Although we develop influenza immunity from an early age, it is insufficient to prevent future infection with antigenically novel strains. One proposed way to generate long-term protective immunity against a broad range of influenza virus strains is to boost responses to the conserved epitopes on the hemagglutinin, the major surface glycoprotein on the influenza virus. Influenza-specific humoral immunity comprises a large fraction of the overall immune memory in humans, and it has been long recognized that preexisting immunity to influenza shapes the response to subsequent influenza infections and vaccinations. However, the mechanisms by which preexisting immunity modulates the response to influenza vaccination are still not completely understood. Using a set of mathematical models, we explore several hypotheses that may contribute to diminished boosting of antibodies to conserved epitopes after repeated vaccinations.

Background: Experimental and epidemiological studies indicate an association between exposure to particulate matter (PM) air pollution and increased risk of type 2 diabetes. In view of the high and increasing prevalence of diabetes, we aimed to quantify the burden of type 2 diabetes attributable to PM2·5 originating from ambient and household air pollution.Methods: We systematically compiled all relevant cohort and case-control studies assessing the effect of exposure to household and ambient fine particulate matter (PM2·5) air pollution on type 2 diabetes incidence and mortality. We derived an exposure–response curve from the extracted relative risk estimates using the MR-BRT (meta-regression—Bayesian, regularised, trimmed) tool. The estimated curve was linked to ambient and household PM2·5 exposures from the Global Burden of Diseases, Injuries, and Risk Factors Study 2019, and estimates of the attributable burden (population attributable fractions and rates per 100 000 population of deaths and disability-adjusted life-years) for 204 countries from 1990 to 2019 were calculated. We also assessed the role of changes in exposure, population size, age, and type 2 diabetes incidence in the observed trend in PM2·5-attributable type 2 diabetes burden. All estimates are presented with 95% uncertainty intervals.Findings: In 2019, approximately a fifth of the global burden of type 2 diabetes was attributable to PM2·5 exposure, with an estimated 3·78 (95% uncertainty interval 2·68–4·83) deaths per 100 000 population and 167 (117–223) disability-adjusted life-years (DALYs) per 100 000 population. Approximately 13·4% (9·49–17·5) of deaths and 13·6% (9·73–17·9) of DALYs due to type 2 diabetes were contributed by ambient PM2·5, and 6·50% (4·22–9·53) of deaths and 5·92% (3·81–8·64) of DALYs by household air pollution. High burdens, in terms of numbers as well as rates, were estimated in Asia, sub-Saharan Africa, and South America. Since 1990, the attributable burden has increased by 50%, driven largely by population growth and ageing. Globally, the impact of reductions in household air pollution was largely offset by increased ambient PM2·5.Interpretation: Air pollution is a major risk factor for diabetes. We estimated that about a fifth of the global burden of type 2 diabetes is attributable PM2·5 pollution. Air pollution mitigation therefore might have an essential role in reducing the global disease burden resulting from type 2 diabetes.Funding: Bill & Melinda Gates Foundation.

Background Indoor allergens are important components of airborne allergens, and studies have linked allergic respiratory diseases (ARDs) and exposure to aeroallergens. Objectives This study sought to identify fungi in house dust from four regions in Baghdad city, and to assess the risks of ARD and asthma associated with indoor fungi. Methods House dust was collected from 84 homes located in four major areas in Baghdad; locations 1, 2, 3 and 4 (north, west, south-west and south of Baghdad, respectively). The dust samples were cultured to identify fungal genera. Results Seventy-nine mold isolates belong to nine genera (Aspergillus, Rhizopus, Penicillium, Chrysonilia, Mucor, Fusarium, Cladosporium, Alternaria and Helminthosporium), as well as five yeast isolates, were identified. Aspergillus was the most common type of mold (56.0%), while Alternaria and Helminthosporium were the least frequent (each with 1%). The distribution of these fungi showed variation according to location, but the difference was not significant. ARD in general and asthma in particular were detected in 50.0 and 14.3% of households, respectively. Both types of allergy were significantly associated with mold genera and with the number of households and rooms, as well as the availability of gardens and trees. Conclusions The study indicated that fungi are significant indoor determinants of ARD and asthma in the city of Baghdad.

Embryonic cells grow in environments that provide a plethora of physical cues, including mechanical forces that shape the development of the entire embryo. Despite their prevalence, the role of these forces in embryonic development and their integration with chemical signals have been mostly neglected, and scrutiny in modern molecular embryology tilted, instead, towards the dissection of molecular pathways involved in cell fate determination and patterning. It is now possible to investigate how mechanical signals induce downstream genetic regulatory networks to regulate key developmental processes in the embryo. Here, we review the insights into mechanical control of early vertebrate development, including the role of forces in tissue patterning and embryonic axis formation. We also highlight recent in vitro approaches using individual embryonic stem cells and self-organizing multicellular models of human embryos, which have been instrumental in expanding our understanding of how mechanics tune cell fate and cellular rearrangements during human embryonic development. Cells in the embryo are subject to autonomous and external mechanical forces that help steer embryonic tissue patterning. Technical developments, such as in vitro models of early embryos, allow probing of the roles of mechanical forces in animal and human embryonic development.

This is the first study of thraustochytrids from the Red Sea coast in Saudi Arabia. One hundred and thirty-four isolates of thraustochytids were cultured from Al-Leith, Jeddah and Yanbu mangroves from this area, and were categorized into 38 morphotypes. Among the isolated thraustochytrids, Aurantiochytrium was the most speciose genus, represented by 36 morphological types. Seventeen strains formed a distinct clade within the genus Aurantiochytrium based on phylogenetic analyses of 18S rDNA sequences. The Aurantiochytrium clade from the Middle East is characterized by the production of high levels of oleic and linoleic acids and may represent undescribed taxa. Four Aurantiochytrium strains were grown on large scale to study their ability to produce docosahexaenoic acid (DHA). These strains produced biomass ranging from 37.7 to 66 g L−1 and the percentages of oil ranged from 40 to 57.2% of the dry weight. Twenty-one fatty acids were recorded from the four strains which included: eight saturated four monounsaturated and nine polyunsaturated. Dominant fatty acids included C16:0 palmitic acid (24.14–37.02% total fatty acid (TFA)), C18:1ω9 oleic acid (3.01–25.07% TFA), C18:2ω6 linoleic acid (1.85–20.85% TFA) and C22:6ω3 DHA (4.87–16.5% TFA). DHA was the predominant polyunsaturated fatty acid in three strains, while linoleic acid was the predominant PUFAs in one strain.

