Studies of dairy cow longevity usually focus on the animal life after first calving, with few studies considering early life conditions and their effects on longevity. The objective was to evaluate the effect of birth conditions routinely collected by Dairy Herd Improvement agencies on offspring longevity measured as length of life and length of productive life. Lactanet provided 712,890 records on offspring born in 5,425 Quebec dairy herds between January 1999 and November 2015 for length of life, and 506,066 records on offspring born in 5,089 Quebec dairy herds between January 1999 and December 2013 for length of productive life. Offspring birth conditions used in this study were calving ease (unassisted, pull, surgery, or malpresentation), calf size (small, medium, or large), and twinning (yes or no). Observations were considered censored if the culling reason was "exported," "sold for dairy production," or "rented out" as well as if the animals were not yet culled at the time of data extraction. If offspring were not yet culled when the data were extracted, the last test-day date was considered the censoring date. Conditional inference survival trees were used in this study to analyze the effect of offspring birth conditions on offspring longevity. The hazard ratio of culling between the groups of offspring identified by the survival trees was estimated using a Cox proportional hazard model with herd-year-season as a frailty term. Five offspring groups were identified with different length of life based on their birth condition. Offspring with the highest length of life [median = 3.61 year; median absolute deviation (MAD) = 1.86] were those classified as large or medium birth size and were also the result of an unassisted calving. Small offspring as a result of a twin birth had the lowest length of life (median = 2.20 year; MAD = 1.69) and were 1.52 times more likely to be culled early in life. Six groups were identified with different length of productive life. Offspring that resulted from an unassisted or surgery calving and classified as large or medium when they were born were in the group with the highest length of productive life (median = 2.03 year; MAD = 1.63). Offspring resulting from a malpresentation or pull in a twin birth were in the group with the lowest length of productive life (median = 1.15 year; MAD = 1.11) and were 1.70 times more likely to be culled early in life. In conclusion, birth conditions of calving ease, calf size, and twinning greatly affected offspring longevity, and such information could be used for early selection of replacement candidates.

Estimated breeding values of a selection index, production, durability, health, and fertility traits from Canadian Ayrshire, Jersey, and Brown Swiss bulls and cows were used to study genetic selection differentials (GSD). The bulls and cows were born from 1950 and 1960, respectively. The GSD for the 3 Canadian dairy populations were studied along the 4-path selection model: sire-to-bull (SB), dam-to-bull (DB), sire-to-cow (SC), and dam-to-cow (DC) pathways. We also determined the variations in realized GSD due to herd and herd × year of conception in addition to the effects of some environmental factors on realized GSD of the SC and DC paths. The mean realized GSD of the DB were higher than those of other paths and were increasing for lifetime performance index, 305-d milk yield, 305-d fat yield, and 305-d protein yield in all 3 dairy cattle populations. We observed no clear trends in realized GSD for type traits in all 3 dairy cattle breeds except for the apparent increasing trends in realized GSD of mammary system, dairy strength, and feet and legs in the DB and SC paths of the Ayrshire breed. No clear patterns were observed in the realized GSD of daughter fertility in the SB, DB, and SC paths of all dairy cattle breeds. Realized GSD for somatic cell score showed increasing and favorable trends in the 3 most influential selection paths (SB, DB, and SC). Year of conception influenced realized GSD of artificial insemination bulls in Ayrshire, Jersey, and Brown Swiss dairy populations. Selection emphases for the SC path generally increased with time. There was considerable variation among herds in selection pressures applied in the SC and DC pathways but no clear association with housing system or region. This study demonstrates that variations exist among herds of minor dairy cattle breeds in their selection for economically important traits. These variations offer opportunities for further improvements in these populations.

Identifying and interpreting pleiotropic loci is essential to understanding the shared etiology among diseases and complex traits. However, this strategy does not account for the complex genetic architectures of traits, such as genetic correlations and heritabilities. Furthermore, the interpretation is challenging because phenotypes often have different characteristics and units. We propose PLEIO (Pleiotropic Locus Exploration and Interpretation using Optimal test), a summary-statistic-based framework to map and interpret pleiotropic loci in a joint analysis of multiple diseases and complex traits. Our method maximizes power by systematically accounting for genetic correlations and heritabilities of the traits in the association test. Any set of related phenotypes, binary or quantitative traits with different units, can be combined seamlessly. In addition, our framework offers interpretation and visualization tools to help downstream analyses. Using our method, we combined 18 traits related to cardiovascular disease and identified 13 pleiotropic loci, which showed four different patterns of associations.

Genetic evaluation records for the Canadian Holstein, Ayrshire, Jersey, and Brown Swiss bulls and cows born from 1950 and 1960, respectively, were used to study the generation intervals (L) along the four-path selection model. The objectives of the study were to determine the L in the four dairy cattle breeds and the effects of some environmental factors and variations among herds or artificial insemination (AI) studs on the L achieved. Total L of the four selection paths was reduced by 55% from 29.2 yr in 1980 to 13.2 yr in 2016 in the Holstein breed. Substantial reductions in total L were also observed in the Ayrshire, Jersey, and Brown Swiss populations between 1980 and 2016. Unlike progeny year of birth, housing system, agricultural region in Québec (region) — as well as their interaction — were not important on realised L of sires and dams used on Canadian dairy farms. There were significant variations among herds and AI studs on the age of sires and dams used for breeding. The considerable variations in realised L among herds and AI studs offer opportunities to increase the annual rate of genetic progress in the four Canadian dairy cattle populations.

The aims of the present study were to provide a portrait of the techno-economic status of dairy herds in Minas Gerais, Brazil, particularly with respect to bulk-tank somatic cell count (BTSCC) data, and to examine the herd-level associations of BTSCC with various economic performance indicators (EPI). Data from 543 herds, 1,052 herd-year records in total, spread over 3 years (2015-2017), from the South and Southwest mesoregions of Minas Gerais State were provided by the Brazilian Support Agency to Micro and Small Companies Division Minas Gerais (SEBRAE). Herds had an average of 82 lactating cows per herd, milk yield of 17 L/cow per day, and availability of financial information via routine monthly economic surveys. The EPI data (revenue, gross margin, GM; net margin, NM; profit; break-even point; and operational profitability) of each herd was measured monthly by SEBRAE personnel, and herd-year averages of all variables were computed. Bulk-tank data (SCC, total bacterial count, content of crude protein and fat) taken by producers or dairy processors were recorded by SEBRAE personal; and corresponding herd-year averages were calculated and included in the SEBRAE database. There were 209 selected herds, which passed all edit checks, and which had data for all 3 years. The EPI (all expressed on a per-cow basis, $/cow per year) were analyzed, including the effects of region, year, log (ln) BTSCC, production level, and herd size, together with the random effect of herd nested within region. A high proportion of herds (94.6%) presented data records (herd-years) with an average BTSCC > 200 × 103 cells/mL: 37.8% of herd-year records had BTSCC between >200 and ≤400, 14.5% with BTSCC between >400 and ≤500, 25% with BTSCC between >500 and ≤750, and 17.3% with BTSCC >750. For each unit increase in ln BTSCC, revenue declined by $228.5/cow per year, GM by $155.6/cow per year, and profit by $138.6/cow per year. Herds with cows of lower production (

