Climacteric ripening of tomato fruit is initiated by a characteristic surge of the production rate of ethylene, accompanied by an increase in respiration rate. As both activities consume O2 and produce CO2 , gas concentration gradients develop in the fruit that cause diffusive transport. This may, in turn, affect respiration and ethylene biosynthesis. Gas diffusion in fruit depends on the amount and connectivity of cells and intercellular spaces in 3D. We investigated micromorphological changes in different tomato tissues during development and ripening by visualizing cells and pores based on high-resolution micro-computed tomography, and computed effective O2 diffusivity coefficients based on microstructural features of the tissues. We demonstrated that mesocarp and septa tissues have larger cells but small and more disconnected pores than the placenta and columella, resulting in relatively lower effective O2 diffusivity coefficients. Cell disintegration occurred in the mesocarp and septa during ripening, indicating lysigenous air pore formation and resulting in a gradual increase of the effective O2 diffusivity. The results suggest that hypoxic conditions caused by the increasing size and, hence, diffusion resistance of the growing fruit may induce an increase of tissue porosity that results in a greatly enhanced O2 diffusivity and, thus, helps to alleviate them. ispartof: NEW PHYTOLOGIST vol:232 issue:5 pages:2043-2056 ispartof: location:England status: published

The evolution of hemoglobin function in the transition from water- to air-breathing has been highly debated but remains unresolved. Here, we characterized the hemoglobin function in five closely related water- and air-breathing catfishes. We identify distinct directions of hemoglobin evolution in the clades that evolved air-breathing, and we show strong selection on hemoglobin function within the catfishes. These findings show that the lack of a general direction in hemoglobin function in the transition from water- to air-breathing may have resulted from divergent selection on hemoglobin function in independent clades of air-breathing fishes.

Rice (Oryza sativa) is a water-intensive crop, and like other plants uses stomata to balance CO2 uptake with water-loss. To identify agronomic traits related to rice stomatal complexes, an anatomical screen of 64 Thai and 100 global rice cultivars was undertaken. Epidermal outgrowths called papillae were identified on the stomatal subsidiary cells of all cultivars. These were also detected on eight other species of the Oryza genus but not on the stomata of any other plant species we surveyed. Our rice screen identified two cultivars that had “mega-papillae” that were so large or abundant that their stomatal pores were partially occluded; Kalubala Vee had extra-large papillae, and Dharia had approximately twice the normal number of papillae. These were most accentuated on the flag leaves, but mega-papillae were also detectable on earlier forming leaves. Energy dispersive X-Ray spectrometry revealed that silicon is the major component of stomatal papillae. We studied the potential function(s) of mega-papillae by assessing gas exchange and pathogen infection rates. Under saturating light conditions, mega-papillae bearing cultivars had reduced stomatal conductance and their stomata were slower to close and re-open, but photosynthetic assimilation was not significantly affected. Assessment of an F3 hybrid population treated with Xanthomonas oryzae pv. oryzicola indicated that subsidiary cell mega-papillae may aid in preventing bacterial leaf streak infection. Our results highlight stomatal mega-papillae as a novel rice trait that influences gas exchange, stomatal dynamics, and defense against stomatal pathogens which we propose could benefit the performance of future rice crops.

Nutrient deficiency alters growth and the production of high-quality nutritious food. In Citrus crops, rootstock technologies have become a key tool for enhancing tolerance to abiotic stress. The use of doubled diploid rootstocks can improve adaptation to lower nutrient inputs. This study investigated leaf structure and ultrastructure and physiological and biochemical parameters of diploid common clementine scions (C) grafted on diploid (2x) and doubled diploid (4x) Carrizo citrange (C/CC2x and C/CC4x) and Citrumelo 4475 (C/CM2x and C/CM4x) rootstocks under optimal fertigation and after 7 months of nutrient deficiency. Rootstock ploidy level had no impact on structure but induced changes in the number and/or size of cells and some cell components of 2x common clementine leaves under optimal nutrition. Rootstock ploidy level did not modify gas exchanges in Carrizo citrange but induced a reduction in the leaf net photosynthetic rate in Citrumelo 4475. By assessing foliar damage, changes in photosynthetic processes and malondialdehyde accumulation, we found that C/CM4x were less affected by nutrient deficiency than the other scion/rootstock combinations. Their greater tolerance to nutrient deficiency was probably due to the better performance of the enzyme-based antioxidant system. Nutrient deficiency had similar impacts on C/CC2x and C/CC4x. Tolerance to nutrient deficiency can therefore be improved by rootstock polyploidy but remains dependent on the rootstock genotype.

Premise of the Study: The global climate during the early Miocene was warmer than the present and preceded the even warmer middle Miocene climatic optimum. The paleo-CO2 records for this interval suggest paradoxically low concentrations (95% confidence) that CO2 during the late early Miocene exceeded 400 ppm. These results revise upwards the likely CO2 concentration at this time, more in keeping with a CO2-forced greenhouse climate. © 2018 Botanical Society of America

Biomass crops are commonly grown in low‐grade land and selection of drought‐tolerant accessions is of major importance to sustain productivity. In this work, we assess phenotypic variation under different environmental scenarios in a series of accessions of Arundo donax, and contrast it with two closely related species, Arundo donaciformis and Arundo plinii. Gas‐exchange and stomatal anatomy analysis showed an elevated photosynthetic capacity in A. plinii compared to A. donax and A. donaciformis with a significant intraspecific variation in A. donax. The three species showed significantly contrasting behaviour of transpiration under developing water stress and increasing vapour pressure deficit (VPD), with A. donax being the most conservative while A. plinii showed an elevated degree of insensitivity to environmental cues. Under optimal conditions, A. donax had the highest estimated leaf area (projected leaf area) and plant dry weight although a significant reduction under water stress was observed for A. donax and A. donaciformis accessions while no differences were recorded for A. plinii between optimal growing conditions (well‐watered [WW]) and reduced soil water availability (water‐stressed [WS]). A. donax displayed a markedly conservative water use behaviour but elevated sensitivity of biomass accumulation under stress conditions. By contrast, in A. plinii, biomass and transpiration were largely insensitive to WS and increasing VPD, though biomass dry weight under optimal conditions was significantly lower than A. donax. We provide evidence of interspecific phenotypic variation within the Arundo genus while the intraspecific phenotypic plasticity may be exploited for further selection of superior clones under disadvantageous environmental conditions. The extensive trade‐off between water use and biomass accumulation present in the three species under stress conditions provides a series of novel traits to be exploited in the selection of superior clones adapted to different environmental scenarios. Non‐destructive approaches are provided to screen large populations for water‐stress‐tolerant A. donax clones., We compared by high‐throughput phenotyping natural populations of three species of the Arundo genus, the giant reed Arundo donax and two of its wild relatives, Arundo donaciformis and Arundo plinii, for drought tolerance. A clearly water‐conservative behavior was observed in A. donax and A. donaciformis. Conversely, A. plinii displayed an overall non‐conservative response to developing water stress. The intraspecific variation we uncovered in A. donax suggests the possibility of selecting natural ecotypes for agronomic use with superior combinations of preferable traits for different environments. In addition, our work stresses the importance of non‐destructive phenotyping approaches for plant‐environment interaction studies.

