International audience; Spicules are mineral-based biocomposites skeletal structures that are widely distributed among phylogenetically distant groups of invertebrates (Porifera, Cnidaria, Mollusca, Echinodermata). Subepidermal spicules are formed under the ectodermal epithelium and are characterized for all groups except mollusks (Aplacophora, Polyplacophora, Bivalvia), their spicules are located on the surface of the body. However, one group of mollusks (Gastropoda: Heterobranchia) have unique subepidermal spicules that have never been detected above the ectodermal epithelium and similarly to those characterized for Porifera, Cnidaria and Echinodermata. Understanding subepidermal spicule formation in mollusks could help solve the question on the origin of spicules. Spicules in nudibranchs have been described for more than 150 years, yet ontogenetic dynamics of spicules have never been studied and the full mechanism of their formation remains unknown. Herein we investigate the spicule formation in different stages of postlarval development of the nudibranch Onchidoris muricata (O.F. Müller, 1776). For the first time, ontogenetic transformations of the spicule complex are described using experiments and different morphological methods. Our studies demonstrate that spicules of O. muricata form in the subepidermal space in early developmental stages immediately after veliger settlement. A single spicule forms inside a huge vacuole within a sclerocyte and remains there throughout the entire life of the specimen. Signs of spicule or sclerocyte migration under the epithelium in postlarval development was not found. Spicules only form during larval settlement, increasing only in size as development furthers. For the first time, spicule mineralization zones were detected at the tips of the spicules as well as the presence of collagen I in the overall composition of the spicules. Thus, our findings suggest that spicules form by an ectodermal cell that emerged under the ectodermal epithelium during the earliest stages of postlarval development.

The head anatomy ofPselaphus heisei(Pselaphitae) is described and documented. The structural features are evaluated in comparison with findings presented in earlier studies on the subfamily, with a special focus on correlations with predacious habits and the groundplan of Pselaphinae. We found the tentorium, labrum, maxillary palps, shape of head, and a system of dorsal pits and sulci highly variable within the subfamily, reflecting multiple transformations, including many homoplasious changes. The following major characters are identified as groundplan features of Pselaphinae: falciform mandibles; small mola; semiglobular neck; ventrolateral antennal articulation; steep clypeal region; setiform labial palpomere 3; tentorium with nearly vertical main branches and lacking laminatentoria; separation of tentorial bridge from tentorial arms; fusion of dorsal tentorial arms with the head capsule; large brain placed in the posterior third of the head; and a triple cluster of well-developed cephalic glands. The last feature supports a hypothesis that multiple and independent cases of adaptations to myrmecophilous habits observed in various lineages of Pselaphinae were possible by re-programming already existing glands to produce appeasement secretions. The cephalic muscle apparatus ofP. heiseiis similar to what is found in other staphylinoid groups, with some exceptions, whereas it is strongly modified in the myrmecophileClaviger testaceus. We propose that the unparalleled structural megadiversity in Pselaphinae is primarily linked with life in the upper soil layers combined with specialized carnivorous habits, with small and agile or mechanically protected arthropods as prey. Within the group, various specialized life habits have evolved, including myrmecophily, termitophily, and also life in deep soil or caves, each with unique morphological adaptations.

This data article presents datasets associated with the research article entitled “The immunological architecture of granulomatous inflammation in central nervous system tuberculosis’’ (Zaharie et al., 2020). The morphology of tuberculosis related granulomas within the central nervous system of human patients was visualized in six different three-dimensional (3D) models. Post-mortem, formalin fixed and paraffin embedded specimens from deceased tuberculous meningitis patients were immunohistochemically stained and 800 serial histologically stained sections were acquired. Images from all sections were obtained with an Olympus BX43 light microscope and structures were identified, labeled and made three-dimensional. The interactive 3D-models allows the user to directly visualize the morphology of the granulomas and to understand the localization of the granulomas. The 3D-models can be used for multiple purposes and provide both an educational source as a gold standard for further animal studies, human research and the development of in silico models on the topic of central nervous system tuberculosis.

