern, with the leaves being the main source of allelochemicals for most species, and the roots, for Bauhinia macrostachya and Inga edulis. In terms of response pattern of the receptor plants, radicle elongation is more sensitive to the effects of the extracts, with hypocotyl elongation being the least sensitive. The effect of the extracts was more intense for Mimosa pudica. These results confirm the importance of the Amazon forest as a source of chemical compounds of interest to mankind, which by itself justifies its preservation.

Nos últimos anos, atenção especial foi dada aos compostos químicos envolvidos na interação entre plantas, especialmente quando se sabe das possibilidades do seu uso em estratégia de manejo de plantas daninhas. A Amazônia, pela sua megabiodiversidade e abundância de espécies vegetais, pode oferecer excelente oportunidade para a descoberta de inovadoras moléculas químicas com potencial de uso na atividade agrícola. Dessa forma, neste trabalho analisou-se, comparativamente, a atividade potencialmente alelopática de três espécies de Copaifera, caracterizando-se as variações na intensidade dos efeitos alelopáticos em função da espécie doadora, da fração da planta e da polaridade dos constituintes químicos. Extratos hexânico e etanólico, preparados a 1,0% a partir de folhas, galhos e cascas de Copaifera duckei, C. martii e C. reticulata, foram testados sobre a germinação de sementes e o desenvolvimento da raiz das plantas daninhas malícia (Mimosa pudica) e mata-pasto (Senna obtusifolia). Observaram-se variações nas intensidades dos efeitos em função das variáveis estudadas. O extrato etanólico de folhas e o de galhos de C. martii e C. reticulata apresentaram alto potencial para inibir a germinação de sementes, sobretudo da espécie malícia. A espécie C. duckei evidenciou baixo potencial alelopático inibitório na germinação das duas espécies receptoras. Cascas, folhas e galhos de C. duckei apresentaram potencial inibitório mais expressivo sobre o desenvolvimento da raiz, com destaque para as folhas. Compostos químicos apolares e polares estão envolvidos na atividade alelopática da espécie C. duckei, com ênfase maior para os compostos apolares. Diferentemente, para C. martii e C. reticulata, compostos polares estão envolvidos, preferencialmente, na atividade inibitória evidenciada por essas espécies, notadamente aqueles localizados nas folhas e cascas. Comparativamente, a tendência observada foi de que a espécie receptora malícia demonstrou maior sensibilidade aos efeitos alelopáticos dos extratos, especialmente no bioensaio de germinação. For the past few years, special attention has been given to chemical composites involved in interaction between plants, especially when the potential of their use in weed management is known. The Amazon region with its enormous biodiversity and abundance of plant species may offer excellent opportunities for the discovery of innovative chemical molecules capable of being used in agriculture. Thus, in this paper, a comparative analysis was made between the potential allelopathic activity of three species of Copaifera, characterizing the variations of the intensity of the allelopathic effects of the donating species, plant fraction and chemical component polarity. Hexanic and ethanolic extracts, prepared at 1.0% from the leaves, branches and barks of Copaifera duckei, C. martii and C. reticulata, were tested on seed germination and development of Mimosa pudica and Senna obtusifolia weed roots. The results showed that the intensity of the effects varied according to the variables studied. The ethanolic extract of C. martii and C. reticulata leaves and barks showed high potential to inhibit seed germination, especially of M. pudica species. The species C. duckei showed low allelopathic inhibitory potential in the germination of two receptor species. Barks, branches and especially leaves of C. duckei presented more expressive inhibitory potential for root development. Polar, and especially non-polar chemical composites, are involved in the allelopathic activity of C. duckei. Differently for C. martii and C. reticulata, the polar composites are preferentially involved in the inhibitory activity evidenced by these species, especially those located on the leaves and barks. Comparatively, it was observed that the weed species receptor tended to show more sensitivity to the allelopathic effects of the extracts, especially in the germination bioassay.