Transpiration is a key component of the watershed water budget. Therefore, determining the transpiration of forest stands with different characteristics have been of interest for watershed hydrology and forest management practices in a wide range of environments. The objectives of this study were to compare transpiration of Anatolian black pine (Pinus nigra Arn. subsp. pallasiana (Lamb.) Holmboe) and sessile oak (Quercus petraea (Matt.) Liebl.) tree stands and to model transpiration based on the measured climatic factors. Stand transpirations were calculated from sap flow measurements made by the trunk heat balance method. We conducted an exploratory factor analysis (PCR) to detect affecting meteorological factors of stand transpiration, and we developed linear regression equations to predict transpiration of pine and oak stands. Mean daily and yearly canopy transpiration (Ec) were 1.05 mm day(-1) and 378.3 mm year(-1) for the pine stand and 3.52 mm day(-1) and 801.7 mm year(-1) for the oak stand. There was a highly positive correlation between daily stand transpiration and wind speed, global radiation, air vapour pressure deficit and air temperature, but a negative correlation with relative humidity for both stands. Soil water potential had little effect on stand transpiration. The model equations accounted for 81% of the variations in transpiration for the pine stand and 85% for the oak stand. Therefore, the transpiration of forest stands should be considered for effective vegetation management practices, as model equations to estimate the transpiration of pine and oak stands in the region. Pines are the dominant conifers in Mediterranean forests. As long-lived sessile organisms that seasonally have to cope with drought periods, they have developed a variety of adaptive responses. However, during last decades, highly intense and long-lasting drought events could have contributed to decay and mortality of the most susceptible trees. Among conifer species, Pinus pinaster Ait. shows remarkable ability to adapt to different environments. Previous molecular analysis of a full-sib family designed to study drought response led us to find active transcriptional activity of stress-responding genes even without water deprivation in tolerant genotypes. To improve our knowledge about communication between above- and below-ground organs of maritime pine, we have analyzed four graft-type constructions using two siblings as rootstocks and their progenitors, Gal 1056 and Oria 6, as scions. Transcriptomic profiles of needles from both scions were modified by the rootstock they were grafted on. However, the most significant differential gene expression was observed in drought-sensitive Gal 1056, while in drought-tolerant Oria 6, differential gene expression was very much lower. Furthermore, both scions grafted onto drought-tolerant rootstocks showed activation of genes involved in tolerance to abiotic stress, and is most remarkable in Oria 6 grafts where higher accumulation of transcripts involved in phytohormone action, transcriptional regulation, photosynthesis and signaling has been found. Additionally, processes, such as those related to secondary metabolism, were mainly associated with the scion genotype. This study aimed to assess the toxicity of glyphosate (GLY; 0, 10, 20 and 30?mg?kg-1) in Solanum lycopersicum L., particularly focusing on the photosynthetic metabolism. By combining ecophysiological, ultrastructural, biochemical and molecular tools, the results revealed that the exposure of tomato plants to GLY led to alterations in leaf water balance regulation [increasing stomatal conductance (gs) and decreasing water use efficiency (WUEi) at higher concentrations] and induced slight alterations in the structural integrity of cells, mainly in chloroplasts, accompanied by a loss of cell viability. Moreover, the transcriptional and biochemical control of several photosynthetic-related parameters was reduced upon GLY exposure. However, in vivo chlorophyll fluorometry and IRGA gas-exchange studies revealed that the photosynthetic yield of S. lycopersicum was not repressed by GLY. Overall, GLY impacts cellular and subcellular homeostasis (by affecting chloroplast structure, reducing photosynthetic pigments and inhibiting photosynthetic-related genes transcription), and leaf structure, but is not reducing the carbon flow on a leaf area basis. Plant functional traits have shown to be relevant predictors of forest functional responses to climate change. However, the trait-based approach to study plant performances and ecological strategies has mostly been focused on trait comparisons at the interspecific and intraspecific levels. In this study, we analyzed traits variation and association at the individual level. We measured wood and leaf traits at different height locations within the crown of five individuals of Pericopsis elata (Harms) Meeuwen (Fabaceae) from the northern tropical forest of the Democratic Republic of the Congo. All traits varied between and within individuals. The between-individual variation was more important for leaf traits (23%&ndash, 48%) than for wood traits (~10%) where the within-individual variation showed to be more important (33%&ndash, 39%). The sample location height within the crown was found to be the driving factor of this within-individual variation. In a gradient from the base to the top of the crown, theoretical specific hydraulic conductivity and specific leaf area decreased while the stomatal density increased. We found significant relationships among traits and between wood and leaf traits. However, these relationships varied with the position within the crown. The relationship between vessel size and vessel density was negative at the bottom part of the crown but positive upward. Also, the negative relationship between stomatal density and stomatal size became stronger with increasing height within the crown. Finally, the positive relationship between specific leaf area and theoretical specific hydraulic conductivity became stronger in higher parts of the crown, suggesting that P. elata constantly adapts its water use with respect to its water supply, more strongly at the top of the crown where the environment is more extreme and less buffered against environmental fluctuations.

Chronic obstructive pulmonary disease (COPD) is a progressive lung disease characterized by pathophysiological factors including airflow limitation, hyperinflation and reduced gas exchange. Treatment consists of lifestyle changes, lung rehabilitation and pharmacological therapies such as long acting beta-2-agonists (LABA) and long acting muscarinic antagonists (LAMA). More recently bronchoscopic treatments are emerging for COPD. Among them endobronchial valves (EBV) and endobronchial coils (EBC), next to endobronchial stents, sclerosing agents, targeted lung denervation and liquid nitrogen metered cryospray. In this review we aim to summarize the new emerging bronchoscopic treatments and their effect sizes compared with lung rehabilitation and pharmacological therapies.

Strawberry and wild strawberry are among the most popular horticultural crops. Due to the development of soilless cultivation systems, the whole-year production of these economically important fruit crops is achievable even in countries with temperate climate. However, the responses of strawberry (Fragaria × ananassa Duch.) and wild strawberry (Fragaria vesca L.) to microclimate conditions in greenhouse gutter cultivation in the autumn–winter season in Poland have not been yet determined. The aim of this study was to analyze the physiological, biochemical, and biometrical responses of two cultivars of strawberry ‘Ostara’ and ‘San Andreas’ and two cultivars of wild strawberry ‘Baron von Solemacher’ and ‘Regina’ grown for 20 weeks, starting from September 17th, in controlled greenhouse conditions on coconut mats in gutters in the autumn–winter season and irradiated with sodium lamps. Strawberry ‘San Andreas’ produced 30% larger leaves and almost three-fold higher fresh and dry weight of biomass than ‘Ostara’. The strawberry plants ‘San Andreas’ had a higher content (20%) of chlorophyll a and 30% of chlorophyll b than ‘Ostara’ plants. Generally, ‘San Andreas’ displayed an overall higher concentration of intercellular CO2 (about 14%) than ‘Ostara’ plants providing higher gas exchange processes. Photosynthetic rate amounted to 13.0 μmol·m−2·s−1 for ‘San Andreas’ that was almost two-fold higher than for ‘Ostara’. ‘San Andreas’ flower and fruit productions were uniform and the six-fold higher individual fruit yield proved the excellent attributes of this cultivar to the greenhouse cultivation. Even though the productivity of the two studied wild strawberry cultivars was similar, ‘Regina’ showed higher values of some parameters than ‘Baron von Solemacher’ (40% larger leaves, 25% higher photosynthetic rate, 10% higher concentration of intercellular CO2). A high nutritional value of fruits is maintained compared to traditional open-air cultivation.

The accumulation of CO2 in the atmosphere and resulting ocean acidification represent a threat to marine ecosystems. While acid–base regulatory capacity is well developed in marine fish, allowing compensation of extra-cellular pH during short-term hypercapnia, the possible energetic costs of such regulation during long-term exposure remain to be established. In this study, juvenile European sea bass (Dicentrarchus labrax) were exposed from 2 days post-hatching to three different ocean acidification scenarios: control (present condition, $$ P_{{{\text{CO}}_{2} }} $$ = 520 µatm, pH 7.9), moderate acidification ( $$ P_{{{\text{CO}}_{2} }} $$ = 950 µatm, pH 7.7), and high acidification ( $$ P_{{{\text{CO}}_{2} }} $$ = 1490 µatm, pH 7.5). After 1.5 years of exposure, fish aerobic metabolic capacities, as well as elements of their oxygen extraction and transport chain, were measured. Compared to control, $$ P_{{{\text{CO}}_{2} }} $$ treatments did not affect fish standard metabolic rate (SMR). However, the most severe acidification condition was associated with a significantly elevated maximum metabolic rate (MMR).This was supported by heavier gill system and higher blood haemoglobin concentration. A reduction of maximum cardiac frequency (fHmax) during incremental warming of anaesthetized fish was also observed in both acidification scenarios. On the other hand, the critical oxygen level (O2crit), the minimum oxygen level required to sustain SMR, did not differ among groups. The increased MMR, associated with maintained SMR, suggests that acid–base compensatory processes, although not increasing maintenance costs, may affect components of bass homeostasis, resulting in new internal physico-chemical conditions. The possibility that these alterations influence metabolic pathways and physiological functions involved in fish aptitude to maximally transport oxygen is discussed.