The investigation of the type series of Oulema septentrionis (Weise, 1880) and Oulema erichsonii (Suffrian, 1841) using Micro-computed X-ray tomography (µCT) revealed that neither species is a melanic variant of Oulema melanopus (Linnaeus, 1758) as has been suggested previously. Lectotypes of Oulema septentrionis (Weise, 1880) and Oulema erichsonii (Suffrian, 1841) are designated based on the study of type material. The male genitalia of O. septentrionis and O. erichsonii differ to an extent in the shape of the median lobe and flagellum that their status as separate species is – cautiously – confirmed by the present study.

Coscinodiscus genus, a type of diatoms with complex frustule, has been widely studied and reported because of their delicate nanostructures. In this paper, a dual beam system focused ion beam and scanning electron microscope (FIB-SEM) were applied to precisely investigate the microstructure of diatom cell walls, in particular, aiming to reveal the hierarchical pores of the valve. The microstructures of the valve, valve mantle and girdle bands were illustrated in details through a series of high resolution images after performing the cross section milling of the specific region. The 3D morphology of frustule valve was reconstructed based on the 2D image series, which demonstrated the presence of cribellum in valve, the external foramen, hexagonal cavity and the internal foramen. The four dense porous membranes and girdle bands were characterized clearly based on FIB-SEM dual beam system. An integrated model of Coscinodiscus frustule could be simulated based on the 2D and 3D results. This study provided a systematic approach to measure the morphological features of diatoms at a nanoscale, which could be applied to other nanoporous structure in three dimensions. © 2017 Chinese Materials Research Society. Leiodidae are the second largest subterranean radiation of beetles at family rank. To explore morphological trends linked with troglobiontic habits and characters with potential phylogenetic significance, the head of the cave-dwelling species Troglocharinus ferreri (Cholevinae, Leptodirini) was examined in detail. Overall, the general pattern is similar to what is found in Catops ventricosus (Cholevini). Shared apomorphic features include a fully exposed anterolateral concavity containing the antennal socket, a distinct bead above this depression, a bilobed lip-like structure anterad the labrum, a flat elevated portion of the ventral mandibular surface, and a ventral process at the proximomesal edge of this mandibular area. The tentorial structures are well-developed as in C. ventricosus, with a large laminatentorium and somewhat shortened dorsal arms. The mouthparts are largely unmodified, with the exception of unusually well-developed extrinsic maxillary muscles. Features of T. ferreri obviously linked with subterranean habits are the complete lack of compound eyes, circumocular ridges, and optic lobes. A series of characters is similar to conditions found in other genera of Leptodirini: the head capsule completely lacks a protruding ocular region, a distinct neck is missing, the transverse occipital crest is indistinct, and the antennae are elongate and lack a distinct club. Two different trends of cephalic transformations occur in troglobiontic Leptodirini, with some genera like Troglocharinus and Leptodirus having elongated head capsules and antennae, and others having broadened, more transverse heads. In contrast, the modifications are more uniform in the closely related Ptomaphagini, with a pattern distinctly differing from Leptodirini: the head is transverse, with a distinctly protruding ocular region, a distinct transverse occipital crest, and a very narrow neck region., The first author wants to express thanks to CSC (No. 201708440281). The presence of a swinging tentorium is a key apomorphy of Myriapoda, but this character has been studied in detail in only few species. Here the tentorium, i.e., the peristomatic skeleton of the preoral chamber, is comparatively studied in three species of the millipede order Sphaerotheriida Brandt, 1833. Since dissections of the fragile tentorial components proved to be difficult, despite the large head size, they were analysed mainly in situ via micro-computed tomography. Our results confirm previous observations of large differences in the tentorial construction in the giant pill-millipedes compared to chilognathan diplopods. The tentorium of Sphaerotheriida consists of a curved, plate-like epipharyngeal bar with distal projections, an elongate and thin hypopharyngeal bar, and a plate-like triangular posterior process; a transverse bar is absent. Only seven muscles attach at the tentorium in giant pill-millipedes, including two antennal muscles and two muscles of the gnathochilarium. Within the order Sphaerotheriida, the composition of the tentorium and its muscular equipment seems to be conserved, except for some variability in the shape of the epipharyngeal bar. As the transverse bar has been considered essential for the mobility of the tentorium in myriapods, its absence in Sphaerotheriida may indicate that their tentorium is not capable of performing a swing. Loss of tentorial mobility may also pertain to the order Glomerida Brandt, 1833, inferred here from the absence of a posterior process. An apparently immobile tentorium in Glomerida and Sphaerotheriida can straightforwardly be correlated with transformations of the head related to their ability of volvation. The different transformations of the tentorium, here hypothesised to cause immobility, may support current assumptions that the ability of volvation evolved convergently in Glomerida and Sphaerotheriida. This conclusion, however, still requires more detailed studies of the head anatomy in Glomerida and Glomeridesmida Cook, 1895.

