Platelets are specialized blood cells that play central roles in physiologic and pathologic processes of hemostasis, inflammation, tumor metastasis, wound healing, and host defense. Activation of platelets is crucial for platelet function that includes a complex interplay of adhesion and signaling molecules. This article gives an overview of the activation processes involved in primary and secondary hemostasis, for example, platelet adhesion, platelet secretion, platelet aggregation, microvesicle formation, and clot retraction/stabilization. In addition, activated platelets are predominantly involved in cross-talk to other blood and vascular cells. Stimulated "sticky" platelets enable recruitment of leukocytes at sites of vascular injury under high shear conditions. Platelet-derived microparticles as well as soluble adhesion molecules, sP-selectin and sCD40L, shed from the surface of activated platelets, are capable of activating, in turn, leukocytes and endothelial cells. This article focuses further on the new view of receptor-mediated thrombin generation of human platelets, necessary for the formation of a stable platelet–fibrin clot during secondary hemostasis. This study investigated the interactive effects of drought stress with biochar on plant growth, the physiological and biochemical attributes of soybean seedlings. A pot trial tested the effect of amending a loamy sandy soil with biochar at three doses: 0%, 3% and 6% in the alleviation of drought stress, imposed by adjusting soil moisture content at three levels (in terms of soil field capacity, FC): 100% FC, 75% FC and 50% FC. Water deficit negatively influenced the growth of soybean seedlings and chlorophyll content of leaves, particularly in non-amended plants whereas biochar amendment boosted the growth and chlorophyll content, particularly under drought stress. Drought stress elevated malondialdehyde (MDA), hydrogen peroxide (H2O2), proline, sucrose and abscisic acid (ABA) contents as well as the antioxidant enzyme activity [peroxidase (POD), catalase (CAT) and superoxide dismutase (SOD)] of soybean leaves but reduced the contents of indole acetic acid (IAA) and gibberellic acid (GA); biochar amendment counteracted the impact of drought. The anaerobic capacity (AC[La]+EPOCfast) method is of interest as it can differentiate between the contributions of phosphocreatine (PCr) and glycolytic energy, assess the anaerobic metabolism during a single effort, and evaluate various exercise modalities. However, no previous studies have investigated whether the superiority of the PCr and glycolytic systems can be assessed in athletes with high anaerobic capacity. This study aimed to compare the AC[La]+EPOCfast method in seven sprinters (SP) and seven middle-distance runners (MD). The participants underwent a graded exercise test and constant-load exercise at 115% ˙VO2max until exhaustion using a bicycle ergometer. AC[La]+EPOCfast was calculated as the sum of oxygen equivalents from the phosphagen (EPCr) and glycolytic (ELa) pathways. This study reveals that there was no significant difference in AC[La]+EPOCfast or maximal accumulated oxygen deficit (MAOD) between the two groups. Although EPCr was not significantly different between groups, ELa was significantly higher in the SP group than in the MD group. In conclusion, the AC[La]+EPOCfast method can be used to evaluate high and low anaerobic capacities. However, further studies on athletes with high aerobic capacity are required. Legumes being a major source of protein have a wide range of economic importance. But the major constraints in growing legumes are poor germination, seedling emergence, non uniform stand establishment and crop mortality leading to lower pulse productivity. Thus, pre-sowing seed treatments are critical parameters which attribute to efficient plant growth and high yield. Uniform seed germination, seedling growth and uniform establishment are the critical stages during the growth of crops. Seed priming is a very promising presowing treatment employed to improve seed germination, better crop establishment, enhance the seed quality and induces tolerance against biotic and abiotic stresses in plants thereby providing a balanced healthy diet to the citizens. Priming is a cost effective and feasible strategy involving controlled hydration of seeds followed by drying to stimulate the pre-germinative metabolic activities to occur promoting rapid germination of seedlings, break dormancy and enhance crop yield. Enzymatic, metabolic and biochemical processes of the primed seeds attributes to rapid and uniform seedling emergence. Seed priming methods comprises of conventional methods including hydro priming, osmopriming, biopriming, chemical priming and the advanced methods including nano priming and priming with physical agents. Legumes have important nutritional and economic values, but their production faces continuous cropping obstacles that seriously affect their yield formation. In order to reduce the negative impact of the continuous cropping obstacles of legumes, it is necessary to understand the response mechanisms of legumes to continuous cropping, the causes of continuous cropping obstacles and the measures to alleviate continuous cropping obstacles. This review aimed to identify the current knowledge gap in the field of continuous cropping obstacles of legumes and provide direction and focus for future research. The continuous cropping obstacles of legumes start with soil degradation, leading to oxidative stress in the plants. This triggers the expression of plant-hormone- and signal-molecule-related genes, activating the defense system and causing continuous cropping obstacles. Although there has been progress in researching these challenges in legume crops, many questions remain. We believe that the exploration of molecular mechanisms of legume crops responding to continuous cropping, rhizosphere signal exchange and soil environment repair mechanisms after long-term continuous cropping of soybean, and the excavation of candidate genes and functional loci related to continuous cropping obstacles in legume crops are breakthroughs for proposing effective continuous cropping obstacle management strategies in the future. Rice (Oryza sativa L.) is a primary energy food for the Asian population. One of the greatest constraints in rice production is soil salinity because rice is very susceptible to salt. Meanwhile, many agricultural lands in Asia are in saline areas. It is important to identify and develop salt-tolerant rice varieties that highly adapt to Asian climates. By combining morphological, physiological, and biochemical assessments for screening the salt tolerance of 116 Asian rice cultivars, we were able to classify them into tolerant, moderate, and sensitive rice cultivars under salinity stress conditions and also understand salt tolerance mechanisms. The rice cultivars that are salt-tolerant include Pokkali from India, TCCP 266 and IR 45427 from the Philippines, and Namyang 7 from Republic of Korea. However, salt-sensitive rice varieties like IR29 and IR58 are from the Philippines, and Daegudo and Guweoldo are from Korea. The salt-tolerant varieties showed signs of tolerance, including a lower percent reduction in germination percentage, root length, root fresh weight, shoot length, plant biomass, and chlorophyll content. In order to maintain the cellular osmotic balance under saline conditions, the salt-tolerant varieties exhibited less membrane damage, a lower Na/K ratio, high proline and sugar accumulation, and lower levels of malondialdehyde (MDA) and hydrogen peroxide (H2O2). Pokkali from India, TCCP 266 and IR 45427 from the Philippines, and Namyang 7 from Republic of Korea are recommended as valuable germplasm resources for Asian rice breeding programs in saline agricultural areas. Background/Aims: Many questions in cancer biology remain unanswered. Perhaps the most important issues remaining to be addressed focus on the molecular basis of carcinogenesis. Today's cancer focus lies on genetics and gene expression, which is unlikely to explain the true