Biological soil crusts (biocrusts) are complex communities of micro‐ and macro‐organisms dwelling at the soil surface in dryland regions worldwide, which contribute to important ecological functions in these areas. Biocrusts may reach different developmental stages, associated to autogenic succession of specific phototrophic organisms. However, notwithstanding the large amount of existing literature, little is known regarding the relationship between biocrust stages of development and soil nutrient dynamics. In this opinion paper, we specifically focused on soil nitrogen, and compared the total nitrogen content and potential nitrogenase activity of different developmental types of biocrust (‐covered) soils. Based on published reports, we looked at a possible relationship between nitrogen content at the soil surface and biocrust development, discussing the potential mechanisms leading to the observed soil nitrogen content heterogeneity. The results showed that a higher nitrogen content remained associated to the presence of moss‐dominated biocrusts, and this biocrust morphotype was characterized by a lower potential nitrogenase activity compared to the cyanobacteria‐ and/or lichen‐dominated biocrusts. We hypothesized that these seemingly contradictory findings might be attributed to three potential mechanisms: nitrogen inheritance, nutrient retention and nutrient transfer, which we hereby examined one by one. Altogether, our opinion supports the theory of biocrust succession from incipient cyanobacteria‐dominated stages to more 'mature' stages dominated by mosses. We stress how the heterogeneous distribution of soil nitrogen, which is closely related with biocrust development and community types, eventually affects regional and even global nitrogen dynamic and storage. The successional ecology of nitrogen cycling in biocrusts and the linkages to ecosystem processes remains unclear. To explore this, four successional stages of natural biocrust with five batches of repeated sampling and three developmental stages of simulated biocrust were studied using relative and absolute quantified multi-omics methods. A consistent pattern across all biocrust was found where ammonium assimilation, mineralization, dissimilatory nitrite to ammonium (DNiRA), and assimilatory nitrate to ammonium were abundant, while denitrification medium, N-fixation, and ammonia oxidation were low. Mathematic analysis showed that the nitrogen cycle in natural biocrust was driven by dissolved organic N and NO3–. pH and SO42– were the strongest variables affecting denitrification, while C:(N:P) was the strongest variable affecting N-fixation, DNiRA, nitrite oxidation, and dissimilatory nitrate to nitrite. Furthermore, N-fixation and DNiRA were closely related to elemental stoichiometry and redox balance, while assimilatory nitrite to ammonium (ANiRA) and mineralization were related to hydrological cycles. Together with the absolute quantification and network models, our results suggest that responsive ANiRA and mineralization decreased during biocrust succession; whereas central respiratory DNiRA, the final step of denitrification, and the complexity and interaction of the whole nitrogen cycle network increased. Shrub encroachment is occurring in many of the world's drylands, but its impacts on ecosystem structure and function are still poorly understood. In particular, it remains unclear how shrub encroachment affects dryland soil surfaces, including biological soil crust (biocrust) communities. In this study, soil surfaces (0–1 cm depth) were sampled from areas of Grewia flava shrubs and Eragrostis lehmanniana and Schmidtia kalahariensis grasses in the southwest Kalahari during two different seasons (March and November). Our hypothesis is that the presence of different vegetation cover types (shrubs versus grasses) alters the microbial composition of soil surfaces owing to their contrasting microenvironments. The results showed that more significant differences in microclimate (light, soil surface temperatures) and soil surface microbial communities were observed between shrubs and grasses than between sampling seasons. Based on high-throughput 16S rRNA gene sequencing, our findings showed that approximately one third (33.5%) of the operational taxonomic units (OTUs) occurred exclusively in soil surfaces beneath shrubs. Soil surfaces with biocrusts in grass areas were dominated by the cyanobacteria Microcoleus steenstrupii , whereas the soil surfaces beneath shrubs were dominated by the proteobacteria Microvirga flocculans. Soil surfaces beneath shrubs are associated with reduced cyanobacterial abundance but have higher total carbon and total nitrogen contents compared to biocrusts in grass areas. These findings infer changes in the relative contributions from different sources of carbon and nitrogen (e.g. cyanobacterial and non-cyanobacterial fixation, plant litter, animal activity). The distinctive microbial composition and higher carbon and nitrogen contents in soil surfaces beneath shrubs may provide a positive feedback mechanism promoting shrub encroachment, which helps to explain why the phenomenon is commonly observed to be irreversible. Microbial communities within biocrusts fulfill important ecological functions, particularly in dryland environments. Identifying the optimal conditions for transporting and storing biocrusts is essential to preserve and accurately analyze their microbial communities. However, the effectiveness of these preservation methods remains poorly understood. In this study, we collected dry biocrusts at different developmental stages (shifting sand, cyanobacteria-dominated, and moss-dominated) from the Qbuqi Desert in northern China. We examined the effects of different storage conditions, i.e. at room temperature (25 ± 2 °C) and low temperature (4 °C and −80 °C) over one month on the microbial community characteristics, including species composition, abundance