It has been suggested that plant carbon (C) use by symbiotic arbuscular mycorrhizal fungi (AMF) may be compensated by higher photosynthetic rates because fungal metabolism creates a strong C sink that prevents photosynthate accumulation and down-regulation of photosynthesis. This mechanism remains largely unexplored and lacks experimental evidence. We report here two experiments showing that the experimental manipulation of the mycorrhizal C sink significantly affected photosynthetic rates of cucumber host plants. We expected that a sudden reduction in sink strength would cause a significant reduction in photosynthetic rates, at least temporarily. Excision of part of the extraradical mycorrhizal mycelium from roots, and causing no disturbance to the plant, induced a sustained (10-40%) decline in photosynthetic rates that lasted from 30 min to several hours in plants that were well nourished and hydrated and in the absence of growth or photosynthesis promotion by mycorrhizal inoculation. This effect was though minor in plants growing at high (700 ppm) atmospheric CO2 . This is first direct experimental evidence for the carbon sink strength effects exerted by arbuscular mycorrhizal symbionts on plant photosynthesis. It encourages further experimentation on mycorrhizal source-sink relations and may have strong implications in large-scale assessments and modelling of plant photosynthesis. International audience; Mutants affected in complex I are useful to understand the role played by mitochondrial electron transport and redox metabolism in cellular homeostasis and signaling. However, their respiratory phenotype is incompletely described and a specific examination of day respiration (Rd) is lacking. Here, we used isotopic methods and metabolomics to investigate the impact of complex I dysfunction on Rd in two respiratory mutants of forest tobacco (Nicotiana sylvestris): cytoplasmic male sterile II (CMSII) and nuclear male sterile 1 (NMS1), previously characterized for complex I disruption. Rd was higher in mutants and the inhibition of leaf respiration by light was lower. Higher Rd values were caused by increased (phosphoenol)pyruvate (PEP) metabolism at the expense of anaplerotic (PEP carboxylase (PEPc) ‐catalyzed) activity. De novo synthesis of Krebs cycle intermediates in the light was larger in mutants than in the wild‐type, although numerically small in all genotypes. Carbon metabolism in mutants involved alternative pathways, such as alanine synthesis, and an increase in amino acid production with the notable exception of aspartate. Our results show that the alteration of NADH re‐oxidation activity by complex I does not cause a general inhibition of catabolism, but rather a re‐orchestration of fluxes in day respiratory metabolism, leading to an increased CO2 efflux.

Aim of this study was to investigate the effect of water shortage in wild jujube plants, Ziziphus lotus (L.) Lam, and how it is related to its ecological success. All leaf anatomical traits were significantly affected following soil drying. Stressed plants displayed more negative stem water potential (psi(w)) and osmotic potential values of ca. -3.5 and -4.5 MPa, respectively, after 30 d. The relative water content declined although it still maintained high values (>= 75%). The net photosynthetic rate, stomatal conductance, and transpiration rate were significantly inhibited as psi(w) decreased. However, the intrinsic water-use efficiency increased as water deficit was intensified; the difference became significant only after 30 d. As a consequence, the effective quantum yield of PSII photochemistry and the photochemical quenching coefficient significantly decreased although the maximal quantum yield of PSII photochemistry ratio remained statisticaly unchanged. Plants could maintain their water status sufficiently by increasing proline 1.7-fold and sucrose 1.8-fold in their leaves, respectively. The largest accumulation of both solutes may avoid photodamages at cellular level and play a critical role in maintaining osmotic adjustment.

Background and Aims Experimental drought is well documented to induce a decline in photosynthetic capacity. However, if given time to acclimate to low water availability, the photosynthetic responses of plants to low soil moisture content may differ from those found in short-term experiments. This study aims to test whether plants acclimate to long-term water stress by modifying the functional relationships between photosynthetic traits and water stress, and whether species of contrasting habitat differ in their degree of acclimation. Methods Three Eucalyptus taxa from xeric and riparian habitats were compared with regard to their gas exchange responses under short- and long-term drought. Photosynthetic parameters were measured after 2 and 4 months of watering treatments, namely field capacity or partial drought. At 4 months, all plants were watered to field capacity, then watering was stopped. Further measurements were made during the subsequent ‘drying-down’, continuing until stomata were closed. Key Results Two months of partial drought consistently reduced assimilation rate, stomatal sensitivity parameters (g1), apparent maximum Rubisco activity (V′cmaxVcmax′) and maximum electron transport rate (J′maxJmax′). Eucalyptus occidentalis from the xeric habitat showed the smallest decline in V′cmaxVcmax′ and J′maxJmax′; however, after 4 months, V′cmaxVcmax′ and J′maxJmax′ had recovered. Species differed in their degree of V′cmaxVcmax′ acclimation. Eucalyptus occidentalis showed significant acclimation of the pre-dawn leaf water potential at which the V′cmaxVcmax′ and ‘true’ Vcmax (accounting for mesophyll conductance) declined most steeply during drying-down. Conclusions The findings indicate carbon loss under prolonged drought could be over-estimated without accounting for acclimation. In particular, (1) species from contrasting habitats differed in the magnitude of V′cmax reduction in short-term drought; (2) long-term drought allowed the possibility of acclimation, such that V′cmax reduction was mitigated; (3) xeric species showed a greater degree of V′cmax acclimation; and (4) photosynthetic acclimation involves hydraulic adjustments to reduce water loss while maintaining photosynthesis.

Following asymmetric pruning of leaves and/or roots, the functional balance of distribution of carbon, but not of nitrogen, in Cunninghamia lanceolata is more readily achieved for the roots and leaves on the same side of the pruning compared with those on the opposite side. Abstract The functional balance between leaves and roots is believed to be mediated by the specific location of shoots and roots, i.e. differences in transport distances and degrees of organ connectivity. However, it remains unknown whether the adaptive responses of trees to biomass removal depend on the relative orientation of leaf and root pruning. Here, we applied five pruning treatments to saplings of Cunninghamia lanceolata (Chinese fir) under field and glasshouse conditions, namely no pruning (control), half of lateral branches pruned, half of lateral roots pruned, half of the branches and roots pruned on the same side of the plant, and half of the branches and roots pruned on opposite sides of the plant. The effects of pruning on the growth, carbon storage and allocation, and physiology of leaves and fine roots on the same and opposite sides of the plant were investigated. Compared with the effect of root-pruning on leaves, fine roots were more limited by carbon availability and their physiological activity was more strongly reduced by shoot pruning, especially when branches on the same side of the plant were removed. Pruning of branches and roots on the opposite side of the plant resulted in the lowest carbon assimilation rates and growth among all treatments. The results of a stable-isotope labeling indicated that less C was distributed to fine roots from the leaves on the opposite side of the plant compared to those on the same side, but N allocation from roots to leaves depended less on the relative root and leaf orientation. The results collectively indicate that the functional responses of C. lanceolata to pruning are not only determined by the source-sink balance model but are also related to interactions between leaves and fine roots. We argue that the connectivity among lateral branches and roots depends on their relative orientation, which is therefore critical for the functional balance between leaves and fine roots.

