In the deep sea, the phylogeny and biogeography of only a few taxa have been well studied. Although more than 200 species in 32 genera have been described for the asellote isopod families Desmosomatidae Sars, 1897 and Nannoniscidae Hansen, 1916 from all ocean basins, their phylogenetic relationships are not completely understood. There is little doubt about the close relationship of these families, but the taxonomic position of a number of genera is so far unknown. Based on a combined morphological phylogeny using the Hennigian method with a dataset of 107 described species and a molecular phylogeny based on three markers (COI, 16S, and 18S) with 75 species (most new to science), we could separate Desmosomatidae and Nannoniscidae as separate families. However, we could not support the concept of the subfamilies Eugerdellatinae Hessler, 1970 and Desmosomatinae Hessler, 1970. Most genera of both families were well supported, but several genera appear as para- or even polyphyletic. Within both families, convergent evolution and analogies caused difficulty in defining apomorphies for phylogenetic reconstructions and this is reflected in the results of the concatenated molecular tree. There is no biogeographic pattern in the distribution as the genera occur over the entire Atlantic and Pacific Ocean, showing no specific phylogeographical pattern. Poor resolution at deep desmosomatid nodes may reflect the long evolutionary history of the family and rapid evolutionary radiations. Metazoans normally possess a single lineage of mitochondria inherited from the mother (♀-type mitochondria) while paternal mitochondria are absent or eliminated in fertilized eggs. In doubly uniparental inheritance (DUI), which is specific to the bivalve clade including the ocean quahog, Arctica islandica, ♂-type mitochondria are retained in male gonads and, in a few species, small proportions of ♂-type mitochondria co-exist with ♀-type in somatic tissues. To the best of our knowledge, we report, for the first time in metazoan, the natural occurrence of male and female individuals with exclusively ♂-type mitochondria in somatic tissues of the bivalve A. islandica. Mitochondrial genomes differ by ~5.5% at DNA sequence level. Exclusive presence of ♂-type mitochondria affects mitochondrial complexes partially encoded by mitochondrial genes and leads to a sharp drop in respiratory capacity. Through a combination of whole mitochondrial genome sequencing and molecular assays (gene presence and expression), we demonstrate that 1) 11% of individuals of an Icelandic population appear homoplasmic for ♂-type mitochondria in somatic tissues, 2) ♂-type mitochondrial genes are transcribed and 3) individuals with ♂-type mitochondria in somatic cells lose 30% of their wild-type respiratory capacity. This mitochondrial pattern in A. islandica is a special case of DUI, highlighted in individuals from both sexes with functional consequences at cellular and conceivably whole animal level., Dégletagne et al. report on an unusual case of doubly uniparental inheritance in the ocean quahog, Arctica isladica. Some individuals in an Icelandic population present homoplasmic presence of male-type mitochondrial genes in somatic tissues, which appears to confer some changes in respiratory capacity in some environmental conditions.

The Southern Ocean is one of the most isolated marine ecosystems, characterized by high levels of endemism, diversity, and biomass. Ascidians are among the dominant groups in Antarctic benthic assemblages; thus, recording the evolutionary patterns of this group is crucial to improve our current understanding of the assembly of this polar ocean. We studied the genetic variation within Cnemidocarpa verrucosa sensu lato, one of the most widely distributed abundant and studied ascidian species in Antarctica. Using a mitochondrial and a nuclear gene (COI and 18S), the phylogeography of fifteen populations distributed along the West Antarctic Peninsula and Burdwood Bank/MPA Namuncurá (South American shelf) was characterized, where the distribution of the genetic distance suggested the existence of, at least, two species within nominal C. verrucosa. When reevaluating morphological traits to distinguish between genetically defined species, the presence of a basal disk in one of the genotypes could be a diagnostic morphological trait to differentiate the species. These results are surprising due to the large research that has been carried out with the conspicuous C. verrucosa with no differentiation between species. Furthermore, it provides important tools to distinguish species in the field and laboratory. But also, these results give new insights into patterns of differentiation between closely related species that are distributed in sympatry, where the permeability of species boundaries still needs to be well understood., The evidence presented here allowed to delimitate two species within the conspicuous Antarctic ascidian C. verrucosa: two monophyletic groups were obtained, and congruent mitochondrial/nuclear species delimitation was demonstrated. Our results once again emphasize that species identities, even for highly abundant and well‐studied species on small local scales, must be assessed rather than assumed.

Parallel phenotypic divergence is the independent differentiation between phenotypes of the same lineage or species occupying ecologically similar environments in different populations. We tested in the Antarctic limpet Nacella concinna the extent of parallel morphological divergence in littoral and sublittoral ecotypes throughout its distribution range. These ecotypes differ in morphological, behavioural and physiological characteristics. We studied the lateral and dorsal outlines of shells and the genetic variation of the mitochondrial gene Cytochrome Oxidase subunit I from both ecotypes in 17 sample sites along more than 2,000 km. The genetic data indicate that both ecotypes belong to a single evolutionary lineage. The magnitude and direction of phenotypic variation differ between ecotypes across sample sites; completely parallel ecotype-pairs (i.e., they diverge in the same magnitude and in the same direction) were detected in 84.85% of lateral and 65.15% in dorsal view comparisons. Besides, specific traits (relative shell height, position of shell apex, and elliptical/pear-shape outline variation) showed high parallelism. We observed weak morphological covariation between the two shape shell views, indicating that distinct evolutionary forces and environmental pressures could be acting on this limpet shell shape. Our results demonstrate there is a strong parallel morphological divergence pattern in N. concinna along its distribution, making this Antarctic species a suitable model for the study of different evolutionary forces shaping the shell evolution of this limpet.

The ocean quahog Arctica islandica is equally attractive to physiologists and environmental scientists due to its long life span of up to 400 years . For physiology, A. islandica is an exciting age model whereas environmental sciences infer past environmental conditions from morphometric and biogeochemical properties of the calcareous shell. In order to evaluate whether and animals from different localities differ, the present study compared genetics and morphometry of six A. islandica populations, from the Norwegian Coast, Kattegat, Kiel bay, White Sea, German Bight and Off Ne Iceland. For genetics, present research found 30 haplotypes of the mitochondrial cytochrome B (cyt B) gene, of which some are widely distributed, whereas others are quite rare. There is no consistent geographical pattern in haplotype distribution, even when a further previously published 11 haplotypes are taken into account. For morphometry, discriminant analysis indicated significant morphometric differences, e.g. in shell shape, between populations which are independent of geographical distance. Moreover, genetic and morphometric patterns appear to be unrelated. Thus, present study reveals morphological differences between populations of A. islandica appear to be driven by the environment rather than underlying genetic differences between populations.

