Many pathogenic bacteria biosynthesize and excrete small molecule metallophores, known as siderophores, that are used to extract ferric iron from host sources to satisfy nutritional need. Native siderophores are often structurally complex multidentate chelators that selectively form high-affinity octahedral ferric iron complexes with defined chirality recognizable by cognate protein receptors displayed on the bacterial cell surface. Simplified achiral analogues can serve as synthetically tractable siderophore mimics with potential utility as chemical probes and therapeutic agents to better understand and treat bacterial infections, respectively. Here, we demonstrate that synthetic spermidine-derived mixed ligand bis-catecholate monohydroxamate siderophores (compounds 1-3) are versatile structural and biomimetic analogues of two native siderophores, acinetobactin and fimsbactin, produced by Acinetobacter baumannii, a multidrug-resistant Gram-negative human pathogen. Objective: The experiment was designed to establish suitable management strategies through the different feeding and breeding approaches on fertility improvement in the experimental repeat breeding (RB) cows. Materials and Methods: 80 RB cows were selected for this experiment. Before grouping, all cows were deworming and then divided into four equal groups, namely GroupTF1 [traditional feed¬ing practice and natural insemination (NI)], GroupTF2 [traditional feeding practice and Artificial insemination (AI)], GroupSF1 [standard (STD) feeding practice and NI], and GroupSF2 (STD feed¬ing practice and AI). These allocated RB cows were fed by traditional and STD feeding methods for 90 days and then inseminated by AI and NI breeding systems. The dominant follicle (DF) diameter, hemato-biochemical elements, and estrogen (E2) hormone were estimated during the insemina¬tion of cows. Estimation of the pregnancy rate was carried out at days 4590 post-insemination in the cows. Results: The pregnancy rate was significantly (p < 0.05) higher in STD feeding practice with NI when compared to traditional feeding practice irrespective of breeding systems, and it was also significantly (p < 0.05) higher in NI than in AI breeding system, irrespective of feeding strategies. The results also showed that the diameter of DF, serum E2, total erythrocyte count, hemoglo¬bin, packed cell volume, total cholesterol, total protein, glucose, calcium, phosphorus, ferric iron, copper, zinc, and magnesium at the time of insemination were significantly (p < 0.01) elevated in the experimental RB cows with STD feeding practice. The diameter of DF and serum E2 were significant (p < 0.01) and positively correlated with all hemato-biochemical elements in the cows at the time of insemination. Conclusion: The results suggest that NI with STD feeding practice may increase fertility in RB cows by improving general health status. Finally, it could support the veterinarians and researchers to define the management strategies using feeding and breeding strategies to prevent repeat breed¬ing syndrome in dairy cows. Aim: Iron uptake and metabolism pathways are promising targets in vaccine development as an alternative strategy for antibiotics. Methods & methods: HemTR, a putative heme receptor of Acinetobacter baumannii, was expressed and its protectivity against A. baumannii was determined singly or in combination with the siderophore receptor, BauA, in mice. Results: High level of IgG was elicited. There was a delay in mice mortality with reduced bacterial loads in internal organs in the sublethal challenge. Protection was better in the HemTR-BauA group in both lethal and sublethal challenges. Passive transfer of anti-HemTR and anti-BauA partially protected mice against A. baumannii infection. Conclusion: HemTR in combination with other iron receptors could contribute to the development of protective vaccines against A. baumannii.

Aims: The present study aimed to evaluate Iron uptake protein-related genes in clinical and environmental Klebsiella pneumoniae isolates. Background: Klebsiella pneumoniae as an opportunistic pathogen cause infections in immunocompromised patients. Iron uptake systems play an important role in the pathogenesis of Klebsiella pneumonia. Objectives: This study was designed to investigate the prevalence of iron uptake coding genes among isolates of Klebsiella pneumonia. Materials and Methods: A total of 300 isolates of Klebsiella pneumonia including 150 clinical isolates and 150 environmental isolates were selected. Finally, the frequency of iroN, iucD, kfuA,hmuR, and ybt [yHPI] genes were detected by PCR method. Results: The frequency of kfuA, iucD, iroN, yHPI in clinical isolates were 33.3%, 16.7%, 24.7%, 15.3%, respectively and these genes among environmental isolates were 20.7%, 6%, 49.3% and 0.7%, respectively. Among the clinical isolates, the most frequency genes were kfuA gene [50 isolates] and after that iroN [37 isolates], iucD [25 isolates] and yHPI [23 isolates], the genes with the most frequency among environmental isolates were iroN gene [74 isolates] and following that kfuA [31 isolates], iucD [9 isolates] and yHPI [1 isolate]. No hmuR positive samples among all clinical or environmental isolates were found. Conclusion: The result of this study showed that because of the high frequency of ferric iron system coding gene kfu among clinical isolates, this system might play an important role in the survival of bacteria against its host.

