The warming trend in the Arctic region is expected to cause drastic changes including permafrost degradation and vegetation shifts. We investigated the spatial distribution of ice content and stable isotopic compositions of water in near-surface permafrost down to a depth of 1 m in the Indigirka River lowlands of northeastern Siberia to examine how the permafrost conditions control vegetation and microtopography in the Taiga–Tundra boundary ecosystem. The gravimetric water content (GWC) in the frozen soil layer was significantly higher at microtopographically high elevations with growing larch trees (i.e., tree mounds) than at low elevations with wetland vegetation (i.e., wet areas). The observed ground ice (ice-rich layer) with a high GWC in the tree mounds suggests that the relatively elevated microtopography of the land surface, which was formed by frost heave, strongly affects the survival of larch trees. The isotopic composition of the ground ice indicated that equilibrium isotopic fractionation occurred during ice segregation at the tree mounds, which implies that the ice formed with sufficient time for the migration of unfrozen soil water to the freezing front. In contrast, the isotopic data for the wet areas indicated that rapid freezing occurred under relatively non-equilibrium conditions, implying that there was insufficient time for ice segregation to occur. The freezing rate of the tree mounds was slower than that of the wet areas due to the difference of such as soil moisture and snow cover depends on vegetation and microtopography. These results indicate that future changes in snow cover, soil moisture, and organic layer, which control underground thermal conductivity, will have significant impacts on the freezing environment of the ground ice at the Taiga–Tundra boundary in northeastern Siberia. The origins of agriculture in Southwest Asia over 10,000 years ago and its subsequent spread into Europe during the Neolithic have been the focus of much archaeological research over the past several decades. Increasingly more sophisticated analytical techniques have allowed for better understanding of the complex interactions that occurred amongst humans, animals, and their environments during this transition. The Aegean Islands are critically situated where Anatolia and the mainland Greece meet, making the region pivotal for understanding the movement of the Neolithic into Europe. Located on the largest Turkish Aegean island of Gökçeada, the site of Uğurlu Höyük dates to the early Neolithic and has been the subject of ongoing excavations and research integrating a rigorous dating program with comprehensive zooarchaeological research. This paper focuses on the combination of bone collagen and tooth enamel stable isotope data with existing archaeological data to develop a fine-resolution picture of the spread of the Neolithic, particularly the importation and management of domestic fauna on Gökçeada, with broader relevance for understanding Aegean-Anatolian interactions. The stable isotope values from the fauna at Uğurlu have been used for both diachronic intrasite analyses and intersite comparisons between contemporaneous mainland sites. Integrating stable isotope and zooarchaeological datasets makes Uğurlu one of the first island sites to provide a comprehensive understanding of the geographic origin of Neolithic livestock populations and the timing of their spread from Anatolia into Europe during the process of Neolithization. Cyclin-dependent kinase 2 (CDK2) is an important member of the CDK family exerting its most important function in the regulation of the cell cycle. It catalyzes the transfer of the gamma phosphate group from an ATP (adenosine triphosphate) molecule to a Serine/Threonine residue of a peptide substrate. Due to the importance of this enzyme, and protein kinases in general, a detailed understanding of the reaction mechanism is desired. Thus, in this work the phosphoryl transfer reaction catalyzed by CDK2 was revisited and studied by means of hybrid quantum mechanics/molecular mechanics (QM/MM) calculations. Our results suggest that the base-assisted mechanism is preferred over the substrate-assisted pathway when one Mg2+ is present in the active site, in agreement with a previous theoretical study. The base-assisted mechanism resulted to be dissociative, with a potential energy barrier of 14.3 kcal/mol, very close to the experimental derived value. An interesting feature of the mechanism is the proton transfer from Lys129 to the phosphoryl group at the second transition state, event that could be helping in neutralizing the charge on the phosphoryl group upon the absence of a second Mg2+ ion. Furthermore, important insights into the mechanisms in terms of bond order and charge analysis were provided. These descriptors helped to characterize the synchronicity of bond forming and breaking events, and to characterize charge transfer effects. Imaging mass cytometry (IMC) is a technique allowing visualization and quantification of over 40 biological parameters in a single experiment with subcellular spatial resolution, however most IMC experiments are limited to endpoint analysis with antibodies and DNA stains. Small molecules containing tellurium are promising probes for IMC due to their cell permeability, synthetic versatility, and most importantly their application to sequential labelling with isotopologous probes (SLIP) experiments. SLIP experiments with tellurium-containing probes allow quantification of intracellular biology at multiple timepoints with IMC. Despite the promise of tellurium in IMC, there are unique challenges in image processing