Diving mammals have evolved a suite of physiological adaptations to manage respiratory gases during extended breath-hold dives. To test the hypothesis that offshore bottlenose dolphins have evolved physiological adaptations to improve their ability for extended deep dives and as protection for lung barotrauma, we investigated the lung function and respiratory physiology of four wild common bottlenose dolphins (Tursiops truncatus) near the island of Bermuda. We measured blood hematocrit (Hct, %), resting metabolic rate (RMR, l O 2 · min -1), tidal volume (V T, l), respiratory frequency (f R, breaths · min -1), respiratory flow (l · min -1), and dynamic lung compliance (C L, l · cmH 2O -1) in air and in water, and compared measurements with published results from coastal, shallow-diving dolphins. We found that offshore dolphins had greater Hct (56 ± 2%) compared to shallow-diving bottlenose dolphins (range: 30-49%), thus resulting in a greater O 2 storage capacity and longer aerobic diving duration. Contrary to our hypothesis, the specific C L (sC L, 0.30 ± 0.12 cmH 2O -1) was not different between populations. Neither the mass-specific RMR (3.0 ± 1.7 ml O 2 · min -1 · kg -1) nor V T (23.0 ± 3.7 ml · kg -1) were different from coastal ecotype bottlenose dolphins, both in the wild and under managed care, suggesting that deep-diving dolphins do not have metabolic or respiratory adaptations that differ from the shallow-diving ecotypes. The lack of respiratory adaptations for deep diving further support the recently developed hypothesis that gas management in cetaceans is not entirely passive but governed by alteration in the ventilation-perfusion matching, which allows for selective gas exchange to protect against diving related problems such as decompression sickness.

Fish regulate ventilation primarily by sensing O2-levels in the water and arterial blood. It is well established that this sensory process involves several steps, but the underlying mechanisms remain frustratingly elusive. Here we examine the effect of increasing lactate ions at constant pH on ventilation in a teleost; specifically the facultative air-breathing catfish Pangasianodon hypophthalmus. At lactate levels within the physiological range obtained by Na-Lactate injections (3.5 ± 0.8 to 10.9 ± 0.7 mmol L−1), gill ventilation increased in a dose-dependent manner to levels comparable to those elicited by NaCN injections (2.0 µmol kg−1), which induces a hypoxic response and higher than those observed in any level of ambient hypoxia (lowest PO2 = 20 mmHg). High lactate concentrations also stimulated air-breathing. Denervation of the first gill arch reduced the ventilatory response to lactate suggesting that part of the sensory mechanism for lactate is located at the first gill arch. However, since a residual response remained after this denervation, the other gill arches or extrabranchial locations must also be important for lactate sensing. We propose that lactate plays a role as a signalling molecule in the hypoxic ventilatory response in fish.

Optimization models of stomatal conductance (g(s)) attempt to explain observed stomatal behaviour in terms of cost-benefit tradeoffs. While the benefit of stomatal opening through increased CO2 uptake is clear, currently the nature of the associated cost(s) remains unclear. We explored the hypothesis that g(s) maximizes leaf photosynthesis, where the cost of stomatal opening arises from nonstomatal reductions in photosynthesis induced by leaf water stress. We analytically solved two cases, CAP and MES, in which reduced leaf water potential leads to reductions in carboxylation capacity (CAP) and mesophyll conductance (g(m)) (MES). Both CAP and MES predict the same one-parameter relationship between the intercellular:atmospheric CO2 concentration ratio (c(i)/c(a)) and vapour pressure deficit (VPD, D), viz. c(i)/c(a) approximate to xi/xi (xi+D), as that obtained from previous optimization models, with the novel feature that the parameter xi is determined unambiguously as a function of a small number of photosynthetic and hydraulic variables. These include soil-to-leaf hydraulic conductance, implying a stomatal closure response to drought. MES also predicts that g(s)/g(m) is closely related to c(i)/c(a) and is similarly conservative. These results are consistent with observations, give rise to new testable predictions, and offer new insights into the covariation of stomatal, mesophyll and hydraulic conductances.

Mitochondrial respiration in the dark (R dark) is a critical plant physiological process, and hence a reliable, efficient and high-throughput method of measuring variation in rates of R dark is essential for agronomic and ecological studies. However, currently methods used to measure R dark in plant tissues are typically low throughput. We assessed a high-throughput automated fluorophore system of detecting multiple O2 consumption rates. The fluorophore technique was compared with O2-electrodes, infrared gas analysers (IRGA), and membrane inlet mass spectrometry, to determine accuracy and speed of detecting respiratory fluxes. The high-throughput fluorophore system provided stable measurements of R dark in detached leaf and root tissues over many hours. High-throughput potential was evident in that the fluorophore system was 10 to 26-fold faster per sample measurement than other conventional methods. The versatility of the technique was evident in its enabling: (1) rapid screening of R dark in 138 genotypes of wheat; and, (2) quantification of rarely-assessed whole-plant R dark through dissection and simultaneous measurements of above- and below-ground organs. Variation in absolute R dark was observed between techniques, likely due to variation in sample conditions (i.e. liquid vs. gas-phase, open vs. closed systems), indicating that comparisons between studies using different measuring apparatus may not be feasible. However, the high-throughput protocol we present provided similar values of R dark to the most commonly used IRGA instrument currently employed by plant scientists. Together with the greater than tenfold increase in sample processing speed, we conclude that the high-throughput protocol enables reliable, stable and reproducible measurements of R dark on multiple samples simultaneously, irrespective of plant or tissue type.

We investigated the drivers of street tree transpiration in boreal conditions, in order to better understand tree water use in the context of urban tree planning and stormwater management. Two streets built in Helsinki in 2002, hemiboreal zone that had been planted either with Tilia x vulgaris or Alnus glutinosa f. pyramidalis were used as the study sites. Tree water use was measured from sap flow over the 2008-2011 period by the heat dissipation method. Penman-Monteith based evapotranspiration models of increasing complexity were tested against the tree water use measurements to assess the role of environmental and tree related factors in tree transpiration. Alnus and Tilia respectively used 1.1 and 0.8 l of water per m(2) of leaf area per day under ample water conditions, but the annual variation was high. The Penman-Monteith evapotranspiration estimate and soil water status changes explained over 80 % of the variation in tree transpiration when the model was parameterized annually. The addition of tree crown surface area in the model improved its accuracy and diminished variation between years and sites. Using single parameterization over all four years instead of annually varying one did not produce reliable estimates of tree transpiration. Tree transpiration, scaled to different canopy cover percentages, implied that the columnar Alnus trees could transpire as much as all annual rainfall at or less than 50 % canopy cover.

To better understand the poor regulation of water loss after leaf development at high relative air humidity (RH), the relative importance of the physiological and anatomical components was analyzed focusing on cultivars with a contrasting sensitivity to elevated RH. The stomatal responsiveness to three closing stimuli (desiccation, abscisic acid feeding, light/dark transition), as well as several stomatal features (density, index, size and pore dimensions) and the cuticular transpiration rate (CTR) were determined in four rose cultivars, grown under moderate (60%) and high (95%) RH. Moreover, the effects of changes in stomatal density and pore dimensions on the stomatal conductance (gs) were quantified using a modified version of the Brown and Escombe equation. Higher water loss, as a result of plant growth at high RH, was primarily caused by an increase in residual gs, and to a lesser extent due to higher CTR. It was estimated that in leaflets subjected to desiccation the enhanced gs in high RH- as compared to moderate RH-grown plants was mostly due to poor stomatal functionality and to a lesser extent the combined result of higher stomatal density and longer pore length. It is concluded that the reduced degree and, specially, the reduced rate of stomatal closure are the primary causes of the large genotypic variation in the control of water loss in high RH-grown plants. Furthermore, it was found that although changes in stomatal length have no influence on stomatal functionality, changed anatomical features per se represent a significant and direct contribution to the increased water loss.

Viticulture is in great need of new rootstocks sharing features of scion growth control and tolerance to major biotic and abiotic stress factors. A two year study was carried out in pots to assess performances of the two new M1 and M3 rootstocks vs. those of the commercial rootstocks 1103P and 101-14. Potted vines of M1, M3, 1103P and 101-14 rootstocks were grown in a calcareous and non calcareous soil and for two consecutive seasons subjected to three N supply levels at 0, 2 and 4 g of N per pot. Vegetative growth, leaf gas exchange, leaf greenness index (GI) and leaf blade nutrition were assessed. M1 and 1103P were the least vigorous genotypes in terms of total pruning weight; M1 also manifested a stronger apical dominance. Both M rootstocks and 101-14 showed increased leaf WUE at both N supply levels which was due to ability to maintain, at increasing N supply, similar leaf assimilation rates while significantly reducing leaf transpiration. Common tendency of any rootstocks was that increasing N supply corresponded to lowered leaf concentration of K, P, Mg and B. M1 was able to combine a series of desirable features including lower vigor, strong apical dominance, higher WUE at increasing N supply and quite well balanced leaf nutritional pattern. In the present trial M3 did not have the expected devigorating effect.

