Search

Governance and regulation of the use of live animals in research and teaching is examined in Australia, Malaysia, Indonesia, the Philippines, Singapore, Thailand, China, Japan and India. A comparison of the systems in different countries will enable the determination of best practice and fit-forpurpose regulation. The most comprehensive government regulation of animal welfare in institutions covers a broad range of animals and institutions are required to have an Institutional Animal Care and Use Committee, whose membership is specified in guidelines or regulations. The work of the Committees is rigorously overseen by government and facilities that use live animals are audited externally. All countries examined have legislation governing the use of live animals in research, although only Australia and Malaysia have a fully equivalent mandated oversight of teaching. Teaching that uses live animals is partly covered in the Philippines, Japan, Singapore and Thailand This paper thus aims to review the regulation of animal use in different Asian jurisdictions in order to determine best practices that are appropriate to those settings. The most comprehensive oversight is provided in Australia and Malaysia that essentially use the same regulatory framework.

Prizes and awards have received limited attention in scholarly research. The present paper engages with the prestigious "UNESCO-Japan Prize on Education for Sustainable Development," which annually honours three outstanding projects related to education for sustainable development (ESD). Drawing on biopolitical theory, the paper explores what this prize can tell us about the global community's quest for sustainable development. While the award is global in scope and alludes to humanity's joint responsibility for the planet, the analysis shows that the winning organisations approach rich and poor populations around the world in entirely different ways; assigning to them different roles, responsibilities and lifestyles. ESD discourse thus displays a remarkable ability to accommodate the lifestyle divide that separates rich and poor; suggesting that inequality has become effectively normalised. Ultimately it is argued that differentiating practices in the global implementation of ESD constitutes an urgent area of further research for comparative and international education.

The method for extracting and separating hydrophobic photosynthetic pigments proposed by Katayama "et al." ("Japanese Journal of Phycology," 42, 71-77, 1994) has been improved to introduce it to student laboratories at the senior high school level. Silica gel powder was used for removing water from fresh materials prior to extracting pigments by a mixture of organic solvents that was also used for chromatographic separation of the pigments. A small silica gel thin-layer plate or a paper strip was used for separating the pigments. The improved method may be applicable to all kinds of plant materials including algae, is easier than most other methods, and can lead to more successful results in separating these pigments by both thin-layer chromatography and paper chromatography. The method has been carried out in student laboratories in some senior high schools and universities in Japan. The results indicate that this laboratory exercise is effective for students to recognise the unity and diversity of plants. Therefore, this laboratory seems to be useful for teaching plant systematics as well as for teaching photosynthesis. (Contains 1 figure and 1 online reference.)

This review examines the effects of nationality on attitudes of students to study science. It compares data from many countries published in refereed research journals, research reports, book chapters, and conference papers. The main aim is to identify the commonalities and variations in the students' attitudes with particular focus on students from developing and developed countries. The search of bibliographical databases yielded a lot of researches. After title and abstract screening and after eliminating duplicates and applying inclusion criteria, this search ended up to include 15. Findings make it clear that students in different parts of the world develop differing attitudes toward science. It is observed that students are positive internationally; however, students in developing countries are more positive than the students in rich and developed countries, especially in Japan and Korea. Biology is most favourite, particularly of girls, whereas boys favour physics. It is also felt that comparing available attitude data is not simple and must be done with caution. This paper describes how the patriarchal structure of Japanese society and its notions of women, femininity, and gendered stereotypes produced strong cultural barriers to increasing the participation of females in science education. Baseline data on attitudes toward science and the perceptions of gender issues in science education, academic major and career choice were collected from 175 university students (124 female, 51 male). Students responded to a Likert scale that included the option "I don't understand the question". All respondents took advantage of the option for items related to gender issues. On some items up to 67% of the males responded that they did not understand the question. Females in science choosing this option did not exceed 19%. In Japan, gender is an invisible, pervasive construct that impacts females' participation in science and science education. In other ways, attitudes toward science among Japanese students mirrored those found in the United States and in other countries. Respondents held the most favorable views of science when they were in elementary school and females preferred biology while males preferred the physical sciences. The exception to the Western pattern of liking science and science teachers is that male non-science majors rather than female non-science majors reported poor academic performance in elementary school, declining attitudes in middle school, and they held the most negative attitudes toward their science teacher and science subjects.

Plant Species Biology will publish a continuing series of papers on the biology and life history of vascular plant species indigenous to or well naturalized in the flora of Japan. This paper sets forth the guidelines and format for contributors to this series. This new series attempts to establish a record as complete as possible concerning all aspects of the life history characteristics as well as significant biosystematic characters of the species under consideration, and keen attention will be given to the species which are becoming very rare and endangered, especially in the lowland areas and aquatic habitats (e.g., rivers and ponds). This paper explores the eugenic thought of Yamanouchi Shigeo (1876-1973), who was trained in plant cytology under the tutelage of botanist and eugenicist John Coulter (1851-1928) in the USA, and later became one of the early and important popularizers of eugenic ideas in Japan. His career demonstrates a direct !ink between Japanese and US eugenics. Despite his academic training and research) at various internationally renowned institutions, numerous publications, and longevity, his life has received little scholarly attention. By the early