Here, we report the draft genome sequences of two Geobacter sp. strains, AOG1 and AOG2, isolated from enrichment cultures using crystalline Fe(III) oxides as electron acceptors. Strains AOG1 and AOG2 possess numerous genes encoding multiheme c -type cytochromes and pilA-N genes encoding the pilin monomer of nanowires in their genomes.

Process understanding of microbial communities containing organohalide-respiring bacteria (OHRB) is important for effective bioremediation of chlorinated ethenes. The impact of iron and sulfate reduction on cis-1,2-dichloroethene (cDCE) and vinyl chloride (VC) dechlorination by a consortium containing the OHRB Dehalococcoides spp. was investigated using multiphase batch experiments. The OHRB consortium was found to contain endogenous iron- and sulfate-reducing bacteria (FeRB and SRB). A biogeochemical model was developed and used to quantify the mass transfer, aquatic geochemical, and microbial processes that occurred in the multiphase batch system. It was determined that the added SRB had the most significant impact on contaminant degradation. Addition of the SRB increased maximum specific substrate utilization rates, k(max), of cDCE and VC by 129% and 294%, respectively. The added FeRB had a slight stimulating effect on VC dechlorination when exogenous SRB were absent, but when cultured with the added SRB, FeRB moderated the SRB's stimulating effect. This study demonstrates that subsurface microbial community interactions are more complex than categorical, guild-based competition for resources such as electron donor.

A convergent total synthesis of the siderophore coelichelin is described. The synthetic route also provided access to acetyl coelichelin and other congeners of the parent siderophore. The synthetic products were evaluated for their ability to bind ferric iron and promote growth of a siderophore-deficient strain of the Gram-negative bacterium Pseudomonas aeruginosa under iron restriction conditions. The results of these studies indicate coelichelin and several derivatives serve as ferric iron delivery vehicles for P. aeruginosa.

Paludiculture, the cultivation of crops on wet or rewetted agricultural peatlands, sustainably integrates productive land use with the provision of multiple ecosystem services. Paludiculture crops thrive under waterlogged conditions that stimulate nitrogen (N) and phosphorus (P) removal from soil and water and convert serious drainage-induced carbon (C) losses to C sequestration. Nutrient uptake by paludicrops can prevent mobilisation after rewetting and provide opportunities for purification of nutrient-rich water. Uncertainty remains, however, if and to what extent N loading and a subsequent increase in biomass productivity affect nutrient cycling as well as emissions of the potent greenhouse gases methane (CH4) and nitrous oxide (N2O). In this study, we use mesocosms with rewetted peat to investigate the effect of different N sources in surface water on biomass production of Typha latifolia, a typical paludiculture crop, and the emissions of CH4 and N2O. Organic (Azolla filiculoides; urea) or mineral (KNO3 ; NH4NO) N was supplied either a single time (steady state) or repeatedly (pulse) to simulate a total surface water load of 150 kg N ha(-1) . We found that N stimulated aboveground and belowground biomass production and nutrient uptake by T. latifolia. These effects were absent in Azolla treatments. Resistance of microorganisms predominating in biohydrometallurgical processes including bacteria of the genus Sulfobaсillus and archaea of the genus Acidiplasma to ferric iron ions was studied. Capabilities of the strains for growth and ferrous iron oxidation in the media containing high concentrations of ferric iron ions (of 250 to 1000 mM) were evaluated. Ferric iron ions significantly inhibited oxidative activity and growth of the studied microorganisms. It was revealed that bacteria of the genus Sulfobacillus were not able to oxidize ferrous iron actively when ferric iron concentration exceeded 500 mM, whereas archaea of the genus Acidiplasma completely oxidized ferrous iron in the medium containing 1000 mM of Fe3+. Growth of the microorganisms was inhibited by relatively low concentrations of ferric iron. Microorganisms did not grow in the medium containing more than 750 mM of Fe3+ and cells of all studied strains lysed in presence of high concentrations of ferric iron. It was shown, that archaea of the genus Acidiplasma of the family Ferroplasmaceae were more resistant to high concentrations of ferric iron than bacteria of the genus Sulfobacillus. The results obtained are important for understanding of the regularities of the formation of microbial communities performing technological processes.

Streptomyces are a distinguished group of gram-positive bacteria mostly acknowledged for their immense contribution to life-saving drugs and lines of compounds with diverse bioactivities. To date, there remains limited studies on Streptomyces with biological activities residing in underexplored ecosystems such as the mangrove forests. For this purpose, the present work aimed at investigating the biological activity of Streptomyces sp. MUSC 11 collected from soil sample in mangrove forests, situated in the State of Pahang, Peninsular of Malaysia. The cultured strain resembled phenotypic and genotypic traits of genus Streptomyces. Investigations of the methanolic extract from Streptomyces sp. MUSC 11 revealed antioxidant activities in form of scavenging free radicals ABTS, DPPH, chelating iron and reducing ferric iron. Besides the antioxidant tests, antioxidant results corresponded well to the presence of phenolic content. In summary, Streptomyces derived from extreme and understudied ecosystem such as the mangrove forests are potential sources of biologically active and therapeutically useful compounds.