associated with tellurium IMC data. Here, we address some of these issues by demonstrating the removal of xenon background signal, combining channels to improve signal-to-noise ratio, and calculating isotope transmission efficiency biases. Three individuals dating to the Great Migration Period (5th century CE) were discovered in a pit at the Hermanov vinograd site in Osijek, Croatia. We were inspired to study these individuals based on their unusual burial context as well as the identification of two different types of artificial cranial deformation in two of the individuals. We combine bioarchaeological analysis with radiographic imaging, stable isotopes analysis, and ancient DNA to analyze their dietary patterns, molecular sex, and genetic affinities in the context of the archaeological data and their bioarchaeological attributes. While all three individuals were adolescent males with skeletal evidence of severe malnutrition and similar diets, the most striking observation is that they had major differences in their genetic ancestry. Results of the genetic analyses of the nuclear ancient DNA data for these individuals indicate that the individual without artificial cranial deformation shows broadly West Eurasian associated-ancestry, the individual with tabular oblique-type has East Asian ancestry and the third individual with circular erect-type has Near Eastern associated-ancestry. Based on these results, we speculate that artificial cranial deformation type may have been a visual indicator membership in a specific cultural group, and that these groups were interacting intimately on the Pannonian Plain during the Migration Period. Resource polymorphism—whereby ancestral generalist populations give rise to several specialised morphs along a resource gradient—is common where species colonise newly formed ecosystems. This phenomenon is particularly well documented in freshwater fish populations inhabiting postglacial lakes formed at the end of the last ice age. However, knowledge on how such differential exploitation of resources across contrasting habitats might be reflected in the biochemical compositions of diverging populations is still limited, though such patterns might be expected. Here, we aimed to assess how fatty acids (FA)—an important biochemical component of animal tissues—diverged across a polymorphic complex of European whitefish (Coregonus lavaretus) and their closely related monomorphic specialist congener vendace (Coregonus albula) inhabiting a series of six subarctic lakes in northern Fennoscandia. We also explored patterns of FA composition in whitefish's predators and invertebrate prey to assess how divergence in trophic ecology between whitefish morphs would relate to biochemical profiles of their key food web associates. Lastly, we assessed how information on trophic divergence provided by differential FA composition compared to evidence of resource polymorphism retrieved from more classical stomach content and stable isotopic (δ13C, δ15N) information. Examination of stomach contents provided high-resolution information on recently consumed prey, whereas stable isotopes indicated broad-scale patterns of benthic-pelagic resource use differentiation at different trophic levels. Linear discriminant analysis based on FA composition was substantially more successful in identifying whitefish morphs and their congener vendace as distinct groupings when compared to the other two methods. Three major FA (myristic acid, stearic acid, and eicosadienoic acid) proved particularly informative, both in delineating coregonid groups, and identifying patterns of pelagic-benthic feeding throughout the wider food web. Myristic acid (14:0) content and δ13C ratios in muscle tissue were positively correlated across fish taxa, and together provided the clearest segregation of fishes exploiting contrasting pelagic and benthic niches. In general, our findings highlight the potential of FA analysis for identifying resource polymorphism in animal populations where this phenomenon occurs, and suggest that this technique may provide greater resolution than more traditional methods typically used for this purpose. The rich and long-lasting Nordic Bronze Age was dependent throughout on incoming flows of copper and tin. The crucial turning point for the development of the NBA can be pinpointed as the second phase of the Late Neolithic (LN II, c. 2000–1700 BC) precisely because the availability and use of metal increased markedly at this time. But the precise provenance of copper reaching Scandinavia in the early second millennium is still unclear and our knowledge about the driving force leading to the establishment of the Bronze Age in southern Scandinavia is fragmentary and incomplete. This study, drawing on a large data set of 210 samples representing almost 50% of all existing metal objects known from this period in Denmark, uses trace element (EDXRF) and isotope analyses (MC-ICP-MS) of copper-based artifacts in combination with substantial typological knowledge to profoundly illuminate the contact directions, networks and routes of the earliest metal supplies. It also presents the first investigation of local recycling or mixing of metals originating from different ore regions. Both continuity and change emerge clearly in the metal-trading networks of the Late Neolithic to the first Bronze Age period. Artifacts in LN II consist mainly of high-impurity copper (so-called fahlore type copper), with the clear exception of British imports. Targeted reuse of foreign artifacts in local production is demonstrated by the presence of British metal in local-style axes. The much smaller range of