BackgroundPatients with chronic inflammatory disease (CID) treated with immunosuppressive medications have increased risk for severe COVID-19. Although mRNA-based SARS-CoV-2 vaccination provides protection in immunocompetent persons, immunogenicity in immunosuppressed patients with CID is unclear.ObjectiveTo determine the immunogenicity of mRNA-based SARS-CoV-2 vaccines in patients with CID.DesignProspective observational cohort study.SettingTwo U.S. CID referral centers.ParticipantsVolunteer sample of adults with confirmed CID eligible for early COVID-19 vaccination, including hospital employees of any age and patients older than 65 years. Immunocompetent participants were recruited separately from hospital employees. All participants received 2 doses of mRNA vaccine against SARS-CoV-2 between 10 December 2020 and 20 March 2021. Participants were assessed within 2 weeks before vaccination and 20 days after final vaccination.MeasurementsAnti-SARS-CoV-2 spike (S) IgG+ binding in all participants, and neutralizing antibody titers and circulating S-specific plasmablasts in a subset to assess humoral response after vaccination.ResultsMost of the 133 participants with CID (88.7%) and all 53 immunocompetent participants developed antibodies in response to mRNA-based SARS-CoV-2 vaccination, although some with CID developed numerically lower titers of anti-S IgG. Anti-S IgG antibody titers after vaccination were lower in participants with CID receiving glucocorticoids (n=17) than in those not receiving them; the geometric mean of anti-S IgG antibodies was 357 (95% CI, 96 to 1324) for participants receiving prednisone versus 2190 (CI, 1598 to 3002) for those not receiving it. Anti-S IgG antibody titers were also lower in those receiving B-cell depletion therapy (BCDT) (n=10). Measures of immunogenicity differed numerically between those who were and those who were not receiving antimetabolites (n=48), tumor necrosis factor inhibitors (n=39), and Janus kinase inhibitors (n=11); however, 95% CIs were wide and overlapped. Neutralization titers seemed generally consistent with anti-S IgG results. Results were not adjusted for differences in baseline clinical factors, including other immunosuppressant therapies.LimitationsSmall sample that lacked demographic diversity, and residual confounding.ConclusionCompared with nonusers, patients with CID treated with glucocorticoids and BCDT seem to have lower SARS-CoV-2 vaccine-induced antibody responses. In this article, we obtain improved Chen-Ricci inequalities for submanifolds of generalized space forms with quarter-symmetric metric connection, with the help of which we completely characterized the Lagrangian submanifold in generalized complex space form and a Legendrian submanifold in a generalized Sasakian space form. We also discuss some geometric applications of the obtained results.

Oman has enormous diversity in important crops of global significance like wheat and barley considered as strategic crops. The country has an ancient cultivation history of barley because of its location on the eastern edge of the Arabian Peninsula and is known to have several landraces spread across all the agro-ecological regions. The present study was undertaken to comprehend genetic diversity in 16 indigenous Omani USDA accessions repatriated recently to select parents for hybridization in national crop improvement programs. The trial was conducted consecutively for two cropping seasons (2017–2018 and 2018–2019) from November to March on the layouts of loamy soil sites under sprinkler irrigation system in augmented design under spacing and crop husbandry practices as per national recommendations. The data on eight quantitative characters and presence (score 1) or absence (score 0) of anthocyanin pigmentation on two plant parts were subjected to both hierarchical cluster analysis (HCA) and principal component analysis (PCA) to know the contribution of characters toward diversity and form prime diverse clusters. The results indicated not only significant differences between accessions (p< 0.05) with respect to the most of quantitative characters but also two significant correlations involving four characters associated with maturity and inflorescence (p< 0.05). The results of two multivariate analyses led to the formation of four diverse clusters with similar constituents of accessions to the most extent, thus supporting each other in discerning diversity. The parents were selected for hybridization for improving characters of growth for higher yield or productivity with anthocyanin-related characters.

This is the first high-throughput amplicon sequencing study of mycobiome associated with leaves of the seagrass Halophila stipulacea. Five leaf samples were collected from the Al-Leith mangroves along the Red Sea coast of Saudi Arabia. Total DNA was extracted from 250 mg of each sample, and the first 300 bp (contains D1-D2 variable regions) of the LSU rDNA amplicon was sequenced with the Illumina MiSeq (bTEFAP). A total of 928,626 reads were obtained from the five samples. The sequence reads belonged to Metazoa (48.1% of the total reads), Viridiplantae (41.1%), Eukaryota (8.8%), Fungi (1.96%), Bacteria (0.09%), and Archaea (0.0001%). Fungi represented between 1.1% and 5.8% of the total reads in the five samples. A total of 18,279 reads (representing 1.96% of the total reads) were recorded from the 5 samples representing 296 molecular species (OTUs) that belong to 13 fungal phyla. At the phylum level, Basidiomycota dominated the community (37.2–51.6%) in three samples, while Neocallimastigomycota (37.5%) and Mucoromycota (42.1%) dominated the community in the fourth and the fifth samples, respectively. High diversity of OTUs (28 molecular species) were recorded from the monokaryotic subkingdom with five unknown basal lineages that are not aligning with any known taxa. Total number of sequence reads of microbial eukaryote organisms (Stramenopiles) from the five samples ranged between 0.16 of total reads in the fifth sample (AL-Hs05) to 2.9% in the first one (AL-Hs01). Majority of the microbial eukaryote reads (93.6%) belong to the phylum Oomycota, followed by Opisthokonts (Fungi/Metazoa group) representing 6.4% of microbial eukaryote reads. Monokaryon phyla (i.e., Chytridiomycota, Mucoromycota, and Neocallimastigomycota) and microbial eukaryotes occupied a major portion of the sequence reads followed by Basidiomycota and Ascomycota. Our results support the findings that the majority of fungi and microbial eukaryotes communities are so far unknown with seven deep branching lineages remain to be cultivated.