Benthic organisms of the Southern Ocean are particularly vulnerable to ocean acidification (OA), as they inhabit cold waters where calcite-aragonite saturation states are naturally low. OA most strongly affects animals with calcium carbonate skeletons or shells, such as corals and mollusks. We exposed the abundant cold-water coral Malacobelemnon daytoni from an Antarctic fjord to low pH seawater (LpH) (7.68 ± 0.17) to test its physiological responses to OA, at the level of gene expression (RT-PCR) and enzyme activity. Corals were exposed in short- (3 days) and long-term (54 days) experiments to two pCO2 conditions (ambient and elevated pCO2 equaling RCP 8.5, IPCC 2019, approximately 372.53 and 956.78 μatm, respectively). Of the eleven genes studied through RT-PCR, six were significantly upregulated compared with control in the short-term in the LpH condition, including the antioxidant enzyme superoxide dismutase (SOD), Heat Shock Protein 70 (HSP70), Toll-like receptor (TLR), galaxin and ferritin. After long-term exposure to low pH conditions, RT-PCR analysis showed seven genes were upregulated. These include the mannose-binding C-Lectin and HSP90. Also, the expression of TLR and galaxin, among others, continued to be upregulated after long-term exposure to LpH. Expression of carbonic anhydrase (CA), a key enzyme involved in calcification, was also significantly upregulated after long-term exposure. Our results indicated that, after two months, M. daytoni is not acclimatized to this experimental LpH condition. Gene expression profiles revealed molecular impacts that were not evident at the enzyme activity level. Consequently, understanding the molecular mechanisms behind the physiological processes in the response of a coral to LpH is critical to understanding the ability of polar species to cope with future environmental changes. Approaches integrating molecular tools into Antarctic ecological and/or conservation research make an essential contribution given the current ongoing OA processes.

The geographic distributions of some coastal marine species have appeared as cosmopolitan ever since they were first scientifically documented. In particular, for many benthic species that are associated with anthropogenic substrata, there is much speculation as to whether or not their broad distributions can be explained by natural mechanisms of dispersal. Here, we focused on two congeneric coastal crustaceans with cosmopolitan distributions—the tube-dwelling amphipodsJassa marmorataandJassa slatteryi. Both species are common elements of marine biofouling on nearly all kinds of artificial hard substrata in temperate to warm seas. We hypothesized that the two species’ modern occurrences across the oceans are the result of human shipping activities that started centuries ago. Mitochondrial DNA sequences of the CO1 fragment of specimens from distinct marine regions around the world were analysed, evaluating genetic structure and migration models and making inferences on putative native ranges of the twoJassaspecies. Populations of both species exhibited considerable genetic diversity with differing levels of geographic structure. For both species, at least two dominant haplotypes were shared among several geographic populations. Rapid demographic expansion and high migration rates between geographically distant regions support a scenario of ongoing dispersal all over the world. Our findings indicate that the likely former native range ofJ. marmoratais the Northwest Atlantic, whereas the likely former native range ofJ. slatteryiis the Northern Pacific region. As corroborated by the genetic connectivity between populations, shipping still appears to be the more successful vector of the two species’ dispersal when compared to natural mechanisms. Historical invasion events that likely started centuries ago, along with current ongoing dispersal, confirm these species’ identities as true “neocosmopolitans”.

The brown shrimp, Crangon crangon, is well adapted to the variable environmental conditions in the southern North Sea. It is very abundant, has high reproduction rates, and holds a key position in coastal ecosystems. This species has very low lipid deposits in the midgut gland, suggesting that the main function of the midgut gland is metabolic turnover rather than energy storage. Based on seasonal gene expression studies and established transcriptome data, we investigated key components of lipid metabolic pathways. Gene expression of triacylglycerol lipase, phospholipase, and fatty acid desaturase were analyzed and compared with that of other digestive enzymes involved in lipid, carbohydrate, and protein catabolism. Our results suggest that gene expression of digestive enzymes involved in lipid metabolism is modulated by the lipid content in the midgut gland and is related to food availability. Brown shrimp seem to be capable of using cellular phospholipids during periods of food paucity but high energetic (lipid) requirements. Two of three isoforms of fatty acid binding proteins (FABPs) from the midgut gland involved in fatty acid transport showed specific mutations of the binding site. We hypothesize that the mutations in FABPs and deficiencies in anabolic pathways limit lipid storage capacities in the midgut gland of C. crangon. In turn, food utilization, including lipid catabolism, has to be efficient to fulfill the energetic requirements of brown shrimp.

En el presente estudio se utilizo un fragmento del rDNA 16S mitocondrial para formular una hipotesis filogenetica para tres familias de cumaceos, Diastylidae, Bodotriidae y Leuconidae. Ademas se han analizado las relaciones intrafamiliares de esta ultima. Los cumaceos es un grupo monofiletico con tanaidaceos e isopodos como taxones externos. De las tres familias analizadas, los Diastylidae fueron la unica con buen apoyo para la monofilia. El genero Leucon se resol- vio parafiletico mientras que el subgenero Crymoleucon fue monofiletico. Ademas, se analizo la estructura genetica de dos especies de leuconidos Leucon antarcticus Zimmer, 1907 y L. intermedius Muhlenhardt-Siegel, 1996 del mar de Weddell y el mar de Ross respectivamente. Ambas especies mostraron diferentes patrones de variabilidad genetica intraespecifica. A diferencia de L. intermedius, para L. antarcticus se observo una distribucion bimodal del mismatch distribution, que se corre- laciona con las distribuciones geograficas y de profundidad entre el mar de Ross y el mar de Weddell. Aunque una evaluacion clara de la especiacion criptica en estas especies requiere trabajo adicional con mas especimenes de mas regiones geograficas y rangos de profundidad mas amplios, las diferencias que se muestran en las secuencias del rDNA 16S solo pueden explicarse por la separacion genetica de poblaciones entre el mar de Weddell y el mar de Ross durante un periodo de tiempo prolongado. EnglishPhylogenetic hypotheses for the peracarid order Cumacea are scarce and have not provided a solution to the full extent. In the present study, a fragment of the mitochondrial 16S rDNA was used to erect a phylogenetic hypothesis for three cumacean families, Diastylidae, Bodotriidae and Leuconidae, along with intra-family relationships of the latter. The Cumacea resolved monophyletic with tanaids and isopods as outgroup taxa. The Diastylidae were the only family with good support for monophyly. The genus Leucon resolved paraphyletic, whereas the subgenus Crymoleucon was monophyletic. Furthermore, the genetic structure was analysed for two leuconid species, Leucon antarcticus Zimmer, 1907 and L. intermedius Muhlenhardt-Siegel, 1996, from the Weddell Sea and the Ross Sea. The two species showed different patterns of intraspecific genetic variability. In contrast to L. intermedius, a bimodal distribution of pairwise genetic distances was observed for L. antarcticus, which is correlated with geographical and depth distributions between the Ross Sea and the Weddell Sea. Although a clear evaluation of cryptic speciation in these species requires additional work on more specimens from more geographic regions and broader depth ranges, differences shown in the sequences of 16S rDNA can only be explained by genetic separation of populations between the Weddell Sea and the Ross Sea for an extended period of time