and diversity. Our findings demonstrated that microbial community differences among the biocrust developmental stages were maintained even after one month of storage at room temperature. Compared to the control (analyzed immediately after sampling), storing biocrusts at 25 °C and 4 °C did not significantly alter biocrust assemblages, whereas storage at −80 °C increased the estimated gene copy number of Proteobacteria in cyanobacteria-dominated biocrusts. Furthermore, our results suggest that biocrust developmental stage and microbial community composition had minimal impact on the effectiveness of different storage methods, though caution should be kept for the prokaryotic communities. Overall, we recommend storing dry-collected biocrusts at room temperature as an effective and economic method for preserving microbial communities, particularly for DNA-based microbial studies from dryland soils with frequent monitoring of biocrust development at remote areas. Overgrazing is a major driver of dryland degradation, however, so far, there is limited understanding on how this process affects biocrust-related microbial community, and especially how the key groups respond to grazing disturbance. In this study, quantitative polymerase chain reaction (qPCR) and high throughout sequencing technologies were used to investigate the bacterial community abundance and diversity in the Horqin Sandland (China) experiencing different livestock grazing disturbances, in order to examine whether a shift in bacterial community (in particular the key biocrust components, cyanobacteria) was involved, and how this was related to biocrust development and altered soil carbon and nitrogen level. Our results revealed that a clear heterogeneous soil bacterial community was associated with grazing disturbance, which inhibited biocrust development. The decreased photosynthetic cyanobacterial abundance (81.81 % in relative abundance and 98.83 % in absolute abundance) and nitrogen-fixing genes (87.72 %), associated with the lower total carbon and nitrogen content (P <0.05), illustrated a low soil carbon and nitrogen-fixing capability and nutrient level in the grazing-disturbed soils. In particular, a switch of nitrogen-fixing dominance from cyanobacteria to proteobacteria was induced by the grazing disturbance. Compared to the grazing-disturbed soils, higher dominant cyanobacterial operational taxonomic units (OTUs) (especially the species of Nostoc and Scytonema) and lower bacterial diversity (e.g., Shannon, Ace, and Chao index) were observed in the undisturbed biocrust soils, highlighting that the dominants rather than diversity played more important roles in biocrust development. Cyanobacterial cultivation emerges as a promising solution by extracting nutrients from wastewater and transferring them to deserts using cyanobacteria-induced biocrust technology. In this study, two biocrust inducing cyanobacteria, Microcoleus vaginatus and Scytonema javanicum were cultivated in synthetic and municipal wastewater, respectively, to assess their capabilities of nutrient removal and biocrust induction. Both cyanobacteria demonstrated effective removal of nitrogen and phosphorus from either synthetic or municipal wastewater, achieving removal rates exceeding 90 %. Sterilization experiments on the municipal wastewater confirmed that indigenous microorganisms did not influence cyanobacterial performance, though the two cyanobacteira exhibited contrasting growth and metabolic behaviors. Specifically, S. javanicum achieved higher biomass accumulation, reaching up to 6.05 ± 0.87 mg Chl- a L−1, whereas M. vaginatus produced more exopolysaccharides, up to 40.98 ± 2.96 mg L−1 in the municipal wastewater. The cyanobacteria harvested from the municipal wastewater effectively promoted biocrust formation, validating the feasibility of transferring nutrients from wastewater to deserts through cyanobacteria-induced biocrusts. Considering their varied growth behaviors, our findings advocate for the use of a combination of diverse cyanobacteria strains to optimize cultivation and inoculation technologies. Cyanobacterial inoculation is a promising technology that can induce biocrust development, for stabilizing degraded soils in dryland environments. However, it is challenging to grow cyanobacteria in dry, saline soils, which limits the application of this technology. In this study, NaCl was added into field-collected induced biocrusts to investigate how salt affects the biocrust community. This fundamental knowledge improves our assessment of microbial salinity preference and adaptation, and can guide researchers to select the appropriate cyanobacterial inoculants for saline soil restoration. The results showed that the biocrusts induced by inoculating Microcoleus vaginatus and Scytonema javanicum could survive 1.0% salinity, although both salt and drought stresses significantly inhibited microbial biomass and metabolism (P < 0.05), and salinity led to an obvious shift in microbial community structure. Compared to salt stress, drought seems to provide a vital ecological protection for the biocrusts, allowing them to be active only under relative low salinity after hydration. In the induced biocrusts, cyanobacteria were always the dominant organisms (even 18 years since inoculation), whereas under elevated salinity, cyanobacterial communities with lower relative abundance had a higher survival percentage of M. steenstrupii and Phormidium sp. Altogether, our results showed that both prokaryotic and eukaryotic communities have obvious salinity preference in the cyanobacteria-induced biocrusts. These results suggest that cyanobacterial inoculation strategies can be adapted in response to the soil salinity conditions. Furthermore, a combined inoculation of M. steenstrupii and Phormidium sp. appears to be favorable approach that is expected to improve cyanobacterial inoculation technology in saline soils.