A set of five concentrations of MgO nanoparticles with zein and without zein-coating were incorporated into three oral mouth washes: Listerine zero, Listerine total control and Oral B in the mass percentages of 0.3%, 0.5%, 1%, 2%, 5% and 10%, in addition to controls with no MgO nanoparticles. The antimicrobial effect of three mouth washes with variable concentrations of MgO was tested against the following organisms: Staphylococcus aureus, Streptococcus mutans, Enterococcus faecalis and Candida albicans using the disc diffusion test (DDT) and direct contact test (DCT). Data were analyzed using one-way ANOVA statistical test.Results: The tested mouthwashes with zein-coated MgO nanoparticles showed significant differences of antimicrobial activity on S. mutans, S. aureus, E. faecalis, and C. albicans in the disc diffusion test. While in the DCT, all tested mouthwashes with MgO nanoparticles with and without zein coating showed antimicrobial activity on all tested microorganisms.Conclusion: Zein-coated MgO nanoparticles may be considered as a potential antimicrobial agent when added to oral mouthwashes. Future analysis, including in vivo studies, is required in order to incorporate zein/MgO nanoparticles into oral mouthwashes that may improve its antibacterial property.Keywords: MgO, nanoparticles, zein polymer, antibacterial, mouthwash

Currently, cancer is among the leading causes of death in the world. It is the correct diagnosis that gives the patient a better chance of healing. The detection of genetic markers, macromolecules that are produced directly by the tumor or by the host, has the potential to be one of the diagnostic tests. This paper is a review article, with the objective of addressing tumor markers c-MYC, n-MYC, protein p-53, protein Kras, and the gene HER-2 neu regarding the diagnosis and prognosis of patients with cancer, studies demonstrated that c-MYC is expressed in several malignant tumors because it affects proliferation and cellular metabolism, P-53 in breast and lung tumors is overexpressed, Regarding p16 it has been suggested that cytoplasmic also overexpression as kind of cancer predictor. K-ras is used to determine survival time and free time for neoplasm. It is an effective marker for monoclonal antibody therapy. The marker HER-2 neu and N-MYC are indicative of poor prognosis, conclusions: application of these markers is being increasingly used for expressing specificity and sensitivity in different areas of tumor characterization, in addition to being related to the therapeutic conduct to be followed in tumor treatment. To enhance the dye removal efficiency by natural enzyme, horseradish peroxidase (HRP) was immobilized onto amine-functionalized superparamagnetic iron oxide and used as a biocatalyst for the oxidative degradation of acid black-HC dye. The anchored enzyme was characterized by vibrating sample magnetometry, Fourier transform infrared spectroscopy, X-ray diffraction, thermogravimetry, scanning electron microscopy, Brunauer–Emmett–Teller and Barrett–Joyner–Halenda methods, nitrogen adsorption–desorption measurements, Zeta potential, energy dispersive X-ray spectroscopy, and transmission electron microscopy. The Michaelis constant values of free and immobilized HRP were determined to be 4.5 and 5 mM for hydrogen peroxide and 12.5 and 10 mM for guaiacol, respectively. Moreover, the maximum values of free and immobilized HRP were 2.4 and 2 U for H2O2, respectively, and 1.25 U for guaiacol. The immobilized enzyme was thermally stable up to 60°C, whereas the free peroxidase was stable only up to 40°C. In the catalytic experiment, the immobilized HRP exhibited superior catalytic activity compared with that of free HRP for the oxidative decolorization and removal of acid black-HC dye. The influence of experimental parameters such as the catalyst dosage, pH, H2O2 concentration, and temperature on the removal efficiency was investigated. The reaction followed second-order kinetics, and the thermodynamic activation parameters were determined.

Stem cell–derived β-like cells (sBC) carry the promise of providing an abundant source of insulin-producing cells for use in cell replacement therapy for patients with diabetes, potentially allowing widespread implementation of a practical cure. To achieve their clinical promise, sBC need to function comparably with mature adult β-cells, but as yet they display varying degrees of maturity. Indeed, detailed knowledge of the events resulting in human β-cell maturation remains obscure. Here we show that sBC spontaneously self-enrich into discreet islet-like cap structures within in vitro cultures, independent of exogenous maturation conditions. Multiple complementary assays demonstrate that this process is accompanied by functional maturation of the self-enriched sBC (seBC); however, the seBC still contain distinct subpopulations displaying different maturation levels. Interestingly, the surface protein ENTPD3 (also known as nucleoside triphosphate diphosphohydrolase-3 [NDPTase3]) is a specific marker of the most mature seBC population and can be used for mature seBC identification and sorting. Our results illuminate critical aspects of in vitro sBC maturation and provide important insights toward developing functionally mature sBC for diabetes cell replacement therapy.

The stock assessment of Klunzinger's mullet, Planiliza klunzengeri was conducted, to estimate its some aspects viz. growth, mortality, recruitment, yield-per-recruit and virtual population analysis in Iraqi marine waters, northwest Arabian Gulf, Iraq, from February 2020 and January 2021. The population parameters were analyzed by the FiSAT software using monthly length-frequency data collected by the Shaheen dhow and from the artisanal fishermen at the fish landing site in Al-Fao city to derive information required for their management. A total of 3319 individuals of P. klunzingeri ranged from 11.0 to 27.0 cm have been collected. Length-weight relationship was calculated as W= 0.026L2.716. The asymptotic length (L∞), growth rate (K) and the growth performance index (Ø') were 29.8 cm, 0.34 and 2.48, respectively. The rates of annual instantaneous of total mortality (Z), fishing mortality (F), natural mortality (M) and present exploitation (Epresent) were 1.19, 0.84, 0.36 and 0.30, respectively. The recruitment pattern of P. klunzingeri was continuous throughout the year, with one peak during May. The results of virtual population analysis revealed that the majority of P. klunzingeri were harvested between 14 cm and 19 cm. The length at first capture (L50) in the current study was higher than the length at first maturity (Lm) of the species in the region. The Epresent was below the biological target reference points (E0.1= 0.770 and Emax= 0.903), referred to the stock of P. klunzingeri is underexploited. Therefore, the harvest level should increase to its maximum sustainable yield level by increasing fishing effort level with monitoring surveys and evaluate the risk associated with fishing effort increases as fishing precautionary approaches.

