Pleurotus ostreatus (oyster mushroom) synthesizes enzymes that degrade lignin, cellulose, and hemicellulose. The objectives of this study were to evaluate the effect of Pleurotus ostreatus spent substrate (POSS) on whole-plant corn silage (WPCS) chemical composition, antioxidant capacity, lignin monomers, and in vitro digestibility, as well as the performance of lactating goats fed corn silage treated with different levels of POSS. In experiment 1, 4 levels of lignocellulolytic enzymes were tested in a complete randomized design: 0, 10, 20, and 30 mg of lignocellulosic enzymes per kilogram of fresh matter, 4 replicates per treatment (vacuum-sealed bags). The bags were opened 60 d after ensiling. In experiment 2, corn silage treated with 3 enzyme levels (0, 10, or 30 mg/kg of fresh matter) was fed to lactating goats as part of the total mixed ration. Nine lactating Saanen goats (62.68 ± 7.62 kg BW; 44 ± 8 d in milk; 2.91 ± 0.81 kg of milk/day, mean ± SD) were assigned to three 3 × 3 Latin squares. Data were analyzed using the GLIMMIX procedure of SAS (version 9.4, SAS Institute Inc.), and means were compared by linear and quadratic orthogonal contrast. In experiment 1, neutral detergent fiber (NDF), acid detergent fiber (ADF), lignin, and cellulose quadratically decreased in the WPCS treated with POSS. At the nadir point, POSS decreased NDF by 14.1%, ADF by 19.5%, lignin by 9.07%, and cellulose by 22.1% compared with the untreated silage. Therefore, POSS led to a quadratic increase in in vitro dry matter digestibility of WPCS (+8.88% at the vertex) compared with the untreated silage. In experiment 2, POSS quadratically increased the in vivo total-tract ADF digestibility. Also, the concentration of polyphenols in the milk of goats linearly increased with the addition of POSS, and no differences were observed among treatments for milk yield and composition. In summary, adding 10 mg of lignocellulolytic enzymes from POSS per kilogram of fresh matter of whole-plant corn at ensiling had a more evident reduction in lignin and cellulose concentration, leading to greater in vitro digestibility, as well as greater in vivo ADF digestibility; however, milk yield was not different among treatments.

Lipopolysaccharides (LPS) are cell wall components from Gram-negative bacteria and are composed of three covalently linked regions: the O-antigen, the core oligosaccharide, and the lipid A moiety, which carries most of their endotoxic activity. The objective of this study was to isolate and compare the lipid A structures from ruminal LPS derived from total mixed ration (TMR)- and pasture-fed cows, by using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). Ruminal bacteria were collected from two rumen-cannulated Holstein cows; one fed a TMR (60:40, forage–concentrate) and the other pasture fed. The representativeness of each sample was validated by comparing the rumen microbiome from the cows in our study to the core rumen microbiome from the previous literature. Lipopolysaccharides from each respective sample were extracted with a phenol–water extraction procedure and purified via ultracentrifugation. To isolate lipid A from the core and O-antigen, pure ruminal LPS samples were hydrolyzed with acetic acid. Lipid A derived from the TMR-fed cow potentially exhibited a tetra-acylated structure, whereas lipid A derived from the pasture-fed cow potentially exhibited a penta-acylated lipid A structure. Both samples were quantified using limulus amebocyte lysate (LAL) assay and exhibited low endotoxic activity, consistent with the MALDI-TOF MS observations. Results indicate that the lipid A acylation pattern differs between diets, and that ruminal bacteria express solely under-acylated lipid A structures contrary to hexa-acylated lipid A, typically expressed by bacteria such as E. coli.

Silage treated with lactic acid bacteria inoculants has been reported to increase ruminal microbial biomass when tested in vitro. Therefore, we tested if alfalfa silage inoculated with Lactobacillus plantarum MTD-1 would improve ruminal N metabolism and increase milk production in high-producing dairy cows. Twenty-eight early lactation Holstein cows (8 ruminally cannulated) were blocked by DIM and milk production; animals were used in a double crossover design consisting of four 28-d periods. Animals in each block were randomly assigned to 2 treatments: a diet containing uninoculated alfalfa silage (control) and a diet containing alfalfa silage inoculated with L. plantarum MTD-1 (LP). Diets were formulated to contain 50% of alfalfa silage, 16% crude protein, and 25% neutral detergent fiber (dry matter basis). Milk production and dry matter intake were recorded in the last 14 d of each period. Milk samples were collected twice at both daily milkings on d 20, 21, 27, and 28 of each period. On d 22, omasal samples were collected from the cannulated animals over a period of 3 d to quantify ruminal digestibility and nutrient flows. Data were analyzed using mixed models of SAS 9.4 (SAS Institute). Compared to the control, cows receiving the LP treatment had greater milk production (40.4 vs. 39.6 kg/d) and lower milk urea nitrogen concentration (11.6 vs. 12.7 mg/dL), despite minor changes in energy-corrected milk. Milk lactose concentration was greater in the milk produced by cows fed the LP treatment, which reflected a tendency for increased milk lactose yield. Although milk true protein concentration was lower for cows in the LP treatment, milk true protein yield was the same on both control and LP treatments. Improvements in milk production of animals under the LP treatment were associated with greater organic matter truly digested in the rumen, especially ruminal neutral detergent fiber digestion. Minor changes were observed in total omasal microbial nonammonia N flow in cows receiving the LP treatment. Therefore, alfalfa silage treated with L. plantarum MTD-1 may improve ruminal fermentation and milk production; however, because of a lack of response in ruminal N metabolism, these changes did not result in greater energy-corrected milk in high-producing dairy cows.

The purpose of this literature review is to evaluate current research into and understanding of whole-plant sorghum silage production and the effect of feeding whole-plant sorghum silage on lactation performance of dairy cows. Sorghum's drought tolerance, water efficiency, and low cost of production make it an intriguing crop in areas where whole-plant corn silage production may be limited. Currently, urban land encroachment and reduced water availability have increased social and economic pressures on farms to improve crop production efficiency. As these challenges become more prevalent, greater reliance on sorghum can be expected because of its ability to produce high dry matter yields while maintaining nutritive value, even under less-than-ideal growing conditions. Moreover, whole-plant sorghum silage provides both physically effective fiber and energy through fiber and grain fractions. Advancements in sorghum genetics and mechanical processing have the potential to alleviate common challenges associated with whole-plant sorghum silage supplementation, such as increased neutral detergent fiber and decreased neutral detergent fiber digestibility, starch concentration, and starch digestibility. These nutritive challenges must be overcome for whole-plant sorghum silage to be a viable alternative to whole-plant corn silage.