cause of most cancers or lead to a cure. Methods: Earlier, we provided a plausible mechanism for this process, specifically, that most cancers develop in response to pathogenic stimuli that induce chronic inflammation, fibrosis, and remodeling of the cellular microenvironment. Collectively, these changes generate a precancerous niche (PCN) in which fibrosis and remodeling are ongoing secondary to persistent inflammation, followed by the deployment of a chronic stress escape strategy (CSES). If the CSES is unsuccessful, the cell undergoes a normal cell to cancer cell transformation (NCCT). Results: Here, we highlight the critical role of fibroblasts as the first cells to undergo neoplastic transformation to a cancerous phenotype which is based on several critical findings. First, persistent disruption of homeostatic crosstalk increases lysyl oxidase activity and lysine oxidation which leads to increased collagen stiffness and decreased elasticity. If unresolved, chronic tissue stress will lead to an escape strategy that involves the recruitment of fibroblasts and fibrocytes from the bone marrow as well as cells undergoing an epithelial-mesenchymal transition (EMT). This yields a heterogeneous pool of cells that express both epithelial and mesenchymal markers and that will ultimately differentiate into cancer-associated fibroblasts (CAFs). Finally, CAFs undergo a mesenchymal-epithelial transition (MET) and express epithelial markers that facilitate their integration into the target tissue. In all fields of biology, understanding technical terminology is a challenge for students. In many cases, this may distract them from focusing on fundamental processes and concepts. Across the biology subfields, much of the vernacular shares similar etymology and morphology. However, students lack the exposure necessary to identify these key features, which often explain the meaning of terms without requiring any context at all. Therefore, instead of encouraging students to memorize many terms independently, it could be more beneficial to show them how words are constructed. Here, I propose an activity designed to help students recognize terms that may be connected, understand how vocabulary is often constructed to reflect its idea, and develop comfortability using these terms themselves in discussions. Through a guided group activity, students will have a chance to break down terms they have previously encountered and to draw connections between novel words. If students are capable of relating words to each other before even knowing what they mean, they may learn more effectively. Without being intimidated by enigmatic vocabulary, they can focus on broader concepts. In addition, when students understand how biological terminology is constructed, they may even dissect new words without needing the context surrounding them. Macrofungi are well known as mushrooms, which belong mostly to Basidiomycota with a few from Ascomycota, and up to now, around 40,000 species have been described. In people's lives, macrofungi are closely related to our economic activities, especially for food and medicine. "One meat, one vegetable and one mushroom" has become a healthy and fashionable dietary structure, and the global edible mushroom production and cultivating area are steadily rising. On the other hand, a large number of mushroom species and new active components have been found, and have become one of the driving forces of innovation of drugs and health products, especially with the development of biochemistry, enzyme engineering, and genetic engineering. Thus, macrofungi in food, medicine, and other aspects have shown a broad prospect. In this Special Issue, research on new species and related molecular phylogenies, mechanisms of hyphae polar growing and basidiocarp formation, biochemistry of edible and medical mushrooms, and some important scientific questions related to the edible mushroom industry are presented, which also reflect the hot areas of common concern on edible and medicinal fungi. Previous research has explored the demands of amateur boxing-specific activity; however, no holistic review of the acute responses to such activity currently exists. This paper aimed to provide a systematic review of the available literature on the acute physiological, endocrine, biochemical, and performance responses to amateur boxing-specific activity. Following a search of EBSCOhost, SPORTDiscus, PubMed and Google Scholar databases, 25 studies were identified as meeting the inclusion criteria for the review. The methodological quality of the included studies was assessed via a modified Downs and Black checklist. Random-effects meta-analysis of standardised mean differences (SMD) revealed large (SMD = 4.62) increases in pre-post blood lactate (BLa), cortisol (SMD = 1.33), myoglobin (Mb) (SMD = 1.43), aspartate transaminase (AST) (SMD = 1.37), and alanine aminotransferase (ALT) (SMD = 0.97), in addition to moderate increases in creatine kinase (CK) (SMD = 0.65). Small pre–post increases in counter-movement jump (CMJ) height (SMD = 0.33) were observed. Consistently greater pre–post alterations were observed in competitive bouts, followed by sparring, and less so in boxing-specific simulations. Considerable physiological, endocrine, and biochemical responses are elicited following amateur boxing. Interestingly, neuromuscular and task-specific performance may not deteriorate following boxing-specific activity. The findings of the review may assist in the designing and periodising of boxing-specific training, dependent on the desired physical adaptations, training phase, and recovery status of the amateur boxer. Highlights Amateur boxing elicits a considerable acute physiological, hormonal, and biochemical response. Such responses are typically greater in competitive bouts, followed by sparring, and less so in simulated activity. The considerable demands of amateur boxing-specific activity do not appear to negatively affect neuromuscular or task-specific performance. Amateur boxers may be conditioned to preserve performance despite the acute demands of the sport, or the lack of performance decrement may reflect the short duration of amateur boxing. Mitochondrial β-oxidation is the most prominent pathway for fatty acid oxidation but alternative oxidative metabolism exists. Fatty acid ω-oxidation is one of these pathways and forms dicarboxylic acids as products. These dicarboxylic acids are metabolized through peroxisomal β-oxidation representing an alternative pathway, which could potentially limit the toxic effects of fatty acid accumulation. Although dicarboxylic acid metabolism is highly active in liver and kidney, its role in physiology has not been explored in depth. In this review, we summarize the biochemical mechanism of the formation and degradation of dicarboxylic acids through ω- and β-oxidation, respectively. We will discuss the role of dicarboxylic acids in different (patho)physiological states with a particular focus on the role of the intermediates and products generated through peroxisomal β-oxidation. Although a substantial body of evidence suggests that large and old trees have reduced metabolic levels, the search for the causes behind this observation has proved elusive. The strong coupling between age and size, commonly encountered in the field, precludes the isolation of the potential causes. We used standard propagation techniques (grafting and air‐layering) to decouple the effects of size from those of age in affecting leaf structure, biochemistry and physiology of two broadleaved trees, Acer pseudoplatanus (a diffuse‐porous species) and Fraxinus excelsior (a ring‐porous species). The first year after establishment of the propagated plants, some of the measurements suggested the presence of age‐related declines in metabolism, while other measurements either did not show any difference or suggested variability across treatments not associated with either age or size. During the second year after establishment, only one of the measured properties (specific leaf area) continued to show some evidence of an