Mastitis is a highly prevalent disease, which negatively affects cow performance, profitability, welfare, and longevity. The objectives of this study were (1) to quantify the impact of the first instance of mastitis, at different stages of lactation, on production and economic performance, and (2) to further quantify the impact of the first instance of mastitis when only cows that remain in the herd for at least 100 d in milk (DIM) and those that remain for 305 DIM are included in the analysis. A retrospective longitudinal study was conducted using data from existing animal health record files and Dairy Herd Improvement records. After editing based on selected inclusion criteria and completeness of health records, data consisted of records from first-lactation Holstein cows, from 120 herds, that calved for the first time between 2003 and 2014, inclusive. Mastitic cows were assigned to 1 of 4 groups based on when in the lactation the first event of mastitis occurred: transition (1-21 DIM), early lactation (22-100 DIM), mid lactation (101-200 DIM), or late lactation (201+ DIM). Mid-lactation and late-lactation mastitic cows were also stratified by cumulative milk yield before the mastitis event. Healthy cows (i.e., no recorded mastitis event) were randomly assigned for each lactation stage, with mid-lactation healthy and late-lactation healthy cows similarly stratified. Production performance (cumulative milk, fat, and protein yield) and economic performance [milk value, margin over feed cost (MOFC), and gross profit] were analyzed using a mixed model with herd as a random effect. Significant losses in cumulative milk yield (-382 to -989 kg) and correspondingly lower fat and protein yields were found in mastitic cows, with transition and late-lactation mastitic cows having the highest losses. Drops in production translated to significant reductions in cumulative milk value (-Can$287 to -Can$591; -US$228 to -US$470), MOFC (-Can$243 to -Can$540; -US$193 to -US$429), and gross profit (-Can$649 to -Can$908; -US$516 to -US$722) for mastitic cows at all stages. Differences between mastitic and healthy cows in the early lactation and transition stages remained for all variables in the 100-DIM analysis, but, aside from gross profit, were nonsignificant in the 305-DIM analysis. Gross profit accounted for all costs associated with mastitis and thus continued to be lower for mastitic cows at all stages, even in the 305-DIM analysis in which culled cows were omitted (-Can$485 to -Can$979; -US$386 to -US$779). The research reflects the performance implications of mastitis, providing more information upon which the producer can make informed culling decisions and maximize both herd profitability and cow longevity.

The goal of this research was to evaluate the microclimate (temperature, relative humidity and ECI-enthalpy comfort index) of commercial loads of broiler chickens at different transport distances: Dist15 (15 km on average) and Dist90 (90 km on average) in the summer and winter seasons and their effects on the production parameters body weight difference (BWD), mortality (%) and bruising prevalence (%). Twelve broiler loads were monitored using dataloggers to record temperature and humidity, with a total of 24 target crates per load. The experiment followed a factorial design [2 seasons (rainy and dry) × 2 distances (Dist15 and Dist90)] with a randomized complete block arrangement, 3 sexes (all males, all females, or mixed shipments) and one shipment per combination. BWD had a heterogeneous distribution throughout the load, and this distribution was not significantly correlated with the mean ECI measured during transport at 12 positions along the load. In terms of comfort, summer is the most critical period for broiler transport. In the interaction between rainy season and Dist90, the highest ECI was scored in the lethal zone (where physiological mechanisms are not enough to control body temperature). Mortality during the rainy season was not significantly different between distances. However, during the dry season, mortality was twice as high as broilers that travelled for 15 km. The prevalence of bruising on carcasses was not affected by the interaction between season and distance. As we know, broiler chicken performance, during transport, can be also related to road conditions, being hard to evaluate the real impact of seasons and distances on animal welfare. Load microclimate can compromise broiler chicken welfare during transport and it does not necessary reflect significant losses pre and post-slaughter.

Crohn's disease (CD) and ulcerative colitis (UC) are the major types of chronic inflammatory bowel disease (IBD) characterized by recurring episodes of inflammation of the gastrointestinal tract. Although it is well established that human leukocyte antigen (HLA) is a major risk factor for IBD, it is yet to be determined which HLA alleles or amino acids drive the risks of CD and UC in Asians. To define the roles of HLA for IBD in Asians, we fine-mapped HLA in 12 568 individuals from Korea and Japan (3294 patients with CD, 1522 patients with UC and 7752 controls). We identified that the amino acid position 37 of HLA-DRβ1 plays a key role in the susceptibility to CD (presence of serine being protective, P = 3.6 × 10-67, OR = 0.48 [0.45-0.52]). For UC, we confirmed the known association of the haplotype spanning HLA-C*12:02, HLA-B*52:01 and HLA-DRB1*1502 (P = 1.2 × 10-28, OR = 4.01 [3.14-5.12]).

Recent genome-wide association studies suggest that the human genetic architecture of complex traits may vary between males and females; however, traditional approaches for association mapping cannot fully account for these between-sex differences... Over the past few years, genome-wide association studies have identified many trait-associated loci that have different effects on females and males, which increased attention to the genetic architecture differences between the sexes. The between-sex differences in genetic architectures can cause a variety of phenomena such as differences in the effect sizes at trait-associated loci, differences in the magnitudes of polygenic background effects, and differences in the phenotypic variances. However, current association testing approaches for dealing with sex, such as including sex as a covariate, cannot fully account for these phenomena and can be suboptimal in statistical power. We present a novel association mapping framework, MetaSex, that can comprehensively account for the genetic architecture differences between the sexes. Through simulations and applications to real data, we show that our framework has superior performance than previous approaches in association mapping.

We evaluated the effects of chronic subclinical mastitis (CSM) caused by different types of pathogens on milk yield and milk components at the cow level. A total of 388 Holstein cows had milk yield measured and were milk sampled three times at intervals of two weeks for determination of SCC and milk composition, and microbiological culture was performed. Cows were considered healthy if all three samples of SCC were ≤200 000 cells/ml and were culture-negative at the third milk sampling. Cows with one result of SCC > 200 000 cells/ml were considered to suffer non-chronic subclinical mastitis whereas cows with at least 2 out of 3 results of SCC > 200 000 cells/ml had CSM. These latter cows were further sorted according to culture results into chronic negative-culture or chronic positive-culture. This resulted in four udder health statuses: healthy, non-chronic, chronicNC or chronicPC. The milk and components yields were evaluated according to the udder health status and by pathogen using a linear mixed effects model. A total of 134 out of 388 cows (34.5%) were chronicPC, 57 cows (14.7%) were chronicNC, 78 cows (20.1%) were non-chronic and 119 cows (30.7%) were considered healthy, which resulted in a grand total of 1164 cow records included in the statistical model. The healthy cows produced more milk than each of the other groups (+2.1 to +5.7 kg/cow/day) and produced higher milk component yields than the chronicPC cows. The healthy cows produced more milk than cows with chronicPC caused by minor (+5.2 kg/cow/day) and major pathogens (+7.1 kg/cow/day) and losses varied from 5.8 to 11.8 kg/cow/day depending on the pathogen causing chronicPC mastitis. Chronic positive-culture cows had a reduction of at least 24.5% of milk yield and 22.4% of total solids yield.

