Optical sensors have shown high capabilities to improve the detection and monitoring of plant disease development. This study was designed to compare the feasibility of different sensors to characterize Fusarium head blight (FHB) caused by Fusarium graminearum and Fusarium culmorum. Under controlled conditions, time-series measurements were performed with infrared thermography (IRT), chlorophyll fluorescence imaging (CFI), and hyperspectral imaging (HSI) starting 3 days after inoculation (dai). IRT allowed the visualization of temperature differences within the infected spikelets beginning 5 dai. At the same time, a disorder of the photosynthetic activity was confirmed by CFI via maximal fluorescence yields of spikelets (Fm) 5 dai. Pigment-specific simple ratio PSSRa and PSSRb derived from HSI allowed discrimination between Fusarium-infected and non-inoculated spikelets 3 dai. This effect on assimilation started earlier and was more pronounced with F. graminearum. Except the maximum temperature difference (MTD), all parameters derived from different sensors were significantly correlated with each other and with disease severity (DS). A support vector machine (SVM) classification of parameters derived from IRT, CFI, or HSI allowed the differentiation between non-inoculated and infected spikelets 3 dai with an accuracy of 78, 56 and 78%, respectively. Combining the IRT-HSI or CFI-HSI parameters improved the accuracy to 89% 30 dai.

Young maize plants were inoculated on unfolded mature leaves and on folded immature leaves with Fusarium graminearum, Fusarium proliferatum, and Fusarium verticillioides suspensions. Infection and symptom development of disease on these asymptomatic mature leaves and immature leaves were then documented. Subcuticular infection was found by the three Fusarium species on both symptomatic and symptomless leaves. The three Fusarium species penetrated the stomata of immature leaves by the formation of appressoria-like structures, infection cushions or by direct penetration. Infection by the three species of Fusarium via stomata is reported here for the first time. The superficial hyphae and re-emerging hyphae of the three species produced conidia. The macroconidia of F. graminearum produced secondary macroconidia and F. proliferatum formed microconidia inside the leaf tissues that sporulated through stomata and trichomes. The infection of maize leaves by the three species of Fusarium and their sporulation may contribute inoculum to cob and kernel infection.

Fifteen day old maize seedlings were inoculated with Fusarium graminearum, Fusarium proliferatum, and Fusarium verticillioides. More than 90 % F. proliferatum and F. verticillioides conidia and 50 % of F. graminearum formed one germ tube whereas the other 50 % of F. graminearum conidia formed two to three germ tubes. The germ tubes of F. graminearum conidia were longer than those of F. proliferatum and F. verticillioides. The three species of Fusarium infected bi-cellular trichomes by adhering and growing along the trichomes or by attaching to the cap cell of the trichomes 48 h after inoculation. Hyphae penetrated into the trichomes at the base, the side or at the top of the cap cells. The hyphae colonized the cap cells and then spread to base cells. Prickle trichomes were infected 72 h after inoculation. The hyphae either wrapped around prickle trichomes or formed a mass of hyphae around the top of prickle trichomes or formed appressorium. Macro trichomes were infected by F. graminearum 7 d after inoculation. Following penetration, the fungus spread to adjacent epidermal cells and to the subcuticle. This investigation provides the first assessment of F. graminearum, F. proliferatum, and F. verticillioides infection via trichomes of maize leaves.

The relationship between initial population densities (Pi) of Meloidogyne hapla on growth of three rose rootstocks (Rosa corymbifera ‘Laxa’, R. multiflora and R. canina ‘Inermis’) and nematode population development was studied. Each plant species was inoculated with ranges of nematode densities of 0, 0.062, 0.125, 0.25, 0.50, 1, 2, 4, 8, 16, 32, 64 and 128 second-stage juvenile/g soil and were allowed to grow for 9 weeks. Seinhorst yield model math formula was fitted to total fresh biomass data of the rootstocks. The tolerance limits (T) were 0.04, 0.09 and 0.01 J2/g soil and the minimum yield (m) 0.65, 0.47 and 0.43 for R. corymbifera ‘Laxa’, R. multiflora and R. canina ‘Inermis’, respectively. The reproductive factor (Pf/Pi) was highest at low initial nematode densities for all rootstocks and then decreased to below maintenance level with increasing initial population densities. Root gall severity consistently increased with initial nematode population density. Furthermore, number of root galling showed a strong positive relationship with final nematode population per gram root fresh weight. The relation between Pi and Pf was also fitted to the Seinhorst population model (Pf = (M*Pi)/Pi + M/a). Rosa multiflora supported the population of M. hapla to a maximum population density (M) of 27.53 J2/g soil with an estimated average maximum multiplication rate (a) of 24.39. For R. corymbifera ‘Laxa’ and R. canina, the maximum multiplication rate was 4.34 and 3.62 and the maximum population density 6.08 and 4.78 J2/g dry soil, respectively. Hence, it was demonstrated that all three rootstocks were susceptible to even low initial nematode densities and therefore are considered good hosts for M. hapla