age‐mediated decline (although much reduced compared to the field), whereas, for some properties (particularly for F. excelsior), even the opposite trend of age‐related increases was apparent. We concluded that (1) our plants suffered from grafting shock during year 1 and they gradually recovered during year 2; (2) the results over 2 years do not support the statement that age directly mediates ageing in either species but instead suggest that size directly mediates ageing processes; and (3) neither shoots nor roots of A. pseudoplatanus showed any evidence of senescence. The multiannual delayed gametophyte of kelp species maintains continuous vegetative growth under unsuitable conditions and can restart gametogenesis under suitable conditions. However, the processes and mechanisms of this phenomenon are still poorly understood. To gain insight into the processes and mechanisms of gametogenesis in multiannual delayed gametophytes of the kelp Saccharina japonica, the ultrastructure changes, photosynthetic and biochemical responses, and dynamics of gene expression were investigated. Data were collected at four time points (0, 3, 6, and 9 days) during gametogenesis. After gametogenesis induction there were notable changes in the ultrastructure, including increases in mitochondria and the Golgi bodies, and changes in chloroplast and endoplasmic reticulum structures. The contents of abscisic acid and H2O2 as well as the activities of pivotal respiratory enzymes (malic dehydrogenase and phosphofructokinase) and key antioxidant enzymes (catalase and ascorbate peroxidase) all increased significantly after 3 days of gametogenesis induction, implying that these changes were closely related to the gametophyte's transition from vegetative growth to gametogenesis. Notably, photosynthetic and respiratory indicators both showed significantly upregulated trends after 6 days of induction, indicating that the gametophyte's metabolism was most active during transformation from vegetative growth into gametogenesis. Time-specific DEGs (differentially expressed genes) were identified during the vegetative and reproductive stages of gametophytes. Gene ontology analysis indicated that "cell part", "cellular process" and "binding" represented the largest categories in the DEG datasets. Moreover, the predominant transcription factor families were bHLH, MYB, and C3H in the DEG datasets. There were 8 genes related to peroxisomes, 12 genes related to proteasomes, 4 genes related to oxidative phosphorylation, and 59 genes related to ubiquitin-mediated proteolysis identified as up- or downregulated DEGs at the two key stages during gametogenesis, the initial developmental stage and the oogonia formation stage. [Objectives] This study was conducted to explore how to improve the waterlogging tolerance of red-seed watermelon through grafting, to provide a theoretical basis for its cultivation in rainy season. [Methods] The effects of flooding stress on the growth and root physiological and biochemical characteristics of grafted and own-rooted red-seed watermelon seedlings were studied using Luffa as rootstocks and "Zhongxin 1" red-seed watermelon as scions. [Results] After flooding stress, the biomass and root activity of grafted seedlings of red-seed watermelon were significantly higher than those of own-rooted seedlings. With the prolongation of flooding stress time, the soluble sugar and proline content showed a trend of first increasing and then decreasing, and the grafted seedlings had a larger increase and a smaller decrease, and were always significantly higher than own-rooted seedlings in the same period. The content of malondialdehyde in the root system of grafted seedlings increased first and then decreased, while it continued to increase in own-rooted seedlings, and the increase in own-rooted seedlings was significantly greater than that in grafted seedlings during the same period. This is a case series about retinal arterial macroaneurysms in three patients recovered from COVID-19. None of them had previous past ocular and systemic history. The first patient was a 47-year-old man, with best-corrected visual acuity of 20/20 in both eyes. He presented a cotton wool spot in the right eye and two peripheral exudative retinal arterial macroaneurysms in the left eye. Laser photocoagulation was performed and best-corrected visual acuity remained stable. The second one was a 62-year-old- woman with acute visual loss in the right eye (best-corrected visual acuity of 20/400). There was vitreous hemorrhage and a peripheral retinal arterial macroaneurysm in the right eye. Laser photocoagulation was performed and best-corrected visual acuity improved to 20/20. The third patient was a 54-year- old woman, with best-corrected visual acuity of 20/20 in the right eye and 20/600 in the left eye. Fundus examination showed multiple layer hemorrhage in the macular region and retinal arterial macroaneurysms in the superior temporal branch. The cases underwent fluorescein angiography, which confirmed the diagnosis of retinal arterial macroaneurysms. [ABSTRACT FROM AUTHOR]

The aim of this study was to investigate the effects of bottled water left in the car and those left in the refrigerator on the physiological and biochemical parameters of male rats. Eighteen male albino rats were randomly divided into control, warmed bottled water (WBW), and cooled bottled water (CBW) groups. The concentrations of bisphenol A (BPA) and antimony (Sb) in the water samples were measured. Results showed that the levels of BPA and Sb were higher in bottled water left in the car and those kept at room temperature (control), relative to the bottled water samples kept in the fridge. Bodyweight, some metabolic indicators, cTnT and CRP levels, hematological parameters, testosterone level, and sperm quality were significantly affected by storage conditions. These results indicate that storage conditions can potentially affect the quality of bottled water, which in turn can affect the biological parameters of living organisms. Moreover, since the concentrations of BPA and Sb were least in bottled water kept in the refrigerator, we recommend that manufacturers and households should endeavor to keep bottled water under cold storage. Compromised productivity and yield loss due to inadequate nutrient abundance or inefficient mineral absorption from the soil are one of the most prevalent agricultural concerns across the world. Essential minerals, required in trace amounts for optimum plant growth and development, are termed as micronutrients. Due to low abundance of these vital elements (iron, zinc, copper, manganese, boron, nickel, molybdenum and chlorine), significant stretches of agricultural land often have limited supply of these micronutrients. As a result, plants, crops and vegetables grown in such soil exhibit unpredictable anatomical, biochemical and metabolic abnormalities associated with deteriorated physiological alterations. This communication details out the genomic, proteomic and metabolomic dynamism mediated by various plant species exposed to micronutrient starvation or grown under micronutrient-limiting environment. Furthermore, deficiency of vital trace elements inhibits the activity of crucial enzymes associated with detoxification of oxidants and regulation of important physiological processes like photosynthesis, respiration, nitrogen assimilation, sugar metabolism, etc. Thus, these plants experience an inherent oxidative stress and are more susceptible to multiple abiotic stresses due to inefficient defence machinery. The differential physiological adaptations mediated under such variable microelement deficiency have also been critically delineated. A pot study was conducted to determine the effects of the combination of plant growth promoting rhizobacteria (PGPR) and biochar on the growth, physiological, and biochemical characteristics of eggplant seedlings under salinity stress. The greenhouse experiment included two salinity levels of NaCl [S0 (0 mM NaCl) and S1 (100 mM NaCl)], three biochar levels [B0 (non-biochar), B1 (5%) and B2 (10%)] and two PGPR [R0 (non-PGPR), R1 (combination of Bacillus megaterium TV-6D, Paenibacillus polymyxa KIN-37, and Pantoea agglomerans RK92). Results showed that plant growth, relative leaf water content (LRWC), and chlorophyll content of eggplant seedlings decreased significantly, while malondialdehyde (MDA), hydrogen peroxide (H2O2), proline, sucrose and ABA content, and electrolyte leakage (EL) increased significantly with increase in salinity levels. Biochar and PGPR applications mitigated the negative influence of salinity stress on plant growth and physiological and biochemical characteristics of eggplant seedlings, enhancing chlorophyll content, plant nutrient element uptake, and antioxidant enzyme activity. The enhanced salinity tolerance due to biochar and PGPR applications could be associated with a significant reduction in Na and Cl uptake, MDA, H2O2 and EL and an increase in LRWC, chlorophyll content, antioxidant activity and plant nutrient element uptake. Therefore, it can be concluded that combining biochar and PGPR could be used to minimize the detrimental impacts of salinity stress conditions in eggplant seedlings. [ABSTRACT FROM AUTHOR]