Summary Phosphatidylserine (PS) is an integral component of eukaryotic cell membranes and organelles. The Drosophila genome contains a single PS synthase (PSS)-encoding gene (Pss) homologous to mammalian PSSs. Flies with Pss loss-of-function alleles show a reduced life span, increased bang sensitivity, locomotor defects, and vacuolated brain, which are the signs associated with neurodegeneration. We observed defective mitochondria in mutant adult brain, as well as elevated production of reactive oxygen species, and an increase in autophagy and apoptotic cell death. Intriguingly, glial-specific knockdown or overexpression of Pss alters synaptogenesis and axonal growth in the larval stage, causes developmental arrest in pupal stages, and neurodegeneration in adults. This is not observed with pan-neuronal up- or down-regulation. These findings suggest that precisely regulated expression of Pss in glia is essential for the development and maintenance of brain function. The objective of the present study was to estimate genetic parameters of milk yield (MY), intake traits, type traits, body condition score (BCS), and number of breedings (NOB) in first lactation Canadian Holsteins with a focus on the possibility of using type traits as an indicator of feed intake. Data were obtained from the Canadian Dairy Network and Valacta. A mixed linear model was fitted under REML for the statistical analysis. The multivariate (five traits) model included the fixed effects of age at calving, stage of lactation, and herd-round-classifier for type traits; age at calving, stage of lactation, and herd–year–season of calving (HYS) for BCS; age at calving and HYS for MY, feed intake, and NOB. Animal and residual effects were fitted as random effects for all traits. Estimates of heritabilities for MY, dry matter intake (DMI), angularity, body depth, stature, dairy strength, final score, BCS, and NOB were 0.41, 0.13, 0.24, 0.30, 0.50, 0.30, 0.22, 0.20, and 0.02, respectively. Genetic correlations between type traits and DMI ranged from 0.16 to 0.60. Results indicate that type traits appear to have the potential to predict DMI as a combination/index of two or more traits.

In April 2014, poor fertility in a major commercial goose breeder operation in California triggered the submission of six live affected Toulouse ganders ( Anser anser ) to the California Animal Health and Food Safety Laboratory, Turlock branch (University of California-Davis). Toulouse were principally affected among all breeds, and their egg fertility dropped from 65.7% to less than 33.9% in the first 40 days of the 2014 breeding season. The flock consisted of 410 adult birds, 90 males and 320 females, between 2 and 5 yr of age. Inspection of the flock revealed that 44.4% of the Toulouse ganders had severe phallic deformities that prevented them from mating. At postmortem examination, severe yellowish fibrocaseous exudate disrupted the architecture of the phallus and occasionally produced fistulating tracts through the wall of the organ. Microscopically, multifocal lymphoid nodules were noted in the mucosa and submucosa of the phallus and were associated with extensive granulomatous reaction, intralesional bacteria, and spermatozoa. Mycoplasma spp. were isolated from the phallus of affected and nonaffected birds, and PCR protocols targeting the 16S-23S ribosomal RNA intergenic spacer regions and the RNA polymerase beta subunit gene were performed to identify the isolates. Three distinct species were identified on sequencing and analysis using the National Center for Biotechnology Information basic local alignment search tool: Mycoplasma cloacale , Mycoplasma anseris , and an unknown novel Mycoplasma sp. Additionally, Pasteurella multocida , in combination with other bacteria, was also isolated from the phallic lesions and identified as serotype 3 with a DNA profile of 1511 (National Veterinary Service Laboratory). This is the first report of these Mycoplasma spp. and other bacteria associated with reproductive disease in ganders in the United States.

Necrotic enteritis (NE) produced by Clostridium perfringens is amongst the most prevalent enteric diseases of chickens and turkeys. However, several other bacterial, parasitic and viral agents can cause clinical signs, gross and microscopic lesions in poultry very similar to those of NE and the diseases produced by those agents need to be differentiated from NE. The main differential diagnoses for C. perfringens NE include bacterial (Clostridium colinum, Clostridium sordellii, Clostridium difficile, Pasteurella multocida, Brachyspira spp.), parasitic (Eimeria spp., Histomonas meleagridis) and viral (Duck Herpesvirus type 1, Avian Paramyxovirus type 1) diseases. Confirmation of the diagnosis of these diseases requires identification of the aetiological agents by morphological, cultural and/or molecular methods.

Phosphatidylserine (PS) is an integral phospholipid component of eukaryotic cell membranes and organelles. The Drosophila genome contains a single PS synthase-encoding gene (Gongpo, Gong for short) with a protein sequence homologous to mammalian PS synthases. In this study, we characterize mutant Gong phenotypes. Adult flies with trans-heterozygous and heterozygous mutations for Gong loss-of-function alleles had reduced life span, increased bang sensitivity, locomotor defects, and increased formation of vacuoles in the brain, a sign associated with neurodegenerative disorders. Similar phenotypes were observed with pan-glial-specific knockdown/overexpression of Gong, but not with pan-neuronal manipulations. We observed defective mitochondria in the mutant adult brain, elevated production of reactive oxygen species, and increased autophagy and apoptotic cell death. These findings suggest that proper expression of Gong in glia is essential for the development and maintenance of brain function. We propose a new mechanism for neurodegenerative disease, triggered by defective PS metabolism in the nervous system.

The flagellate parasite Histomonas meleagridis causes a severe disease in turkeys. Since nitarsone, the last antiflagellate feed additive, was withdrawn from the market, H. meleagridis has gained increasing clinical and economic importance, and frequently entire turkey flocks are lost to the disease. Even before the antiflagellate market withdrawal, H. meleagridis has been recognized as a noteworthy disease in commercial turkeys. The aim of this study was to compile and analyze data on histomoniasis outbreaks diagnosed by the California Animal Health and Food Safety System in the years 2000 through 2014 while nitarsone was still available. Sixty-two cases were included in the study. In all cases, the parasite was detected by histopathology. Five cases were from breeder flocks, 44 from commercial meat turkey flocks, and 13 from other flocks such as backyard flocks or pet turkeys. There were between one and eight cases each year with no clear trend over time. Most cases occurred in the warmer months between April and October with a clear peak in August. Turkeys in the flocks with the disease were aged between 2 wk and 15 mo, with a median age of 9 wk. In cases for which mortality was reported, daily mortality ranged between 0.04% and 2% with an average of 0.34% and a median of 0.15%. Five-week mortality in infected houses on three infected ranches was between 1.3% and 68%. In 12 cases, the parasite was observed in organs other than ceca and liver. These included spleen, kidney, bursa of Fabricius, proventriculus, lung, pancreas, and crop. In 58 cases, histomoniasis was considered the most significant and primary diagnosis; in four cases histomoniasis was regarded as an incidental finding. In 14 other outbreaks, detailed information was available. In 11 cases, more than one house on the ranch was affected; in five cases the disease spread to all houses. Total mortality in turkeys from affected flocks ranged between 12% and 65%. In eight flocks, the birds were marketed with a weight that was considered too light. In conclusion, the present study shows that even with nitarsone available as antiflagellate feed, additive histomoniasis was a serious disease of turkeys in the United States with an epidemiology similar to that observed in Europe.

The objective of this study was to examine the effect of feeding systems [component and total mixed rations (TMR)] and dietary grain sources (barley, commercial concentrate, corn grain, and high-moisture corn) on lactation characteristics and milk composition. A total of 852,242 test-day records, information on animal characteristics, feed composition, and feeding systems from 104,129 Holstein cows in 4,319 herds covering a period of 5 yr were obtained from Quebec's Dairy Herd Improvement Association (Valacta). We performed descriptive statistics and graphical representations of the data for each type of feeding system and grain source by parity (1 to 3). The milk records were binned in 15-d in milk blocks. Mixed models using a combination of forward and backward stepwise selections were developed to predict milk and milk component yields. The TMR-fed cows had greater yield of milk, fat, protein, and lactose and lower milk urea N (MUN) concentration than component-fed cows at all parities. Cows fed a TMR had higher peak milk yields and greater persistency after peak lactation compared with component-fed cows. In addition, greater yields of milk fat and protein from peak to mid-lactation were found in TMR- versus component-fed cows. In general, greater milk fat and protein yields as well as lower MUN concentration were observed in cows fed corn grain or high-moisture corn compared with barley or commercial concentrate, but parity influenced these relationships. The feeding system by day in milk blocks interaction was significant in models of milk and components yields for all parities, but only for second-lactation cows for MUN concentration. This means that effect of TMR and component feeding differs with stage of lactation. In conclusion, feeding TMR and corn-based diets are associated with greater yield of milk and milk components under commercial conditions.