Organogenesis is a complex coordinated process of cell proliferation, growth, migration, and apoptosis. Differential growth rates, particularly during cardiogenesis, play a role in establishing morphology. Studies using stereological and cell sorting methods derive averages of morphogenetic parameters for an organ. To understand tissue composition and differential growth, the researcher must determine a number of morphogenetic parameters in the developing organ. Such measurements require sectioning to enable identification of organ borders, tissue components and cell types, three‐dimensional (3D)‐reconstruction of sections to visualize morphology and a 3D‐measurement scheme to build local morphogenetic information. Although thick the section confocal microscopy partially solves these issues, information loss at the section surface hampers the reconstruction of 3D morphology. Episcopic imaging provides the correct morphology but lacks histological procedures to identify multiple cell types. The 3D‐measurement scheme is based on systematic sampling, with overlapping sample volumes, of the entire organ in the aligned image stack. For each sample volume, morphogenetic variables are calculated and results projected back to the cube (boxel) at the sample volume center. Boxel size determines spatial resolution of the final quantitative 3D‐reconstruction whereas size of the sample volume determines the precision of the morphogenetic information. The methods described here can be used to measure tissue volume, proliferation and cell size, to determine contribution and distribution of cell types in a tissue and to display this information in a quantitative 3D‐reconstruction. The thoracic morphology of the troglobiontic leiodid species Troglocharinus ferreri (Cholevinae, Leptodirini) is described and documented in detail. The features are mainly discussed with respect to modifications linked with subterranean habits. Troglocharinus is assigned to the moderately modified pholeuonoid morphotype. The body is elongated and slender compared to epigean leiodids and also cave-dwelling species of Ptomaphagini. The legs are elongated, especially the hindlegs, though to a lesser degree than in the most advanced troglobiontic species. The prothorax is moderately elongated but otherwise largely unmodified. Its muscular system is strongly developed, with more muscle bundles that in free-living staphylinoid or hydrophiloid species. The pterothorax is greatly modified, especially the metathoracic flight apparatus. The meso- and metathoracic elements of the elytral locking device are well-developed, whereas the other notal parts are largely reduced. The mesonotum is simplified, with the triangular scutellar shield as the only distinctly developed part. The mesothoracic musculature is strongly reduced, with only 6 muscles compared to 12 or 13 in free-living staphylinoid or hydrophiloid species. The metanotum is greatly reduced, without a recognizable subdivision into prescutum scutum and scutellum. It is strongly narrowing laterally and lacks notal wing processes and other wing-related elements, but well-developed alacristae are present. The wings are reduced to small membranous flap-like structures inserted at the posterior end of the metanotum. A metapostnotum is not developed. Like in the case of the head, cave dwelling species of the related Ptomaphagini and Leptodirini show different trends of adaptations, with a compact ovoid or navicular body shape in the former, and a distinct trend towards elongation of the body and appendages in the latter tribe. Structural affinities of the thoraces of T. ferreri and the troglobiontic trechine carabid Sinaphaenops wangorum are mainly due to the reduced flight apparatus. Adult head structures are well known in the coleopteran