This study aimed to evaluate levels of yerba mate (Ilex paraguariensis) extract (YME), as a feed additive in the diets of growing lambs on serum biochemical parameters and hematological indices, animal performance, body metrics and carcass traits. Thirty-six entire (nine per treatment), male growing lambs, weighing 23.8 &plusmn, 3.7 kg, were fed the experimental diets which were treatments consisting of increasing levels of YME (0, 1, 2, and 4% inclusion on a dry matter [DM] basis) during an experimental period of 53 days. The experiment was carried out in a randomized block design, which initial body weight was used as blocking factor and the results were analyzed by orthogonal contrasts (linear, quadratic, and cubic). Yerba mate extract did not change the general health status of the animals, however, inclusions of up to 2% of the extract increased globulins (p = 0.05) and white blood cell count, as segmented neutrophils (p = 0.02) and lymphocytes (p = 0.04). Additionally, inclusion of up to 2% YME increased dry matter intake, final weight gain, total and daily gain (p &lt, 0.05), also tended to increase ribeye area and reduce fat thickness (p &lt, 0.10), however, YME above 2% of inclusion reduced animal productive parameters (p &lt, 0.05). In conclusion, levels up to 2% of YME were beneficial to the health and productive parameters of growing lambs.

Thirty-six uncastrated male lambs (90 d old) were fed experimental diets, which treatments consisted of 0%, 1%, 2%, and 4% inclusion of YME. Animals were fed for 50 d before slaughter. Muscle and meat samples were collected for metabolomics and meat quality analysis, respectively. The experiment was carried out in a randomized block design and analyzed using orthogonal contrasts. There was a quadratic effect of YME inclusion in tenderness (P < 0.05) and a positive linear effect on meat lightness (P < 0.05). No qualitative changes (P > 0.05) on individual metabolites were observed; however, changes in the quantitative metabolic profile were observed, showing that animals fed 1% and 2% of YME have a greater concentration of desirable endogenous muscle antioxidants, with direct impact on metabolic pathways related to beta-alanine metabolism and glutathione metabolism. Therefore, YME dietary supplementation up to 2% of the diet to lambs had little to no effects on the majority of meat quality traits evaluated; moreover, 4% of YME inclusion negatively affected feed intake and meat quality traits. The objectives of this study were: 1) to compare the effects of live yeast (LY), yeast fermentation product (YFP), a mix of Lactobacillus acidophilus and Propionibacterium freudenreichii (MLP), and Lactobacillus plantarum included as additives in dairy cows’ diets on in vitro ruminal fermentation and gas production (GP); and 2) to evaluate the effects of L. plantarum as direct-fed microbials (DFM) in dairy cows’ diets on in vitro ruminal fermentation, GP, nutrient digestibility, and N metabolism. Three experiments were carried out: Exp. 1 had the objective to compare all additives regarding ruminal fermentation parameters: an Ankom GP system was used in a completely randomized design, consisting of four 48 h incubations, and eight replications per treatment. There were eight treatments: a basal diet without additive (CTRL) or with one of the following additives: LY, YFP, MLP, or L. plantarum at four levels (% of diet Dry Matter (DM)): 0.05% (L1), 0.10% (L2), 0.15% (L3), and 0.20% (L4). In Exp. 2, a batch culture was used to evaluate ruminal fermentation, and CO2 and CH4 production using the same treatments and a similar experimental design, except for having 16 replications per treatment. Based on Exp. 1 and 2 results, Exp. 3 aimed at evaluating the effects of the L. plantarum on ruminal true nutrient digestibility and N utilization in order to evaluate the use of L. plantarum as DFM. The treatments CTRL, MLP, L1, and L2 were used in a replicated 4 × 4 Latin square design using a dual-flow continuous culture system. Data were analyzed using linear and nonlinear regression; treatment means were compared through contrasts, and L treatments in Exp. 1 and 2 were tested for linear, quadratic, and cubic effects. In Exp. 1, all treatments containing additives tended to reduce OM digestibility as well as reduced total volatile fatty acids (VFA) concentration and total GP. The YFP had greater OM digestibility than LY, and MLP treatment had greater total VFA concentration compared to L. plantarum treatments. In Exp. 2, additives reduced CO2 production, and there were no major differences in CH4. In Exp. 3, all additives reduced NH3-N concentration. In conclusion, pH and lactate concentration were not affected in all three experiments regardless of additive tested, suggesting that these additives may not improve ruminal fermentation by pH modulation; and L. plantarum may improve ruminal N metabolism when used as DFM in high-producing dairy cows’ diets, mainly by reducing NH3-N concentration.

The objective of this study was to examine the enzyme activities of an enzymatic complex produced by Pleurotus ostreatus in different pH and the effects of adding increased application rates of this enzymatic complex on the fermentation profile, chemical composition, and in situ ruminal disappearance of whole-plant corn silage (WPCS) at the onset of fermentation and 30 d after ensiling. The lignocellulolytic enzymatic complex was obtained through in vitro cultivation of P. ostreatus. In the first experiment, the activities of laccase, lignin peroxidase (LiP), manganese peroxidase, endo- and exo-glucanase, xylanase, and mannanase were determined at pH 3, 4, 5, and 6. In the second experiment, five application rates of enzymatic complex were tested in a randomized complete block design (0, 9, 18, 27, and 36 mg of lignocellulosic enzymes/kg of fresh whole-plant corn [WPC], corresponding to 0, 0.587, 1.156, 1.734, and 2.312 g of enzymatic complex/kg of fresh WPC, respectively). There were four replicates per treatment (vacuum-sealed bags) per opening time. Bags were opened 1, 2, 3, and 7 d after ensiling (onset of fermentation period) and 30 d after ensiling to evaluate the fermentation profile, chemical composition, and in situ dry matter and neutral fiber detergent disappearance of WPCS. Laccase had the greatest activity at pH 5 (P < 0.01), whereas manganese peroxidase and LiP had the greatest activity at pH 4 (P < 0.01; P < 0.01). There was no effect of the rate of application of enzymatic complex, at the onset of fermentation, on the fermentation profile (P > 0.21), and chemical composition (P > 0.36). The concentration of water-soluble carbohydrate quadratically decreased (P < 0.01) over the ensiling time at the onset of fermentation, leading to a quadratic increase of lactic acid (P = 0.02) and a linear increase of acetic acid (P = 0.02) throughout fermentation. Consequently, pH quadratically decreased (P < 0.01). Lignin concentration linearly decreased (P = 0.04) with the enzymatic complex application rates at 30 d of storage; however, other nutrients and fermentation profiles did not change (P > 0.11) with the enzymatic complex application rates. Addition of lignocellulolytic enzymatic complex from P. ostreatus cultivation to WPC at ensiling decreased WPCS lignin concentration 30 d after ensiling; however, it was not sufficient to improve in situ disappearance of fiber and dry matter.