Geoffrey Burnstock was a biomedical scientist who gained renown for his discovery that adenosine 5′-triphosphate (ATP) functions as an extracellular signalling molecule. Born in London and educated at King's and University Colleges, he did postdoctoral work at Mill Hill and Oxford. He moved in 1959 to the Department of Zoology at the University of Melbourne because he sensed there a greater freedom to challenge established thinking in physiology. His group found that transmission from sympathetic and parasympathetic autonomic nerves to smooth muscle was in some places not mediated by the accepted chemical messengers (noradrenaline and acetylcholine). He amassed evidence that ATP was this non-adrenergic, non-cholinergic (NANC) transmitter, using biochemical, histological and electrophysiological approaches: heretically, he styled this 'purinergic transmission'. Geoff further upset dogma in the 1970s by proposing 'co-transmission' in which some nerves released ATP in addition to either noradrenaline or acetylcholine. He distinguished pharmacologically P1 receptors (activated best by adenosine and blocked by xanthines) and P2 receptors (activated best by purine nucleotides such as ATP) and he proposed in 1985 that the latter embraced P2X (ion channel) and P2Y (G protein-coupled) subtypes: about ten years later these categories were substantiated by cDNA cloning. From 1975 until his retirement in 1997, Geoff was head of Anatomy and Embryology at University College London (UCL), which he developed energetically into a large and strong research department. Later, as head of the Autonomic Research Institute at the Royal Free (part of UCL), he continued to collaborate extensively, and founded several journals and international professional societies. Clusia is a remarkable genus of Neotropical woody plants as its members engage in either C3 photosynthesis or employ, to varying degrees, crassulacean acid metabolism (CAM) photosynthesis. Information about the evolutionary history of CAM in Clusia is scarce. Restriction site-associated sequencing of 64 species (20% of the genus) provided strong support for most of the previously recognized nine lineages. Ancestral reconstruction using maximum parsimony or maximum likelihood under a one-rate model suggested that CAM evolved at least four times independently from a most recent common ancestor (MRCA) with C3, whereas a maximum likelihood two-rate model suggested that CAM was already present in the MRCA followed by reversions to C3 in several lineages. Phylogenetic generalized least square analysis assessed variation in seven leaf anatomical traits and CAM activity measured as δ 13C. Results indicate that CAM is highly correlated with palisade mesophyll layer thickness and cell size. In addition, correlation between 19 bioclimatic variables and δ 13C was evaluated. It was found that CAM is positively correlated with habitats with a more severe dry season and greater precipitation seasonality. Since CAM is weakly and/or only periodically expressed in many Clusia spp. and thus not readily reflected in δ 13C, future analysis of phylogenetically-informed CAM expression in Clusia must include physiological measurements such as CO2 exchange and/or diel changes in leaf acidity for each species under investigation. Transglutaminases (TGases) are calcium-dependent enzymes that catalyse an acyl-transfer reaction between primary amino groups and protein-bound Gln residues. They are widely distributed in nature, being found in vertebrates, invertebrates, microorganisms, and plants. TGases and their functionality have been less studied in plants than humans and animals. TGases are distributed in all plant organs, such as leaves, tubers, roots, flowers, buds, pollen, and various cell compartments, including chloroplasts, the cytoplasm, and the cell wall. Recent molecular, physiological, and biochemical evidence pointing to the role of TGases in plant biology and the mechanisms in which they are involved allows us to consider their role in processes such as photosynthesis, plant fertilisation, responses to biotic and abiotic stresses, and leaf senescence. In the present paper, an in-depth description of the biochemical characteristics and a bioinformatics comparison of plant TGases is provided. We also present the phylogenetic relationship, gene structure, and sequence alignment of TGase proteins in various plant species, not described elsewhere. Currently, our knowledge of these proteins in plants is still insufficient. Calcium (Ca2+) is a ubiquitous and fundamental signaling component that is utilized by cells to regulate a diverse range of cellular functions, such as insulin secretion from pancreatic β-cells of the islets of Langerhans. Cyclic ADP-ribose (cADPR), synthesized from NAD+ by ADP-ribosyl cyclase family proteins, such as the mammalian cluster of differentiation 38 (CD38), is important for intracellular Ca2+ mobilization for cell functioning. cADPR induces Ca2+ release from endoplasmic reticulum via the ryanodine receptor intracellular Ca2+ channel complex, in which the FK506-binding protein 12.6 works as a cADPR-binding regulatory protein. Recently, involvements of the CD38-cADPR signal system in several human diseases and animal models have been reported. Simple Summary: Drought is considered a critical threat to crop growth and sustainable agriculture worldwide, and it also greatly impacts insect development and population growth. Brown planthopper (BPH), Nilaparvata lugens (Stål), is the predominant rice crop pest in China, and the damaging effects of BPH are enhanced by its strong migratory and reproductive capacities. Our results provide useful information about the effect of drought stress on the poor population growth and negative physiological changes in BPH. Negative changes to water balance and osmotic pressure can cause a decline in the quality of BPH; the GST content of BPH feeding on drought-stressed rice was significantly higher than BPH feeding on non-stressed control plants, and the length of flight muscle sarcomeres and mitochondrial content were decreased in BPH feeding on drought-stressed rice. These findings suggest that water management greatly impacts the physiology and population growth of BPH, and provide a basis for understanding physiological and population-wide responses in BPH during drought stress, which may be helpful in understanding the relationship between drought stress and BPH infestation. Some studies have demonstrated increased reproductive capacity in drought-stressed insects; however, physiological changes in the brown planthopper (BPH), Nilaparvata lugens (Stål), during periods of drought are unclear. In this study, BPH fed on drought- stressed rice had lower population numbers than BPH feeding on non-stressed rice. Water content, osmotic pressure of hemolymph and total amino acid content of BPH were significantly lower when BPH fed on drought-stressed rice compared to the non-stressed