The kaolin (s.l.) in many tropical soils is distinctly different from most specimen and industrial kaolins. Crystal structure is highly disordered and crystal sizes are much smaller than for specimen and industrial kaolins. The crystal habit of soil kaolins is diverse ranging through euhedral hexagonal and anhedral platy, spheroidal and tubular. The specific surface of soil kaolins is higher than for most industrial kaolins. Consequently, cation and anion retention capacities are higher although there is no compelling evidence that the high structural disorder of soil kaolin is associated with a greater specific reactivity of the kaolin surface. A small amount of ferric iron substitutes for octahedral Al but there is generally little or no substitution of other cations. Soil kaolin commonly contains minor amounts of potassium but this is present in mica layers within, attached or associated with kaolin crystals so that potassium is not within the kaolin structure. Due to its considerable chemical reactivity soil kaolin helps provide an effective substrate to support agriculture and other land uses in the tropics.

More than 500 siderophores that bind ferric iron have been characterized and grouped by type based on their chemical structure. The chrome azurol S (CAS) assay is a universal colorimetric method that detects siderophores independent of their structure. In this assay, siderophores scavenge iron from an Fe-CAS-hexadecyltrimethylammonium bromide complex, and subsequent release of the CAS dye results in a color change from blue to orange. Solution-based experiments with CAS result in a quantitative measure of siderophore production, while an observable color change on CAS agar plates can be performed for qualitative detection of siderophores. Cross-feeding assays are another useful method to detect and characterize siderophores produced by bacteria. Under iron-limiting conditions, cross-feeding assays test the ability of an indicator strain to grow when supplied with a specific siderophore (from a test strain) to which it has a cognate receptor required for import into the cell. The cross-feeding assay can be tested with a variety of wild-type strains, siderophore biosynthesis mutants, and siderophore receptor mutants.

This review discusses the chemical mechanisms of ascorbate-dependent reduction and solubilization of ferritin's ferric iron core and subsequent release of ferrous iron. The process is accelerated by low concentrations of Fe(II) that increase ferritin's intrinsic ascorbate oxidase activity, hence increasing the rate of ascorbate radical formation. These increased rates of ascorbate oxidation provide reducing equivalents (electrons) to ferritin's core and speed the core reduction rates with subsequent solubilization and release of Fe(II). Ascorbate-dependent solubilization of ferritin's iron core has consequences relating to the interpretation of 59Fe uptake sourced from 59Fe-lebelled holotransferrin into ferritin. Ascorbate-dependent reduction of the ferritin core iron solubility increases the size of ferritin's iron exchangeable pool and hence the rate and amount of exchange uptake of 59Fe into ferritin, whilst simultaneously increasing net iron release rate from ferritin. This may rationalize the inconsistency that ascorbate apparently stabilizes 59Fe ferritin and retards lysosomal ferritinolysis and whole cell 59Fe release, whilst paradoxically increasing the rate of net iron release from ferritin. This capacity of ascorbate and iron to synergise ferritin iron release has pathological significance, as it lowers the concentration at which ascorbate activates ferritin's iron release to within the physiological range (50–250 μM). These effects have relevance to inflammatory pathology and to the pro-oxidant effects of ascorbate in cancer therapy and cell death by ferroptosis.

Soap Lake, located in Washington State, is a meromictic soda lake that was the subject of a prior National Science Foundation funded Microbial Observatory. Several organisms inhabiting this lake have been identified as producers of siderophores that are unique in structure. Two isolates found to be of the species Halomonas, SL01 and SL28, were found to produce suites of amphiphilic siderophores consisting of a peptidic head-group, which binds iron appended to fatty acid moieties of various lengths. The ability for siderophores to self-assemble into vesicles was determined for three suites of amphiphilic siderophores of unique structure (two from SL01 and one from SL28). These siderophores resemble the amphiphilic aquachelin siderophores produced by Halomonas aquamarina strain DS40M3, a marine bacterium. Vesicle self-assembly studies were performed by dynamic light scattering and epifluorescence microscopy. The addition of ferric iron (Fe3+) at different equivalents, where an equivalence of iron is defined as equal to the molarity of the siderophore, demonstrated vesicle formation. This was suggested by both dynamic light scattering and epifluorescence microscopy. Bacteria thriving under saline and alkaline conditions are capable of producing unique siderophores that self-assemble in micelles and vesicles due to ferric iron chelation.

Siderophores are small organic molecules produced by microorganisms under iron-limiting conditions which enhance the uptake of iron to the microorganisms. In environment, the ferric form of iron is insoluble and inaccessible at physiological pH (7.35-7.40). In the field of agriculture, different types of siderophores promote the growth of several plant species and increase their yield by enhancing the Fe uptake to plants. Siderophores acts as a potential biocontrol agent against harmful phyto-pathogens and holds the ability to substitute hazardous pesticides. Heavy-metal-contaminated samples can be detoxified by applying siderophores, which explicate its role in bioremediation. Siderophores can detect the iron content in different environments, exhibiting its role as a biosensor. In the medical field, siderophore uses the "Trojan horse strategy" to form complexes with antibiotics and helps in the selective delivery of antibiotics to the antibiotic-resistant bacteria. Certain iron overload diseases for example sickle cell anemia can be treated with the help of siderophores. Other medical applications of siderophores include antimalarial activity, removal of transuranic elements from the body, and anticancer activity. The aim of this review is to discuss the important roles and applications of siderophores in different sectors including ecology, agriculture, bioremediation, biosensor, and medicine.