The objective of this study was to adapt existing in vitro methodologies to determine the extent of intestinal digestion of corn oil (CO), canola oil (CA), and beef tallow (BT) via manipulation of incubation length and concentrations of lipase, bile, and calcium within a buffer solution. Unless otherwise stated, 0.5 g of each lipid source were incubated separately and in triplicate, with triplicate batch culture runs for each treatment in 40 mL of 0.5 M KH2PO4 (pH = 7.6) for 24 h with pancreatin (8 g/L), bovine bile (2.5 g/L), and CaCl2 (10 mM). Individually, concentrations of pancreatin, bile, and CaCl2, as well as incubation length were tested. To examine the use of this assay to estimate in vitro total tract digestion, a KH2PO4 solution with concentrated amounts to reach the same final concentrations of pancreatin, bile, and Ca were used as the third step in a three-step total tract digestibility procedure. Free glycerol and free fatty acid (FFA) concentrations were measured using colorimetric assays as indicators of digestion. Data wereanalyzed as a completely randomized block design (block = run), using the Glimmix procedure of SAS. For each lipid source, free glycerol increased with increasing pancreatin; however, FFA was lowest at 0 g/L pancreatin but was similar at 6, 8, and 10 g/L. Both glycerol and FFA were greater for 2.5 and 5 g/L of bile than for 0 g/L for each lipid source. Calcium concentration did not affect glycerol or FFA for either CO or CA; however, glycerol and FFA for BT were greater when calcium was included at 5 and 10 mM than at 0 mM. For all fat sources, free glycerol and FFA increased after 1 h until 12 h, but did not increase from 12 to 24 h. When a concentrated mixture was used following fermentation and acidification steps, digestibility using FFA concentration increased as compared to just adding buffer; however, free glycerol concentration was indeterminable. Thus, free glycerol and FFA can be used as indicators of lipid digestion when a lipid source is incubated for at least 12 h in a buffer solution containing 8 g/L pancreatin, 2.5 g/L bile, and 5 mM Ca when only estimating in vitro intestinal digestion; however, when utilizing this assay in a three-step in vitro total tract digestibility procedure, only FFA can be used.

No. of bitstreams: 0 Previous issue date: 2021-04-01 Bacterial cultures, enzymes, and yeast-derived feed additives are often included in commercial dairy rations due to their effects on ruminal fermentation. However, the effects of these additives when fed together are not well understood. The objective of this study was to evaluate the changes in ruminal fermentation when a dairy ration is supplemented with combinations of bacterial probiotics, enzymes and yeast. Our hypotheses were that ruminal fermentation would be altered, indicated through changes in volatile fatty acid profile and nutrient digestibility, with the inclusion of (1) an additive, (2) yeast, and (3) increasing additive doses. Treatments were randomly assigned to 8 fermenters in a replicated 4 × 4 Latin square with four 10 d experimental periods, consisting of 7 d for diet adaptation and 3 d for sample collection. Basal diets contained 52:48 forage:concentrate and fermenters were fed 106 g of dry matter per day divided equally between two feeding times. Treatments were: control (CTRL, without additives); bacterial culture/enzyme blend (EB, 1.7 mg/d); bacterial culture/enzyme blend with a blend of live yeast and yeast culture (EBY, 49.76 mg/d); and a double dose of the EBY treatment (2×, 99.53 mg/d). The bacterial culture/enzyme blend contained five strains of probiotics (Lactobacillus animalis, Propionibacterium freudenreichii, Bacillus lichenformis, Bacillus subtilis, and Enterococcus faecium) and three enzymes (amylase, hemicellulase, and xylanase). On d 8-10, samples were collected for pH, redox, volatile fatty acids, lactate, ammonia N, and digestibility measurements. Statistical analysis was performed using the GLIMMIX procedure of SAS. Repeated measures were used for pH, redox, VFA, NH3-N, and lactate kinetics data. Orthogonal contrasts were used to test the effect of (1) additives, ADD (CTRL vs. EB, EBY, and 2X); (2) yeast, YEAST (EB vs. EBY, and 2X); and (3) dose, DOSE (EBY vs. 2X). No effects (P > 0.05) were observed for pH, redox, NH3-N, acetate, isobutyrate, valerate, total VFA, acetate:propionate, nutrient digestibility or N utilization. Within the 24 h pool, the molar proportion of butyrate increased (P = 0.03) with the inclusion of additives when compared to the control while the molar proportion of propionate tended to decrease (P = 0.07). In conclusion, the inclusion of bacterial cultures, enzymes and yeast in the diet increased butyrate concentration; but did not result in major changes in ruminal fermentation. The objective of this study was to evaluate the effect of inclusion of condensed tannins (CT) from black wattle (Acacia mearnsii) on feed intake, ruminal protozoa population, ruminal fermentation, and nutrient digestibility in Jersey steers. Five ruminally-cannulated steers were used in a 5x5 Latin square design, with five periods of 20 days each (14 days for diet adaptation and six days for sample collection per period). Treatments were composed of dietary inclusion levels of condensed tannins at 0, 5, 10, 15, and 20 g/kg of diet dry matter. Intakes of dry matter, organic matter, ether extract, crude protein, neutral detergent fiber, and total digestible nutrients were not affected by condensed tannins. The ruminal pH was reduced linearly with tannin levels. Ruminal ammonia nitrogen concentration was not affected by tannins. Tannins reduced the molar proportion of acetate and did not affect the ruminal protozoal population, which might be related to the low doses used. Digestibilities of dry matter, organic matter, and neutral detergent fiber were not altered, however, there was a linear reduction in crude protein digestibility. Based on these results, CT extracts from black wattle are not recommended for improving nutrient utilization in steers at the tested levels.

Acute and subacute ruminal acidosis (SARA) are common nutritional problems in both beef and dairy cattle. Therefore, the objective of this review is to describe how ruminal Gram-negative bacteria could contribute to the pathogenesis of ruminal acidoses, by releasing lipopolysaccharides (LPS; a component of their cell wall) in the ruminal fluid. When cattle consume excessive amounts of highly fermentable carbohydrates without prior adaptation, normal fermentation become disrupted. The fermentation of these carbohydrates quickly decreases ruminal pH due to the accumulation of short-chain fatty acids and lactate in the rumen. As a consequence, ruminal epithelium may be damaged and tissue function could be impaired, leading to a possible translocation of pathogenic substances from the rumen into the bloodstream. Such changes in fermentation are followed by an increase in Gram-positive bacteria while Gram-negative bacteria decrease. The lyses of Gram-negative bacteria during ruminal acidosis increase LPS concentration in the ruminal fluid. Because LPS is a highly proinflammatory endotoxin in the circulatory system, past studies have raised concerns regarding ruminal LPS contribution to the pathogenesis of ruminal acidosis. Although animals that undergo these disorders do not always have an immune response, recent studies showed that different Gram-negative bacteria have different LPS composition and toxicity, which may explain the differences in immune response. Given the diversity of Gram-negative bacteria in the rumen, evaluating the changes in the bacterial community during ruminal acidosis could be used as a way to identify which Gram-negative bacteria are associated with LPS release in the rumen. By identifying and targeting ruminal bacteria with possible pathogenic LPS, nutritional strategies could be created to overcome, or at least minimize, ruminal acidosis.