peer-reviewed The reduction of milk production caused by subclinical mastitis in dairy cows was evaluated through the regression of test-day milk yield on log-transformed somatic cell counts (LnSCC). Official test-day records (n = 1,688,054) of Holstein cows (n = 87,695) were obtained from 719 herds from January 2010 to December 2015. Editing was performed to ensure both reliability and consistency for the statistical analysis, and the final data set comprised 232,937 test-day records from 31,692 Holstein cows in 243 herds. A segmented regression was fitted to estimate the cutoff point in the LnSCC scale where milk yield started to be affected by mastitis. The statistical model used to explain daily milk yield included the effect of herd as a random effect and days in milk and LnSCC as fixed effects regressions, and analyses were performed by parity and stage of lactation. The cutoff point where milk yield starts to be affected by changes in LnSCC was estimated to be around 2.52 (the average of all estimates of approximately 12,400 cells/mL) for Holsteins cows from Brazilian herds. For first-lactation cows, milk losses per unit increase of LnSCC had estimates around 0.68 kg/d in the beginning of the lactation [5 to 19 d in milk (DIM)], 0.55 kg/d in mid-lactation (110 to 124 DIM), and 0.97 kg/d at the end of the lactation (289 to 304 DIM). For second-lactation cows, milk losses per unit increase of LnSCC had estimates around 1.47 kg/d in the beginning of the lactation (5 to 19 DIM), 1.09 kg/d in mid-lactation (110 to 124 DIM), and 2.45 kg/d at the end of the lactation (289 to 304 DIM). For third-lactation cows, milk losses per unit increase of LnSCC had estimates around 2.22 kg/d in the beginning of the lactation (5 to 19 DIM), 1.13 kg/d in mid-lactation (140 to 154 DIM), and 2.65 kg/d at the end of the lactation (289 to 304 DIM). Daily milk losses caused by increased LnSCC were dependent on parity and stage of lactation, and these factors should be considered when estimating losses associated with subclinical mastitis.

The objective of the present study was to estimate heritabilities of milk fatty acids (FA) and genetic and phenotypic correlations among milk FA and milk production traits in Canadian Holsteins. One morning milk sample was collected from each of 3,185 dairy cows between February and June 2010 from 52 commercial herds enrolled in Valacta (Ste-Anne-de-Bellevue, Quebec, Canada). Individual FA percentages (g/100 g of total FA) were determined for each sample by gas chromatography. After editing the data, 2,573 cows from 46 herds remained. Genetic parameters were estimated using multitrait animal models fitted under REML. The model included fixed effects of age at calving and stage of lactation each nested within parity and random effects of herd-year-season of calving, animal, and residual. The pedigree of animals with data was traced back 5 generations on both the male and female sides to account for relationships among animals. The estimates of heritability for individual FA ranged from 0.01 to 0.39, with standard errors ranging from 0.01 to 0.06. Generally, monounsaturated FA (MUFA) and saturated FA (SFA) showed higher heritability estimates than polyunsaturated FA (PUFA). Overall, SFA were negatively genetically correlated with MUFA and PUFA, whereas genetic correlations between MUFA and PUFA were positive. The SFA showed positive associations with fat yield and fat percentage, whereas unsaturated FA were negatively associated with fat yield and fat percentage. Bovine milk FA composition could be improved through genetic selection, and selection for MUFA or against SFA could alter the bovine milk fat profile in a desirable direction.

Several health disorders, such as milk fever, displaced abomasum, and mastitis, as well as impaired reproductive performance, are known risk factors for the removal of affected cows from a dairy herd. While cow-level risk factors are well documented in the literature, herd-level associations have been less frequently investigated. The objective of this study was to investigate the effect of cow- and herd-level determinants on variations in culling risk in Quebec dairy herds: whether herd influences a cow's culling risk. For this, we assessed the influence of herd membership on cow culling risk according to displaced abomasum, milk fever, and retained placenta. A retrospective longitudinal study was conducted on data from dairy herds in the Province of Quebec, Canada, by extracting health information events from the dairy herd health management software used by most Quebec dairy producers and their veterinarians. Data were extracted for all lactations starting between January 1st and December 31st, 2010. Using multilevel logistic regression, we analysed a total of 10,529 cows from 201 herds that met the inclusion criteria. Milk fever and displaced abomasum were demonstrated to increase the cow culling risk. A minor general herd effect was found for the culling risk (i.e. an intra-class correlation of 1.0% and median odds ratio [MOR] of 1.20). The proportion of first lactation cows was responsible for this significant, but weak herd effect on individual cow culling risk, after taking into account the cow-level factors. On the other hand, the herd's average milk production was a protective factor. The planning and management of forthcoming replacement animals has to be taken into consideration when assessing cow culling risks and herd culling rates.

The relationship between cows' health, reproductive performance or disorders and their longevity is well demonstrated in the literature. However these associations at the cow level might not hold true at the herd level, and herd-level variables can modify cow-level outcomes independently of the cows' characteristics. The interaction between cow-level and herd-level variables is a relevant issue for understanding the culling of dairy cows. However it requires the appropriate group-level variables to assess any contextual effect. Based on 10 years of health and production data, the objectives of this paper are:(a) to quantify the culling rates of dairy herds in Quebec; (b) to determine the profiles of the herds based on herd-level factors, such as demographics, reproduction, production and health indicators, and whether these profiles can be related to herd culling rates for use as potential contextual variables in multilevel modelling of culling risk. A retrospective longitudinal study was conducted on data from dairy herds in Quebec, Canada, by extracting health information events from the dairy herd health management software used by most Quebec producers and their veterinarians. Data were extracted for all lactations taking place between January 1st, 2001 and December 31st, 2010. A total of 432,733 lactations from 156,409 cows out of 763 herds were available for analysis. Thirty cow-level variables were aggregated for each herd and years of follow-up, and their relationship was investigated by Multiple Factor Analysis (MFA). The overall annual culling rate was 32%, with a 95% confidence interval (CI) of [31.6%,32.5%]. The dairy sale rate by 60 days in milk (DIM) was 3.2% [2.8%,3.6%]. The annual culling rate within 60 DIM was 8.2% [7.9%,8.4%]. The explained variance for each axis from the MFA was very low: 14.8% for the first axis and 13.1% for the second. From the MFA results, we conclude there is no relationship between the groups of herd-level indicators, demonstrating the heterogeneity among herds for their demographics, reproduction and production performance, and health status. However, based on Principal Component Analysis (PCA), the profiles of herds could be determined according to specific, single, herd-level indicators independently. The relationships between culling rates and specific herd-level variables within factors were limited to livestock sales, proportion of first lactation cows, herd size, proportion of calvings occurring in the fall, longer calving intervals and reduced 21-day pregnancy rates, increased days to first service, average age at first calving, and reduced milk fever incidence. The indicators found could be considered as contextual variables in multilevel model-building strategies to investigate cow culling risk.