lead isotope ratios among locally crafted compared to imported artifacts is also likely due to mixing. In the latter half of Nordic LN II (1800–1700 BC), the first signs emerge of a new and distinct type of copper with low impurity levels, which gains enormously in importance later in NBA IA. From an environmental perspective optimised dairy systems, which follow current regulations, still have low nitrogen (N) use efficiency, high N surplus (kg N ha-1) and enable ad-hoc delivery of direct and indirect reactive N losses to water and the atmosphere. The objective of the present study was to divide an intensive dairy farm into N attenuation capacity areas based on this ad-hoc delivery. Historical and current spatial and temporal multi-level datasets (stable isotope and dissolved gas) were combined and interpreted. Results showed that the farm had four distinct attenuation areas: high N attenuation: characterised by ammonium-N (NH4+-N) below 0.23 mg NH4+-N l-1 and nitrate (NO3--N) below 5.65 mg NO3--N l-1 in surface, drainage and groundwater, located on imperfectly to moderately-well drained soils with high denitrification potential and low nitrous oxide (N2O) emissions (av. 0.0032 mg N2O-N l-1); moderate N attenuation: characterised by low NO3--N concentration in drainage water but high N2O production (0.0317 mg N2O-N l-1) and denitrification potential lower than group 1 (av. δ15N-NO3-: 16.4‰, av. δ18O-NO3-: 9.2‰), on well to moderately drained soils; low N attenuation—area 1: characterised by high NO3--N (av. 6.90 mg NO3--N l-1) in drainage water from well to moderately-well drained soils, with low denitrification potential (av. δ15N-NO3-: 9.5‰, av. δ18O-NO3-: 5.9‰) and high N2O emissions (0.0319 mg N2O l-1); and low N attenuation—area 2: characterised by high NH4+-N (av. 3.93 mg NH4+-N l-1 and high N2O emissions (av. 0.0521 mg N2O l-1) from well to imperfectly drained soil. Knowing what animals eat is fundamental to our ability to understand and manage biodiversity and ecosystems, but researchers often must rely on indirect methods to infer trophic position and food intake. Using an approach that combines evidence from stable isotope analysis and DNA metabarcoding, we assessed the diet and trophic position of Anthene usamba butterflies, for which there are no known direct observations of larval feeding. An earlier study that analyzed adults rather than caterpillars of A. usamba inferred that this butterfly was aphytophagous, but we found that the larval guts of A. usamba and two known herbivorous lycaenid species contain chloroplast 16S sequences. Moreover, chloroplast barcoding revealed high sequence similarity between chloroplasts found in A. usamba guts and the chloroplasts of the Vachellia drepanolobium trees on which the caterpillars live. Stable isotope analysis provided further evidence that A. usamba caterpillars feed on V. drepanolobium, and the possibilities of strict herbivory versus limited omnivory in this species are discussed. These results highlight the importance of combining multiple approaches and considering ontogeny when using stable isotopes to infer trophic ecology where direct observations are difficult or impossible. Nitrate can be reduced to other N inorganic species via denitrification and incorporated into organic matter by immobilization; however, the effect of biotic/abiotic and redox condition on immobilization and denitrification processes from a single system are not well documented. We hypothesize nitrate (NO3-) transformation pathways leading to the formation of dissolved- and solid-phase organic N are predominantly controlled by abiotic reactions, but the formation of soluble inorganic N species is controlled by redox condition. In this study, organic matter in the form of leaf compost (LC) was spiked with 15NO3- and incubated under oxic/anoxic and biotic/abiotic conditions at pH 6.5. We seek to understand how variations in environmental conditions impact NO3- transformation pathways through laboratory incubations. We find production of NH4+ is predominantly controlled by redox whereas NO3- conversion to dissolved organic nitrogen (DON) and immobilization in solid-phase N are predominantly controlled by abiotic processes. Twenty % of added 15N-NO3- was incorporated into DON under oxic conditions, with abiotic processes accounting for 85% of the overall incorporation. Nitrogen immobilization processes resulted in N concentrations of 4.1–6.6 μg N (g leaf compost)-1, with abiotic processes accounting for 100% and 66% of the overall (biotic+abiotic) N immobilization under anoxic and oxic conditions, respectively. 15N-NMR spectroscopy suggests 15NO3- was immobilized into amide/aminoquinones and nitro/oxime under anoxic conditions. A fraction of the NH4+ was produced abiotically under anoxic conditions (~10% of the total NH4+ production) although biotic organic N mineralization contributed to most of NH4+ production. Our results also indicate Fe(II) did not act as an electron source in biotic-oxic incubations; however, Fe(II) provided electrons for NO3- reduction in biotic-anoxic incubations although it was not the sole electron source. It is clear that, under the experimental conditions of this investigation, abiotic and redox processes play important roles in NO3- transformations. As climatic conditions change (e.g., frequency/intensity of rainfall), abiotic reactions that shift transformation pathways and N species concentrations from those controlled by biota might become more prevalent. Accelerator Mass Spectrometry (AMS) is one of the most sensitive analysis techniques to measure long-lived radionuclides, reaching detection limits for isotopic