Salinity is one of the largest stresses blocking horizontal and vertical expansion in agricultural lands. Establishing salt-tolerant genotypes is a promising method to benefit from poor water quality and salinized lands. An integrated method was developed for accomplishing reliable and effective evaluation of traits stability of salt-tolerant wheat. The study aims were to estimate the genetic relationships between explanatory traits and shoot dry matter (SDM), and determine the traits stability under three salinity levels. Morphophysiological and biochemical traits were evaluated as selection criteria for SDM improvement in wheat for salinity tolerance. Three cultivars and three high-yielding doubled haploid lines (DHLs) were used. Three salt (NaCl) levels (control (washed sand), 7 and 14 dS m−1) were applied for 45 days (at the first signs of death in the sensitive genotypes). All morphophysiological traits gradually decreased as salinity levels increased, excluding the number of roots. Decreases were more visible in sensitive genotypes than in tolerant genotypes. All biochemical traits increased as salinity levels increased. Variance inflation factors (VIFs) and condition number exhibited multicollinearity for membrane stability index and polyphenol oxidase activity. After their removal, all VIFs were 0.60%) and genetic gain (>20%) and highly significant genetic correlation, co-heritability, and selection efficiencies for SDM. CHL and CAT could be used as selection criteria for salinity tolerance in wheat-breeding programs. The tolerated line (DHL21) with the check cultivar (Sakha 93) can be also recommended as novel genetic resource for improving salinity tolerance of wheat.

Rose-ringed parakeet (P. krameri) is commonly found native psittacid in Pakistan. It is most popular companion bird in Pakistan. It is an unprotected species as per The Punjab Wildlife Act, 1974. The objectives of the present study were to study its population paradigm and basic nesting biology in the Gujar Khan, Punjab, Pakistan. Previously, no studies were carried out to assess its live and vacant nest cavities, nesting site, nest height, cavity position on substrate, and potential roosts in the area. Moreover, this study also assesses the potential threats and factors in this area and conservation of the parakeet. Results show that in the study area (36.77%) active and (63.22%) inactive nests were found. A total of 10 tree species were utilized to make nests. It was found Ficus bengalensis, Syzygium cumini, Morus alba, Melia azedarach and Broussonetia papyrifera as the most used tree species for nesting due to more frequencies and old ages. Parakeets make nest at 3-11 m height. The preferred nest height (42.48%) was in the range of 7-9 m, while (5.88%) least at 3-5 m height. For safety reasons they made more nests (45.09%) on the tree trunk followed by terminal (31.37%) and at fork (23.52%). Due to smaller thriving population we found only (mean=7±4) parakeets during roost counting. Severe cutting of trees, destruction of its habitat and poaching for selling in the bird market are the main causes of its population decline. Government must change its status from unprotected to protected species and should ban its dealing for pet.

This study evaluated the genetic diversity among 18 inbred lines of Pumpkin (Cucurbita moschata) using HFO-TAG (high frequency oligonucleotide–targeting active gene) markers. The inbred lines were selected from six different populations in Saudi Arabia and Egypt according to genetic distance and population origin. A total of 132 alleles were detected. Sixteen HFO-TAG markers averaged 8.25 alleles per primer with 125 alleles showing polymorphisms that averaged 7.81 alleles per primer. A cluster analysis of the means of the HFO-TAG data created three groups with similarity coefficients between 0.34 and 0.74. In general, the 18 inbred lines were separated according to pedigree and population origin. Hybrids from the chosen inbred lines, S1 L-1 with E L-2, S1 L-2 with E L-3, and S1 L-3 with E L-1, may be the basis for improved hybridization programs and can be used in future hybridization program

