Introduction The functional relevance of intra-species diversity in natural microbial communities remains largely unexplored. The guts of two closely related honey bee species, Apis cerana and A. mellifera, are colonised by a similar set of core bacterial species composed of host-specific strains, thereby providing a good model for an intra-species diversity study. Objective and method We used shotgun metagenomic sequencing to assess the functional relevance of intra-species diversity of A. cerana and A. mellifera gut microbiota. Results Comparative analysis showed that the strain composition of the core bacterial species was host-specific. These core bacterial species presented distinctive functional profiles between the hosts. However, the overall functional profiles of the A. cerana and A. mellifera gut microbiomes were similar; this was further supported by the consistency of the honey bees’ gut metabolome, as the gut microbiota of different honey bee species showed rather similar metabolic profiles in the cross-species gut bacteria colonisation assay. Moreover, this experiment also demonstrated that the gut microbiota of A. cerana and A. mellifera could cross colonise between the two honey bee species. Conclusion In summary, our findings revealed functional differences in most core gut bacteria between the guts of A. cerana and A. mellifera, which may be associated with their inter-species diversity. However, the functional profiles of the overall gut microbiomes between the two honey bee species converge, probably as a result of the overlapping ecological niches of the two species. Our findings provide critical insights into the evolution and functional roles of the mutualistic microbiota of honey bees and reveal that functional redundancy could stabilise the gene content diversity at the strain-level within the gut community.

Brain transcriptional regulatory network for behavior demonstrates that brain gene expression in the honey bee can be accurately predicted from the expression transcription factors (TFs), but roles for specific TFs are less understood. Mushroom bodies (MBs) are important for learning, memory and sensory integration in the honey bee brain. A transcription factor, Mblk-1, expressed preferentially in the large-type Kenyon cells of the honeybee MBs is predicted to be involved in brain function by regulating transcription of its target genes in honey bee. However its function and the mechanism of regulation in behavior of honey bee is still obscure. Here we show that Mblk-1 had significantly higher expression in the brains of forager bees relative to nurse bees. Mblk-1 was significantly inhibited in bees fed siRNA. In addition, inhibition of Mblk-1 decreased sucrose responsiveness in foragers. Finally, we determined that Mblk-1 regulated the mRNA of AmGR1. These findings suggest that Mblk-1 may target AmGR1 to regulate the sucrose responsiveness of foragers. This article is protected by copyright. All rights reserved.

As ancient bacterial endosymbionts of eukaryotic cells, mitochondria have retained their own circular DNA as well as protein translation system including mitochondrial ribosomes (mitoribosomes). In recent years, methodological advancements in cryoelectron microscopy and mass spectrometry have revealed the extent of the evolutionary divergence of mitoribosomes from their bacterial ancestors and their adaptation to the synthesis of 13 mitochondrial DNA encoded oxidative phosphorylation complex subunits. In addition to the structural data, the first assembly pathway maps of mitoribosomes have started to emerge and concomitantly also the assembly factors involved in this process to achieve fully translational competent particles. These transiently associated factors assist in the intricate assembly process of mitoribosomes by enhancing protein incorporation, ribosomal RNA folding and modification, and by blocking premature or non-native protein binding, for example. This review focuses on summarizing the current understanding of the known mammalian mitoribosome assembly factors and discussing their possible roles in the assembly of small or large mitoribosomal subunits.

Microplastics (MPs) are an emerging threat to ecological conservation and biodiversity; however, little is known of the types and possible impacts of MPs in pollinators. To examine whether MPs were present in honeybees, we analyzed the honeybee samples collected in fields from six provinces in China. Four types MPs were identified in honeybee including polystyrene (PS) by Raman spectroscopic analysis, and these plastic polymers were detected in 66.7% bee samples. Then, we assessed the physical and biological impacts of PS of three sizes (0.5, 5, and 50 μm) on bees for 21 days. Next, we measured how the presence of PS affected the Israeli acute paralysis virus proliferation, a small RNA virus associated with bee colony decline. Experimental evidence showed that a large mass of PS was ingested and accumulated within the midgut and enhanced the susceptibility of bees to viral infection. Not only histological analysis showed that PS, especially 0.5 μm PS, damaged the midgut tissue and was subsequently transferred to the hemolymph, trachea, and Malpighian tubules, but also qPCR and transcriptomic results indicated that genes correlated with membrane lipid metabolism, immune response, detoxification, and the respiratory system were significantly regulated after PS ingestion. Our results highlight neglected MP contamination to the bees, a pollination ecosystem stressed by the anthropogenic pollution, and have implications for human health via ingestion of bee products.

Curcumin (Cur) is an active ingredient of Curcuma longa. Cur has many pharmacological effects, such as anti-inflammation, anti-oxidation, anticoagulation, hypolipidemic, anti-angiogenesis and anti-cancer. An injectable curcumin loaded chitosan/hydroxyapatite bone cement (Cur-CS/HA) was prepared as a bone scaffold and drug delivery. Tween 20, a nonionic surfactant, was incorporated into the cement to improve the solubility of curcumin. Extensive application of antibiotics leads to their ubiquitous occurrence in coastal aquatic environments. However, it remains largely unknown whether antibiotics can be bioaccumulated and biotransformed in major mariculture organisms such as sea cucumbers and toxicokinetic models for Echinodermata are lacking. In this study, laboratory exposure experiments on juvenile sea cucumber (Apostichopus japonicus) were performed for seven antibiotics (sulfadiazine, sulfamethoxazole, trimethoprim, enrofloxacin, ofloxacin, clarithromycin, and azithromycin). Field sea cucumber and surrounding seawater samples were also analyzed. Results show that the sea cucumbers tend to accumulate high concentrations of the antibiotics with kinetic bioconcentration factors (BCFs) up to 1719.7 L·kg-1 for ofloxacin. The BCFs determined in the laboratory agree well with those estimated from the field measurements. Seven biotransformation products (BTPs) of the antibiotics were identified, four of which were not reported previously in aquatic organisms. The BTPs were mainly found in the digestive tract, indicating its high capacity in the biotransformation. A multicompartmental toxicokinetic model based on the principles of passive diffusion was developed, which can successfully predict time-course concentrations of the antibiotics in different compartments of the juvenile sea cucumbers. The findings may offer a scientific basis for assessing health risks and guiding healthy mariculture of sea cucumbers.

