Atopic and normal individuals were immunized with the hemocyanin from the horseshoe crab (LPH) by subcutaneous or nasal routes. Subjects were divided into 3 groups: (1) atopic, (2) normal, and (3) ‘indeterminant’. The ‘indeterminant’ group was comprised of subjects who did not fit the criteria for designation as either atopic or normal. Antibody production in serum and saliva was characterized by hemagglutination, radioimmunodiffusion, radioimmunoprecipitation, and direct skin testing. Ten of 11 subjects immunized subcutaneously produced detectable antibody activity in one or more immunoglobulin classes. Only two subjects developed positive wheal-and-flare skin reactions. Salivary IgA antibody was not detected in any subjects in spite of high levels of serum IgA antibody. Seven of 11 subjects immunized by the nasal route produced serum antibody limited to a single class. Skin tests in all these subjects were negative. Also, salivary antibodies were not detected in these individuals.