Background The superior parietal lobule (SPL) is involved in somatosensory and visuospatial integration with additional roles in attention, written language, and working memory. A detailed understanding of the exact location and nature of associated white matter tracts could improve surgical decisions and subsequent postoperative morbidity related to surgery in and around this gyrus. Objective To characterize the fiber tracts of the SPL based on relationships to other well-known neuroanatomic structures through diffusion spectrum imaging (DSI)-based fiber tracking validated by gross anatomical dissection as ground truth. Methods Neuroimaging data of 10 healthy, adult control subjects was obtained from a publicly accessible database published in Human Connectome Project for subsequent tractographic analyses. White matter tracts were mapped between both cerebral hemispheres, and a lateralization index was calculated based on resultant tract volumes. Post-mortem dissections of 10 cadavers identified the location of major tracts and validated our tractography results based on qualitative visual agreement. Results We identified 9 major connections of the SPL: U-fiber, superior longitudinal fasciculus, inferior longitudinal fasciculus, inferior fronto-occipital fasciculus, middle longitudinal fasciculus, extreme capsule, vertical occipital fasciculus, cingulum, and corpus callosum. There was no significant fiber lateralization detected. Conclusion The SPL is an important region implicated in a variety of tasks involving visuomotor and visuospatial integration. Improved understanding of the fiber bundle anatomy elucidated in this study can provide invaluable information for surgical treatment decisions related to this region.

Background The COVID‐19 pandemic has been accompanied by the largest mobilization of therapeutic convalescent plasma (CCP) in over a century. Initial identification of high titer units was based on dose–response data using the Ortho VITROS IgG assay. The proliferation of severe acute respiratory syndrome coronavirus 2 serological assays and non‐uniform application has led to uncertainty about their interrelationships. The purpose of this study was to establish correlations and analogous cutoffs between multiple serological assays. Methods We compared the Ortho, Abbott, Roche, an anti‐spike (S) ELISA, and a virus neutralization assay. Relationships relative to FDA‐approved cutoffs under the CCP emergency use authorization were identified in convalescent plasma from a cohort of 79 donors from April 2020. Results Relative to the neutralization assay, the spearman r value of the Ortho Clinical, Abbott, Roche, anti‐S ELISA assays was 0.65, 0.59, 0.45, and 0.76, respectively. The best correlative index for establishing high‐titer units was 3.87 signal‐to‐cutoff (S/C) for the Abbott, 13.82 cutoff index for the Roche, 1:1412 for the anti‐S ELISA, 1:219 by the neutralization assay, and 15.9 S/C by the Ortho Clinical assay. The overall agreement using derived cutoffs compared to a neutralizing titer of 1:250 was 78.5% for Abbott, 74.7% for Roche, 83.5% for the anti‐S ELISA, and 78.5% for Ortho Clinical. DISCUSSION Assays based on antibodies against the nucleoprotein were positively associated with neutralizing titers and the Ortho assay, although their ability to distinguish FDA high‐titer specimens was imperfect. The resulting relationships help reconcile results from the large body of serological data generated during the COVID‐19 pandemic.

In this study, the population dynamics of the Arabian yellow fin sea bream, Acanthopagrus arabicus in the Iraqi marine waters, Arabian Gulf were assessed, to contributing towards the development of management plans for its sustainable exploitation. A. arabicus is one of the species caught in large quantities as commercial by artisanal fishers. Fish samples were collected by the Shaheen steel-hulled dhow and from the artisanal fishermen from February 2020 to January 2021. The length-weight relationship was described as W = 0.030L2.867, indicating a negative allometric growth pattern. Of 3484 individuals ranging in total length from 13.0 to 41.0 cm were used to evaluate the growth and mortality parameters. The asymptotic length (L∞), growth rate (K) and growth performance index (O') were 44.6 cm, 0.34 and 2.830, respectively. The total mortality (Z) was calculated as 1.17, while the natural and fishing mortality rates were 0.75 and 0.42, respectively, and the current exploitation rate (Epresent) was 0.36. Length at first capture (L50) was 21.9 cm. The recruitment pattern occurs throughout the year, with two recruitment peaks. The yield per recruit analysis indicates that the current exploitation rate was below the economic yield target and the maximum sustainable yield, indicating that this stock underexploited. Virtual population analysis results showed that the mid-lengths (16-22 cm) experienced the highest fishing mortality. So, for management purposes, more yields could be obtained by increasing the fishing activities on this species for a substantial harvest.

Background The medial occipital lobe, composed of the lingual gyrus and cuneus, is necessary for both basic and higher level visual processing. It is also known to facilitate cross-modal, nonvisual functions, such as linguistic processing and verbal memory, after the loss of the visual senses. A detailed cortical model elucidating the white matter connectivity associated with this area could improve our understanding of the interacting brain networks that underlie complex human processes and postoperative outcomes related to vision and language. Methods Generalized q-sampling imaging tractography, validated by gross anatomic dissection for qualitative visual agreement, was performed on 10 healthy adult controls obtained from the Human Connectome Project. Results Major white matter connections were identified by tractography and validated by gross dissection, which connected the medial occipital lobe with itself and the adjacent cortices, especially the temporal lobe. The short- and long-range connections identified consisted mainly of U-shaped association fibers, intracuneal fibers, and inferior fronto-occipital fasciculus, inferior longitudinal fasciculus, middle longitudinal fasciculus, and lingual–fusiform connections. Conclusions The medial occipital lobe is an extremely interconnected system, supporting its ability to perform coordinated basic visual processing, but also serves as a center for many long-range association fibers, supporting its importance in nonvisual functions, such as language and memory. The presented data represent clinically actionable anatomic information that can be used in multimodal navigation of white matter lesions in the medial occipital lobe to prevent neurologic deficits and improve patients' quality of life after cerebral surgery.