Siderophores are small molecules produced by bacteria that bind ferric iron in the surrounding environment with extraordinary affinity. A new study provides evidence that a simple animal host, Caenorhabditis elegans, co-opts siderophores to promote its own iron acquisition and growth.

Melanomacrophage centres (MMCs), located in different organs of non-mammalian vertebrates, play a role in the destruction, detoxification or recycling of endogenous and exogenous materials. Cytochrome P450 monoxygenase 1A (CYP1A) is involved in xenobiotics biotransformation, and its liver expression is considered as a biomarker for detecting exposure to environmental pollutants. Atlantic bluefin tuna (ABFT), Thunnus thynnus L., liver samples were collected from: wild animals caught in the eastern Atlantic; juveniles reared in the central Adriatic; juveniles reared in the northern Adriatic; adults reared in the western Mediterranean. The samples were processed for basic histology, histochemistry and for CYP1A immunodetection. An unexpected high density of MMCs, containing ferric iron and lipofuscin–ceroids, was detected in the juveniles sampled in the northern Adriatic Sea. These individuals showed also a strong anti-CYP1A immunopositivity in hepatocytes and in the epithelium of bile ducts. This study supports the utility of MMCs as biomarkers of fish ‘health status’ and gives concern for a potential contaminant accumulation in ABFT.

WOS: 000411471800018, PubMed ID: 28872486, We compared the efficacy of ferrous sulfate (divalent) and ferric polymaltose (trivalent) compounds for the prophylaxis of iron-deficiency anemia (IDA). Study infants included exclusively breast milk-fed term infants. Subjects were divided randomly into 2 groups at 4 months of age and group 1 (n=56) received divalent and group 2 (n=56) received trivalent iron (Fe) preparation at a dose of 2 mg/kg/d for 5 months. At 9 months of age, after a 5-month prophylaxis, a significant increase was observed in hemoglobin (Hb), hematocrit, serum Fe levels, and transferrin saturation in both groups. However, group 1 had significantly higher Hb, hematocrit, mean corpuscular volume, Fe, and transferrin saturation than group 2 (11.7 +/- 0.6 g/dL, 34.6% +/- 1.7%, 76.2 +/- 2.9 fL, 55.5 +/- 1.8 mcg, 20.8 +/- 3.9 g/L, respectively in group 1 vs. 11.3 +/- 0.5 g/dL, 33.5% +/- 1.5%, 74.7 +/- 3.2 fL, 42.5 +/- 1.8 mcg, 14.1 +/- 7.5 g/L, respectively in group 2). No significant difference was found in ferritin values between the groups. Fe deficiency was found in 17 (30.3%) of the subjects in group 1, and 23 (41%) of subjects in group 2 whereas 5 (8.9%) subjects had IDA in group 1 and 12 (12%) in group 2 which were insignificant between groups. No significant difference was found with regard to side effects between 2 Fe preparations. Although divalent Fe therapy led to a higher Hb and serum Fe level, both ferrous and ferric Fe preparations were effective for prophylactic use in the prevention of Fe deficiency and IDA with comparable side effects.

Iron acquisition systems are critical for bacterial pathogenesis and thus have been proposed as attractive targets for iron-dependent pathogen control. Of these systems, high-affinity iron acquisition mediated by siderophore, a small iron chelator, is the most efficient iron-scavenging mechanism in gram-negative bacteria. Campylobacter does not produce any siderophores but has the ability to utilize exogenous siderophores. In particular, the enterobactin (Ent)-mediated iron scavenging is tightly linked to Campylobacter pathogenesis. To date, Ent, a triscatecholate with the highest known affinity for ferric iron, is a well-characterized siderophore used by Campylobacter for iron acquisition during in vivo infection. Here, we describe the key methods used to characterize Ent-mediated high affinity iron acquisition system in Campylobacter jejuni.

Summary Siderophores are chelators with extremely strong affinity for ferric iron and are best known for their capacity to feed microorganisms with this metal. Despite their preference for iron, they can also chelate numerous other metals with variable affinities. There is also increasing evidence that metals other than iron can activate the production of siderophores by bacteria, thereby implicating siderophores in the homeostasis of metals other than iron and especially heavy metal tolerance. This article considers this new concept that siderophores play a role in protecting bacteria against metal toxicity and discusses the possible contribution of these chelators to the transport of biological relevant metals in addition to iron.