The search for new and potent cholinesterase inhibitors is an ongoing quest mobilizing many organic chemistry groups around the world as these molecules have been shown to treat the late symptoms of Alzheimer's disease as well as to act as neuroprotecting agents. In this work, we disclose the synthesis of novel 2-acetamidopurine nucleosides and, for the first time, regioselective N(7)-glycosylation with 2-acetamido-6-chloropurine, promoted by trimethylsilyl triflate, was accomplished by tuning the reaction conditions (acetonitrile as solvent, 65 degrees C, 5h) starting from 1-acetoxy bicyclic glycosyl donors, or by direct coupling of a methyl glucopyranoside with the nucleobase to obtain only N(7) nucleosides in reasonable yield (55-60%). The nucleosides as well as their sugar precursors were screened for acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) inhibition. While none of the compounds tested inhibited AChE, remarkably, some of the N(7) nucleosides and sugar bicyclic derivatives showed potent inhibition towards BChE. Nanomolar inhibition was obtained for one compound competing well with rivastigmine, a drug currently in use for the treatment of Alzheimer's disease. Experimental results showed that the presence of benzyl groups on the carbohydrate scaffold and the N(7)-linked purine nucleobase were necessary for strong BChE inactivation. A preliminary evaluation of the acute cytotoxicity of the elongated bicyclic sugar precursors and nucleosides was performed indicating low values, in the same order of magnitude as those of rivastigmine.

A alelopatia é um importante mecanismo que influencia a estabilidade de agroecossistemas. A identificação desse caráter, em muitos casos, realiza-se via análise dos efeitos de extratos brutos polares. No presente trabalho, caracterizou-se a atividade alelopática do cipó-d'alho (Mansoa standleyi - Bignoniaceae), analisando-se, comparativamente, o extrato hidroalcoólico e o óleo essencial das folhas da planta, procurando-se estabelecer a necessidade de se considerar, em estudos dessa natureza, a abordagem envolvendo extratos apolares. Realizaram-se bioensaios de germinação e desenvolvimento da radícula e do hipocótilo de malícia (Mimosa pudica), em períodos de dez dias, empregando-se concentrações de 0,5%, 1,0% e 2,0%. Foram identificados, ainda, os principais constituintes químicos do óleo essencial. Os resultados indicaram que tanto o extrato hidroalcoólico como o óleo essencial apresentaram potencial para inibir a germinação e o desenvolvimento da radícula e do hipocótilo. O extrato hidroalcoólico manifestou maior potencial inibitório sobre a germinação, enquanto o óleo essencial promoveu inibições mais expressivas sobre o desenvolvimento da radícula e do hipocótilo. Os efeitos estiveram positivamente associados à concentração, com efeitos máximos e mínimos obtidos nas concentrações de 2,0% e 0,5%, respectivamente. Compostos sulfurados, como o dissulfeto de dialila (42,15%) e o trissulfeto de dialila (11,25%), isoladamente ou em associação, estão envolvidos nos efeitos alelopáticos promovidos pelo óleo essencial. Adicionalmente, os resultados obtidos apontam para a necessidade de se considerar a utilização de extratos apolares quando da análise da atividade alelopática de uma dada planta, especialmente se não houver informações sobre a produção de óleo essencial pela planta prospectada. Allelopathy is an important mechanism that affects the stability of agro-eco systems. The identification of this character, in several cases, is accomplished via analysis of the effects of polar crude extracts. The allelopathic activity of the species "cipó-de-alho"(Mansoa standleyi - Bignoniaceae) is characterized in the present work, by comparing the effects of the hydroalcoholic extract and essential oil of the leaves, attempting to establish the need to consider in research of this nature the approach involving non polar extracts. Bioassays of the weed plant 'malicia'' (Mimosa pudica) radicle and hypocotyl germination and growth were carried out in periods of 10 days, using concentrations of 0.5%; 1.0% and 2.0%. The chemical constituents of the essential oil were also identified. The results showed that both the hydroalcoholic extract and the essential oil have the potential to inhibit radicle and hypocotyl germination and growth. The hydroalcoholic extract showed higher potential to inhibit germination, while the essential oil promoted more intense inhibitions on radicle and hypocotyl growth. The effects were positively related with concentration, with maximum and minimum effects being obtained at concentrations of 2.0% and 0.5%, respectively. The sulfur compounds diallyl disulfide (42.15) and diallyl trisulfide (11.25%), alone or in association, are involved with the allelophatic effects caused by the essential oil. In addition, the results point to the need to consider using non polar extracts when evaluating the allelopathic activity of a given plant, especially when there is no information about the production of essential oil by the prospected plant.