twentieth century. most biologists in Japan, as in the USA. began accepting Mendelian evolutionary theory and rejecting the Lamarckian notion of inheritance of acquired characteristics. However. Yamanouchi Shigeo's eugenic view represents a paradox: he was a Mendelian cytologist sympathetic to Lamarckism. Was his 'nurture'-oriented eugenic view unscientific? Is that why he was largely ignored in the history of botany in Japan? This study attempts to answer these questions and to analyse the origins and distinct features of Yamanouchi's eugenic ideas by situating Yamanouchi's eugenic thought historically and culturally. After examining his scientific papers, popular writings, and documents of various organizations to which he belonged. I argue that Yamanouchi's 'softer' (or less biologically deterministic) perspective may have reflected the Japanese desire to catch up with the dominant 'race' by using eugenics without accepting permanent inferior status. In this paper, I revise the taxonomy of the lace bug genus Agramma Stephens, 1829 (Hemiptera: Heteroptera: Tingidae: Tinginae) from Japan. Two species are recognized and re-described: A. japonicum (Drake, 1948) described previously from Japan, and A. abruptifrons Golub, 1990, newly recorded from Japan. The previous records of A. nexile (Drake, 1948) from Japan are confirmed as misidentifications of A. japonicum. The host plant relationship is discussed for A. abruptifrons and A. japonicum. Photographs of living individuals for A. abruptifrons and A. japonicum are presented. A key is provided to facilitate the identification of the two species of Agramma distributed in Japan. Umeko Tsuda (1864-1929), a pioneering educator for Japanese women and the founder of Tsuda College, was a scientist. As an English teacher at the Peeresses School in Tokyo, the young Tsuda was granted a leave of absence by the government to study "teaching method" at Bryn Mawr College, a women's college near Philadelphia. During her stay in Bryn Mawr(1889-1892), however, she majored not in pedagogy but in biology, despite the fact that the Peeresses School officially banned science education for noble women. Following the vision of the feminist Dean Carrey Thomas, Bryn Mawr College offered full-fledged professional education in science comparable to that of Johns Hopkins University. Bryn Mawr's Biology Department was growing; there, Tsuda took courses from such notable biologists as Edmund B. Wilson, Jacques Loeb, and the future Nobel Laureate Thomas H. Morgan. In her third year, under Morgan, she carried out experimental research on the development of the frog's egg, which was published in a British scientific journal as their joint paper two years later. Tsuda was considered one of the best students in the department, and Bryn Mawr offered her opportunities for further study. However, after much consideration, she chose to return to Japan. Although Tsuda gave up a possibly great career as a biologist in American academe, she knew that it was almost impossible for a woman to pursue a scientific career in Meiji Japan and wanted to develop her dream of establishing an English school for women. Her experience of "forbidden" scientific study at Bryn Mawr seems to have given her great confidence in realizing her feminist ideal of enlightening Japanese women at the women's school she founded in 1900, the forerunner of Tsuda College.

The red swamp crayfish Procambarus clarkii (Girard, 1852) has been responsible for negative impacts on native benthic fauna and flora in invaded freshwater ecosystems around the world, including Japan. We need to clarify the reproductive biology in the invaded habitats as basic information to effectively control the introduced populations, but the reproductive biology of P. clarkii in Japan (especially in western Japan) has not been well studied. We conducted monthly samplings of P. clarkii from November 2015 to November 2016 in a pond (which does not freeze, even in winter) in Okayama, western Japan, both by using shrimp cage traps and by hand nets to examine aspects of the reproduction, including a form alternation (i.e. Form I and II). We also reared spawning females in the laboratory and calculated the accumulated water temperature during the period to Stage-3 juveniles after spawning. The total number of individuals caught throughout the study period was 6,319 (2,601 males and 2,777 females, with eight of unknown sex and 933 juveniles). The males were all Form I with a breeding status from September to November 2016. We first found ovigerous females in July 2016 and females carrying hatchlings in October 2016, even in January 2016. The form alternation in males was confirmed not only in the cheliped length, but in the hook length. In laboratory observations, approximately 52 days with approximately 1,222 °C degree-days of the accumulated water temperature were necessary to Stage-3 juveniles after spawning. Bembecia kaszabi(Capu§e, 1973)is newly recorded from Kyushu, Japan. This record is the first discovery of the genus Bembecia from Japan. The article reports that critics questioned the claims made by biologist Haruko Obokata and coworkers including anaesthetist Charles Vacanti in their stem cell biology study that reprogrammed mature mice cells into an embryonic state by brief exposure to an acid solution, published in "Nature."

The study of biomagnetic fields originating in a biological body is called biomagnetism. Among various fields of biomagnetism, this paper reviews the research and clinical works in magnetocardiogram, neuromagnetism, magneto-oculogram, magnetopneumogram and magnetic measurement for cell motility carried out in Japan.

Publication data relating to a rare and obscure Japanese journal Lansania Journal of arachnology and zoology (1929–1941) are examined. Available facts, together with a substantial body of circumstantial and anecdotal evidence suggest that many planned issues, including several cited by independent sources as having been published, were not published. Some biographical data relating to the editor, Kyukichi Kishida (1888–1968), are provided. Titles of all papers known to have been published in Lansania, with page numbers and claimed publication dates are presented, together with a list of 113 new zoological names proposed in the journal. Known library holdings of the journal worldwide are indicated. Details are provided of unpublished manuscripts in proof obtained from Kishida in the 1960s. A new species of Microgastrinae (Hymenoptera: Braconidae) parasitoid wasp, Microgaster godzilla Fernandez-Triana & Kamino, is described from Japan. From a biological and morphological perspective this is a very unusual species. It represents only the third known microgastrine to be aquatic, and the first one to be found entering the water. The female wasp searches for its hosts, aquatic larvae of Elophila turbata (Lepidoptera: Crambidae), mostly by walking over floating plants, but occasionally diving underwater for several seconds to force the larva out of its case, when it is quickly parasitized (parasitization was always observed above water). The unique searching behaviour of M. godzilla as well as its parasitization of aquatic larvae was filmed and it is presented here. The wasp has simple tarsal claws, which are elongate and