Cystic hydatid disease or cystic echinococcosis (CE) is a globally endemic zoonosis caused by the larval cyst stage of the tapeworm Echinococcus granulosus. Concomitant presence of CE and hepatocellular carcinoma (HCC) is a rare clinical scenario. A 70-year-old male patient presented with acute abdominal pain to the surgical outpatient department. On evaluation, a cystic lesion with solid components and free fluid in the abdomen was observed, which led to multiple differentials in the working diagnosis. A CT showed the mass to have a delayed enhancement. Surgical exploration revealed a partially ruptured hydatid cyst with daughter cysts in the abdominal cavity and a solid-component mass lesion. We proceeded with a right partial hepatectomy. Pathological evaluation revealed a pale mass lesion with a large collapsed cyst. HCC with unusual dense fibrillar fibrosis and cystic interface with normal parenchyma was observed. This case connects the multimodal assessment of radiology, surgery and pathology.

Introduction Legume crops are an important source of protein and oil for human health and in fixing atmospheric N2 for soil enrichment. With an objective to accelerate much-needed genetic analyses and breeding applications, draft genome assemblies were generated in several legume crops; many of them are not high quality because they are mainly based on short reads. However, the superior quality of genome assembly is crucial for a detailed understanding of genomic architecture, genome evolution, and crop improvement. Objectives Present study was undertaken with an objective of developing improved chromosome-length genome assemblies in six different legumes followed by their systematic investigation to unravel different aspects of genome organization and legume evolution. Methods We employed in situ Hi-C data to improve the existing draft genomes and performed different evolutionary and comparative analyses using improved genome assemblies. Results We have developed chromosome-length genome assemblies in chickpea, pigeonpea, soybean, subterranean clover, and two wild progenitor species of cultivated groundnut (A. duranensis and A. ipaensis). A comprehensive comparative analysis of these genome assemblies offered improved insights into various evolutionary events that shaped the present-day legume species. We highlighted the expansion of gene families contributing to unique traits such as nodulation in legumes, gravitropism in groundnut, and oil biosynthesis in oilseed legume crops such as groundnut and soybean. As examples, we have demonstrated the utility of improved genome assemblies for enhancing the resolution of “QTL-hotspot” identification for drought tolerance in chickpea and marker-trait associations for agronomic traits in pigeonpea through genome-wide association study. Genomic resources developed in this study are publicly available through an online repository, ‘Legumepedia’. Conclusion This study reports chromosome-length genome assemblies of six legume species and demonstrates the utility of these assemblies in crop improvement. The genomic resources developed here will have significant role in accelerating genetic improvement applications of legume crops.

Introduction Kikuchi-Fujimoto disease is a benign and self-limiting systemic disorder of unknown aetiology characterised by fever, superficial lymphadenopathy and leukopenia. In highly endemic & low-resource country like India, it is frequently misdiagnosed as tuberculosis. Case report Both the cases were diagnosed as necrotizing lymphadenitis by fine-needle aspiration cytology. Tuberculin skin prick test (TST) was positive for one case and negative for the other case. Cartridge based nucleic acid amplification test (CBNAAT) from lymph node aspirate was negative for mycobacterium tuberculosis in both the cases, later on histopathology of lymph node showed diagnosis of Kikuchi-Fujimoto disease. Conclusion Kikuchi Fujimoto is a self-limiting disease systemic disease of unknown aetiology. A definite diagnosis can be established by incisional/excisional biopsy of the lymph node. When dealing with cases of tubercular lymphadenitis, Kikuchi-Fujimoto disease should be kept as differential diagnosis.

Gastrointestinal stromal tumours (GISTs) are the most common mesenchymal neoplasms of the gastrointestinal tract. Dedifferentiation in these tumours occurs rarely, and when it does occurs most commonly after prolonged treatment with imatinib. We report the case of a 64-year-old man who presented with a mass of 8×7×3 cm dimensions involving the duodenum and head of the pancreas. On histopathology, areas of anaplastic tumour cells were negative for DOG-1, c-kit, CD-34, desmin and panCK along with a molecular level study showing wild-type KIT and PDGFRA (platelet-derived growth factor receptor alpha) gene. Based on focal GIST-like areas and CD117 positivity and absence of prior therapy, the diagnosis of a de novo dedifferentiated GIST was made. These tumours need to be reported as they pose a diagnostic challenge and their predicted response rated to targeted molecular therapies are unclear as compared with their c-kit positive counterparts.

With progressive climate change and the associated increase in mean temperature, heat stress tolerance has emerged as one of the key traits in the product profile of the maize breeding pipeline for lowland tropics. The present study aims to identify the genomic regions associated with heat stress tolerance in tropical maize. An association mapping panel, called the heat tolerant association mapping (HTAM) panel, was constituted by involving a total of 543 tropical maize inbred lines from diverse genetic backgrounds, test-crossed and phenotyped across nine locations in South Asia under natural heat stress. The panel was genotyped using a genotyping-by-sequencing (GBS) platform. Considering the large variations in vapor pressure deficit (VPD) at high temperature (Tmax) across different phenotyping locations, genome-wide association study (GWAS) was conducted separately for each location. The individual location GWAS identified a total of 269 novel significant single nucleotide polymorphisms (SNPs) for grain yield under heat stress at a p value of –5. A total of 175 SNPs were found in 140 unique gene models implicated in various biological pathway responses to different abiotic stresses. Haplotype trend regression (HTR) analysis of the significant SNPs identified 26 haplotype blocks and 96 single SNP variants significant across one to five locations. The genomic regions identified based on GWAS and HTR analysis considering genomic region x environment interactions are useful for breeding efforts aimed at developing heat stress resilient maize cultivars for current and future climatic conditions through marker-assisted introgression into elite genetic backgrounds and/or genome-wide selection.