An iron-reducing, moderately thermophilic, acidophilic actinobacterium, strain IC-180T, isolated from a solfataric field in Hakone, Japan, was subjected to polyphasic taxonomic analysis. Strain IC-180T was a motile, short rod-shaped, Gram-positive bacterium that was able to grow at temperatures of 35–58 °C (optimally at 50 °C) and at pH 2.0–4.5 (optimally at pH 3.0). The strain grew aerobically and heterotrophically. It also grew anaerobically or autotrophically by dissimilatory reduction of ferric iron. No oxidation of ferrous iron was observed. Major cellular fatty acids detected were iso-C16 : 0, anteiso-C17 : 0 and iso-C18 : 0; the major menaquinone was MK-9(H8). Phosphatidyl-N-methylethanolamine and an unknown ninhydrin-positive phosphoglycolipid were detected. The total DNA G+C content was 74.1 mol%. 16S rRNA gene sequence comparisons revealed that strain IC-180T was a member of the order Acidimicrobiales and clustered coherently with uncultured actinobacteria from a geothermal site and a bioreactor operated under moderately thermophilic conditions. This cluster could be distinguished from the two other clusters comprising the families of this order, Acidimicrobiaceae and Iamiaceae, respectively. Based on the properties of strain IC-180T determined in this polyphasic taxonomic study, this strain represents a novel species in a new genus in the order Acidimicrobiales, for which the name Aciditerrimonas ferrireducens gen. nov., sp. nov. is proposed; the type strain is IC-180T ( = JCM 15389T = DSM 45281T).

The composition was studied of the microbial association involved in tank biooxidation of the concentrate of a refractory pyrrhotite-containing pyrite-arsenopyrite gold-arsenic ore from the Olympiadinskoe deposit at 50°C. The two Sulfobacillus thermosulfidooxidans strains predominant in the association were phylogenetically different from the strains used as inocula. The isolates were found to differ significantly both from each other and from the strains that dominated in the processes of biooxidation of a similar concentrate by traditional tank technology at 39°C or at 39°C with treatment of the concentrate with ferric iron prior to biooxidation. These results indicate the strain and species diversity of sulfobacilli in microbial associations involved in biooxidation of the concentrates under different technological modes.

Acidithiobacillus thiooxidans A01 is widely used in bioleaching processes and commonly thrives in most metal-rich environments. However, interactions between different heavy metals remain obscure. In this study, we elaborated the effect of ferric iron on the growth and gene expression of At. thiooxidans A01 under the stress of nickel. The results showed that 600 mM Ni2+ completely inhibited the growth and sulfur metabolism of At. thiooxidans A01. However, trace amounts of Fe3+ (0.5 mM) facilitated the growth of At. thiooxidans A01 in the presence of 600 mM Ni2+. With the addition of 5 mM Fe3+, the maximum cell density reached 1.84 &times, 108 cell/mL, and pH value was 0.95. In addition, metal resistance-related and sulfur metabolism genes were significantly up regulated with extra ferric iron. Taking the whole process into account, the promoting effect of Fe3+ addition can be attributed to the following: (1) alleviation of the effects of Ni2+ toxicity and restoring the growth of At. thiooxidans A01, (2) a choice of multiple pathways to export nickel ion and producing precursor of chelators of heavy metals. This can suggest that microorganisms may widely exhibit metabolic activity in iron-rich environments with heavy metals. Our study will facilitate the technique development for the processing of ore bodies with highly challenging ore compositions.

Purpose Dissimilatory iron-reducing bacteria have been described by both culture-dependent and -independent methods in various environments, including freshwater, marine sediments, natural wetlands, and contaminated aquifers. However, little is known about iron-reducing microbial communities in paddy soils. The goal of this study was to characterize iron-reducing microbial communities in paddy soil. Moreover, the effect of dissolved and solid-phase iron (III) species on the iron-reducing microbial communities was also investigated by enrichment cultures.

Palaeozoic bryozoan colonies display a large variety of skeletal elements. Various types of rod-like styles are mainly built by laminated and non-laminated (hyaline) skeleton. Typical styles consist of hyaline cores and surrounding laminated sheaths. They usually protrude on the colony surface as spines. In a new bryozoan genus from the Middle Devonian of the Western Sahara described here, styles do not protrude on the colony surface and are embedded within the laminated skeleton. They consist of fibrous material. This new type of styles, unknown from other bryozoans, is here called a “cryptostyle”. A special characteristic of these structures is their intensive reddish-brown colouration. This colouration is apparently caused by the presence of ferric iron between the individual fibres. The function of cryptostyles was apparently weight reduction and stabilising the skeleton. The general morphology of the new bryozoan, Cryptostyloecia hexapuncta gen. et n. sp., implies its systematic position within ptilodictyine cryptostomes, specifically the Family Ptilodictyidae Zittel, 1880.