control; however, glucose content and glutathione S-transferase (GST) activity were significantly higher in BPH fed on drought-stressed rice. The expression of Vitellogenin and Exuperantia in BPH fed on drought-stressed rice was higher than that in BPH feeding on non-stressed control plants. The size of myofibrils and the abundance of mitochondria in BPH flight muscles were significantly lower in BPH fed on drought-stressed rice compared to non-stressed plants. These results indicate that water management impacts the physiology of BPH, which may be useful in understanding the relationship between drought stress and this damaging herbivore. The auxin-inducible degradation system in C. elegans allows for spatial and temporal control of protein degradation via heterologous expression of a single Arabidopsis thaliana F-box protein, transport inhibitor response 1 (AtTIR1). In this system, exogenous auxin (Indole-3-acetic acid; IAA) enhances the ability of AtTIR1 to function as a substrate recognition component that adapts engineered degron-tagged proteins to the endogenous C. elegans E3 ubiquitin ligases complex [SKR-1/2-CUL-1-F-box (SCF)], targeting them for degradation by the proteosome. While this system has been employed to dissect the developmental functions of many C. elegans proteins, we have found that several auxin-inducible degron (AID)-tagged proteins are constitutively degraded by AtTIR1 in the absence of auxin, leading to undesired loss-of-function phenotypes. In this manuscript, we adapt an orthogonal auxin derivative/mutant AtTIR1 pair [C. elegans AID version 2 (C.e.AIDv2)] that transforms the specificity of allosteric regulation of TIR1 from IAA to one that is dependent on an auxin derivative harboring a bulky aryl group (5-Ph-IAA). We find that a mutant AtTIR1(F79G) allele that alters the ligand-binding interface of TIR1 dramatically reduces ligand-independent degradation of multiple AID*-tagged proteins. In addition to solving the ectopic degradation problem for some AID-targets, the addition of 5-Ph-IAA to culture media of animals expressing AtTIR1(F79G) leads to more penetrant loss-of-function phenotypes for AID*-tagged proteins than those elicited by the AtTIR1-IAA pairing at similar auxin analog concentrations. The improved specificity and efficacy afforded by the mutant AtTIR1(F79G) allele expand the utility of the AID system and broaden the number of proteins that can be effectively targeted with it. Dementias, including the type associated with Alzheimer's disease (AD), are on the rise worldwide. Similarly, type 2 diabetes mellitus (T2DM) is one of the most prevalent chronic diseases globally. Although mechanisms and treatments are well-established for T2DM, there remains much to be discovered. Recent research efforts have further investigated factors involved in the etiology of AD. Previously perceived to be unrelated diseases, commonalities between T2DM and AD have more recently been observed. As a result, AD has been labeled as "type 3 diabetes". In this review, we detail the shared processes that contribute to these two diseases. Insulin resistance, the main component of the pathogenesis of T2DM, is also present in AD, causing impaired brain glucose metabolism, neurodegeneration, and cognitive impairment. Dysregulation of insulin receptors and components of the insulin signaling pathway, including protein kinase B, glycogen synthase kinase 3β, and mammalian target of rapamycin are reported in both diseases. T2DM and AD also show evidence of inflammation, oxidative stress, mitochondrial dysfunction, advanced glycation end products, and amyloid deposition. The impact that changes in neurovascular structure and genetics have on the development of these conditions is also being examined. With the discovery of factors contributing to AD, innovative treatment approaches are being explored. Investigators are evaluating the efficacy of various T2DM medications for possible use in AD, including but not limited to glucagon-like peptide-1 receptor agonists and peroxisome proliferator-activated receptor-gamma agonists. Furthermore, there are 136 active trials involving 121 therapeutic agents targeting novel AD biomarkers. Military personnel are required to complete physically demanding tasks when performing work and training, which may be quantified through the physical stress imposed (external load) or the resultant physiological strain (internal load). The aim of this narrative review is to provide an overview of the techniques used to monitor work and training load in military settings, summarise key findings, and discuss important practical, analytical, and conceptual considerations. Most investigations have focused upon measuring external and internal load in military training environments; however, limited data exist in operational settings. Accelerometry has been the primary tool used to estimate external load, with heart rate commonly used to quantify internal load. Supplemental to heart rate, psychophysiological and biochemical measures have also been investigated to elucidate aspects of internal load. Broadly, investigations have revealed that military training requires personnel to perform relatively large volumes of physical activity (e.g. averaging ∼15,000 steps·day−1) of typically low-moderate intensity activity (<6 MET), although considerable temporal and inter-individual variability is observed from these gross mean estimates. There are limitations associated with these measures and, at best, estimates of external and internal load can only be inferred. These limitations are particularly pertinent for military tasks such as load carriage and manual material handling, which often involve complex activities performed individually or in teams, in a range of operational environments, with multiple layers of protection, over a protracted duration. Allium mongolicum Regel is a high‐quality plant resource but highly perishable with short shelf life of 1–2 days. Changes in postharvest quality and microbiological population on A. mongolicum regulated by poly (butylene adipate‐co‐terephthalate)/poly (butylene succinate), poly (butylene adipate‐co‐terephthalate)/polycaprolactone, poly (butylene adipate‐co‐terephthalate)/poly (L‐lactide) and poly (butylene adipate‐co‐terephthalate)/polypropylene MAP were investigated during near‐freezing storage. The results showed all four MAPs could reduce the respiration rate, water loss, decay rate and malondialdehyde content significantly and maintain the morphology, color and typical flavor of A. mongolicum effectively which also had high level acceptable commercial score as well as ascorbic acid and chlorophyll contents. However, the preservation effect packed with poly (butylene adipate‐co‐terephthalate)/polypropylene was significantly better than the other three films in which it could establish an optimal atmospheric condition with O2 concentration of 0.34%–0.53% and CO2 concentration of 4.53%–6.13% and the A. mongolicum had the highest level of postharvest quality, the lowest level of physiological metabolism and surface microorganisms, and its shelf life could reach 18 days. Practical applications: Allium mongolicum Regel has delicious taste, high nutritional value, and bioactive ingredients good for human health but with short shelf life which is a great challenge to the storage and sales. Therefore, maintaining postharvest quality and extending shelf life are significant to the industry development for A. mongolicum. However, studies on its preservation are not available. Modified atmosphere packaging is a cheap, safe and effective method to keep produce fresh. In this study, the shelf life of A. mongolicum was extended significantly in the treatment of poly (butylene adipate‐co‐terephthalate)/polypropylene MAP. It provided an effective solution for the storage problem and had great potential in industry for A. mongolicum. Furthermore, it provided a theoretical basis for the storage and industry development by studying the correlations of the O2, CO2 and water vapor transmission rate of package film, O2, CO2 concentration