A novel approach towards modeling proteins in aqueous solutions by applying perturbed-chain statistical associating fluid theory (PC-SAFT) is proposed in this work. Lysozyme and bovine serum albumin (BSA) were chosen as model proteins due to a vast database available in literature. However, the huge majority of these literature data had been measured at certain ionic strengths, which is disadvantageous in order to derive pure-component parameters for proteins. Thus, in a first step within this work osmotic coefficients of dialyzed lysozyme and of dialyzed BSA in pure water were measured at 298.15 K. The data were very different for dialyzed and non-dialyzed solutions and further showed a strong dependence on pH and ionic strength. In a second step, the pure-component parameters of lysozyme and BSA were adjusted to the measured osmotic coefficients and to solution densities of dialyzed lysozyme or dialyzed BSA. PC-SAFT explicitly takes into account the complex association behavior between protein and water as well as among the proteins. Therefore, the number of protein association sites of one protein was determined from the sum of association sites of the single amino-acid constituents of a protein available from previous work (Ind. Eng. Chem. Res 50 (2011) 141–151). Similarly, the two PC-SAFT parameters “segment number” and “segment diameter” of a protein were summed or averaged from the single amino-acid constituents. This approach allowed modeling densities and osmotic coefficients of protein solutions with reasonable accuracy compared to experimental data.

Hyperiid amphipods are predatory pelagic crustaceans that are particularly prevalent in high-latitude oceans. Many species are likely to have co-evolved with soft-bodied zooplankton groups such as salps and medusae, using them as substrate, for food, shelter or reproduction. Compared to other pelagic groups, such as fish, euphausiids and soft-bodied zooplankton, hyperiid amphipods are poorly studied especially in terms of their distribution and ecology. Hyperiids of the genus Themisto, comprising seven distinct species, are key players in temperate and cold-water pelagic ecosystems where they reach enormous levels of biomass. In these areas, they are important components of marine food webs, and they are major prey for many commercially important fish and squid stocks. In northern parts of the Southern Ocean, Themisto are so prevalent that they are considered to take on the role that Antarctic krill play further south. Nevertheless, although they are around the same size as krill, and may also occur in swarms, their feeding behaviour and mode of reproduction are completely different, hence their respective impacts on ecosystem structure differ. Themisto are major predators of meso- and macrozooplankton in several major oceanic regions covering shelves to open ocean from the polar regions to the subtropics. Based on a combination of published and unpublished occurrence data, we plot out the distributions of the seven species of Themisto. Further, we consider the different predators that rely on Themisto for a large fraction of their diet, demonstrating their major importance for higher trophic levels such as fish, seabirds and mammals. For instance, T. gaudichaudii in the Southern Ocean comprises a major part of the diets of around 80 different species of squid, fish, seabirds and marine mammals, while T. libellula in the Bering Sea and Greenland waters is a main prey item for commercially exploited fish species. We also consider the ongoing and predicted range expansions of Themisto species in light of environmental changes. In northern high latitudes, sub-Arctic Themisto species are replacing truly Arctic, ice-bound, species. In the Southern Ocean, a range expansion of T. gaudichaudii is expected as water masses warm, impacting higher trophic levels and biogeochemical cycles. We identify the many knowlegde gaps that must be filled in order to evaluate, monitor and predict the ecological shifts that will result from the changing patterns of distribution and abundance of this important pelagic group.

Hyperiidean amphipods are a major prey for fish and seabirds. In the Southern Ocean, they are particularly abundant, with distributions ranging from the Polar Frontal Zone to Antarctic shelf waters. The species Hyperiella dilatata has previously been reported to show a peculiar anti-predatory behaviour: It captures chemically protected, gymnosome pteropods in the water column and carries them on its dorsum, like a backpack. We report this association at four oceanic sampling sites between latitudes 45° and 71° S. Molecular barcodes of both hosts and pteropods are provided and compared with those of other hyperiidean and pteropod specimens. Morphological identifications as well as molecular analyses show a so far undocumented association of Hyperiella antarctica with the pteropod Spongiobranchaea australis in the Polar Frontal Zone (Lazarev Sea). H. dilatata carried Clione limacina antarctica specimens in the Weddell Sea, as recorded previously for the Ross Sea. Lengths of the abducted pteropods varied between 1 and 5 mm, with the biggest pteropod measuring more than half the host’s size. One of the abducting amphipods was a female carrying eggs. The formation of such tandem is known to be very efficient as protection from visually hunting icefish in the crystal-clear coastal waters around the Antarctic continent; however, in the open ocean, this behaviour was so far undocumented. Here, we develop hypotheses on its origin and function.

An integrative inventory of the amphipod scavenging fauna (Lysianassoidea), combining morphological identifications with DNA barcoding, is provided here for the Filchner area situated in the south-eastern Weddell Sea. Over 4400 lysianassoids were investigated for species richness and relative abundances, covering 20 different stations and using different sampling devices, including the southernmost baited traps deployed so far (76°S). High species richness was observed: 29 morphospecies of which 5 were new to science. Molecular species delimitation methods were carried out with 109 cytochrome c oxidase I gene (COI) sequences obtained during this study as well as sequences from specimens sampled in other Antarctic regions. These distance-based analyses (trees and the Automatic Barcode Gap Discovery method) indicated the presence of 42 lineages; for 4 species, several (cryptic) lineages were found. More than 96% of the lysianassoids collected with baited traps belonged to the species Orchomenella pinguides s. l. The diversity of the amphipod scavenger guild in this ice-bound ecosystem of the Weddell Sea is discussed in the light of bottom–up selective forces. In this southernmost part of the Weddell Sea, harbouring spawning and nursery grounds for silverfish and icefishes, abundant fish and mammalian food falls are likely to represent the major food for scavengers. Finally, the importance of biodiversity surveys in the context of the establishment of a marine protected area in this region (Weddell Sea MPA) is highlighted and how future studies can contribute to a better understanding the ecological role of scavengers in this system is discussed.