Rose rosette disease (RRD) caused by rose rosette emaravirus (RRV) is a major issue in the U.S. rose industry with no effective method for its management. This study evaluated the effect of foliar application of acibenzolar-S-methyl (ASM), a plant systemic acquired resistance inducer, in reducing RRD disease severity on Rosa species cv. Radtkopink (‘Pink Double Knock Out’) under greenhouse conditions, and the effect of ASM on plant growth under commercial nursery production conditions. ASM at 50- or 100-mg/liter concentrations at weekly intervals significantly reduced RRD severity compared with the untreated control in two of the three greenhouse trials (P < 0.05). The plants in these trials were subsequently pruned and observed for symptoms, which further indicated that application of ASM at 50- or 100-mg/liter concentrations lowered disease severity compared with the untreated control (P < 0.05) in these two trials. Plants treated with ASM at 50- or 100-mg/liter concentrations had delayed incidence of RRD compared with the nontreated controls. Plants treated with ASM at the 50- or 100-mg/liter rate in all three trials either did not have RRV present or the virus was present in fewer leaf samples than untreated controls as indicated by quantitative reverse transcription PCR analysis. Overall, plants treated with ASM at the 50-mg/liter concentration had 36 to 43% reduced RRD incidence compared with the water control. The treatment of two cultivars of rose, ‘Radtkopink’ and ‘Meijocos’ (‘Pink Drift’), with weekly foliar applications of ASM at the three rates (0.5, 0.75, and 1.0 oz/A) indicated that ASM had no negative effect on flowering or plant growth at even the highest rate of application.

Background The emergence of resistance to commonly used antibiotics by human infections occurred mostly due to their overuse, that prompted individuals to pursue novel and innovative treatments. The phytochemical characteristics, antibacterial activity, and cytotoxicity of MCF7 cells were evaluated in two Pleurotus spp. mycelial extracts in this work. Methods Pleurotus djamor and Pleurotus florida mycelial extracts from pure cultures were tested for antibacterial activity by a well-diffusion assay and antimicrobial activity against mold fungi was evaluated for biomass inhibition. Mycelial extracts were obtained from dichloromethane extracts and their biophysical characteristics are analyzed by UV–vis spectrum and FTIR analysis. By spraying detection reagents onto TLC plates, the chemicals in dichloromethane extraction of chosen mushroom fungus mycelia were identified. Using the MTT test, the cytotoxic effect of dichloromethane extracts of selected mushroom fungi was evaluated on MCF7 Cell lines. Results Mycelial extracts of P. djamor and P. florida exhibited significant antimicrobial effect on the bacterial and fungal pathogens tested. Dichloromethane mycelial extracts were obtained using soxhlet extraction which response positive for various phytochemical analysis. Detection of metabolites in thin layer chromatography using spray reagents documented one of few first accounts on flavonoids, anthroquinone and terpenoid compounds in P. djamor and P. florida. P. djamor and P. florida had dose-dependent antiproliferative activity against MCF7 cells, with an inhibitory impact of 55.72% and 64.47% percent at 125 μg/mL, respectively. Conclusion The study has reported the identification with the potent biological activity of some of the key bioactive components present in DCM extracts from the mycelia of P. djamor and P. florida.

Because of their high strength-to-weight ratio, composites outperform traditional materials. Natural fibers are increasingly being used in composites due to environmental concerns. Their hygroscopic character, on the other hand, has a negative impact on the mechanical properties of the composites. The need to analyses the water uptake characteristics of natural fiber hybrid composites has arisen as a result of composites' use in decking, flooring, and outdoor facilities. The objective of this paper is to study the water absorption behavior and observed that banana/glass fiber composite keeps on increasing. The conclusion is that banana fiber absorbs more water than kenaf/glass fiber composites, and there are numerous causes for trapping of natural fiber water consumption.

Fossil fuels are used as a fuel for wide range of applications around the world, as per the statistics it is found that the depletion rate of fossil fuel is reducing rapidly. To satisfy the needs of mankind we are in search of an alternative fuel. The present research work is carried out using pongamia pinnata and silkworm oil, on a four-stroke diesel engine as an alternative fuel to find the efficiency of the diesel engine using above specified mixture along with fossil fuel with various blends at different flow rates of engine and calorimeter cooling water. In present investigation the biodiesel is prepared from pongamia oil, and it is blended with silkworm oil. The performance parameters of single cylinder 4 S diesel engine are evaluated.

Epidermal growth factor receptor (EGFR) is a receptor tyrosine kinase widely expressed in many cancers such as non-small cell lung cancer (NSCLC), pancreatic cancer, breast cancer, and head and neck cancer. Mutations such as L858R in exon 21, exon 19 truncation (Del19), exon 20 insertions, and others are responsible for aberrant activation of EGFR in NSCLC. First-generation EGFR tyrosine kinase inhibitors (TKIs) such as gefitinib and erlotinib have clinical benefits for EGFR-sensitive (L858R and Del19) NSCLC patients. However, after 10-12 months of treatment with these inhibitors, a secondary T790M mutation at the gatekeeper position in the kinase domain of EGFR was identified, which limited the clinical benefits. Second-generation EGFR irreversible inhibitors (afatinib and dacomitinib) were developed to overcome this T790M mutation. However, their lack of selectivity toward wild-type EGFR compromised their clinical benefits due to serious adverse events. Recently developed third-generation irreversible EGFR TKIs (osimertinib and lazertinib) are selective toward driving mutations and the T790M mutation, while sparing wild-type EGFR activity. The latest studies have concluded that their efficacy was also compromised by additional acquired mutations, including C797S, the key residue cysteine that forms covalent bonds with irreversible inhibitors. Because second- and third-generation EGFR TKIs are irreversible inhibitors, they are not effective against C797S containing EGFR triple mutations (Del19/T790M/C797S and L858R/T790M/C797S). Therefore, there is an urgent unmet medical need to develop next-generation EGFR TKIs that selectively inhibit EGFR triple mutations via a non-irreversible mechanism.