strongly curved, similar to those found in the related genus Hygroplitis; they seem to represent an adaptation for gripping to the substrate when entering the water. The new species is described based on morphological, molecular (DNA barcoding), biological and ethological data. Additionally, we provide detailed diagnoses to recognize M. godzilla from all other described species of Microgaster and Hygroplitis in the Palearctic, Nearctic and Oriental regions. The mature larva of Cionus olivieri Rosenschoeld, 1838 is described and illustrated in detail for the first time. It is compared with those known from the same genus and other genera in the tribe Cionini and with those of the hypothesized sister tribe Mecinini in the Curculioninae. The larvae of Cionus have three distinctive diagnostic features: the reduced number of setae on the epicranium (only two or three des and one or two fs) and on the epipharyngeal lining (only two als, two ams, and no mes); i.e., distinctly fewer than the most frequent number of setae in weevils, and mandibles dentate or angulate internally near the base. If considered together with Stereonychus Suffrian, 1854, the other genus of Cionini with larvae studied in detail, it is preliminarily suggested that mature larvae of this tribe might be characterized by six main diagnostic features: (1) labial palpi one-segmented, (2) labral rods absent, (3) pedal areas swollen to form large lobes or prolegs, (4) mandible with sharp apical teeth, (5) reduced number of fs on frons, only one or two fs, and (6) reduced number of epipharyngeal setae (two or three als and two or three ams, but no mes). It was noticed that C. helleri Reitter, 1904 from Japan, a very distinct species in the genus for some characters of the adult, also possesses distinctive characters in the larva which are uncommon among known cionines. New biological data on C. olivieri with the discovery of its host plant, Verbascum songaricum (Scrophulariaceae), in central Asia are also reported. [ABSTRACT FROM AUTHOR]

We compiled historical reports of megamouth sharks Megachasma pelagios (mostly fishery by‐catch and strandings) from 1976 to 2018 (n = 117) and found that they are distributed globally (highest latitude, 36°) with three hotspots: Japan, Taiwan and the Philippines. Despite possible biases due to variability in fishing effort, more individuals were reported at higher latitudes in the summer, suggesting seasonal, latitudinal migrations. Sex ratios were female‐biased in Japan, but more even in Taiwan and the Philippines, suggesting some sexual segregation. Females (total length, LT = 3.41–7.10 m) were larger than males (LT = 1.77–5.39 m) and matured at a larger LT (5.17 m) than males (4.26 m). Also, we reviewed the systematics, feeding ecology and swimming behaviour of Megachasma pelagios based on the literature. Our review shows that, compared with their morphology, anatomy and genetics, behavioural ecology of this species remains largely unknown and electronic tagging studies are warranted. Purpose: In the early 2010s, Japanese society recognized and experienced a panic about increasing infertility and people's lack of knowledge about human reproduction. This chapter focuses on several graphs that misrepresented or distorted scientific findings that were used in the campaign related to this panic and explores (1) how the graphs were made, used, and authorized, and (2) how they contributed to changes in discourses and policies. Methodology/approach: Literature survey. Findings: (1) The graphs were made in the field of obstetrics, gynecology, and reproductive medicine by questionable methods, including falsifying, trimming, and misunderstanding of data. (2) Researchers in the field of fertility study relied on secondary and tertiary sources thus ignoring and compounding errors. (3) Such inauthentic research was approved and politically mobilized by professional organizations, rather than being penalized or criticized. (4) Discourse based on such unscientific knowledge may have encouraged a pronatalist policy of promoting early marriage and education about human fertility and life planning, targeted at teenage girls. Research limitations/implications: Any society suffering from a low birthrate can experience similar phenomena. This study focuses on Japan, but it has wider implications about how low integrity and quality of the presentation of medical research can cause these issues elsewhere in the world. Social implications: This chapter includes a warning against biological explanations that contain unscientific connotations about gender. Originality/value of study: This study confirms how gender-related policy in 2010s Japan was influenced by science that lacked research integrity and was of sub-standard quality. The mirine plant bug genus Phytocoris Fallén of Japan is reviewed; twelve species are recognized. Eight new species are proposed, diagnosed, and described: P. amateras, P. hasegawai, P. izanagii, P. izanamiae, P. kerzhnerianus, P. minakatai, P. miyamotoi, and P. protobothrops. Four known species are diagnosed, and two poorly recognized species, P. ohataensis Linnavuori and P. pallidicollis Kerzhner, are also redescribed. Phytocoris scotinus Kerzhner is proposed as a junior synonym of P. ohataensis. The male and female genitalic structures are figured and the biological information is documented for most treated species. Mutations in LIPH cause of autosomal recessive woolly hair/hypotrichosis (ARWH), and the 2 missense mutations c.736T>A (p.Cys246Ser) and c.742C>A (p.His248Asn) are considered prevalent founder mutations for ARWH in the Japanese population. To reveal genotype/phenotype correlations in ARWH cases in Japan and the haplotypes in 14 Japanese patients from 14 unrelated Japanese families. 13 patients had woolly hair, and 1 patient had complete baldness since birth. An LIPH mutation search revealed homozygous c.736T>A mutations in 10 of the patients. Compound heterozygous c.736T>A and c.742C>A mutations were found in 3 of the patients, and homozygous c.742C>A mutation in 1 patient. The phenotype of mild hypotrichosis with woolly hair was restricted to the patients with the homozygous c.736T>A mutation. The severe phenotype of complete baldness was seen in only 1 patient with homozygous c.742C>A. Haplotype analysis revealed that the alleles containing the LIPH c.736T>A mutation had a haplotype identical to that reported previously, although 4 alleles out of 5 chromosomes containing the LIPH c.742C>A mutation had a different haplotype from the previously reported founder allele. These alleles with c.742C>A are thought to be the third founder LIPH mutation causing ARWH. To accurately determine the prevalence of the founder mutations, we investigated allele frequencies of those mutations in 819 Japanese controls. Heterozygous c.736T>A mutations were found in 13 controls (allele frequency: 0.0079; carrier rate: 0.016), and heterozygous c.742C>A mutations were found in 2 controls (allele frequency: 0.0012; carrier rate: 0.0024). In conclusion, this study confirms the more accurate allele frequencies of the pathogenic founder mutations of LIPH and shows that there is a third founder mutation in Japan. In addition, the present findings suggest that the mutation patterns of LIPH might be associated with hypotrichosis severity in ARWH. [ABSTRACT FROM AUTHOR]

