Tree roots penetrate the soil to several meters depth, but the role of subsoils for the supply of nutrient elements such as phosphorus (P) to the trees is poorly understood. Here, we tested the hypothesis that increased P deficiency in the topsoil results in an increased microbial recycling of P from the forest subsoil. We sampled soils from four German temperate forest sites representing a gradient in total P stocks. We analyzed the oxygen isotopic composition of HCl-extractable phosphate (δ18OP) and identified differences in P speciation with increasing soil depth using X-ray absorption near-edge structure (XANES) spectroscopy. We further determined microbial oxygen demand with and without nutrient supply at different soil depths to analyse nutrient limitation of microbial growth and used nanoscale secondary ion mass spectrometry (NanoSIMS) to visualize spatial P gradients in the rhizosphere. We found that δ18OP values in the topsoil of all sites were close to the isotopic signal imparted by biological cycling when oxygen isotopes in phosphate are exchanged by enzymatic activity. However, with increasing soil depth and increasing HCl-P concentrations, δ18Ο values continuously decreased towards values expected for primary minerals in parent material at depths below 60 cm at sites with high subsoil P stocks and below more than 2 m at sites with low subsoil P stocks, respectively. For these depths, XANES spectra also indicated the presence of apatite. NanoSIMS images showed an enrichment of P in the rhizosphere in the topsoil of a site with high P stocks, while this P enrichment was absent at a site with low P stocks and in both subsoils. Addition of C, N and P alone or in combination revealed that microbial activity in subsoils of sites with low P stocks was mostly P limited, whereas sites with high P stocks indicated N limitation or N and P co-limitation. We conclude that subsoil P resources are recycled by trees and soil microorganisms. With continued weathering of the bedrock and mobilisation of P from the weathered rocks, P cycling will proceed to greater depths, especially at sites characterised by P limitation. The study of ecosystem processes over multiple scales of space and time is often best achieved using comparable data from multiple sites. Yet, long-term ecological observatories have often developed their own data collection protocols. Here, we address this problem by proposing a set of ecological protocols suitable for widespread adoption by the ecological community. Scientists from the European ecological research community prioritized terrestrial ecosystem parameters that could benefit from a more consistent approach to data collection within the resources available at most long-term ecological observatories. Parameters for which standard methods are in widespread use, or for which methods are evolving rapidly, were not selected. Protocols were developed by domain experts, building on existing methods where possible, and refined through a process of field testing and training. They address above-ground plant biomass; decomposition; land use and management; leaf area index; soil mesofaunal diversity; soil C and N stocks, and greenhouse gas emissions from soils. These complement existing methods to provide a complete assessment of ecological integrity. These protocols offer integrated approaches to ecological data collection that are low cost and are starting to be used across the European Long Term Ecological Research community. Litter-feeding soil animals are notoriously neglected in conceptual and mechanistic biogeochemical models. Yet, they may be a dominant factor in decomposition by converting large amounts of plant litter into faeces. Here, we assess how the chemical and physical changes occurring when litter is converted into faeces alter their fate during further decomposition with an experimental test including 36 combinations of phylogenetically distant detritivores and leaf litter of contrasting physicochemical characteristics. We show that, across litter and detritivore species, litter conversion into detritivore faeces enhanced organic matter lability and thereby accelerated carbon cycling. Notably, the positive conversion effect on faeces quality and decomposition increased with decreasing quality and decomposition of intact litter. This general pattern was consistent across detritivores as different as snails and woodlice, and reduced differences in quality and decomposition amongst litter species. Our data show that litter conversion into detritivore faeces has far-reaching consequences for the understanding and modelling of the terrestrial carbon cycle. By performing an ecologically relevant experiment with variety in litter composition and detritivore species, Joly et al. find that litter conversion into faeces has a profound effect on organic matter lability and decomposition. Microscale processes occurring at biogeochemical interfaces in soils and sediments have fundamental impacts on phenomena at larger scales. To obtain the organo-mineral associations necessary for the study of biogeochemical interfaces, bulk samples are usually fractionated into microaggregates or micrometer-sized single particles. Such fine-grained mineral particles are often prepared for nanoscale secondary ion mass spectroscopy (NanoSIMS) investigations by depositing them on a carrier. This introduces topographic differences, which can strongly affect local sputtering efficiencies. Embedding in resin causes undesired C impurities. We present a novel method for preparing polished cross-sections of micrometer-sized primary soil particles that overcomes the problems of topography and C contamination. The particles are coated with a marker layer, embedded, and well-polished. The interpretation of NanoSIMS data is assisted by energy-dispersive X-ray spectroscopy on cross-sections prepared by a focused ion beam. In the cross-sections, organic assemblages on the primary soil particles become visible. This novel method significantly improves the quality of NanoSIMS measurements on grainy mineral samples, enabling better characterization of soil biogeochemical interfaces.
