Metazoans normally possess a single lineage of mitochondria inherited from the mother (♀-type mitochondria) while paternal mitochondria are absent or eliminated in fertilized eggs. In doubly uniparental inheritance (DUI), which is specific to the bivalve clade including the ocean quahog, Arctica islandica, ♂-type mitochondria are retained in male gonads and, in a few species, small proportions of ♂-type mitochondria co-exist with ♀-type in somatic tissues. To the best of our knowledge, we report, for the first time in metazoan, the natural occurrence of male and female individuals with exclusively ♂-type mitochondria in somatic tissues of the bivalve A. islandica. Mitochondrial genomes differ by ~5.5% at DNA sequence level. Exclusive presence of ♂-type mitochondria affects mitochondrial complexes partially encoded by mitochondrial genes and leads to a sharp drop in respiratory capacity. Through a combination of whole mitochondrial genome sequencing and molecular assays (gene presence and expression), we demonstrate that 1) 11% of individuals of an Icelandic population appear homoplasmic for ♂-type mitochondria in somatic tissues, 2) ♂-type mitochondrial genes are transcribed and 3) individuals with ♂-type mitochondria in somatic cells lose 30% of their wild-type respiratory capacity. This mitochondrial pattern in A. islandica is a special case of DUI, highlighted in individuals from both sexes with functional consequences at cellular and conceivably whole animal level., Dégletagne et al. report on an unusual case of doubly uniparental inheritance in the ocean quahog, Arctica isladica. Some individuals in an Icelandic population present homoplasmic presence of male-type mitochondrial genes in somatic tissues, which appears to confer some changes in respiratory capacity in some environmental conditions.

The limpet Nacella concinna is a dominant macroinvertebrate along the coastal Antarctic Peninsula with two ecotypes inhabiting intertidal and subtidal areas, respectively. The ecological aim of the study was to understand whether higher stress competence and migratory energy expenses in intertidal Antarctic limpets shorten their lifetime and limit the shell growth rate compared to their sublittoral conspecific. We evaluated shell morphometry, age and internal shell growth bands in a large number of intertidal and subtidal N. concinna shells in Potter Cove, South Shetland Islands. Comparisons of their morphometrics showed that intertidal limpets are relatively shorter and less wide, and have higher shell mass, i.e. at common shell height, intertidal shells are relatively thicker and heavier than those of subtidal specimens. Internal shell growth bands showed alternating wide opaque (faster growth in summer) and thin translucent bands (slow growth in winter). The maximum age read was close to 20 years for both groups. Comparisons of von Bertalanffy growth curves showed for shell length and shell width lower growth rate k in intertidal animals than in subtidal ones associated to a great variability, with no differences in other growth parameters. However, when shell height vs. age is considered, no differences were observed for any growth parameter. Curtailed variability of growth rates in the intertidal population reflects either a limitation of the food reserves or feeding time, or an energy gap for shell growth due to the costs for migratory movements and stress defense.

Bivalve sclerochronological records with annually resolved growth bands are applicable proxies in reconstructing features of the hydro-climate system. Here we evaluate the relationship between growth indices of A. islandica, previously collected at approximately 82 m depth in the North Atlantic, and seasonal subsurface temperature at various depths for the 1900–2005 period. Correlations with sea surface temperature at the collection site are not significant during winter and weak for the remaining seasons. The strongest in-phase correlations persist for summer and autumn below 56 m water depth, whereas weaker correlations are lagged by one or two years. We also observe similarities with distant water bodies in the North Atlantic sector, and a corresponding large-scale oceanographic pattern that increases significantly with water depth along the trajectory of the North Atlantic Current. We suggest that by investigating the relationship with the temperature signal at various depths locally and at large-scale increases the reliability and application of bivalve shells as marine archives.

Benthic organisms of the Southern Ocean are particularly vulnerable to ocean acidification (OA), as they inhabit cold waters where calcite-aragonite saturation states are naturally low. OA most strongly affects animals with calcium carbonate skeletons or shells, such as corals and mollusks. We exposed the abundant cold-water coral Malacobelemnon daytoni from an Antarctic fjord to low pH seawater (LpH) (7.68 ± 0.17) to test its physiological responses to OA, at the level of gene expression (RT-PCR) and enzyme activity. Corals were exposed in short- (3 days) and long-term (54 days) experiments to two pCO2 conditions (ambient and elevated pCO2 equaling RCP 8.5, IPCC 2019, approximately 372.53 and 956.78 μatm, respectively). Of the eleven genes studied through RT-PCR, six were significantly upregulated compared with control in the short-term in the LpH condition, including the antioxidant enzyme superoxide dismutase (SOD), Heat Shock Protein 70 (HSP70), Toll-like receptor (TLR), galaxin and ferritin. After long-term exposure to low pH conditions, RT-PCR analysis showed seven genes were upregulated. These include the mannose-binding C-Lectin and HSP90. Also, the expression of TLR and galaxin, among others, continued to be upregulated after long-term exposure to LpH. Expression of carbonic anhydrase (CA), a key enzyme involved in calcification, was also significantly upregulated after long-term exposure. Our results indicated that, after two months, M. daytoni is not acclimatized to this experimental LpH condition. Gene expression profiles revealed molecular impacts that were not evident at the enzyme activity level. Consequently, understanding the molecular mechanisms behind the physiological processes in the response of a coral to LpH is critical to understanding the ability of polar species to cope with future environmental changes. Approaches integrating molecular tools into Antarctic ecological and/or conservation research make an essential contribution given the current ongoing OA processes.

The mitochondrial oxidative stress theory of aging posits that membrane susceptibility to peroxidation and the organization of the electron transport system (ETS) linked with reactive oxygen species (ROS) generation are two main drivers of lifespan. While a clear correlation has been established from species comparative studies, the significance of these characteristics as potential modulators of lifespan divergences among populations of individual species is still to be tested. The bivalve Arctica islandica, the longest-lived non-colonial animal with a record lifespan of 507 years, possesses a lower mitochondrial peroxidation index (PI) and reduced H2O2 efflux linked to complexes I and III activities than related species. Taking advantage of the wide variation in maximum reported longevities (MRL) among 6 European populations (36–507 years), we examined whether these two mitochondrial properties could explain differences in longevity. We report no relationship between membrane PI and MRL in populations of A. islandica, as well as a lack of intraspecific relationship between ETS complex activities and MRL. Individuals from brackish sites characterized by wide temperature and salinity windows had, however, markedly lower ETS enzyme activities relative to citrate synthase activity. Our results highlight environment-dependent remodeling of mitochondrial phenotypes.

