Larix gemelinii is an important tree species in the Great Khingan Mountains in Northeast China with a high economic and ecological value for its role in carbon sequestration and as a source of lumber and nuts. However, the ectomycorrhizal (EM) fungal diversity and community composition of this tree remain largely undefined. We examined EM fungal communities associated with L. gemelinii from three sites in the Great Khingan Mountains using Illumina Miseq to sequence the rDNA ITS2 region and evaluated the impact of spatial, soil, and climatic variables on the EM fungal community. A total of 122 EM fungal operational taxonomic units (OTUs) were identified from 21 pooled-root samples, and the dominant EM fungal lineages were /tricholoma, /tomentella-thelephora, /suillus-rhizopogon, and /piloderma. A high proportion of unique EM fungal OTUs were present; some abundant OTUs largely restricted to specific sites. EM fungal richness and community assembly were significantly correlated with spatial distance and climatic and soil variables, with mean annual temperature being the most important predictor for fungal richness and geographic distance as the largest determinant for community turnover. Our findings indicate that L. gemelinii has a rich and distinctive EM fungal community contributing to our understanding of the montane EM fungal community structure from the perspective of a single host plant that has not been previously reported.

Despite the numerous studies on the large-scale patterns of species richness, the spatial variation and determinants of species richness for the alpine plants are still an outstanding question and critical to future biodiversity conservation. The genus Saxifraga is a typical alpine plant group with high species richness in the Himalaya-Hengduan Mountain (HHM) regions, China. We performed simple regression models and variance partitioning to assess the importance of different factors, especially soil-related ones, in driving Saxifraga richness patterns. The results showed that environmental energy, habitat heterogeneity, and soil heterogeneity together dominated the spatial variation of species richness. The coarse fragment volume (CRFVOL) of soil, elevation range, and soil heterogeneity are positively related to Saxifraga richness. Soil slightly outperforms habitat heterogeneity in predicting the spatial variation of Saxifraga species richness with an explanatory power of 39.3% and 36.6%, respectively. Environmental energy, such as the maximum temperature of the warmest quarter, is negatively correlated with species richness and explains 44.8% of spatial variation of Saxifraga richness. Multiple regression models, including three variables, each representing energy, soil, and habitat heterogeneity, can only explain 53.1% variation of species richness. Variance partitioning outscored 26% of the shared effects of the three variables, while the independent effect of each variable is less than 10%. These results indicated that the energy, soil, and habitat heterogeneity together are primary determinants of the spatial variation of Saxifraga species richness. However, there are probably other hidden factors predicting species richness variation due to the low explanatory power of the multiple regression models. Our study emphasizes the significance of soil properties in determining species richness patterns in China, especially for alpine plant groups. The negative association of species richness with temperature suggests a potential threat of alpine biodiversity loss in HHM from future warming.

Summary Elucidating interactions of above-ground and below-ground communities in different habitat types is essential for understanding biodiversity maintenance and ecosystem functioning. Using 454 pyrosequencing of ITS2 sequences we examined the relationship between subtropical mountain forest soil fungal communities, abiotic conditions, and plant communities using correlation and partial models. Ridge and valley habitats with differing fungal communities were delineated. Total, saprotrophic and pathogenic fungal richness were significantly correlated with plant species richness and/or soil nutrients and moisture in the ridge habitat, but with habitat convexity or basal area of Castanopsis eyrei in the valley habitat. Ectomycorrhizal (EM) fungal richness was significantly correlated with basal area of C. eyrei and total EM plants in the ridge and valley habitats, respectively. Total, saprotrophic, pathogenic and EM fungal compositions were significantly correlated with plant species composition and geographic distance in the ridge habitat, but with various combinations of plant species composition, plant species richness, soil C : N ratio and pH or no variables in the valley habitat. Our findings suggest that mechanisms influencing soil fungal diversity and community composition differ between ridge and valley habitats, and relationships between fungal and woody plant assemblages depend on habitat types in the subtropical forest ecosystem.

In order to explore the diversity, antimicrobial activity and enzyme-producing activity of marine actinobacteria isolated from mangrove sediments in Indian Ocean.Eight sediments collected from mangrove sediments in Indian Ocean were treated by the plate dilution method and spread on 24 isolation media only containing sole carbon source for energy. Marine actinobacteria were isolated and identified by 16S rRNA gene sequence analysis. The antimicrobial activity and enzyme-producing activity of isolated strains were further detected by spot planting method.In total 139 representative strains were selected from 521 isolates, and they were further sequenced and performed phylogenetic analysis based on their 16S rRNA gene sequences. There were 35 strains identified as potential novel species. Antimicrobial activity was detected in Bacillus subtilis, Candida albicans, Escherichia coli, Staphylococcus aureus, Aspergillus niger. Enzyme-producing activity for protease cellulase, amylase and esterase were 36.5%, 26.5%, 22.4% and 15.9%, respectively.Diverse marine actinobacteria were discovered in mangrove sediment in Indian Ocean, which have antimicrobial and enzyme activity.

By adopting antimicrobial spectrum test, BOXAIR-PCR, physiological and biochemical, and 16S rDNA sequencing analysis, this paper analyzed the diversity of 55 antagonistic bacterial strains isolated from the rhizosphere of 10 cash crops. There was a high diversity of the antagonism of the strains. Based on BOXAIR-PCR, all the strains were clustered into 7 groups at the similarity level of 72.1%, and divided into 25 groups at the similarity level of 85.0%. All the strains belonged to Bacillus, Paenibacillus, Brevibacillus, Pseudomonas and Alcaligenes, respectively. The antagonistic bacteria isolated from the rhizosphere had high genetic diversity and high diversity in antagonistic activity.

Taking non-transgenic parental rice as the control, and by using 13C pulse-chase labeling method coupled with phospholipid fatty acid (PLFA) analysis, this paper studied the effects of transgenic Bt rice on the microbial diversity in rhizosphere soil. The results showed that in the rhizosphere soils of transgenic Bt rice and its non-transgenic parent, saturated PLFAs and branched PLFAs were the main, followed by monounsaturated PLFAs, and polyunsaturated PLFAs. A significantly lower amount of Gram-positive bacterial PLFAs and a higher amount of Gram-negative bacterial PLFAs were observed in the rhizosphere soil of transgenic Bt rice at its seedling, booting, and heading stages, as compared with the control. In the whole growth period of rice, transgenic Bt gene had no significant effects on the fungal and actinomycete PLFAs in rhizosphere soil, and no significant difference was observed in the rhizosphere soil 13C-PLFA content between transgenic Bt rice and its non-transgenic parent. These findings indicated that the insertion of exogenous cry1Ab gene into rice only had temporary effects on the microbial community composition in the rhizosphere soil of rice.

As a new DNA fingerprinting technique, denaturing gradient gel electrophoresis (DGGE) can be used to analyze the microbial diversity in different environmental samples. Three biofilters treating air loaded with ammonia will be studied with respect to microbial diversity by applying cultivation-independent molecular methods DGGE. The principal objectives of this paper include the development and application of a suitable set of molecular tools to deliver information about the possible microbial community changes within carriers during treatment of ammonia-contaminated gas. Carriers were sampled at different time during operation of biofilters. After extraction and purification of the genomic DNA, the 16S rRNA genes (V3 region) were amplified by using the specific primers. These amplified DNA fragments were then separated by paralleled DGGE. The profile of DGGE showed that the same carrier at different time had the different bands' patterns. The structural diversity of the microbial community was examined by the Shannon index of general diversity H. Shannon indexes of three kinds of carriers all showed a decrease with time, which had a similar trend to NH3 removal efficiency.