Nanoparticles have a diameter of up to 100 nm and a higher surface-to-volume ratio, enabling more active surface atoms to contribute to implementations and improve material properties. In nanoparticle preparation, the ability to control particle size, shape, and morphology is important. The most important tool for studying nanomaterials is XRD, it is a vital characterization tool in solid-state chemistry and materials science. For any compound, XRD is a simple method for determining the unit cell's size and shape. This study explains how copper oxide nanoparticles are formed in Cissus vitiginea leaves. The antioxidant function and XRD study of the synthesized CuONPs were also investigated. According to the XRD results, the copper oxide nanoparticles formed by reducing Cu2 + ions by Cissus vitiginea leaf extract are crystalline in nature. CuONPs have an average crystalline size of ~32.32 nm, according to the Debye-Scherrer formula. CuONPs have higher antioxidant activity than plant extract and are closest to ascorbic acid in terms of standard.

This work identified a summary of new chalcone derivatives (E)-2-(4-acetamidophenoxy)-N-(3-(3-(4-substitutedphenyl) acryloyl) phenyl) acetamide by the condensation of 2-(4-acetamidophenoxy)-N-(3-acetylphenyl) acetamide with various aromatic aldehydes. Synthesized compounds were characterized by IR and 1H NMR spectroscopy. Several bacterial species and also candida albicans were tested for the antimicrobial activity. Compared to the standard drug streptomycin and clotrimazole against bacterial and fungal species, 5e, 5c and 5d have shown strong antibacterial and moderate antifungal activity. The aim of the anti - microbial agent enzyme was to investigate and describe the interactions of the identified hits within the target enzyme binding pocket using the synthesized composite against glucosamine-6 phosphorus synthase. The docking results enhanced the behaviour of new derivatives as promising antimicrobials. The simulation of (E)-2-(4-acetamidophenoxy)-N-(3-(3-(2-chlorophenyl) acryloyl) phenyl acetamide (5a) in BRCA1 resulted in the creation of two hydrogen bond interactions with bond distance (2.13 A) and it was observed that the best binding energy value for −9.07Kcal / mol

The present investigation “Influence of liquid and carrier based biofertilizers on growth, yield and quality of tomato (Solanum lycopersicum L.)”was laid out in randomized block design with two factors and three levels comprising of nine treatment combinations in three replications at Horticultural Research Station, Dr. Y.S.R. Horticultural University, Venkataramannagudem, West Godavari during Rabi, 2018. The application of 80 % recommended dose of fertilizers along with liquid biofertilizers resulted in significant higher differences in number of fruits per cluster (4.90), number of fruits per plant (60.0), fruit set % (98.34), fruit length (5.83 cm), fruit width (5.45 cm), average fruit weight (96.96 g), fruit yield per plant (7.27 kg), TSS (4.670 Brix), ascorbic acid content (16.53 mg/100 g), lycopene content (31.72 mg/100g) and titrable acidity (0.53 %) when compared to other treatment combinations.

Epithelial to mesenchymal transition (EMT) is pivotal in embryonic development and wound healing, whereas in cancer it inflicts malignancy and drug resistance. The recognition of an EMT-like process in glioma is relatively new and its clinical and therapeutic significance has, as yet, not been fully elucidated. Here, we aimed to delineate the clinical significance of the EMT-like process in glioma and its therapeutic relevance to rabeprazole. We investigated the expression profiles of EMT-associated proteins in primary glioma biopsies through Western blotting and immunohistochemistry, and correlated them with various clinicopathological features and data listed in the cancer genome atlas (TCGA). In addition, the anticancer efficacy of rabeprazole and its therapeutic relevance to EMT along with temozolomide chemo-sensitization were assessed using multiple cell-based assays, Western blotting and confocal imaging. For in vivo assessment, we used a stereotaxic C6-rat glioma model. Expression analysis of EMT-associated proteins in glioma biopsies, in conjunction with clinicopathological and TCGA dataset analyses, revealed non-canonical expression of E/N-cadherin and upregulation of GFAP, vimentin and β-catenin. The increased expression of EMT-associated proteins may attribute to glioma malignancy and a poor patient prognosis. Subsequent in vitro studies revealed that rabeprazole treatment attenuated glioma cell growth and migration, and induced apoptosis. Rabeprazole suppressed EMT by impeding AKT/GSK3β phosphorylation and/or NF-κB signaling and sensitized temozolomide resistance. Additional in vivo studies showed restricted tumor growth and inhibited expression of EMT-associated proteins after rabeprazole treatment. Our data revealed (i) a clinical association of the EMT-like process with glioma malignancy and a poor survival and (ii) an anticancer and temozolomide sensitizing effect of rabeprazole by repressing EMT.

In this study, Manilkara zapota seed oil was used for biodiesel production. The free fatty acid content of the oil was determined as 5.91 mg KOH/g and hence a two-step acid-base transesterification reaction was followed to convert M. zapota seed oil into biodiesel. The performance variables such as reaction temperature, alcohol to oil ratio, experimental duration, and catalyst weight were optimized using Taguchi orthogonal array. The study revealed a 40°C reaction temperature, 6:1 alcohol-to-oil ratio, 150 min experimental duration, and 1 wt.% NaOH catalyst that were optimal performance variables to obtain maximum conversion efficiency of 97.08%. Among the four performance variables, the alcohol-to-oil ratio was found to be the most effective variable with 84.36% of the contribution. The synthesized Manilkara zapota methyl ester (MZME) procured from seed oil with optimized performance variables matches with the standards of ASME D 6751.