There is growing evidence of shared risk alleles for complex traits (pleiotropy), including autoimmune and neuropsychiatric diseases. This might be due to sharing among all individuals (whole-group pleiotropy) or a subset of individuals in a genetically heterogeneous cohort (subgroup heterogeneity). Here we describe the use of a well-powered statistic, BUHMBOX, to distinguish between those two situations using genotype data. We observed a shared genetic basis for 11 autoimmune diseases and type 1 diabetes (T1D; P1 × 10(-4)) and for 11 autoimmune diseases and rheumatoid arthritis (RA; P1 × 10(-3)). This sharing was not explained by subgroup heterogeneity (corrected PBUHMBOX0.2; 6,670 T1D cases and 7,279 RA cases). Genetic sharing between seronegative and seropostive RA (P1 × 10(-9)) had significant evidence of subgroup heterogeneity, suggesting a subgroup of seropositive-like cases within seronegative cases (PBUHMBOX = 0.008; 2,406 seronegative RA cases). We also observed a shared genetic basis for major depressive disorder (MDD) and schizophrenia (P1 × 10(-4)) that was not explained by subgroup heterogeneity (PBUHMBOX = 0.28; 9,238 MDD cases).

Recent genome-wide association studies have identified more than 200 regions that affect susceptibility to inflammatory bowel disease (IBD). However, identified common variants account for only a fraction of IBD heritability and largely have been identified in populations of European ancestry. We performed a genome-wide association study of susceptibility loci in Korean individuals, comprising a total of 1505 IBD patients and 4041 controls. We identified 2 new susceptibility loci for IBD at genome-wide significance: rs3766920 near PYGO2-SHC1 at 1q21 and rs16953946 in CDYL2 at 16q23. In addition, we confirmed associations, in Koreans, with 28 established IBD loci ( P -4 ). Our findings support the complementary value of genetic studies in different populations.

Between April 2013 and April 2015, seven flocks belonging to three different major commercial egg producers inCalifornia experienced a mild increase in mortality 2 to 3 wk after administration of Salmonella Enteritidis bacterins. Strains of chickens involved were H&N (flock A1, A2, B2, C1, C2, and C3) and Lohmann white (flock B1). Vaccination was administered individually through injection either in the breast muscles or subcutis in the legs between 11 and 18 wk of age in all flocks. Clinical signs ranged from inapparent to lameness, reluctance to walk, greenish diarrhea, and retching-like symptoms. The mortality ranged from 0.16% to 1.38% per week, with the highest peaks occurring usually 2 to 3 wk postvaccination, and then declined rapidly. Postmortem examinations revealed enlarged livers with disseminated hemorrhages and pale foci of necrosis. Also, severe extensive hemorrhages in the intestine, heart, and proventriculus were observed in a few birds. Various degrees of productive, exudative giant cell granulomatous myositis were observed invading deeply the muscles and subcutis at the site of vaccination. The myositis was always associated with optically empty vacuoles positive for neutral lipids by Oil Red O stain. Droplets of Oil Red O material were also noticed in the affected livers and intestines. Congo red stain highlighted the presence of amyloid in moderate to severe amounts in the breast muscles and moderate amounts in livers, spleens, and intestines. Salmonella antigens were detected in the injection sites and livers by immunohistochemical staining. No viruses or toxic substances were recovered from the liver, spleen, intestine, and pectoral muscles, and the few bacteria isolated were interpreted as secondary postmortem invaders. In addition, livers and bile tested for hepatitis E virus were negative by reverse-transcriptase polymerase chain reaction.

Trichodinid ciliophorans are opportunistic parasites of many species of fish, amphibians, and molluscs, but yet never reported in association with lesions in birds. Postmortem and histopathological evaluation of a commercial adult Toulouse gander and female goose, and a wild Mallard drake revealed the presence of severe pathological parasitic colonization of their reproductive tracts. Histopathological findings included moderate to severe granulocytic inflammation, acanthosis, accentuation of the rete pegs, and proliferative hyperplastic squamous metaplasia of the mucosa of the ejaculatory ducts and groove, sulcus spermaticus, glandular part of the phallus (cavum penis), and oviduct in association with large numbers of ciliated protozoa anchored to the tissues or free in the lumen. These protozoa had characteristic morphological features analogous to the family of Trichodinidae. The source of this parasitism could not be determined. To our knowledge, this is the first report of trichodinosis associated with pathology in birds.

A total of 850 Holstein cows from 13 commercial dairy herds were involved in the present study to compare the effects of 2 different dry period (DP) management strategies on health and reproductive parameters. Cows were assigned to either a short (SDP; 35-d) or a conventional (CDP; 60-d) DP management within each herd, based on previous 305-d milk yield, parity (414 primiparous and 436 multiparous), and estimated calving interval. Cows assigned to CDP were fed a dry cow ration from dry-off until 21 d prepartum, and were then switched to a precalving ration. Cows assigned to SDP were fed the precalving ration throughout their DP. Rations were specific to each herd. A significant treatment × parity interaction was found for culling rate. Dry period management did not affect culling rate for second-lactation cows but a significantly higher culling rate occurred in multiparous CDP cows compared with SDP (42.6 vs. 31.6% ± 3.7 for CDP and SDP, respectively). Management used in the DP did not affect incidence of severe ketosis, displaced abomasum, milk fever, and mastitis, although incidence of these metabolic disorders were lower in second-lactation than third- or greater-lactation cows. The incidence of mild ketosis (evaluated by milk ketone concentration) was lower following SDP, probably as a result of better energy balance. On the other hand, the incidence of retained placenta was higher in multiparous cows assigned to SDP, but the reason for this increase remains unclear. Nevertheless, this did not lead to increased incidence of metritis. Moreover, DP management did not influence reproductive measures, including days in milk at first breeding, number of breedings per conception, as well as conception rates at first and second services. Regarding days open, overall, all 13 herds were not significantly affected by treatment, but 1 herd clearly showed opposite results to the 12 others. Our results indicate that a short DP management strategy could facilitate transition from one lactation to the next by decreasing the incidence of mild ketosis, with no major negative effects on other health parameters and reproduction. The variation in results observed among herds suggests that other management practices influence the response observed following a short or conventional DP, emphasizing the need for other field studies.

A high-performance liquid chromatography (HPLC) unit equipped with size exclusion column and a refractive index detector was used for simultaneous monitoring, identification, and quantitation of the reaction components from lipase-catalyzed transesterification of three oils. The procedure simultaneously separated and detected the unreacted triacylglycerols (TAG), diacyl-, and monoacyl-glycerol (DAG and MAG) co-products, residual alcohol as well as free fatty acid (FFA) based on retention times. During the routine histologic evaluation of an outbreak of inclusion body hepatitis (IBH) in Mississippi broilers, a high incidence of renal enlargement and glomerulonephropathy was observed in the birds presenting classic hepatic pathology. Characteristic intranuclear adenoviral inclusion bodies were demonstrated in the livers of these birds, and fowl adenovirus was identified by viral isolation and by PCR. The glomerular lesions were consistent with proliferative or membranoproliferative forms of glomerulonephritis. Histomorphometric evaluations were performed to generate a more quantitative analysis of altered glomerular size and cellularity, to detect statistically significant borderline changes, and to get a clearer insight into the incidence of the glomerular alterations. Marked increases in both the average glomerular size (area) and the total glomerular cellularity were observed for the affected glomeruli relative to normal controls. The average glomerular area values for normal glomeruli in the peripheral subcapsular cortical and central cortical kidney regions were 1791 microm2 and 5302 microm2, respectively. In contrast, glomerular measurements for kidneys exhibiting glomerulonephritis by routine histopathology, had average values for the two regions of 4429 microm2 and 11,063 microm2. The average glomerular cell counts for the two regions in controls were 44 and 107 cells/ glomeruli, while averages for birds with glomerulonephritis were 85 and 193 cells/glomeruli. The proportion of IBH-associated glomeruli greater than two standard deviations above the mean glomerular size of the normal controls was 52% for the central region and 62% for the peripheral region.