in package and postharvest physiology and quality of A. mongolicum during storage. The experiment was implemented to assess the influence of dietary supplementation of laying quails with creatine monohydrate (CrM), L-carnitine (CAR) and their mixture (CrMCAR) as antioxidants against oxidative stress (OS) induced by 2.5 ppm lead acetate (LA) in drinking water on productive, physiological and microbial aspects. In total, 400 laying quail females at 10 wk of age were divided into a randomized design with 5 groups and 4 replicates of 20 birds each. Birds were fed ad libitum with a balanced diet for 8 wk. The control group was kept under no-stress conditions and was given fresh water without any additives (G1). While birds in other groups were exposed to OS induced experimentally by 2.5 ppm LA in drinking water with no feed additive (G2) or supplemented with 500 mg/kg CrM (G3) or 500 mg/kg CAR (G4) or combination of 250 mg/kg each of CrM and CAR (CrMCAR, G5) to feed mixture. Compared to G2, G5 demonstrated the reduction (P ≤ 0.05) of feed conversion ratio, feed intake, mortality and ileal total coliform, as well as serum and egg malondialdehyde and serum lipid hydroperoxide, uric acid, glucose, cholesterol, enzymatic activities (alanine aminotransferase, aspartate transaminase, alkaline phosphatase, creatine phosphokinase, γ-glutamyl transferase), and heterophils/lymphocytes ratio. In the meanwhile, there was an increase (P ≤ 0.05) in egg production, egg mass, and weight with the improvement of egg quality, serum sex hormones level and ileal lactic acid bacteria for G5 followed by G4 and G3. Moreover, G5 enhanced (P ≤ 0.05), the total antioxidant capacity of egg and serum glutathione, superoxide dismutase, catalase, glutathione peroxidase, protein and calcium levels. Therefore, dietary CrMCAR, CAR and CrM have analogous influence to control by improving the antioxidant and physiological parameters which resulted in better productive performance and egg characteristics of stressed quails. DEPTOR plays vital roles in the regulation of cell proliferation and survival by directly modulating the activity of mTORC1/2. However, the physiological role of DEPTOR in lung tumorigenesis, as well as its clinical significance, remains elusive. In this study, we revealed that decreased DEPTOR expression correlated with increased tumor size, poor differentiation, and worse survival in patients with lung cancer. DEPTOR depletion promoted cell proliferation, survival, migration, and invasion in human lung cancer cells. Mechanistically, DEPTOR bound to the kinase domain of EGFR via its PDZ domain to inactivate EGFR signal. Thus, DEPTOR depletion not only directly activated mTORC1/2, but also relieved the inhibition of EGFR to subsequently activate mTOR signals, leading to the induction of cell proliferation and survival. Additionally, activated EGFR-mTOR signals upregulated the expression of ZEB1 and SLUG to induce epithelial-mesenchymal transition, resulting in enhanced migration and invasion. Importantly, Deptor deletion accelerated KrasG12D;p53fl/fl-induced lung tumorigenesis and shortened mouse life span via the activation of EGFR-mTOR signals. Complete blood count, serum chemistry values, and biological reference intervals were compared between two age groups (34–49 and 84–120 days old) of healthy male and female laboratory raised natal multimammate mice (Mastomys natalensis). Blood was collected via cardiocentesis under isoflurane anesthesia. Data sets of machine automated complete blood counts and clinical chemistries were analyzed. Significant differences between sex and age groups of the data sets were defined. Physiology can either be a product of or affect the behavior and ecology of individual animals. Blood plasma chemistry contains many components that are indicative of several aspects of a bird's physiological state at the time of sampling. Plasma chemistry is routinely used in clinical settings. However, despite its potential application in ecological studies, few birds have published reference values for these parameters from wild populations. Here, we analyzed the blood plasma chemistry for 17 parameters in Short-toed Snake Eagles (Circaetus gallicus) captured in southern Spain. Most values did not differ between age classes and sexes. However, we found differences between our captive and wild populations of fledgling eagles. These differences arose primarily in parameters associated with nutritional condition, as we expected due to captive individuals being fed ad libitum prior to blood draws. We also compared urea levels and a body condition index with hatching date and found a much stronger relationship with urea than the body condition index. Our results demonstrate the necessity to obtain blood chemistry reference values from wild populations and not just captive individuals, and also illustrate how these parameters can complement other traditional data collected when handling birds in the field. Thermoregulation has substantial implications for human health. Traditional central space heating and cooling systems are less efficient due to wasted energy spent on the entire building and ignore individual thermophysiological comfort. Emerging textiles based on innovations in materials chemistry and physics, nanoscience, and nanotechnology have now facilitated thermoregulation in a far more personalized and energy-saving manner. In this tutorial review, we discuss the latest technological advances in thermoregulatory textiles. First, we outline the basic mechanisms behind the physiological chemistry processes for both internal and external thermoregulation in the human body. Then, we systematically elaborate on typical smart passive and active thermoregulatory textiles considering current working mechanisms, materials engineering towards practical applications. In light of burgeoning commercial trends, we offer important insights into green chemistry for the sustainable development of smart thermoregulatory textiles. Prospectively, we propose an autonomous textile thermoregulation system that could intelligently provide personalized thermophysiological comfort in a self-adaptive manner in the era of Internet of Things (IoT). The discussion of interdisciplinary interactions of energy, environmental science, and nanotechnology in this review will further promote development of the thermoregulatory textile field in both academia and industry, ultimately realizing personalized thermoregulation and a sustainable energy future. All vertebrates share a canonical retina with light-sensitive photoreceptors in the outer retina. These photoreceptors are of two kinds: rods and cones, adapted to low and bright light conditions, respectively. They both show a peculiar morphology, with long outer segments, comprised of ordered stacks of disc-shaped membranes. These discs host numerous proteins, many of which contribute to the visual transduction cascade. This pathway converts the light stimulus into a biological signal, ultimately modulating synaptic transmission. Recently, the zebrafish (Danio rerio) has gained popularity for studying the function of vertebrate photoreceptors. Effective treatment regimens for triple-negative breast cancer (TNBC) are relatively scarce due to a lack of specific therapeutic targets. Epidermal growth factor receptor (EGFR) signaling is highly active in TNBC and is associated with poor prognosis. Most EGFR antagonists, which significantly improve outcome in lung and colon cancer, have shown limited clinical effects in breast cancer. However, limiting EGFR expression in TNBC is a potential strategy for improving the clinical efficacy of EGFR antagonists. Here, we found that the gamma-aminobutyric acid type A receptor π subunit (GABRP), as a membrane protein enriched in TNBC stem cells, interacted with EGFR and significantly sustained its expression, resulting in stemness maintenance and chemotherapy resistance. Silencing GABRP induced down-regulation of EGFR signaling, which hindered cell stemness and enhanced sensitivity to chemotherapies, including paclitaxel, doxorubicin, and