The west Antarctic Peninsula (WAP) region has undergone significant changes in temperature and seasonal ice dynamics since the mid-twentieth century, with strong impacts on the regional ecosystem, ocean chemistry and hydrographic properties. Changes to these long-term trends of warming and sea ice decline have been observed in the 21st century, but their consequences for ocean physics, chemistry and the ecology of the high-productivity shelf ecosystem are yet to be fully established. The WAP shelf is important for regional krill stocks and higher trophic levels, whilst the degree of variability and change in the physical environment and documented biological and biogeochemical responses make this a model system for how climate and sea ice changes might restructure high-latitude ecosystems. Although this region is arguably the best-measured and best-understood shelf region around Antarctica, significant gaps remain in spatial and temporal data capable of resolving the atmosphere-ice-ocean-ecosystem feedbacks that control the dynamics and evolution of this complex polar system. Here we summarise the current state of knowledge regarding the key mechanisms and interactions regulating the physical, biogeochemical and biological processes at work, the ways in which the shelf environment is changing, and the ecosystem response to the changes underway. We outline the overarching cross-disciplinary priorities for future research, as well as the most important discipline-specific objectives. Underpinning these priorities and objectives is the need to better define the causes, magnitude and timescales of variability and change at all levels of the system. A combination of traditional and innovative approaches will be critical to addressing these priorities and developing a co-ordinated observing system for the WAP shelf, which is required to detect and elucidate change into the future. Multiple toxic and bioactive compounds produced by Alexandrium spp. cause adverse effects on bivalves, but these effects are frequently difficult to attribute to a single compound class. To disentangle the effect of neurotoxic vs lytic secondary metabolites, we exposed blue mussels to either a paralytic shellfish toxin (PST) producing Alexandrium spp. strain, or to an exclusively lytic compound (LC) producing strain, or a strain containing both compound classes, to evaluate the time dependent effects after 3 and 7 days of feeding. Tested parameters comprised signs of paralysis, feeding activity, and immune cell integrity (hemocyte numbers and viability; lysosomal membrane destabilization) and function (ROS production). Both compound classes caused paralysis and immune impairment. The only effect attributable exclusively to PST was increased phagocytic activity after 3 days and impaired feeding activity after 7 days, which curtailed toxin accumulation in digestive glands. Lysosomal membrane destabilization were more closely, but not exclusively, matched with LC exposure. Effects on circulating hemocyte integrity and immune related functions were mostly transient or remained stable within 7 days; except for increased lysosomal labialization and decreased extracellular ROS production when mussels were exposed to the toxin combination. M. edulis displays adaptive fitness traits to survive and maintain immune capacity upon prolonged exposure to environmentally relevant concentrations of PST and/or LC producing Alexandrium strains. The gills and mantle are the first tissues exposed to altered oxidative conditions during dissolved Fe uptake. The aim of this work was to characterize Fe affected oxidative metabolism on the respiratory tissues of Mya arenaria. After 9 days of exposure to elevated Fe levels (500 μM of Fe as Fe-EDTA), a significant increase in the total Fe and labile Fe pool content was found in the gills. Dichlorofluorescein diacetate oxidation rate was higher after 17 days of exposure as compared to controls. Thiobarbituric acid reactive substances content increased 1.9- and 3.7-fold over controls on day 9 and 17, respectively. Both, catalase activity and nitrate and nitrite content decreased significantly on days 9 and 17 compared with controls. Similar effects were observed in mantle, but catalase activity was not affected. Background: Scavenger guilds are composed of a variety of species, co-existing in the same habitat and sharing the same niche in the food web. Niche partitioning among them can manifest in different feeding strategies, e.g. during carcass feeding. In the bentho-pelagic realm of the Southern Ocean, scavenging amphipods (Lysianassoidea) are ubiquitous and occupy a central role in decomposition processes. Here we address the question whether scavenging lysianassoid amphipods employ different feeding strategies during carcass feeding, and whether synergistic feeding activities may influence carcass decomposition. To this end, we compared the relatively large species Waldeckia obesa with the small species Cheirimedon femoratus, Hippomedon kergueleni, and Orchomenella rotundifrons during fish carcass feeding (Notothenia spp.). The experimental approach combined ex situ feeding experiments, behavioural observations, and scanning electron microscopic analyses of mandibles. Furthermore, we aimed to detect ecological drivers for distribution patterns of scavenging amphipods in the Antarctic coastal ecosystems of Potter Cove. In Potter Cove, the climate-driven rapid retreat of the Fourcade Glacier is causing various environmental changes including the provision of new marine habitats to colonise. While in the newly ice-free areas fish are rare, macroalgae have already colonised hard substrates. Assuming that a temporal dietary switch may increase the colonisation success of the most abundant lysianassoids C. femoratus and H. kergueleni, we aimed to determine their consumption rates (g food x g amphipods−1 x day−1) and preferences of macroalgae and fish.Results: We detected two functional groups with different feeding strategies among scavenging amphipods during carcass feeding: carcass 'opener' and 'squeezer'. Synergistic effects between these groups were not statistically verified under the conditions tested. C. femoratus switched its diet when fish was not available by consuming macroalgae (about 0.2 day−1) but preferred fish by feeding up to 80% of its own mass daily. Contrary, H. kergueleni rejected macroalgae entirely and consumed fish with a maximal rate of 0.8 day−1.Conclusion: This study reveals functional groups in scavenging shallow-water amphipods and provides new information on coastal intraguild niche partitioning. We conclude that the dietary flexibility of C. femoratus is a potential ecological driver and central to its success in the colonisation of newly available ice-free Antarctic coastal habitats.

A diesel spill occurring at Carlini Station (King George Island (Isla 25 de Mayo), South Shetland Islands) in 2009 started the study of the fate of the hydrocarbons and their effect on the bacterial communities of the Potter Cove ecosystem. Soils and sediments were sampled across the 200-meter long diesel plume towards Potter Cove four and 15 months after the spill. The sampling revealed a second fuel leakage from an underground pipeline at the spill site. The hydrocarbon fraction spilt over frozen and snow-covered ground reached the sea and dispersed with the currents. Contrary, diesel that infiltrated unfrozen soil emained detectable for years, and was seeping with ground water towards coastal marine sediments. Structural changes of the bacterial communities as well as hydrocarbon, carbon and nitrogen contents were investigated in sediments in front of the station, two affected terrestrial sites, and a terrestrial non-contaminated reference site. Bacterial communities (16S rRNA gene clone libraries) changed over time in contaminated soils and sediments. At the underground seepage site of highest contamination (5812 to 366 μgg−1dw hydrocarbons fromsurface to 90-cm depth), communities were dominated by Actinobacteria (18%) and a betaproteobacteriumclosely related to Polaromonas naphthalenivorans (40%). At one of the spill sites, affected exclusively at the surface, contamination disappeared within one year. The same bacterial groups were enriched at both contaminated sites. This response at community level suggests that the cold-adapted indigenous microbiota in soils of the West Antarctic Peninsula have a high potential for bioremediation and can support soil cleaning actions in the ecosystem. Intensive monitoring of pollution and site assessment after episodic fuel spills is required for decision-making towards remediation strategies. Fil: Vázquez, Susana Claudia. Hypoxia in freshwater ecosystems is spreading as a consequence of global change, including pollution and eutrophication. In the Patagonian Andes, a decline in precipitation causes reduced lake water volumes and stagnant conditions that limit oxygen transport and exacerbate hypoxia below the upper mixed layer. We analyzed the molecular and biochemical response of the North Patagonian bivalve Diplodon chilensis after 10 days of experimental anoxia (< 0.2 mg O2/L), hypoxia (2 mg O2/L), and normoxia (9 mg O2/L). Specifically, we investigated the expression of an alternative oxidase (AOX) pathway assumed to shortcut the regular mitochondrial electron transport system (ETS) during metabolic rate depression (MRD) in hypoxia-tolerant invertebrates. Whereas, the AOX system was strongly upregulated during anoxia in gills, ETS activities and energy mobilization decreased [less transcription of glycogen phosphorylase (GlyP) and succinate dehydrogenase (SDH) in gills and mantle]. Accumulation of succinate and induction of malate dehydrogenase (MDH) activity could indicate activation of anaerobic mitochondrial pathways to support anoxic survival in D. chilensis. Oxidative stress [protein carbonylation, glutathione peroxidase (GPx) expression] and apoptotic intensity (caspase 3/7 activity) decreased, whereas an unfolded protein response (HSP90) was induced under anoxia. This is the first clear evidence of the concerted regulation of the AOX and ETS genes in a hypoxia-tolerant freshwater bivalve and yet another example that exposure to hypoxia and anoxia is not necessarily accompanied by oxidative stress in hypoxia-tolerant mollusks. International audience; Delineating the physiological and biochemical causes of aging process in the animal kingdom is a highly active area of research not only because of potential benefits for human health but also because aging process is related to life history strategies (growth and reproduction) and to responses of organisms to environmental conditions and stress. In this synthesis, we advocate studying bivalve species as models for revealing the determinants of species divergences in maximal longevity. This taxonomic group includes the longest living metazoan on earth (Arctica islandica), which insures the widest range of maximum life span when shorter living species are also included in the comparative model. This model can also be useful for uncovering factors modulating the pace of aging in given species by taking advantages of the wide disparity of lifespan among different populations of the same species. For example, maximal lifespan in different populations of A islandica range from approximately 36 years to over 500 years. In the last 15 years, research has revealed that either regulation or tolerance to oxidative stress is tightly correlated to longevity in this group which support further investigations on this taxon to unveil putative mechanistic links between Reactive Oxygen Species and aging process.