BACKGROUND: The first national severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) serosurvey in India, done in May-June, 2020, among adults aged 18 years or older from 21 states, found a SARS-CoV-2 IgG antibody seroprevalence of 0·73% (95% CI 0·34-1·13). We aimed to assess the more recent nationwide seroprevalence in the general population in India. METHODS: We did a second household serosurvey among individuals aged 10 years or older in the same 700 villages or wards within 70 districts in India that were included in the first serosurvey. Individuals aged younger than 10 years and households that did not respond at the time of survey were excluded. Participants were interviewed to collect information on sociodemographics, symptoms suggestive of COVID-19, exposure history to laboratory-confirmed COVID-19 cases, and history of COVID-19 illness. 3-5 mL of venous blood was collected from each participant and blood samples were tested using the Abbott SARS-CoV-2 IgG assay. Seroprevalence was estimated after applying the sampling weights and adjusting for clustering and assay characteristics. We randomly selected one adult serum sample from each household to compare the seroprevalence among adults between the two serosurveys. FINDINGS: Between Aug 18 and Sept 20, 2020, we enrolled and collected serum samples from 29 082 individuals from 15 613 households. The weighted and adjusted seroprevalence of SARS-CoV-2 IgG antibodies in individuals aged 10 years or older was 6·6% (95% CI 5·8-7·4). Among 15 084 randomly selected adults (one per household), the weighted and adjusted seroprevalence was 7·1% (6·2-8·2). Seroprevalence was similar across age groups, sexes, and occupations. Seroprevalence was highest in urban slum areas followed by urban non-slum and rural areas. We estimated a cumulative 74·3 million infections in the country by Aug 18, 2020, with 26-32 infections for every reported COVID-19 case. INTERPRETATION: Approximately one in 15 individuals aged 10 years or older in India had SARS-CoV-2 infection by Aug 18, 2020. The adult seroprevalence increased approximately tenfold between May and August, 2020. Lower infection-to-case ratio in August than in May reflects a substantial increase in testing across the country. FUNDING: Indian Council of Medical Research.

Erratic rainfall often results in intermittent drought and/or waterlogging and limits maize (Zea mays L.) productivity in many parts of the Asian tropics. Developing climate-resilient maize germplasm possessing tolerance to these key abiotic stresses without a yield penalty under optimal growing conditions is a challenge for breeders working in stress-vulnerable agro-ecologies in the region. Breeding stress-resilient maize for rainfed stress-prone ecologies is identified as one of the priority areas for CIMMYT-Asia maize program. We applied rapid cycle genomic selection (RCGS) on two multiparent yellow synthetic populations (MYS-1 and MYS-2) to improve grain yield simultaneously under drought and waterlogging conditions using genomic-estimated breeding values (GEBVs). Also, the populations were simultaneously advanced using recurrent phenotypic selection (PS) by exposing them to managed drought and waterlogging and intermating tolerant plants from the two selection environments. Selection cycles per se (C1, C2, and C3) of the two populations developed using RCGS and PS approach and their test-cross progenies were evaluated separately in multilocation trials under managed drought, waterlogging, and optimal moisture conditions. Significant genetic gains were observed with both GS and PS, except with PS in MYS-2 under drought and with GS in MYS-1 under waterlogging. Realized genetic gains from GS were relatively higher under drought conditions (110 and 135 kg ha−1 year−1) compared to waterlogging (38 and 113 kg ha−1 year−1) in both MYS-1 and MYS-2, respectively. However, under waterlogging stress PS showed at par or better than GS as gain per year with PS was 80 and 90 kg ha−1, whereas with GS it was 90 and 43 kg ha−1 for MYS-1 and MYS-2, respectively. Our findings suggested that careful constitution of a multiparent population by involving trait donors for targeted stresses, along with elite high-yielding parents from diverse genetic background, and its improvement using RCGS is an effective breeding approach to build multiple stress tolerance without compromising yield when tested under optimal conditions.

Turbinaria ornata, tropical brown algae found in the South pacific and Indian Ocean ecosystems. In accordance with recent studies, Turbinaria ornata J. has potent anti-inflammatory effects. Therefore, this study is aimed to explore the biological activities of ethanolic extract of Turbinaria ornata J by analyzing the presence of phytochemical components, antioxidant property, antimicrobial activity and the wound healing activity. From the results, phytochemical analysis of ethanolic extract of T. ornata showed the presence of alkaloids, saponins, oils, total phenolic and total flavanoid content were estimated to be 0.683 Abs and 0.433 Abs respectively. Anti-oxidant activity of the ethanolic extract of T. ornata extract showed remarkable DPPH radical scavenging activity of about 58.8% at 200μg/ml and total anti-oxidant activity of 0.257 absorbance at 100μg/ml concentration, as compared to that of their respective controls. The ethanolic extract of T. ornata exhibited the maximum zone of inhibition against the clinical pathogens like Pseudomonas aeuruginosa, Escherichia coli, Staphylococcus aureus, Candida albicans and Methicillin-resistant Staphylococcus aureus with their potent anti-microbial activity. Wound healing effects of the ethanolic extract of T. ornata was analysed by using Zebra fish model. The results showed the rapidand significant regeneration of the wounded caudal fin on day 14. Therefore, the preliminary results of this study strongly supports the ethanolic extract of T. ornata may be effective in wound healing and regeneration of the wounded tissues.

Background Biomedical research, recently, focus more on searching for biomasses that contain extractable biologically active components for formulating new drugs. Halophytes growing in hyper saline conditions are expected to produce stress alleviating bioactive compounds. These phytochemicals could be the better raw materials for formulating new drugs. Methods The purpose of this work was to describe physiologically active compounds from Acanthus ilicifolius and Heliotropium curassavicum that had antimicrobial, antioxidant and nutraceutical properties utilizing a variety of solvents. Analysis of bioactive compounds included the application of common phytochemical screening assays, proximate analysis, FTIR analysis and antioxidant assays. The disc diffusion technique was used to determine the antibacterial activity of the plant extracts. Results Highest extraction yield was observed with methanol. A. ilicifolius methanolic extracts included a variety of bioactive components, including alkaloids, saponins, phenolics, flavonoids, steroids, cardiac glycosides, tannins, and terpenoids. H. curassavicum extracts showed the presence of all the phytochemicals except cardiac glycosides. The overall phenolic concentration and antioxidant capacity of A. ilicifolius were substantially greater. The antimicrobial assays explored that among the tested bacterial pathogens viz., Bacillus subtilis, Escherichia coli, Klebsiella pneumoniae, Staphylococcus aureus, Streptococcus pneumoniae, significant level of control was observed in E. coli, K. pneumoniae and B. subtilis, which were significantly susceptible to both the plant extracts at a concentration of 50 μg/ml. Conclusion The reports from the current investigation explored the possibility of utilizing these halophytes in nutraceutical formulations. The current study sheds light on the possibility of halophytes as natural secondary metabolites and bioactive chemicals with potential for antimicrobials.