To evaluate the relationships between coral calcification, thermal stress, and sedimentation and eutrophication linked to human impact (hereafter referred to as “land development”) by river discharge, we analyzed growth characteristics in the context of a paleoenvironment that was reconstructed from geochemical signals in modern and fossil (1.2 cal kyr BP and 3.5 cal kyr BP, respectively) massive Porites corals from Nagura Bay (“Nagura”) and from modern Porites corals from the estuary of the Todoroki River, Shiraho Reef (“Todoroki”). Both sites are on Ishigaki Island, Japan, and Nagura is located approximately 12 km west of Todoroki. At Nagura, the individual corals provide time windows of 13 (modern), 10 (1.2 cal kyr BP), and 38 yr in length (3.5 cal kyr BP). Here, we present the coral annual calcification for Nagura and Todoroki, and (bi) monthly resolved records of Sr/Ca (a proxy of sea surface temperature (SST)) and Ba/Ca (a proxy of sedimentation and nutrients related to land development) for Nagura. At Nagura, the winter SST was cooler by 2.8°C in the 1.2 cal kyr BP, and the annual and winter SSTs in the 3.5 cal kyr BP were cooler by 2.6°C and 4.6°C, respectively. The annual periodicity of Ba/Ca in modern coral is linked to river discharge and is associated with land development including sugar cane cultivation. Modern coral calcification also has declined with SST warming and increasing Ba/Ca peaks in winter. However, calcification of fossil corals does not appear to have been influenced by variations in Sr/Ca and Ba/Ca. Modern coral growth characteristics at Nagura and Todoroki indicate that coral growth is both spatially and temporally influenced by river discharge and land development. At Nagura, our findings suggest that land development induces negative thermal sensitivity for calcification in winter due to sugar cane harvest, which is a specifically modern phenomenon. The objective of this study was to estimate influenza vaccine effectiveness (VE) against medically attended, laboratory-confirmed influenza during the 2011–2012 season in Japan using a test-negative case-control study design. The effect of co-circulating non-influenza respiratory viruses (NIRVs) on VE estimates was also explored. Nasopharyngeal swab samples were collected from outpatients with influenza-like illnesses (ILIs) in a community hospital in Nagasaki, Japan. Thirteen respiratory viruses (RVs), including influenza A and B, were identified from the samples using a multiplex polymerase chain reaction. The difference in VE point estimates was assessed using three different controls: ILI patients that tested negative for influenza, those that tested negative for all RVs, and those that tested positive for NIRVs. The adjusted VE against medically attended, laboratory-confirmed influenza using all influenza-negative controls was 5.3% (95% confidence interval [CI], −60.5 to 44.1). The adjusted VEs using RV-negative and NIRV-positive controls were −1.5% (95% CI, −74.7 to 41) and 50% (95% CI, −43.2 to 82.5), respectively. Influenza VE was limited in Japan during the 2011–2012 season. Although the evidence is not conclusive, co-circulating NIRVs may affect influenza VE estimates in test-negative case-control studies. The Nabe-kaburi is a unique burial method, the purpose of which is shrouded in mystery. The burials were performed during the 15th to 18th centuries in eastern Japan, and involved covering the heads of the deceased with iron pots or mortars. The identification of leprosy-specific osteological lesions among some of the excavated remains has led to the suggestion that Nabe-kaburi burials were a reflection of the social stigma against certain infectious diseases, such as leprosy, tuberculosis or syphilis. However, molecular evidence for the presence of disease has been lacking. The goal of this study was to detect Mycobacterium leprae (M. leprae) DNA in archaeological human skeletal remains from Nabe-kaburi burials. The paleopathological data from three Nabe-kaburi burials were re-evaluated before small samples were taken from affected and control areas. DNA was extracted and used as a template to target the M. leprae-specific DNA using a combination of whole genome amplification, PCR analysis and DNA sequencing. M. leprae DNA fragments were detected in the two sets of skeletal remains that had also shown paleopathological evidence of leprosy. These findings provide definitive evidence that some of the Nabe-kaburi burials were performed for people affected by leprosy. Demonstration of the presence of M. leprae DNA, combined with archeological and anthropological examinations, will aid in solving the mystery of why Nabe-kaburi burials were performed in medieval Japan. Although Japanese herbal medicines (JHMs) are widely used in Japan, only a few studies have investigated their effects on nonalcoholic steatohepatitis (NASH). In the present study, we examined the effect of 4 kinds of JHMs [sho-saiko-to (TJ-9), inchin-ko-to (TJ-135), juzen-taiho-to (TJ-48), and keishi-bukuryo-gan (TJ-25)] on a mouse model of NASH. Db/db mice were divided into 6 groups: control diet (control), methionine- and choline-deficient diet (MCD), and MCD diet supplemented with TJ-9, TJ-135, TJ-48, and TJ-25 (TJ-9, TJ-135, TJ-48, and TJ-25, respectively). All mice were sacrificed after 4 weeks of treatment, and biochemical, pathological, and molecular analyses were performed. Serum alanine aminotransferase levels and liver histology, including necroinflammation and fibrosis, were significantly alleviated in the TJ-9 and TJ-48 groups compared with the MCD group. The expression level of transforming growth factor (TGF)-β1 mRNA in the liver was significantly suppressed by TJ-48. Although the differences were not statistically significant, the expression levels of tumor necrosis factor (TNF)-α and interleukin (IL)-6 were lower, and those of peroxisome proliferators-activated receptor (PPAR)γ were higher in the TJ-9 and/or TJ-48 groups than in the MCD group. Similarly, even though the results were not statistically significant, malondialdehyde levels in liver tissues were lower in the TJ-9 and TJ-48 groups than in the MCD group. We showed that JHMs, especially TJ-9 and TJ-48, inhibited the necroinflammation and fibrosis in the liver of a mouse model of NASH, even though the mechanisms were not fully elucidated. Epidemiological studies suggest that retroviruses, including human immunodeficiency virus type 1, are associated with cardiomyopathy and myocarditis, but a causal relationship remains to be established. We encountered unusual cardiomyocyte hypertrophy and mitosis in Japanese native fowls infected with subgroup A of the avian leukosis viruses (ALVs-A), which belong to the genus Alpharetrovirus of the family Retroviridae and mainly induce lymphoid neoplasm in chickens. The affected hearts were evaluated by histopathology and immunohistochemistry, viral isolation, viral genome sequencing and experimental infection. There was non-suppurative myocarditis in eighteen fowls and seven of them had abnormal cardiomyocytes, which were distributed predominantly in the left ventricular wall and showed hypertrophic cytoplasm and atypical large nuclei. Nuclear chains and mitosis were frequently noted in these cardiomyocytes