The kinetics of microbial ferrous-iron oxidation have been well studied as it is a critical sub-process in bioleaching of sulphide minerals. Exhaustive studies in continuous culture have been carried out recently, investigating the effects of conditions relevant to heap bioleaching on the microbial ferrous-iron oxidation by Leptospirillum ferriphilum [1-3]. It was postulated that ferric-iron, which is known to be inhibitory, also acts as a stress stimulus, promoting microbial growth at higher total iron concentration. This paper investigates this phenomenon further, by comparing tests run with pure ferrous-iron feeds against those where the feed is partially oxidised to ferric at comparable concentrations. The findings clearly suggest that, contrary to reactor theory, it is indeed ferrous iron concentration in the reactor feed that determines biomass concentration and that ferric iron concentration has little effect on microbial growth. Further mathematical analysis shows that the phenomenon can be explained on the basis of the Pirt equation and the particular reaction conditions employed in the test work.

Aeromonas salmonicida is an important fish pathogen whose virulence is beginning to be understood at the molecular level. The primary virulence property of this organism is a surface protein array (A-layer) of tetragonal symmetry. Biochemical and immunochemical analysis of this structure and its isolated A-protein monomer have shown that this virulence property is highly conserved. The surface protein array protects A. salmonicida from the fishes' defense mechanisms and so gives the organism a pathogenic advantage. This advantage is further enhanced by the ability of A. salmonicida to sequester ferric iron from the high affinity iron chelators lactoferrin and transferrin. This allows the pathogen to grow under the iron-limiting growth conditions normally found in fish tissues, and clearly is another important virulence attribute.

In this study, ferrous iron oxidation rates of a Leptospirillum ferriphilum dominated culture were determined over the temperature range of 2-50oC at pH below one. The results show that at pH 0.9 the culture oxidizes iron within the temperature range of 10°C to 45°C. Using the Arrhenius equation, an Ea value of 89.9 ± 6.75 kJ/mol was calculated. From the data fitted to Ratkowsky Equation, the optimum, minimum and maximum temperatures were 35 ± 1.5, 9.96 ± 1.72 and 42.93 ± 0.64 °C for this culture, respectively. The redox potential of the solution becomes more positive, which was the maximum (650-700 mV) at temperatures between 19-40 oC due to completing biological oxidation and increasing in ferric iron concentration.

Summary Protein is known to aid iron absorption from foods. The possible complexation occurring between iron and an in-vitro protein digest was studied by pH titration and by measuring the concentration of sulphydryl, amino and carboxyl groups in the presence and absence of ferrous and ferric iron. Complexation of iron was shown to occur with the protein digestion products and the carboxyl groups were identified as a site of complexation.

Mycoprotein is the biomass of the fungus Fusarium venenatum. The ability of mycoprotein to enhance production of dialyzable iron following in-vitro digestion or extraction was studied, with the aim of identifying the components responsible. Digested mycoprotein, extracted mycoprotein and digested chicken breast muscle all produced about ten times as much dialyzable iron as the control. All of the dialyzable iron from mycoprotein was ferric. Essentially all the dialyzable iron passed through a 1 kDa molecular weight cut-off membrane. Iron binding components were extracted from mycoprotein using weak acid but not water. Extraction of mycoprotein produced both dialyzable and non-dialyzable components, which bound ferric iron. Analysis of the digests and the dialyzable fraction of acid extraction showed that they contained siderophores, which contributed to the formation of dialyzable iron.

Cobblestone, glass beads and active carbon were selected as bacterial supports to study immobilization of Acidithiobacillus ferrooxidans in packed bed reactors. The production of ferric iron was then investigated in these immobilized reactors in batch and continuous operation modes. The results show that stable biofilm forms in cobblestone and active carbon supports, thus these two kinds of supports are suitable for immobilization of A. ferrooxidans. In batch culture, ferric iron productivity in reactor with cobblestone as supports is 0.61 g/(L·h), which is 1.49 times higher than that in suspended culture reactor. In continuous operation mode, the maximum ferric iron productivity in reactor with cobblestone as supports is 1.54 g/(L·h), which is 3.76 times higher than that in suspended culture reactor. The maximum ferric iron productivity in reactor with active carbon as supports is 1.89 g/(L·h), which is 4.61 times higher than that in suspended culture reactor. In addition to bacteria, the results of X-ray diffraction and scanning electronic microscope analysis show that there is a lot of exopolysaccharide, jarosite and ammoniojarosite in biofilm, which plays important role in the formation of biofilm.

Natural resistance to infection, which does not depend on antibiotics, is a powerful protective mechanism common to all mankind that has been responsible for the survival of our species during countless millennia in the past. The normal functioning of this complex system of phagocytic cells and tissue fluids is entirely dependent on an extremely low level of free ionic iron (10(-18) M) in tissue fluids. This low-iron environment is maintained by the unsaturated iron-binding proteins transferrin and lactoferrin, which depend on well-oxygenated tissues, where a relatively high oxidation-reduction potential (Eh) and pH are essential for the binding of ferric iron. Freely available iron is derived from iron overload, free haem compounds, or hypoxia in injured tissue leading to a fall in Eh and pH. This can severely damage or abolish normal bactericidal mechanisms in tissue fluids leading to overwhelming growth of bacteria or fungi. The challenge for clinical medicine is to reduce or eliminate the presence of freely available iron in clinical disease. In injured or hypoxic tissue, treatment with hyperbaric oxygen might prove very useful by increasing tissue oxygenation and restoring normal bactericidal mechanisms in tissue fluids, which would be of huge benefit to the patient.