The growth, mortality and stock parameters of greenback mullet, Planiliza subviridis from Iraqi marine waters, northwest Arabian Gulf was assessed using FiSAT II software for length-frequency data collected from February 2020 to January 2021. P. subviridis is one of the species caught in large quantities as commercial by artisanal fishers. Fish samples were collected by the Shaheen steel-hulled dhow and from the artisanal fishermen. The total length and body weight relationship of fish was estimated as W= 0.034L2.670, indicating negative allometric growth. Of 3350 specimens, growth and mortality parameters were evaluated. The asymptotic length (L∞), growth rate (K) and growth performance index (Ø') were 33.8 cm, 0.30 and 2.535, respectively. The total mortality rate (Z), natural mortality rate (M), and fishing mortality rate (F) were 1.11, 0.74 and 0.38, respectively. The present exploitation rate (Epresent) of P. subviridis computed as 0.34. Length at first capture (L50) was 17.47 cm. Recruitment of P. subviridis was observed throughout the year, with a peak during July. The yield per recruit analysis indicates that the current exploitation rate was below the biological target reference points (E0.1 and Emax), which refers to the stock of P. subviridis is underexploited. Virtual population analysis results showed that mid-lengths (16-22 cm) experienced the highest fishing mortality. The length at first capture (L50) was higher than the length at first maturity (Lm) of the species. So, for management purposes, more yields could be obtained by increasing the fishing activities on this species for a substantial harvest.

Spergularia iraqensis sp. nov. is described as a new species from Iraq. This species has been collected from Diyala Province in the central east of Iraq; it is closely related to Spergularia rubra (L.) J. Presl & C. Presl, 1819 and Spergularia bocconei (Scheele) Graebn., 1919. The distinguishing of the morphological characteristics of the new species alongside the two similar species are discussed with photographs, and an identification key is given for Spergularia iraqensis and other closely related species.

Background The middle frontal gyrus (MFG) is involved in attention, working memory, and language-related processing. A detailed understanding of the subcortical white matter tracts connected within the MFG can facilitate improved navigation of white matter lesions in and around this gyrus and explain the postoperative morbidity after surgery. We aimed to characterize the fiber tracts within the MFG according to their connection to neuroanatomic structures through the use of diffusion spectrum imaging-based fiber tractography and validate the findings by gross anatomic dissection for qualitative visual agreement. Methods Tractography analysis was completed using diffusion imaging data from 10 healthy, adult subjects enrolled in the Human Connectome Project. We assessed the MFG as a whole component according to its fiber connectivity with other neural regions. Mapping was completed on all tracts within both hemispheres, with the resultant tract volumes used to calculate a lateralization index. A modified Klingler technique was used on 10 postmortem dissections to demonstrate the location and orientation of the major tracts. Results Two major connections of the MFG were identified: the superior longitudinal fasciculus, which connects the MFG to parts of the inferior parietal lobule, posterior temporal lobe, and lateral occipital cortex; and the inferior fronto-occipital fasciculus, which connected the MFG to the lingual gyrus and cuneus. Intra- and intergyral short association, U-shaped fibers were also identified. Conclusions Subcortical white matter pathways integrated within the MFG include the superior longitudinal fasciculus and inferior fronto-occipital fasciculus. The MFG is implicated in a variety of tasks involving attention and memory, making it an important cortical region. The postoperative neurologic outcomes related to surgery in and around the MFG could be clarified in the context of the anatomy of the fiber bundles highlighted in the present study.

Embryogenesis is guided by a limited set of signaling pathways that are reused at different times and places throughout development. How a context dependent signaling response is generated has been a central question of developmental biology, which can now be addressed with in vitro model systems. Our previous work in human embryonic stem cells (hESCs) established that pre-exposure of cells to WNT/{beta}-catenin signaling is sufficient to switch the output of ACTIVIN/SMAD2 signaling from pluripotency maintenance to mesendoderm (ME) differentiation. A body of previous literature has established the role of both pathways in ME differentiation. However, our work demonstrated that the two signals do not need to be present simultaneously and that hESCs have a means to record WNT signals. Here we demonstrate that hESCs have accessible chromatin at SMAD2 binding sites near pluripotency and ME-associated target genes and that WNT priming does not alter SMAD2 binding. Rather our results indicate that stable transcriptional output at ME genes results from WNT-dependent production of an additional SMAD2 co-factor, EOMES. We show that expression of EOMES can replace WNT signaling in ME differentiation, providing a mechanistic basis for WNT-priming and memory in early development.

Background and objective: Hypertension is a multifactorial disease where numerous constitutive, genetic, and environmental factors interplay. Among the constitutive factors, age is a major determent continuously reported to be associated with a significant increase in the prevalence of hypertension. In addition to age, Helicobacter pylori (H. pylori) infection was also shown to be associated. On the other hand, Vitamin D (Vit D) plays an important role in the development of hypertension. In the current study, it was investigated whether H. pylori interacts with Vit D levels to influence hypertension. Methods: This cross-sectional study was conducted on seven hundred and eighty-two “a priori” healthy individuals equally divided according to hypertension status. To study the association between Vit D, H. pylori, and hypertension, a multivariate logistic regression model was used while correcting for different confounding factors. Power analysis was also performed Results: Approximately half of the participants were hypertensive and had Vit D insufficiency and they were also matched for age. Using a multiple logistic regression model, the results showed an inversely proportionate association between H. pylori infection and the risk of hypertension (OR=0.38, P Conclusion: H. pylori infection and Vit D deficiency could predict hypertension. The odds of hypertension development were double when the participants were negative for H. pylori infection and had vitamin D deficiency.