Fusarium head blight (FHB) epidemics in wheat and contamination with Fusarium mycotoxins has become an increasing problem over the last decades. This prompted the need for non-invasive and non-destructive techniques to screen cereal grains for Fusarium infection, which is usually accompanied by mycotoxin contamination. This study tested the potential of hyperspectral imaging to monitor the infection of wheat kernels and flour with three Fusarium species. Kernels of two wheat varieties inoculated at anthesis with F. graminearum, F. culmorum, and F. poae were investigated. Hyperspectral images of kernels and flour were taken in the visible-near infrared (VIS-NIR) (400&ndash, 1000 nm) and short-wave infrared (SWIR) (1000&ndash, 2500 nm) ranges. The fungal DNA and mycotoxin contents were quantified. Spectral reflectance of Fusarium-damaged kernels (FDK) was significantly higher than non-inoculated ones. In contrast, spectral reflectance of flour from non-inoculated kernels was higher than that of FDK in the VIS and lower in the NIR and SWIR ranges. Spectral reflectance of kernels was positively correlated with fungal DNA and deoxynivalenol (DON) contents. In the case of the flour, this correlation exceeded r = &minus, 0.80 in the VIS range. Remarkable peaks of correlation appeared at 1193, 1231, 1446 to 1465, and 1742 to 2500 nm in the SWIR range.

Background Plant pathogenic fungi of the genus Fusarium infect a wide array of crops and produce numerous health-threatening mycotoxins. Recently, we found that larvae of the common pest of stored products Tenebrio molitor preferably fed on grains colonized with Fusarium proliferatum. We draw the hypothesis that the increased attractiveness of infected grains for mealworms facilitates dispersal of the fungus. In this work we examined the dissemination of F. proliferatum and further Fusarium spp. by adults of T. molitor. Results Mealworm beetle Tenebrio molitor transmitted Fusarium species F. avenaceum, F. culmorum, F. poae, and F. proliferatum to wheat grains with varying efficiency. F. proliferatum was disseminated most efficiently: 20 days after feeding on Fusarium cultures, the beetles still transmitted F. proliferatum to most grains exposed to feeding. The transmission of F. culmorum gradually declined over time and the transmission of the other Fusarium spp. ceased completely 20 d after beetles feeding of fungal cultures. Propagules of F. proliferatum and F. culmorum were traceable in beetles' feces for 20 days while no colonies of F. poae and F. avenaceum were detectable after 5 days. Because F. proliferatum was transmitted by mealworms most efficiently, this species was further investigated. Mealworm beetles T. molitor preferred feeding on grains colonized with F. proliferatum as compared to uninfected grains. Male beetles infected with F. proliferatum transmitted the fungus by copulation. Conclusions Efficient dissemination of F. proliferatum by mealworm beetle together with the feeding preference of the beetle for grains colonized with F. proliferatum show that the chemical phenotype of the fungus responsible for the enhanced attractiveness of infected grains is subjected to positive selection. This indicates that adaptation of F. proliferatum to transmission by insects involved an alteration of insects' feeding preferences.

Fusarium graminearum is a devastating fungal pathogen that causes significant yield and quality losses in cereals. We utilized a diversity set of barley (140 genotypes) to explore vital resistance alleles against this aggressive pathogen. The resistance assessment was carried out on spikes and leaves via artificial inoculations under control conditions. The phenotypic data was subjected to genome-wide association analysis using a genetic map based on DArT and SNP markers. This analysis revealed eleven and nine marker trait associations for leaf disease scoring (LDS) and spike disease scoring (SDS), respectively. The strongest QTL for LDS was found on chromosome 1H where a minor allele of wild origin decreased disease symptoms by 78%. The major QTL allele for SDS was linked with marker locus SCRI_RS174710 on chromosome 5H. In addition, four favorable epistatic interactions effects were found in decreasing disease symptoms. Overall, three QTL were common for LDS and SDS, which indicates a partial genetic relatedness of these resistances in barley. The QTL alleles for LDS and SDS will help to establish organ specific resistances in cultivated barley.