The objectives of this study were to evaluate the effects of lipopolysaccharide (LPS) dosing on bacterial fermentation and bacterial community composition (BCC), to set up a subacute ruminal acidosis (SARA) nutritional model in vitro, and to determine the best sampling time for LPS dosing in a dual-flow continuous culture system. Diets were randomly assigned to 6 fermentors in a replicated 3 × 3 Latin square with three 11-d experimental periods that consisted of 7 d for diet adaptation and 4 d for sample collection. Treatments were control diet (CON), wheat and barley diet (WBD) to induce SARA, and control diet + LPS (LPSD). Fermenters were fed 72 g of dry matter/d. The forage:concentrate ratio of CON was 65:35. The WBD diet was achieved by replacing 40% of dry matter of the CON diet with 50% ground wheat and 50% ground barley. The LPS concentration in LPSD was 200,000 endotoxin units, which was similar to that observed in cows with SARA. The SARA inducing and LPS dosing started at d 8. The BCC was determined by sequencing the V4 region of the 16S rRNA gene using the Illumina MiSeq platform (Illumina Inc., San Diego, CA). The LPSD and CON maintained pH above 6 for the entire experimental period, and the WBD kept pH between 5.2 and 5.6 for 4 h/d, successfully inducing SARA. Digestibility of neutral detergent fiber and crude protein in LPSD were not different from WBD but tended to be lower than CON. Lipopolysaccharide dosing had no effect on pool of VFA concentrations and profiles but decreased bacterial N; the pattern changes of VFA and LPS in LPSD started to increase and be similar to WBD 6 h after LPS dosing. Pool of LPS concentration was around 11-fold higher in WBD and 4-fold higher in LPSD than CON. In the solid fraction, the BCC of LPSD was different from WBD and tended to be different from CON. In the liquid fraction, the BCC was different among treatments. The LPS dosing increased the relative abundance of Succinimonas, Anaeroplasma, Succinivibrio, Succiniclasticum, and Ruminobacter, which are main gram-negative bacteria related to starch digestion. Our results suggest that LPS dosing does not affect pH alone. However, LPS could drive the development of SARA by affecting bacteria and bacterial fermentation. For future studies, samples are suggested to be taken 6 h after LPS dosing in a dual-flow continuous culture system.

The objective of this study was to evaluate the effects of five levels of condensed tannins (CT) from black wattle (Acacia mearnsii) in the diets of lactating dairy cows on intake, nutrient digestibility, ruminal microbial protein synthesis, milk production, composition, oxidative profile, and blood metabolites. Five Holstein cows (88 &plusmn, 26.8 days in milk) were allocated in a 5 x 5 Latin square design for a period of 20 days (14 days of diet adaptation and six for sampling). Treatments were the inclusion levels of CT at 0, 5, 10, 15 and 20 g/kg of dry matter (DM) in the diet. There was no effect of CT on DM intake. The digestibility of DM and neutral detergent fiber changed quadratically, with the maximum values at 12.2 and 11.4 g/kg of DM, respectively. There was no effect on ruminal microbial protein synthesis and milk production, however, milk casein concentration was reduced linearly. There was no effect on the milk oxidative profile. Inclusion of CT at levels up to 20 g/kg of DM did not affect intake or microbial protein synthesis, however, added CT depressed the production of energy corrected milk and milk casein concentration.

We evaluated Haemonchus contortus (HC) and Trichostrongylus colubriformis (TC) infection on the ruminal microbial community of Santa Ines lambs to better understand the pathophysiology of parasite infections and the interactions among gastrointestinal nematodes and gut resident microbiota. In this study, 18 six months of age lambs were maintained for 34 days in individual pens divided into three treatments that included animals infected with HC and TC, and control (infection-free). Haematological, ruminal parameter and microbial nitrogen absorbed by pune derivatives, as well as enteric methane emission (CH4), were analysed, and the rumen microbial taxonomic and functional profile assessed by shotgun metagenomics. The analysis showed that total protein, albumin, urea, and butyrate level were lower in animals infected by both parasites, while HC infection also decreased the haemoglobin level. Both infected groups (TC and HC) increased the enteric methane emission (CH4). TC and HC infections increased the diversity and richness of functional microbial genes. Most alterations in the rumen microbiome composition of infected groups are associated with the suppression of microbes involved in microbial homeostasis maintenance and expansion of the archaeal community in the infected animals. Infection led to an increased abundance of nitrogen, amino acid, protein, and energy metabolism genes. Overall, TC and HC infection increased the enteric methane emission, negatively affected taxon's responsible for maintenance de rumen homeostasis and modulate some important genes related to protein and energy metabolism.

The effects of ruminal protozoa (RP) concentration on methane emissions from ruminants were evaluated in a meta-analysis using 67 publications reporting data from 85 in vivo experiments. Experiments included in the database reported methane emissions (g/kg DMI) and RP (log10 cells/mL) from the same group of animals. Quantitative data including diet chemical composition, ruminal fermentation, total tract digestibility, and milk production; and qualitative information including methane mitigation strategies, animal type, and methane measurement methods were also collected. The studies were conducted in dairy cows (51%), beef steers (32%) and small ruminants (32%). 70% of the studies reported a reduction in methane emissions. Supplemental lipids reduced methane emissions 95% of the time. The relationship between methane emissions and RP concentration was evaluated as a random coefficient model with the experiment as a random effect and weighted by the inverse pooled SEM squared, including the possibility of covariance between the slope and the intercept. A quadratic effect of RP concentration on methane emissions was detected: CH4= -28.8 + 12.2 × RP-0.64 × RP2. To detect potential interfering factors in the relationship, the influence of several qualitative and quantitative factors were separately tested. Acetate, butyrate, and isobutyrate molar proportions had positive relationships with methane emissions and influenced the relationship between RP concentration and methane emissions, where the presence of ruminal fermentation variables reduced the effects of RP concentration in methane emissions. Total tract digestibility of DM, OM, and CP had negative relationships while NDF digestibility had a positive relationship with methane emissions; however, they only changed the magnitude of intercept and slope of RP and RP2 for the relationship. For dairy cows, milk fat and protein concentrations had positive relationships and milk yield had a negative relationship with methane emissions and changed the magnitude of intercept and slope of RP and RP2 for the relationship.