Non-destructive methods have been widely recognized for evaluating fruit quality traits of many horticultural crops and food processing industry. Destructive (analytical) test, and non-destructive evaluation of the quality traits were investigated and compared for ‘Red Rose’ tomato (Solanum lycopersicum L.) fruit grown under protected environment. Fresh tomato fruit at five distinctive maturity stages namely; breaker (BK), turning (TG), pink (PK), light-red (LR), and red (RD) were labeled and scanned using the handheld near infra-red (NIR) enhanced spectrometer at a wavelength range of 285–1200 nm. The labeled tomato samples were then measured analytically for flesh firmness, lycopene, β-carotene, total phenolic content (TPC) and total flavonoids content (TFC). The results revealed that quality traits could be estimated using NIR spectroscopy with a relatively high coefficient of determination (R2): 0.834 for total phenolic content, 0.864 for lycopene, 0.790 for total flavonoid content, 0.708 for β-carotene; and 0.679 for flesh firmness. The accumulation of Lyco and β-Car rapidly increased in tomatoes harvested between the TG and the LR maturity stages. Harvesting tomatoes at BK maturity stage resulted in significantly higher flesh firmness than harvesting at the later maturity stages. Tomato fruits had the lowest TPC and TFC contents at the earliest maturity stage (BK), while they had intermediate TPC and TFC levels at LR and RD maturity stages. NIR spectroscopic measurements of fruit firmness and lipophilic antioxidants in tomato fruit at various maturity stages were partially in accordance with those estimated by destructive (analytical) methods. Based on these findings, we recommend using non-destructive NIR spectroscopy as an effective tool for predicting tomato fruit quality during harvest stage and postharvest processing.

The use of synthetic antioxidants has been associated with serious concerns for human and environmental health. During ripening stages, tomato fruit is exposed to different abiotic stresses which not only influence its nutritional, mechanical, and functional properties at harvest, but also affect the quality and shelf life of the fruit during storage. This study investigated the pattern of changes in dietary antioxidants during various ripening stages of tomato fruit (cv. Red Rose) and their impact on storage behavior of the fruit during cold storage. Tomato fruits were harvested at mature green, breaker, turning, pink, light-red and red stages of maturity. Then, they were analysed for flesh firmness, soluble solids content, titratable acidity, total sugars, pH, dry matter content, lipophilic (lycopene, β-carotene, and total carotenoids), and hydrophilic (ascorbic acid, phenolic and flavonoids) antioxidants. Additional fruits were harvested at each maturity stage and divided into three equal lots, then were subjected to low-temperature (10 ± 1 °C) storage with 80 ± 5% RH, for 7, 14, and 21 days. Flesh firmness, and the levels of dietary antioxidants were analysed following the subsequent storage periods. The results revealed that the peak of hydrophilic antioxidants such as ascorbic acid, phenolic compounds, and flavonoids was between the ‘pink’ and the ‘light-red’ stages of fruit maturity. Whereas tomatoes harvested at the ‘red’ stage of maturity had the highest levels of lycopene and β-carotene. Both the stage of fruit maturity at harvest and duration of cold storage influenced flesh firmness, organoleptic and functional properties of ‘Red Rose’ tomato fruit. In conclusion, the results of the current investigation have practical implications in formulating foods with improved functional properties at processing industries.

Despite numerous reports on the role of nitric oxide (NO) in regulating plants growth and mitigating different environmental stresses, its participation in sulfur (S) -metabolism remains largely unknown. Therefore, we studied the role of NO in S acquisition and S-assimilation in tomato seedlings under low S-stress conditions by supplying NO to the leaves of S-sufficient and S-deficient seedlings. S-starved plants exhibited a substantial decreased in plant growth attributes, photosynthetic pigment chlorophyll (Chl) and other photosynthetic parameters, and activity of enzymes involved in Chl biosynthesis (δ-aminolevulinic acid dehydratase), and photosynthetic processes (carbonic anhydrase and RuBisco). Also, S-deficiency enhanced reactive oxygen species (ROS) (superoxide and hydrogen peroxide) and lipid peroxidation (malondialdehyde) levels in tomato seedlings. Contrarily, foliar supplementation of NO to S-deficient seedlings resulted in considerably reduced ROS formation in leaves and roots, which alleviated low S-stress-induced lipid peroxidation. However, exogenous NO enhanced proline accumulation by increasing proline metabolizing enzyme (Δ1-pyrroline-5-carboxylate synthetase) activity and also increased NO, hydrogen sulfide (a gasotransmitter small signaling molecule) and S uptake, and content of S-containing compounds (cysteine and reduced glutathione). Under S-limited conditions, NO improved S utilization efficiency of plants by upregulating the activity of S-assimilating enzymes (ATP sulfurylase, adenosine 5-phosphosulfate reductase, sulfide reductase and O-acetylserine (thiol) lyase). Under S-deprived conditions, improved S-assimilation of seedlings receiving NO resulted in improved redox homeostasis and ascorbate content through increased NO and S uptake. Application of 2-(4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxy l-3-oxide (an NO scavenger) invalidated the effect of NO and again caused low S-stress-induced oxidative damage, confirming the beneficial role of NO in seedlings under S-deprived conditions. Thus, exogenous NO enhanced the tolerance of tomato seedlings to limit S-triggered oxidative stress and improved photosynthetic performance and S assimilation.