Amphipoda from the superfamily Lysianassoidea Dana, 1849 play an important role in Southern Ocean benthic food webs due to their high biomass, abundance and predominantly scavenging mode of feeding. Our knowledge on the lysianassoid fauna, even in well-studied areas of the Western Antarctic Peninsula, is incomplete. Here we report the findings of an integrated study of lysianassoid amphipods of Potter Cove, King George Island/Isla 25 de Mayo (KGI), combining morphological and molecular species identification (COI barcoding) methods, investigating more than 41,000 specimens from baited traps. Background: Scavenger guilds are composed of a variety of species, co-existing in the same habitat and sharing the same niche in the food web. Niche partitioning among them can manifest in different feeding strategies, e.g. during carcass feeding. In the bentho-pelagic realm of the Southern Ocean, scavenging amphipods (Lysianassoidea) are ubiquitous and occupy a central role in decomposition processes. Here we address the question whether scavenging lysianassoid amphipods employ different feeding strategies during carcass feeding, and whether synergistic feeding activities may influence carcass decomposition. To this end, we compared the relatively large species Waldeckia obesa with the small species Cheirimedon femoratus, Hippomedon kergueleni, and Orchomenella rotundifrons during fish carcass feeding (Notothenia spp.). The experimental approach combined ex situ feeding experiments, behavioural observations, and scanning electron microscopic analyses of mandibles. Furthermore, we aimed to detect ecological drivers for distribution patterns of scavenging amphipods in the Antarctic coastal ecosystems of Potter Cove. In Potter Cove, the climate-driven rapid retreat of the Fourcade Glacier is causing various environmental changes including the provision of new marine habitats to colonise. While in the newly ice-free areas fish are rare, macroalgae have already colonised hard substrates. Assuming that a temporal dietary switch may increase the colonisation success of the most abundant lysianassoids C. femoratus and H. kergueleni, we aimed to determine their consumption rates (g food x g amphipods−1 x day−1) and preferences of macroalgae and fish.Results: We detected two functional groups with different feeding strategies among scavenging amphipods during carcass feeding: carcass 'opener' and 'squeezer'. Synergistic effects between these groups were not statistically verified under the conditions tested. C. femoratus switched its diet when fish was not available by consuming macroalgae (about 0.2 day−1) but preferred fish by feeding up to 80% of its own mass daily. Contrary, H. kergueleni rejected macroalgae entirely and consumed fish with a maximal rate of 0.8 day−1.Conclusion: This study reveals functional groups in scavenging shallow-water amphipods and provides new information on coastal intraguild niche partitioning. We conclude that the dietary flexibility of C. femoratus is a potential ecological driver and central to its success in the colonisation of newly available ice-free Antarctic coastal habitats.

Large-amplitude internal waves (LAIW) cause intense drops in sea water temperature (6–9 °C) and acidity (0.6 pH units) within minutes, posing a major disturbance factor for shallow water corals in the Andaman Sea. During periods of elevated temperatures, however, they may provide a vital source of relief to heat-stressed corals. Hypoxia in freshwater ecosystems is spreading as a consequence of global change, including pollution and eutrophication. In the Patagonian Andes, a decline in precipitation causes reduced lake water volumes and stagnant conditions that limit oxygen transport and exacerbate hypoxia below the upper mixed layer. We analyzed the molecular and biochemical response of the North Patagonian bivalve Diplodon chilensis after 10 days of experimental anoxia (< 0.2 mg O2/L), hypoxia (2 mg O2/L), and normoxia (9 mg O2/L). Specifically, we investigated the expression of an alternative oxidase (AOX) pathway assumed to shortcut the regular mitochondrial electron transport system (ETS) during metabolic rate depression (MRD) in hypoxia-tolerant invertebrates. Whereas, the AOX system was strongly upregulated during anoxia in gills, ETS activities and energy mobilization decreased [less transcription of glycogen phosphorylase (GlyP) and succinate dehydrogenase (SDH) in gills and mantle]. Accumulation of succinate and induction of malate dehydrogenase (MDH) activity could indicate activation of anaerobic mitochondrial pathways to support anoxic survival in D. chilensis. Oxidative stress [protein carbonylation, glutathione peroxidase (GPx) expression] and apoptotic intensity (caspase 3/7 activity) decreased, whereas an unfolded protein response (HSP90) was induced under anoxia. This is the first clear evidence of the concerted regulation of the AOX and ETS genes in a hypoxia-tolerant freshwater bivalve and yet another example that exposure to hypoxia and anoxia is not necessarily accompanied by oxidative stress in hypoxia-tolerant mollusks. The population genetic structure of Calanoides natalis (ex Calanoides carinatus; Copepoda, Calanoida), an ecologically important component of African upwelling systems, was studied in order to (i) search for potential cryptic species, (ii) describe spatial patterns in the distribution of genetic variance and (iii) identify potential barriers to gene flow. Samples were obtained in the eastern Atlantic Ocean from the Iberian Peninsula to Namibia. Analysis of mitochondrial (cytochrome c oxidase subunit I; COI) and nuclear (citrate synthase; CS) marker genes revealed a genetically cohesive population of C. natalis with a prevalent shift in allele frequencies. The discovery of a deep split solely present in the mitochondrial dataset does not point to cryptic speciation, but rather suggests the occurrence of nuclear mitochondrial pseudogenes or incomplete reproductive isolation upon secondary contact. Genetic differentiation between the northern and southern hemisphere was significant, which may point to a potential, but permeable barrier close to the equator. No vertical genetic structuring was detected in the northern Benguela implying that horizontal differentiation was more pronounced than vertical structuring. Retention mechanisms and the oxygen minimum zone did not have a strong impact on genetic differentiation of C. natalis in the Benguela region.