This study was carried out to evaluate the effects of tannin supplementation on ruminal microbiota of sixteen lambs infected and non-infected with Haemonchus contortus and Trichostrongylus colubriformis. Animals were fed with hay, concentrate and supplemented with Acacia mearnsii (A. mearnsii). The animals were divided into four treatments: two control groups without infection, either receiving A. mearnsii (C+) or not (C-), and two infected groups, one with A. mearnsii (I+) and another without A. mearnsii (I-). Ruminal short-chain fatty acids (SCFA) and metagenome sequencing of ruminal microbiota were used to evaluate the effect of tannin and infection on ruminal microbiome. The objective was to identify the non-linear model with the best fit for cumulative gas production from fermentation of fresh alfalfa, with or without tannin extract, incubated with rumen fluid from five different species of ruminants. Fifteen animals (Taurine and Zebuine cattle, water buffaloes, sheep and goats) were used as inoculum donors. During incubation, 500 mg of fresh alfalfa, with or without 150 mg of acacia tannin extract, were used as substrate in the semi-automated gas production technique. Experimental design was completely randomized in a factorial arrangement with five inoculum sources (ruminant specie) and two treatments (with or without tannin extract). We used the PROC NLMIXED to fit ten mathematical models and the best one was chosen based on the lowest AIC and MSE and highest R2. Lastly, the best model was validated using the cross validation technique. The model with the best fit was the Groot model (AIC 1255.5; MSE 174.01; R2 0.9496) comparatively to others methods and the most part of error is from random effect (97.7%). Tannin inclusion reduced parameters potential gas production (A) and time to produce half of total gas production (T1) (P > 0.0001); however, no difference was observed on the gas production rate (k) (P > 0.1181). When no tannin was added, differences between the two cattle category were observed. Comparing water buffaloes’ inoculum with Taurine inoculum, no differences were observed for “A,” however, this parameter differed among water buffaloes and Zebuine cattle. In conclusion, Groot model had the best fit on in vitro bioassay with alfalfa substrate and treated or not with tannin extract. The tannin extract reduced the potential gas production; however, it did not change the gas production rate. For evaluation of alfalfa by cumulative gas production technique, the potential gas production was changed by using different animal categories as inoculum donor.

Several studies have evaluated the effects of complete or partial ruminal protozoa (RP) inhibition; however, to this date, no practical suppressant has been identified and used in large scale. This meta-analysis quantitatively evaluates the effectiveness of multiple strategies on inhibiting RP numbers and their influence on ruminal fermentation and animal performance. This study compared 66 peer-reviewed articles (16 manuscripts for complete and 50 manuscripts for partial RP inhibition that used supplemental phytochemicals and lipids, published from 2000 to 2018, to inhibit RP in vivo. Data were structured to allow a meta-analytical evaluation of differences in response to different treatments (complete RP inhibition, phytochemicals, and lipids). Data were analyzed using mixed models with the random effect of experiment and weighted by the inverse of pooled standard error of the mean (SEM) squared. Supplemental phytochemicals and LCFA had no effects on inhibiting RP numbers; however, supplemental MCFA had a potent antiprotozoal effect. Both complete and partial RP (supplemental phytochemicals and lipids) inhibition decreased methane production, total tract digestibility of OM and NDF, and ruminal NH3-N concentration and increased propionate molar proportion. Methane production, molar proportions of acetate and propionate, total tract NDF digestibility were affected by the interaction of treatment (supplemental phytochemicals and lipids) and RP numbers. Supplemental phytochemicals and lipids can be effective in reducing methane production when RP numbers is below 7 Log10 cells/mL, especially by supplemental saponins, tannins, and MCFA. In terms of animal performance, supplemental tannins could be recommended to control methane emissions without affecting animal performance. However, their negative effects on total tract digestibility could be a drawback when feeding tannins to ruminants. The negative effects of supplemental lipids on milk fat composition should be considered when feeding lipids to ruminants. In conclusion, ruminal protozoa play important roles on methanogenesis, fiber digestion, and ruminal NH3-N concentration, regardless of experimental diets and conditions; supplemental phytochemicals and lipids can be effective on reducing methane production when RP numbers is below 7 Log10 cells/mL. Among these partial RP inhibition strategies, supplemental tannins could be recommended to control methane production.

The objective of this study was to determine intake, nutrient availability, and animal selection of major forage species in sheep supplemented with zinc sulfate or propylene glycol in Caatinga-native pastures during the rainy season. Twenty-four mixed Santa In&ecirc, s sheep, all non-castrated males, with initial weight of 19.3 &plusmn, 2.52 kg and 4 &plusmn, 0.35 months of age, were distributed in a complete randomized design into three treatments: Control (CT)&mdash, concentrate supplemented at 0.7% of body weight, CT + 300 mg of Zn day&minus, 1, CT + 2.5 mL of propylene glycol/kg LW0.75&middot, day&minus, 1. Measurements were done in four periods during the rainy season, with 28 days of interval between each measurement. Differences were observed in the composition of the ruminal extrusa samples from pastures for crude protein (CP) (192 to 131 g kg&minus, 1), in vitro dry matter digestibility (IVDMD) (537 to 441 g kg&minus, 1), and in vitro organic matter digestibility (IVOMD) (468 to 359 g kg&minus, 1) in March and June, respectively. There was no effect for treatments, neither for the treatment x period interaction on organic matter (OM), CP, and fibrous fraction intake (p &gt, 0.05). Organic matter intake (OMI) was, on average 23.9% greater in March compared to June. CP intake decreased monthly (p &lt, 0.05). Fibrous fraction intake was greater in March (p &lt, 0.05), with reductions of 34.8, 33.3, and 39.4% in June, respectively, for neutral detergent fiber (NDF), acid detergent fiber (ADF), and cellulose (CEL) fractions. There was no effect of treatments (p &gt, 0.05) on selection of vegetal species present in the pasture. On the other hand, the proportion between ingested species changed over the experimental period. Greater intakes were found in April compared to May, except for Zizyphus joazeiro intake, which was greater in March (p &lt, 0.05). In conclusion, based on the finding of this study, Zn and propylene glycol (PG) supplementation did not improve sheep nutrient intake when grazing Caatinga-native pasture in the rainy season. Caatinga-native pasture biomass has adequate protein and digestible organic matter levels during early rainy season. Over this period, however, the advanced maturity of the plants and the reduced availability of pasture may result in variations of intake by the animals. In the months of April to June, a reduced energy supply is caused by reduced nutritive values of pastures, which contributes to inefficient protein utilization and reduced feed intake.

Flaxseed (FS) and chia seed (CS) are oilseeds rich in omega-3 fatty acids, which may change meat and milk composition when added to ruminants' diets and may have health benefits for humans. Literature on the effects of CS supplementation on ruminal metabolism is nonexistent. A dual-flow continuous culture fermenter system consisting of 6 fermenters was used to assess the effect of FS and CS supplementation in an alfalfa hay-based diet on ruminal fermentation, nutrient digestibility, microbial protein synthesis, and long-chain fatty acid flow. Diets were randomly assigned to fermenters in a replicated 3 × 3 Latin square design, with 3 consecutive periods of 10 d each, consisting of 7 d for diet adaptation and 3 d for sample collection. Each fermenter was fed a total of 72 g of DM/d divided in 6 equal portions. Treatments were 1) alfalfa hay + calcium soaps of palm oil fatty acid (MEG; 69.3 g DM/d of alfalfa hay plus 2.7 g DM/d of calcium soaps of palm oil fatty acid), 2) alfalfa hay + FS (FLAX; 68.4 g DM/d of alfalfa hay plus 3.6 g DM/d of ground FS), and 3) alfalfa hay + CS (CHIA; 68.04 g DM/d of alfalfa hay plus 3.96 g DM/d of ground CS). Dietary treatments had similar amounts of total fat, and fat supplements were ground to 2-mm diameter. Effluents from the last 3 d of incubation were composited for analyses. Data were analyzed using the MIXED procedure of SAS. Ruminal apparent and true nutrient digestibility of all nutrients did not differ ( > 0.05) among treatments. Compared with MEG, FLAX and CHIA increased the flows of C18:3 -3, C20:4 -6, and total PUFA ( 0.05) by treatments. Lastly, there were no differences ( > 0.05) among diets for total VFA concentration and molar proportions of individual VFA. Results from this study indicate that FS and CS supplementation did not impair ruminal fermentation, digestibility, microbial efficiency, and ruminal N metabolism. Overall, CS appears to be as effective as FS as a fat source when added to ruminants' diets using a dual-flow continuous culture system.