Many underutilized tree species are good sources of food, fodder and possible therapeutic agents. Balanites aegyptiaca (L.) Delile belongs to the Zygophyllaceae family and is popularly known as “desert date”, reflecting its edible fruits. This tree grows naturally in Africa, the Middle East and the Indian subcontinent. Local inhabitants use fruits, leaves, roots, stem and root bark of the species for the treatment of various ailments. Several research studies demonstrate that extracts and phytochemicals isolated from desert date display antioxidant, anticancer, antidiabetic, anti-inflammatory, antimicrobial, hepatoprotective and molluscicidal activities. Mesocarp of fruits, seeds, leaves, stem and root bark are rich sources of saponins. These tissues are also rich in phenolic acids, flavonoids, coumarins, alkaloids and polysterols. Some constituents show antioxidant, anticancer and antidiabetic properties. The objective of this review is to summarize studies on diverse bioactive compounds and the beneficial properties of B. aegyptiaca.

Owing to the active use of rare-earth elements in many areas, it is necessary to study their behavior in the environment and their biological impact on plants. Despite the role of melatonin and sulfur in plant growth, development and abiotic stress tolerance; it is still not clear how they have a strong regulatory influence and synergistic effect on growth, physiological and biochemical characteristics of plants under different environmental stresses. Therefore, this study highlights how melatonin and sulfur together potentially involved in a reversal of lanthanum-inhibited photosynthetic and growth responses in tomato seedlings. Here, we reported that seedlings grown in a medium containing 150 μM lanthanum exhibited increased overproduction of reactive oxygen species (ROS) and lipid peroxidation together with increased Chlorophyll degradation, and activity of chlorophyllase, proline dehydrogenase and glycolate oxidase (GOx), and decreased photosynthesis and growth. However, the application of melatonin and sulfur showed significant responses on tomato seedlings, although the response of their combined treatment was more effective by further increasing photosynthesis and growth under lanthanum toxicity. Melatonin supplied with sulfur suppressed ROS formation, lipid peroxidation and activity of GOx, and increased photosynthesis by upregulating activities of carbonic anhydrase and ribulose-1,5-bisphosphate carboxylase/oxygenase. Also, sulfur supplementation with melatonin to seedlings resulted in an elevation in the accumulation of Chl and proline by increasing δ-aminolevulinic acid and activity of δ-aminolevulinic acid dehydratase and Δ1-pyrroline-5-carboxylate synthetase activity. The administration of melatonin with sulfur substantially induced upregulation of enzymes (superoxide dismutase, ascorbate peroxidase, monodehydroascorbate reductase, dehydroascorbate reductase and glutathione reductase) activities involved in the antioxidant system, thereby mitigating ROS-induced oxidative damage. Thus, this study provides strong evidence that melatonin and sulfur have strong regulatory influence and synergistic role in alleviating the adverse effect of lanthanum-toxicity by increasing photosynthesis and growth.

The present study reports on the effect of humic and salicylic acids on the growth, yield, and fruit quality of three red sweet pepper (Capsicum annuum) cultivars: Barbero, Ferrari, and Imperio. The plants were grown in a greenhouse and the leaves were treated with humic or salicylic acids at 0, 0.5, 1.0, and 1.5 g·L−1 at 20, 40, and 60 days after transplanting. Foliar application of humic or salicylic acids significantly increased vegetative growth, fruit yield, and quality of the three cultivars as compared with the control plants. However, salicylic acid treatment proved more effective than humic acid treatment. Red sweet pepper plants of all three cultivars sprayed with 1.5 g·L−1 salicylic acid showed the greatest vegetative growth; fruit yield components, such as fruit number, diameter, and fresh and dry weights; and fruit quality traits, such as vitamin C content, total soluble solid content, titratable acidity, and total sugar content, than the plants in all other treatments. There were significant differences (P ≤ 0.05) among cultivars in response to humic and salicylic acid foliar application; ‘Ferrari’ showed significantly higher yield and productivity than ‘Barbero’ or ‘Imperio’. ‘Ferrari’ plants sprayed with 1.5 g·L−1 salicylic acid showed the highest fruit weight (202.41 g) and flesh thickness (68 mm), both of which are preferred by consumers, and therefore, have increased market value. This treatment also increased total yield by 27.7% (16.03 t·ha−1), 15.9% (12.38 t·ha−1), and 17.9% (11.88 t·ha−1) in ‘Barbero’, ‘Ferrari’, and ‘Imperio’, respectively. Therefore, salicylic acid foliar application is recommended for enhancing fruit yield and quality of greenhouse-grown red sweet pepper.