The present study enumerates the effectivity (in vivo) of ethanolic leaf extract of Azadirachta indica against circulating microfilariae (mf) of Dirofilaria immitis in Canis familiaris. An ethanolic extract was prepared from the leaves of A. indica (EEA) and its efficacy was evaluated after oral treatment in dogs infected with the filarial nematode, D. immitis, the causative agent of canine cardiopulmonary dirofilariasis. Before treatment, all the infected dogs were vigilantly supervised for any natural fluctuation of mf count. Two doses; 25 and 50 mg/kg body weight/twice a day; both for 15 days were selected for in vivo tests along with a control group that received an empty capsule during the study period. The highest reduction of circulating mf was counted on day 60, showing mf reduction of 77.9% and 86.7% respectively for the two doses. Thereafter mf density increased with a minor change and maintained a reduction of 49.5% and 64.1% on day 180 respectively. Additionally, no appreciable side effects in the treated dogs were recorded as evident from serum toxicity parameter analyses. In conclusion, the ethanolic formulation of A. indica leaves possesses considerable effectiveness against D. immitis in vivo with no toxic modification in the host after exposure. Thus, EEA appears to be a good alternative remedy against heartworm infection in infected dogs.

Aim: The present study was conducted to evaluate oil palm dura progenies developed from Zambia and Cameroon germplasm sources for bunch components for the selecting of high oil yielding mother palms. Methodology: Total 77 dura genotypes were evaluated by considering 15 different bunch components. Bunch analysis data were collected from tenth year onwards after planting. The genetic diversity, Principal component analysis, correlation and path analysis of the genotypes were estimated. Results: Study revealed that oil-to bunch ratio depends on fruit-to-bunch ratio and mesocarp-fruit ratio. The ANOVA value showed a significant variability among the genotypes for most of the bunch traits and the identified genotype P35 (24.53 %) was highest oil yielder. Positive significant correlation (p≤ 0.01) of mesocarp-to-fruit ratio and oil-to-dry mesocarp with oil-to-bunch ratio were noted in this study. Based on path analysis, the information obtained from this study is the oil-to-wet mesocarp ratio (0.697) showed the highest significant positive direct effect on oil-to-bunch ratio followed by mesocarp-to-fruit ratio (0.570). Interpretation: This study may support for the future selection of mother palms to utilize in oil palm improvement programmes. Introgression into existing breeding materials and breeding for seed production utilising selected palms.

Drought affects the rainfed groundnut (Arachis hypogaea L.) at different phases of development and it is the serious threats on groundnut productivity causing losses than any other abiotic factor under rainfed agriculture. In the world's semiarid regions, groundnut accounts for 90% of worldwide production. Drought mainly affects the pace and pattern of nutrient and water intake from the soil, affecting the architecture of the groundnut root system. Plant selections with desirable root trait have been a major focus in developing drought resistant Groundnut cultivars. In 2019, 60 groundnut genotypes were cultivated in root block design with two different soil water treatments, as well as in the field during the year under same circumstances. The purpose of this study was to see how different groundnut cultivars fared in terms of yield, yield contributing features, root characters, and their relationships with drought tolerance. Drought resistant genotypes had thicker roots, larger roots, and a deeper root system than susceptible genotypes. Recent series in groundnut genotypes of 60 numbers were sown during kharif 2019 (july-september) under rainfed condition (It includes life irrigation and rainfall received during cropping season). Groundnut genotypes were semi spreading with the duration of 110-120 days. Observation on root morphological character viz., roots length, root volume after 20 days of stress imposition of the crop and yield parameters were observed at the harvest. Among the 60 genotypes, 20 genotypes (VG 17008, VG 17046, VG 18005, VG 18102, VG 18077, VG 19572, VG 19709, VG 18111, VG19561, VG19576, VG 19620, VG 19681, VG 19688 etc.,) similarly, yield character were observed for 60 genotypes and all the genotypes given above recorded higher value in Total number of pods per plant, Number of double seeded pods per plant, Pod yield per plant, Harvest index and Total dry matter production. The methods used in this study identified correlation between yield character and root characters. Groundnut genotypes by assessing yield metrics and their relationship with root trait. These findings lay the groundwork for future study aimed at deciphering the molecular pathways underpinning Groundnut drought resistance.

CRISPR-Cas systems are RNA-guided nucleases that provide adaptive immune protection in bacteria and archaea against intruding genomic materials. Cas9, a type-II CRISPR effector protein, is widely used for gene editing applications since a single guide RNA can direct Cas9 to cleave specific genomic targets. The conformational changes associated with RNA/DNA binding are being modulated to develop Cas9 variants with reduced off-target cleavage. Previously, we showed that proline substitutions in the arginine-rich bridge helix (BH) of Streptococcus pyogenes Cas9 (SpyCas9-L64P-K65P, SpyCas92Pro) improve target DNA cleavage selectivity. In this study, we establish that kinetic analysis of the cleavage of supercoiled plasmid substrates provides a facile means to analyze the use of two parallel routes for DNA linearization by SpyCas9: (i) nicking by HNH followed by RuvC cleavage (the TS (target strand) pathway) and (ii) nicking by RuvC followed by HNH cleavage (the NTS (nontarget strand) pathway). BH substitutions and DNA mismatches alter the individual rate constants, resulting in changes in the relative use of the two pathways and the production of nicked and linear species within a given pathway. The results reveal coordinated actions between HNH and RuvC to linearize DNA, which is modulated by the integrity of the BH and the position of the mismatch in the substrate, with each condition producing distinct conformational energy landscapes as observed by molecular dynamics simulations. Overall, our results indicate that BH interactions with RNA/DNA enable target DNA discrimination through the differential use of the parallel sequential pathways driven by HNH/RuvC coordination.