and immunohistochemistry for proliferating cell nuclear antigen supported the enhancement of mitotic activity. ALVs were isolated from all affected cases and phylogenic analysis of envSU genes showed that the isolates were mainly classified into two different clusters, suggesting viral genome diversity. In ovo experimental infection with two of the isolates was demonstrated to cause myocarditis and cardiomyocyte hypertrophy similar to those in the naturally occurring lesions and cardiac hamartoma (rhabdomyoma) in a shorter period of time (at 70 days of age) than expected. These results indicate that ALVs cause myocarditis as well as cardiomyocyte abnormality in chickens, implying a pathogenetic mechanism different from insertional mutagenesis and the existence of retrovirus-induced heart disorder. During recent decades, over 40% of Japanese estuarine tidal flats have been lost due to coastal developments. Local populations of the saltmarsh sesarmid crab Clistocoeloma sinense, designated as an endangered species due to the limited suitable saltmarsh habitat available, have decreased accordingly, being now represented as small remnant populations. Several such populations in Tokyo Bay, have been recognised as representing distributional limits of the species. To clarify the genetic diversity and connectivity among local coastal populations of Japanese Clistocoeloma sinense, including those in Tokyo Bay, mitochondrial DNA analyses were conducted in the hope of providing fundamental information for future conservation studies and an understanding of metapopulation dynamics through larval dispersal among local populations. All of the populations sampled indicated low levels of genetic diversity, which may have resulted from recent population bottlenecks or founder events. However, the results also revealed clear genetic differentiation between two enclosed-water populations in Tokyo Bay and Ise-Mikawa Bay, suggesting the existence of a barrier to larval transport between these two water bodies. Since the maintenance of genetic connectivity is a requirement of local population stability, the preservation of extant habitats and restoration of saltmarshes along the coast of Japan may be the most effective measures for conservation of this endangered species. Background: Norovirus (NoV) is a major cause of gastroenteritis during the autumn and winter seasons in Japan as well as in other temperate climate regions. Most outbreaks are thought to occur by secondary attacks through person-to-person infection by fecal-oral route. Severe cases are found in young children or patients with chronic diseases. Clarifying the patterns of epidemic diffusion is important for considering effective monitoring and surveillance as well as possible prevention. Methods: We considered the predominant viral genotype from the laboratory result obtained from Infectious Agents Surveillance Report (IASR) of National Institute of Infectious Diseases (NIID). We investigated the increase of NoV cases nationwide for the 2006–07 to 2008–09 seasons using sentinel gastroenteritis data collected from about 3000 pediatric clinics on National Epidemiological Surveillance of Infectious Diseases (NESID) acquired from the kriging method in the geographic information system (GIS). Results: During these three seasons, the majority of the detected virus was GII.4, which ranged from 60.4 to 88.9%. The number of cases (per sentinel site) at the peak week was 22.81 in the 2006–07 season and it decreased in the following seasons. NoV cases began to increase earlier in the southern areas and gradually extended into the northern areas, similarly, over the seasons. The average period from when the increase of cases was detected in the southern area to when it reached the northern area was 12.7 weeks. Conclusion: The decrease of the number of sentinel cases at the peak week may suggest the development of herd immunity after a period of high prevalence. Although the NoV epidemic is thought to be associated with cold weather, its cases first increased in the southern area with relatively warm temperature, indicating there are other climate factors involved. Background: Angiostrongylus cantonensis (Chen, 1935) is a parasite of murid rodents and causative agent of human neuro-angiostrongyliasis. In 2011, the Ogasawara Islands in the western North Pacific were assigned a World Natural Heritage site status. The occurrence of A. cantonensis is well documented in the Chichijima, Hahajima, and Anijima Islands. However, the occurrence of A. cantonensis in the other islands of the Ogasawara Islands has not been reported. Methodology/Principal Findings: Between March 2010 and July 2011, 57 Rattus norvegicus and 79 R. rattus were collected from 9 islands (the Hahajima group: Anejima, Imoutojima, Meijima, Mukohjima, and Hirajima; Chichijima group: Minamijima; Mukojima group: Nakoudojima and Yomejima; and Iwojima group: Iwojima). Adult nematodes were found in the pulmonary artery of 46 R. norvegicus collected in the 5 islands of the Hahajima group (Anejima, Meijima, Imoutojima, Hrajima, and Mukohjima Islands). These nematodes were identified by molecular analysis as A. cantonensis. Comparison of the mitochondrial DNA sequences confirmed that all the samples from the Ogasawara Islands shared only a single lineage of A. cantonensis, which has been previously detected in the Okinawa, Hawaii, and Brazil. Conclusions/Significance: We describe new endemic foci of rat angiostrongyliasis in the Hahajima group (Anejima, Meijima, Imoutojima, Hirajima, and Mukohjima Islands) of the Ogasawara Islands. These findings indicate that the endemic foci of A. cantonensis are widely distributed in the Ogasawara Islands. Although human cases have not yet been reported in the Ogasawara Islands, the widespread detection of A. cantonensis could be of importance from the perspective of public health. Background: EML4-ALK fusion gene is found in only a small subset (2–6%) of non-small cell lung cancer. There is an urgent need to establish a rational diagnostic algorithm to identify this rare but important fusion in lung cancer. Methods: We performed a comprehensive analysis of EGFR/KRAS mutation and ALK rearrangement in a total of 360 surgically resected lung cancers. ALK rearrangement was examined by 3 analyses: multiplex reverse transcription-PCR, fluorescent in situ hybridization (FISH), and immunohistochemistry (IHC) with the intercalated antibody-enhanced polymer method. A scoring system was used for IHC (iScore). A test set (202 patients with unselected lung cancer) was used for proposing a diagnostic algorithm. This diagnostic algorithm was validated in 158 patients with EGFR and KRAS mutation-negative adenocarcinoma. Results: ALK rearrangement was identified in 2 patients (1.0%) from the test set and both adenocarcinomas were negative for EGFR and KRAS mutations. The results of FISH and RT-PCR were completely matched. The highest iScore 3 was found only in the 2 positive cases. A diagnostic algorithm was proposed: IHC screening for ALK rearrangement followed by confirmatory FISH. In the validation set, 8 cases (5.1%) had iScore 3 and were positive for FISH, while the other cases had iScore 0 and were negative for FISH. Conclusions: Screening for ALK rearrangement by IHC followed by confirmatory