Honey bees play a vital role in providing pollination services for agricultural crops and wild flowering plants. However, the spillover risk of their pathogens to other pollinators or wild insects is becoming a cause for concern. There is some evidence that stingless bees can carry honey bee viruses, but little is known about the presence of honey bee viruses in stingless bees in China. Here, we investigate the occurrence of major honey bee pathogens including bacteria, fungi, and viruses in stingless bees (Apidae: sp.). Our results show that the stingless bees (Apidae: sp.) were mainly infected with DWV-A, but no DWV-B and DWV-C. Phylogenetic analysis on fragments of lp, RdRp, and VP3 of DWV-A indicated that genetic variation in VP3 might an important indicator for host-specific viruses, but it requires further study. Our results indicated that DWV-A is not only the major strain of virus currently circulating in managed bee colonies in China and globally, but in stingless bee species as a whole.

Although effects of dietary condensed tannins (CT) on growth and health of fish have been evaluated, limited information is available on shrimp. This study investigated the effects of CT on growth, nutrient digestibility, antioxidant response and intestinal histomorphology of Litopenaeus vannamei. A 56-day feeding trial was conducted by randomly distributing 600 shrimp into 15 tanks with 40 shrimp per tank and fed five diets containing 0 (G0), 0.5 (G0.5), 1 (G1), 2 (G2) and 4 (G4) g/kg of grape seed CT respectively. Shrimp were fed to apparent satiation during the entire feeding trial and feces were collected daily for the last 14 days to determine nutrient digestibilities. Hepatopancreas and intestine samples were collected to analyze antioxidant-related genes expression and histomorphology. Results indicated that dietary CT linearly increased final body weight, weight gain rate and specific growth rate (0.05 0.05). This study was performed to assess the effects of dietary aflatoxin B1 (AFB1) on the growth, antioxidant and immune response, digestive enzyme activities, and intestinal morphology of Lateolabrax maculatus during a 56-day feeding trial. Four diets were formulated including 0, 0.1, 0.5, and 1.0 mg/kg of AFB1. Each diet was randomly assigned to 3 fish tanks with 40 fish per tank. Results indicated that the fish’s final body weight, weight gain rate, specific growth rate, feed intake, condition factor, viscerosomatic index, hepatosomatic index, and intestinesomatic index decreased (p < 0.01) as dietary AFB1 increased. AFB1 levels in diets increased (p < 0.05) serum total antioxidant capacity (TAOC), superoxide (SOD), catalase, malondialdehyde (MDA), alkaline phosphatase (AKP), and lysozyme (LZM), and increased (p < 0.05) the TAOC, SOD, MDA, AKP, LZM, and immunoglobulin M in the livers of the fish. Dietary AFB1 decreased (p < 0.05) intestinal trypsin activity and induced intestinal injury. In summary, dietary AFB1 up to 1.0 mg/kg was toxic to L. maculatus as judged by reduced growth, enhanced antioxidant and immune response, decreased intestinal trypsin activity, and impaired intestinal morphology.

Benzotriazole ultraviolet stabilizers (BUVSs) have received increasing attention due to their widespread usage, ubiquitous detection and their adverse ecological effect. However, information about the bioaccumulation potential of BUVSs and their joint exposure with heavy metals remains scarce. In this study, we investigated the bioaccumulation kinetics of 6 frequently reported BUVSs in common carp under different Cu concentration for 48 d, and their tissue-specific distribution patterns (liver, kidney, gill, and muscle tissues) were also evaluated. The bioconcentration factors (BCFs) and half-lives (t1/2) in the tissues ranged from 5.73 (UV-PS) to 1076 (UV-327), and 2.19 (UV-PS) to 31.5 (UV-320) days, respectively. The tissue-specific concentration and BCF values followed the order of liver > kidney > gill > muscle with or without Cu exposure. An increase in BCF with rising Cu concentration was observed, which is caused by the decreased depuration rate (k2) in more than half of treatment groups. These results indicated that BUVSs accumulated in fish and provides important insight into the risk assessment of this group of chemicals.

Chemodynamic therapy (CDT) is a promising therapeutic modality with transition metal ions and endogenous H2O2 as reagents, but its efficiency is impaired by low endogenous H2O2 levels and nonregeneration of metal ions. Most intracellular H2O2 supplement strategies use oxidases and are intensively dependent on oxygen participation. The hypoxia microenvironments of solid tumors weaken their performance. Here, we develop a near-infrared II light powered nanoamplifier to improve the local oxygen level and to enhance CDT. The nanoamplifier CPNP-Fc/Pt consists of ferrocene (Fc)- and cisplatin prodrug (Pt(IV))-modified conjugated polymer nanoparticles (CPNPs). CPNP has a donor-acceptor structure and demonstrates a good photothermal effect under 1064 nm light irradiation, which accelerates blood flow and efficiently elevates the local oxygen content. In response to intracellular glutathione, Pt(II) is released from CPNP-Fc/Pt and triggers enzymatic cascade reactions with nicotinamide adenine dinucleotide phosphate oxidase (NADPH oxidase) and superoxide dismutase to convert oxygen into H2O2. The enhanced oxygen level results in efficient intracellular H2O2 supply. Fc is reacted with H2O2 and converted to Fc+ via the Fenton reaction, with the generation of hydroxyl radicals for CDT. Unlike free metal ions, the Fe(III) in Fc+ is reduced to Fe(II) by intracellular NAD(P)H, which achieves the regeneration of Fc. The sufficient intracellular H2O2 supply and efficient Fc regeneration effectively enhance the Fenton reaction and demonstrate good in vivo CDT results with tumor growth suppression. This design offers a promising strategy to enhance CDT efficiency in the hypoxia microenvironment of solid tumors.