The objective of this symposium at the First International Congress of Respiratory Biology (ICRB) was to enhance communication between comparative biologists and cancer researchers working on O(2) sensing via the HIF pathway. Representatives from both camps came together on August 13-16, 2006, in Bonn, Germany, to discuss molecular adaptations that occur after cells have been challenged by a reduced (hypoxia) or completely absent (anoxia) supply of oxygen. This brief "critters-to-cancer" survey discusses current projects and new directions aimed at improving understanding of hypoxic signaling and developing therapeutic interventions.

Environmental factors can affect the rate of ageing and shape the lifespan in marine ectotherms. The mechanisms and the degree of - environmental influence on aging can best be studied in species with wide ranging biogeographic distribution. One of the biomarkers of physiological ageing is the fluorescent age pigment lipofuscin, which accumulates over lifetime in tissues of bivalves. We compared lipofuscin accumulation rate in muscles and respiratory tissues of the extremely long lived bivalve Arctica islandica from five geographically distinct populations (Northern Norway, White Sea, Kiel Bay, German Bight and Iceland). Maximum investigated chronological age across different populations in the present study differed from 40 years in Kiel Bay to 192 years at Iceland. An inverse association between lipofuscin deposition rate and recorded maximum age was observed through inter-population comparisons. In most cases lipofuscin accumulated exponentially over age in a tissue specific manner. The age specific lipofuscin content was significantly higher in respiratory than muscles tissues in all populations. Cellular lipofuscin granule area can be used as indicator of aging across A. islandica populations with the variance in granule accumulation depending on the annual variations of salinity in different marine regions, but not on the habitat specific thermal envelope.

After 96 h goldfish exposure to 10, 25 or 50 mg/L of Ni(2+) no Ni accumulation was found in the brain, but lipid peroxide concentration was by 44% elevated in the brain, whereas carbonyl protein content was by 45-45% decreased in the heart. High molecular mass thiol concentration was enhanced by 30% in the heart, while in the brain low molecular mass thiol concentration increased by 28-88%. Superoxide dismutase activity was by 27% and 35% increased in the brain and heart, respectively. Glutathione peroxidase activity was lowered to 38% and 62% of control values in both tissues, whereas catalase activity was increased in the heart by 15-45%, accompanied by 18-29% decreased glutathione reductase activity. The disturbances of free radical processes in the brain and heart might result from Ni-induced injuries to other organs with more prominent changes in the heart, because of close contact of this organ with blood, whereas the blood-brain barrier seems to protect the brain.

Owing to its extremely long life span and occurrence in the entire North Atlantic, the Arctic boreal Arctica islandica has become of particular significance for monitoring the environment, because information on past environmental conditions is archived in morphological and biogeochemical properties of the calcareous shell. To evaluate whether such properties are comparable between different localities, shell and soft body morphometry of six A. islandica populations, Norwegian Coast, Kattegat, Kiel Bay (Baltic), White Sea, German Bight (North Sea), and off NE Iceland, were compared. Discriminant analysis indicated distinct differences between populations, albeit not related to geographical distance, but more likely to local hydrography, bottom morphology, and food regime.

A high input of lithogenic sediment from glaciers was assumed to be responsible for high Fe and Mn contents in the Antarctic soft shell clam Laternula elliptica at King George Island. Indeed, withdrawal experiments indicated a strong influence of environmental Fe concentrations on Fe contents in bivalve hemolymph, but no significant differences in hemolymph and tissue concentrations were found among two sites of high and lower input of lithogenic debris. Comparing Fe and Mn concentrations of porewater, bottom water, and hemolymph from sampling sites, Mn appears to be assimilated as dissolved species, whereas Fe apparently precipitates as ferrihydrite within the oxic sediment or bottom water layer prior to assimilation by the bivalve. Hence, we attribute the high variability of Fe and Mn accumulation in tissues of L. elliptica around Antarctica to differences in the geochemical environment of the sediment and the resulting Fe and Mn flux across the benthic boundary.

Molecular technologies are more frequently applied in Antarctic ecosystem research and the growing amount of sequence-based information available in databases adds a new dimension to understanding the response of Ant- arctic organisms and communities to environmental change. We apply molecular techniques, including finger- printing, and amplicon and metagenome sequencing, to understand biodiversity and phylogeography to resolve adaptive processes in an Antarctic coastal ecosystem from microbial to macrobenthic organisms and communities. Interpretation of the molecular data is not only achieved by their combination with classical methods (pigment analyses or microscopy), but furthermore by combining molecular with environmental data (e.g., sediment characteristics, biogeochemistry or oceanography) in space and over time. The studies form part of a long-term ecosystem investigation in Potter Cove on King-George Island, Antarctica, in which we follow the effects of rapid retreat of the local glacier on the cove ecosystem. Longevity and ageing are two sides of a coin, leaving the question open as to which one is the cause and which one the effect. At the individual level, the physiological rate of ageing determines the length of life (= individual longevity, as long as death results from old age and not from disease or other impacts). Individual longevity depends on the direct influence of environmental conditions with respect to nutrition, and the possibility for and timing of reproduction, as well as on the energetic costs animals invest in behavioural and physiological stress defence. All these environmental effectors influence hormonal and cellular signalling pathways that modify the individual physiological condition, the reproductive strategy, and the rate of ageing. At the species level, longevity (= maximum lifespan) is the result of an evolutionary process and, thus, largely determined by the species' behavioural and physiological adaptations to its ecological niche. Specifically, reproductive and breeding strategies have to be optimized in relation to local environmental conditions in different habitats. As a result of adaptive and evolutionary processes, species longevity is genetically underpinned, not necessarily by a few ageing genes, but by an evolutionary process that has hierarchically shaped and optimized species genomes to function in a specific niche or environmental system. Importantly, investigations and reviews attempting to unravel the mechanistic basis of the ageing process need to differentiate clearly between the evolutionary process shaping longevity at the species level and the regulatory mechanisms that alter the individual rate of ageing.