Background DNA barcoding has demonstrated that many discrete phenotypes are in fact genetically distinct (pseudo)cryptic species. Genetically identical, isogenic individuals, however, can also express similarly different phenotypes in response to a trigger condition, e.g. in the environment. This alternative explanation to cryptic speciation often remains untested because it requires considerable effort to reject the hypothesis that the observed underlying genetic homogeneity of the different phenotypes may be trivially caused by too slowly evolving molecular markers. The widespread squat lobster Munida gregaria comprises two discrete ecotypes, gregaria s. str. and subrugosa, which were long regarded as different species due to marked differences in morphological, ecological and behavioral traits. We studied the morphometry and genetics of M. gregaria s. l. and tested (1) whether the phenotypic differences remain stable after continental-scale sampling and inclusion of different life stages, (2) and whether each phenotype is underpinned by a specific genotype. Results A total number of 219 gregaria s. str. and subrugosa individuals from 25 stations encompassing almost entire range in South America were included in morphological and genetic analyses using nine unlinked hypervariable microsatellites and new COI sequences. Results from the PCA and using discriminant functions demonstrated that the morphology of the two forms remains discrete. The mitochondrial data showed a shallow, star-like haplotype network and complete overlap of genetic distances within and among ecotypes. Coalescent-based species delimitation methods, PTP and GMYC, coherently suggested that haplotypes of both ecotypes forms a single species. Although all microsatellite markers possess sufficient genetic variation, AMOVA, PCoA and Bayesian clustering approaches revealed no genetic clusters corresponding to ecotypes or geographic units across the entire South-American distribution. No evidence of isolation-by-distance could be detected for this species in South America. Conclusions Despite their pronounced bimodal morphologies and different lifestyles, the gregaria s. str. and subrugosa ecotypes form a single, dimorphic species M. gregaria s. l.. Based on adequate geographic coverage and multiple independent polymorphic loci, there is no indication that each phenotype may have a unique genetic basis, leaving phenotypic plasticity or localized genomic islands of speciation as possible explanations. Molecular technologies are more frequently applied in Antarctic ecosystem research and the growing amount of sequence-based information available in databases adds a new dimension to understanding the response of Ant- arctic organisms and communities to environmental change. We apply molecular techniques, including finger- printing, and amplicon and metagenome sequencing, to understand biodiversity and phylogeography to resolve adaptive processes in an Antarctic coastal ecosystem from microbial to macrobenthic organisms and communities. Interpretation of the molecular data is not only achieved by their combination with classical methods (pigment analyses or microscopy), but furthermore by combining molecular with environmental data (e.g., sediment characteristics, biogeochemistry or oceanography) in space and over time. The studies form part of a long-term ecosystem investigation in Potter Cove on King-George Island, Antarctica, in which we follow the effects of rapid retreat of the local glacier on the cove ecosystem. We formulate and encourage new approaches to integrate molecular tools into Antarctic ecosystem research, environmental conservation ac- tions, and polar ocean observatories. Fil: Abele, Doris. Traditionally, many small-sized copepod species are considered to be widespread, bipolar or cosmopoli- tan. However, these large-scale distribution patterns need to be re-examined in view of increasing evi- dence of cryptic and pseudo-cryptic speciation in pelagic copepods. Here, we present a phylogeographic study of Oithona similis s.l. populations from the Arctic Ocean, the Southern Ocean and its northern boundaries, the North Atlantic and the Mediterrranean Sea. O. similis s.l. is considered as one of the most abundant species in temperate to polar oceans and acts as an important link in the trophic network between the microbial loop and higher trophic levels such as fish larvae. Two gene frag- ments were analysed: the mitochondrial cytochrome oxidase c subunit I (COI), and the nuclear ribosomal 28 S genetic marker. Seven distinct, geographically delimitated, mitochondrial lineages could be identi- fied, with divergences among the lineages ranging from 8 to 24%, thus representing most likely cryptic or pseudocryptic species within O. similis s.l. Four lineages were identified within or close to the borders of the Southern Ocean, one lineage in the Arctic Ocean and two lineages in the temperate Northern hemi- sphere. Surprisingly the Arctic lineage was more closely related to lineages from the Southern hemi- sphere than to the other lineages from the Northern hemisphere, suggesting that geographic proximity is a rather poor predictor of how closely related the clades are on a genetic level.

Understanding phylogenetic relationship and patterns of contemporary population genetic structure is a prerequisite for conservation and management of potential fishery resources. In this study we report the isolation and characterization of 11 polymorphic microsatellite loci for the squat lobster Munida gregaria from around the Falkland Islands/Islas Malvinas. The number of alleles ranged from 3 to 31, observed heterozygosity varied from 0.130 to 0.870. Cross-amplification was 100 % successfully in the species/morph M. subrugosa and 36.4 % in another congeneric species M. gracilis. This set of microsatellites is useful for studies focused on taxonomy, genetic diversity and genetic connectivity further may provide stock assessment information for monitoring this important fishery resource.

Brachyuran and anomuran decapod crabs do not occur in the extremely cold waters of the Antarctic continental shelf whereas caridean and other shrimp-like decapods, amphipods and isopods are highly abundant. Differing capacities for extracellular ion regulation, especially concerning magnesium, have been hypothesised to determine cold tolerance and by that the biogeography of Antarctic crustaceans. Magnesium is known to have a paralysing effect, which is even more distinct in the cold. As only few or no data exist on haemolymph ionic composition of Sub-Antarctic and Antarctic crustaceans, haemolymph samples of 12 species from these regions were analysed for the concentrations of major inorganic ions (Na+, K+, Ca2+, Mg2+, Cl−, SO4 2−) by ion chromatography. Cation relationships guaranteed neuromuscular excitability in all species. Sulphate and potassium correlated positively with magnesium concentration. The Antarctic caridean decapod as well as the amphipods maintained low (6–20% of ambient sea water magnesium concentration), Sub-Antarctic brachyuran and anomuran crabs as well as the Antarctic isopods high (54–96% of ambient sea water magnesium concentration) haemolymph magnesium levels. In conclusion, magnesium regulation may explain the biogeography of decapods, but not that of the peracarids.

Colossendeis megalonyx Hoek, 1881 is a widespread and abundant pycnogonid in the Southern Ocean which has also been reported from the South Atlantic and South Pacific Oceans. Its strictly benthic lifestyle is expected to promote genetic differentiation among populations and ultimately facilitate speciation. On the other hand, the reported eurybathy and unknown larval stages of this species may allow Colossendeis megalonyx to maintain genetic continuity between isolated shallow-water habitats by active dispersal through the deep sea or by passive rafting on floating substrates. Thus, it remains unknown whether and to which extent geographically separated populations of Colossendeis megalonyx maintain gene flow in the Southern Ocean. We sampled 96 specimens of Colossendeis megalonyx from three stations in the Atlantic Sector of the Southern Ocean and one station from the South American continental shelf (Burdwood Bank). The genetic structure of nominal Colossendeis megalonyx as well as its phylogenetic position within the genus Colossendeis were assessed using a fragment of the cytochrome c oxidase subunit 1 gene. Our data strongly support that nominal Colossendeis megalonyx consists of at least five cryptic and one pseudocryptic mitochondrial lineages, four of which appear to be geographically restricted. Two lineages occurred at locations separated by more than 1,000 km in the Antarctic, thus indicating high levels of gene flow or recent colonization. No haplotype sharing across the Polar Frontal Zone was observed. Our results strongly suggest that cryptic speciation occurred within the genus Colossendeis. The wide biogeographic distribution range of Colossendeis megalonyx and perhaps that of other Antarctic pycnogonids should therefore be regarded with caution.

This study analyzes the performance of a method to isolate microsatellites from largely unknown target genomes. This reporter genome protocol (RGP) utilizes naturally occurring repeat motifs in genomes of distantly related organisms as hybridization probes. The RGP proved very successful in all 13 enrichment reactions from eight marine target species (comprising a dinoflagellate, a diatom, and six arthropod species), yielding on average 85.5% positive colonies. The RGP naturally screens for all repeat types occurring in the reporter genomes and is therefore less biased than standard protocols that typically test few, short motifs only. Using the genomes of Mus musculus, Drosophila melanogaster, and Homo sapiens as reporter genomes in this study, 133 different di- to hexanucleotide repeat types were obtained. Success of the RGP did not depend on overall microsatellite density in the reporter genome but increased with genetic distance between target and reporter genomes because hybridization to conserved nonrepeat regions is less frequent. Relative abundance of repeat types in the reporter genome had a significant effect on repeat type frequencies in the target library. Altogether, the RGP greatly simplifies the isolation of microsatellites from unknown genomes and makes microsatellite markers more attractive for a wide range of studies.