The novel coronavirus named SARS-CoV-2 has emerged at the end of 2019, which causes coronavirus disease (COVID-2019). Recent case reports of SARS-CoV-2 infected individuals have revealed the onset of Parkinson's disease (PD) symptoms in patients who do not have a family history of the PD.. However, till recently, no genetic impact or mechanisms that may induce Parkinsonism inCOVID-19 patients or after COVID-19 have been found.. This study aimed to detect the commonly dysregulated genes, transcriptional regulators, and pathways between PD and SARS-CoV-2 infections. We integrated genome-wide transcriptomic datasets from PBMC samples from COVID-19 and PD and associated pathways. Our study revealed 81 upregulated and 48 downregulated differentially expressed genes (DEGs) shared between PD and SARS-CoV-2 infections. These dysregulated genes were involved in key pathways "mitochondrion structure organization", "cell activation in immune response", and "signalling by interleukins". Our analysis showed RELA, TP53 and SP1 TFs that may regulate the upregulated DEGs. We have discovered key dysregulated genes and characterized the biological processes of commonly dysregulated in SARS-CoV-2 infections and PD, which could be used for the design of personalized treatment of PD following COVID-19.

BACKGROUND Eupatilin, an active flavone separated from Artemisia species, has various biological activity such as anti-inflammatory activity. The aim of the present study was to find out the influence of eupatilin on lipopolysaccharide (LPS)-induced acute lung injury (ALI) in rats. MATERIAL AND METHODS The administration of LPS was used to induce ALI; eupatilin was given 1 hour before the LPS administration. Lung structural damage of rats was analyzed by hematoxylin and eosin staining and the wet/dry lung ratio. The related inflammatory factors and lung injury markers were examined by enzyme-linked immunosorbent assay. The oxidative stress factors were analyzed by corresponding kits. The expression of peroxisome proliferator-activated receptor-alpha (PPAR-alpha) was assayed by western blot and immunohistochemical staining. RESULTS The results showed that eupatilin alleviated LPS-induced structural damage and decreased the wet/dry lung ratio concentration-dependently. Eupatilin decreased the level of surfactant protein (SP)-A, SP-D, and inflammatory factors such as interleukin (IL)-6, tumor necrosis factor (TNF)-alpha, and monocyte chemo-attractant protein (MCP)-1. LPS trigged nitric oxide (NO) generation, improved the production of malondialdehyde (MDA) and lactate dehydrogenase (LDH) and decreased the activity of superoxide dismutase (SOD), which were reversed when rats treated with eupatilin in a concentration-dependent way. Besides, the expression of PPAR-a was increased under the treatment of eupatilin. CONCLUSIONS Collectively, eupatilin alleviated LPS-induced ALI through inhibiting inflammation and oxidative stress in a concentration-dependent way, which was likely to be closely related with the activation of PPAR-alpha.

Seipin is an oligomeric integral endoplasmic reticulum (ER) protein involved in lipid droplet (LD) biogenesis. To study the role of seipin in LD formation, we relocalized it to the nuclear envelope and found that LDs formed at these new seipin-defined sites. The sites were characterized by uniform seipin-mediated ER-LD necks. At low seipin content, LDs only grew at seipin sites, and tiny, growth-incompetent LDs appeared in a Rab18-dependent manner. When seipin was removed from ER-LD contacts within 1 h, no lipid metabolic defects were observed, but LDs became heterogeneous in size. Studies in seipin-ablated cells and model membranes revealed that this heterogeneity arises via a biophysical ripening process, with triglycerides partitioning from smaller to larger LDs through droplet-bilayer contacts. These results suggest that seipin supports the formation of structurally uniform ER-LD contacts and facilitates the delivery of triglycerides from ER to LDs. This counteracts ripening-induced shrinkage of small LDs.

Condensed tannins (CT) have been observed to be beneficial to maintain glucose homeostasis and improve carbohydrate utilization for livestock, whereas little information is available about the effects of CT on glycometabolism of fish. Three diets were formulated to contain approximately 15 % carbohydrate (G1), 39 % carbohydrate (G2), and G2 supplemented with 0.1 % CT. A total of 225 juvenile Lateolabrax japonicus were randomly assigned to 3 groups (triplicate tanks per group) with 25 fish in each tank and fed to apparent satiation during 56-d feeding trial. Results indicated that fish fed G2 had lower (p < 0.05) final body weight, weight gain rate, feed intake, specific growth rate and serum insulin concentration, but higher (p < 0.05) feed efficiency ratio and serum glucose, total antioxidant capacity, superoxide dismutase, immunoglobulin M and lipopolysaccharide than those fish fed G1 or G3. The activity of pyruvate kinase (PK) was lower (p < 0.05) but glucokinase (GK) was higher (p < 0.05) in G2 than in G1 or G3. Gene expression of insulin like growth factor Ⅰ and PK were down-regulated (p < 0.05), whereas GK was up-regulated (p < 0.05) in G2 than in G1 or G3. No difference (p> 0.05) in these gene expressions were observed between G1 and G3. It was concluded that L. japonicus fed diet with 39 % carbohydrate exhibited depressed growth performance, increased antioxidant and immune capacity, and hyperglycemia. Supplementation of 0.1 % CT resulted in improved growth performance and decreased serum glucose concentration, mainly due to the positive effects of CT on the activity and gene expression of hepatic glycolysis and gluconeogenesis enzymes of L. japonicus.

Chinese sacbrood virus (CSBV) is a serious threat to eastern honeybees (Apis cerana), especially larvae. However, the pathological mechanism of this deadly disease remains unclear. Here, we employed mRNA and small RNA (sRNA) transcriptome approach to investigate the microRNAs (miRNAs) and small interfering RNAs (siRNAs) expression changes of A. cerana larvae infected with CSBV under natural condition. We found that serine proteases involved in immune response were down-regulated, while the expression of siRNAs targeted to serine proteases were up-regulated. In addition, CSBV infection also affected the expression of larvae cuticle proteins such as larval cuticle proteins A1A and A3A, resulting in increased susceptibility to CSBV infection. Together, our results provide insights into sRNAs that they are likely to be involved in regulating honeybee immune response.

Varroa destructor is by far the most serious threat to the western honey bee, Apis mellifera. A screen bottom board, a cultural method for mite control, is a modified bottom board with a screen that allows mites to fall onto a sticky board, or the grass or soil below the screen. Whether or not a screen bottom board can reduce varroa significantly has been controversial. Most studies show a trend of lower varroa populations in colonies with these boards, but the results are usually not statistically significant. To understand whether the negative results have been due to small sample sizes, or because the board is actually ineffective, we conducted a meta-analysis with seven published studies with a total of 145 colonies. Meta-analysis showed that the confidence intervals of the combined effect sizes were negative with a Hedges&rsquo, g of &minus, 1.09 (SE 0.39, 95% CI &minus, 2.0 to &minus, 0.19, p &lt, 0.01), which suggests that the varroa population in colonies with screen bottom boards is significantly lower compared to those with traditional wooden floors. We thus conclude that the screen bottom board does have a significantly negative impact on the varroa population and can be part of tool kits for mite control.

