A Zr‐based metal‐organic framework PCN‐777 with good biocompatibility was synthesized by solvothermal method using 2,4,6‐Tris(4‐Carboxyphenyl)‐1,3,5‐Triazine and ZrOCl2 ⋅ 8H2O as raw materials. Its structure was identified and the results showed that it had good crystallinity. Then the anti‐inflammatory drug ibuprofen sodium was loaded into PCN‐777 to establish an IBUNa@PCN‐777 compound and the maximum loading capacity was tested. When the mass ratio of IBUNa to PCN‐777 was 4 : 1, the loading of IBUNa in IBUNa@PCN‐777 reached the maximum value 72.53 wt %. Biocompatibility investigation indicated PCN‐777 and IBUNa@PCN‐777 had good biocompatibility. Moreover, the drug release behavior of IBUNa@PCN‐777 testified the drug release at pH 7.4(>80 %) was greater than that at pH 2.0(<20 %). indicated that IBUNa@PCN‐777 had good pH response in simulated digestive environments. A Zr‐based metal‐organic framework was synthesized by solvothermal method. It was designated as UiO‐67(bpy) since its structure matched well with UiO‐67 according to the X‐ray diffraction results. The cytotoxicity assay of UiO‐67(bpy) was measured and it revealed that UiO‐67(bpy) had excellent biocompatibility. UiO‐67(bpy) was used to load anti‐inflammatory drugs, and the loading capacity of diclofenac sodium (DS) of UiO‐67(bpy) reached 30.08 wt %. Furthermore, the DS@UiO‐67(bpy) exhibited a pH‐controlled drug deliver behavior. The DS released in simulated intestinal environment (pH 7.4) was about 20 times of that in simulated gastric environment (pH 2.0) after 12 h. Drug delivery carriers with a high drug loading capacity and biocompatibility, especially for controlled drug release, are urgently needed due to the side effects and frequently dose in the traditional therapeutic method. In our work, a Zr-based metal-organic framework named ZJU-801, which is isoreticular with NU-801, has been designed and further demonstrated as an excellent drug delivery system (DDS) with a high drug loading of 41.7 %. Such a high drug loading capacity may be ascribed to the appropriate match of the size and the large pore volume of this kind of Zr MOF material. Compared with DS@NU-801, this DDS has successfully achieved on-command heating-activated drug release, which was probably attributed to the bulkier ligand, the better stability, and the intense π-π interaction between ZJU-801 and diclofenac sodium (DS) demonstrated comprehensively by SEM, powder X-ray diffraction (PXRD), FTIR and 13C solid-state NMR spectroscopy as well as computer simulations. It is worth noting that premature drug release was avoided effectively without any complicated post-modifications. The low cytotoxicity and good biocompatibility of our DDS were certificated by the in vitro favorable results from an MTT assay, a WST-1 assay, and confocal microscopy imaging. A biocompatible Zr-based metal-organic framework UiO-66-PDC was synthesized basing on 2,5-pyridinedicarboxylic acid as the ligand. Powder X-ray diffraction (PXRD) was used to characterize the structure and estimate the water stability, exhibiting the ideal structure and the excellent water stability of UiO-66-PDC. The anti-inflammatory drug ibuprofen sodium (IBUNa) was selected and loaded into UiO-66-PDC to construct an oral drug delivery system. The loading amount of IBUNa reached to about 27.60 ​wt%. The drug release behaviors of MOF were investigated and the material exhibited pH-response release which the drugs were delivered more in intestinal environment (pH 7.4) than that in gastric environment (pH 2.0). A metal–organic framework HKUST-1 with admirable biocompatibility was selected to load antibiotic tetracycline hydrate (TCN) obtaining TCN@HKUST-1, which had excellent antibacterial activity against E.coli and S.aureus. TCN@HKUST-1 revealed utilizable potentiality in the field of biological antibacterial. In this work, metal–organic framework HKUST-1 with excellent biocompatibility was synthesized to entrap the antibacterial tetracycline hydrate (TCN) obtaining TCN@HKUST-1 system. The drug loading amount of the system was 54.90 wt%. TCN@HKUST-1 showed excellent antibacterial effects against both Staphylococcus aureus and Escherichia coli due to the antibacterial Cu2+ and TCN, revealing utilizable potentiality in the field of antibacterial application.