FISH is a rational diagnostic algorithm. Background: For the pending National Claims Database in Japan, researchers will not have access to death information in the enrollment files. We developed and evaluated a claims-based definition of death. Methodology/Principal Findings: We used healthcare claims and enrollment data between January 2005 and August 2009 for 195,193 beneficiaries aged 20 to 74 in 3 private health insurance unions. We developed claims-based definitions of death using discharge or disease status and Charlson comorbidity index (CCI). We calculated sensitivity, specificity and positive predictive values (PPVs) using the enrollment data as a gold standard in the overall population and subgroups divided by demographic and other factors. We also assessed bias and precision in two example studies where an outcome was death. The definition based on the combination of discharge/disease status and CCI provided moderate sensitivity (around 60%) and high specificity (99.99%) and high PPVs (94.8%). In most subgroups, sensitivity of the preferred definition was also around 60% but varied from 28 to 91%. In an example study comparing death rates between two anticancer drug classes, the claims-based definition provided valid and precise hazard ratios (HRs). In another example study comparing two classes of anti-depressants, the HR with the claims-based definition was biased and had lower precision than that with the gold standard definition. Conclusions/Significance: The claims-based definitions of death developed in this study had high specificity and PPVs while sensitivity was around 60%. Fatty acid has been suggested to be involved in development of diabetes. However, its association is unclear among Japanese populations, which consume large amounts of fish rich in n-3 polyunsaturated fatty acids. The present cross-sectional study examined the association of individual dietary fatty acids and dietary fatty acid patterns with abnormal glucose metabolism among 1065 Japanese employees, aged 18–69 years. Impaired glucose metabolism is defined if a person has a history of diabetes, current use of anti-diabetic drug, fasting plasma glucose of 110 mg/dl (≥6.1 mmol/L) or greater, or hemoglobin A1C of 6.0% (≥42 mmol/mol) or greater. Dietary intake was assessed with a self-administered diet history questionnaire. Dietary fatty acid patterns were extracted by principal component analysis. Odds ratios of impaired glucose metabolism according to tertile categories of each fatty acids and dietary fatty acid patterns were estimated using logistic regression with adjustment for potential confounding variables. A higher intake of polyunsaturated fatty acid, n-6 fatty acid, linoleic acid, and oleic acid were significantly associated with a decreased prevalence of impaired glucose metabolism (P for trend = 0.03, 0.01, 0.02, and 0.04, respectively). Alpha-linolenic acid was marginally significantly associated with a decreased prevalence of impaired glucose metabolism (P for trend = 0.12). Of three fatty acid patterns identified, a higher plant oil pattern score, which characterized by high intake of alpha-linolenic acid, linoleic acid, and oleic acid, was associated with a decreased prevalence of impaired glucose metabolism (P for trend = 0.03). Background: Despite advances in our understanding of the countercyclical association between economic contraction and suicide, less is known about the levels of and changes in inequalities in suicide. The authors examined social and geographical inequalities in suicide in Japan from 1975 through 2005. Methods: Based on quinquennial vital statistics and census data, the authors analyzed the entire population aged 25–64 years. The total number of suicides was 75,840 men and 30,487 women. For each sex, the authors estimated odds ratios (ORs) and 95% credible intervals (CIs) for suicide using multilevel logistic regression models with “cells” (cross-tabulated by age and occupation) at level 1, seven different years at level 2, and 47 prefectures at level 3. Prefecture-level variance was used as an estimate of geographical inequalities in suicide. Results: Adjusting for age and time-trends, the lowest odds for suicide was observed among production process and related workers (the reference group) in both sexes. The highest OR for men was 2.52 (95% CI: 2.43, 2.61) among service workers, whereas the highest OR for women was 9.24 (95% CI: 7.03, 12.13) among security workers. The degree of occupational inequalities increased among men with a striking change in the pattern. Among women, we observed a steady decline in suicide risk across all occupations, except for administrative and managerial workers and transport and communication workers. After adjusting for individual age, occupation, and time-trends, prefecture-specific ORs ranged from 0.76 (Nara Prefecture) to 1.36 (Akita Prefecture) for men and from 0.79 (Kanagawa Prefecture) to 1.22 (Akita Prefecture) for women. Geographical inequalities have increased primarily among men since 1995. Conclusions: The present findings demonstrate a striking temporal change in the pattern of social inequalities in suicide among men. In Japan, Japanese cedar pollen dispersal is one of the major causes of pollinosis. Sydowia japonica is an ascomycetous fungus that grows exclusively on the male strobili of Japanese cedar, suggesting a possible mechanism for controlling pollen dispersal. To evaluate this possibility, eleven isolates of S. japonica were collected from around Japan and used as an inoculum to male strobili of Japanese cedar. The treatment demonstrated that the fungus infected only the pollen and prevented pollen dispersal. The fungus did not cause any additional symptoms to other parts of Japanese cedar, such as needles, stems, and buds. All S. japonica isolates collected around Japan could serve to control pollen dispersal. Periodic observation of the fungal pathogenesis with stereomicroscope and scanning electron microscope showed that hyphal fragments and conidia of S. japonica germinated on the surface of male strobili, and the germ tube entered pollen sacs through opening microsporophylls. Within the pollen sacs, the hyphae penetrated pollen gradually, such that all pollen was infected by the fungus by approximately one month before the pollen dispersal season. The infected pollen was destroyed due to the fungal infection and was never released. Our data suggests a novel approach of preventing pollen dispersal using pollen-specific fungal infection. Hepatitis C virus (HCV) infection is the major cause of hepatocellular carcinoma (HCC) in Japan. We previously identified the association of SNP rs2596542 in the 5' flanking region of the MHC class I polypeptide-related sequence A (MICA) gene with the risk of HCV-induced HCC. In the current study, we performed detailed functional analysis of 12 candidate SNPs in the promoter region and found that a SNP rs2596538 located at 2.8 kb upstream of the MICA gene affected the binding of a nuclear protein(s) to the genomic segment including this SNP. By electrophoretic mobility shift assay (EMSA) and chromatin immunoprecipitation (ChIP) assay, we identified that transcription factor Specificity Protein 1 (SP1) can bind to the protective G allele, but not to the risk A allele. In addition, reporter