Social organisms combat pathogens through individual innate immune responses or through social immunity—behaviors among individuals that limit pathogen transmission within groups. Although we have a relatively detailed understanding of the genetics and evolution of the innate immune system of animals, we know little about social immunity. Addressing this knowledge gap is crucial for understanding how life-history traits influence immunity, and identifying if trade-offs exist between innate and social immunity. Hygienic behavior in the Western honey bee, Apis mellifera, provides an excellent model for investigating the genetics and evolution of social immunity in animals. This heritable, colony-level behavior is performed by nurse bees when they detect and remove infected or dead brood from the colony. We sequenced 125 haploid genomes from two artificially selected highly hygienic populations and a baseline unselected population. Genomic contrasts allowed us to identify a minimum of 73 genes tentatively associated with hygienic behavior. Many genes were within previously discovered QTLs associated with hygienic behavior and were predictive of hygienic behavior within the unselected population. These genes were often involved in neuronal development and sensory perception in solitary insects. We found that genes associated with hygienic behavior have evidence of positive selection within honey bees (Apis), supporting the hypothesis that social immunity contributes to fitness. Our results indicate that genes influencing developmental neurobiology and behavior in solitary insects may have been co-opted to give rise to a novel and adaptive social immune phenotype in honey bees.

Understanding salt tolerance in tomato (Solanum lycopersicum L.) landraces will facilitate their use in genetic improvement. The study assessed the morpho-physiological variability of Hail tomato landraces in response to different salinity levels at seedling stages and recommended a tomato salt-tolerant landrace for future breeding programs. Three tomato landraces, Hail 548, Hail 747, and Hail 1072 were tested under three salinity levels: 75, 150, and 300 mM NaCl. Salinity stress reduced shoots’ fresh and dry weight by 71% and 72%, and roots were 86.5% and 78.6%, respectively. There was 22% reduced chlorophyll content, carotene content by 18.6%, and anthocyanin by 41.1%. Proline content increased for stressed treatments. The 300 mM NaCl treatment recorded the most proline content increases (67.37 mg/g fresh weight), with a percent increase in proline reaching 61.67% in Hail 747. Superoxide dismutase (SOD) activity decreased by 65% in Hail 548, while it relatively increased in Hail 747 and Hail 1072 treated with 300 mM NaCl. Catalase (CAT) activity was enhanced by salt stress in Hail 548 and recorded 7.6%, increasing at 75 and 5.1% at 300 mM NaCl. It revealed a reduction in malondialdehyde (MDA) at the 300 mM NaCl concentration in both Hail 548 and Hail 1072 landraces. Increasing salt concentrations showed a reduction in transpiration rate of 70.55%, 7.13% in stomatal conductance, and 72.34% in photosynthetic rate. K+/Na+ ratios decreased from 56% for 75 mM NaCl to 85% for 300 mM NaCl treatments in all genotypes. The response to salt stress in landraces involved some modifications in morphology, physiology, and metabolism. The landrace Hail 548 may have better protection against salt stress and observed protection against reactive oxygen species (ROS) by increasing enzymatic “antioxidants” activity under salt stress.

Garcinia morella (Gaertn.) Desr. is an evergreen tree that yields edible fruits, oil, and resin. It is a source of “gamboge”, a gum/resin that has a wide range of uses. The fruits, leaves, and seeds of this tree are rich in bioactive compounds, including xanthones, flavonoids, phenolic acids, organic acids, and terpenoids. Evidence from different studies has demonstrated the antioxidant, antifungal, antiviral, hepatoprotective, anticancer, anti-inflammatory, antibacterial, and larvicidal activities of the fruit, leaf, and seed extracts of G. morella. This review summarizes the information on the phytochemicals of G. morella and the biological activities of its active constituents.