Surgical cotton production has drastically been increased in the past few years due to excessive use by medical health professionals especially in countries like India, which is among the top three exporters of cotton worldwide. The effluent generated from surgical cotton industries differ from textile effluents by the conspicuous absence of dyeing chemicals. This wastewater has a high concentration of suspended particles, COD, dissolved ions, organic carbon, and alkaline pH. Several studies have been published on the treatment of textile effluents and the degradation of dyeing chemicals, while the treatment studies on surgical cotton wastewater have been rarely reported in spite of their potential to cause pollution in receiving land/water bodies. Activated sludge microbes have been extensively studied and well documented in the treatment of several industrial effluent but does not match to the production of valuable biomass from algae. The global energy demand has prompted the scientific community to investigate and explore the possibility of using algae for energy production with simultaneous wastewater treatment. To the best of the authors' knowledge, no research articles have been published which compare the effectiveness of activated sludge microorganisms, microalgae, and macroalgae in removing contaminants from real wastewater. To date, there is a knowledge gap in understanding and selecting the right choice of biological system for effective and economical effluent treatment. In an attempt to minimize this gap, carbon removal by microalgae, macroalgae, and activated sludge microbes were investigated on real effluent from surgical cotton industries. It was observed that the strain of Chlorella vulgaris could dissipate 83% of COD from real wastewater, while consortia of macroalgae (consisting predominantly of Ulvaceae and Chaetomorpha) and activated sludge microbes could remove 81% and 69% of the carbon, respectively. The microalgal growth (in terms of wet weight) increased from 0.15 to 0.3 g, whereas the macroalgal wet weight increased from 1.5 to 3 g in over 7 days of batch experiments conducted in triplicates. This indicated the superlative performance of microalgae over activated sludge microbes in carbon dissipation.

Recently, in India, the marine finfish culture is expanding rapidly without adequate knowledge and research information on the coastal and marine finfish disease epidemics. Therefore, for the benefit of farmers, this study was designed to find the prevalence of Vibriosis outbreaks in marine finfish farms. In this study, 167 moribund fishes that show outward symptoms of lethargy, popeye, pale gills, skin erosions, abdominal edema, and hemorrhagic spots on different parts of the body were collected from 26 marine finfish farms (Lates calcarifer, Rachycentron canadum, and Trachinotus blochii) located in the states of Tamil Nadu, Andhra Pradesh, and Kerala, India. A total of 173 bacterial isolates were retrieved from all the fish samples, of which 144 isolates demonstrated a Vibrio-like character during biochemical and Vibriostatic 0/129 examinations. The phenotypic, 16s RNA gene sequencing and histopathological analysis confirm that the isolated bacterial strains were Vibriospecies. In the collected fish samples, the total prevalence of Vibrio species was 83.2% and the remaining 16.8% were other bacterial species. Among these bacteria, V. harveyi was the most predominant bacterium (34.1%) that cause infection in all the three fish species whereas Photobacterium damselae was found to be the least (2.9%) one. In the pathogenicity study, the highest mortality of 83% was observed after 168 h, when the seabass fingerling exposure to V. harveyi at 106 CFU/ml, and established similar clinical signs to the disease specimens of farmed fish. The in vitro antibacterial study confirms that the tested probiotic effectively inhibits the V. harveyi at 125 µl (108 CFU/ml). The above results inspire further in-depth studies on the probiotic efficacy to control the V. harveyi in finfish farms.

Phyllanthus is a large genus of shrubs, trees and rare herbs of the family Euphorbiaceae, comprising more than 600 species [1]. The aim of this project was to identify the phytochemicals present in P. niruri and the antidiabetic activity of the ethanolic extracts of the plant. The phytochemical analysis for alkaloids, flavonoids, anthraquinone, phenols, saponins, tannins, alkaloids, total phenolics, total flavonoids, carbohydrates and amino acids were made by following standard procedures. The antidiabetic property of the plant extract was evaluated using insulin as standards through inhibition of alphaamylase enzyme activity. The presence of saponins, alkaloids, tannins, flavonoids, phenols, terpenoids, carbohydrates, coumarins, anthraquinones and amino acids was observed. The total phenolic content in the extract was measured using catechol (standard) and a graph was plotted with concentration on X axis and absorbance on the Y axis. The total phenolic content was calculated. The calibration curves of α-amylase inhibition of Insulin and P. niruri was used to calculate the IC50 value.

Context and Aim: A plethora of studies have revealed that there is a lack of general consensus in the precise anatomic detailing of the canals and the major foramen in both the maxillary and mandibular teeth while emphasizing the significance of the same for a successful endodontic treatment. The aim of the present study was to determine the variations in canal morphology, shapes, and positions of major foramen in maxillary and mandibular teeth. Materials and Methods: In the present study, 420 extracted human maxillary and mandibular teeth with completely formed apices were included. The specimens were cleaned for surface debris including remnants of periodontal tissue and were stored in saline. Examination of the apices was performed with the help of stereomicroscope for their exact shapes and configurations and precise anatomic location. Comparison of different parameters within the group was done using Chi-square test while P < 0.05 was considered statistically significant. Results: The results of the present study indicated that apical foramen in mandibular teeth showed higher degree of deviation with a prevalence of 70.2% while flat shape of apical foramen was observed only in maxillary teeth. Furthermore, maximum deviation in maxillary teeth was recorded in the canines with a prevalence of 90% while, in mandibular teeth, maximum deviation was recorded with mandibular second premolars with a prevalence of 79.12%. Conclusions: The results of the present study revealed that deviation of apical foramen from the root apex was seen in 68.2% of the specimens. Furthermore, deviation was greater in the mandibular than the maxillary teeth.

In this study, the optimisation of a process for producing medium-chain-length polyhydroxyalkanoate (mcl-PHA) by Pseudomonas putida KT2440 when fed with a polyethene (PE)-derived fatty acid mixture was investigated. The PE was pyrolysed to produce a hydrocarbon wax that was subsequently oxidised to produce a mixture of fatty acids, purified, and used as a PHA substrate for the growth and selection of microorganisms. Based on the shaken flask screening, a production strain, i.e., Pseudomonas putida KT2440, was selected for conducting bioreactor studies. Feeding PE-derived fatty acids in a 20-L setup resulted in high mcl-PHA yields (83.0 g L−1 CDW with 65% PHA in 25 h). Furthermore, life-cycle assessment (LCA) was conducted to determine the environmental advantages of the proposed process and its impacts compared to those of other technologies for treating PE-derived waste streams. We conclude that processing waste PE into PHA, rather than incineration, produces biodegradable material while also reducing the additional emissions that arise from traditional PE waste treatment processes, such as incineration to gain energy.

