The actions of a chromatographically identified extract of the marine dinoflagellate Ostreopsis lenticularis, named ostreotoxin-3 (OTX-3), were studied on frog isolated neuromuscular preparations. OTX-3 (1–10μg ml−1) applied to cutaneous pectoris nerve-muscle preparations depolarized skeletal muscle fibres and caused spontaneous contractions. The depolarization was neither reversed by prolonged washing nor by (+)-tubocurarine. OTX-3 decreased the amplitude of miniature end plate potentials (m.e.p.ps) but did not affect their frequency. Extracellular recording of compound action potentials revealed that OTX-3 affected neither excitability nor conduction along intramuscular nerve branches. End-plate potentials (e.p.ps) elicited by nerve stimulation were reduced in amplitude by OTX-3 and even showed reversed polarity in junctions deeply depolarized by the toxin. Membrane depolarization induced by OTX-3 was decreased about 70% in muscles pretreated for 30min with 10μm tetrodotoxin. In contrast, muscles pretreated with 5μm μ-conotoxin GIIIA were completely insensitive to OTX-3-induced depolarization. OTX-3 did not affect e.p.p. amplitude and the quantal content of e.p.ps in junctions in which muscle depolarization was abolished by μ-conotoxin GIIIA. OTX-3 is a novel type of sodium-channel activating toxin that discriminates between nerve and skeletal muscle membranes.

Dibromosceptrin and clathrodin are alkaloid compounds purified from tropical marine sponges of the genus Agelas. Experiments done using the whole cell configuration of the patch clamp technique revealed that these compounds have neurotoxic activity. Both compounds decreased by 27–40% the average maximum amplitudes of pharmacologically isolated inward sodium currents in cells isolated from chick embryo sympathetic ganglia. Current-voltage data, fitted using Boltzmann's equation, did not show any effect of these agents on the voltage dependence of current activation. However, the voltage dependence of current inactivation was shifted toward more negative potentials by dibromosceptrin, changing by an average of 20 mV the voltage for 50% inactivation. In contrast, clathrodin shifted this voltage dependence of inactivation toward more positive potentials and changed the voltage for 50% inactivation by 14 mV. Time for current reactivation was not altered by clathrodin but was slightly prolonged by dibromosceptrin. Similarly, dibromosceptrin was more effective than clathrodin in delaying the time course of current decay. Thus, these two alkaloids appear to be new sodium channel neurotoxins acting through different mechanisms, dibromosceptrin modifying the channel inactivation characteristics and clathrodin probably influencing channel ionic conductance.

Pharmacological studies on the body wall musculature of the sedentary polychaete Sabellastarte magnifica show a potential neurotransmitter role for monoamines and neuropeptides in this organism. All catecholamines induced contraction of longitudinal muscle strips, while serotonin and the neuropeptides FMRFamide and substance P caused a relaxation of both resting and active muscle. In addition, we demonstrate catecholaminergic and serotonergic pathways in the nervous system of this sabellid, using immunohistochemistry and catecholamine-induced fluorescence. The presence of neuropeptide-containing fibers in the nervous system of this polychaete has been previously reported. Together these results suggest that catecholamines act as excitatory transmitters on the longitudinal muscle cells of the body wall of S. magnifica, while serotonin and FMRFamide, and possible substance P, are inhibitory transmitters. The possibility of coexistence of serotonin and FMRFamide within the same neuronal cell bodies and fibers of this polychaete is also explored. © 1992 Wiley-Liss, Inc.

Immunohistochemical studies of the nervous system of Sabellastarte magnifica, a sedentary polychaete, showed the presence of neuropeptide expressing cells and fibers within the double ventral nerve cord. Immunoreactivity to cholecystokinin, neuropeptide Y, enkephalins, substance P, and FMRFamide was found to be present in specific populations of cells, identifiable by their location and by the neuropeptide they expressed. Fibers expressing the various neuropeptides were also observed in particular locations within the nerve cord. This characteristic distribution of the various neuron subgroups and fiber pathways may represent functional circuits within the nervous system of this annelid.

The contractility of guinea pig ileum and frog skeletal muscle is inhibited in solutions containing 0.4 to 2.5 M formamide (FMD). Contrary to mammalian vis ceral muscle, this blocking action is not reversed when frog muscles are transferred back to isotonic Ringer's after FMD treatment. Under these conditions the water content of the skeletal muscles is markedly increased and electronmicrographs show a swellingof the transverse tubules. These changes are not observed when frog muscles are transferred to ethylene glycol solutions that are isosmotic with the FMD containing Ringer's solution. In addition, over 50% of the contractility is recovered in these muscles. These observations provide direct evidence of the occurrence of an osmotic shock in frog muscles transferred from FMD solutions to isotonic Ringer's. It is concluded that the resulting alterations in the triad structure and function are re sponsible for the irreversibility of the FMD uncoupling action in these muscles.

Buchtal and collaborators reported thirty years ago an excitatory action of low (10 −3 M) concentrations of ATP on frog muscle, as well as in increase in the sensitivity of the muscle to ACh. These effects have been re-investigated employing both intra-and extra-cellular recording and the technique of iontophoretic drug application. ATP at a concentration of 10 −4 M decreases the recorded resting potential by about 35%. The depolarizing action of ATP is more pronounced in the tibial end of the frog sartorius muscle than in the nerve free pelvic end. In addition ATP, added to the bath and electro-osmotically applied, increases the depolarizing action of ACh. This potentiating effect is particularly marked in denervated muscles.