suborders Archostemata and Adephaga, whereas the available information is very fragmentary in the megadiverse Polyphaga, including the successful superfamily Staphylinoidea. In the present study, the cephalic morphology of the cholevine species Catops ventricosus is described in detail and documented. The results were compared to conditions occurring in other polyphagan lineages, especially staphylinoid and scarabaeoid representatives. Specific external features documented in Catops and potential autapomorphies of Leiodidae include a five-segmented antennal club with a reduced eighth antennomere and the presence of periarticular grooves filled with sensilla on antennomeres 7, 9, and 10. The firm connection of the head and pronotum is possibly an apomorphy of Cholevinae. The monophyly of Cholevinae excluding Eucatopini and Oritocatopini is supported by the apical maxillary palpomere as long as or shorter than the subapical one, and the presence of cryptic pore plates on the surface of these palpomeres—a feature described and documented here for the first time. The internal cephalic structures of Catops are mostly plesiomorphic, as for instance the complete tentorium. The pattern of the muscles is similar to what is found in other staphylinoid taxa. The unusual maxillary muscle “Mx” is likely a groundplan apomorphy of the clade Staphyliniformia + Scarabaeoidea. M. hypopharyngomandibularis (M13) was identified in Catops and is ancestral for Coleoptera, even though it is often missing. A distinctive feature of embryonic tendon development is the steady increase in collagen fibril diameter and associated improvement of tissue mechanical properties. A potential mechanical stimulus for these changes is slow stretching of the tendon during limb growth. Testing this hypothesis in vivo is complicated by the presence of other developmental processes including muscle development and innervation. Here we used a cell culture tendon-like construct to determine if slow stretch can explain the increases in fibril diameter and mechanical properties that are observed in vivo. Non-stretched constructs had an ultrastructural appearance and mechanical properties similar to those of early embryonic tendon. However, slowly stretching during 4 days in culture increased collagen fibril diameter, fibril packing volume, and mechanical stiffness, and thereby mimicked embryonic development. 3D EM showed cells with improved longitudinal alignment and elongated nuclei, which raises the hypothesis that nuclear deformation could be a novel mechanism during tendon development. The fine analysis of synaptic contacts is usually performed using transmission electron microscopy (TEM) and its combination with neuronal labeling techniques. However, the complex 3D architecture of neuronal samples calls for their reconstruction from serial sections. Here we show that focused ion beam/scanning electron microscopy (FIB/SEM) allows efficient, complete, and automatic 3D reconstruction of identified dendrites, including their spines and synapses, from GFP/DAB-labeled neurons, with a resolution comparable to that of TEM. We applied this technology to analyze the synaptogenesis of labeled adult-generated granule cells (GCs) in mice. 3D reconstruction of dendritic spines in GCs aged 3–4 and 8–9 weeks revealed two different stages of dendritic spine development and unexpected features of synapse formation, including vacant and branched dendritic spines and presynaptic terminals establishing synapses with up to 10 dendritic spines. Given the reliability, efficiency, and high resolution of FIB/SEM technology and the wide use of DAB in conventional EM, we consider FIB/SEM fundamental for the detailed characterization of identified synaptic contacts in neurons in a high-throughput manner.