Recent rapid climate warming at the western Antarctic Peninsula (WAP) results in elevated glacial melting, enhanced sedimentary run-off, increased turbidity and impact of ice-scouring in shallow coastal areas. Discharge of mineral suspension from volcanic bedrock ablation and chronic physical disturbance are anticipated to influence sessile filter feeders such as the Antarctic soft shell clam Laternula elliptica (King and Broderip 1831). We investigated effects of sedimentary run-off on the accumulation of trace metals, and together with physical disturbance, the cumulative effect on oxidative stress parameters in younger and older L. elliptica from two stations in Potter Cove (King George Island, Antarctica) which are distinctly impacted by turbidity and ice-scouring. Fe, Mn, Sr, V and Zn concentrations were slightly higher in sediments of the station receiving more sediment run-off, but not enriched in bivalves of this station. The only element that increased in bivalves experimentally exposed to sediment suspension for 28 days was Mn. Concentration of the waste accumulation biomarker lipofuscin in nervous tissue was higher in L. elliptica from the “exposed” compared to the “less exposed” site, whereas protein carbonyl levels in bivalve mantle tissue were higher at the less sediment impacted site. Tissue metal content and lipofuscin in nervous tissue was generally higher in older compared to younger individuals from both field stations. We conclude that elevated sediment ablation does not per se result in higher metal accumulation in L. elliptica. Instead of direct absorbance from sediment particles, metal accumulation in gills seems to indicate uptake of compounds dissolved in the water column whereas metals in digestive gland appear to originate from enriched planktonic or detritic food. Accumulation of cellular waste products and potentially reactive metals over lifetime presumably alters Laternula elliptica physiological performance with age and may contribute to higher stress susceptibility in older animals.

Increase in oxidative damage and decrease in cellular maintenance is often associated with aging, but, in marine ectotherms, both processes are also strongly influenced by somatic growth, maturation and reproduction. In this study, we used a single cohort of the short-lived catarina scallop Argopecten ventricosus, to investigate the effects of somatic growth, reproduction and aging on oxidative damage parameters (protein carbonyls, TBARS and lipofuscin) and cellular maintenance mechanisms (antioxidant activity and apoptosis) in scallops, caged in their natural environment. The concentrations of protein carbonyls and TBARS increased steeply during the early period of fast growth and during reproduction in one-year-old scallops. However, oxidative damage was transient, and apoptotic cell death played a pivotal role in eliminating damage in gill, mantle and muscle tissues of young scallops. Animals were able to reproduce again in the second year, but the reduced intensity of apoptosis impaired subsequent removal of damaged cells. In late survivors low antioxidant capacity and apoptotic activity together with a fast accumulation of the age pigment lipofuscin was observed. Rates of oxygen consumption and oxidative stress markers were strongly dependent on somatic growth and reproductive state but not on temperature. Compared to longer-lived bivalves, A. ventricosus seems more susceptible to oxidative stress with higher tissue-specific protein carbonyl levels and fast accumulation of lipofuscin in animals surviving the second spawning. Superoxide dismutase activity and apoptotic cell death intensity were however higher in this short-lived scallop than in longer-lived bivalves. The life strategy of this short-lived and intensely predated scallop supports rapid somatic growth and fitness as well as early maturation at young age at the cost of fast cellular degradation in second year scallops.

Glacial retreat and subglacial bedrock erosion are consequences of rapid regional warming on the West Antarctic Peninsula. Sedimentation of fine-grained eroded particles can impact the physiology of filter-feeding benthic organisms. We investigated the effect of increasing concentrations of sediment on the oxygen consumption of suspension feeding species, the ascidians Molgula pedunculata, Cnemidocarpa verrucosa, Ascidia challengeri, and the pennatulid Malacobelemnon daytoni in Potter Cove (South Shetland Islands, Antarctica). In A. challengeri and C. verrucosa, oxygen consumption increased gradually up to a critical sediment concentration (C crit) where species oxygen consumption was maximal (O max in mg O2 g−1dm day−1) and further addition of sediments decreased respiration. C crit was 200 mg L−1 for A. challengeri (O max of 0.651 ± 0.238) and between 100 and 200 mg L−1 for C. verrucosa (O max of 0.898 ± 0.582). Oxygen consumption of M. pedunculata increased significantly even at low sediment concentrations (15–50 mg sediment L−1). Contrary to the ascidians, sediment exposure did not affect oxygen consumption of the sea pen. The tiered response to sedimentation in the four species corroborates recent field observations that detected a reduction in the abundance of the sensitive ascidian M. pedunculata from areas strongly affected by glacial sediment discharge, whereas sea pens are increasing in abundance. Our investigation relates consequences (population shifts in filter-feeder communities) to causes (glacial retreat) and is of importance for modelling of climate change effects in Antarctic shallow coastal areas.

Environmental factors are known to modify the life history of marine ectotherms. In a 16-month laboratory experiment, we investigated the influence of temperature and presence of predators on life-history parameters including shell growth, survival and the energy investment in reproduction and body mass of the short-lived (2–3 years) scallop Argopecten ventricosus. In parallel, A. ventricosus was maintained in the field at the Pacific coast of Baja California, Mexico, to compare growth, survival and reproductive effort under natural conditions. For the laboratory treatments, scallops were reared at simulated field temperatures (SFT), 5 °C above SFT and in the presence of predators. Elevated water temperatures caused higher growth and gonad production, although at the cost of increased mortality. Presence of predators induced energy allocation to muscle rather than gonad growth, deferred spawning and extended survival. Field scallops exhibited higher growth, higher reproductive investment and were able to reproduce twice, whereas all laboratory scallops died after the first spawning. The natural variability of environmental parameters such as food and temperature may thus support optimal growth in the field, and when animals are protected from predators, reproduction in the second year of life.

In Arctica islandica, a long lifespan is associated with low metabolic activity, and with a pronounced tolerance to low environmental oxygen. In order to study metabolic and physiological responses to low oxygen conditions vs. no oxygen in mantle, gill, adductor muscle and hemocytes of the ocean quahog, specimens from the German Bight were maintained for 3.5 days under normoxia (21 kPa = controls), hypoxia (2 kPa) or anoxia (0 kPa). Tissue levels of anaerobic metabolites octopine, lactate and succinate as well as specific activities of octopine dehydrogenase (ODH) and lactate dehydrogenase (LDH) were unaffected by hypoxic incubation, suggesting that the metabolism of A. islandica remains fully aerobic down to environmental oxygen levels of 2 kPa. PO2-dependent respiration rates of isolated gills indicated the onset of metabolic rate depression (MRD) below 5 kPa in A. islandica, while anaerobiosis was switched on in bivalve tissues only at anoxia. Tissue-specific levels of glutathione (GSH), a scavenger of reactive oxygen species (ROS), indicate no anticipatory antioxidant response takes place under experimental hypoxia and anoxia exposure. Highest specific ODH activity and a mean ODH/LDH ratio of 95 in the adductor muscle contrasted with maximal specific LDH activity and a mean ODH/LDH ratio of 0.3 in hemocytes. These differences in anaerobic enzyme activity patterns indicate that LDH and ODH play specific roles in different tissues of A. islandica which are likely to economize metabolism during anoxia and reoxygenation.