The reduction in milk production caused by subclinical mastitis in dairy cattle was assessed through the regression of test day milk yield on log-transformed somatic cell counts (LnSCC). Data was obtained from Valacta, Quebec, and a total of 312,756 test day records from Ayrshire cows and 1,869,785 test day records from Holstein cows were included in the analyses. A segmented regression was fitted to estimate the cutoff point in the LnSCC scale where milk yield starts to be affected by mastitis. The statistical model used to explain daily milk yield included the effects of herd–year-season of test (random), days in milk, age at calving and LnSCC, and analyses were performed by breed, parity and stage of lactation. The cutoff point where milk yield starts to be affected by changes in LnSCC was estimated from data to be around 2 (approximately 7400 cells/mL) for Canadian Ayrshires and Holsteins. Milk losses per unit increase in LnSCC varied from 0.55 to 0.84 kg/day in first lactation Ayrshires, from 0.33 to 0.55 kg/day in first lactation Holsteins, from 0.74 to 2.45 kg/day in adult Ayrshires and from 0.77 to 1.78 kg/day in adult Holsteins. The objective of this study was to characterize body condition score (BCS) in dairy cattle recorded under commercial conditions in Quebec. There were 354 958 BCS records from Holstein cows and 15 334 records from Ayrshire cows available, from first to fifth parity, recorded by producers using a scale of 1 to 5. A classification model was fitted including fixed effects of herd, year of calving, month of calving, age at calving, and days in milk (DIM); another model substituted the Wilmink function on days in milk to model lactational BCS curves. Both models used the spatial power covariance structure to account for correlation between BCS recorded on the same cow along the lactation and were fitted using the Mixed procedure of SAS software (version 9.1.3). Body condition score was significantly affected by month of calving, age at calving and DIM. Average BCS (SD) for Holstein cows from first to fifth lactation were: 2.95 (0.46), 2.90 (0.53), 2.93 (0.56), 2.94 (0.57), and 2.93 (0.57), respectively; the estimates for Ayrshire cows from first to fifth lactation were: 2.99 (0.47), 3.06 (0.54), 3.15 (0.56), 3.11 (0.57), 3.13 (0.58). Lactation curves for BCS were generated for each breed and parity. The routine collection of BCS in individual cows is recommended to ensure close monitoring of energy balance during the lactation. The BCS information analyzed in this study is suitable for modeling changes throughout the lactation with the application of the Wilmink function. The entire sequence of the pig 3β-hydroxysteroid dehydrogenase (3β-HSD) gene has recently become known. This gene is deemed to be important in androstenone metabolism in pig liver, and its defective expression has been shown to be related to androstenone accumulation in adipose tissue and the development of boar taint. The aim of the present work was to do the following: 1) define the structure of the pig 3β-HSD gene and 2) compare 3β-HSD DNA sequences from pigs of different breeds, which vary in adipose tissue androstenone levels, with the purpose of identifying a polymorphism that might be responsible for differential 3β-HSD expression. The 5′ flanking and the coding region of 3β-HSD were cloned and sequenced by conventional techniques. The 3β-HSD coding regions were identical in pigs of different breeds and in animals with high and low androstenone levels. Significant sequence variations were found in the 5′ flanking region of the 3β-HSD gene, where differences in the number of TTAT repeats and 3 SNP were observed. The SNP were associated with the number of the TTAT repeats. These variations in the DNA sequence of the 3β-HSD gene were not associated with the androstenone level in s.c. adipose tissue but were breed-dependent. The results of this work might be used for detection of the presence of Meishan genes in Western pig breeds, especially if the phenotype is not clearly established. © 2007 American Society of Animal Science. Only bacteria can synthesize vitamin B12, and this requires adequate Co supply. The natural source of vitamin B12 in human diets comes from animal products, especially those from ruminants. This study aimed to describe variability regarding vitamin B12 concentration in milk among and within commercial dairy herds in early lactation. A secondary objective was to explore potential causes for this variability such as genetic variation and diet characteristics. In total, 399 dairy cows (135 primiparous and 264 multiparous; 386 Holstein and 13 Jersey cows) in 15 commercial herds were involved. Milk samples were taken at 27.4±4.1 and 55.4±4.1d in milk. Neither parity (primiparous vs. multiparous) nor sampling time affected milk concentrations of vitamin B12. Nevertheless, vitamin B12 concentration in milk was highly variable among and within dairy herds. The lowest vitamin B12 concentration in milk of cows was observed in the Jersey herd. Among herds, vitamin B12 concentration in milk ranged from 2,309 to 3,878 pg/mL; one glass (250mL) of milk from those herds would provide between 23 and 40% of the vitamin B12 recommended daily allowance. Among individual cows, however, this provision varied between 16 and 57% of the recommendation. In spite of the limited size of the studied population, the heritability value was 0.23, suggesting that genetic selection could modify milk vitamin B12 concentration. We observed a positive relationship between milk vitamin B12 concentration and dietary acid detergent fiber content and a negative relationship between milk concentration of vitamin B12 and dietary crude protein content.

The objective was to provide an overview of heifer growth in Quebec and analyze its relationship with age at first calving. Heifer body weights, wither heights, body condition scores, and breeding events collected from 1993 to 2003 were obtained and combined with first lactation data, mature body weights, type classification final score and frame/capacity, and estimated breeding values for milk, conformation, frame/capacity, and Lifetime Profit Index. Analyses were restricted to 39 animal-level variables. The resulting data set consisted of 44 989 Holsteins and 2294 Ayrshires with data available for at least one variable. The average age at first calving was 26.5 and 27.1 mo for Holsteins and Ayrshires, respectively. For Holsteins, average daily gains in body weights were 0.79, 0.89, 0.87, and 0.70 kg d-1 for 0 to 4 mo (birth to weaning), 4 to 10 mo (to target onset of puberty), 10 to 14 mo (to first breeding), and 14 mo to pre-calving periods of growth, respectively. Body weights at 14 mo, conception, and post calving were 398, 464, and 601 kg, respectively, and relative to mature body weight were 59, 68 and 89%, respectively. Correlations with age at first calving, corrected for herd and year of birth effects, were positive for body weight at conception (0.69), body weight at first calving (0.31), and number of breedings (0.37), and negative for the average daily gain from conception to post calving (-0.38) and body weight at 14 mo relative to mature body weight (-0.20). For Ayrshires, body weights and average daily gains were lower than for Holsteins, but body weights relative to mature body weight and correlations were generally similar. Holsteins and Ayrshires calved substantially earlier and at a heavier body weight compared with Quebec data from the 1980s. The results of this study suggest that heifer growth may not be a limiting factor in reducing the average age at first calving in Québec. Key words: Dairy cattle, field-recorded data, heifer, growth

Survival analysis was used to study the effects of composite and descriptive linear type traits on functional herd life of Quebec Holsteins. Functional herd life was defined as the length of life from first calving to death, culling, or censoring, and was adjusted for 305-d milk production. The dataset contained information from 331,105 cows from Quebec province calving for the first time between 1981 and 1995; 58% of the records had type information. Weibull models were fitted to analyze the data. The hazard function was described as the product of a baseline hazard function and the time-independent effects of age at first calving and type, and the time-dependent effects of year of calving, stage of lactation x lactation number, annual change in herd size, 305-d milk production, and herd-year (random). Analyses were done one at a time for each type trait. The strongest relationships between survival and composite type traits were found for final score, mammary system, and feet and legs. Among the linear type traits, the highest impact on functional herd life was found for traits related to the udder.