cisplatin. We also identified that retigabine, an FDA-approved drug for adjunctive treatment of seizures, increased the sensitivity of EGFR to gefitinib in gefitinib-resistant cells. Our findings show that GABRP can sustain the stemness of TNBC via modulating EGFR expression, suggesting that GABRP may be a potential therapeutic target that can address EGFR inhibitor resistance in TNBC. Plants of Bambusa vulgaris Schrad. ex Wendl are a viable option to generate environmental, social and economic benefits. However, there are problems in the propagation via organogenesis of this species such as the low in vitro multiplication coefficient, which limits its propagation. The research was aimed to determine the effect of 6-benzylaminopurine and the immersion time on the multiplication, morphophysiology and biochemistry of B. vulgaris shoots in temporary immersion systems. Morphophysiological and biochemical variables were evaluated in shoots grown at different BAP concentrations (0; 6.6; 13.2 and 26.4) at immersion times of one, two or three minutes, for 30 days of cultivation. In those systems, the lower concentration of BAP (6.6 µM) and immersion time of two minutes favored the emission of new shoots. It was associated with an increase in the content of total chlorophylls, lignin and total phenols in shoots. Based on these results, a protocol was established for the propagation via organogenesis of B. vulgaris plants in Temporary Immersion Systems, with a high emission of new shoots in vitro, which can be applied to other bamboo species. Background: Secondary plasma cell leukaemia (sPCL) is an aggressive form of multiple myeloma (MM), but the mechanism underlying MM progresses into PCL remains unknown.Methods: Gene expression profiling of MM patients and PCL patients was analysed to identify the molecular differences between the two diseases. Cox survival regression and Kaplan-Meier analysis were performed to illustrate the impact of integrin subunit alpha 6 (ITGA6) on prognosis of MM. Invasion assays were performed to assess whether ITGA6 regulated the progression of MM to PCL.Results: Gene expression profiling analyses showed that cell metastasis pathways were enriched in PCL and ITGA6 was differentially expressed between PCL and MM. ITGA6 expression was an independent prognostic factor for event-free survival (EFS) and overall survival (OS) of MM patients. Moreover, the stratification ability of the International Staging System (ISS) of MM was improved when including ITGA6 expression. Functional studies uncovered that increased ITGA6 reduced the myeloma cell invasion. Additionally, low expression of ITGA6 resulted from epigenetic downregulating of its anti-sense non-coding RNA, ITGA6-AS1.Conclusion: Our data reveal that ITGA6 gradually decreases during plasma cell dyscrasias progression and low expression of ITGA6 contributes to myeloma metastasis. The physiology and biochemistry of young Opuntia spp. cladodes relate with their Crassulacean acid metabolism, which extends over the day-night cycle in four phases, is species-dependent and is affected by water availability. This study aimed to assess the interaction among species, time-of-day, and the soil water potential (Ψ W) on biochemical and physiological characteristics of cladodes of Opuntia species. Three-week-old cladodes were harvested at 7:00 a.m. and 3:00 p.m. from plants with or without irrigation for 30 d (−0.17 and −5.72 MPa soil Ψ W), from O. albicarpa , O. ficus-indica , O. hyptiacantha , O. megacantha , and O. streptacantha. The experimental design was a factorial 5 x 2 x 2 (species, sampling time and soil Ψ W). The experimental unit was one cladode per plant, and six repetitions were evaluated. Total acids, glucose, fructose, sucrose, starch, total phenolics, free amino acids, and soluble proteins concentrations were evaluated, as well as acid invertase and neutral invertase activities. The interaction among species x soil Ψ W and species x time of the day was significant (P ≤ 0.05) in all variables evaluated. An exception was the species x soil Ψ W on starch concentration (P = 0.1827). The biochemical and physiological characteristics of Opuntia cladodes were modified by the time of the day and soil Ψ W interaction, but most of the characteristics were positively or inversely affected depending on the species, frequently displaying a descending trend following O. streptacantha , O. hyptiacantha , O. megacantha , O. albicarpa and O. ficus-indica. The determination of hematological values is used to obtain knowledge about the health conditions of animal species. The bigheaded Amazon River turtles, (Peltocephalus dumerilianus) are considered one of the least known testudine species concerning their biology and health status. Herein, we determined the hematological and plasma biochemical parameters of 17 (eight males and nine females) adult P. dumerilianus to provide reference interval values for clinically healthy individuals. We collected the blood samples by puncturing the femoral vein using long heparinized hypodermic syringes. Sexual dimorphism for individuals was determined by external observation of the shape of the plastron. The average values obtained for the ten hematological and biochemical parameters analyzed were red blood cell count = 0.32 million µL-1; hematocrit = 20.6 %; hemoglobin = 8.5 g dL-1; mean corpuscular volume = 681.6 fL; mean corpuscular hemoglobin = 267.8 pg; mean corpuscular hemoglobin concentration = 41.9 g dL-1; glucose = 80.6 mg dL-1, total protein = 4.1 g dL-1, triglycerides = 388.9 mg dL-1, and total cholesterol = 79.3 mg dL-1. Despite the sexual dimorphism evidenced for the species, there was no significant statistical difference between males and females for both hematological and biochemical parameters analyzed herein. Based on these results, the population is considered healthy, with parameter values coinciding with previously reported reference ranges for testudines species in the region. The results obtained in this study can be used for assessing the health status of other Amazonian turtle populations, especially in actions aimed at cultivation strategies, management, and species conservation. Background: Impaired blood pressure (BP) recovery with orthostatic hypotension on standing occurs in 20% of older adults. Low BP is associated with low cerebral blood flow but mechanistic links to postural instability and falls are not established. We investigated whether posture-related reductions in cerebral tissue oxygenation (tSO2) in older adults impaired stability upon standing, if a brief sit before standing improved tSO2 and stability, and if Low-tSO2 predicted future falls.Method: Seventy-seven older adults (87 ± 7 years) completed (i) supine-stand, (ii) supine-sit-stand, and (iii) sit-stand transitions with continuous measurements of tSO2 (near-infrared spectroscopy). Total path length (TPL) of the center of pressure sway quantified stability. K-cluster analysis grouped participants into High-tSO2 (n = 62) and Low-tSO2 (n = 15). Fall history was followed up for 6 months.Results: Change in tSO2 during supine-stand was associated with increased TPL (R = -.356, p = .001). When separated into groups and across all transitions, the Low-tSO2 group had significantly lower tSO2 (all p < .01) and poorer postural stability (p < .04) through 3 minutes of standing compared to the High-tSO2 group. There were no effects of transition type on tSO2 or TPL for the High-tSO2 group, but a 10-second sitting pause improved tSO2 and enhanced postural stability in the Low-tSO2 group (all p < .05). During 6-month follow-up, the Low-tSO2 group had a trend (p < .1) for increased fall risk.Conclusions: This is the first study to show an association between posture-related cerebral hypoperfusion and quantitatively assessed instability. The production of bulk chemicals mostly depends on exhausting petroleum sources and leads to emission of greenhouse gases. Within the last decades the urgent need for alternative sources has increased and the development of bio-based processes received new attention. To avoid the competition