Marine bivalves, such as the Antarctic soft shell clam Laternula elliptica (King and Broderip 1831), experience a wide range of environmental influences including the permanent contact to the surrounding microbial community due to filter feeding or strong physiochemical disturbances in a changing environment. Such impacts are anticipated to influence L elliptica physiology, including the immune system, especially under the current climate change conditions at the Western Antarctic Peninsula (WAP). To understand basal immune defence reactions in L elliptica, we investigated in vitro hemocyte responses upon bacterial challenge or after stimulation with different substances of microbial origin (pathogen associated molecular patterns, PAMPS). To study the age-dependent in vivo response of hemocyte abundance, starvation and injury experiments were undertaken with young and old individuals. Hemocytes of L. elliptica feature common immune functions such as phagocytosis and ROS generation, as well as agglutination of bacterial cells. ROS generation response to different stimuli was generally low and lower in larger/older animals compared to smaller/younger individuals. Physiological conditions such as size/age, starvation and injury modulated the abundance of hemocytes in L. elliptica. Implications are that current and future climate change conditions in West Antarctic coastal ecosystems scenarios may lead to changes in species survival and population composition. Recent rapid changes of air temperature on the western side of the Antarctic Peninsula results in increased sediment discharge and ice scouring frequencies in coastal regions. These changes are bound to especially affect slow growing, sessile filter feeders such as the Antarctic bivalve, Laternula elliptica, a long-lived and abundant key species with circumpolar distribution. We investigated the effect of sedimentation and ice scouring on small/young and large/old individuals at two closely located stations, distinctly influenced by both types of disturbance. Small individuals dealt better with disturbance in terms of their respiratory response to sediment exposure, reburrowing ability, and survival after injury, compared to larger animals. At the more disturbed station L. elliptica population density was lower, but larger animals reburrowed faster after iceberg disturbance and reduced their metabolic rate under strong sediment coverage, compared to larger animals of the less disturbed station, indicating that an adaptation or learning response to both types of disturbance may be possible. Smaller individuals were not influenced. Laternula elliptica seems capable of coping with the rapidly changing environmental conditions. Due to a decrease in population density and mean population lifespan, L. elliptica could however lose its key role in the bentho-pelagic carbon flux in areas of high sediment deposition.

Summary 1. The oxidative stress theory of ageing predicts that animals living longer will have less cumulative oxidative damage together with structural characteristics that make them more resistant to oxidative damage itself. 2. Although a general relationship between body size, metabolism and longevity does not exist in marine invertebrates, they are generally characterized by low rates of metabolism and reactive oxygen species (ROS) formation associated with lower antioxidant enzyme activities compared to vertebrates. 3. Birds and mammals have very similar size-affected metabolic rates and their metabolic intensity explains only some of the variation in maximum lifespan potential (MLSP). Within each class, smaller animals have higher rates of metabolism and ROS production and membranes that are more susceptible to oxidative damage and autocatalytic propagation of free radicals than larger ones. 4. Although the high variation in life-history strategies is accompanied by substantial variation in MLSP, there is a consistent positive correlation between rates of ROS formation and antioxidant levels among most animals examined so far for these traits. The consensus of these studies is that ROS and antioxidant levels are inversely related to MLSP. 5. The lack of a clear stoichiometric relation between variables contributing to oxidative stress limits our capacity to infer longevity consequences from measures of pro-oxidant or antioxidant status among or within species.

Marine invertebrates with open circulatory system establish low and constant oxygen partial pressure (Po2) around their tissues. We hypothesized that as a first step towards maintenance of low haemolymph and tissue oxygenation, the Po2in molluscan mantle cavity water should be lowered against normoxic (21 kPa) seawater Po2, but balanced high enough to meet the energetic requirements in a given species. We recorded Po2in mantle cavity water of five molluscan species with different lifestyles, two pectinids ( Aequipecten opercularis , Pecten maximus ), two mud clams ( Arctica islandica , Mya arenaria ), and a limpet ( Patella vulgata ). All species maintain mantle cavity water oxygenation below normoxic Po2. Average mantle cavity water Po2correlates positively with standard metabolic rate (SMR): highest in scallops and lowest in mud clams. Scallops show typical Po2frequency distribution, with peaks between 3 and 10 kPa, whereas mud clams and limpets maintain mantle water Po2mostly 2time series. Adjustment of mantle cavity Po2to lower than ambient levels through controlled pumping prevents high oxygen gradients between bivalve tissues and surrounding fluid, limiting oxygen flux across the body surface. The patterns of Po2in mantle cavity water correspond to molluscan ecotypes.

During the austral spring and summer months, the Antarctic limpet Nacella concinna colonizes intertidal environments in the Western Antarctica Peninsula region. The species is divided into a permanently sub-littoral and a seasonally intertidal, migratory subpopulation. We investigate the physiological differentiation between the two limpet groups to identify cellular and molecular changes that accompany adaptation of stenothermal Antarctic invertebrates to life under more stressful intertidal habitat conditions. A major difference between the two groups is the significantly higher concentrations of heavy metals (Fe, Al, Zn) from ingested sediments in sub-littoral limpet digestive glands (DG), associated with higher rates of reactive oxygen species (ROS) formation in this organ. ROS formation is accompanied by significantly higher SOD activity in sub-littoral limpet DG. These high SOD activities are, however, not conserved during either air exposure or hypoxic stress exposure of the sub-littoral limpets, when ROS production is slowed due to the absence of oxygen. The intertidal animals maintain higher levels of SOD and also conserve catalase activity at higher levels during hypoxia or air exposure compared to sub-littoral individuals under the same exposure conditions. More oxidized redox potential in gills and foot muscle and higher antioxidant enzyme activities in gills indicate that intertidal limpets maintain more oxygenated tissues during air exposure, in keeping with shell-lifting for oxygen up-take by the gills of intertidal limpets which migrate up the shore in the spring and down in the autumn. An increase of the redox ratio (GSSG/GSH) and accumulation of the lipid oxidation derived malonedialdehyde in intertidal limpet foot muscle during 12 h of exposure to air shows that indeed this tissue becomes more oxidized before the limpets eventually contract their shells tightly to minimize water loss and eventually become anaerobic. Intertidal limpets obviously avoid early onset of anaerobic energy production seen in their sub-littoral congeners when exposed to air and are still able to maintain tissue redox ratio balance when exposed to air.

Owing to its extraordinary lifespan and wide geographical distribution along the continental margins of the North Atlantic Ocean, the ocean quahog Arctica islandica may become an important indicator species in environmental change research. To test for applicability and ''calibrate'' the Arctica-indicator, metabolic properties of A. islandica specimens were compared across different climatic and oceanographic regions. Fully saline populations from Iceland to the North Sea as well as animals from polyhaline and low salinity, environments, the White Sea and the Baltic were included in the study. This calibration centrally includes recordings of growth-age relationships in different populations. Shells were used as age recorders by counting annual growth bands. As a result of this study, we propose a general respiration model that links individual metabolic rates of A. islandica from five populations: Norwegian coast, Kattegat, Kiel Bay (Baltic Sea), White Sea and German Bight (North Sea), to body mass, water temperature and site. Temperature exerts distinct site specific effects on respiration rate, which is indicated by Q10 values ranging from 4.48 for German Bight to 1.15 for Kiel Bay animals. Individual age, occurrence of apneal respiratory gaps, parasite infestation and salinity do not affect respiration rate. Respiration of Arctica islandica is significantly below the average of 59 bivalve species when compared at the same temperature and animal mass. This respiration model principally enables the coupling of A. islandica life history and population dynamics to regional oceanographic temperature models.