Background Nephrolithiasis is a systemic metabolic disease with a high prevalence worldwide and is closely related to lipid-mediated oxidative stress and inflammation. Orthosiphon stamineus Benth. (OS) is a traditional medicinal herb mainly containing flavonoids, caffeic acid derivatives, and terpenoids, which has the effect of treating urinary stones. However, the active ingredients of OS for the treatment of kidney stones and their regulatory mechanisms remain unknown. As a powerful antioxidant, flavonoids from herbs can mitigate calcium oxalate stone formation by scavenging radical. Thus, this work focused on EtOAc extract of OS (EEOS, mainly flavonoids) and aimed to reveal the potential intrinsic mechanism of EEOS in the treatment of kidney stones disease. Methods Firstly, 75% ethanol extract of OS was further extracted with EtOAc to obtain EtOAc extract containing 88.82% flavonoids. Secondly, the extract was subjected to component analysis and used in animal experiments. Then, an untargeted lipidomics based on ultrahigh performance liquid chromatography coupled with TripleTOF 5600 mass spectrometer (UPLC-QTOF-MS) was performed to test the lipid changes of kidneys in the control group, model group and EEOS treatment groups. Finally, multivariate statistical analysis was used to identify differences between the lipid profiles of mice in the model group and the EEOS group. Results Fifty-one lipid metabolites were significantly different between the mice in the model group and the EEOS intervention group, including glycerophosphocholines, glycerophosphoethanolamines, glycerophosphoinositols, and glycerophosphoglycerols. And the composition of glycerophospholipids-esterified ω-3 polyunsaturated fatty acids and glycerophospholipid subclasses in the kidneys of the EEOS group significantly changed compared to model group. Conclusions The EEOS can inhibit the stones formation by improving oxidative stress and inflammation mediated by glycerophospholipid metabolism. This study reveals the potential mechanism of EEOS for kidney stones treatment at the lipid molecule level, providing a new direction for further study of the efficacy of OS.

Chinese sacbrood virus (CSBV) is a serious threat to eastern honeybees (Apis cerana), especially larvae. However, the pathological mechanism of this deadly disease is remains unclear. Here, we employed an mRNA-seq and sRNA-seq approach in healthy and CSBV-infected 3rd Apis cerana larval. Gene ontology (GO) and KEGG analysis of 203 differentially expressed genes showed that CSBV infection affected host development by up-regulating the expression of larval cuticle proteins, such as larval cuticle proteins A1A and A3A, resulting in elevated susceptibility to CSBV. In addition, viral infection not only affected the expression of serine protease related to the melanization pathway and but also altered fatty acid metabolism and biosynthesis, thus progressed to disturb host immune response. Interestingly, GO annotation and KEGG analysis on target genes of CSBV-specific siRNA (vsiRNAs) showed that serine/threonine kinase activity and serine-type endopeptidas as well as fatty acid biosynthesis were significantly enriched (P < 0.05). Among these vsiRNAs, vsiRNA-1441 with relatively high expression targeted extracellular serine/threonine protein kinase. This study provides new evidence that CSBV attacks a distinct immune response pathway and mediates the expression of cuticle protein to gain the more chance of proliferation.IMPORTANCE Chinese sacbrood virus (CSBV) is the major factor threatening the health of Apis cerana. Although honeybees have antiviral defense mechanisms including RNA-interference (RNAi), virus had evolved resistance strategy like virus-derived small interfering RNAs (vsiRNA) produced during the viral infection, which can alter host immune by hijacking the host RNAi pathway. To gain insights at the transcription level into the mechanisms vsiRNA altering immune response and how this variation is connected with CSBV infection, we performed the first combined mRNA and small RNA-seq analyses of healthy and CSBV-infected Apis cerana larvae. Together, the omics approach enabled us to obtain comprehensive results that CSBV infection changed the expressions of cuticle proteins and related serine protease, and leading to the larvae unable to pupate. We report the infestation of small hive beetle, Aethina tumida, in a honeybee, Apis cerana, in South China. This is the first record for domestic Chinese honey bee infested with small hive beetle.

Despite organophosphate esters (OPEs) are ubiquitous in the environment, limited information is available about their tissue-specific accumulation potential in marine fish and exposure risks. Ten fish species from the coastal area of Laizhou Bay, North China, were sampled and analyzed to investigate tissue levels, bioaccumulation, and human exposure risks of 20 OPEs. Seventeen OPEs were detected in fish tissues with total concentration ranging from 6.6-107 ng/g dry weight. The average log bioaccumulation factor (BAF) values of OPEs ranged from 2.8 to 4.4 in livers, 2.3-3.8 in muscles, 2.5-3.9 in gills, and 2.8-4.4 in kidneys. The log BAF values of OPEs significantly increased with increasing their log KOW values (r = 0.55-0.63, p < 0.001). The estimated daily intake of OPEs ranged from 1.7-12.0 and 3.1-22.1 ng/kg bw/d for rural and urban residents, respectively. The hazard quotients of OPEs were in the range of 4 × 10 -5 to 6.7 × 10 -4 and 7 × 10 -5 to 1.2 × 10 -3 for rural and urban residents, respectively. Results showed that the human health risks of OPEs associated with fish consumption is at low level.

Honey bee viruses are associated with honey bee colony decline. Israeli acute paralysis virus (IAPV) is considered to have a strong impact on honey bee survival. Phylogenetic analysis of the viral genomes from several regions of the world showed that various IAPV lineages had substantial differences in virulence. Chronic bee paralysis virus (CBPV), another important honey bee virus, can induce two significantly different symptoms. However, the infection characteristics and pathogenesis of IAPV and CBPV have not been completely elucidated. Here, we constructed infectious clones of IAPV and CBPV using a universal vector to provide a basis for studying their replication and pathogenesis. Infectious IAPV and CBPV were rescued from molecular clones of IAPV and CBPV genomes, respectively, that induced typical paralysis symptoms. The replication levels and expression proteins of IAPV and CBPV in progeny virus production were confirmed by qPCR and Western blot. Our results will allow further dissection of the role of each gene in the context of viral infection while helping to study viral pathogenesis and develop antiviral drugs using reverse genetics systems.