This study reports the successful isolation of highly informative microsatellite marker sets for two marine serolid isopod species. For Serolis paradoxa (Fabricius, 1775), 13, and for Septemserolis septemcarinata (Miers, 1875), eight polymorphic microsatellite markers were isolated using the reporter genome enrichment protocol. The number of alleles per locus (N(A) ) and the observed heterozygosity (H(O) ) encompass a wide range of variation within S. paradoxa (N(A) 3-31, H(O) 6-89%) and S. septemcarinata (N(A) 2-18, H(O) 9-94%). The suitability of the newly isolated markers for population genetic studies is evaluated.

A new arcturid isopod of the genus Chaetarcturus is described from the deep-sea of the Weddell Sea. C. cryophilus n. sp. is characterized by blunt supraocular spines, a large number of acute dorsal spines on the body and strong caudal spines on the pleotelson. The similar Antarctic species C. tenuispinatus has long and acute supraocular spines. The frequent discovery of new Antarctic arcturids suggests that the number of known species is only a fraction of the biodiversity of this group.

We report the successful isolation of 10 polymorphic microsatellite markers for one species of the marine isopod species complex Ceratoserolis trilobitoides (Eights, 1833) from the Southern Ocean. The number of alleles per locus ranged from 6 to 30 for the 148 specimens analysed and heterozygosity ranged from 0.34 to 0.98. Seven microsatellites amplified successfully in a cryptic sister species, which is restricted to the Antarctic Peninsula. This novel marker set provides the opportunity to study and monitor population structure, demography and gene flow patterns for a benthic model taxon in a region that is now subject to rapid climate change.

During diving explorations of anchialine cave systems on Abaco Island, Bahamas, we collected five larvae that represent different developmental stages of remipede crustaceans. Based on four early naupliar stages and a post-naupliar larva, it is possible for the first time to reconstruct the postembryonic development of Remipedia some 25 years after their discovery. These specimens begin to fill in some critical gaps in our knowledge of this important group of crustaceans.

Mitochondrial DNA (mtDNA) is strictly maternally inherited in metazoans. The major exception to this rule has been found in many bivalve species which allow the presence of different sex-linked mtDNA molecules. This mechanism, named Doubly Uniparental Inheritance (DUI), is characterized by the presence of 2 mtDNAs: the female mtDNA is found in somatic tissue and female gonads whereas the male mtDNA is usually found in male gonads and sperm. In this study we highlight the existence of two divergent mitochondrial haplotypes with a low genetic difference around 6-8% in Arctica islandica, a long-lived clam belonging to the Arcticidae, a sister group to the Veneridae in which DUI has been found. Phylogenetic analysis on cytochrome b and 16S sequences from somatic and gonadic tissues of clams belonging to different populations reveal the presence of the “divergent” type in male gonads only and the “normal” type in somatic tissues and female gonads. This peculiar segregation of divergent mtDNA types speaks for the occurrence of the DUI mechanism in Arctica islandica. This example also highlights the difficulties to assess the presence of such particular mitochondrial inheritance system and underlines the possible misinterpretations in phylogeographic and phylogenetic studies of bivalve species linked to the presence of two poorly differentiated mitochondrial genomes.

SUMMARY: The genus Glyptonotus is most commonly regarded as monospecific, with Glyptonotus antarcticus Eights 1852 being its only constituent. Two more forms (G. acutus, G. antarcticus var. obtusus) that had been described based on morphological evidence have received little attention in the literature, though no formal attempt to evaluate their taxonomic status has been undertaken. In a survey of within-species genetic diversity, 23 specimens of the benthic Antarctic isopod Glyptonotus antarcticus from three sampling areas around the Antarctic had a high genetic variability in their mitochondrial LSU (16S) gene. Eleven unique mitochondrial haplotypes were found, two at the Antarctic Peninsula (AP), two in the Ross Sea (RS) and seven in the Eastern Weddell Sea (EWS). Average haplotype variation within sampling areas (AP, RS, EWS) was one order of magnitude less than between sampling areas. In the EWS, however, two highly differentiated haplotypes co-exist. These four groups of haplotypes may represent cryptic, but reproductively isolated species rather than a single species.

Despite its extreme environmental conditions the deep sea harbors a unique and species-rich fauna of mostly unknown age and phylogeny. Asellote isopods have undergone their most impressive radiation in the deep sea, being found at all depths down to the deepest trench. Here we present the first molecular evidence for the phylogenetic origin of this remarkable array of deep-sea crustaceans, based on 30 new DNA-sequences of the complete 18s rRNA gene of specimens collected at depths down to 4543 m in the South Atlantic and South Polar Ocean. The results show that most of these isopod lineages belong to a single ancient clade. They evolved in situ in large oceanic depths and survived several climatic changes, but the lack of fossils and of a suitable molecular clock model prevents a precise dating of this radiation. The monophyly of typical deep-sea families, for example the Haploniscidae, Ischnomesidae or Munnopsidae, is well supported by different methods of analysis, while the monophyly of the Janiridae is rejected.

The shortening of telomeres as a causative factor in ageing is a widely discussed hypothesis in ageing research. The study of telomere length and its regenerating enzyme telomerase in the longest-lived non-colonial animal on earth, Arctica islandica, should inform whether the maintenance of telomere length plays a role in reaching the extreme maximum lifespan (MLSP) of > 500 years in this species. Since longitudinal measurements on living animals cannot be achieved, a cross-sectional analysis of a short-lived (MLSP 40 years from the Baltic Sea) and a long-lived population (MLSP 226 years Northeast of Iceland) and in different tissues of young and old animals from the Irish Sea was performed. A high heterogeneity of telomere length was observed in investigated A. islandica over a wide age range (10–36 years for the Baltic Sea, 11–194 years for Irish Sea, 6–226 years for Iceland). Constant telomerase activity and telomere lengths were detected at any age and in different tissues; neither correlated with age or population habitat. Stable telomere maintenance might contribute to the long lifespan of A. islandica. Telomere dynamics are no explanation for the distinct MLSPs of the examined populations and thus the cause of it remains to be investigated.

Arctica islandica is known as the longest-lived non-colonial metazoan species on earth and is therefore increasingly being investigated as a new model in aging research. As the mitochondrial genome is associated with the process of aging in many species and bivalves are known to possess a peculiar mechanism of mitochondrial genome inheritance including doubly uniparental inheritance (DUI), we aimed to assess the genomic variability of the A. islandica mitochondrial DNA (mtDNA). We sequenced the complete mitochondrial genomes of A. islandica specimens from three different sites in the Western Palaearctic (Iceland, North Sea, Baltic Sea). We found the A. islandica mtDNA to fall within the normal size range (18 kb) and exhibit similar coding capacity as other animal mtDNAs. The concatenated protein sequences of all currently known Veneroidea mtDNAs were used to robustly place A. islandica in a phylogenetic framework. Analysis of the observed single nucleotide polymorphism (SNP) patterns on further specimen revealed two prevailing haplotypes. Populations in the Baltic and the North Sea are very homogenous, whereas the Icelandic population, from which exceptionally old individuals have been collected, is the most diverse one. Homogeneity in Baltic and North Sea populations point to either stronger environmental constraints or more recent colonization of the habitat. Our analysis lays the foundation for further studies on A. islandica population structures, age research with this organism, and for phylogenetic studies. A new arcturid species of unusual size (> 6 cm) is described from a locality in the Drake Passage at 3720 m depth. Due to the unique combination of characters found in these isopods, the new genus Furcarcturus is proposed. Sequences of the 16S rRNA gene obtained from eight species indicate that the new species does not belong to groups of species currently placed in the genera Antarcturus and Chaetarcturus. More sequences are needed to corroborate arcturid phlylogeny.