The river valleys in Southwest China are characterized by a dry–hot climate and relatively rich soils, and host valley-type savannas that are dominated by deciduous species. However, little is known about the ecological adaptations of Chinese savanna plants to the local environments. We hypothesize that Chinese savanna species mainly possess a drought-avoiding strategy by having a deciduous leaf habit and have productive leaf traits. To test this hypothesis, we measured 26 anatomical, morphological, physiological, and chemical traits for 33 woody species from a valley savanna in Southwest China and compared them with the literature data of other dry and wet tropical tree species and a global dataset. We found that Chinese savanna species showed drought avoidance adaptations and exhibited productive leaf traits, such as thin and dense leaves with high ratio of palisade to spongy mesophyll, leaf nutrient concentrations and photosynthetic capacity. Correlations of photosynthetic capacity with N, P, and stomatal conductance across Chinese savanna species were consistent with global patterns reported for seed plants. However, the Chinese savanna species had consistently greater carbon gain at a given specific leaf area, N, P, and stomatal conductance, suggesting higher nutrient- and intrinsic water use efficiencies. These results suggest that paradoxically, Chinese savanna species are adapted to the stressful dry–hot valley habitat by having productive leaves.

To compare the effect of arbuscular mycorrhiza (AM) and P-supplement on N uptake and N assimilation under well-watered or drought-stressed conditions, Glomus intraradices-colonised, P-supplemented non-mycorrhizal (P) and non-mycorrhizal (control) plants of Lolium perenne were exposed to 12 days of water treatment. Leaf water potential (Ψ w), photosynthetic ability, and N and P nutritional status were measured at the beginning (day 0) and end (day 12) of water treatment. N absorption, amino acid and protein synthesis were quantified using the isotopic tracer 15N at day 12. Under well-watered conditions, growth response and physiological parameters were similar in AM and P plants, as compared to controls. Drought (10% water) significantly decreased these parameters in all three treatments. As compared to control plants, the negative impact of water deficit on the Ψ w, photosynthesis, biomass, and N and P content was highly alleviated in AM plants, while only slightly improved or remained the same level in P plants. The effect of AM symbiosis on N absorption and N assimilation was greater than that of the P supplement under well-watered and drought-stressed conditions, and this effect was highly enhanced under drought-stressed conditions. At terminal drought stress on day 12, the effect of AM colonisation on de novo synthesis of amino acids and proteins was 4.4- and 4.8-fold higher than that of the P supplement. These results indicate that the AM symbiosis plays an integrative role in N nutrition by alleviating the negative impacts of drought on N or P uptake and N assimilation, whereas the efficiency of a direct P supplement is very limited under drought-stressed conditions.

1. The object of this research was to investigate the effects of high altitude with supplementary oxygen during the last stage of incubation of broiler eggs laid at low altitude and incubated at low and high altitude. We analysed thyroid hormones and haematological variables. 2. The treatment groups were: low altitude (LA), high altitude with oxygen supplementation in the hatcher (HA-OX) and high altitude non-oxygen-supplemented (HA-NOX). 3. High altitude affected relative egg weight loss and early embryonic mortality. The hatchability of fertile eggs was lower at high than at low altitude. 4. Oxygen supplementation into the hatcher cabinet during the last stage of incubation decreased late embryonic mortality ratio (LEM(1)) and improved survival rates of embryos incubated at high altitude. 5. Eggs incubated at low altitude had a higher hatched chick weight and relative chick weight than those incubated at high altitude. Hatched chick weight and relative chick weight did not change with oxygen supplementation at high altitude. 6. High altitude caused an increase in plasma T(3) and T(4) concentrations as well as in the ratio of T(3):T(4) in embryos. High altitude newly hatched chicks showed a higher T(3):T(4) ratio than low altitude chicks; this ratio decreased with oxygen supplementation at high altitude. Altitude and oxygen supplementation did not affect the mean plasma T(4). 7. Newly-hatched chicks incubated at high altitude showed a higher plasma haematocrit (PCV) than the newly-hatched chicks from eggs incubated at low altitude. High altitude without supplementation increased haemoglobin (Hb), while oxygen supplementation returned the value to low altitude values.

The existence of an efficient but also safe hydraulic system seems to be essential for plant survival under water limiting conditions. To investigate any common pattern in this safetyefficiency trade-off, static (xylem anatomy and vulnerability to xylem cavitation) and dynamic (K(plant), soil to leaf hydraulic conductance and (leaf), leaf water potential) hydraulic properties of nine Mediterranean species belonging to four functional groups (semi-deciduous malacophyllous, sclerophylls, deciduous and herbaceous) were studied across two altitude sites. Static parameters did not show any pattern, but a strong exponential relationship between K(plant) and (leaf) was evident for all the studied species. Furthermore, each species is represented by a different part and/or range of the K(plantleaf) relationship, indicative of its adaptive mechanisms and capacity for survival under water stress conditions. The use of K(plantleaf) relationship as a tool for understanding the mechanisms of plant responses to water stress is discussed. Tree Physiology

The negative effects of drought on plant growth, development of natural plant communities and crop productivity are well established, but some of the responses remain poorly characterized, particularly the effect of long-term drought on photosynthetic capacity. We hypothesized that long-term drought results in a decline in leaf photosynthetic capacity, and not just a decrease in diffusive conductance. To test this hypothesis, we studied the effect of drought, slowly developed over 3.5 months, in leaves of eight potted mango (Mangifera indica L.) trees. We found that photosynthesis was not only limited by stomatal closure, but was also downregulated as a consequence of a strong decrease in photosynthetic capacity assessed by the measurements of maximal net photosynthesis (Amax) and the light-saturated rate of photosynthetic electron transport (Jmax). The rapid recovery of Amax and Jmax, after only 1 week of rewatering, the maintenance of a stable pool of leaf nitrogen throughout the trial, and the decrease in quantum efficiency of open centers of photosystem II, indicate that the photosynthetic machinery escaped photodamage in the drought-treated trees and was simply downregulated during drought. The hexose-to-sucrose ratio was higher in leaves from drought-treated trees than in control leaves, suggesting that photosynthetic capacity decreased as a consequence of sink limitation. Feed intake and satiation in fish are regulated by a number of factors, of which dissolved oxygen concentration (DO) is important. Since fish take up oxygen through the limited gill surface area, all processes that need energy, including food processing, depend on their maximum oxygen uptake capacity. Maximum oxygen uptake capacity relative to body weight in bigger fish is smaller than in smaller fish because the gill surface area is allometrically related to body weight. In this study, effects of DO concentration and body weight on maximum feed intake, growth and hematological parameters of Nile tilapia (Oreochromis niloticus) were investigated. Two weight classes of fish (21 g and 147 g) were used. For each class, six tanks were employed of which half were exposed to one of two DO levels (about 3.0 mg/L and 5.6 mg/L). Fish were fed to apparent satiation twice per day with a commercial diet. Striga hermonthica (Del.) Benth. is a parasitic weed on tropical cereals causing serious yield losses in Africa. The use of host crop varieties with improved resistance and tolerance against this parasite is a key component of an integrated control strategy. Breeding for tolerance is however seriously hampered by the absence of reliable and yet practical selection measures. The observation that the photosynthetic rate of tolerant genotypes is less sensitive to Striga infection was used as a starting point to search for suitable selection measures. In a greenhouse pot experiment the effect of Striga infection on the photosynthesis of four sorghum (Sorghum bicolor [L.] Moench) genotypes, differing in Striga tolerance level, was measured at three moments in time (26, 48 and 75 days after sowing). Genotypes were CK60-B, E36-1, Framida and Tiemarifing. Measurements involved CO2-assimilation (A) and three chlorophyll fluorescence characteristics (electron transport rate through photosystem II [ETR], photochemical [Pq] and non-photochemical quenching [NPq]). Striga infection negatively affected A, ETR and Pq. Based on A and Pq, genotypes with superior levels of tolerance (Tiemarifing) could be discriminated from genotypes with superior level of resistance (Framida). Both A and Pq showed high heritabilities and consequently clear and predictable differences between genotypes. Using discriminative ability, heritability and cost effectiveness as main criteria, photochemical quenching (Pq) was concluded to possess the highest potential to serve as indirect selection measure for host plant tolerance to Striga. Screening should preferably be conducted at relatively high Striga infestation levels, between Striga emergence and host plant flowering.