Spectral vegetation indices (SVIs) have been shown to be useful for an indirect detection of plant diseases. However, these indices have not been evaluated to detect or to differentiate between plant diseases on crop plants. The aim of this study was to develop specific spectral disease indices (SDIs) for the detection of diseases in crops. Sugar beet plants and the three leaf diseases Cercospora leaf spot, sugar beet rust and powdery mildew were used as model system. Hyperspectral signatures of healthy and diseased sugar beet leaves were assessed with a non-imaging spectroradiometer at different developing stages and disease severities of pathogens. Significant and most relevant wavelengths and two band normalized differences from 450 to 950 nm, describing the impact of a disease on sugar beet leaves were extracted from the data-set using the RELIEF-F algorithm. To develop hyperspectral indices for the detection of sugar beet diseases the best weighted combination of a single wavelength and a normalized wavelength difference was exhaustively searched testing all possible combinations. The optimized disease indices were tested for their ability to detect and to classify healthy and diseased sugar beet leaves. With a high accuracy and sensitivity healthy sugar beet leaves and leaves, infected with Cercospora leaf spot, sugar beet rust and powdery mildew were classified (balanced classification accuracy: 89%, 92%, 87%, 85%, respectively). Spectral disease indices were also successfully applied on hyperspectral imaging data and on non-imaging data from a sugar beet field. Specific disease indices will improve disease detection, identification and monitoring in precision agriculture applications.

Fusarium proliferatum is a fungal pathogen causing ear rot of maize. The fungus infects a range of other plants but the economic impact of these diseases has not been established. Recently, F. proliferatum and its mycotoxin fumonisin were found in wheat grains. Here we report that seed-borne infection of wheat with F. proliferatum resulted in systemic colonization of wheat plants and contamination of wheat grains with fumonisins and beauvericin. F. proliferatum strains originating from different hosts were able to infect wheat via seeds. Colonization of wheat plants with the fungus was highest in the stems, followed by leaves; one third of the strains reached kernels, causing accumulation of fumonisins and beauvericin to 15–55 µg kg-1. The results show that seed-borne infection of wheat with F. proliferatum can lead to contamination of wheat kernels with mycotoxins fumonisins and beauvericin.

Fusarium species involved in the Fusarium head blight complex in Western Europe were investigated for their potential to infect and colonize non-damaged wheat leaves and to produce conidia on senescing wheat leaves incubated at high relative humidity. Fusarium avenaceum, Fusarium culmorum, Fusarium graminearum, Fusarium poae and Fusarium tricinctum did not directly penetrate the leaf tissue after conidia germination on the leaf surface. Germ tubes grew on the host surface for 24–36 hr forming a mycelial network. After invading the host, some species formed runner hyphae between cell wall layers or underneath the cuticular layer. Macroscopic symptoms developed on leaves and stems from 7 d post inoculation. Inside leaf tissues, hyphae thickened in diameter and were both inter- and intra-cellular. Fusarium tricinctum formed sporophores which erupted through the leaf surface releasing numerous conidia. Incubation of senescing leaves at 100 % relative humidity for 48 hr resulted in sporulation of all Fusarium spp.

Interspecies interactions between Fusarium avenaceum, Fusarium culmorum, Fusarium graminearum, Fusarium poae, and Fusarium tricinctum were studied during early growth stages of isolates on model surfaces. Additionally, germination and germ tube growth of the pathogens were studied on attached and detached wheat leaves at 10 °C and 22 °C. Two-species interactions between Fusarium isolates during germination and germ tube growth were assessed after 8 hours of incubation. All species except F. tricinctum germinated and grew faster at higher than lower temperature. All species were able to germinate with more than one germ tube per conidium cell; and germination and germ tube growth were faster on leaves than on glass surface. Interactions among Fusarium species during germination and germ tube growth were predominantly competitive with macroconidia-producing species being more competitive. It is concluded that the type of conidia as well as environmental factors influence the competitiveness of Fusarium species during early stages of growth.

Near-range and remote sensing techniques have demonstrated a high potential in detecting diseases and in monitoring crop stands for sub-areas with infected plants. The occurrence of plant diseases depends on specific environmental and epidemiological factors; diseases, therefore, often have a patchy distribution in the field. This review outlines recent insights in the use of non-invasive optical sensors for the detection, identification and quantification of plant diseases on different scales. Most promising sensor types are thermography, chlorophyll fluorescence and hyperspectral sensors. For the detection and monitoring of plant disease, imaging systems are preferable to non-imaging systems. Differences and key benefits of these techniques are outlined. To utilise the full potential of these highly sophisticated, innovative technologies and high dimensional, complex data for precision crop protection, a multi-disciplinary approach—including plant pathology, engineering, and informatics—is required. Besides precision crop protection, plant phenotyping for resistance breeding or fungicide screening can be optimized by these innovative technologies.