Since the discovery that honey bee viruses play a role in colony decline, researchers have made major breakthroughs in understanding viral pathology and infection processes in honey bees. Work on virus transmission patterns and virus vectors, such as the mite Varroa destructor, has prompted intense efforts to manage honey bee health. However, little is known about the occurrence of honey bee viruses in bee predators, such as vespids. In this study, we characterized the occurrence of 11 honey bee viruses in five vespid species and one wasp from four provinces in China and two vespid species from four locations in France. The results showed that all the species from China carried certain honey bee viruses, notably Apis mellifera filamentous virus (AmFV), Deformed wing virus (DWV), and Israeli acute paralysis virus (IAPV), furthermore, in some vespid colonies, more than three different viruses were identified. In France, DWV was the most common virus, Sacbrood virus (SBV) and Black queen cell virus (BQCV) were observed in one and two samples, respectively. Phylogenetic analyses of IAPV and BQCV sequences indicated that most of the IAPV sequences belonged to a single group, while the BQCV sequences belonged to several groups. Additionally, our study is the first to detect Lake Sinai virus (LSV) in a hornet from China. Our findings can guide further research into the origin and transmission of honey bee viruses in Vespidae, a taxon of ecological, and potentially epidemiological, relevance.

Condensed tannins (CT) were demonstrated to mitigate oxidative stress and maintain intestinal health of fish, but little information is available to clarify the underlying regulation mechanisms. This study was conducted to evaluate the effects of CT supplementation on growth, antioxidant capacity and intestinal barrier function of Lateolabrax maculatus via in vivo oxidized fish oil-induced and in vitro hydrogen peroxide-induced stress models. Information about bioaccumulation and tissue distribution of antibiotics in wild marine fish is still limited. In the present study, tissue levels, bioaccumulation and distribution patterns of 9 sulfonamide (SA), trimethoprim (TMP), 5 fluoroquinolone (FQ), and 4 macrolide (ML) antibiotics were investigated in gill, muscle, kidney, and liver tissues of seven wild fish species collected from Laizhou Bay, North China in 2016. All the 19 antibiotics were detected in these fish tissues with the total concentrations ranging from 22ng/g dry weight (dw) to 500ng/g dw. An accelerated solvent extraction (ASE) with in-cell clean-up method coupled to ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) was developed to determine 21 antibiotics in sea cucumber. The analytes include 10 sulfonamides, 4 fluoroquinolones, 3 amphenicols, 2 beta-lactams, 1 lincosamide and trimethoprim. Optimal parameters of ASE method were obtained at 80 °C, 1 static cycle of 5 min with methanol/acetonitrile (1/1, v/v) using 2 g of C18 as adsorbent. Recoveries at 50.1–129.2% were achieved with RSD under 20%. Method detection limits ranged from 0.03 to 2.9 μg kg−1. Compared to the reported ultrasound-assisted extraction method, the proposed method offered comparable extraction efficiency for sulfonamides from sea cucumber, but higher for other categories of antibiotics. This validated method was then successfully applied to sea cucumber samples and 9 antibiotics were detected with the highest concentration up to 57.7 μg kg−1 for norfloxacin.

Fluoroquinolones (FQs) have attracted wide concerns due to their pseudo-persistent and universal presence in natural water. Here we exposed zebrafish separately to two FQs (enrofloxacin (ENR) and ofloxacin (OFL)) in different copper (Cu) concentrations for 20 days (d) in a flow-through system, followed by a 11 d depuration period in clean water to investigate compound specific bioaccumulation and tissue distribution. Two FQs could accumulate in zebrafish, and the high concentration was observed in liver. Moreover, the levels of FQs in different treatment groups were higher than the corresponding control fish group. The uptake rates (k1), elimination rates (k2), BCF value, and half-lives (t1/2) of FQs ranged from 0.02 to 3.28 d−1, 0.01 to 0.97 d−1, 0.33 to 109.33, 9.90 to 69.31 d, respectively. With Cu exposure concentration's increasing, k1 values in three tissues (liver, skin and gill) obviously decreased. The exposure concentration affected the BCF value significantly, but didn’t change their relative compositions in liver, gill, skin, and muscle after long time exposure. BCF values of ENR were always a little bit higher than those of OFL in almost all the tissues (liver, skin and gill) in the low Cu concentration treatments, whereas, in the high Cu concentration treatments the bioconcentration factors (BCF) values of ENR were lower than the values of OFL. The exposure of Cu played an important role in the FQs bioconcentration and BCF. These results are meaningful for improved understanding and prediction of the behavior and fate of metallic and antibiotics in aqueous environments.

Root foraging and root physiology such as exudation of carboxylates into the rhizosphere are important strategies for plant phosphorus (P) acquisition. We used 100 chickpea (Cicer arietinum) genotypes with diverse genetic backgrounds to study the relative roles of root morphology and physiology in P acquisition. Plants were grown in pots in a low-P sterilized river sand supplied with 10 mu g P g(-1) soil as FePO4, a poorly soluble form of P. There was a large genotypic variation in root morphology (total root length, root surface area, mean root diameter, specific root length and root hair length), and root physiology (rhizosheath pH, carboxylates and acid phosphatase activity). Shoot P content was correlated with total root length, root surface area and total carboxylates per plant, particularly malonate. A positive correlation was found between mature leaf manganese (Mn) concentration and carboxylate amount in rhizosheath relative to root DW. This is the first study to demonstrate that the mature leaf Mn concentration can be used as an easily measurable proxy for the assessment of belowground carboxylate-releasing processes in a range of chickpea genotypes grown under low-P, and therefore offers an important breeding trait, with potential application in other crops.

