Trabalho de Projeto do Mestrado Integrado em Engenharia Biomédica apresentado à Faculdade de Ciências e Tecnologia Os sistemas de libertação controlada de fármacos à base de polímeros têm sido estudados, por contribuírem para uma maior especificidade, redução da toxicidade, e melhorarem as características de solubilidade dos fármacos com características hidrofóbicas. O fenómeno de self-assembly de polímeros tem papel de destaque por permitir o processo a encapsulação do fármaco. Para um efetivo sistema libertador é necessário também garantir a estabilidade do mesmo desde a administração até à libertação do fármaco, por ação de um estímulo externo, de forma rápida e eficiente. A biocompatibilidade e biodegradabilidade dos copolímeros é também fundamental para a eficácia do sistema formado. Devido à sua inocuidade na zona do visível e capacidade de penetração tecidular, a luz como estímulo externo, revela-se uma estratégia muito interessante para provocar a destruição de nanopartículas.Assim, a presente dissertação descreve um sistema de libertação controlada à base de copolímeros de bloco PEG-PLA, com um segmento sensível ao oxigénio singuleto que tem a capacidade de encapsular e libertar a doxorrubicina usada como fármaco. Aqui, as nanopartículas do tipo micelar, pela ação das porfirinas, reagem à luz de elevado comprimento de onda (superior a 600 nm) de modo a libertar o material encapsulado de forma rápida. Assim sendo, a este processo de libertação poderá trazer benefícios ao nível da seletividade de fármacos anti tumorais com toxicidade elevada, permitindo que a sua especificidade seja alcançada num alvo específico que seja irradiado. Os objetivos do projeto centram-se no desenvolvimento de copolímeros de bloco unidos pelo segmento sensível ao oxigénio singuleto, SegS, procedido pela análise da capacidade de self-assembly dos copolímeros preparados. Posteriormente determinou-se a capacidade de encapsulação das nanoestruturas e realizou-se o estudo de libertação da doxorrubicina após incidência de luz vermelha.Os resultados sugerem que as nanopartículas desenvolvidas são eficientes no sentido em que apresentaram baixa polidispersividade e capacidade de encapsulação da doxorrubicina segundo o método de diálise. Após irradiação por laser de 650 nm, as nanopartículas PEG5k-seg-PLA1k, evidenciaram uma taxa de cinética de libertação significativa nos instantes iniciais que tornam o sistema de libertação de fármacos promissor na terapia fotodinâmica. Controlled drug delivery systems based on polymers have been studied because they increase specificity, reduce toxicity and improve the solubility characteristics of drugs with hydrophobic characteristics. The self-assembly technique of polymers stands out because it allows the encapsulation of the drug. For an effective release system, it is also necessary to guarantee the drug stability from the administration until the drug release, by the action of an external stimulus, quickly and efficiently. The copolymers biocompatibility and biodegradability is also important to the effectiveness of the system. Due to its innocuity in the visible zone and capacity of tissue penetration, light as an external stimulus, proves to be a very interesting strategy to cause the destruction of nanoparticles.Thus, the present dissertation describes a controlled release system based on PEG-PLA block copolymers with a singlet oxygen sensitive segment that has the ability to encapsulate and release doxorubicin used as a drug. Here, micellar nanoparticles, by the action of porphyrins, react to high wavelength light (greater than 600 nm) in order to release the encapsulated material quickly. Thus, this release process may brings benefits to the level of selectivity of antitumor drugs with high toxicity, allowing their specificity to be achieved in a specific target that is irradiated.The objectives of this project are the development of block copolymers bound by the singlet oxygen-sensitive segment, SegS, carried out by analyzing the self-assembly capacity of the prepared copolymers. Subsequently, the nanostructures were encapsulated and the doxorubicin release study was performed after red light.The results suggest that the developed nanoparticles are efficient since they presented low polydispersity and doxorubicin encapsulation capacity according to the dialysis method. After 650 nm laser irradiation, the PEG5k-seg-PLA1k nanoparticles evidenced a significant rate of release kinetics at the initial instants that makes the drug delivery system promising in photodynamic therapy.