Thermophilic microorganisms can reduce Fe(III), Mn(IV), Cr(VI), U(VI), Tc(VII), Co(III), Mo(VI), Au(I, III), and Hg(II). Ferric iron and Mn(IV) can be used as electron acceptors during growth; the physiological role of the reduction of the other metals is unclear. The process of microbial dissimilatory reduction of Fe(III) is the most thoroughly studied. Iron-reducing prokaryotes have been found in virtually all of the recognized types of terrestrial ecosystems, from hot continental springs to geothermally heated subsurface sediments. Thermophilic iron reducers do not belong to a phylogenetically homogenous group and include representatives of many bacterial and archaeal taxa. Iron reducing thermophiles can couple Fe(III) reduction with oxidation of a wide spectrum of organic and inorganic compounds. In the thermophilic microbial community, they can fulfil both degradative and productive functions. Thermophilic prokaryotes probably carried out global reduction of metals on Earth in ancient times, and, at the same time, they are promising candidates for use in modern biotechnological processes.

Predominance and mineral stability diagrams are most useful if they are based on a full chemical speciation. The diagrams can then include a wide range of reactions of importance in environmental chemistry including adsorption and ion exchange reactions. They can also take into account the possible interactions between a large number of components and are able to model changes at a fixed total concentration of each component rather than under the commonly used but less realistic assumption of a fixed activity. A "hunt-and-track" algorithm is described that enables predominance and mineral stability diagrams to be constructed using any general-purpose speciation program. Examples are given using PHREEQC and Orchestra.

This review focuses on the properties of ferric iron surface receptors of Gram negative bacteria. We discuss the different strategies to acquire iron, and the fundamental role of these receptors in pathogenicity. The structure of some of these receptors, iron transport and regulation mechanisms are presented here.

The finely ground seeds of Canavalia ensiformis (jack bean) and Brachystegen eurycoma (achi seed) were used in the treatment of sewage sludge. They were found to improve sludge filterability and compare favourably with the traditional, ferric chloride conditioner. The optimum dosage for each sample can be determined by drawing a tangent to the corresponding curve of each sample at the point where there is a sharp break between the straight and curvilinear portion. In this work, the optimum values for C. ensiformis and B. eurycoma are 19.50% (0.39 g/cm3) and 21.5% (0.4 g/cm3) respectively. For ferric chloride, the optimum value is 25% (0.5 g/cm3). These results show that ferric chloride is still a better conditioner than both the other conditioners. However, during sludge filtration, cake and filtrate concentration increased with increasing dosages of both C. ensiformis and B. eurycoma samples. This indicates that they would be useful as conditioners if the primary product of filtration is the procurement of low filtrate concentration or high-quality cake.

The aim of this work was to investigate the production of extracellular polymeric substances (EPS) by Aeromonas hydrophila grown under anaerobic conditions. EPS composition was studied for planktonic cells, cells attached to carbon fibre supports using a soluble ferric iron source and cells grown with a solid ferric iron mineral (gossan). Conventional spectrophotometric methods, Fourier transform infrared (FTIR) and confocal laser scanning microscopy (CLSM) were used to determine the main components in the biofilm extracted from the cultures. The key EPS components were proteins, indicating their importance for electron transfer reactions. Carbohydrates were observed mostly on the mineral and contained terminal mannosyl and/or terminal glucose, fucose and N-acetylgalactosamine residues.

The ability of Staphylococcus epidermidis strains to grow in the presence of human transferrin and varying amounts of ferric iron was studied. At initial bacterial densities up to 10(4) cfu ml(-1), none of the three strains grew when transferrin iron saturation was below the full saturation point, whereas the bacteria grew consistently when transferrin was fully iron-saturated and there was non-transferrin-bound iron in the medium. Precultivation of the bacteria under iron-restricted conditions to induce siderophore production did not abolish the growth dependence on non-transferrin-bound iron. At initial bacterial densities of 10(6) cfu ml(-1), the bacteria proliferated consistently also in the presence of partially saturated transferrin. The results indicate that at low bacterial densities, S. epidermidis cannot utilise transferrin-bound iron for growth and that its proliferation is dependent on non-transferrin-bound iron.

The siderophore production of various isolates of Phialocephala fortinii was assessed quantitatively as well as qualitatively in batch assays under pure culture conditions at different pH values and iron(III) concentrations. We found a distinct effect of both of these parameters on siderophore synthesis and as well as on fungal growth. In comparative analyses of two of the isolates, maximum siderophore production was found at a pH in the range of pH 4.0 to 4.5 while, under the experimental conditions employed, the optimal concentration of ferric iron was determined to be between 20–40 μg iron (III) l−1 (0.36–0.72 μM, respectively). HPLC analysis of the culture filtrate of most of the isolates of P. fortinii revealed the excretion of ferricrocin as main hydroxamate siderophore, followed by ferrirubin and ferrichrome C. The pattern of release of these three substances proved to be dependent on pH and iron(III) concentration of the culture medium, and to be specific for each isolate under investigation.