Chromosomal instability leading to aneuploidy is pervasive in early human embryos1-3 and is considered as a major cause of infertility and pregnancy wastage4,5. Here we provide several lines of evidence that blastocysts containing aneuploid cells are worthy of in vitro fertilization transfer. First, we show clinically that aneuploid embryos can lead to healthy births, suggesting the presence of an in vivo mechanism to eliminate aneuploidy. Second, early development and cell specification modelled in micropatterned human 'gastruloids' grown in confined geometry show that aneuploid cells are depleted from embryonic germ layers, but not from extraembryonic tissue, by apoptosis in a bone morphogenetic protein 4 (BMP4)-dependent manner. Third, a small percentage of euploid cells rescues embryonic tissue in mosaic gastruloids when mixed with aneuploid cells. Finally, single-cell RNA-sequencing analysis of early human embryos revealed a decline of aneuploidy beginning on day 3. Our findings challenge two current dogmas: that a single trophectoderm biopsy at blastocyst stage to perform prenatal genetic testing can accurately determine the chromosomal make-up of a human embryo, and that aneuploid embryos should be withheld from embryo transfer in association with in vitro fertilization.

Introduction:The Sultanate of Oman is rich in diversity of the most important crops like wheat, which not only has a global significance but is also regarded as one of the strategic crops in the country. The country has an ancient cultivation history of both bread wheat (Triticum aestivumL.s.l.) and durum wheat (Triticum turgidumsub sp.durum) because of its characteristic location on the eastern edge of the Arabian Peninsula. Wheat landraces constitute the prime genetic resources of cultivated wheat not only in Oman but also in several MENA (the Middle East and North Africa) countries. Indigenous landraces have paramount significance for their potential utilization in crop improvement and conservation programs. Hence, the present study was undertaken to subject 17 indigenous durum wheat accessions for analyses of diversity to select parents for hybridization in national crop improvement programs.Materials and Methods:The trial was conducted consecutively for two cropping seasons (2017-2018 and 2018-2019) during winter from November to March on the layouts of a loamy soil site under sprinkler irrigation system in Augmented Design with five check varieties replicated five times randomized and distributed throughout the experimental area under spacing and crop husbandry practices as per national recommendations. The data on 9 quantitative (Plant descriptors) and 6 qualitative traits on the presence (score 1) or absence (score 0) of pigmentation on 6 plant parts were collected. These traits were subjected to both Principal Component Analysis (PCA) and Hierarchical Cluster Analysis (HCA) to comprehend the contribution of these characters towards diversity and form prime diverse clusters from 17 indigenous durum wheat landraces to select appropriate parents for crossing.Results:The results indicated that indigenous durum wheat accessions were significantly different (p>0.05) with respect to all the quantitative characters except the number of tillers. Of 36 combinations of associations among 9 agro-morphological characters’ studied, only six correlations involving four charactersviz. tiller no., spikelets/ spike, grains/spike, and grain length were found significant (pConclusion:The indigenous durum wheat landraces / accessions were found to be more diverse and potential for use in the national crop improvement programs for higher productivity.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has caused the global COVID-19 pandemic. Rapidly spreading SARS-CoV-2 variants may jeopardize newly introduced antibody and vaccine countermeasures. Here, using monoclonal antibodies (mAbs), animal immune sera, human convalescent sera and human sera from recipients of the BNT162b2 mRNA vaccine, we report the impact on antibody neutralization of a panel of authentic SARS-CoV-2 variants including a B.1.1.7 isolate, chimeric strains with South African or Brazilian spike genes and isogenic recombinant viral variants. Many highly neutralizing mAbs engaging the receptor-binding domain or N-terminal domain and most convalescent sera and mRNA vaccine-induced immune sera showed reduced inhibitory activity against viruses containing an E484K spike mutation. As antibodies binding to spike receptor-binding domain and N-terminal domain demonstrate diminished neutralization potency in vitro against some emerging variants, updated mAb cocktails targeting highly conserved regions, enhancement of mAb potency or adjustments to the spike sequences of vaccines may be needed to prevent loss of protection in vivo.

Background: Esophageal adenocarcinoma (EAC) is a deadly disease with limited treatment options. STING is a transmembrane protein that activates transcription of interferon genes, resulting in stimulation of APCs and enhanced CD8+ T-cell infiltration. The present study evaluates STING agonists, alone and in combination with radiation to determine durable anticancer activity in solid tumors. Materials and Methods: Esophagojejunostomy was performed on rats to induce reflux leading to the development of EAC. At 32 weeks post operatively, rats received intratumorally either 50 μg STING (ADU-S100) or placebo (PBS), +/– 16Gy radiation. Drug activity was evaluated by pre- and post- treatment MRI, histology, immunofluorescence and RT-PCR. Results: Mean MRI tumor volume decreased by 30.1% and 50.8% in ADU-S100 and ADU-S100 + radiation animals and increased by 76.7% and 152.4% in placebo and placebo + radiation animals, respectively (P < 0.0001). Downstream gene expression, pre- to on- and post- treatment, demonstrated significant upregulation of IFNβ, TNFα, IL-6, and CCL-2 in the treatment groups vs. placebo. On- or post- treatment, radiation alone, ADU-S100 alone, and ADU-S100 + radiation groups demonstrated enhanced PD-LI expression, induced by upregulation of CD8+ T-cells (p < 0.01). Conclusions: ADU-S100 +/– radiation exhibits potent antitumor activity and a promising immunomodulatory profile in a de novo EAC.

Background: In this study, we assessed the in vitro antischistosomal activity of the active ingredients of Allium sativum (allicin) and Curcuma longa (curcumin) on Schistosoma mansoni. Methods: This study was conducted in Faculty of Science, Port said University, Egypt (2018). Adult worms were exposed to a range of concentrations of AL or CU, and worm survival was assessed 24 h post-exposure to calculate the lethal concentration of the compounds. Scanning electron microscopy was used to assess ultrastructural changes in the surface of AL- or CU- treated worms. The genotoxicities of AL and CU on S. mansoni were determined by DNA fragmentation analysis. Results: We determined the concentrations of AL and CU required to kill 50% of S. mansoni (LC50). The LC50 of AL was 8.66 µL/mL, whereas 100% mortality of S. mansoni was achieved by AL at concentrations of 50 µL/mL. The LC50 of CU was 87.25 µL/mL, with the highest mortality of 91.3% seen after 24 h exposure to 100 µg/mL CU. Ultrastructural studies revealed that exposure to either AL or CU led to mild or severe surface damage to S. mansion, respectively. The degree of damage in the worms was sex-dependent. Interestingly, while CU exposure resulted in DNA fragmentation in S. mansoni worms, we observed no genotoxic effects of AL. Conclusion: Both AL and CU exhibit antischistosomal activity; the study provided evidence suggesting that these compounds act through distinct mechanisms. These promising results encourage further investigation into these compounds as potential antischistosomal agents, either alone or as complementary treatments to praziquantel.