In Germany, maize is one of the most important agriculture commodities, a major component in animal feed as well as an essential substrate producing biogas. Maize ear rot poses a major impact worldwide as it is caused by several Fusarium spp., most of which have the ability to produce mycotoxins. Despite of the expansion of the maize acreage in recent years, limited information is available concerning the incidence of Fusarium ear rot in Germany. Therefore, in a two-year survey maize crops were sampled in the major maizeproducing areas in Germany to establish the severity of Fusarium ear rot and the biodiversity of Fusarium species. In 2006, the frequency of kernels infected by Fusarium spp. ranged from 0.7 % to 99.7 %; the average incidence was 32.4 %. Thirteen Fusarium species were isolated from maize kernels, with F. verticillioides, F. graminearum and F. proliferatum being the predominant species. In 2007, the highest incidence of Fusarium ear rot was 64 %; the mean level of infection was 21.7 %. F. graminearum was by far the most frequent species isolated from all sampled fields in 2007. In addition, F. crookwellense, F. subglutinans and F. avenaceum were also frequently isolated. In particular, the fumonisin-producing species F. verticillioides an F. proliferatum were less frequent than in 2006. The year-to-year variability in the frequency of Fusarium species and in the overall infection rate may be explained by significant differences in temperature and precipitation during the growth periods.

Fusarium Head Blight (FHB) is one the most important diseases in small grain cereals and often is caused by a complex of Fusarium species. Some of these species are able to produce one or several mycotoxins. The spatial distribution of the disease and associated mycotoxins was examined in this study. Results were mapped and analysed with a geographic information system (GIS). Correlations between the incidence of the deoxynivalenol (DON) producing Fusarium species and DON contamination of kernels were rather weak. The level of DON contamination seemed to be less influenced by the frequency of DON producing Fusarium species than by other factors.

Adhesion to the host surface is the first step for successful plant pathogen development and has been reported to be associated with both passive and active processes. For conidia of Venturia inaequalis, which depend on leaf wetness for germination, this process has not yet been described. Conidia of V. inaequalis adhered to wet hydrophobic surfaces immediately after contact to the surface, hours before initiation of germination. Attachment of nongerminated conidia was much better on hydrophobic surfaces, such as apple leaves and polystyrene, than on hydrophilic glass. Conidia released adhesive material localized in a droplet named spore tip glue (STG) at the spore apex which interacted with a contact surface only when water was present. Histochemical investigations indicated the presence of proteins and carbohydrates in STG, lectin labeling the presence of β-galactose and N-acetylglucosaminyl residues. Transmission electron microscopy revealed two phases in the STG at the tip of dry mature conidia; as STG was present on the outer side of the intact fungal cell wall its formation should be associated with the secretion of glue through pores of the conidial wall. Surface-active substances affected the adhesion of conidia to hydrophobic surfaces stressing the importance of hydrophobic interactions. The use of protein biosynthesis inhibitors did not affect adhesion of conidia indicating that the adhesive material was preformed. It is concluded that the coincidence of STG, contact to a hydrophobic surface, and free water are essential for the adhesion of V. inaequalis conidia.

Background The detection and characterization of resistance reactions of crop plants against fungal pathogens are essential to select resistant genotypes. In breeding practice phenotyping of plant genotypes is realized by time consuming and expensive visual rating. In this context hyperspectral imaging (HSI) is a promising non-invasive sensor technique in order to accelerate and to automate classical phenotyping methods. A hyperspectral microscope was established to determine spectral changes on the leaf and cellular level of barley (Hordeum vulgare) during resistance reactions against powdery mildew (Blumeria graminis f.sp. hordei, isolate K1). Experiments were conducted with near isogenic barley lines of cv. Ingrid, including the susceptible wild type (WT), mildew locus a 12 (Mla12 based resistance) and the resistant mildew locus o 3 (mlo3 based resistance), respectively. The reflection of inoculated and non-inoculated leaves was recorded daily with a hyperspectral linescanner in the visual (400 – 700 nm) and near infrared (700 – 1000 nm) range 3 to 14 days after inoculation. Results Data analysis showed no significant differences in spectral signatures between non-inoculated genotypes. Barley leaves of the near-isogenic genotypes, inoculated with B. graminis f.sp. hordei differed in the spectral reflectance over time, respectively. The susceptible genotypes (WT, Mla12) showed an increase in reflectance in the visible range according to symptom development. However, the spectral signature of the resistant mlo-genotype did not show significant changes over the experimental period. In addition, a recent data driven approach for automated discovery of disease specific signatures, which is based on a new representation of the data using Simplex Volume Maximization (SiVM) was applied. The automated approach - evaluated in only a fraction of time revealed results similar to the time and labor intensive manually assessed hyperspectral signatures. The new representation determined by SiVM was also used to generate intuitive and easy to interpretable summaries, e.g. fingerprints or traces of hyperspectral dynamics of the different genotypes. Conclusion With this HSI based and data driven phenotyping approach an evaluation of host-pathogen interactions over time and a discrimination of barley genotypes differing in susceptibility to powdery mildew is possible.