The objective of this study was to evaluate different doses of phenolic compounds extracted from honey bees propolis on ruminal protozoa population, microbial protein synthesis, ruminal fermentation, solids’ passage rate, and total tract digestibility in water buffaloes. Four crossbred non-lactating female water buffaloes averaging 543.9732.1 kg of body weight fitted with rumen cannulas were used in a 4 � 4 latin square design. The phenolic compounds were dosed in the rumen via rumen cannula in four doses: 0, 16.95, 33.9, and 50.85 mg/d; respectively. The total mixed ration consisted of 800 g/kg corn silage and 200 g/kg concentrate. Phenolic compounds linearly reduced the Entodinium protozoa population (Po0.01) and quadratically increased ruminal acetate concentration (Po0.05) and there was a trend to increase microbial protein synthesis in the rumen (P¼0.07). Phenolic compounds did not change total digestibility of dry matter and nutrients, solids' passage rate, and the efficiency of microbial protein synthesis (P40.05). It can be concluded that phenolic compounds reduce the Entodinium protozoa population in water buffaloes and change ruminal fermentation pattern, favoring acetate fermentation.

Simple Summary Lipopolysaccharide (LPS) is a component of the outer membranes of Gram-negative bacterial cell wall made of three covalently linked regions: the O-antigen, the core oligosaccharide, and the endotoxin lipid A moiety, which carries the endotoxic activity of LPS. Among Gram-negative bacteria there is significant structural diversity in the lipid A region. Specifically, the number of lipid A acyl chains directly correlates with the ability to induce cytokine production whereas the hexa-acylated forms usually are the most immunostimulant ones, contrary to penta- or tetra- acylated forms that result in weak inflammatory host responses. Ruminal bacteria are predominantly Gram-negative, and their respective LPS presence has been suggested to be associated with ruminal acidosis, a metabolic disorder of cattle with negative effects on health and production. In the rumen, the most predominant phylum is Bacteroidetes which exhibit weak host immunological response compared to widely used Escherichia coli LPS. This review aims to present accumulated knowledge regarding ruminal LPS, pointing out the differences in ruminal LPS compared to widely known LPS, and introduce hypotheses that could contribute to further understanding and planning strategies to tackle ruminal acidosis. Abstract The objective of this review is to present the need for the development of a comprehensive ruminal lipopolysaccharide (LPS) extraction, purification and analysis protocol and state hypotheses that could contribute to planning novel strategies against ruminal acidosis. Lipopolysaccharide is an immunostimulatory molecule of Gram-negative bacterial outer membranes and has been reported to contribute to ruminal acidosis in cattle. Bacterial death and lysis are normal processes, and thus LPS is normally present in ruminal fluid. However, ruminal LPS concentration is much greater during subacute ruminal acidosis (SARA). Contrary to the widely known LPSs, ruminal LPS seems to be composed of a variety of LPS chemotypes that may interact with each other resulting in an LPS “mixture”. Hypotheses regarding the influence of each specific ruminal bacterial specie to innate immunity during SARA, and the representativeness of the exclusive use of the Escherichia coli LPS to rumen epithelial tissue challenges, could expand our knowledge regarding SARA. In addition, possible correlation between the monomeric Toll-like Receptor 4 (TRL4) and the antagonistic penta-acylated lipid A of LPS could contribute to novel strategies to tackle this nutrition disorder.

The objective of this study was to evaluate the effects of Lactobacillus inoculants on fermentation, losses, and aerobic stability of a total mixed ration (TMR) silage. A TMR, formulated to meet the requirements of dairy cows producing 25 kg of milk/d, was applied with the following treatments prior to ensiling: 1) Control (CON), 2) Lactobacillus buchneri (10(5) cfu/g of fresh forage; LB), and 3) Lactobacillus plantarum (10(5) cfu/g of fresh forage; LP). TMR silages were ensiled for 15 and 60 d in silos equipped with an apparatus for determination of gravimetric DM, gas, and effluent losses. The experiment was performed in a complete randomized design with a 3 × 2 factorial arrangement of the treatments, with 5 replicates per treatment. Chemical changes, microbial counts, fermentation profile, and aerobic stability were measured after opening the silos. Data were submitted to ANOVA, and means were compared by Tukey and T-test and statistical significance was declared at P ≤ 0.05. After 15 d of ensiling, the inclusion of inoculant decreased NDF (P < 0.05) and butyric acid concentrations (P < 0.05) in TMR. LP had the lowest aerobic stability (P < 0.05) and the greatest loss of DM (P < 0.03). Ensiling for 60 d increased ammonia nitrogen (NH(3)-N), lactic acid bacteria (LAB), aerobic stability, and concentrations of lactic and acetic acid (P < 0.01) and lowered (P < 0.02) total fermentation losses compared to 15 d across all treatments. After 60 d of ensiling, LP lowered pH to the greatest extent. Treatment had no effect on concentrations of DM, CP, ADF, ash, and EE, as well as in vitro DM digestibility. In conclusion, inoculants containing LP or LB did not improve fermentation profile, did not prolong the aerobic stability, nor reduced losses. Furthermore, the 15-d ensiling period was insufficient for adequate bacterial activity.

Histone deacetylase (HDAC) inhibition attenuates inflammation in rodents and short-chain fatty acids (SCFAs) are effective HDAC inhibitors. Therefore, the objective of this study was to evaluate the role of the SCFAs sodium propionate (SP) and sodium butyrate (SB) as HDAC-dependent regulators of inflammatory gene expression in bovine mammary epithelial cells (MAC-Ts). We postulated that SP and SB would decrease inflammation in MAC-Ts by inhibiting HDAC activity and increasing histone H3 acetylation and consequently decreasing inflammatory gene expression. For this study, MAC-Ts stimulated with lipopolysaccharide (LPS) were used as a model for bovine mammary epithelial cell inflammation. MAC-Ts were cultured in a basal medium. Cell lysates were incubated with SP or SB (0 to 5 mM) for 2 h prior to HDAC substrates incubation for an additional 2 h and HDACs activity was determined. Next, cells were pretreated with SP or SB (0 to 3.0 mM) for 2 h prior to LPS (1 µg/mL) stimulation for an additional 2 h and assessed for histone H3 acetylation. Then, cells were pretreated with SP or SB (1 mM) for 24 h prior to LPS (1 µg/mL) stimulation for an additional 2 h and RNA was isolated for inflammatory gene expression evaluation by PCR array and gene validation was performed using quantitative real-time PCR. One-way ANOVA followed by Tukey post hoc analysis was conducted and statistical significance set at P 0.05) overall inflammatory gene expression. Under our experimental conditions, findings suggest that in MAC-Ts, SCFAs regulate epigenetic markers on nucleosomal DNA in addition to regulation of inflammatory gene events independent of HDAC activity. Nevertheless, examination of SCFAs and/or HDACs inhibitors in bovine mammary gland is worth being further investigated to delineate the potential impact of HDAC inhibition and histones hyperacetylation on mammary gland tissue inflammation.