The recent decline in honey bee (Hymenoptera: Apidae) colony health worldwide has had a significant impact on the beekeeping industry as well as on pollination-dependent crop sectors in North America and Europe. The pollinator crisis has been attributed to many environmental and anthropological factors including less nutrient rich agricultural monocultures, pesticide exposure, new parasite and pathogen infestations as well as beekeeper management and weather. Canadian beekeepers have indicated that issues with honey bee queens are the most significant factor affecting their colony health. In Canada, beekeepers manage colony losses by relying on the importation of foreign bees, particularly queens from warmer climates, to lead new replacement colonies. Unfortunately, the risks associated with imported queens include the introduction of new and potentially resistant pests and diseases, undesirable genetics including bees with limited adaptations to Canada’s unique climate and bees negatively affected by transportation. Importing a large proportion of our queens each year also creates an unsustainable dependency on foreign bee sources, putting our beekeeping and pollination sectors at an even greater risk in the case of border closures and restrictions. Increasing the domestic supply of queens is one mitigation strategy that could provide Canadian beekeepers, farmers and consumers with a greater level of agricultural stability through locally bred, healthier queens. Our study is the first rigorous analysis of the economic feasibility of Canadian queen production. We present the costs of queen production for three case study operations across Canada over two years as well as the profitability implications. Our results show that for a small to medium sized queen production operation in Canada, producing queen cells and mated queens can be profitable. Using a mated queen market price ranging from $30 to $50, a producer selling mated queens could earn a profit of between $2 and $40 per queen depending on price and the cost structure of his operation. If the producer chose to rear queens for his own operation, the cost savings would also be significant as imported queen prices continue to rise. Our case studies reveal that there is potential for both skilled labour acquisition over time in queen production as well as cost savings from economies of scale. Our queen producers also reduced their production costs by re-using materials year to year. Domestic queen production could be one viable strategy to help address the current pollinator crisis in Canada.

Simple Summary Behavioral characteristics of living organisms may affect the metabolism and its underlying molecular basis. The lifestyles of some modern communities include prolonged light exposure at night, and a high-fat/high-sugar-containing diet is frequently investigated. The molecular mechanisms of this unhealthy behavior might involve Adropin and some related nuclear receptors. This study examines the effect of long-term continuous light exposure and high fat/sucrose (HFS) diet on Adropin expression, RORα, Rev-erb-α nuclear receptors, key enzymes of lipid metabolism, and energy homeostasis in a rat model. The results of this study demonstrate the deleterious effects of this modern behavior on the metabolism, histology of many organs and general health. In conclusion CL and WD produced widespread derangements of energy homeostasis and lipid metabolism. The pathogenesis of this disturbance includes, at least in part, the Adropin hormone with the involvement of the RORα and Rev-erb-α nuclear receptors. Future therapeutic potential may involve Adropin. Abstract Long-term continuous light exposure (CL) and western diet (WD) effects on Adropin expression, RORα, and Rev-erb-α nuclear receptors and energy homeostasis were studied in rats. Thirty-two male Wistar rats (250–290 g) were enrolled for 3 months in the following groups (n = 8/group): (a) Normal control group (NC), (b) CL group, (c) WD group, and (d) CL + WD group. Then, indirect calorimetry and food intake (FI) were measured. Finally, Adropin, hormone-sensitive lipase (HSL), adipocyte triglyceride lipase (ATGL), and free fatty acids (FFA) were measured. Additionally, the histopathology and gene expression of Enho, RORα, and Rev-erb-α genes were done. The CL alone elevated the Adropin plasma level and gene expression, increased RORα expression, and decreased the Rev-erb-α nuclear receptor expression mainly in the liver and kidney. Besides, CL increased the total energy expenditure (TEE) and decreased the respiratory quotient. WD alone or in combination with the CL reversed gene expression of Enho, RORα, and Rev-erb-α. Combined CL and WD increased the TEE, reduced the food intake, increased the ATGL, and reduced the Adropin level in addition to widespread degenerative changes in the liver, spleen, and renal tissues. The deleterious effects of CL and WD on energy homeostasis may include Adropin with the involvement of the RORα and Rev-erb-α nuclear receptors.

This study assessed effects of irrigation water regimes and humic acid (HA) application on vegetative growth, yield, tuber quality and water use efficiency (WUE) of potato. Five irrigation treatments were applied at three developmental stages; (WR1) control (100 % crop evapotranspiration, 100 % ETc) at all plant growth, (WR2) 75 % ETc at all stages, (WR3) 75 % ETc at stage S1 (vegetative growth), (WR4) 75 % ETc at stage S2 (tuber initiation), and (WR5) 75 % ETc at stage S3 (tuber bulking). HA was applied at a rate of 1.5 g L−1 30 day after seed pieces planting. Plants experiencing water stress at S1 were shorter with fewer branches and lower fresh and dry vine weights. Water stress imposed at S2 significantly reduced tuber number, size, and yield. HA application increased vegetative growth, tuber weight, yield, WUE, and tuber quality (specific gravity and starch content). Thus, applying 1.5 g L−1 HA during vegetative growth and a 75 % ET water regime at S3 can increase potato production and tuber quality while reducing water use.