between the use of sugars as food or fuel, other feedstocks with high availability and low cost are needed, which brought acetogenic bacteria into focus. This group of anaerobic organisms uses mixtures of CO2, CO and H2 for the production of mostly acetate and ethanol. Also methanol, a cheap and abundant bulk chemical produced from methane, is a suitable substrate for acetogenic bacteria. In methylotrophic acetogens the methyl group is transferred to the Wood-Ljungdahl pathway, a pathway to reduce CO2 to acetate via a series of C1-intermediates bound to tetrahydrofolic acid. Here we describe the biochemistry and bioenergetics of methanol conversion in the biotechnologically interesting group of anaerobic, acetogenic bacteria. MicroRNAs (miRNAs) are noncoding small RNAs that regulate various pathophysiological cellular processes. Here, we report that expression of the miR-378 family was significantly induced by metabolic inflammatory inducers, a high-fructose diet, and inflammatory cytokine tumor necrosis factor-α. Hepatic miRNA profiling revealed that expression of miR-378a was highly upregulated, which, in turn, targeted the 3'-untranslated region of PPARα mRNA, impaired mitochondrial fatty acid β-oxidation, and induced mitochondrial and endoplasmic reticulum stress. More importantly, the upregulated miR-378a can directly bind to and activate the double-strand RNA (dsRNA)-dependent protein kinase R (PKR) to sustain the metabolic stress. In vivo, genetic depletion of miR-378a prevented PKR activation and ameliorated inflammatory stress and insulin resistance. Counterbalancing the upregulated miR-378a using nanoparticles encapsulated with an anti-miR-378a oligonucleotide restored PPARα activity, inhibited PKR activation and ER stress, and improved insulin sensitivity in fructose-fed mice. Our study delineated a novel mechanism of miR-378a in the pathogenesis of metabolic inflammation and insulin resistance through targeting metabolic signaling at both mRNA (e.g., PPARα) and protein (e.g., PKR) molecules. This novel finding of functional interaction between miRNAs (e.g., miR-378a) and cellular RNA binding proteins (e.g., PKR) is biologically significant because it greatly broadens the potential targets of miRNAs in cellular pathophysiological processes. Background: Microglia are highly motile phagocytic cells in the healthy brain with surveillance and clearance functions. Although microglia have been shown to engulf cellular debris following brain insult, less is known about their phagocytic function in the absence of injury. Propofol can inhibit microglial activity, including phagocytosis. Milk fat globule epidermal growth factor 8 (MFG-E8), as a regulator of microglia, plays an essential role in the phagocytic process. However, whether MFG-E8 affects the alteration of phagocytosis by propofol remains unknown.Methods: Microglial BV2 cells were treated with propofol, with or without MFG-E8. Phagocytosis of latex beads was evaluated by flow cytometry and immunofluorescence. MFG-E8, p-AMPK, AMPK, p-Src, and Src levels were assessed by western blot analysis. Compound C (AMPK inhibitor) and dasatinib (Src inhibitor) were applied to determine the roles of AMPK and Src in microglial phagocytosis under propofol treatment.Results: The phagocytic ability of microglia was significantly decreased after propofol treatment for 4 h (P < 0.05). MFG-E8 production was inhibited by propofol in a concentration- and time-dependent manner (P < 0.05). Preadministration of MFG-E8 dose-dependently (from 10 to 100 ng/ml) reversed the suppression of phagocytosis by propofol (P < 0.05). Furthermore, the decline in p-AMPK and p-Src levels induced by propofol intervention was reversed by MFG-E8 activation (P < 0.05). Administration of compound C (AMPK inhibitor) and dasatinib (Src inhibitor) to microglia blocked the trend of enhanced phagocytosis induced by MFG-E8 (P < 0.05).Conclusions: These findings reveal the intermediate role of MFG-E8 between propofol and microglial phagocytic activity. Background: Neonatal hypoxic-ischemic (HI) brain injury is a leading cause of acute mortality and chronic disability in newborns. Our previous studies demonstrated that HI insult significantly increased microRNA-210 (miR-210) in the brain of rat pups and inhibition of brain endogenous miR-210 by its inhibitor (LNA) provided neuroprotective effect in HI-induced brain injury. However, the molecular mechanisms underpinning this neuroprotection remain unclear.Methods: We made a neonatal HI brain injury model in mouse pups of postnatal day 7 to uncover the mechanism of miR-210 in targeting the ten eleven translocation (TET) methylcytosine dioxygenase 2 that is a transcriptional suppressor of pro-inflammatory cytokine genes in the neonatal brain. TET2 silencing RNA was used to evaluate the role of TET2 in the neonatal HI-induced pro-inflammatory response and brain injury. MiR-210 mimic and inhibitor (LNA) were delivered into the brain of mouse pups to study the regulation of miR-210 on the expression of TET2. Luciferase reporter gene assay was performed to validate the direct binding of miR-210 to the 3' untranslated region of the TET2 transcript. Furthermore, BV2 mouse microglia cell line was employed to confirm the role of miR-210-TET2 axis in regulating pro-inflammatory response in microglia. Post-assays included chromatin immunoprecipitation (ChIP) assay, co-immunoprecipitation, RT-PCR, brain infarct assay, and neurobehavioral test. Student's t test or one-way ANOVA was used for statistical analysis.Results: HI insult significantly upregulated miR-210, downregulated TET2 protein abundance, and increased NF-κB subunit p65 acetylation level and its DNA binding capacity to the interleukin 1 beta (IL-1β) promoter in the brain of mouse pups. Inhibition of miR-210 rescued TET2 protein level from HI insult and miR-210 mimic decreased TET2 protein level in the brain of mouse pups, suggesting that TET2 is a functional target of miR-210. The co-immunoprecipitation was performed to reveal the role of TET2 in HI-induced inflammatory response in the neonatal brain. The result showed that TET2 interacted with NF-κB subunit p65 and histone deacetylase 3 (HDAC3), a co-repressor of gene transcription. Furthermore, TET2 knockdown increased transcriptional activity of acetyl-p65 on IL-1β gene in the neonatal brain and enhanced HI-induced upregulation of acetyl-p65 level and pro-inflammatory cytokine expression. Of importance, TET2 knockdown exacerbated brain infarct size and neurological deficits and counteracted the neuroprotective effect of miR-210 inhibition. Finally, the in vitro results demonstrated that the miR-210-TET2 axis regulated pro-inflammatory response in BV2 mouse microglia cell line.Conclusions: The miR-210-TET2 axis regulates pro-inflammatory cytokine expression in microglia, contributing to neonatal HI brain injury. This study was designed to investigate the protective role of kefir and insulin consumption on cellular activities, histochemical components, histological architecture, and integrity of the liver by measuring IL-6 and TNF- α in high fat-fed STZ- induced diabetic male Wistar rats. From a cell biological and histochemical point of view, both kefir and insulin have beneficial various effects on cellular activities and the histochemical components such as DNA, RNA, total protein, collagen, polysaccharides, and phospholipids materials synthesis in the examined normal and diabetic hepatocytes of the male rats. Biochemically, kefir and insulin have various biochemical effects on the serum IL-6 and TNF-α levels. From a pathological point of view, we can say the beneficial various cell biological and histochemical effects of kefir, pathologically may make kefir, to some extent, completely repair all the pathological side effects of type1 diabetes on the hepatocytes in diabetic rats. Also, insulin repair some of the pathological side effects of diabetes on the hepatocytes. The positive results of using kefir and insulin in treating the pathogen effects of diabetes make it possible to obtain positive clinical applications/implications.