Here, we describe a robust method for mapping the number and type of neuro-chemically distinct synaptic inputs that a single reconstructed neuron receives. We have used individual hypoglossal motor neurons filled with Neurobiotin by semi-loose seal electroporation in thick brainstem slices. These filled motor neurons were then processed for excitatory and inhibitory synaptic inputs, using immunohistochemical-labeling procedures. For excitatory synapses, we used anti-VGLUT2 to locate glutamatergic pre-synaptic terminals and anti-PSD-95 to locate post-synaptic specializations on and within the surface of these filled motor neurons. For inhibitory synapses, we used anti-VGAT to locate GABAergic pre-synaptic terminals and anti-GABA-A receptor subunit α1 to locate the post-synaptic domain. The Neurobiotin-filled and immuno-labeled motor neuron was then processed for optical sectioning using confocal microscopy. The morphology of the motor neuron including its dendritic tree and the distribution of excitatory and inhibitory synapses were then determined by three-dimensional reconstruction using IMARIS software (Bitplane). Using surface rendering, fluorescence thresholding, and masking of unwanted immuno-labeling, tools found in IMARIS, we were able to obtain an accurate 3D structure of an individual neuron including the number and location of its glutamatergic and GABAergic synaptic inputs. The power of this method allows for a rapid morphological confirmation of the post-synaptic responses recorded by patch-clamp prior to Neurobiotin filling. Finally, we show that this method can be adapted to super-resolution microscopy techniques, which will enhance its applicability to the study of neural circuits at the level of synapses.

The dendritic spines on pyramidal cells represent the main postsynaptic elements of cortical excitatory synapses and they are fundamental structures in memory, learning and cognition. In the present study, we used intracellular injections of Lucifer yellow in fixed tissue to analyse over 19 500 dendritic spines that were completely reconstructed in three dimensions along the length of the basal dendrites of pyramidal neurons in the parahippocampal cortex and CA1 of patients with Alzheimer's disease. Following intracellular injection, sections were immunostained for anti-Lucifer yellow and with tau monoclonal antibodies AT8 and PHF-1, which recognize tau phosphorylated at Ser202/Thr205 and at Ser396/404, respectively. We observed that the diffuse accumulation of phospho-tau in a putative pre-tangle state did not induce changes in the dendrites of pyramidal neurons, whereas the presence of tau aggregates forming intraneuronal neurofibrillary tangles was associated with progressive alteration of dendritic spines (loss of dendritic spines and changes in their morphology) and dendrite atrophy, depending on the degree of tangle development. Introduction Traditionally, the origin of the third germ layer and its special formation of coelomic cavities by enterocoely is regarded to be an informative character in phylogenetic analyses. In early deuterostomes such as sea urchins, the mesoderm forms through a single evagination pinching off from the apical end of the archenteron which then gives off mesocoela and metacoela on each side. This echinoid-type coelom formation has conventionally been assumed to be ancestral for Deuterostomia. However, recent phylogenetic analyses show that Echinodermata hold a more derived position within Deuterostomia. In this regard a subgroup of Hemichordata, namely enteropneusts, seem to host promising candidates, because they are supposed to have retained many ancestral deuterostome features on the one hand, and furthermore share some characteristics with chordates on the other hand. In enteropneusts a wide range of different modes of coelom formation has been reported and in many cases authors of the original observations carefully detailed the limitations of their descriptions, while these doubts disappeared in subsequent reviews. In the present study, we investigated the development of all tissues in an enteropneust, Saccoglossus kowalevskii by using modern morphological techniques such as complete serial sectioning for LM and TEM, and 3D-reconstructions, in order to contribute new data to the elucidation of deuterostome evolution. Results Our data show that in the enteropneust S. kowalevskii all main coelomic cavities (single protocoel, paired mesocoela and metacoela) derive from the endoderm via enterocoely as separate evaginations, in contrast to the aforementioned echinoid-type. The anlagen of the first pair of gill slits emerge at the late kink stage (~96 h pf). From that time onwards, we documented a temporal left-first development of the gill slits and skeletal gill rods in S. kowalevskii until the 2 gill slit juvenile stage. Conclusions The condition of coelom formation from separate evaginations is recapitulated in the larva of amphioxus and can be observed in crinoid echinoderms in a similar way. Therefore, coelom formation from separated pouches, rather than from a single apical pouch with eventual subdivision is suggested as the ancestral type of coelom formation for Deuterostomia. Left-right asymmetries are also present in echinoderms (rudiment formation), cephalochordates (larval development), tunicates (gut coiling) and vertebrates (visceral organs), and it is known from other studies applying molecular genetic analyses that genes such as nodal, lefty and pitx are involved during development. We discuss our findings in S. kowalevskii in the light of morphological as well as molecular genetic data.