A clonal variant of serotype M1 group A streptococcus, strain 90-131, disseminated to several continents, where it was associated with severe systemic infections and toxic shock. Although this strain harbours the speA gene and is efficiently internalized by human epithelial cells, clinical isolates often fail to express the erythrogenic toxin under laboratory growth conditions. Cultures of strain 90-131 were observed to phase vary between small, dry, compact and larger, more mucoid colonies. The former were shown to be poorly internalized by epithelial cells. Analysis of RNA by Northern hybridization demonstrated that the emm1, hasA and speA genes were weakly transcribed in cultures derived from the small colonies and highly transcribed in those derived from the large colonies. An insertion mutation in mga (the multigene activator) downregulated the invasion of epithelial cells and the transcription of emm1 and hasA, but had little impact on the transcription of speA. These are the first data to suggest the existence of a common regulatory circuit linking intracellular invasion, M protein, hyaluronic acid capsule and erythrogenic toxin expression by group A streptococcus. Moreover, the genetic instability of toxin expression exhibited by this serotype may impact on laboratory studies that attempt to associate toxin production with toxic shock.

In Quebec first calving occurs on average at 27 mo, whereas the target is 23 to 24.5 mo to maximize herd profitability. The aim of this study was to quantify current and future heifer growth using individual heifer random regressions and to generate indicators (such as heifer weight and height at 15 and 24 mo, average daily gain before and after 15 mo, age at which optimal weight for breeding is attained, i.e., 55% of mature weight, and reliability of the 15- and 24-mo weight predictions) that could be used as a practical on-farm tool. Dairy heifer weight estimated by heart girth circumference and height measured at the withers (from 0 to 27 mo) were obtained from the Valacta database (DHI agency, Ste-Anne-de-Bellevue, QC, Canada) from 1995 to 2012. Indicators were calculated based on the current situation of Holstein (HO), Ayrshire (AY), Jersey (JE), and Brown Swiss (BS) heifer growth in Quebec. Heifers with less than 2 records were excluded from the analysis. Mature weights were determined by weight at calving of cows from third or greater lactation for a given breed and were 710 kg for HO, 625 kg for AY, 470 kg for JE, and 670 kg for BS. Estimated weights at 15 and 24 mo were 425 and 627, 334 and 482, 297 and 429, and 379 and 560 kg for HO, AY, JE, and BS, respectively, which are heavy enough for breeding and calving, except for AY. Relative reliabilities of the 15- and 24-mo weight predictions were on average 89 and 60%, respectively, based on measurements up to 15 mo. For HO, AY, JE, and BS, wither heights at 15 and 24 mo were 134 and 143, 125 and 134, 122 and 131, and 130 and 140 cm, respectively. Age at optimal breeding weight was 13.6, 15.5, 12.6, and 14.5 mo for HO, AY, JE, and BS, respectively. These data suggest that it is realistic to expect a first calving at 24 mo for HO, JE, and BS. A growth delay was observed for AY; average daily gain was 655 and 538 g/d before and after 15 mo, respectively. The average daily gain before and after 15 mo was 848 and 747 g/d for HO, 603 and 486 g/d for JE, and 775 and 662 g/d for BS, respectively. These indicators could be calculated for an individual heifer and on a herd-level basis and used on farm as a management tool for reducing age at first breeding and at first calving.

Survival analysis methodologies were used to study herd life in Canadian Holstein cows. Herd life was defined as true herd life or the length of time between first calving and censoring. True herd life adjusted for 305-d milk production was defined as functional herd life. Lifetime record (censored or completed) were from 331,147 Holstein cows registered in the Programme d'Analyse des Troupeaux Laitiers du Québec (PATLQ) that calved for the first time between March 1, 1981 and March 31, 1995. The Weibull (proportional hazards) model used to analyze true herd life and functional herd life contained a Weibull baseline hazard function and the time-dependent effects of year of first calving, lactation number by stage of lactation, annual change in herd size and herd-year (random), and the time-independent effects of the milk recording option (supervised or not) and age at first calving. The model for functional herd life included also the time-dependent effect of herd-year-parity class of 305-d milk production. Genetic differences between sires with regard to the hazard function of their daughters was clearly demonstrated. The hazard rate followed a different pattern in later lactations, particularly in the first 240 d in milk. Older age at first calving was found to be associated with higher risks of culling. Changes in herd size had a small impact on the hazard function of animals. The hazard decreased as production of the cow increased. Heritability in the log scale was 0.09 for true herd life and 0.08 for functional herd life, but when heritability was expressed on the original scale, the estimates for the two traits were 0.19 and 0.15, respectively. The difference in the median survival between a bull with an estimated transmitting ability of 0.6 and another bull with an estimated transmitting ability of 1.3 was 690 d or 1.7 lactations. Rank correlations between the official estimated transmitting abilities for true herd life and functional herd life and those obtained in this study were 0.62 and 0.66, respectively.

A clonal variant of serotype M1 group A streptococcus (designated M1inv+) has been linked to severe and invasive infections, including sepsis, necrotizing fasciitis and toxic shock. High frequency internalization of cultured epithelial cells by the M1inv+ strain 90-226 is dependent upon the M1 protein. Invasion of HeLa cells was blocked by an anti-M1 antibody, invasion by an M1- strain (90-226 emm1::km) was greatly reduced, and latex beads bound to M1 protein were readily internalized by HeLa cells. Beads coated with a truncated M1 protein were internalized far less frequently. Scanning electron microscopy indicated that streptococci invade by a zipper-like mechanism, that may be mediated by interactions with host cell microvilli. Initially, internalized streptococci and streptococci undergoing endocytosis are associated with polymerized actin. Later in the internalization process, streptococcal-containing vacuoles are associated with the lysosomal membrane glycoprotein, LAMP-1.

The M1inv+ subclone of M1 group A streptococci that spread globally in the late 1980s and early 1990s was previously identified by restriction fragment length polymorphism (RFLP), M protein, and SpeA exotoxin sequence analyses. Strains representing this subclone were characterized with regard to carriage of bacteriophage and capacity to invade cultured human epithelial cells. The M1inv+ subclone was found to harbor two entirely different prophages, phage T13 and phage T14, which together supplement its genome with nearly 70 kb of DNA. Phage T14 encodes the SpeA exotoxin and is closely related to the classic converting phage T12. Plaque-forming characteristics and RFLP analyses of phages T13 and T14 were compared to each other and to phage T12. Other subclones of M1, isolated in the 1970s to the early 1980s, lacked both prophages. The M1inv+ subclone was previously reported to be efficiently internalized by human epithelial cells. This potential was confirmed and expanded by comparing a variety of clinical isolates. The capacity for high-frequency invasion of epithelial cells was not transmitted to a laboratory strain of group A streptococci by the above-mentioned bacteriophages.

Termination of packaging of the λ chromosome involves completion of translocation of the DNA into the head shell, and conversion of the translocation complex into a cleavage complex. The cleavage reaction introduces staggered nicks into the downstream cosN to generate the right cohesive end of the chromosome. cosQ, a site adjacent to cosN, was found to be required for nicking the bottom strand of cosN; bottom strand nicking was also sequence-specific for bps at the nick site. Nicking of the top strand of cosN (cosNL) was stimulated by cosQ, but fidelity and efficiency of cosNL nicking were largely dictated by other cos subsites (i.e. cosB and I2). Aberrant top-strand cleavage within cosQ was observed in the absence of I2, and nicking at a site 8 nt 5′ to the normal cosNL nick site occurred in the absence of cosB. The presence of cosQ was found to be insufficient to arrest DNA translocation in vivo, indicating that cosQ, per se, is not a packaging stop signal. A model is presented in which the role of cosQ is to depolarize the asymmetric arrangement of terminase protomers in the translocation complex so that protomers are configured to match the 2-fold rotational symmetry of cosN.