Bivalves are newly discovered models of natural aging. This invertebrate group includes species with the longest metazoan lifespan approaching 400 y, as well as species of swimming and sessile lifestyles that live just for 1 y. Bivalves from natural populations can be aged by shell growth bands formed at regular intervals of time. This enables the study of abiotic and biotic environment factors (temperature, salinity, predator and physical disturbance) on senescence and fitness in natural populations, and distinguishes the impact of extrinsic effectors from intrinsic (genetic) determinants of animal aging. Extreme longevity of some bivalve models may help to analyze general metabolic strategies thought to be life prolonging, like the transient depression of metabolism, which forms part of natural behaviour in these species. Thus, seasonal food shortage experienced by benthic filter feeding bivalves in polar and temperate seas may mimic caloric restriction in vertebrates. Incidence of malignant neoplasms in bivalves needs to be investigated, to determine the implication of late acting mutations for bivalve longevity. Finally, bivalves are applicable models for testing the implication of heterozygosity of multiple genes for physiological tolerance, adaptability (heterozygote superiority), and life expectancy.

Antarctic intertidal zones are extremely harsh environments, and the Antarctic limpet Nacella concinna is one of the most conspicuous components of the macrofauna, colonizing such areas at King George Island, South Shetlands. The limpet subpopulation encompasses an intertidal and a subtidal ecomorph; these differ with respect to shell height and inner volume. We studied mor- phological, behavioural and physiological traits that distinguish both N. concinna subpopulations. We found a higher volume to circumference ratio of intertidal versus subtidal limpets to go along with a drastically reduced loss of body water during air exposure in intertidal specimens. Smaller inter- tidal limpets were less susceptible to desiccation than larger individuals. Further, the intertidal snails avoided accumulation of anaerobic metabolites (succinate, lactate and propionate), whereas subtidal limpets switched to anaerobic fermentation when exposed to air for >12 h. Only acetate accumulated in both types of snails after 12 h. Mean pO2 in shell water was higher in intertidal (2.6 kPa) than sub- tidal individuals (1.3 kPa) when submersed under water, and only the intertidal snails appeared able to respire air on emersion. These differences document strong physiological diversification between shore levels and shell morphs in the Antarctic limpet subpopulation at King George Island.

SUMMARY The decline of cellular and especially mitochondrial functions with age is,among other causes, held responsible for a decrease in physiological fitness and exercise capacity during lifetime. We investigated size- and age-related changes in the physiology of exercising specimens of the short lived swimming scallop Aequipecten opercularis (maximum life span 8 to 10 years)from the Isle of Man, UK. A. opercularis swim mainly to avoid predators, and a decrease in swimming abilities would increase the risk of capture and lower the rates of survival. Bigger (older) individuals were found to have lower mitochondrial volume density and aerobic capacities (citrate synthase activity and adenylates) as well as less anaerobic capacity deduced from the amount of glycogen stored in muscle tissue. Changes in redox potential, tissue pH and the loss of glutathione in the swimming muscle during the exercise were more pronounced in young compared to older individuals. This indicates that older individuals can more effectively stabilize cellular homeostasis during repeated exercise than younger animals but with a possible fitness cost as the change in physiology with age and size might result in a changed escape response behaviour towards predators.

Melting of coastal glaciers at the West Antarctic Peninsula (WAP) causes shorter winter sea ice duration, intensified ice scouring, sediment erosion and surface freshening in summer, which alters coastal productivity and feeding conditions for the benthos. The soft shell clam Laternula elliptica is a fast growing and abundant filter feeder in coastal Antarctica and a key element for bentho-pelagic carbon recycling. Our aim was to assess the cellular growth and maintenance capacity of small and large clams during natural winter food shortage (seasonal sampling) and in response to experimental starvation exposure. We measured tissue specific proliferation rates and the expression of cell cycling genes and the iron binding protein Le-ferritin in freshly collected specimens in spring (Nov 2008) and at the end of summer (March 2009). For the experimental approach, we focused on 14 cell cycling and metabolic genes using the same animal size groups. Mantle tissue of young bivalves was the only tissue showing accelerated proliferation in summer (1.7 % of cells dividing per day in March) compared to 0.4% dividing cells in animals collected in November. In mantel, siphon and adductor muscle proliferation rates were higher in younger compared to older indiciduals. At transcript level, Le-cyclin D was upregulated in digestive gland of older animals collected in spring (Nov) compared to March indicating enhanced cell proliferation. Likewise, during experimental starvation Le-cyclin D expression increased in large clam digestive gland, whereas Le-cyclin D and the autophagic factor beclin1 decreased in digestive gland of smaller starved clams. The paper corroborates earlier findings of size and age dependent differences in the metabolic response and gene expression patterns in L. elliptica under energetic deprivation. Age structure of shallow water populations can potentially change due to differences in cellular response between young and old animals as environmental stress levels increase.

Background: Reactive oxygen (ROS) and nitrogen (RNS) species are produced during normal unstressed metabolic activity in aerobic tissues. Most analytical work uses tissue homogenates, and lacks spatial information on the tissue specific sites of actual ROS formation. Live-imaging techniques (LIT) utilize target-specific fluorescent dyes to visualize biochemical processes at cellular level. Results: Together with oxidative stress measurements, here we report application of LIT to bivalve gills for ex-vivo analysis of gill physiology and mapping of ROS and RNS formation in the living tissue. Our results indicate that a) mitochondria located in the basal parts of the epithelial cells close to the blood vessels are hyperpolarized with high Delta Psi m, whereas b) the peripheral mitochondria close to the cilia have low (depolarized)Delta Psi m. These mitochondria are densely packed (mitotracker Deep Red 633 staining), have acidic pH (Ageladine-A) and collocate with high formation of nitric oxide (DAF-2DA staining). NO formation is also observed in the endothelial cells surrounding the filament blood sinus. ROS (namely H2O2, HOO center dot and ONOO- radicals, assessed through C-H(2)DFFDA staining) are mainly formed within the blood sinus of the filaments and are likely to be produced by hemocytes as defense against invading pathogens. On the ventral bend of the gills, subepithelial mucus glands contain large mucous vacuoles showing higher fluorescence intensities for O-2 is approximately equal to than the rest of the tissue. Whether this O-2 is approximately equal to production is instrumental to mucus formation or serves antimicrobial protection of the gill surface is unknown. Cells of the ventral bends contain the superoxide forming mucocytes and show significantly higher protein carbonyl formation than the rest of the gill tissue. Conclusions: In summary, ROS and RNS formation is highly compartmentalized in bivalve gills under unstressed conditions. The main mechanisms are the differentiation of mitochondria membrane potential and basal ROS formation in inner and outer filament layers, as well as potentially antimicrobial ROS formation in the central blood vessel. Our results provide new insight into this subject and highlight the fact that studying ROS formation in tissue homogenates may not be adequate to understand the underlying mechanism in complex tissues.