In the current paper, the effect of fear in three species Beddington–DeAngelis food chain model is investigated. A three species food chain model incorporating Beddington-DeAngelis functional response is proposed, where the growth rate in the first and second level decreases due to existence of predator in the upper level. The existence, uniqueness and boundedness of the solution of the model are studied. All the possible equilibrium points are determined. The local as well as global stability of the system are investigated. The persistence conditions of the system are established. The local bifurcation analysis of the system is carried out. Finally, numerical simulations are used to investigate the existence of chaos and understand the effect of varying the system parameters. It is observed that the existence of fear up to a critical value has a stabilizing effect on the system; otherwise it works as an extinction factor in the system.

Soil microorganisms might be assessed for their capabilities of plant growth promotion in order to identify heat tolerant strategies for crop production. The planned study was conducted to determine the potential of heat tolerant plant growth promoting rhizobacteria (PGPR) in mitigating heat stress effects in tomato. Bacillus cereus was evaluated for plant growth promoting activities and assessed for 1-aminocyclopropane-1-carboxylate (ACC-deaminase) (0.76&ndash, C0.9 &mu, M/mg protein/h), and exopolysaccharide (0.66&ndash, C0.91 mg/mL) under normal and heat stressed conditions. Plant growth regulators were evaluated through High Performance Liquid Chromatography. Bacterial inoculation effects on important physiological and biochemical parameters were evaluated under normal and heat stressed conditions in growth chamber. The morphological-physiological traits significantly revealed drastic effects on both of un-inoculated tomato varieties under heat stress conditions. Bacterial augmentation significantly promoted shoot, root length, leaf surface area, fresh and dry weight. Heat stress enhanced extracellular polymeric substances (EPS) production and cleavage of ACC into a-ketobutyrate and ammonia due to ACC-deaminase producing bacteria that significantly reduced the adverse effects of heat on tomato growth. In conclusion, the applied plant growth promoting rhizobacteria (PGPR) bacterial strain proved as potential candidate for improving tomato crop growing under heat stressed conditions. However, it is highly suggested to validate the current results by conducting field trials.

We studied the effects of two Salmonella enterica serovar Typhimurium (host-adapted) strains (14028 and 4/74) and three S. Choleraesuis (non-host-adapted) strains (A50, A45, and B195) in human monocytes between 2 and 24 h postinfection (p.i.) to investigate whether differences in immune response may explain the much higher prevalence of sepsis in individuals infected with S. Choleraesuis. Both serovars significantly increased the production of cytokines associated with acute sepsis (tumor necrosis factor alpha [TNF-α], interleukin β [IL-β], and IL-6), but temporal differences occurred between these serovars and between different S. Choleraesuis strains. Generally, all S. Choleraesuis strains induced significantly higher production of inflammatory cytokines than S. Typhimurium strains (P < 0.01 to 0.05). All S. Choleraesuis strains very significantly increased IL-10 production by monocytes at 6 and 24 h p.i. in comparison to S. Typhimurium strains (P < 0.01). In addition, ∼80% of monocytes were viable at 24 h p.i. with S. Choleraesuis A50, compared to only ∼40% following S. Typhimurium infection. Using S. Typhimurium 14028 and S. Choleraesuis A50 as examples of these two serovars, we also showed differential expression of genes within the Janus tyrosine kinase (JAK) and signal transducer and activator of transcription (STAT) (JAK/STAT) pathway via quantitative PCR (qPCR) microarray analysis. High serum IL-10 concentration and monocyte survival have been reported as markers of the development of human sepsis. We therefore conclude that high production of IL-10 by monocytes may, in part, explain the greater propensity for S. Choleraesuis to induce human sepsis and that this may be greater in strains such as A50, which induces both high IL-10 production and monocyte survival.