A new complex, [Fe(acac)L2], bearing inidazolidine nitroxyl-free radical ligand (L-) was recently synthesised for biological studies. It proved to be biologically active against African sleeping sickness, plasmodium falciparum (malaria), leishmaniasis and chaga disease causative agents. Three ESR well resolved peaks indicated the presence of a free (unpaired) and chemically active electron in the complex. The structural complex ferric iron was found at the centre of two electric gradient whose the biggest is suggested to be initiated by the unpaired charge. No distinction between different cis isomers could be made.

Synthetic andradite (Ca3Fe2Si3O12) has been compressed to loading pressures >21 GPa and heated to ∼1000°C by a YAG laser in a Diamond Anvil Cell (DAC). After quenching to room temperature, X-ray diffraction of the sample, still held at 21 GPa, showed that andradite had transformed to a cubic perovskite type polymorph with a = 3.460(4) Å. Upon decompression the perovskite phase transformed into an amorphous phase. The density of the perovskite polymorph (Ca3Fe2Si3O12) is ∼13.6% greater than that of isochemical andradite at 21 GPa. Ferric iron replaces Ca2+ and Si4+ in the perovskite structure (Fe3+ + Fe3+ = Si4+ + Ca2+), giving a formula unit: (Ca,Fe3+)(Si,Fe3+)O3. The new Fe3+-rich Ca-perovskite may provide new insight into the controls on the electrical conductivity of the lower mantle.

DNA is an excellent complexing agent for ferric iron. However it does not provide an adequate matrix for 57Fe nuclear resonance absorption at room temperature. Cellular ferritin with large particle size displays a broad resonance at ambient temperature. Therefore only ferritin could provide, if at all, the essential prerequisite for the MIRAGE effect.

Although iron therapy is very common, few studies have assessed iron absorption or iron kinetics in general, particularly with Fe(3+). This was the primary objective of this study, with assessment of tolerance as a secondary objective. Eight nonsmoking iron-deficient females without other associated pathologies, with an average age of 21.62 +/- 1.69 years, were studied. The diagnosis of iron deficiency was established by determination of sideraemia (28.7 +/- 13.5 microg/dl), iron binding capacity (380.5 +/- 70.2 microg/dl) and ferritin (5 +/- 1.4 microg/L) on the prestudy visit. The women remained in the Phase I Unit during days 0, 1, 2 and 3 of the study. A serum iron concentration curve was obtained daily from blood samples drawn at 0, 1, 2, 4, 6, 8 and 12 hours postdose. Therapy with ferric natural protein (ferrimannitol ovoalbumin) 40mg twice daily was started on day 1 of the study and continued for 30 days. The following parameters were evaluated: area under the curve (AUC(0-12h)), peak iron concentration (C(max)) and time to reach peak concentration (t(max)). Serum iron concentration-time curves were practically flat on day 0, but showed an increase following ferric therapy (Friedman, p0.05), confirming the usefulness of the postabsorption test and good absorption of ferric iron (Fe(3+)). The increase in AUC (314.65 +/- 67.9 to 1174.44 +/- 1071.8 microg/dl.h) and C(max) (49 +/- 24.4 to 146 +/- 101.9 microg/dl) from day 1 and the correlation between both parameters (r(2)0.85, p/= 0.008) supported the use of either parameter. The t(max) ranged from 4 to 6 hours. Ferric therapy was well tolerated by all patients. This study shows that ferric iron contained in this preparation is well absorbed and provides a satisfactory method for evaluation of iron absorption.

An optimal treatment has yet to be determined. There are relatively little publications on the responsiveness to IV iron therapy in children with IRIDA. Patients and Method: This study analyzed responses to IV iron sucrose therapy given to 11 children, ranging in age from 2 to 13 years (mean 4.8 years), with iron-deficiency anemia who were unresponsive to oral iron therapy. Results: The hemoglobin and ferritin values (mean) of the 11 children with IRIDA were 7.7 g/dL and 4.8 ng/mL at diagnosis. Both hemoglobin and ferritin levels increased to 9.5 g/dL, and 24 ng/mL, respectively, at 6 weeks after the first therapy. Although the level of hemoglobin was steady at 6 months after the first, and 6 weeks after the second therapy, the ferritin levels continued to increase up to 30 ng/mL and 47 ng/mL at 6 months after the first and 6 weeks after the second therapy, respectively. Conclusion: We recommend that IRIDA should be considered in patients presenting with iron-deficiency anemia of unknown cause that is unresponsive to oral iron therapy. Our results suggest that IV iron therapy should be administered only once in cases of IRIDA. Continued administration of IV iron would be of no benefit to increase hemoglobin levels. On the contrary, ferritin levels may continue to increase resulting in untoward effects of hyperferritinemia.