Novel extracted natural stabilizer for the polymer was investigated in this study. The natural anti-aging was extracted from plants and it is demonstrated that this type of stabilizers are suitable for biodegradable polymers. Gynostemma pentaphyllum (0.01–0.03 wt%) were extracted and added to the poly(vinyl chloride) blend. The achieved polymeric films were exposed to an accelerated UV light aging process for 300 hours at ambient temperature. The efficiency of tested natural stabilizer was linked to blank PVC films. FTIR spectroscopy, weight loss, electronic microscope and scanning electron microscopy were performed. Using small amount of natural stabilizer is significantly beneficial because it does not influence the PVC properties.

This is the first report of thraustochytrids from Saudi Arabia. A total of 108 isolates of thraustochytrid were cultured from Syhat mangroves, Arabian Gulf, Saudi Arabia. Isolated thraustochytrids belonged to five genera: Aplanochytrium, Aurantiochytrium, Schizochytrium, Thraustochytrium and Ulkenia. Cultured thraustochytrids isolated from decaying leaves of Avicennia marina (77 isolates), sediment (15), seawater (10) and decaying thalli of Sargassum (6). Of the 108 isolates, three strains (SY25, SY38 and SY52) were selected based on their high biomass productivity and high percentages of PUFAs. Phylogenetic analyses based on 18S rDNA placed the three strains within the Aurantiochytrium clade with high statistical support. Species of Aurantiochytrium formed six separate clades, the two strains (SY38 and SY52) formed a separate clade that is a sister clade to the one that contains the type species A. limacinum, while SY25 grouped with Aurantiochytrium sp. TA4, that is also isolated from mangroves in Iran, Arabian Gulf. The strains (SY38 and SY52) shared the phylogenetic placement, their morphology and fatty acid profile. The strain SY25 have different shape of sporangia that divide to give zoospores directly, sporogenous cells are surrounded by thick gelatinous sheath and produce high levels of Linoleic and Oleic essential unsaturated fatty acids. The three studied strain produced high levels of Palmitic acid (ranged between 31.1 and 65.3 % of total fatty acids) that can be further optimized for biofuel production.

Recent advances in human naive pluripotent stem cell culture have demonstrated their ability to generate trophectoderm and descendant trophoblast cell types. Moreover, the same cells when cultured in three-dimensional configurations self-organize to generate blastocyst-like structures called blastoids. These discoveries represent a major step forward in modeling early human embryonic development.

The direct association between some microbial species and cancers, such as in colorectal cancer (CRC), has been disclosed. The aim of this study was to evaluate the changes in intestinal microbiota in subjects with CRC compared with healthy group. Three-hundred fecal specimens were gathered from patients with CRC and 300 from healthy individuals during March 2014 to October 2019 from two hospitals in Tehran. The informed consent form and the questionnaire were completed by the patients. Following the identification of Lactobaccilus acidophilus (L. acidophilus), Lactobacillus palntarom (L. palntarom), and Enterococcus faecalis (E. faecalis), the number of bacteria was determined using quantitative real-time PCR (qPCR). The patients’ age range was 20–76 years (mean: 55.34 ± 3.66). The qPCR clarified that number of E. faecalis was 2.2-fold higher in patients with CRC compared to healthy population (p = 0.0013). Additionally, the number of L. acidophilus and L. plantarom was 3.4-fold (p < 0.0001) and 4.8-fold (p < 0.0001) higher in healthy population. The inhibitory effect of intestinal microflora against the CRC development was proposed by observation of the changes in intestinal microbiota and determining their composition in subjects with CRC compared with that of healthy individuals. Microbiota was considered as a goal for the prevention and treatment of CRC. The relationship between microbiota and human health would be known deeper; this knowledge provides insights into the management of intestinal microbiota and therapeutics.

Background Middle East respiratory syndrome coronavirus (MERS-CoV) has continued to cause sporadic outbreaks of severe respiratory tract infection over the last 8 years. Methods Complete genome sequencing using next-generation sequencing was performed for MERS-CoV isolates from cases that occurred in Riyadh between 2015 and 2019. Phylogenetic analysis and molecular mutational analysis were carried out to investigate disease severity. Results A total of eight MERS-CoV isolates were subjected to complete genome sequencing. Phylogenetic analysis resulted in the assembly of 7/8 sequences within lineage 3 and one sequence within lineage 4 showing complex genomic recombination. The isolates contained a variety of unique amino acid substitutions in ORF1ab (41), the N protein (10), the S protein (9) and ORF4b (5). Conclusion Our study shows that MERS-CoV is evolving. The emergence of new variants carries the potential for increased virulence and could impose a challenge to the global health system. We recommend the sequencing every new MERS-CoV isolate to observe the changes in the virus and relate them to clinical outcomes.

This work aimed to measure serum vascular endothelial growth factor (VEGF) levels before and after Conventional transarterial chemoembolization (cTACE) versus drug-eluting beads (DEB)-TACE and evaluate its efficacy in predicting response to therapy and tumor recurrence.114 patients with unresectable hepatocellular carcinoma complicating hepatitis C virus-related cirrhosis were included. They underwent cTACE (58) or DEB-TACE (56). VEGF serum levels were measured before and on days 1 and 30 after TACE. Patients with complete response (CR) after TACE were followed-up for one year. Statistical analysis was done.VEGF level was higher than baseline after cTACE (VEGF serum levels may predict response to therapy in patients treated by DEB-TACE or cTACE but it has no relation to tumor recurrence.