O extrato bruto etanólico foi preparado na proporção de 100 g de material vegetal seco para 500 mL de etanol. A partir deste, os extratos foram solubilizados em solução tampão e dimetil sulfóxido (DMSO), nas concentrações 10,0; 7,5; 5,0 e 2,5 mg/mL. Foi avaliado o crescimento do sistema radicular, da parte aérea e das células do metaxilema radicular das plântulas. Os extratos etanólicos de folhas e frutos de B. oxyclada exerceram atividade inibitória no crescimento radicular de plântulas de capim-arroz e amendoim-bravo, com efeito dependente da concentração. Foi observada redução no crescimento da parte aérea apenas nas plântulas de amendoim-bravo. A redução no crescimento radicular das plântulas de amendoim-bravo pode estar relacionada à diminuição no alongamento das células do metaxilema. Banisteriopsis oxyclada apresenta propriedades itotóxicas e pode ser considerada uma alternativa no controle das plantas daninhas estudadas, com base em produtos naturais.

Os métodos usuais de controle de plantas daninhas não atendem mais as atuais e futuras exigências da sociedade em relação à preservação dos recursos naturais e da qualidade de vida. Uma alternativa para essa questão seriam os metabólitos secundários produzidos pelas plantas, que apresentam pouco risco para o ambiente e para os interesses da sociedade. Os objetivos deste trabalho foram isolar, identificar e caracterizar a atividade alelopática de substâncias químicas produzidas pela gramínea forrageira Brachiaria humidicola. Analisaram-se os efeitos alelopáticos dos extratos, frações e substâncias isoladas sobre a germinação e o desenvolvimento da radícula das invasoras malícia, fedegoso e mata-pasto, em bioensaios monitorados em períodos de 10 dias, em condições de 25 ºC e fotoperíodo de 12 horas, para a germinação, e 24 horas, para o desenvolvimento da radícula. A partir do extrato hidrometanólico, foi isolado e identificado o ácido p-cumárico. Os efeitos alelopáticos estiveram positivamente relacionados à concentração do ácido, à espécie de planta daninha e à característica da espécie analisada. Comparativamente, fedegoso e malícia se mostraram mais sensíveis aos efeitos alelopáticos. A germinação e o desenvolvimento da radícula do mata-pasto não foram afetados pelo ácido p-cumárico nas concentrações de 1,0 a 8,0 mg L¹. O alongamento da radícula se mostrou mais sensível aos efeitos alelopáticos do ácido pcumárico do que a germinação das sementes. The traditional methods of weed control in cultivated pasture do not meet society's present and future needs of protecting natural resources and life quality. An alternative to this problem could be the use of secondary metabolites produced by plants. These products present few risks to the environment and meet society interests. Therefore, this research was carried out to isolate, identify and characterize the allelopathic activity of chemical compounds produced by the forage grass Brachiaria humidicola. The allelopathic effects of the extracts, fractions and compound were tested on seed germination and root elongation of the weeds Mimosa pudica, Senna obtusifolia and Senna occidentalis. Germination bioassays were developed under 25 ºC and a photoperiod of 12 hours. For root elongation, the bioassay conditions were 25 ºC and photoperiod of 24 hours. Hydromethanolic extract was used as a source for isolating and identifying p-coumaric acid. The allelopathic effects were positively related to p-coumaric acid concentration, weed species and the evaluated parameter. Comparatively, S. occidentalis and M. pudica showed the greatest sensitivity to the allelopathic effects. For S. obtusifolia no allelopathic effects promoted by p-coumaric acid on seed germination or on root elongation could be detected under the concentration of 1.0 and 8.0 mg L-1. Root elongation was more sensitive to p-coumaric acid allelophatic effects than seed germination.

Four Lentinula edodes strains (Le10, 46, K2, Assai) were assessed for their antagonistic effect on four filamentous fungus species of agricultural importance (Helminthosporium euphorbiae, Helminthosporium sp, Fusarium solani and Phomopsis sojae) and on Alagoas serotype of Vesicular Stomatitis Virus (VSA). The L. edodes strains studied had variable effects on the filamentous fungi and on VSA. The K2 and Le10 strains were antagonistic on the fungi assessed and the 46 and K2 strains were efficient on the Vesicular Stomatitis Virus. The results widened the list of beneficial effects of L. edodes on the control and prevention of animal pathogenic virus and filamentous fungi. Quatro linhagens de Lentinula edodes (Le10, 46, K2, ASSAI) foram avaliadas quanto ao seu efeito inibitório sobre quatro espécies de fungos filamentosos de importância agrícola (Helminthosporium euphorbiae, Helminthosporium sp., Fusarium solani, Phomopsis sojae) e sobre o sorotipo Alagoas vírus da estomatite vesicular (VSA). Foi observado que as linhagens de L. edodes estudadas apresentaram variabilidade quanto ao seu efeito, tanto sobre os fungos filamentosos quanto sobre o vírus VSA. As linhagens K2 e Le10 apresentaram-se antagônicas sobre os fungos e as linhagens 46 e K2 foram eficientes na inibição do vírus VSA. Os resultados obtidos permitem ampliar a lista de efeitos benéficos de algumas linhagens de L. edodes no controle e prevenção de vírus patogênicos animais e de fungos filamentosos.