23 pages; International audience; In the Southern Ocean, that is areas south of the Polar Front, long-term oceanographic cooling, geographic separation, development of isolating current and wind systems, tectonic drift and fluctuation of ice sheets amongst others have resulted in a highly endemic benthic fauna, which is generally adapted to the long-lasting, relatively stable environmental conditions. The Southern Ocean benthic ecosystem has been subject to minimal direct anthropogenic impact (compared to elsewhere) and thus presents unique opportunities to study biodiversity and its responses to environmental change. Since the beginning of the century, research under the Census of Marine Life and International Polar Year initiatives, as well as Scientific Committee of Antarctic Research biology programmes, have considerably advanced our understanding of the Southern Ocean benthos. In this paper, we evaluate recent progress in Southern Ocean benthic research and identify priorities for future research. Intense efforts to sample and describe the benthic fauna, coupled with coordination of information in global databases, have greatly enhanced understanding of the biodiversity and biogeography of the region. Some habitats, such as chemosynthetic systems, have been sampled for the first time, while application of new technologies and methods are yielding new insights into ecosystem structure and function. These advances have also highlighted important research gaps, notably the likely consequences of climate change. In a time of potentially pivotal environmental change, one of the greatest challenges is to balance conservation with increasing demands on the Southern Ocean's natural resources and services. In this context, the characterization of Southern Ocean biodiversity is an urgent priority requiring timely and accurate species identifications, application of standardized sampling and reporting procedures, as well as cooperation between disciplines and nations.

There has been a significant body of literature on species flock definition but not so much about practical means to appraise them. We here apply the five criteria of Eastman and McCune for detecting species flocks in four taxonomic components of the benthic fauna of the Antarctic shelf: teleost fishes, crinoids (feather stars), echinoids (sea urchins) and crustacean arthropods. Practical limitations led us to prioritize the three historical criteria (endemicity, monophyly, species richness) over the two ecological ones (ecological diversity and habitat dominance). We propose a new protocol which includes an iterative fine-tuning of the monophyly and endemicity criteria in order to discover unsuspected flocks. As a result nine « full » species flocks (fulfilling the five criteria) are briefly described. Eight other flocks fit the three historical criteria but need to be further investigated from the ecological point of view (here called « core flocks »). The approach also shows that some candidate taxonomic components are no species flocks at all. The present study contradicts the paradigm that marine species flocks are rare. The hypothesis according to which the Antarctic shelf acts as a species flocks generator is supported, and the approach indicates paths for further ecological studies and may serve as a starting point to investigate the processes leading to flock-like patterning of biodiversity. © 2013 Lecointre et al., SCOPUS: ar.j, info:eu-repo/semantics/published

Research investigating the genetic basis of physiological responses has significantly broadened our understanding of the mechanisms underlying organismic response to environmental change. However, genomic data are currently available for few taxa only, thus excluding physiological model species from this approach. In this study we report the transcriptome of the model organism Hyas araneus from Spitsbergen (Arctic). We generated 20,479 transcripts, using the 454 {GS} {FLX} sequencing technology in combination with an Illumina HiSeq sequencing approach. Annotation by Blastx revealed 7159 blast hits in the {NCBI} non-redundant protein database. The comparison between the spider crab H. araneus transcriptome and {EST} libraries of the European lobster Homarus americanus and the porcelain crab Petrolisthes cinctipes yielded 3229/2581 sequences with a significant hit, respectively. The clustering by the Markov Clustering Algorithm (MCL) revealed a common core of 1710 clusters present in all three species and 5903 unique clusters for H. araneus. The combined sequencing approaches generated transcripts that will greatly expand the limited genomic data available for crustaceans. We introduce the {MCL} clustering for transcriptome comparisons as a simple approach to estimate similarities between transcriptomic libraries of different size and quality and to analyze homologies within the selected group of species. In particular, we identified a large variety of reverse transcriptase (RT) sequences not only in the H. araneus transcriptome and other decapod crustaceans, but also sea urchin, supporting the hypothesis of a heritable, anti-viral immunity and the proposed viral fragment integration by host-derived {RTs} in marine invertebrates.

Eurythenes gryllus is one of the most widespread amphipod species, occurring in every ocean with a depth range covering the bathyal, abyssal and hadal zones. Previous studies, however, indicated the existence of several genetically and morphologically divergent lineages, questioning the assumption of its cosmopolitan and eurybathic distribution. For the first time, its genetic diversity was explored at the global scale (Arctic, Atlantic, Pacific and Southern oceans) by analyzing nuclear (28S rDNA) and mitochondrial (COI, 16S rDNA) sequence data using various species delimitation methods in a phylogeographic context. Nine putative species-level clades were identified within E. gryllus. A clear distinction was observed between samples collected at bathyal versus abyssal depths, with a genetic break occurring around 3,000 m. Two bathyal and two abyssal lineages showed a widespread distribution, while five other abyssal lineages each seemed to be restricted to a single ocean basin. The observed higher diversity in the abyss compared to the bathyal zone stands in contrast to the depth-differentiation hypothesis. Our results indicate that, despite the more uniform environment of the abyss and its presumed lack of obvious isolating barriers, abyssal populations might be more likely to show population differentiation and undergo speciation events than previously assumed. Potential factors influencing species’ origins and distributions, such as hydrostatic pressure, are discussed. In addition, morphological findings coincided with the molecular clades. Of all specimens available for examination, those of the bipolar bathyal clade seemed the most similar to the ‘true’ E. gryllus. We present the first molecular evidence for a bipolar distribution in a macro-benthic deep-sea organism.

Understanding patterns of contemporary population genetic structure and diversity is critically important for cold-adapted Southern Ocean benthic species along the Scotia Arc to distinguish between natural and anthropogenically-mediated range expansions and detect population declines due to the rapid climate change. We report the isolation and characterization of 10 polymorphic microsatellite loci for the Antarctic endemic giant isopod Glyptonotus antarcticus from the Eastern Scotia Arc (n = 30). The number of alleles per locus ranged from 3 to 11 with no evidence of significant linkage disequilibrium between loci. These markers will prove useful in studying the fine-scale processes, which have shaped the spatial distribution of variation, and hypothesis-testing in the Scotia Arc region of Antarctica.