Bovine mammary epithelial cells (MAC-Ts) are a common cell line for the study of mammary epithelial inflammation; these cells are used to mechanistically elucidate molecular underpinnings that contribute to bovine mastitis. Bovine mastitis is the most prevalent form of disease in dairy cattle that culminates in annual losses of two billion dollars for the US dairy industry. Thus, there is an urgent need for improved therapeutic strategies. Histone deacetylase (HDAC) inhibitors are efficacious in rodent models of inflammation, yet their role in bovine mammary cells remain unclear. HDACs have traditionally been studied in the regulation of nucleosomal DNA, in which deacetylation of histones impact chromatin accessibility and gene expression. Using MAC-T cells stimulated with tumor necrosis factor α (TNF-α) as a model for mammary cell inflammation, we report that inhibition of HDACs1 and 2 (HDAC1/2) attenuated TNF-α-mediated inflammatory gene expression. Of note, we report that HDAC1/2-mediated inflammatory gene expression was partly regulated by c-Jun N-terminal kinase (JNK) and extracellular signal-regulated kinase (ERK) phosphorylation. Here, we report that HDAC1/2 inhibition attenuated JNK and ERK activation and thus inflammatory gene expression. These data suggest that HDACs1 and 2 regulate inflammatory gene expression via canonical (i.e., gene expression) and noncanonical (e.g., signaling dependent) mechanisms. Whereas, further studies using primary cell lines and animal models are needed. Our combined data suggest that HDAC1/2-specific inhibitors may prove efficacious for the treatment of bovine mastitis.

Camelina is an oil seed crop that belongs to the Brassica family (Cruciferae). Camelina meal is a by-product from the biofuel industry that contains on average 38% crude protein and between 10 to 20% of residual fat, which limits the inclusion levels of camelina meal in dairy cow diets as the main protein supplement. Thus, we conducted a solvent extraction on ground camelina seed on a laboratory scale. The objectives of this study were (1) to assess the effects of replacing canola meal (CM) with solvent-extracted camelina meal (SCAM) in lactating dairy cow diets; and (2) to determine the effects of SCAM on microbial fermentation and AA flow in a dual-flow continuous culture system. Diets were randomly assigned to 6 fermentors in a replicated 3 × 3 Latin square with three 10-d experimental periods consisting of 7 d for diet adaptation and 3 d for sample collection. Treatments were 0, 50, and 100% SCAM inclusion, replacing CM as the protein supplement. Diets contained 55:45 forage:concentrate, and fermentors were fed 72 g of dry matter/d equally divided in 2 feeding times. On d 8, 9, and 10 of each period, samples were collected for analyses of pH, volatile fatty acids (VFA), N metabolism, NH3-N, digestibility, and AA flow. Statistical analysis was performed using the MIXED procedure of SAS (SAS Institute Inc., Cary, NC), and linear and quadratic effects of SCAM inclusion were assessed. Total VFA concentration and pH were not affected by diets. Molar proportion of acetate decreased, whereas molar proportion of propionate increased with SCAM inclusion. Total branched-chain VFA concentration was the least in fermentors fed diet 0, and greatest in fermentors fed diet 50. Digestibility of NDF decreased in fermentors fed SCAM diets, and dry matter, organic matter, and crude protein true digestibility were similar across diets. Concentration of NH3-N linearly decreased, and non-NH3-N linearly increased with SCAM inclusion. Bacterial efficiency (calculated as g of bacterial N flow/kg of organic matter truly digested) tended to be greater in fermentors fed diet 100. Outflow of Arg linearly increased with SCAM inclusion, whereas overall AA flow was not affected by diet. In conclusion, replacing CM with SCAM increased propionate molar proportion and non-NH3-N flow, and decreased NH3-N flow and concentration, which may improve animal energy status and N utilization. Inclusion of SCAM did not change most AA flow, indicating that it can be a potential replacement for CM.

An experiment was conducted to evaluate whether a reduction in dietary crude protein (CP) and genotype affects animal performance, feed efficiency and carcass traits of beef cattle at different stages of the feedlot. Twenty-two Nellore and 22 crossbred F1 Angus × Nellore (Nellore: 8 months, initial body weight (iBW) = 212.7 kg; Crossbred: 8 months, iBW = 226.1 kg) were used in this experiment. The experiment was conducted in a complete randomized design with six replicates, in a 2 × 3 factorial design. The factors were two genotypes (Nellore and Crossbred) and three CP concentrations (100, 120 and 140 g/kg dry matter [DM]). The experimental period lasted 224 day, being divided into two stages (Backgrounding = 112 day, and Finishing = 112 day). At the end of the experiment, all animals were slaughtered to evaluate their carcass characteristics. Similar patterns were observed during backgrounding and finishing phases on intake and average daily gain (ADG) regarding genotype. Intakes of DM, organic matter, CP, neutral detergent fibre, non-fibre carbohydrates and total digestible nutrients, ADG (kg/day) and feed efficiency were greater in Crossbred animals than Nellore in both phases. During the backgrounding phase, dietary CP did not influence DM intake. However, a linear effect of CP on ADG was observed in the backgrounding phase. During finishing, Nellore cattle fed 100 g CP/kg DM presented lower ADG than Nellore fed 120 and 140 g CP/kg DM. Crossbred animals were heavier than Nellore at the beginning and end of the trial. Crossbred animals also gained more carcass, retained more CP and fat, and had a greater subcutaneous fat thickness during the experiment compared with Nellore. There was a quadratic effect of dietary CP on retained CP and dressing. The current study found that crossbred animals (F1 Angus × Nellore) not only had greater performance, but also better carcass traits compared with Nellore, representing an option for increasing productivity. Also, it is possible to adjust diets according to phase. During the backgrounding stage, 140 g CP/kg DM should be used, being reduced to 120 g CP/kg DM during the finishing stage of feedlot for Nellore and to 100 g CP/kg DM for Crossbred, without affecting performance adversely.