13 páginas, 7 figuras, 1 tabla -- PAGS nros. 456-468, Endoglin is a type I membrane protein expressed as a disulphide-linked homodimer on human vascular endothelial cells whose haploinsufficiency is responsible for the dominant vascular dysplasia known as hereditary hemorrhagic telangiectasia (HHT). Structurally, endoglin belongs to the zona pellucida (ZP) family of proteins that share a ZP domain of ∼ 260 amino acid residues at their extracellular region. Endoglin is a component of the TGF-β receptor complex, interacts with the TGF-β signalling receptors types I and II, and modulates cellular responses to TGF-β. Here, we have determined for the first time the three-dimensional structure of the ∼ 140 kDa extracellular domain of endoglin at 25 Å resolution, using single-particle electron microscopy (EM). This reconstruction provides the general architecture of endoglin, which arranges as a dome made of antiparallel oriented monomers enclosing a cavity at one end. A high-resolution structure of endoglin has also been modelled de novo and found to be consistent with the experimental reconstruction. Each subunit comprises three well-defined domains, two of them corresponding to ZP regions, organised into an open U-shaped monomer. This domain arrangement was found to closely resemble the overall structure derived experimentally and the three modelled de novo domains were tentatively assigned to the domains observed in the EM reconstruction. This molecular model was further tested by tagging endoglin's C terminus with an IgG Fc fragment visible after 3D reconstruction of the labelled protein. The laser scanning confocal microscope (LSCM) is an extremely useful tool that allows fluorescently labelled cells to be visualized in whole-mount preparations. This is particularly advantageous, for example, in studying the dendritic trees of neurons with respect to their environment. One of the most popular, and easiest, ways to visualize a cell is to inject it intracellularly with the fluorophore Lucifer Yellow (LY). However, the argon gas lasers of most LSCM's are not well matched to the excitation spectrum of aqueous LY. When this largely inappropriate excitation is combined with standard filters, designed for fluorescein fluorescence rather than Lucifer Yellow, the resulting image is poor. We report that clearing LY-injected neurons in methyl salicylate and mounting them in Entellan, a non-aqueous medium of high refractive index, enhances their visualization on a Bio-Rad LSCM with standard fluorescein (FITC) filters to an unexpected degree. This technique also leads to a substantial reduction in photobleaching.

Limbed vertebrates have functionally integrated postcranial axial and appendicular systems derived from two distinct populations of embryonic mesoderm. The axial skeletal elements arise from the paraxial somites, the appendicular skeleton and sternum arise from the somatic lateral plate mesoderm, and all of the muscles for both systems arise from the somites. Recent studies in amniotes demonstrate that the scapula has a mixed mesodermal origin. Here we determine the relative contribution of somitic and lateral plate mesoderm to the avian scapula from quail-chick chimeras. We generate 3D reconstructions of the grafted tissue in the host revealing a very different distribution of somitic cells in the scapula than previously reported. This novel 3D visualization of the cryptic border between somitic and lateral plate populations reveals the dynamics of musculoskeletal morphogenesis and demonstrates the importance of 3D visualization of chimera data. Reconstructions of chimeras make clear three significant contrasts with existing models of scapular development. First, the majority of the avian scapula is lateral plate derived and the somitic contribution to the scapular blade is significantly smaller than in previous models. Second, the segmentation of the somitic component of the blade is partially lost; and third, there are striking differences in growth rates between different tissues derived from the same somites that contribute to the structures of the cervical thoracic transition, including the scapula. These data call for the reassessment of theories on the development, homology, and evolution of the vertebrate scapula.