Pathogenesis of Pseudoperonospora cubensis causing downy mildew of cucumber resulted in changes in the metabolic processes within cucumber leaves including the transpiration rate. Due to the negative correlation between transpiration rate and leaf temperature, digital infrared thermography permitted a non-invasive monitoring and an indirect visualization of downy mildew development. Depending on the stage of pathogenesis and the topology of chloroses and necroses, infection resulted in a typical temperature pattern. Spatial heterogeneity of the leaf temperature could be quantified by the maximum temperature difference (MTD) within a leaf. The MTD increased during pathogenesis with the formation of necrotic tissue and was related to disease severity as described by linear and quadratic regression curves. Under controlled conditions, changes in temperature of infected leaves allowed the discrimination between healthy and infected areas in thermograms, even before visible symptoms of downy mildew appeared. Environmental conditions during thermographic measurement, in particular air temperature and humidity, as well as water content and age of the leaf influenced the temperature of its surface. Conditions enhancing the transpiration rate facilitated the detection of changes in leaf temperature of infected leaves at early stages of infection. As modified by environmental conditions, MTD alone is not suitable for the quantification of downy mildew severity in the field.

Disease progress of downy mildew on cucumber leaves, caused by the obligate biotrophic pathogen Pseudoperonospora cubensis, was shown to be associated with various changes in transpiration depending on the stage of pathogenesis. Spatial and temporal changes in the transpiration rate of infected and noninfected cucumber leaves were visualized by digital infrared thermography in combination with measurements of gas exchange as well as microscopic observations of pathogen growth within plant tissue and stomatal aperture during pathogenesis. Transpiration of cucumber leaf tissue was correlated to leaf temperature in a negative linear manner (r = -0.762, P < 0.001, n = 18). Leaf areas colonized by Pseudoperonospora cubensis exhibited a presymptomatic decrease in leaf tem perature up to 0.8°C lower than noninfected tissue due to abnormal stomata opening. The appearance of chlorosis was associated with a cooling effect caused by the loss of integrity of cell membranes leading to a larger amount of apoplastic water in infected tissue. Increased water loss from damaged cells and the inability of infected plant tissue to regulate stomatal opening promoted cell death and desiccation of dying tissue. Ultimately, the lack of natural cooling from necrotic tissue was associated with an increase in leaf temperature. These changes in leaf temperature during downy mildew development resulted in a considerable heterogeneity in temperature distribution of infected leaves. The maximum temperature difference within a thermogram of cucumber leaves allowed the discrimination between healthy and infected leaves before visible symptoms appeared.

It is well accepted that agricultural production must be increased considerably in the foreseeable future to meet the food and feed demands of a rising human population and increasing livestock production. Crop protection plays a key role in safeguarding crop productivity against competition from weeds, animal pests, pathogens and viruses. The loss potential of these pest groups and the actual losses—i.e. losses despite the present crop protection practices—have been estimated for wheat, rice, maize, barley, potatoes, soybeans, sugar beet and cotton for the period 1996–1998 on a regional basis for 17 regions. Among crops the loss potential of pests worldwide varied from less than 50% (on barley) to more than 80% (on sugar beet and cotton). Actual losses are estimated at 26–30% for sugar beet, barley, soybean, wheat and cotton, and 35%, 39% and 40% for maize, potatoes and rice, respectively. Overall, weeds had the highest loss potential (32%) with animal pests and pathogens being less important (18% and 15%, respectively). Dans les conditions agricoles et climatiques specifiques du Rheinland (DE), 48 essais au champ sur trois ans ont demontre la possibilite d'appliquer la strategie fongicide basee sur un seuil d'intervention du IPM Wheat Model. Les donnees sous-jacentes pour les seuils d'intervention et le dosage de fongicides appliques sont decrits. Les agents pathogenes des cereales concernes, surtout Mycosphaerella graminicola et Puccinia recondita, ont ete controles dans les premiers stades du developpement de l'epidemie en utilisant des doses reduites de fongicides. Le IPM Wheat Model donne une lutte de grande efficacite, en terme d'incidence et de severite de la maladie, aussi bien en cas de faible ou de forte pression de la maladie. Les niveaux de rendement consecutifs a la reduction effective des maladies etaient presque les memes que ceux de la modalite sans maladie. L'augmentation totale du rendement variait entre 15 et 30% avec une moyenne globale d'environ 20%, et a donc permis un benefice financier de 50-100 EUR ha - 1 lors des annees considerees.