Summary Influenza viruses remain a major public health threat. Seasonal influenza vaccination in humans primarily stimulates pre-existing memory B cells, leading to a transient wave of circulating antibody-secreting plasmablasts1–3. This recall response contributes to “original antigenic sin,” the selective boosting of antibody specificities from prior exposures to influenza virus antigens4. It remains unclear whether such vaccination can also induce germinal centre (GC) reactions in the draining lymph node (LN) where diversification and maturation of recruited B cells can occur5. Here we used ultrasound-guided fine needle aspiration to serially sample the draining LNs and investigate the dynamics and specificity of GC B cell responses after influenza vaccination in humans. We show that influenza vaccine-binding GC B cells can be detected as early as 1 week after vaccination. In 3 out of 8 participants, we detected vaccine-binding GC B cells up to 9 weeks after vaccination. Between 12% and 88% of the responding GC B cell clones overlapped with those detected among early circulating plasmablasts. These shared B cell clones had high frequencies of somatic hypermutation (SHM) and encoded broadly cross-reactive monoclonal antibodies (mAbs). In contrast, vaccine-induced B cell clones detected only in the GC compartment exhibited significantly lower SHM frequencies and predominantly encoded strain-specific mAbs, suggesting a naïve B cell origin. Electron microscopy-based epitope mapping revealed that some of these strain-specific mAbs recognized epitopes that were not targeted by the early plasmablast response. Our results indicate that influenza virus vaccination of humans can elicit a GC reaction to which B cell clones targeting novel epitopes are more likely to be recruited, thereby broadening the spectrum of vaccine-induced protective antibodies against this rapidly mutating pathogen.

Small cell lung cancer (SCLC) is an aggressive neuroendocrine cancer characterized by initial chemosensitivity followed by emergence of chemoresistant disease. To study roles for MYCN amplification in SCLC progression and chemoresistance, we developed a genetically engineered mouse model of MYCN-overexpressing SCLC. In treatment-naïve mice, MYCN overexpression promoted cell cycle progression, suppressed infiltration of cytotoxic T cells, and accelerated SCLC. MYCN overexpression also suppressed response to cisplatin–etoposide chemotherapy, with similar findings made upon MYCL overexpression. We extended these data to genetically perturb chemosensitive patient-derived xenograft (PDX) models of SCLC. In chemosensitive PDX models, overexpression of either MYCN or MYCL also conferred a switch to chemoresistance. To identify therapeutic strategies for MYCN-overexpressing SCLC, we performed a genome-scale CRISPR–Cas9 sgRNA screen. We identified the deubiquitinase USP7 as a MYCN-associated synthetic vulnerability. Pharmacological inhibition of USP7 resensitized chemoresistant MYCN-overexpressing PDX models to chemotherapy in vivo. Our findings show that MYCN overexpression drives SCLC chemoresistance and provide a therapeutic strategy to restore chemosensitivity.

In the present study, the start codon targeted (SCoT) technique was used for measuring the genetic variability between two fish species of family Moronidae (Dicentrarchus punctus and Dicentrarchus labrax), these species were collected from the Mediterranean Sea, Port Said, Egypt. Eleven SCoT primers were used in the study of generating different lengths of amplicons that ranged from 150bp to 1800bp (SCoT-2, SCoT-3, SCoT-4, SCoT-6, SCoT-7, SCoT-8, SCoT-9, SCoT-10, SCoT-28, SCoT-35 and SCoT-46). The polymorphism percent ranged from 18% with SCOT-28 primer to 75% with SCOT-6. The genetic similarity between the two species was 75.5%. Protein analysis using Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) was also performed in this study producing a total of 22 bands that ranged in size from 19KD to 200KD. Each of the two species produced 22 bands.

Pomegranate fruit extract contains many polyphenols and flavonoids of diverse biological importance including anticancer potential. In cancer, the angiogenesis process facilitates solid cancer growth and metastasis. Here, the antiangiogenic effect of pomegranate fruit extract against human pancreatic cancer (Suit-2) and colon (colo205) cell lines in the chick chorioallantoic membrane (CAM) model was studied along with the effect of pomegranate fruit extract on fibroblast growth factor (FGF2). Pomegranate fruit extract significantly reduced the tumor weight and hemoglobin content in CAM models of pancreatic Suit-2 and colon colo205.

Aging and aging-associated diseases are issues with unsatisfactory answers in the medical field. Aging causes important physical changes which, even in the absence of the usual risk factors, render the cardiovascular system prone to some diseases. Although aging cannot be prevented, slowing down the rate of aging is entirely possible to achieve. In some traditional medicine, medicinal herbs such as Ginseng, Radix Astragali, Ganoderma lucidum, Ginkgo biloba, and Gynostemma pentaphyllum are recognized by the “nourishing of life” and their role as anti-aging phytotherapeutics is increasingly gaining attention. By mainly employing PubMed here we identify and critically analysed 30 years of published studies focusing on the above herbs' active components against aging and aging-associated conditions. Although many plant-based compounds appear to exert an anti-aging effect, the most effective resulted in being flavonoids, terpenoids, saponins, and polysaccharides, which include astragaloside, ginkgolide, ginsenoside, and gypenoside specifically covered in this review. Their effects as antiaging factors, improvers of cognitive impairments, and reducers of cardiovascular risks are described, as well as the molecular mechanisms underlying the above-mentioned effects along with their potential safety. Telomere and telomerase, PPAR-α, GLUTs, FOXO1, caspase-3, bcl-2, along with SIRT1/AMPK, PI3K/Akt, NF-κB, and insulin/insulin-like growth factor-1 pathways appear to be their preferential targets. Moreover, their ability to work as antioxidants and to improve the resistance to DNA damage is also discussed. Although our literature review indicates that these traditional herbal medicines are safe, tolerable, and free of toxic effects, additional well-designed, large-scale randomized control trials need to be performed to evaluate short- and long-term effects and efficacy of these medicinal herbs.