Limbal niche cells located in the limbal Palisades of Vogt are mesenchymal stem cells that reside next to limbal basal epithelial cells. Limbal niche cells are progenitors that express embryonic stem cell markers such as Nanog, Nestin, Oct4, Rex1, Sox2 and SSEA4, mesenchymal cell markers such as CD73, CD90 and CD105, and angiogenesis markers such as Flk‐1, CD31, CD34, VWF, PDGFRβ and α‐SMA, but negative for CD45. In addition, the stemness of limbal niche cells can be maintained during their cell culture in a three‐dimension environment. Furthermore, expanded limbal niche cells have the capability to undergo adipogenesis, chondrogenesis, osteogenesis and endogenesis in vitro, indicating that they are in fact a valuable resource of adult progenitors. Furthermore studies on how the limbal niche cells regulate the aforementioned stemness and corneal fate decision are warranted, as those investigations will shed new light on how mesenchymal progenitors reverse limbal stem cell deficiency and lead to new methods for limbal niche cell treatment.

Aethina tumida is a parasite and predator of honeybee causing severe loss to the bee industry. No effective and environmentally friendly methods are available to control this pest at present. Chemosensory genes play key roles in insect behavior which can potentially be used as targets for developing environmentally friendly pest control agents. In this study, the putative chemosensory genes in antennae and forelegs of A. tumida involved in olfaction or contact chemical communication of adults were investigated using RNA transcriptome sequencing and PCR methods. Based on transcriptomic data, unigenes encoding 38 odorant receptors (ORs), 24 ionotropic receptors (IRs), 14 gustatory receptors (GRs), 3 sensory neuron membrane proteins (SNMPs), 29 odorant binding proteins (OBPs), and 22 chemosensory proteins (CSPs) were identified. The analyses of tissue expression profiles revealed that genes encoding 38 ORs, 13 antennal IRs, 11 GRs, 1 SNMP, 24 OBPs, and 12 CSPs were predominately expressed in antennae. No significant differences in expression levels of these genes were found between males and females. Genes encoding 5 non-NMDA iGluRs, 3 GRs, 2 SNMPs, 5 OBPs, and 12 CSPs were predominately expressed in forelegs. RT-PCR assays for SNMPs, OBPs and CSPs further revealed that 3 OBPs (AtumOBP3, 26 and 28) and 3 CSPs (AtumCSP7, 8 and 21) were highly expressed in antennae. Our results enrich the gene inventory of A. tumida and facilitate the discovery of potential novel targets for developing new pest control measures.

Understanding the effects of above- and below-ground interactions on seedling growth is pivotal for identifying the key drivers of secondary forest succession. However, it is still unclear whether the effects of above- and below-ground interactions of plants are consistent for seedling growth of deciduous and evergreen species. There are two types of broadleaved forests (i.e., tropical lowland rainforest and tropical deciduous monsoon rainforest) in the low-elevation (&lt, 800 m) areas of Hainan Island in China. Here, 32 seedling transplanting plots (1 × 1 m2) were established in the tropical lowland rainforest and the tropical deciduous monsoon rainforest, respectively. Four treatments (each with 16 replicates) were carried out to reduce above- and below-ground interactions of plants in the low-elevation forests: removal of vegetation (R), root trenching (T), removal of vegetation and root trenching (R + T), and no vegetation removal or trenching (as the control) (C). Seedlings of four deciduous species and four evergreen species were planted to observe their performance in the experiments. The relative growth rates (RGR) of the seedlings were measured to distinguish the relative effects of above- and below-ground interactions. The photosynthetically active radiation (PAR) was measured as a proxy for above-ground interaction and the root biomass was used as a proxy for below-ground interaction. The relationships between seedling RGR and PAR/root biomass were examined. Results showed that: (1) R and R+T treatments significantly increased the seedlings RGR, but T treatment had no effect on the RGR, (2) the growth rates of deciduous species were greater than those of the evergreen species, and (3) seedling growth rates were increased with more PAR. Our study suggests that above-ground vegetation removal had a stronger effect than trenching on the growth and assembly of tree seedlings in the low-elevation tropical rainforests.

Populus cathayana was employed as a model species to study sexual differences in plant growth, physiological, biochemical, and ultrastructural responses to manganese (Mn) stress with two different nitrogen sources (NH4 + and NO3 −). Results showed that NH4 +-N (3.75 mM) and NO3 −-N (3.75 mM) significantly increased plant growth, gas exchange capacity, as well as superoxide dismutase (SOD) and peroxidase (POD) activities in the leaves of both male and female P. cathayana. In addition, NH4 +-N supply could improve tolerance to Mn stress in both male and female P. cathayana with higher photosynthetic capabilities, biomass accumulation, antioxidant enzyme activities, superoxide radicals (O2 −) and thiobarbituric acid-reactive substances (TBARS) in leaves. Under the same condition, female plants grown under NO3 − supply exhibited significantly lower biomass accumulation, photosynthetic capabilities, and antioxidant enzyme activities than male plants because of higher Mn accumulation in both above- and belowground organs and higher ROS releasing rate in leaves. Moreover, damage to cellular ultrastructure was also observed in leaves of males and females exposed to Mn stress, but more strongly in females, with more visible degradation of mitochondrion in plant leave cells. The difference in the preferences for different N sources between male and female P. cathayana under Mn stress provides important guidance in fertilizing strategy to minimise intraspecific competition between female and male plants.