construct containing the G allele was found to exhibit higher transcriptional activity than that containing the A allele. Moreover, SNP rs2596538 showed stronger association with HCV-induced HCC (P = 1.82×10−5 and OR = 1.34) than the previously identified SNP rs2596542. We also found significantly higher serum level of soluble MICA (sMICA) in HCV-induced HCC patients carrying the G allele than those carrying the A allele (P = 0.00616). Coral bleaching, triggered by elevated sea-surface temperatures (SSTs) has caused a decline in coral cover and changes in the abundances of corals on reefs worldwide. Coral decline can be exacerbated by the effects of local stressors like turbidity, yet some reefs with a natural history of turbidity can support healthy and resilient coral communities. However, little is known about responses of coral communities to bleaching events on anthropogenically turbid reefs as a result of recent (post World War II) terrestrial runoff. Analysis of region-scale coral cover and species abundance at 17–20 sites on the turbid reefs of Okinawa Island (total of 79 species, 30 genera, and 13 families) from 1995 to 2009 indicates that coral cover decreased drastically, from 24.4% to 7.5% (1.1%/year), subsequent to bleaching events in 1998 and 2001. This dramatic decrease in coral cover corresponded to the demise of Acropora species (e.g., A. digitifera) by 2009, when Acropora had mostly disappeared from turbid reefs on Okinawa Island. In contrast, Merulinidae species (e.g., Dipsastraea pallida/speciosa/favus) and Porites species (e.g., P. lutea/australiensis), which are characterized by tolerance to thermal stress, survived on turbid reefs of Okinawa Island throughout the period. Our results suggest that high turbidity, influenced by recent terrestrial runoff, could have caused a reduction in resilience of Acropora species to severe thermal stress events, because the corals could not have adapted to a relatively recent decline in water quality. Background: The marine epiphytic dinoflagellate genus Gambierdiscus produce toxins that cause ciguatera fish poisoning (CFP): one of the most significant seafood-borne illnesses associated with fish consumption worldwide. So far, occurrences of CFP incidents in Japan have been mainly reported in subtropical areas. A previous phylogeographic study of Japanese Gambierdiscus revealed the existence of two distinct phylotypes: Gambierdiscus sp. type 1 from subtropical and Gambierdiscus sp. type 2 from temperate areas. However, details of the genetic diversity and distribution for Japanese Gambierdiscus are still unclear, because a comprehensive investigation has not been conducted yet. Methods/Principal Finding: A total of 248 strains were examined from samples mainly collected from western and southern coastal areas of Japan during 2006–2011. The SSU rDNA, the LSU rDNA D8–D10 and the ITS region were selected as genetic markers and phylogenetic analyses were conducted. The genetic diversity of Japanese Gambierdiscus was high since five species/phylotypes were detected: including two reported phylotypes (Gambierdiscus sp. type 1 and Gambierdiscus sp. type 2), two species of Gambierdiscus (G. australes and G. cf. yasumotoi) and a hitherto unreported phylotype Gambierdiscus sp. type 3. The distributions of type 3 and G. cf. yasumotoi were restricted to the temperate and the subtropical area, respectively. On the other hand, type 1, type 2 and G. australes occurred from the subtropical to the temperate area, with a tendency that type 1 and G. australes were dominant in the subtropical area, whereas type 2 was dominant in the temperate area. By using mouse bioassay, type 1, type 3 and G. australes exhibited mouse toxicities. Conclusions/Significance: This study revealed a surprising diversity of Japanese Gambierdiscus and the distribution of five species/phylotypes displayed clear geographical patterns in Japanese coastal areas. We conducted a survey of adults in Kotohira, a town of about 10,000 people located in the Nakatado District of Kagawa Prefecture, Japan. The survey was distributed to 8184 individuals, and effective responses were received from 3863 persons (response rate, 47.2%) during the survey period. Results regarding numbness and pain showed numbness alone in 7.7%, pain alone in 7.2%, both numbness and pain in 6.0%, and neither numbness nor pain in 79.6%. Spine and spinal cord damage was reported present by 5.4%, and absent by 94.6%. Analysis using the Short-Form Health Survey questionnaire, with comparison between subjects reporting both numbness and pain in the extremities and subjects with either numbness or pain alone, showed lower scores for in Short-Form Health Survey subscales (physical functioning, role [physical, emotional], bodily pain, vitality, and mental health). Subjects with numbness alone generally reported no disability in daily life. In a secondary survey, analysis of neurological findings by specialists identified 6 cases of “pain following spinal cord damage” in which spinal cord-related pain developed in the hands or feet. To discover susceptibility genes of late-onset Alzheimer’s disease (LOAD), we conducted a 3-stage genome-wide association study (GWAS) using three populations: Japanese from the Japanese Genetic Consortium for Alzheimer Disease (JGSCAD), Koreans, and Caucasians from the Alzheimer Disease Genetic Consortium (ADGC). In Stage 1, we evaluated data for 5,877,918 genotyped and imputed SNPs in Japanese cases (n = 1,008) and controls (n = 1,016). Genome-wide significance was observed with 12 SNPs in the APOE region. Seven SNPs from other distinct regions with p-values <2×10−5 were genotyped in a second Japanese sample (885 cases, 985 controls), and evidence of association was confirmed for one SORL1 SNP (rs3781834, P = 7.33×10−7 in the combined sample). Subsequent analysis combining results for several SORL1 SNPs in the Japanese, Korean (339 cases, 1,129 controls) and Caucasians (11,840 AD cases, 10,931 controls) revealed genome wide significance with rs11218343 (P = 1.77×10−9) and rs3781834 (P = 1.04×10−8). SNPs in previously established AD loci in Caucasians showed strong evidence of association in Japanese including rs3851179 near PICALM (P = 1.71×10−5) and rs744373 near BIN1 (P = 1.39×10−4). The associated allele for each of these SNPs was the same as in Caucasians. These data demonstrate for the first time genome-wide significance of LOAD with SORL1 and confirm the role of other known loci for LOAD in Japanese. This note reviews the work done by Ju-shey Ho, currently Professor Emeritus at California State University, Long Beach, CA, USA, on the systematics of symbiotic copepods from aquatic animals in Japanese waters. Since 1980, he has reported 110 species of symbiotic copepods from Japanese fish and marine invertebrates, including those representing one new family, seven new genera and 41 new species, and has greatly contributed to clarifying the symbiotic copepod fauna of Japan. Research using symbiotic copepods as bioindicators of the phylogeny and evolution of host animals was conducted by him for the first time in Japan. He also made significant contributions to the taxonomy and biology of caligid copepods, a group that poses a serious threat to the aquaculture industry, found on farmed fish in Japan. This