Zinc (Zn) plays a role in the antioxidant capacity and immunity of aquatic animals. A twelve-week feeding experiment was performed to estimate the impact of dietary zinc on antioxidant enzyme-related gene expression, antioxidant enzyme activity and non-specific immune functions of soft-shelled turtles, Pelodiscus sinensis. Six fishmeal-based experimental diets with 32.45% protein were formulated, which contained 35.43, 46.23, 55.38, 66.74, 75.06 and 85.24 mg/kg Zn, respectively. Catalase (CAT), glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) levels improved with an elevation in dietary Zn from 35.43 to 55.38 mg/kg and then reduced when dietary Zn was further elevated. The expression levels of Nrf2 and antioxidant-related genes CuZnSOD, MnSOD, CAT, GPX1, GPX2, GPX3 and GPX4 escalated with elevating Zn concentration up to 55.38 mg/kg in diets and then reduced as dietary Zn elevated. The expression levels of Kelch-like ECH-associating protein 1 (keap1) showed a reverse trend with that of Nrf2. The contents of malondialdehyde (MDA) in the 55.38 and 66.74 mg/kg Zn diet-fed groups were the lowest. Alkaline phosphatase activity (AKP), superoxide anion (O2−), lysozyme activity and total antioxidant capacity (T-AOC) improved with an escalation in dietary Zn concentration up to 66.74 mg/kg. Optimal dietary Zn improved antioxidant capability, immunity, and antioxidant enzyme-related gene expression. The dietary Zn demand for soft-shelled turtles were 60.93 and 61.63 mg/kg, based on second regression analysis of SOD and T-AOC activity, respectively.

The objective was to investigate the anti-urolithiatic effect of ethanolic extract of Cassia auriculate on ethylene glycol (EG) induced urolithiasis in experimental rats. The animals were divided into five groups of six animals each. Except the normal group all the other groups received ethylene glycol 0.75% and Ammonium chloride 1% v/v in water induce orally for 28 days. Normal groups received plain water orally. The standard group received cystone 750mg/kg b.w orally. Test groups received EECA 200mg/kg and 400mg/kg b.w orally. On the 28th day, blood and urine samples were collected and used for estimation of biochemical parameters such as calcium, phosphates, oxalates, creatinine, uric acid, SOD, and CAT followed by histopathological studies. Treatment with EECA was found to exert dose dependent anti urolithiatic action. Increased urine volume in EECA treated groups as compared to diseased group was indicative of diuretic property. Elevated calcium, phosphate and oxalate levels in diseased group animals were found to be decreased in animals treated with EECA. Increased levels of serum calcium, creatinine and uric acid were considerably brought down towards normal values in proportion to EECA doses administered. Antioxidant parameters like SOD and CAT were decreased along with significant increase in MDA levels which is the main product of lipid peroxidation in EG treated rats. However the rats treated with EECA showed significant improvement in these parameters. Hence it is concluded that the ethanolic extract of Cassia auriculata possess anti-urolithiatic activity.

Highly efficient methodology was developed for the construction of functionalized Kojic acid involving Click linker via 1,3-dipolar cycloaddition and their cytotoxicity against MCF-7, MIAPaCa-2 and DU145 mammalian cell lines were evaluated. Preliminary studies on structure-activity-relationship (SAR) revealed that substitution at C-2 of kojic acid as well as C-5 of 1,2,3-triazole motif played a major role in the activity profile. Kojic acid 1,2,3-triazole analogue 3 b containing an alkyl chain (n = 6) exhibited two fold potent activity than the parent compound, kojic acid against MCF-7 and MIA PaCa-2 cell lines. It induced apoptosis in these cell lines via ID1/PARP1 mediated pathway. The structures of the new analogues of kojic acid 1,2,3-triazole were confirmed by the detailed spectroscopic data analysis.

In India, Labeo rohita is widely cultured and consumed freshwater fish. Aeromonads are etiological agents of major bacterial fish diseases like furunculosis, haemorrhagic septicaemia, skin ulcers, fin/tail rot and dropsy, causing significant economic losses in carp culture. Aeromonas species are widely distributed in aquatic environment which is considered as important vehicle of Aeromonas infections to fish and humans. Some of the Aeromonas spp. causes gastroenteritis, septicaemia, peritonitis, meningitis and eye infections in humans. In the present study Aeromonas species were isolated from diseased freshwater fish Labeo rohita collected from two districts viz., West Godavari and SPSR Nellore of Andhra Pradesh, India. Background: Inflammation has an important role in the initiation and progression of carcinoma cervix. The measurement of inflammatory biomarkers is a cost-effective method of identifying patients at high risk of recurrence after treatment. This study was done to identify the influence of pre-treatment inflammatory biomarkers on survival in patients treated with radical chemo-radiation or radiation. Methods: Patients with biopsy proven carcinoma cervix treated with Radiotherapy or Chemo-radiation from January 1st, 2016 to September 30th, 2017 were included. Pre-treatment complete blood counts, differential counts, serum albumin and C-reactive protein (CRP) were obtained. Neutrophil lymphocyte ratio (NLR), platelet lymphocyte ratio (PLR) and CRP Albumin ratio (CAR) were calculated. The best cut off values for NLR, PLR, serum albumin, CRP and CAR were found out from receiver operating characteristic (ROC) curves. OS (Overall Survival) and DFS (Disease Free Survival) were estimated using Kaplan -Meier method. The prognostic value of inflammatory biomarkers on survival was assessed by cox regression model. Results: Sixty-three patients were included. The median follow up was 42.5 months. The best cut off values for NLR, PLR, albumin, CRP and CAR from the ROC curve were 2.36, 122.725, 3.95, 0.65 and 0.8 respectively. The three-year OS and DFS probability were 68.3% and 63.5% respectively. Patients with CAR >0.8 had 5.7 times more risk of death and 6.01 times risk of relapse or progression compared to patients with CAR ≤0.8. Conclusion: Patients with a CRP-Albumin ratio of more than 0.8 are at significant risk of relapse and death after chemoradiation or radiation.