15 pages; International audience; The Dumont d'Urville Sea (East Antarctic region) has been less investigated for DNA barcoding and molecular taxonomy than other parts of the Southern Ocean, such as the Ross Sea and the Antarctic Peninsula. The Collaborative East Antarctic MARine Census (CEAMARC) took place in this area during the austral summer of 2007e2008. The Australian vessel RSVAurora Australis collected very diverse samples of demersal and benthic organisms. The specimens were sorted centrally, and then distributed to taxonomic experts for molecular and morphological taxonomy and identification, especially barcoding. The COI sequences generated from CEAMARC material provide a sizeable proportion of the Census of Antarctic Marine Life barcodes although the studies are still ongoing, and represent the only source of sequences for a number of species. Barcoding appears to be a valuable method for identification within most groups, despite low divergences and haplotype sharing in a few species, and it is also useful as a preliminary taxonomic exploration method. Several new species are being described. CEAMARC samples have already provided new material for phylogeographic and phylogenetic studies in cephalopods, pycnogonids, teleost fish, crinoids and sea urchins, helping these studies to provide a better insight in the patterns of evolution in the Southern Ocean.

Climate warming involves not only a rise of air temperature means, but also more frequent heat waves in many regions on earth, and is predicted to intensify physiological stress especially in extremely changeable habitats like the intertidal. We investigated the heat-shock response (HSR) and enzymatic antioxidant defense levels of Patagonian shallow-water limpets, adapted to distinct tidal exposure conditions in the sub- and intertidal. Limpets were sampled in the temperate Northern Patagonia and the subpolar Magellan region. Expression levels of two Hsp70 genes and activities of the antioxidants superoxide dismutase (SOD) and catalase (CAT) were measured in submerged and 2- and 12-h air-exposed specimens. Air-exposed Patagonian limpets showed a tiered HSR increasing from South to North on the latitudinal gradient and from high to low shore levels on a tidal gradient. SOD activities in the Magellan region correlated with the tidal rhythm and were higher after 2 and 12 h when the tide was low at the experimental site compared to the 6 h value taken at high tide. This pattern was observed in intertidal and subtidal specimens, although subtidal individuals are little affected by tides. Our study shows that long-term thermal adaptation shapes the HSR in limpets, while the oxidative stress response is linked to the tidal rhythm. Close to the warm border of their distribution range, energy expenses to cope with stress might become overwhelming and represent one cause why the limpets are unable to colonize the shallow intertidal zone.

In this study we provide eight polymorphic microsatellite markers for the two South American patel- logastropods Nacella magellanica and N. deaurata. Allelic diversity ranged from 5 to 57 alleles per locus. Observed heterozygosity varied between 0.1 and 0.98. Three of the four loci designed for N. magellanica cross-amplified also with N. deaurata, and two loci vice versa. Six of the microsatellites successfully cross-amplified with the sister taxon N. mytilina. This set of microsatellites provides a suitable tool for population genetic and phylogeographic studies.

In this study, we analyse the evolutionary dynamics and phylogenetic implications of gene order rearrangements in five newly sequenced mitochondrial (mt) genomes and four published mt genomes of isopod crustaceans. The sequence coverage is nearly complete for four of the five newly sequenced species, with only the control region and some tRNA genes missing, while in Janira maculosa only two thirds of the genome could be determined. Mitochondrial gene order in isopods seems to be more plastic than that in other crustacean lineages, making all nine known mt gene orders different. Especially the asellote Janira is characterized by many autapomorphies. The following inferred ancestral isopod mt gene order exists slightly modified in modern isopods: nad1, tnrL1, rrnS, control region, trnS1, cob, trnT, nad5, trnF. We consider the inferred gene translocation events leading to gene rearrangements as valuable characters in phylogenetic analyses. In this first study covering major isopod lineages, potential apomorphies were identified, e.g., a shared relative position of trnR in Valvifera. We also report one of the first findings of homoplasy in mitochondrial gene order, namely a shared relative position of trnV in unrelated isopod lineages. In addition to increased taxon sampling secondary structure, modification in tRNAs and GC-skew inversion may be potentially fruitful subjects for future mt genome studies in a phylogenetic context.

Species integrity is maintained only if recurrent allelic exchange between subpopulations occurs by means of migrating specimens. Predictions of this gene flow on the basis of observed or assumed mobility of marine species have proven to be error-prone. Using one mitochondrial gene and seven microsatellite markers, we studied the genetic structure and gene flow in Septemserolis septemcarinata, a strictly benthic species lacking pelagic larvae and the ability to swim. Suitable shallow-water habitats around three remote islands (South Georgia, Bouvet, and Marion Island) are geographically disjunct, isolated by more than 2,000 km of uninhabitable deep sea (east–west) and also separated by the Polar Front (north–south), which serves as a strong demarcation line in many marine taxa. Although we did find genetic differentiation among the three island populations, our results also revealed that a scenario with recent gene flow explains our data best. A model assuming no gene flow after initial colonization of the islands performs significantly worse. The tests also favor an asymmetric gene flow pattern (west to east ≫ east to west) thus mirroring the directionality of major oceanographic currents in the area. We conclude that rare long-distance dispersal rather than vicariance or human-mediated transport must be responsible for the observed patterns. As a mechanism, we propose passive rafting on floating substrata in the Antarctic Circumpolar Current. The results demonstrate that the effectiveness of a physical barrier is not solely a function of its physical parameters but strongly depends on how organisms interact with their environment.

The taxonomy of two sympatric surf clams Donax marincovichi Coan, 1983 and Donax obesulus Reeve, 1854, inhabiting the coastal Humboldt Current Upwelling System is revisited. Because both species are exploited by artisanal fisheries, it is essential to verify that they are indeed distinct species that have to be managed separately. In this study, both taxa were sampled across their shared distributional area and specimens were indentified according to their respective morphological characteristics. Although width/height and height/length ratios revealed significant differences within sampling areas, the two morphotypes were frequently incongruent for taxonomically important morphometric parameters. In addition, they showed no significant mitochondrial genetic differentiation within or among populations and exhibited indistinguishable sperm ultrastructure. We conclude that the two morphotypes do not represent distinct species and should be included together under D. obesulus.

A population of stone crab (Lithodidae) was encountered on the continental slope off Antarctica in the Bellingshausen Sea between 1,123 and 1,304 m water depths using the ROV-Isis during leg 166 of the RV James Clark Ross, in January 2007. Specimens were video recorded and one specimen was retrieved by ROV for morphological and molecular identification. Based on morphology and molecular data from the mitochondrial COI gene, this specimen identified as P. birsteini, Macpherson, 1988a. The significance of the molecular data and their implications for biogeography and evolution of lithodids in the Southern Ocean are briefly discussed.