The responses of water relations, stomatal conductance (gs) and growth parameters of tomato (Lycopersicon esculentum Mill. cv. Royesta) plants to nitrogen fertilisation and drought were studied. The plants were subjected to a long-term, moderate and progressive water stress by adding 80 % of the water evapotranspirated by the plant the preceding day. Well-watered plants received 100 % of the water evapotranspirated. Two weeks before starting the drought period, the plants were fertilised with Hoagland’s solution with 14, 60 and 110 mM NO3 − (N14, N60 and N110, respectively). Plants of the N110 treatment had the highest leaf area. However, gs was higher for N60 plants and lower for N110 plants. At the end of the drought period, N60 plants showed the lowest values of water potential (Ψw) and osmotic potential (Ψs), and the highest values of pressure potential (Ψp). N60 plants showed the highest Ψs at maximum Ψp and the highest bulk modulus of elasticity.

The CO2 concentration near Rubisco and, therefore, the rate of CO2 assimilation, is influenced by both leaf anatomical factors and biochemical processes. Leaf anatomical structures act as physical barriers for CO2 transport. Biochemical processes add or remove CO2 along its diffusion pathway through mesophyll. We combined a model that quantifies the diffusive resistance for CO2 using anatomical properties, a model that partitions this resistance and an extended version of the Farquhar-von Caemmerer-Berry model. We parametrized the model by gas exchange, chlorophyll fluorescence and leaf anatomical measurements from three tomato cultivars. There was generally a good agreement between the predicted and measured light and CO2 response curves. We did a sensitivity analysis to assess how the rate of CO2 assimilation responds to changes in various leaf anatomical properties. Next, we conducted a similar analysis for assumed diffusive properties and curvature factors. Some variables (diffusion pathway length in stroma, diffusion coefficient of the stroma, curvature factors) substantially affected the predicted CO2 assimilation. Understanding the seasonality of photosynthesis in boreal evergreen trees and its control by the environment requires separation of the instantaneous and slow responses, as well as the dynamics of light reactions, carbon reactions, and respiration. We determined the seasonality of photosynthetic light response and respiration parameters of Scots pine (Pinus sylvestris L.) in the field in southern Finland and in controlled laboratory conditions. CO2 exchange and chlorophyll fluorescence were measured in the field using a continuously operated automated chamber setup and fluorescence monitoring systems. We also carried out monthly measurements of photosynthetic light, CO2 and temperature responses in standard conditions with a portable IRGA and fluorometer instrument. The field and response measurements indicated strong seasonal variability in the state of the photosynthetic machinery with a deep downregulation during winter. Despite the downregulation, the photosynthetic machinery retained a significant capacity during winter, which was not visible in the field measurements. Light-saturated photosynthesis (P sat) and the initial slope of the photosynthetic light response (α) obtained in standard conditions were up to 20% of their respective summertime values. Respiration also showed seasonal acclimation with peak values of respiration in standard temperature in spring and decline in autumn. Spring recovery of all photosynthetic parameters could be predicted with temperature history. On the other hand, the operating quantum yield of photosystem II and the initial slope of photosynthetic light response stayed almost at the summertime level until late autumn while at the same time P sat decreased following the prevailing temperature. Comparison of photosynthetic parameters with the environmental drivers suggests that light and minimum temperature are also decisive factors in the seasonal acclimation of photosynthesis in boreal evergreen trees.

In coastal environments plants have to cope with various water sources: rainwater, water table, seawater, and mixtures. These are usually characterized by different isotopic signatures ( (18)O/ (16)O and D/H ratios). Xylem water reflects the isotopic compositions of the water sources. Additionally, water-use efficiency (WUE) can be assessed with carbon isotope discrimination (Delta) analyses. Gas exchange, Delta of leaf dry matter, and isotopic composition (delta (18)O) of xylem water were measured from June to August 2001 in herbaceous perennials of mobile dunes (Ammophila littoralis, Elymus farctus) and sclerophyllous shrubs and climbers (Arbutus unedo, Pistacia lentiscus, Phillyrea angustifolia, Qercus ilex, Juniperus oxycedrus, Smilax aspera) of consolidated dunes. Assimilation rates were rather low and did not show clear seasonal patterns, possibly due to limited precipitation and generally low values of stomatal conductance. The lowest values were shown in S. aspera. Different physiological patterns were found, on the basis of delta (18)O and Delta analyses. Values of delta (18)O of xylem water of phanerophytes were remarkably constant and matched those of the water table, indicating dependence on a reliable water source; values of Delta were relatively high, indicating low intrinsic WUE, with the exception of J. oxycedrus. Surprisingly, very high delta (18)O values were found for the xylem water from S. aspera in August. This suggests retrodiffusion of leaf water to xylem sap in the stem or direct uptake of water by leaves or stems, owing to dew or fog occurrence. Low Delta values indicated high WUE in S. aspera. Contrasting strategies were shown by the species of mobile dunes: E. farctus relied on superficial water and exhibited low WUE, accordingly to its therophyte-like vegetative cycle; on the contrary, A. littoralis used deeper water sources, showing higher WUE in relation to its long-lasting vegetative habit.

The Finnish growing season is particularly short, with an intensive growth period, unfavourable rainfall distribution and frequently occurring fluctuations in climate that affect crop growth and yield formation. A three-year study was conducted in the field to determine the contribution of alterations in canopy structure, tillering and stem elongation among dwarf (D), semidwarf (SD) and tall (T) oat (Avena sativa L.) lines to yield formation. Yield components, leaf characteristics and straw traits were measured from six oat lines (D lines Pal and Grane, SD lines Hja 76416 and Salo, and T lines Veli and Jalostettu maatiainen) separately on the main shoot and tillers. Results indicated that long leaf area duration and high leaf area index were associated with increased grain yield probably due to more persistent and active assimilation. Also, higher number of leaves increased the grain yield. Higher peduncle, straw and node weights associating with increased grain yield may result from more abundant assimilate reserves; however, the longer the straw and peduncle, the lower the grain yield, which may result from increased lodging of SD and T lines. The traits contributing most to the grain yield varied greatly from year to year. It is concluded that no single dominant trait determined grain yield, since yield is a product of several different traits. Kuuden kauralinjan pääversoista ja sivuversoista mitattiin satokomponentit sekä lehdistörakennetta ja kortta kuvaavia ominaisuuksia. Saatujen tulosten perusteella kääpiöt versoivat enemmän, mutta niiden sadontuottokyky ei parantunut versontakyvyn myötä. Määritetyistä ominaisuuksista korkeaan satoon olivat yhteydessä pitkään yhteyttämiskykyisenä säilyvä kasvusto ja suuri lehtialaindeksi, mikä saattaa johtua pitempään jatkuvasta tehokkaasta yhteyttämisestä. Myös suuri lehtilukumäärä lisäsi satoa. Kääpiöillä oli yleensä hieman vähemmän lehtiä kuin pitkäkortisilla lajikkeilla ja erityisesti Pal-lajikkeella epätyydyttävä kasvustorakenne. Predicting the large-scale consequences of drought in contrasting environments requires that we understand how drought effects differ among species originating from those environments. A previous meta-analysis of published experiments suggested that the effects of drought on both stomatal and non-stomatal limitations to photosynthesis may vary consistently among species from different hydroclimates. Here, we explicitly tested this hypothesis with two short-term water stress experiments on congeneric mesic and xeric species. One experiment was run in Australia using Eucalyptus species and the second was run in Spain using Quercus species as well as two more mesic species. In each experiment, plants were grown under moist conditions in a glasshouse, then deprived of water, and gas exchange was monitored. The stomatal response was analysed with a recently developed stomatal model, whose single parameter g1 represents the slope of the relationship between stomatal conductance and photosynthesis. The non-stomatal response was partitioned into effects on mesophyll conductance (gm), the maximum Rubisco activity (Vcmax) and the maximum electron transport rate (Jmax). We found consistency among the drought responses of g1, gm, Vcmax and Jmax, suggesting that drought imposes limitations on Rubisco activity and RuBP regeneration capacity concurrently with declines in stomatal and mesophyll conductance. Within each experiment, the more xeric species showed relatively high g1 under moist conditions, low drought sensitivity of g1, gm, Vcmax and Jmax, and more negative values of the critical pre-dawn water potential at which Vcmax declines most steeply, compared with the more mesic species. These results indicate adaptive interspecific differences in drought responses that allow xeric tree species to continue transpiration and photosynthesis for longer during periods without rain.