Murine/LPS models of Gram negative sepsis indicate that vasoactive intestinal peptide (VIP) has therapeutic potential. We investigated the unknown effect of VIP on JAK/STAT proteins and genes in human monocytes infected with Salmonella Typhimurium 14028. S. Typhimurium 14028 increased expression of both IL-6 receptor (IL-6R) and interferon gamma receptor 1 (IFNγR1) on monocytes but co-culture of infected monocytes with VIP (10-7 M) only decreased expression of IFNγR1 (P < 0.05). In contrast, S. Typhimurium 14028 infection or co-culture with VIP had no effect on IL-10 receptor expression on the monocyte surface. However, S. Typhimurium 14028 down regulated IFNGR1 gene expression and this was not altered by co-culture with VIP, suggesting that changes in IFNγR1 protein may be due to an effect on cytoplasmic transport. 15 JAK/STAT genes, out of 84 studied, were up-regulated by S. Typhimurium 14028 infection and five were down-regulated. Co-culture with VIP significantly decreased expression of two genes (IFNG and IL-20) and increased expression of three genes (SOCS1, SOCS3 and STAT4) (P < 0.05). S. Typhimurium 14028 also increased expression of PTPN1, which dephosphorylates JAK2 and TYK2. This was unaltered by co-culture with VIP but S. Typhimurium 14028-induced expression of ISG15, associated with susceptibility to Gram negative infection, was further increased by VIP. We conclude that the effect of VIP on JAK/STAT genes may preclude its therapeutic use in human Gram negative sepsis.

We present a novel way to select for highly polygenic traits. For millennia, humans have used observable phenotypes to selectively breed stronger or more productive livestock and crops. Selection on genotype, using single-nucleotide polymorphisms (SNPs) and genome profiling, is also now applied broadly in livestock breeding programs; however, selection on protein/peptide or mRNA expression markers has not yet been proven useful. Here we demonstrate the utility of protein markers to select for disease-resistant hygienic behavior in the European honey bee (Apis mellifera L.). Robust, mechanistically-linked protein expression markers, by integrating cis- and trans- effects from many genomic loci, may overcome limitations of genomic markers to allow for selection. After three generations of selection, the resulting marker-selected stock outperformed an unselected benchmark stock in terms of hygienic behavior, and had improved survival when challenged with a bacterial disease or a parasitic mite, similar to bees selected using a phenotype–based assessment for this trait. This is the first demonstration of the efficacy of protein markers for industrial selective breeding in any agricultural species, plant or animal.

Fumagillin is a potent fungal metabolite first isolated from Aspergillus fumigatus. It is widely used in apiculture and human medicine against a variety of microsporidian fungal infections. It has been the subject of research in cancer treatments by employing its angiogenesis inhibitory properties. The toxicity of fumagillin has limited its use for human applications and spurred the development of analogues using structure-activity relationships relating to its angiogenesis properties. These discoveries may hold the key to the development of alternative chemical treatments for use in apiculture. The toxicity of fumagillin to humans is important for beekeeping, because any residues remaining in hive products pose a direct risk to the consumer. The analytical methods published to date measure fumagillin and its decomposition products but overlook the dicyclohexylamine counterion of the salt form widely used in apiculture.

This also represents a completely novel way to select for highly polygenic traits.

International audience; AbstractFumagilin-B® is the only currently registered chemical treatment available to combat nosema disease in apiculture. Fumagillol, the basic hydrolysis product of fumagillin, two semisynthetic fumagillin analogues, and four in-house purely synthetic compounds which were designed to mimic the mode of action of fumagillin against the methionine aminopeptidase type 2 (MetAP-2) enzyme, was observed to exhibit statistically significant biological activity against Nosema ceranae-infected caged bees. None of these compounds were, however, as effective as Fumagilin-B®. The commercially available thymol and enilconazole also exhibited activity against N. ceranae, with thymol being the most promising chemical treatment other than Fumagilin-B®. High cumulative bee mortality was associated with the therapeutic dosage of Fumagilin-B® during our study, suggesting the need for continued investigation.

International audience; AbstractBoth commercially available fumagillin-based treatments for honey bees (Apis mellifera), Fumagilin-B® as well as Fumidil-B®, contain the reportedly genotoxic and tumorigenic compound dicyclohexylamine (DCH) as the counter ion in the fumagillin-DCH salt. The effect of DCH, purified fumagillin (containing no DCH), and the commercial formulation Fumagilin-B® (containing both fumagillin as well as DCH) on the mortality of caged Nosema ceranae-infected honey bees was investigated. A statistically significant risk of bee mortality associated with oral exposure to DCH was observed. DCH is also known to be significantly more stable than fumagillin in honey under a variety of temperature conditions, both in the presence and absence of light. The presence of DCH in the hive is therefore a potential concern for bee health and also for food safety.