Neste artigo, a partir de uma perspectiva psicanalítica, realiza-se uma discussão acerca da evolução clínica do tratamento de um menino de sete anos que apresentava um quadro de inibição da escrita, com o objetivo de evidenciar como, no decorrer do processo de análise, ocorreu uma modificação no que diz respeito ao agenciamento da angústia vivida pelo paciente. Conclui-se que, a partir de certas intervenções pontuais, a angústia inicialmente apresentada pelo paciente diante da falha da função paterna deu lugar à angústia frente ao processo de subjetivação da castração, culminando na assunção de uma tomada de posição na partilha sexual.

Xanthomonas oryzae pv. oryzae (Xoo), a pathogen responsible for rice bacterial leaf blight, produces biofilm to protect viable Xoo cells from antimicrobial agents. A study was conducted to determine the potency of Acacia mangium methanol (AMMH) leaf extract as a Xoo biofilm inhibitor. Four concentrations (3.13, 6.25, 9.38, and 12.5 mg/mL) of AMMH leaf extract were tested for their ability to inhibit Xoo biofilm formation on a 96-well microtiter plate. The results showed that the negative controls had the highest O.D. values from other treatments, indicating the intense formation of biofilm. This was followed by the positive control (Streptomycin sulfate, 0.2 mg/mL) and AMMH leaf extract at concentration 3.13 mg/mL, which showed no significant differences in their O.D. values (1.96 and 1.57, respectively). All other treatments at concentrations of 6.25, 9.38, and 12.5 mg/mL showed no significant differences in their O.D. values (0.91, 0.79, and 0.53, respectively). For inhibition percentages, treatment with concentration 12.5 mg/mL gave the highest result (81.25%) followed by treatment at concentrations 6.25 and 9.38 mg/mL that showed no significant differences in their inhibition percentage (67.75% and 72.23%, respectively). Concentration 3.13 mg/mL resulted in 44.49% of biofilm inhibition and the positive control resulted in 30.75% of biofilm inhibition. Confocal laser scanning microscopy (CLSM) analysis of Xoo biofilm inhibition and breakdown showed the presence of non-viable Xoo cells and changes in aggregation size due to increase in AMMH leaf extract concentration. Control slides showed the absence of Xoo dead cells. Resumo Xanthomonas oryzae pv. oryzae (Xoo), um patogênico responsável pela influência bacteriana na folha do arroz, produz biofilme para proteger células Xoo viáveis de agentes antimicrobianos. Foi conduzido um estudo para determinar a potência do extrato de folha de Acacia mangium methanol (AMMH) como um inibidor de biofilme Xoo. Quatro concentrações (3,13, 6,25, 9,38 e 12,5 mg/mL) de extrato de folha de AMMH foram testadas quanto à sua capacidade de inibir a formação de biofilme Xoo em uma placa de microtitulação de 96 poços. Os resultados mostraram que os controles negativos tiveram o maior valor de OD do que os outros tratamentos, indicando a intensa formação de biofilme. Isso foi seguido do controle positivo (sulfato de estreptomicina, com concentração de 0,2 mg/mL, e extrato de folha de AMMH, com concentração de 3,13 mg/mL), que não apresentou diferenças significativas nos seus valores OD (1,96 e 1,57, respectivamente). Todos os outros tratamentos com concentrações de 6,25, 9,38, e 12,5 mg/mL não tiveram diferenças significativas nos seus valores OD (0,91, 0,79, e 0,53, respectivamente). Para percentagens de inibição, o tratamento com concentração 12,5 mg/mL apresentou o maior resultado (81,25%), seguido do tratamento em concentrações de 6,25 e 9,38 mg/mL, que não mostraram diferenças significativas na sua percentagem de inibição (67,75 e 72,23%, respectivamente). Concentração 3,13 mg/mL resultou em 44,49% de inibição do biofilme, e o controle positivo resultou em 30,75% de inibição do biofilme. Análise por microscopia confocal de leitura a laser de inibição e separação de biofilme Xoo revelou a presença de células Xoo não viáveis e alterações no tamanho da agregação por causa do aumento na concentração de extrato de folha de AMMH. Slides de controle mostraram a ausência de células Xoo mortas.