Current understanding of physiological mechanisms governing stomatal behavior under water stress conditions is still incomplete and controversial. It has been proposed that coordination of stomatal kinetics with xylem vulnerability to cavitation [vulnerability curve (VC)] leads to different levels of isohydry/anisohydry in different plant species/cultivars. In this study, this hypothesis is tested in Vitis vinifera cultivars displaying contrasting stomatal behavior under drought stress. The cv Montepulciano (MP, near-isohydric) and Sangiovese (SG, anisohydric) were compared in terms of stomatal response to leaf and stem water potential, as possibly correlated to different petiole hydraulic conductivity (k(petiole)) and VC, as well as to leaf water relations parameters. MP leaves showed almost complete stomatal closure at higher leaf and stem water potentials than SG leaves. Moreover, MP petioles had higher maximum k(petiole) and were more vulnerable to cavitation than SG. Water potential at the turgor loss point was higher in MP than in SG. In SG, the percentage reduction of stomatal conductance (PLg(s)) under water stress was almost linearly correlated with corresponding percentage loss of k(petiole) (PLC), while in MP PLg(s) was less influenced by PLC. Our results suggest that V. vinifera near-isohydric and anisohydric genotypes differ in terms of xylem vulnerability to cavitation as well as in terms of k(petiole) and that the coordination of these traits leads to their different stomatal responses under water stress conditions.

The photosynthetic response was studied in two clones (Populus deltoides × maximowiczii Eridano and Populus × euramericana I-214), known for their differential response to ozone (O3) in terms of visible symptoms, when exposed to O3 (60 nl l−1 5 h day−1, 7 and 15 days). The photosynthetic ability was tested using gas exchange and chlorophyll fluorescence analysis. O3 caused a decrease in the CO2 assimilation rate at light saturation level in mature leaves of both clones. Alterations of Chl fluorescence parameters, in particular the Fv/Fm ratio and non-photochemical quenching were also observed. The effects were similar for both clones and it could not be concluded that differential effects on electron transport capacity were responsible for the observed reduction in photosynthesis. The reduction of photosynthetic rate in Eridano was due mainly to a reduced mesophyll activity, as evidenced by the increase in intercellular CO2 concentration and the minimal changes in stomatal conductance. In contrast, in I-214, stomatal effects were primarily responsible, although effects on the mesophyll cannot be excluded. Data obtained indicate that the effects observed at the mesophyll level may be attributed to indirect effects caused by membrane disorders.

International audience; The static chamber method (non-flow-through-non-steady-state chambers) is the most common method to measure fluxes of methane (CH4) from soils. Laboratory comparisons to quantify errors resulting from chamber design, operation and flux calculation methods are rare. We tested fifteen chambers against four flux levels (FL) ranging from 200 to 2300 μg CH4 m−2 h−1. The measurements were conducted on a calibration tank using three quartz sand types with soil porosities of 53% (dry fine sand, S1), 47% (dry coarse sand, S2), and 33% (wetted fine sand, S3). The chambers tested ranged from 0.06 to 1.8 m in height, and 0.02 to 0.195 m3 in volume, 7 of them were equipped with a fan, and 1 with a vent-tube. We applied linear and exponential flux calculation methods to the chamber data and compared these chamber fluxes to the reference fluxes from the calibration tank. The chambers underestimated the reference fluxes by on average 33% by the linear flux calculation method (Rlin), whereas the chamber fluxes calculated by the exponential flux calculation method (Rexp) did not significantly differ from the reference fluxes (p < 0.05). The flux under- or overestimations were chamber specific and independent of flux level. Increasing chamber height, area and volume significantly reduced the flux underestimation (p < 0.05). Also, the use of non-linear flux calculation method significantly improved the flux estimation; however, simultaneously the uncertainty in the fluxes was increased. We provide correction factors, which can be used to correct the under- or overestimation of the fluxes by the chambers in the experiment.

Although the mechanisms of nodule N2 fixation in legumes are now well documented, some uncertainty remains on the metabolic consequences of water deficit. In most cases, little consideration is given to other organs and, therefore, the coordinated changes in metabolism in leaves, roots, and nodules are not well known. Here, the effect of water restriction on exclusively N2-fixing alfalfa (Medicago sativa L.) plants was investigated, and proteomic, metabolomic, and physiological analyses were carried out. It is shown that the inhibition of nitrogenase activity caused by water restriction was accompanied by concerted alterations in metabolic pathways in nodules, leaves, and roots. The data suggest that nodule metabolism and metabolic exchange between plant organs nearly reached homeostasis in asparagine synthesis and partitioning, as well as the N demand from leaves. Typically, there was (i) a stimulation of the anaplerotic pathway to sustain the provision of C skeletons for amino acid (e.g. glutamate and proline) synthesis; (ii) re-allocation of glycolytic products to alanine and serine/glycine; and (iii) subtle changes in redox metabolites suggesting the implication of a slight oxidative stress. Furthermore, water restriction caused little change in both photosynthetic efficiency and respiratory cost of N2 fixation by nodules. Estimation of total leaf area (LAT) is important to express biochemical properties in plant ecology and remote sensing studies. A measurement of LAT is easy in broadleaf species, but it remains challenging in coniferous canopies. We proposed a new geometrical model to esti- mate Norway spruce LAT and compared its accuracy with other five published methods. Further, we assessed vari- ability of the total to projected leaf area conversion factor (CF) within a crown and examined its implications for remotely sensed estimates of leaf chlorophyll content (Cab). We measured morphological and biochemical properties of three most recent needle age classes in three vertical can- opy layers of a 30 and 100-year-old spruce stands. Newly introduced geometrical model and the parallelepiped model predicted spruce LAT with an error \5 % of the average needle LAT, whereas two models based on an elliptic approximation of a needle shape underestimated LAT by up to 60 %. The total to projected leaf area conversion factor varied from 2.5 for shaded to 3.9 for sun exposed needles and remained invariant with needle age class and forest stand age. Erroneous estimation of an average crown CF by 0.2 introduced an error of 2-3 l gc m -2 into the crown averaged Cab content. In our study, this error represents 10-15 % of observed crown averaged Cab range (33-53 l gc m -2 ). Our results demonstrate the importance of accurate LAT estimates for validation of remotely sensed estimates of Cab content in Norway spruce canopies.