A total of 206 963 official lactation records and 128 182 owner sampler lactation records from the Programme d'Analyse des Troupeaux Laitiers du Québec herds were used to estimate survival rates of Holstein cows up to different parities by milk recording option and year of first calving. Survival up to a given parity was computed only for cows having the opportunity to reach that parity. Average productive herd life up to and including the eighth parity was computed for cows calving for the first time from 1981 to 1985. The overall productive herd life was 33.13 mo for owner sampler herds and 32.97 mo for official herds, and the respective replacement rates per year were 36.22% and 36.40%. Average herd composition in Quebec Holstein herds was: 33.4% of cows in parity 1, 24.2% in parity 2, 17.1% in parity 3, 11.4% in parity 4, 7.0% in parity 5, 3.9% in parity 6, 1.9% in parity 7, and 1.1% in parity 8. Cows in official herds had longer calving intervals than those in owner sampler herds. First calf heifers have been culled by official producers earlier in lactation than by owner sampler producers. The linear double-stranded DNA molecules in lambda virions are generated by nicking of concatemeric intracellular DNA by terminase, the lambda DNA packaging enzyme. Staggered nicks are introduced at cosN to generate the cohesive ends of virion DNA. After nicking, the cohesive ends are separated by terminase; terminase bound to the left end of the DNA to be packaged then binds the empty protein shell, i.e., the prohead, and translocation of DNA into the prohead occurs. cosB, a site adjacent to cosN, is a terminase binding site. cosB facilitates the rate and fidelity of the cosN cleavage reaction by serving as an anchoring point for gpNu1, the small subunit of terminase. cosB is also crucial for the formation of a stable terminase-DNA complex, called complex I, formed after cosN cleavage. The role of complex I is to bind the prohead. Mutations in cosB affect both cosB functions, causing mild defects in cosN cleavage and severe packaging defects. The lethal cosB R3- R2- R1- mutation contains a transition mutation in each of the three gpNu1 binding sites of cosB. Pseudorevertants of lambda cosB R3- R2- R1- DNA contain suppressor mutations affecting gpNu1. Results of experiments that show that two such suppressors, Nu1ms1 and Nu1ms3, do not suppress the mild cosN cleavage defect caused by the cosB R3- R2- R1- mutation but strongly suppress the DNA packaging defect are presented. It is proposed that the suppressing terminases, unlike the wild-type enzyme, are able to assemble a stable complex I with cosB R3- R2- R1- DNA. Observations on the adenosine triphosphatase activities and protease susceptibilities of gpNu1 of the Nu1ms1 and Nu1ms3 terminases indicate that the conformation of gpNu1 is altered in the suppressing terminases.

First lactation classification records for 17 non-production traits in New Zealand dairy cows, and survival from first to second and to third lactation were analyzed in order to estimate the correlations amongst the 17 traits and with cow survival. Four of the traits were farmer-scored, 13 were scored by inspectors. The data were collected on heifers calving between 1987 and 1991, to allow expression of survival through to 3rd lactation. Analyses were carried out by breed of sire (of cow), for Holstein, Ayrshire and Jersey sires and combined, with breed and heterosis effects included. Approximately 20% of the cows were crossbred, mainly combinations of Jersey and Holstein. The 17 traits were scored on a 1 to 9 scale; a Snell transformation of the scores was used for the analyses using mixed model REML methods. The overall rate of survival to 2nd lactation was 0.88 and to 3rd lactation was 0.74. The genetic correlations between the producer-scored traits and survival to 2nd and to 3rd lactations were mainly in the range 0.3 to 0.6 whereas the genetic correlations between the inspector-scored, ‘conformation’ traits were not different from zero. Heritabilities for the producer-scored traits ranged from 0.1 to 0.37, for the inspector-scored traits heritabilities ranged from 0.076 to 0.45.

A total of 304,001 artificial insemination outcomes in up to 7 lactations from 142,389 Holstein cows, daughters of 5,349 sires and 101,433 dams, calving between January 1995 and December 2007 in 1,347 herds were studied by a reaction norm model. The (co)variance components for days to first service (DFS), days open, nonreturn rate in the first service (NRFS), and number of services per conception were estimated by 6 models 3 Legendre polynomial degrees for the genetic effects and adjustment or not for the level of fat plus protein (FP) production recorded at day closest to DFS. For all traits and type of FP adjustment, a second degree polynomial showed the best fit. The use of the adjusted FP model did not increase the level of genetic (co)variance components except for DFS. The heritability for each of the traits was low in general (0.03-0.10) and increased from the first to fourth calving; nevertheless, very important variability was found for the estimated breeding value (EBV) of the sires. The genetic correlations (rg) were close to unity between adjacent calvings, but decreased for most distant parities, ranging from rg=0.36 (for DFS) to rg=0.63 (for NRFS), confirming the existence of heterogeneous genetic (co)variance components and EBV across lactations. The results of the eigen decomposition of rg shows that the first eigenvalue explained between 82 to 92% and the second between 8 to 14% of the genetic variance for all traits; therefore, a deformation of the overall mean trajectory for reproductive performance across the trajectory of the different calving could be expected if selection favored these eigenfunctions. The results of EBV for the 50 best sires showed a substantial reranking and variation in the shape of response across lactations. The more important aspect to highlight, however, is the difference between the EBV of the same sires in different calvings, a characteristic known as plasticity, which is particularly important for DFS and NRFS. This component of fertility adds another dimension to selection for fertility that can be used to change the negative genetic progress of reproductive performance presented in this population of Holstein cows. The use of a reaction norm model should allow producers to obtain more robust cows for maintenance of fertility levels along the whole productive life of the cows. © 2013 American Dairy Science Association.

Phage lambda, like a number of other large DNA bacteriophages and the herpesviruses, produces concatemeric DNA during DNA replication. The concatemeric DNA is processed to produce unit-length, virion DNA by cutting at specific sites along the concatemer. DNA cutting is co-ordinated with DNA packaging, the process of translocation of the cut DNA into the preformed capsid precursor, the prohead. A key player in the lambda DNA packaging process is the phage-encoded enzyme terminase, which is involved in (i) recognition of the concatemeric lambda DNA; (ii) initiation of packaging, which includes the introduction of staggered nicks at cosN to generate the cohesive ends of virion DNA and the binding of the prohead; (iii) DNA packaging, possibly including the ATP-driven DNA translocation; and (iv) following translocation, the cutting of the terminal cosN to complete DNA packaging. To one side of cosN is the site cosB, which plays a role in the initiation of packaging; along with ATP, cosB stimulates the efficiency and adds fidelity to the endonuclease activity of terminase in cutting cosN. cosB is essential for the formation of a post-cleavage complex with terminase, complex I, that binds the prohead, forming a ternary assembly, complex II. Terminase interacts with cosN through its large subunit, gpA, and the small terminase subunit, gpNu1, interacts with cosB. Packaging follows complex II formation. cosN is flanked on the other side by the site cosQ, which is needed for termination, but not initiation, of DNA packaging. cosQ is required for cutting of the second cosN, i.e. the cosN at which termination occurs. DNA packaging in lambda has aspects that differ from other lambda DNA transactions. Unlike the site-specific recombination system of lambda, for DNA packaging the initial site-specific protein assemblage gives way to a mobile, translocating complex, and unlike the DNA replication system of lambda, the same protein machinery is used for both initiation and translocation during lambda DNA packaging.