We investigated the shielding against solar ultraviolet radiation and inducible damage, as well as the short-term response of whole animal metabolic rate in two Antarctic shallow water amphipod species. Light absorbance by the carapace of Gondogeneia antarctica and Djerboa furcipes was higher in the UVR (UVB + UVA) range (42.1% and 54.5% on average respectively) compared to the PAR (photosynthetically active radiation) range (38.1% and 50.1% respectively) of the solar spectrum. Bands of higher absorbance correlated with maximal absorbance ranges of sunscreening compounds indicating mycosporine-like amino acids (MAAs) and carotenoids to be innate compounds of the exoskeleton of these species. Though the antioxidant enzyme catalase was photoinhibited, protein damage products did not accumulate under experimental exposure to a daily dose of 6.84 kJ m � 2 d � 1 UVB, 66.24 kJ m � 2 d � 1 UVA and 103.14 kJ m � 2 d � 1 PAR. Animal oxygen consumption during UV-exposure was measured as an indicator of immediate behavioural and physiological stress response. UVB as well as UVA induced a response with altered and highly variable respiratory intensity. Our findings indicate that sub-lethal UVR exposure causes increased oxygen consumption in polar amphipods due to radiation avoidance, shelter seeking behaviour, and presumably also from cellular repair processes. 2007 Elsevier Ltd. We studied the seasonal variation on aerobic metabolism and the response of oxidative stress parameters in the digestive glands of the subpolar limpet Nacella ( P. ) magellanica . Sampling was carried out from July (winter) 2002 to July 2003 in Beagle Channel, Tierra del Fuego, Argentina. Whole animal respiration rates increased in early spring as the animals spawned and remained elevated throughout summer and fall (winter: 0.09 ± 0.02 μmol O 2 h − 1 g − 1 ; summer: 0.31 ± 0.06 μmol O 2 h − 1 g − 1 ). Oxidative stress was assessed at the hydrophilic level as the ascorbyl radical content / ascorbate content ratio (A / AH − ). The A / AH − ratio showed minimum values in winter (3.7 ± 0.2 10 − 5 AU) and increased in summer (18 ± 5 10 − 5 AU). A similar pattern was observed for lipid radical content (122 ± 29 pmol mg − 1 fresh mass [FW] in winter and 314 ± 45 pmol mg − 1 FW in summer), iron content (0.99 ± 0.07 and 2.7 ± 0.6 nmol mg − 1 FW in winter and summer, respectively) and catalase activity (2.9 ± 0.2 and 7 ± 1 U mg − 1 FW in winter and summer, respectively). Since nitrogen derived radicals are thought to be critically involved in oxidative metabolism in cells, nitric oxide content was measured and a significant difference in the content of the Fe–MGD–NO adduct in digestive glands from winter and summer animals was observed. Together, the data indicate that both oxygen and nitrogen radical generation rates in N. ( P. ) magellanica are strongly dependent on season.

Climatic change in the Antarctic Peninsula has driven profound shifts in the seabed., The Antarctic Peninsula (AP) is one of the three places on Earth that registered the most intense warming in the last 50 years, almost five times the global mean. This warming has strongly affected the cryosphere, causing the largest ice-shelf collapses ever observed and the retreat of 87% of glaciers. Ecosystem responses, although increasingly predicted, have been mainly reported for pelagic systems. However, and despite most Antarctic species being benthic, responses in the Antarctic benthos have been detected in only a few species, and major effects at assemblage level are unknown. This is probably due to the scarcity of baselines against which to assess change. We performed repeat surveys of coastal benthos in 1994, 1998, and 2010, analyzing community structure and environmental variables at King George Island, Antarctica. We report a marked shift in an Antarctic benthic community that can be linked to ongoing climate change. However, rather than temperature as the primary factor, we highlight the resulting increased sediment runoff, triggered by glacier retreat, as the potential causal factor. The sudden shift from a “filter feeders–ascidian domination” to a “mixed assemblage” suggests that thresholds (for example, of tolerable sedimentation) and alternative equilibrium states, depending on the reversibility of the changes, could be possible traits of this ecosystem. Sedimentation processes will be increasing under the current scenario of glacier retreat, and attention needs to be paid to its effects along the AP.

Mitochondrial DNA (mtDNA) is strictly maternally inherited in metazoans. The major exception to this rule has been found in many bivalve species which allow the presence of different sex-linked mtDNA molecules. This mechanism, named Doubly Uniparental Inheritance (DUI), is characterized by the presence of 2 mtDNAs: the female mtDNA is found in somatic tissue and female gonads whereas the male mtDNA is usually found in male gonads and sperm. In this study we highlight the existence of two divergent mitochondrial haplotypes with a low genetic difference around 6-8% in Arctica islandica, a long-lived clam belonging to the Arcticidae, a sister group to the Veneridae in which DUI has been found. Phylogenetic analysis on cytochrome b and 16S sequences from somatic and gonadic tissues of clams belonging to different populations reveal the presence of the “divergent” type in male gonads only and the “normal” type in somatic tissues and female gonads. This peculiar segregation of divergent mtDNA types speaks for the occurrence of the DUI mechanism in Arctica islandica. This example also highlights the difficulties to assess the presence of such particular mitochondrial inheritance system and underlines the possible misinterpretations in phylogeographic and phylogenetic studies of bivalve species linked to the presence of two poorly differentiated mitochondrial genomes.

Iron stable isotope signatures (δ56Fe) in hemolymph (bivalve blood) of the Antarctic bivalve Laternula elliptica were analyzed by Multiple Collector-Inductively Coupled Plasma-Mass Spectrometry (MC-ICP-MS) to test whether the isotopic fingerprint can be tracked back to the predominant sources of the assimilated Fe. An earlier investigation of Fe concentrations in L. elliptica hemolymph suggested that an assimilation of reactive and bioavailable Fe (oxyhydr)oxide particles (i.e. ferrihydrite), precipitated from pore water Fe around the benthic boundary, is responsible for the high Fe concentration in L. elliptica (Poigner et al., 2013b). At two stations in Potter Cove (King George Island, Antarctica) bivalve hemolymph showed mean δ56Fe values of −1.19 ± 0.34‰ and −1.04 ± 0.39‰, respectively, which is between 0.5‰ and 0.85‰ lighter than the pool of easily reducible Fe (oxyhydr)oxides of the surface sediments (−0.3‰ to −0.6‰). This is in agreement with the enrichment of lighter Fe isotopes at higher trophic levels, resulting from the preferential assimilation of light isotopes from nutrition. Nevertheless, δ56Fe hemolymph values from both stations showed a high variability, ranging between −0.21‰ (value close to unaltered/primary Fe(oxyhydr)oxide minerals) and −1.91‰ (typical for pore water Fe or diagenetic Fe precipitates), which we interpret as a “mixed” δ56Fe signature caused by Fe assimilation from different sources with varying Fe contents and δ56Fe values. Furthermore, mass dependent Fe fractionation related to physiological processes within the bivalve cannot be ruled out. This is the first study addressing the potential of Fe isotopes for tracing back food sources of bivalves.

Effects of acute cold exposure (at 1 degrees C and 5 degrees C) on tissue redox state and oxidative stress parameters, as well as the onset of hypoxic signaling were investigated in the North Sea eelpout, Zoarces viviparus. Activation of the transcription factor HIF-1 (hypoxia inducible factor) was detected in liver samples after acute cold exposure. At this temperature the cellular redox milieu was significantly reduced (below -270 mV) as compared to controls (-250 to -267 mV). Increased levels of oxidative stress parameters (TBARS and protein carbonyls) were observed mainly during recovery at control temperature (12 degrees C). This increase in oxidative stress parameters, in spite of maintained antioxidant capacity, indicates that acute cold stress and recovery mimic ischemia/reperfusion events as found in mammals. Notably the non-enzymatic antioxidant defense (e.g. glutathione) may play an important role for eelpout ROS scavenging capacity under cold stress.