The efficacy of Metarhizium anisopliae in combination with sublethal concentrations of oils and potassium-oleate for biological control of whiteflies was tested under controlled conditions. Three commercial products (Biola ® , Naturen ® , Neudosan ® ) and five experimental formulations of plant oils were tested. The efficacy of M. anisopliae against Trialeurodes vaporariorum and Bemisia tabaci without additives was about 50%. At 1/20 of their recommended dosages, all compounds tested significantly increased the efficacy of M. anisopliae for the control of T. vaporariorum , with the formulated sunflower oil Biola ® giving the highest synergistic effect, reaching nearly 100% control. Not only was the level of control increased but also the speed of action was improved, resulting in a higher reliability of control. Fifteen wheat varieties commercially grown in Kenya were tested for their susceptibility to head blight and mycotoxin accumulation after inoculation with Fusarium graminearum in pot experiments. The strains of the pathogen used had been isolated from wheat collected in different growing areas of Kenya. Head blight susceptibility was assessed as the percentage of spikelets bleached and area under disease progress curve; kernel colonization by fungal mycelium was determined as ergosterol content. All varieties were found to be moderately to highly susceptible. However, the varieties differed in head blight susceptibility (29–68% of spikelets bleached; mean 54%), fungal colonization (67–187 μg/g ergosterol content; mean 111 μg/g) and the resulting mycotoxin contamination [deoxynivalenol (DON) 5–31 μg/g; mean 13.5 μg/g]. Grain weight reductions due to head blight ranged from 23 to 57% (mean 44%). The varieties could be therefore divided into partially resistant and highly susceptible genotypes. The kernels of highly susceptible varieties had higher mycotoxin and ergosterol contents. However, the kernels of some varieties contained more fungal mycelium (ergosterol) without the corresponding high amounts of DON, suggesting that they possess some resistance to DON accumulation. Less susceptible varieties showed resistance to fungal spread, as indicated by a slow disease development and lower content of fungal biomass.

Details of our long-term research programme concerning the epidemiology of Fusarium spp. and mycotoxin production are summarized. Evaluation of the occurrence of Fusarium spp., mainly on winter wheat (Triticum aestivum), was carried out by investigating Fusarium infection and mycotoxin contamination. Two to 15% of grains were infested during 1995–1998 at three climatologically differing localities of the Rhineland, Germany. Disease progress was accelerated by rainfall during the flowering season. The species most frequently isolated were Fusarium avenaceum, F. poae, F. culmorum and F. graminearum. The mean deoxynivalenol (DON) content varied from 19μgkg−1 (1995) to 310μgkg−1 (1998) and was not always correlated with disease severity. Organic farming systems showed lower rates of infection with ear blight and lower mycotoxin contamination than conventional farming systems.

Actual and potential crop losses of eight major food and cash crops have been estimated by evaluating data from literature and field experiments. Total losses were calculated from yield reductions due to pathogens, animal pests and weeds on a regional, continental and global level. Since 1965, worldwide production of most crops has increased considerably. Simultaneously, crop losses in wheat, potatoes, barley and rice increased by 4 to 10 percent, in maize, soybean, cotton and coffee losses remained unchanged or slightly decreased. The efficacy of crop protection practices was calculated as the percentage of potential losses prevented by control. The efficacy is highest in cotton (55 percent), it reaches only 34 to 38 percent in the food crops rice, wheat and maize. The variability among cropping areas is high: In Western Europe, 61 percent of potential crop losses is prevented, in North America and Oceania 44, in all other regions 38 percent. Due to the small share of Western Europe in worldwide production of 8 percent, the efficacy of actual crop protection worldwide is only 40 percent. In view of population growth and rising food demand crop production has to be increased substantially. As potential loss rates often increase with attainable yields high productivity largely depends on effective crop protection management. Scenarios for the production of food crops by the year 2025 in developed and in developing countries are given. Recent and future developments in crop protection can contribute to establish sustainability in agriculture and to preserve natural resources. However, although effective control methods have been developed for most biotic yield constraints, the use of crop protection products is regulated by economic considerations rather than by food demand.

A total of 27Fusarium culmorum isolates from Germany and 41F. graminearum isolates from Kenya were investigated for aggressiveness and mycotoxin production on wheat ears. In addition, ergosterol content of the kernels from ears inoculated withF. graminearum was determined and theF. culmorum isolates were tested for mycotoxin productionin vitro. For both pathogens, isolates markedly differed in aggressiveness. 59% and 37% of theF. culmorum isolates produced NIV and DON, respectively,in vivo andin vitro. The DON-producing isolates also produced 3-acDONin vitro. The more aggressive isolates produced mainly DON while the less aggressive isolates produced mainly NIV. 12% and 85% of theF. graminearum isolates produced NIV and DON, respectively. The highly aggressive isolates produced higher amounts of DON, aggressiveness being highly correlated to DON content in the kernels. NIV-producing isolates were less aggressive. Ergosterol content of kernels was moderately correlated to aggressiveness but highly correlated to DON content. Disease severity was associated with kernel weight reduction.