Heat waves and droughts are projected to become more widespread as a result of climate change. At the same time, CO2 levels are predicted to have doubled by 2100. The stomatal regulation and gas exchange characteristics were assessed in 25 days old plants of green gram (var Co 8) by exposing them to six different treatments namely, T1: a [CO2] + a T+ irrigation (100%), T2: a [CO2] + a T+ irrigation (50%), T3: a [CO2] + e T (40ºC) + irrigation (100%), T4: e [CO2] – 800 ppm + a T+ irrigation (100%), T5: a [CO2] + combined stress [e T (40ºC) + irrigation (50%) T6: e [CO2] – 800 ppm + combined stress [e T (40ºC) + irrigation (50%)]. The experiment was carried out using Completely Randomized Design (CRD) with three replications. All gas exchange parameters viz., ((photosynthesis rate, stomatal conductance, and transpiration rate) were determined before imposing stress and two weeks after imposing stress. Stomatal characters was examined two weeks after imposing stress. Elevated CO2 stress caused a reduction in stomatal frequency accompanied by larger stomatal size. The study revealed the positive effect of higher CO2 concentration on gas exchange traits of the C3 crops viz., green gram.

Fanged sea bream, Sparidentex jamalensis Amir et al. (Pak J Zool 46(2):471–477, 2014) has been recently recorded for the first time in Indian waters from Korapuzha, Kadalundi and Moorad estuaries, Calicut, Kerala. Previously this species was reported only from Pakistan waters globally and till date there are no other reports available on the distribution of this species elsewhere. The morphometric measurements and meristic counts of the specimens collected in the present study were similar to those specimens reported from type locality with few variations. The variations observed in comparison with specimens from Pakistan are the counts of dorsal-fin spines (11–12 vz 11); dorsal-fin rays (11–12 vz 11); anal-fin rays (81/2 – 9 vz 8) pored lateral-line scales (45- 48 vz 45- 47) and in the number of gill rakers in the upper limb (6- 9 vz 6- 7). The difference observed in morphometric and meristic characters owes to the origin of species from two different geographical regions or genetic differences or environmental impact, or combination of all the above. The present report indicates an extension of the geographical distribution range of S. jamalensis from its localities. Identification of the species was further confirmed using DNA barcoding.

Greengram (Vigna radiata L. Wilczek) is the third most important pulse crop and drought is the most severe constraint to greengram growth and productivity. The present study was conducted to identify the drought tolerant greengram genotypes. Four greengram varieties used for standarization of drought stress using Polyethyleneglycol (PEG) 6000. The effect of water stress caused by different concentration of PEG 6000 are control (0 MPa), -0.4MPa, -0.5 MPa, -0.6MPa and -0.7 MPa. Increasing PEG concentration decrease the germination percentage, root length, shoot length, fresh weigh and dry weight of seedlings. At -0.5 MPa shows 50% seedling mortality , So control and -0.5 MPa level of drought stress was used for screening the greengram genotypes. Under PEG induced drought situations, parameters such as germination percentage, growth indices and proline content were recorded in all greengram genotypes. Compared to control, PEG induced drought stress (-0.5MPa) decrease all these parameters studied, where as drought has increased the proline content in all greengram genotypes screened. Among the greengram genotypes VGG17019 and VGG17004 posses higher germination percentage, GSI and proline content indicates high level of tolerance to drought stress.

In children following asthma and epilepsy, diabetes is the third most prevalent chronic condition. More recently, the cumulative incidence of diabetes has started to grow significantly among children and adults. This has been partly influenced by the obesity pandemic in youth. Understandably, the health agencies and countries coping with the disease's important morbidity with possible grave complications have been placing an economic strain. More treatment discoveries have since broadened the list of alternatives for the drugs available. The use of plants to treat different human conditions is stated in Ayurveda and other Indian literature. India has approximately 48000 plant species and several thousand of them have claimed to have medicinal properties. Ants diabetic properties have been seen in research on plants described or historically used in ancient literature in the last few decades. The area of herbal medicine has grown exponentially in recent years, and both in the development and developed countries these medications are increasingly common due to their natural sources and fewer side effects. This study is aimed to provide information about the plant's anti-diabetes function based on the main evidence gathered from web search pages from different literature and science publications.

Platelet-rich fibrin (PRF) is a fibrin matrix in which platelet cytokines, growth factors, and cells are trapped and may be released after a certain time and that can serve as a resorbable membrane. It can be obtained from blood with the help of a simple process. PRF is basically a concentrate of growth factors that promote wound healing and regeneration, which is used in various disciplines of dentistry to repair various lesions and regenerate dental and oral tissues. Although the use of PRF is well-documented, its use in pediatric dentistry remains unexplored, due to its restrictions and on account of the fact that it is a blood-derived product. PRF has been used globally to enhance tissue healing. This article provides an insight into the use of PRF in pediatric dentistry, its benefits, limitations, and recommendations.

Objective The mandible has been used for sex determination and forensic identification due to its unique anatomy and morphology. Every part of the mandible is unique, including the shape and size of the temperomandibular joint, the ramus, the body and the symphysis region, and the inferior alveolar canal. In addition, the position and placement of the teeth within their sockets are unique for every individual. Sample Population A study was conducted on 20 males and females to estimate sexual dimorphism using anthropometric measurements obtained by cone beam computed tomography images of the mandible of Karnataka and Kerala populations. Materials and Methods The mean, standard deviation, and standard error along with the confidence interval of different measurements were documented. Various measurements between the sexes were compared using Student's t-test. Association between categorical variables was analyzed using the Chi-square test. A p-value Results The p-value of mRBr L, GA L, and GA R among females was significantly higher in the Karnataka population when compared with that in the Kerala population. The Karnataka State population showed significantly higher values of RL R, BiGBr, and BiCBr among males, and the p-value of the Kerala population for GGL L and GGL R was significant and higher among the males. Conclusion The data derived from the above study suggest that the mandibular anthropometric measurements used may be applied for forensic sex estimation.