We compared physiological ageing parameters in 2 scallops, the temperate Aequipecten opercularis and the Antarctic Adamussium colbecki. These 2 species are phylogeneti- cally closely related and display a similar lifestyle but have distinctly different maximum lifespans (MLSP). A. opercularis does not live longer than 8 to 10 yr, whereas A. colbecki lives over 18 yr. The development of several physiological ageing parameters over time, chosen according to the 'free rad- ical theory of ageing', was compared in the 2 species to identify differences in the ageing process. In the shorter-lived A. opercularis, activities of the mitochondrial enzymes citrate synthase and cytochrome c oxidase and of the antioxidant enzyme catalase showed a more pronounced decrease with increasing age than in the longer-lived A. colbecki. In line with this finding, lipofuscin accumu- lation increased more distinctly in A. opercularis than in A. colbecki, while tissue protein content decreased in A. opercularis but increased in A. colbecki. Its better preservation of mitochondrial and antioxidant enzyme activities and the avoidance of waste accumulation may enable A. colbecki to live longer than A. opercularis. Mitochondrial function investigated in A. opercularis showed only minor changes with age, and mitochondrial H2O2 generation rates were low at all ages. We relate our findings to the 'free radical-rate of living' theory, to the 'uncoupling to survive' hypothesis, and to the particular lifestyle of these scallops.

SUMMARYThe interplay between antioxidants, heat shock proteins and hypoxic signaling is supposed to be important for passive survival of critical temperature stress, e.g. during unfavorable conditions in hot summers. We investigated the effect of mild (18°C), critical (22°C) and severe(26°C) experimental heat stress, assumed to induce different degrees of functional hypoxia, as well as the effect of recovery following heat stress on these parameters in liver samples of the common eelpout Zoarces viviparus.Upon heat exposure to critical and higher temperatures we found an increase in oxidative damage markers such as TBARS (thiobarbituric reactive substances)and a more oxidized cellular redox potential, combined with reduced activities of the antioxidant enzyme superoxide dismutase at 26°C. Together, these point to higher oxidative stress levels during hyperthermia. In a recovery-time series, heat-induced hypoxia and subsequent reoxygenation upon return of the fishes to 12°C led to increased protein oxidation and chemiluminescence rates within the first 12 h of recovery, therein resembling ischemia/reperfusion injury in mammals.HSP70 levels were found to be only slightly elevated after recovery from sub-lethal heat stress, indicating minor importance of the heat shock response in this species. The DNA binding activity of the hypoxia-inducible transcription factor (HIF-1) was elevated only during mild heat exposure(18°C), but appeared impaired at more severe heat stress. We suppose that the more oxidized redox state during extreme heat may interfere with the hypoxic signaling response.

High oxygen solubility at cold-water temperature is frequently considered to be responsible for an apparently elevated level of antioxidant protection in marine ectotherms from polar environments. However, tissue oxidative stress is in most cases a function of elevated or variable pO2, rather than of an elevated tissue oxygen concentration. This review summarizes current knowledge on pro- and antioxidant processes in marine invertebrates and fish, and relates reactive oxygen species (ROS) formation in polar ectotherms to homeoviscous adaptations of membrane and storage lipids, as well as to tissue hypoxia and re-oxygenation during physiological stress.

Daphnia was collected from five subarctic ponds which differed greatly in their DOC contents and, consequently, their underwater light (UV) climates. Irrespective of which Daphnia species was present, and contrary to expectations, the ponds with the lowest DOC concentrations (highest UV radiation levels) contained Daphnia with the highest eicosapentaenoic acid (EPA) concentrations. In addition, EPA concentrations in these Daphnia generally decreased in concert with seasonally increasing DOC concentrations. Daphnia from three of the ponds was also tested for its tolerance to solar ultraviolet radiation (UVR) with respect to survival. Daphnia pulex from the clear water pond showed, by far, the best UV-tolerance, followed by D. longispina from the moderately humic and D. longispina from the very humic pond. In addition, we measured sublethal parameters related to UV-damage such as the degree to which the gut of Daphnia appeared green (as a measure of their ability to digest algae), and whether their guts appeared damaged. We developed a simple, noninvasive scoring system to quantify the proportion of the gut in which digestive processes were presumably active. This method allowed repeated measurement of the same animals over the course of the experiment. We demonstrated, for the first time, that sublethal damage of the gut precedes mortality caused by exposure to UVR. In a parallel set of experiments we fed UV-exposed and non-exposed algae to UV-exposed and non-exposed daphnids. UVR pretreatment of algae enhanced the negative effects of exposure to natural solar UV-irradiation in Daphnia. These UV-related effects were generally not specific to the species of Daphnia.

To understand the adaptation of euphausiid (krill) species to oxygen minimum zones (OMZ), respiratory response and stress experiments combining hypoxia/reoxygenation exposure with warming were conducted. Experimental krill species were obtained from the Antarctic (South Georgia area), the Humboldt Current system (HCS, Chilean coast), and the Northern California Current system (NCCS, Oregon). Euphausia mucronata from the HCS shows oxyconforming pO2-dependent respiration below 80% air saturation (18 kPa). Normoxic subsurface oxygenation in winter posed a “high oxygen stress” for this species. The NCCS krill, Euphausia pacifica, and the Antarctic krill, Euphausia superba maintain respiration rates constant down to low critical pO2 values of 6 kPa (30% air saturation) and 11 kPa (55% air saturation), respectively. Antarctic krill had low antioxidant enzyme activities, but high concentrations of the molecular antioxidant glutathione (GSH) and was not lethally affected by 6 h exposure to moderate hypoxia. Temperate krill species had higher SOD (superoxide dismutase) values in winter than in summer, which relate to higher winter metabolic rate (E. pacifica). In all species, antioxidant enzyme activities remained constant during hypoxic exposure at the typical temperature for their habitat. Warming by 7°C above their typical temperature in summer increased SOD activities and GSH levels in E. mucronata (HCS), but no oxidative damage occurred. In winter, when the NCCS is well mixed and the OMZ is deeper, +4°C of warming combined with hypoxia represents a lethal condition for E. pacifica. In summer, when the OMZ expands upwards (100 m subsurface), antioxidant defences counteracted hypoxia and reoxygenation effects in E. pacifica, but only at mildly elevated temperature (+2°C). In this season, experimental warming by +4°C reduced antioxidant activities and the combination of warming with hypoxia again caused mortality of exposed specimens. We conclude that a climate change scenario combining warming and hypoxia represents a serious threat to E. pacifica and, as a consequence, NCCS food webs.

Lipid peroxidation in Laternula elliptica was assessed by detecting lipid radicals by electronic paramagnetic resonance. The values were compared with data from the temperate mud clam Mya arenaria. Lipid radical content was higher in the Antarctic bivalve than in the temperate mud clam, even within the range of its habitat temperature. The rate of generation of lipid radicals was affected by the iron content in the samples. The iron content in individual samples of digestive glands in L. elliptica ranged from 3 to 6 nmol g(-1) fresh weight (fwt) and in M. arenaria from 0.6 to 2.7 nmol g(-1) fwt. Arrhenius plots, developed from the rates obtained in the presence of 25 microM iron, showed no significant differences between the activation energy calculated for digestive glands of L. elliptica and M. arenaria. The Fe3+ reduction rate in L. elliptica was higher than in M. arenaria (4.7 +/- 0.9 vs. 1.8 +/- 0.4 nmol mg(-1) protein min(-1), respectively). L. elliptica had a higher content of alpha-tocopherol and beta-carotene than M. arenaria. Our data suggest that increased lipid radical content in the membranes of cold-adapted organisms could be related to iron content.