The occurrence and distribution of 25 antibiotics from 5 categories in Yancheng coastal area of the South Yellow Sea were investigated using solid-phase extraction coupled with high-performance liquid chromatography tandem mass spectrometry. Results showed that these antibiotics were widely present in this region with the total concentration up to 1349.2ng/L. Fluoroquinolones and sulfonamides were the most abundant categories contributing 46.5%, and 21.4% to the total antibiotics burden. Trimethoprim was the antibiotic detected in all the samples. The detection rates of erythromycin-H2O, sulfamethoxazole and florfenicol were 70.0%, 56.7% and 53.4%, respectively. The distribution of antibiotics demonstrated a seaward decreasing trend with the attenuation rate ranging from 0.07 to 0.19km-1 in this region. The Hippo pathway regulates cell proliferation, apoptosis, and stem cell self-renewal, and its inactivation in animal models causes organ enlargement followed by tumorigenesis. Hippo pathway deregulation occurs in many human cancers, but the underlying mechanisms are not fully understood. Here, we report tyrosine phosphorylation of the Hippo pathway tumor suppressor LATS1 as a mechanism underlying its regulation by cell adhesion. A tyrosine kinase library screen identified Src as the kinase to directly phosphorylate LATS1 on multiple residues, causing attenuated Mob kinase activator binding and structural alteration of the substrate-binding pocket in the kinase domain. Cell matrix adhesion activated the Hippo pathway effector transcription coactivator YAP partially through Src-mediated phosphorylation and inhibition of LATS1. Aberrant Src activation abolished the tumor suppressor activity of LATS1 and induced tumorigenesis in a YAP-dependent manner. Protein levels of Src in human breast cancer tissues correlated with accumulation of active YAP dephosphorylated on the LATS1 target site. These findings reveal tyrosine phosphorylation of LATS1 by Src as a novel mechanism of Hippo pathway regulation by cell adhesion and suggest Src activation as an underlying reason for YAP deregulation in tumorigenesis. Despite numerous studies that report the glucose derived glycoconjugates as antitumor candidates, using mannose as sugar motif for specific tumor targeting remains less studied. In this research, two novel mannose-conjugated platinum complexes 4a and 4b that target the Warburg effect were designed, synthesized and evaluated for their antitumor activities in vitro and in vivo. Compared with oxaliplatin, both complexes exhibited substantial enhancement in water solubility as well as excellent or comparative cytotoxicity in six human cancer cell lines. Cytotoxicity assessments on Glucose transporter 1 (GLUT1) down-regulated or overexpressed cells and platinum accumulation study demonstrated that cellular uptake of compound 4a was regulated by GLUT1. In particular, 4a induced apoptosis in HT29 cells by suppressing expression of Bcl-2 and Bcl-XL, which preliminary explained the mechanism origin of antitumor effect. As indicated by its maximum tolerated dose-finding assay and in vivo anticancer activity, compound 4a exhibits better safety and efficacy profile than oxaliplatin. The findings of this study indicate the possibility of subjecting mannose-conjugated platinum complexes as lead compounds for further preclinical evaluation.

Little information is available about the bioaccumulation and biomagnification of antibiotics in marine food webs. Here we investigate the levels and trophic transfer of 9 sulfonamide (SA), 5 fluoroquinolone (FQ), and 4 macrolide (ML) antibiotics, as well as trimethoprim in nine invertebrate and ten fish species collected from a marine food web in Laizhou Bay, North China in 2014 and 2015. All the antibiotics were detected in the marine organisms, with SAs and FQs being the most abundant antibiotics. Benthic fish accumulated more SAs than invertebrates and pelagic fish, while invertebrates exhibited higher FQ levels than fish. Generally, SAs and trimethoprim biomagnified in the food web, while the FQs and MLs were biodiluted. Trophic magnification factors (TMF) were 1.2 – 3.9 for SAs and trimethoprim, 0.3 – 1.0 for FQs and MLs. Limited biotransformation and relatively high assimilation efficiencies are the likely reasons for the biomagnification of SAs. The pH dependent distribution coefficients (logD) but not the lipophilicity (logKOW) of SAs and FQs had a significant correlation (r = 0.73; p < 0.05) with their TMFs. Although the calculated estimated daily intakes (EDI) for antibiotics suggest that consumption of seafood from Laizhou Bay is not associated with significant human health risks, this study provides important insights into the guidance of risk management of antibiotics.

Chromosome segregation in mitosis requires the removal of catenation between sister chromatids. Timely decatenation of sister DNAs at mitotic centromeres by topoisomerase IIα (TOP2A) is crucial to maintain genomic stability. The chromatin factors that recruit TOP2A to centromeres during mitosis remain unknown. Here, we show that histone H2A Thr‐120 phosphorylation (H2ApT120), a modification generated by the mitotic kinase Bub1, is necessary and sufficient for the centromeric localization of TOP2A. Phosphorylation at residue‐120 enhances histone H2A binding to TOP2A in vitro. The C‐gate and the extreme C‐terminal region are important for H2ApT120‐dependent localization of TOP2A at centromeres. Preventing H2ApT120‐mediated accumulation of TOP2A at mitotic centromeres interferes with sister chromatid disjunction, as evidenced by increased frequency of anaphase ultra‐fine bridges (UFBs) that contain catenated DNA. Tethering TOP2A to centromeres bypasses the requirement for H2ApT120 in suppressing anaphase UFBs. These results demonstrate that H2ApT120 acts as a landmark that recruits TOP2A to mitotic centromeres to decatenate sister DNAs. Our study reveals a fundamental role for histone phosphorylation in resolving centromere DNA entanglements and safeguarding genomic stability during mitosis.

Snail is a master inducer of epithelial-mesenchymal transition (EMT) and metastasis, however, Snail protein is labile and is quickly degraded through the predominate ubiquitination-mediated proteasome pathway. Deubiquitinases (DUBs) can counteract the Snail degradation process to maintain high level of Snail protein in cancer cells. In this study, we screened a cDNA library containing 79 DUBs, and discovered that a panel of DUBs consisting of USP13, USP28, USP29, USP37, OTUD6A, and DUB3 can markedly stabilize Snail protein, with USP29 displaying the strongest activity to prevent Snail degradation. Mechanistically, USP29 enhances the interaction of Snail and SCP1, resulting in simultaneous dephosphorylation and deubiquitination of Snail and thereafter cooperative prevention of Snail degradation. Biologically, ectopic expression of USP29 promotes gastric cancer cell migration, and depletion of Snail abolishes USP29-mediated cell migration; and USP29 can be induced by major EMT and metastatic inducing factors such as TGFβ, TNFα, and hypoxia. More importantly, high expression levels of Snail, USP29, and SCP1 are associated with poor survival and prognosis. Collectively, these data indicate that Snail is a crucial substrate for USP29 to promote cell migration and USP29/SCP1 complex may be new therapeutic targets to treat metastatic cancer.