Nous avons etudie le mecanisme selon lequel des abeilles (Apis mellifera), selectionnees pour le caractere « suppression de la reproduction des acariens » (abeilles SMR), pouvaient agir negativement sur le succes reproductif (S.R.) de l'acarien Varroa destructor, notre hypothese etant que les abeilles SMR detectent et eliminent les nymphes qui ont des acariens reproducteurs, donnant ainsi l'impression que les acariens restant sur les nymphes au stade preeclosion se reproduisent mal. Nous avons utilise la fertilite des acariens et le nombre de descendants femelles viables comme mesure du S.R. Nous avons aussi etudie s'il existe un facteur physiologique associe au couvain d'ouvrieres SMR qui reduit le S.R. des acariens. Nous avons compare des colonies selectionnees pour le caractere SMR, fournies par Harbo, avec des colonies selectionnees poule comportement hygienique (abeilles HYG) elevees a l'universite du Minnesota. Pour tester si les abeilles SMR pouvaient detecter et eliminer les nymphes infestees, nous avons introduit des acariens des colonies sources SMR et HYG dans du couvain d'ouvrieres SMR et HYG recemment opercule. Au 10 e jour apres l'introduction des acariens, les trois colonies SMR avaient elimine significativement plus de nymphes infestees que les trois colonies HYG, quelle que fut la source des acariens (P < 0,0001) (Fig. 1). Ensuite nous avons releve le S.R. des acariens sur les nymphes restantes (Tab. I). Les acariens dans les colonies SMR etaient significativement moins fertiles (moins d'acariens ont pondu au moins un oeuf) que ceux des colonies HYG (P = 0,014). Ils ont aussi produit significativement moins de descendantes viables (definies par la presence d'un acarien adulte fille et d'un acarien adulte mâle sur une nymphe au stade du bourrelet alaire gris) que ceux des colonies HYG (P = 0,030). Pour etudier l'action du couvain SMR sur le S.R. des acariens, nous avons place du couvain infeste dans une etuve reglee a la temperature et a l'hygrometrie de la ruche (Tab. II). Les acariens etaient significativement moins fertiles (P = 0,033) et ont produit significativement moins de descendantes viables (P = 0,026) sur le couvain SMR que sur le couvain HYG. Les resultats confirment donc notre hypothese: les abeilles SMR detectent, desoperculent et eliminent significativement plus de nymphes infestees que les abeilles HYG. Il ne semble pas que les abeilles SMR eliminent selectivement les nymphes avec acariens a S.R. eleve et laissent les nymphes avec acariens a S.R. faible, parce que l'etude en etuve a montre que tous les acariens avaient un S.R. plus faible sur le couvain SMR que sur le couvain HYG. En consequence la combinaison des abeilles SMR, qui eliminent une forte proportion d'abeilles infestees, et le faible succes reproductif des acariens sur le couvain SMR en general pourraient expliquer le faible S.R. des acariens restants. D'autres experiences sont necessaires pour determiner l'action physiologique du couvain SMR sur le succes reproductif des acariens et savoir si les caracteres HYG et SMR sont regules par les memes genes.

We compared colonies selectively bred for both hygienic behavior and Suppression of Mite Reproduction (HYG/SMR) with colonies bred solely for hygienic behavior (HYG) and unselected control colonies. Colonies were evaluated for strength, brood viability, removal of freeze-killed brood, honey production, mite loads on adult bees and within worker brood, and mite reproductive success on worker brood for two years in two locations. By autumn in both years, the HYG/SMR colonies had significantly fewer mites on adult bees and in worker brood compared to the control colonies, and the HYG colonies had intermediate mite populations. Contrary to expectation, there were no differences among the lines in mite reproductive success. Further studies are required to determine if the genes and neural mechanisms that regulate the SMR trait are the same or different from those regulating hygienic behavior.

Vasoactive intestinal peptide (VIP) is a pleiotropic, highly conserved, peptide found in many different biological systems throughout invertebrate phyla. VIP is produced by cells of the immune system but also inhibits many different inflammatory products produced by these immune cells, including cytokines and chemokines. VIP inhibits these immune mediators by affecting transcriptional regulators such as NFκB and activator protein 1 which transcribes genes responsible for the production of inflammatory mediators in response to pathogens or cytokines. In this review, the therapeutic potential of VIP will be discussed in the context of transcriptional regulation of immune cells in in vitro and in vivo animal models.