This experiment aimed to determine the effects of camelina seed (CS) supplementation at different dietary fat levels on ruminal bacterial community composition and how it relates to changes in ruminal fermentation in a dual-flow continuous culture system. Diets were randomly assigned to 8 fermenters (1,200–1,250 mL) in a 2 × 2 factorial arrangement of treatments in a replicated 4 × 4 Latin square with four 10-day experimental periods that consisted of 7 days for diet adaptation and 3 days for sample collection. Treatments were: (1) no CS at 5% ether extract (EE, NCS5); (2) no CS at 8% EE (NCS8); (3) 7.7% CS at 5% EE (CS5); and (4) 17.7% CS at 8% EE (CS8). Megalac was used as a control to adjust EE levels. Diets contained 55% orchardgrass hay and 45% concentrate, and fermenters were equally fed a total of 72 g/day (DM basis) twice daily. The bacterial community was determined by sequencing the V4 region of the 16S rRNA gene using the Illumina MiSeq platform. Sequencing data were analyzed using mothur and statistical analyses were performed in R and SAS. The most abundant phyla across treatments were the Bacteroidetes and Firmicutes, accounting for 49 and 39% of the total sequences, respectively. The bacterial community composition in both liquid and solid fractions of the effluent digesta changed with CS supplementation but not by dietary EE. Including CS in the diets decreased the relative abundances of Ruminococcus spp., Fibrobacter spp., and Butyrivibrio spp. The most abundant genus across treatments, Prevotella, was reduced by high dietary EE levels, while Megasphaera and Succinivibrio were increased by CS supplementation in the liquid fraction. Correlatively, the concentration of acetate was decreased while propionate increased; C18:0 was decreased and polyunsaturated fatty acids, especially C18:2 n-6 and C18:3 n-3, were increased by CS supplementation. Based on the correlation analysis between genera and fermentation end products, this study revealed that CS supplementation could be energetically beneficial to dairy cows by increasing propionate-producing bacteria and suppressing ruminal bacteria associated with biohydrogenation. However, attention should be given to avoid the effects of CS supplementation on suppressing cellulolytic bacteria.

The objectives of this study were to evaluate the feeding of coconut oil (CO), in which lauric acid (La) comprises about 50% of the fatty acid composition, as a practical rumen protozoa (RP) suppressing agent, to assess whether the source of La affects ruminal fermentation and animal performance and to test whether suppressing RP improves N utilization, nutrient digestion, nutrient flow at the omasal canal, and milk production. Fifteen multiparous Holstein cows (3 fitted with ruminal cannulas) and 15 primiparous Holstein cows (3 fitted with ruminal cannulas) were used in a replicated 3 × 3 Latin square experiment with 14 d of adaptation and 14 d of sample collection. Diets were fed as total mixed ration and contained (dry matter basis) 10% corn silage, 50% alfalfa silage, and 40% concentrate. The control diet contained 3% (dry matter basis) calcium soaps of palm oil fatty acids (Megalac, Church & Dwight Co. Inc., Princeton, NJ) as a ruminally inert fat source and had no added La or CO. Diets with La and CO were formulated to contain equal amounts of La (1.3%, dry matter basis). Dry matter intake was not affected by treatment. Both CO and La reduced RP numbers by about 40%. Lauric acid reduced yield of milk and milk components; however, CO did not affect yield of milk and yields of milk components. Both La and CO caused small reductions in total VFA concentration; CO increased molar proportion of ruminal propionate, reduced ruminal ammonia and branched-chain volatile fatty acids, suggesting reduced protein degradation, and reduced milk urea N and blood urea N concentrations, suggesting improved protein efficiency. Lauric acid reduced total-tract apparent digestibility of neutral detergent fiber and acid detergent fiber as well as ruminal apparent digestibility of neutral detergent fiber and acid detergent fiber as measured at the omasal canal; however, CO did not alter fiber digestion. Microbial protein flow at the omasal canal, as well as the flow of N fractions at the omasal canal, did not differ among treatments. Results from this experiment have confirmed that dietary La is not a practical agent for suppressing RP population in dairy cows, mainly because of its negative effects on fiber digestion and ruminal fermentation. Intake of CO appeared to reduce ruminal and improve protein efficiency, but did not improve milk production, milk composition, or increase microbial outflow from the rumen. Based on the results of this study, a 40% reduction of RP population is not sufficient to improve N utilization in dairy cows.

This experiment aimed to estimate the energy and protein requirements for Holstein × Gyr calves up to 60 days of age fed with milk replacer and amino acid supplement. Fifty male calves were used, of which seven were randomly allocated into the reference group and slaughtered at 8 days of age, seven were randomly allocated into the maintenance group slaughtered at 30 days of age, and the 36 remaining calves were included in the experiment at 8 days of age and randomly supplied with four dietary methionine+cysteine:lysine ratios (RMCL; 44%, 48%, 52%, and 56%) through amino acid supplement added as 1 kg/day of milk replacer reconstituted at 13.8% of dry matter. Different RMCL were tested for the models, and there were no significant effects on energy and protein requirements. The net energy requirement for maintenance was 75.2 kcal/empty bodyweight (EBW)0.75.day, with an energy use efficiency for maintenance of 67.38%. The prediction equation of net energy requirement for gain (Mcal/day) was energy retained = 0.0879 × EBW0.75 × empty bodyweight gain (EBWG)0.7580, with an energy use efficiency for gain of 47.57%. The estimated requirements for metabolisable protein for maintenance were 4.83 g/EBW0.75.day. The estimated equation for net protein requirements for gain (g/day) was NPg = EBWG × 246.73 × EBW–0.1204, with a protein use efficiency for gain of 71.55%. The estimated requirements for energy and proteins were greater than the values reported for calves fed with milk. Milk replacers are less efficiently used by calves up to 60 days of age when compared with whole milk.

The objective of this study was to evaluate the effects of increasing dietary crude protein (CP) levels and also comparing the effects of static versus oscillating dietary CP on ruminal nutrient digestibility, ruminal fermentation, nitrogen (N) metabolism, and microbial efficiency in beef cattle diets using a dual-flow continuous culture system. Eight fermenters (1,223 ± 21 mL) were used in a replicated 4 x 4 Latin square design with periods lasting 12 d each (8 d for adaptation and 4 d for sampling). Dietary treatments were: 1) 10% CP, 2) 12% CP, 3) 14% CP, and 4) 10 and 14% CP diets oscillating at 48-h intervals. Experimental diets consisted of 50% orchard hay and 50% concentrate. Fermenters were fed 72 g/d and solid and liquid dilution rates were adjusted to 5.5 and 11%/h, respectively. Data were analyzed using the MIXED procedure in SAS with α = 0.05. Apparent and true ruminal digestibilities of dry matter and organic matter were not affected (P > 0.05) by increasing dietary CP, nor by oscillating dietary CP. Total volatile fatty acids concentration and molar proportions of acetate, propionate, butyrate, valerate, iso-butyrate and iso-valerate were not affected (P > 0.05) by increasing or oscillating dietary CP. Ruminal NH3-N concentration increased linearly (P < 0.01) in response to increasing dietary CP. Total N, non-ammonia N, and rumen undegraded protein flows did not differ among treatments or between oscillating dietary CP and static 12% CP. Microbial N and NH3-N flows and microbial efficiency did not differ when comparing oscillating versus static CP (P > 0.05). However, there was a quadratic effect (P < 0.05) for these variables when dietary CP was increased. These results indicate that either ruminal microorganisms do not respond to oscillating CP levels or are capable of coping with 48-h periods of undernourishment.