The inoculation of wheat ears with 27 isolates ofFusarium culmorum in growth stage 65 reduced 1000-grain weights by 14 to 61%. For the phytopathological characterisation of isolates the virulence on primary wheat leaves and the growth rate an potato-dextrose-agar were assessed. Deoxynivalenol-producing isolates ofF. culmorum reduced the 1000-grain weight more than nivalenol-producing isolates.

Beetles (Coleoptera) are the most diverse animal group on earth and interact with numerous symbiotic or pathogenic microbes in their environments. The red flour beetle Tribolium castaneum is a genetically tractable model beetle species and its whole genome sequence has recently been determined. To advance our understanding of the molecular basis of beetle immunity here we analyzed the whole transcriptome of T. castaneum by high-throughput next generation sequencing technology. Here, we demonstrate that the Illumina/Solexa sequencing approach of cDNA samples from T. castaneum including over 9.7 million reads with 72 base pairs (bp) length (approximately 700 million bp sequence information with about 30× transcriptome coverage) confirms the expression of most predicted genes and enabled subsequent qualitative and quantitative transcriptome analysis. This approach recapitulates our recent quantitative real-time PCR studies of immune-challenged and naïve T. castaneum beetles, validating our approach. Furthermore, this sequencing analysis resulted in the identification of 73 differentially expressed genes upon immune-challenge with statistical significance by comparing expression data to calculated values derived by fitting to generalized linear models. We identified up regulation of diverse immune-related genes (e.g. Toll receptor, serine proteinases, DOPA decarboxylase and thaumatin) and of numerous genes encoding proteins with yet unknown functions. Of note, septic-injury resulted also in the elevated expression of genes encoding heat-shock proteins or cytochrome P450s supporting the view that there is crosstalk between immune and stress responses in T. castaneum. The present study provides a first comprehensive overview of septic-injury responsive genes in T. castaneum beetles. Identified genes advance our understanding of T. castaneum specific gene expression alteration upon immune-challenge in particular and may help to understand beetle immunity in general.

Meloidogyne hapla is one of the most damaging plant-parasitic nematodes in temperate regions. This nematode has a wide host range with more than 500 plant taxa including roses. In Ethiopia, rose production has developed over the past 10 years to the second most important export market after coffee. Considering the high damage potential of M. hapla, infestation of roses in Ethiopia with this nematode could result in major economic losses. Therefore, awareness of this nematode species is extremely important. During two surveys conducted in August 2011 and April 2012, M. hapla was detected in soil samples from six out of nine rose producing farms located in the districts of Ziway, Holleta, Sebeta, and Menagesha. At infested farms, rose plants appeared stunted and less productive and often showed symptoms of chlorosis and wilting. Identification was based on morphological and morphometrical characters of females, males, and second-stage juveniles, which were all within the range of variability known for this species (4). Shape of juvenile stylet knobs, shape of male head, and perennial pattern of the females with characteristic punctuations between the anus and tail terminus were also typical for M. hapla. The morphological identification was confirmed by sequence analysis of the D2-D3 expansion segment of the 28S rDNA gene following amplification with the primers D2A (5′-ACAAGTACCGTGAGGGAAAGTT-3′) and D3B (5′-TCGGAAGGAACCAGCTACTA-3′) (1). PCR products were purified and sequenced at the Macrogene sequencing facility service (Amsterdam, The Netherlands). Sequences were deposited in GenBank (KJ645427 to 33). The sequences were compared with previously published sequences in NCBI database and showed 96 to 100% sequence similarity with M. hapla accession nos. GQ130139, DQ328685, KF430798, and DQ145641. Unfortunately, comparison of sequences did not provide further information about the origin of this Ethiopian population, if it is native to Ethiopia or was imported with infected plant material. To the best of our knowledge, this is the first record of M. hapla occurring in Ethiopia. M. hapla is known as a serious pest of roses in colder climate regions. In Africa, it was previously reported from Tanzania (3) and South Africa (2). Thus, it appears that this species has now become also established in Ethiopia at higher altitudes (1,400 to 2,100 m above sea level) within the urban hinterland of Addis Ababa. Species of Fusarium have significant agro-economical and human health-related impact by infecting diverse crop plants and synthesizing diverse mycotoxins. Here, we investigated interactions of grain-feeding Tenebrio molitor larvae with four grain-colonizing Fusarium species on wheat kernels. Since numerous metabolites produced by Fusarium spp. are toxic to insects, we tested the hypothesis that the insect senses and avoids Fusarium-colonized grains. We found that only kernels colonized with F. avenaceum or Beauveria bassiana (an insect-pathogenic fungal control) were avoided by the larvae as expected. Kernels colonized with F. proliferatum, F. poae or F. culmorum attracted T. molitor larvae significantly more than control kernels. The avoidance/preference correlated with larval feeding behaviors and weight gain. Interestingly, larvae that had consumed F. proliferatum- or F. poae-colonized kernels had similar survival rates as control. Larvae fed on F. culmorum-, F. avenaceum- or B. bassiana-colonized kernels had elevated mortality rates. HPLC analyses confirmed the following mycotoxins produced by the fungal strains on the kernels: fumonisins, enniatins and beauvericin by F. proliferatum, enniatins and beauvericin by F. poae, enniatins by F. avenaceum, and deoxynivalenol and zearalenone by F. culmorum. Our results indicate that T. molitor larvae have the ability to sense potential survival threats of kernels colonized with F. avenaceum or B. bassiana, but not with F. culmorum. Volatiles potentially along with gustatory cues produced by these fungi may represent survival threat signals for the larvae resulting in their avoidance. Although F. proliferatum or F. poae produced fumonisins, enniatins and beauvericin during kernel colonization, the larvae were able to use those kernels as diet without exhibiting increased mortality. Consumption of F. avenaceum-colonized kernels, however, increased larval mortality; these kernels had higher enniatin levels than F. proliferatum or F. poae-colonized ones suggesting that T. molitor can tolerate or metabolize those toxins. peerReviewed