The greater wax moth, Galleria mellonella Linnaeus (Lepidoptera: Galleriinae), is a ubiquitous pest of the honeybee, and poses a serious threat to the global honeybee industry. G. mellonella pheromone system is unusual compared to other lepidopterans and provides a unique olfactory model for pheromone perception. To better understand the olfactory mechanisms in G. mellonella, we conducted a transcriptomic analysis on the antennae of both male and female adults of G. mellonella using high-throughput sequencing and annotated gene families potentially involved in chemoreception. We annotated 46 unigenes coding for odorant receptors, 25 for ionotropic receptors, two for sensory neuron membrane proteins, 22 for odorant binding proteins and 20 for chemosensory proteins. Expressed primarily in antennae were all the 46 odorant receptor unigenes, nine of the 14 ionotropic receptor unigenes, and two of the 22 unigenes coding for odorant binding proteins, suggesting their putative roles in olfaction. The expression of some of the identified unigenes were sex-specific, suggesting that they may have important functions in the reproductive behavior of the insect. Identification of the candidate unigenes and initial analyses on their expression profiles should facilitate functional studies in the future on chemoreception mechanisms in this species and related lepidopteran moths.

The presence and concentrations of 25 antibiotics in Dalian coastal water of the Bohai Sea were investigated using solid-phase extraction coupled with high-performance liquid chromatography tandem mass spectrometry. Results showed that antibiotics were widely detected in this region with total concentration ranging from 22.6 to 2402.4 ng/L. Enrofloxacin and trimethoprim were 100% detected followed by sulfamethoxazole with a detection rate of 90.9%. No significant correlations were found between antibiotics concentrations and sample parameters such as dissolved organic carbon, salinity, and distance from the coast, suggesting that concentrations and distributions of the antibiotics in this area were source-dependent. Antibiotic concentration in the sample from an offshore cage-culture area was the highest. Based on composition profiles, mariculture was supposed to be an important source of antibiotics. According to the assessment, individual antibiotic posed low to moderate risk, while the antibiotic mixture presented high risk. Enrofloxacin, clarithromycin and sulfamethoxazole, the top three contributors to the mixture risk quotients for each site, need priority control in this area. Besides, levels of enrofloxacin were high enough to exert a selective pressure on bacteria that may lead to an increase in the prevalence of resistance.

A representative congener of polybrominated diphenyl ethers in the environment, 2,2′,4,4′-tetrabromodiphenyl ether (BDE-47), is associated with male reproductive toxicity, yet the underlying mechanisms remain largely unclear. In this study, mice were administered environmentally relevant concentrations of BDE-47 for six weeks. Histopathological observations showed that BDE-47 induced inflammatory reactions and damaged the testes. By conducting an integrated proteomic and metabolomic analysis coupled with a bioinformatic analysis using ingenuity pathway analysis (IPA) methods, we found that BDE-47 mainly affected the molecules involved in free radical scavenging, cell death and survival, neurological disease, and inflammatory response. IPA canonical pathways showed inflammatory and apoptosis pathways, including hepatic fibrosis/hepatic stellate cell activation, the GP6 signaling pathway, tight junction signaling, acute phase response signaling, LXR/RXR activation, unfolded protein response, and FXR/RXR activation, which are related to male reproductive toxicity. Key transcriptional regulator networks were activated via a focus on upstream regulator analysis. The expression of MYC and Clu as the core transcriptional factor and targeted protein, respectively, was verified. It is further proposed that MYC may contribute to the etiology of male reproductive toxicity. These findings will improve our understanding of the mechanisms responsible for BDE-47-induced male reproductive toxicity, which may promote the discovery of useful biomarkers indicative of BDE-47 exposure.

The effects of dietary sodium butyrate (SB) on serum metabolic parameters, intestinal antioxidant status, immune response and resistance to ammonia stress in Pelteobagrus fulvidraco were investigated. The fish were fed with diets containing 0, 250, 500, 1000 or 2000 mg/kg SB for 56 days followed by a 72-h ammonia challenge (2.65 mg/L unionized ammonia-N). The actin cytoskeleton powers membrane deformation during many cellular processes, such as migration, morphogenesis, and endocytosis. Membrane phosphoinositides, especially phosphatidylinositol 4,5-bisphosphate [PI(4,5)P-2], regulate the activities of many actinbinding proteins (ABPs), including profilin, cofilin, Dia2, N-WASP, ezrin, and moesin, but the underlying molecular mechanisms have remained elusive. Moreover, because of a lack of available methodology, the dynamics of membrane interactions have not been experimentally determined for any ABP. Here, we applied a combination of biochemical assays, photobleaching/activation approaches, and atomistic molecular dynamics simulations to uncover the molecular principles by which ABPs interact with phosphoinositide-rich membranes. We show that, despite using different domains for lipid binding, these proteins associate with membranes through similar multivalent electrostatic interactions, without specific binding pockets or penetration into the lipid bilayer. Strikingly, our experiments reveal that these proteins display enormous differences in the dynamics of membrane interactions and in the ranges of phosphoinositide densities that they sense. Profilin and cofilin display transient, low-affinity interactions with phosphoinositide-rich membranes, whereas F-actin assembly factors Dia2 and N-WASP reside on phosphoinositide-richmembranes for longer periods to performtheir functions. Ezrin and moesin, which link the actin cytoskeleton to the plasma membrane, bindmembranes with very high affinity and slow dissociation dynamics. Unlike profilin, cofilin, Dia2, and N-WASP, they do not require high "stimulus-responsive" phosphoinositide density for membrane binding. Moreover, ezrin can limit the lateral diffusion of PI(4,5)P-2 along the lipid bilayer. Together, these findings demonstrate that membrane-interaction mechanisms of ABPs evolved to precisely fulfill their specific functions in cytoskeletal dynamics.

This experiment was conducted to investigate the effects of dietary arginine levels on growth performance, body composition, serum biochemical indices and resistance ability against ammonia-nitrogen stress in juvenile yellow catfish (Pelteobagrus fulvidraco). Five isonitrogenous and isolipidic diets (42% protein and 9% lipid) were formulated to contain graded levels of arginine (2.44%, 2.64%, 2.81%, 3.01% and 3.23% of diet), by supplementing L-Arginine HCl. Seven hundred juvenile yellow catfish with an initial average body weight of 1.13 ± 0.02 g were randomly divided into 5 groups with 4 replicates of 35 fish each and each group was fed one of the diets. After 56 d feeding, fish were exposed to 100 mg/L of ammonia-nitrogen for 72 h. The results showed that weight gain (WG) and specific growth rate (SGR) in 2.64% and 2.81% groups were significantly higher than those in 3.23% group (P 0.05).
