Abstract

Background: Recently it is shown that Mycobacterium tuberculosis Beijing genotype are associated with drug resistance. Thus a simple and rapid method is required for identification and differentiation of Beijing family isolates. Therefore the aim of this study is to evaluate discriminatory power of different loci in both 12 MIRU-VNTR and 15 MIRU-VNTR Methods for Beijing genotype. Methods: In total 105 Mycobacterium tuberculosis strains were identified by spoligotyping, then genomic analysis was carried out by using 12 and 15 MIRU-VNTR methods. Allelic diversity for each locus was calculated using Hunter Gaston Discriminatory Index (HGI). Results: With spoligotyping 20 out of 105 isolates (19.05%) belonged to the Beijing family. Investigating all strains with 12 and 15 MIRU-VNTR showed that 11 loci were highly discriminative (HGI≥0.6) and QUB26 had the highest discriminatory power (HGI=0.84). Number of repeats in QUB11b was specific for Beijing genotype, and only MIRU16 was highly discriminator for this genotype. Abstract

Introduction: Beijing strains constitute more than 1/4 of Mycobacterium tuberculosis (MT) genotypes.Beijing genotype is considered an important genotype because of its reasonable characteristics such asassociation with multi-drugs resistance TB. The aim of this study was to identify the genetic pattern ofMT Beijing genotype using spoligotyping, variable number tandem repeat (VNTR) typing and RFLP-IS6110methods.Materials ands Methods: 238 MT culture positive specimens (2007-2008) were genotyped byspoligotyping. Thereafter, the isolated Beijing genotype was subjected to VNTR typing and RFLPanalysis. The results of Spoligotyping were analyzed using SPOLDB4 database. VNTR typing was usedto identify alleles diversity in 9 locus (MPTR-A, ETR-A, ETR-B, ETR-C, ETR-D, ETR-E, ETR-F,QUB11B, QUB3232) of the isolated Beijing strains. The allelic diversity of VNTR was measured usingHunter Gaston Index (HGI).Results: The spoligotyping of MT isolates revealed the following 8 patterns: Haarlem (27.7%), CAS1(25.2%), EAI3 (21.8%), CAS2 (6.7%), T1 (6.3%), Beijing (5.5%), U (5%), T (0.4), EAI2 (1.2%). ByVNTR typing, the QUB3232 locus was identified as the most distinctive (HGI≥0.6); ETR-F, ETR-E andQUB11b loci; as median distinctive loci (HGI≥0.4-0.6) and other loci as the weakest distinctive locus forBeijing families.The Beijing strains demonstrated diverse patterns in RFLP, 13/13(100%) and VNTR10/13(77%).Conclusion: The ariety of genetic patterns revealed the reactivation in the studied population. AsRFLP method is a time consuming and costly effective method, VNTR and spoligotyping method can beused as alternative methods for molecular epidemiology of MT.