Fusarium spp. are important pathogens of winter wheat, causing yield reduction and decreasing grain quality due to mycotoxin production. Therefore, the occurrence of Fusarium spp. at different locations with two systems of cultivation was investigated. Different isolation conditions were tested in order to validate the sometimes low isolation frequency.

Near-range and remote sensing techniques are excellent alternatives to destructive methods for measuring beneficial effects of fungicides on plant physiology. Different noninvasive sensors and imaging techniques have been used and compared to measure the effects of three fungicidal compounds (bixafen, fluoxastrobin and prothioconazole) on wheat (Triticum aestivum L.) physiology under disease-free conditions in the greenhouse. Depending on the fungicidal treatment, changes in green leaf area and yield parameters were observed. Chlorophyll fluorescence of leaves was useful for measuring differences in the effective quantum yield of PSII. Reflectance measurements of wheat leaves were highly sensitive to changes in plant vitality. The spectral vegetation indices were useful for determining the differences among treatments in terms of leaf senescence, pigments and water content. The analysis of ear and leaf surface temperature was reliable for detecting effects of fungicides on plant senescence. Using nondestructive sensors, it was possible to assess a delay in senescence of wheat due to fungicide application. Furthermore, it was deduced that sensors and imaging methods are useful tools to estimate the effects of fungicides on wheat physiology. Physiological parameters measured by the sensors were actually more sensitive than yield parameters to assess the effect caused by fungicide application on wheat physiology.

Hyperspectral imaging (HSI) offers high potential as a non-invasive diagnostic tool for disease detection. In this paper leaf characteristics and spectral reflectance of sugar beet leaves diseased with Cercospora leaf spot, powdery mildew and leaf rust at different development stages were connected. Light microscopy was used to describe the morphological changes in the host tissue due to pathogen colonisation. Under controlled conditions a hyperspectral imaging line scanning spectrometer (ImSpector V10E) with a spectral resolution of 2.8 nm from 400 to 1000 nm and a spatial resolution of 0.19 mm was used for continuous screening and monitoring of disease symptoms during pathogenesis. A pixel-wise mapping of spectral reflectance in the visible and near-infrared range enabled the detection and detailed description of diseased tissue on the leaf level. Leaf structure was linked to leaf spectral reflectance patterns. Depending on the interaction with the host tissue, the pathogens caused disease-specific spectral signatures. The influence of the pathogens on leaf reflectance was a function of the developmental stage of the disease and of the subarea of the symptoms. Spectral reflectance in combination with Spectral Angle Mapper classification allowed for the differentiation of mature symptoms into zones displaying all ontogenetic stages from young to mature symptoms. Due to a pixel-wise extraction of pure spectral signatures a better understanding of changes in leaf reflectance caused by plant diseases was achieved using HSI. This technology considerably improves the sensitivity and specificity of hyperspectrometry in proximal sensing of plant diseases.
