We investigated the distribution of neurons in the substantia nigra pars reticulata (SNr) which received cortical input. The activities of single SNr neurons were studied extracellulary in awake monkeys. SNr neurons showed excitatory and/or inhibitory responses to cortical stimulation. These responses were considered to be mediated by the subthalamic nucleus and striatum, respectively. The neurons receiving inhibitory input from the motor, premotor and supplementary motor areas (Motor-related cortical areas) were located in the lateral part of the SNr, whereas those with input from the medial, dorsal and orbital areas of the prefrontal cortex (PFmdo) were frequently found in the rostro-medial part of this nucleus. SNr neurons with inhibitory input from the ventral periprincipal area (PSv) were mainly distributed in the intermedio-lateral portion, with some degree of overlap with input from other cortical areas. The distribution of the excitatory input was almost similar to that of inhibitory one, but the excitatory input from the PSv was much stronger than that from the PFmdo. Some SNr neurons receiving cortical input were proved to project to the thalamus. Our results support the existence of several parallel organization of the cortico-basal ganglia loop circuits [G.E. Alexander, M.R. DeLong, P.L. Strick, Parallel organization of functionally segregated circuits linking basal ganglia and cortex, Ann. Rev. Neurosci., 9, 1986, pp. 357-381.], but interaction between the loops can not be ignored.

Electrical stimulation of the mediodorsal nucleus (MD) of the thalamus elicited field potentials in the gyrus proreus (PRO), frontalis (FR), rectus (RE) and cinguli anterior (CIant) of the ipsilateral prefrontal and adjacent cortical areas in cats. The results of a laminar field potential analysis indicate that the field potentials can be regarded as a combination of deep and superficial thalamocortical responses. By injecting horseradish peroxidase (HRP) into the MD, HRP-labeled terminals were distributed in the prefrontal and adjacent cortical areas where the field potentials were elicited. Densely labeled terminals in cortical layer I were distributed where the superficial thalamocortical responses were prominent, while those in layers III-V were distributed in the areas where the deep thalamocortical responses were prominent.

A few studies have been performed in primate basal ganglia–thalamo–prefrontal pathways. Nevertheless, their electrophysiological properties and anatomical arrangements remain obscure. The present report investigated the involvement of primate nigro-thalamo-cortical projections in discrimination of visual signals with behavioral meaning. We tested the extracellular unit activity of mediodorsal (MD) and ventral anterior (VA) thalamic neurons monosynaptically receiving inhibitory input from the substantia nigra pars reticulata (SNr) and projecting to the frontal cortex in Japanese monkeys performing a delayed conditional go/no-go discrimination task. In the task two colored stimuli (S1, S2) intervened by delay period required the monkeys lifting a lever (go) or not (no-go); the same and different colored pairs of S1 and S2 meant go and no-go signals, respectively. Prominent task-relevant responses were sustained activity with color preference to S1 during delay period and S2-related activity with different firing rates between go and no-go trials. In particular, a high proportion of such go/no-go differential S2-related activity was found in thalamic relay neurons, receiving input from the caudolateral SNr and projecting to the prefrontal area (PSv) ventral to the principal sulcus, in the rostrolateral MD. The findings suggest that the caudolateral SNr–rostrolateral MD–PSv pathways may be possible conduits of signals coding the behavioral meaning of the visual stimuli and thus may be responsible for generating similar neuronal activity in the PSv.

Projections from the cerebellar nuclei (CN) and the internal segment of the globus pallidus (GPi) to areas 6 and 4 via the thalamus were examined electrophysiologically in monkeys. In addition to the well-known pallido-thalamo-cortical projection to area 6, some thalamic neurons with CN input were found to project to area 6. Seventeen neurons in VLc, area X and the dorsal margin of VPLo were activated antidromically by stimulation of supplementary motor or premotor areas, and orthodromically by CN stimulation. Twelve neurons in area X and the medial part of VPLo were found to receive CN input and to project to the arcuate premotor area (APA). Four neurons in the VLo-X border region which were identified to project to APA were inhibited by GPi stimulation.