The rate of photosynthesis (A) of plants exposed to water deficit is a function of stomatal (gs) and mesophyll (gm) conductance determining the availability of CO2 at the site of car- boxylation within the chloroplast. Mesophyll conductance often represents the greatest impediment to photosynthetic uptake of CO2, and a crucial determinant of the photosyn- thetic effects of drought. Abscisic acid (ABA) plays a fundamental role in signalling and co- ordination of plant responses to drought; however, the effect of ABA on gm is not well- defined. Rose, cherry, olive and poplar were exposed to exogenous ABA and their leaf gas exchange parameters recorded over a four hour period. Application with ABA induced reductions in values of A, gs and gm in all four species. Reduced gm occurred within one hour of ABA treatment in three of the four analysed species; indicating that the effect of ABA on gm occurs on a shorter timescale than previously considered. These declines in gm val- ues associated with ABA were not the result of physical changes in leaf properties due to altered turgor affecting movement of CO2, or caused by a reduction in the sub-stomatal con- centration of CO2 (Ci). Increased [ABA] likely induces biochemical changes in the properties of the interface between the sub-stomatal air-space and mesophyll layer through the actions of cooporins to regulate the transport of CO2. The results of this study provide further evi- dence that gm is highly responsive to fluctuations in the external environment, and stress signals such as ABA induce co-ordinated modifications of both gs and gm in the regulation of photosynthesis.

International audience; Leaf hydraulic conductance (Kleaf) plays a significant part in plant-water regulation. In walnut leaves, Kleaf is stimulated by irradiance and closely relates to the accumulation of JrPIP2s aquaporin transcripts, but it is independent of stomatal aperture. To provide an insight into the early molecular events occurred during light-induced Kleaf, a large-scale transcriptomic analysis consisting of the cDNA-amplified fragment length polymorphism (AFLP) was carried out on walnut leaves maintained under irradiance or in darkness. Of the total 12 000 transcript-derived fragments (TDFs) obtained using cDNA-AFLP with 128 primer pairs, 187 TDFs were selected after sequencing, and only 93 (49 %) that had been ascribed known functions through BLAST searching of the GenBank databases. Most of these TDFs correspond to genes whose protein products are involved in cellular regulation (57.9 %) and global metabolism (39.8 %). To validate cDNA-AFLP expression patterns, 30 TDFs were further analyzed using real-time quantitative polymerase chain reaction. Moreover, exposure of leaves to irradiance was accompanied by the modification of the Ca2+-signaling pathway, ubiquitin-proteasome pathway, vesicle trafficking process and expression of multiple transcription factors.

Estimating leaf stomatal conductance for CO 2 diffusion ( g sc ) is pivotal for further estimation of crop transpiration as well as energy and mass balances between air and plant in greenhouses. In this study, we tested two models, i.e. the Jarvis model and a new version of BWB–Leuning model (BWB–Leuning–Yin model), for estimating g sc in four greenhouse crops (cucumber, chrysanthemum, tulip and lilium), using data from extensive experiments conducted under a wide range of environmental conditions from 2003 to 2010. The models were parameterized from a subset of the experimental data. The remaining data sets for model validation were classified into two groups: group one was from experiments conducted during the similar seasons at the same sites as those for model parameterization, whereas group two was from experiments in different seasons and sites. When using data of group one, both models gave satisfactory estimations of g sc under both ample water supply and water stress conditions. When using data of group two, the BWB–Leuning–Yin model gave better estimations of g sc than the Jarvis model did. Functions or parameters of the Jarvis model, if applied to independent environmental conditions, have to be re-derived. Compared to its earlier versions, the BWB–Leuning–Yin model gave better estimated g sc under low light intensities and low CO 2 concentrations, suggesting that the new version is especially suitable for estimation of g sc for crops grown in low-investment greenhouses where levels of light and CO 2 are lower than ambient levels. In addition, analytical algorithms for the coupled FvCB model and diffusional conductance as adopted in the BWB–Leuning–Yin model make parameterization and simultaneous estimation of net photosynthetic rate and g sc an easy task, using readily obtained information.

Background: Anthropogenic disturbance of old-growth tropical forests increases the abundance of early successional tree species at the cost of late successional ones. Quantifying differences in terms of carbon allocation and the proportion of recently fixed carbon in soil CO2 efflux is crucial for addressing the carbon footprint of creeping degradation. Methodology: We compared the carbon allocation pattern of the late successional gymnosperm Podocarpus falcatus (Thunb.) Mirb. and the early successional (gap filling) angiosperm Croton macrostachyus Hochst. es Del. in an Ethiopian Afromontane forest by whole tree (CO2)-C-13 pulse labeling. Over a one-year period we monitored the temporal resolution of the label in the foliage, the phloem sap, the arbuscular mycorrhiza, and in soil-derived CO2. Further, we quantified the overall losses of assimilated C-13 with soil CO2 efflux. Principal Findings: C-13 in leaves of C. macrostachyus declined more rapidly with a larger size of a fast pool (64% vs. 50% of the assimilated carbon), having a shorter mean residence time (14 h vs. 55 h) as in leaves of P. falcatus. Phloem sap velocity was about 4 times higher for C. macrostachyus. Likewise, the label appeared earlier in the arbuscular mycorrhiza of C. macrostachyus and in the soil CO2 efflux as in case of P. falcatus (24 h vs. 72 h). Within one year soil CO2 efflux amounted to a loss of 32% of assimilated carbon for the gap filling tree and to 15% for the late successional one. Conclusions: Our results showed clear differences in carbon allocation patterns between tree species, although we caution that this experiment was unreplicated. A shift in tree species composition of tropical montane forests (e. g., by degradation) accelerates carbon allocation belowground and increases respiratory carbon losses by the autotrophic community. If ongoing disturbance keeps early successional species in dominance, the larger allocation to fast cycling compartments may deplete soil organic carbon in the long run. DFG/Gu 406/19-1

Plants of several species, if grown at high relative air humidity (RH >= 85%), develop stomata that fail to close fully in case of low leaf water potential. We studied the effect of a reciprocal change in RH, at different stages of leaf expansion of Rosa hybrida grown at moderate (60%) or high (95%) RH, on the stomatal closing ability. This was assessed by measuring the leaf transpiration rate in response to desiccation once the leaves had fully expanded. For leaves that started expanding at high RH but completed their expansion after transfer to moderate RH, the earlier this switch took place the better the stomatal functioning. Leaves initially expanding at moderate RH and transferred to high RH exhibited poor stomatal functioning, even when this transfer occurred very late during leaf expansion. Applying a daily abscisic acid (ABA) solution to the leaves of plants grown at continuous high RH was effective in inducing stomatal closure at low water potential, if done before full leaf expansion (FLE). After FLE, stomatal functioning was no longer affected either by the RH or ABA level. The results indicate that the degree of stomatal adaptation depends on both the timing and duration of exposure to high RH. It is concluded that stomatal functionality is strongly dependent on the humidity at which the leaf completed its expansion. The data also show that the effect of ambient RH and the alleviating role of ABA are restricted to the period of leaf expansion.

Due to anthropogenic influences, solar UV-B irradiance at the earth’s surface is increasing. To determine the effects of enhanced UV-B radiation on photosynthetic characteristics of Prunus dulcis, two-year-old seedlings of the species were submitted to four levels of UV-B stress, namely 0 (UV-Bc), 4.42 (UV-B1), 7.32 (UV-B2) and 9.36 (UV-B3) kJ m−2 d−1. Effects of UV-B stress on a range of chlorophyll (Chl) fluorescence parameters (FPs), Chl contents and photosynthetic gas-exchange parameters were investigated. UV-B stress promoted an increase in minimal fluorescence of dark-adapted state (F0) and F0/Fm, and a decrease in variable fluorescence (Fv, Fv/Fm, Fv/F0 and F0/Fm) due to its adverse effects on photosystem II (PSII) activity. No significant change was observed for maximal fluorescence of dark-adapted state (Fm). Enhanced UV-B radiation caused a significant inhibition of net photosynthetic rate (P N) at UV-B2 and UV-B3 levels and this was accompanied by a reduction in stomatal conductance (g s) and transpiration rate (E). The contents of Chl a, b, and total Chl content (a+b) were also significantly reduced at increased UV-B stress. In general, adverse UV-B effects became significant at the highest tested radiation dose 9.36 kJ m−2 d−1. The most sensitive indicators for UV-B stress were Fv/F0, Chl a content and P N. Significant P