study is dedicated to the environmental monitoring of radionuclides released in the course of the Fukushima nuclear accident. The activity concentrations of β− -emitting 90Sr and β−/γ-emitting 134Cs and 137Cs from several hot spots in Japan were determined in soil and vegetation samples. The 90Sr contamination levels of the samples were relatively low and did not exceed the Bq⋅g−1 range. They were up four orders of magnitude lower than the respective 137Cs levels. This study, therefore, experimentally confirms previous predictions indicating a low release of 90Sr from the Fukushima reactors, due to its low volatility. The radiocesium contamination could be clearly attributed to the Fukushima nuclear accident via its activity ratio fingerprint (134Cs/137Cs). Although the correlation between 90Sr and 137Cs is relatively weak, the data set suggests an intrinsic coexistence of both radionuclides in the contaminations caused by the Fukushima nuclear accident. This observation is of great importance not only for remediation campaigns but also for the current food monitoring campaigns, which currently rely on the assumption that the activity concentrations of β−-emitting 90Sr (which is relatively laborious to determine) is not higher than 10% of the level of γ-emitting 137Cs (which can be measured quickly). Monitoring of harmful algal bloom (HAB) species in coastal waters is important for assessment of environmental impacts associated with HABs. Co-occurrence of multiple cryptic species such as toxic dinoflagellate Ostreopsis species make reliable microscopic identification difficult, so the employment of molecular tools is often necessary. Here we developed new qPCR method by which cells of cryptic species can be enumerated based on actual gene number of target species. The qPCR assay targets the LSU rDNA of Ostreopsis spp. from Japan. First, we constructed standard curves with a linearized plasmid containing the target rDNA. We then determined the number of rDNA copies per cell of target species from a single cell isolated from environmental samples using the qPCR assay. Differences in the DNA recovery efficiency was calculated by adding exogenous plasmid to a portion of the sample lysate before and after DNA extraction followed by qPCR. Then, the number of cells of each species was calculated by division of the total number of rDNA copies of each species in the samples by the number of rDNA copies per cell. To test our procedure, we determined the total number of rDNA copies using environmental samples containing no target cells but spiked with cultured cells of several species of Ostreopsis. The numbers estimated by the qPCR method closely approximated total numbers of cells added. Finally, the numbers of cells of target species in environmental samples containing cryptic species were enumerated by the qPCR method and the total numbers also closely approximated the microscopy cell counts. We developed a qPCR method that provides accurate enumeration of each cryptic species in environments. In this study, we analyzed viral metagenomes (viromes) in the sedimentary habitats of three geographically and geologically distinct (hado)pelagic environments in the northwest Pacific; the Izu-Ogasawara Trench (water depth = 9,760 m) (OG), the Challenger Deep in the Mariana Trench (10,325 m) (MA), and the forearc basin off the Shimokita Peninsula (1,181 m) (SH). Virus abundance ranged from 106 to 1011 viruses/cm3 of sediments (down to 30 cm below the seafloor [cmbsf]). We recovered viral DNA assemblages (viromes) from the (hado)pelagic sediment samples and obtained a total of 37,458, 39,882, and 70,882 sequence reads by 454 GS FLX Titanium pyrosequencing from the virome libraries of the OG, MA, and SH (hado)pelagic sediments, respectively. Only 24−30% of the sequence reads from each virome library exhibited significant similarities to the sequences deposited in the public nr protein database (E-value <10−3 in BLAST). Among the sequences identified as potential viral genes based on the BLAST search, 95−99% of the sequence reads in each library were related to genes from single-stranded DNA (ssDNA) viral families, including Microviridae, Circoviridae, and Geminiviridae. A relatively high abundance of sequences related to the genetic markers (major capsid protein [VP1] and replication protein [Rep]) of two ssDNA viral groups were also detected in these libraries, thereby revealing a high genotypic diversity of their viruses (833 genotypes for VP1 and 2,551 genotypes for Rep). A majority of the viral genes predicted from each library were classified into three ssDNA viral protein categories: Rep, VP1, and minor capsid protein. The deep-sea sedimentary viromes were distinct from the viromes obtained from the oceanic and fresh waters and marine eukaryotes, and thus, deep-sea sediments harbor novel viromes, including previously unidentified ssDNA viruses. After a hiatus of some decades, gender transition was recognised and regulated by the legal and medical professions in Japan from the mid-1990s. Since the first of these gender reassignment procedures were carried out, the courts and the legislature have repeatedly had to come to terms with new situations. Decisions have been made on such matters as changing an individual's gender on their family register and other official documents, recognition of a transgendered person's marriage and, most recently, how to register the child of a marriage involving a transgendered parent. Assisted reproductive technologies have complicated this picture. Although there are gaps between the legal and medical discourses on gender transition, both the medical and legal professions operate according to assumptions about what constitutes a proper family. There are ever-widening gaps between the actual forms and practices of families in Japan, the cultural representations of alternative family forms, and the official policies of the legal and medical establishments. The Japanese crayfish ( Cambaroides japonicus), the only native crayfish in Japan, is endangered and has experienced rapid population declines. We surveyed the habitat requirements of Japanese crayfish at the reach and microhabitat scales in semi-natural low-gradient streams. Habitat use by Japanese crayfish differed between the spatial scales. Reach-scale analysis revealed that the bed slope was the only positive predictor of crayfish density. This finding indicates that relatively high-gradient reaches, such as headwater reaches, are an important habitat for the conservation of Japanese crayfish in low-gradient streams. Microhabitat-scale analysis showed that crayfish density was positively affected by substrate coarseness and the presence of instream vegetation cover (bank vegetation, woody debris, and leaf patches), whereas it was negatively affected by distance from the stream edge. Coarse substrates and vegetation cover may function as shelters from water flows and predators during low flow periods. The use of stream-edge areas may allow quick access to refugia and enable the crayfish to avoid unexpected flood disturbance and predation. These habitat characteristics should therefore be preserved for the conservation of Japanese crayfish, and scale-dependent habitat characteristics should be considered in future conservation plans.