ratios down to 10−15–10−16 in special cases. Its application portfolio covers nearly every field of environmental research, considering processes in the atmosphere, biosphere, hydrosphere, cryosphere, lithosphere and the cosmosphere. Normally, AMS measures the content of isotopes in comparison to a validated standard. However, in some cases like for example 60Fe, well characterized standard materials are difficult to produce due to the extreme rareness of the isotope. We report here on the manufacturing of a set of 60Fe standards, obtained by processing irradiated copper from a beam dump of the high-power proton accelerator (HIPA) at the Paul Scherrer Institute (PSI). The isotopic ratios of the standards have been adjusted via a dilution series of a master solution, isotopic content of which has been characterized by Multi Collector–Inductively Coupled Plasma–Mass Spectrometry (MC-ICP-MS). In total, we produced three samples with isotopic ratios of 1.037(6)·10−8, 1.125(7)·10−10 and 1.234 (7)·10-12, respectively. The latter had already been applied in three pioneering AMS studies investigating the remaining signal of injected matter of nearby super novae explosions in sediment archives. The origin of the tin used for the production of bronze in the Eurasian Bronze Age is still one of the mysteries in prehistoric archaeology. In the past, numerous studies were carried out on archaeological bronze and tin objects with the aim of determining the sources of tin, but all failed to find suitable fingerprints. In this paper we investigate a set of 27 tin ingots from well-known sites in the eastern Mediterranean Sea (Mochlos, Uluburun, Hishuley Carmel, Kfar Samir south, Haifa) that had been the subject of previous archaeological and archaeometallurgical research. By using a combined approach of tin and lead isotopes together with trace elements it is possible to narrow down the potential sources of tin for the first time. The strongly radiogenic composition of lead in the tin ingots from Israel allows the calculation of a geological model age of the parental tin ores of 291 ± 17 Ma. This theoretical formation age excludes Anatolian, central Asian and Egyptian tin deposits as tin sources since they formed either much earlier or later. On the other hand, European tin deposits of the Variscan orogeny agree well with this time span so that an origin from European deposits is suggested. With the help of the tin isotope composition and the trace elements of the objects it is further possible to exclude many tin resources from the European continent and, considering the current state of knowledge and the available data, to conclude that Cornish tin mines are the most likely suppliers for the 13th–12th centuries tin ingots from Israel. Even though a different provenance seems to be suggested for the tin from Mochlos and Uluburun by the actual data, these findings are of great importance for the archaeological interpretation of the trade routes and the circulation of tin during the Late Bronze Age. They demonstrate that the trade networks between the eastern Mediterranean and some place in the east that are assumed for the first half of the 2nd millennium BCE (as indicated by textual evidence from Kültepe/Kaneš and Mari) did not exist in the same way towards the last quarter of the millennium. The black caiman is one of the largest neotropical top predators, which means that it could play a structuring role within swamp ecosystems. However, because of the difficulties inherent to studying black caimans, data are sorely lacking on many aspects of their general biology, natural history, and ecology, especially in French Guiana. We conducted a detailed study of the Agami Pond black caiman population using a multidisciplinary approach. The aim was to better understand the species’ dietary ecology and movements in the pond, and thus its functional role in pond system. We gathered natural history data, tracked caiman movements using satellite transmitters, and characterized feeding ecology via stable isotope analysis. Our study was carried out over three sampling periods and spanned both wet and dry seasons, which differ in their hydrological and ecological conditions. Our results show that black caiman abundance and age demographics differed between seasons in Agami Pond. In the dry season, Agami Pond is one of the only areas within the marsh to hold water. It thus contains large quantities of different fish species, which form the basis of the black caiman’s diet. Caiman body size, a proxy for age class, was around 1.5 meters. During the wet season, which corresponds to the breeding period for migratory birds (e.g., Agami herons), adult black caimans are present in Agami Pond. Adults were most abundant in the inundated forest. There, most individuals measured up to 2 meters. They also exhibited a particular “predatory” behavior near bird nests, preying on fallen chicks and adults. Juveniles and subadults were present during both seasons in the pond’s open waters. These behavioral observations were backed up by stable isotope analysis, which revealed ontogenetic variation in the caiman’s isotopic values. This isotopic variation reflected variation in diet that likely reduced intraspecific competition between adults and young. The telemetry and microchip data show that different age classes had different movement patterns and that seasonal variation in the pond may influence caiman prey availability and reproductive behavior. The new information gathered should help predict this species’ responses to potential ecosystem disturbance (e.g., water pollution, habitat destruction) and inform the development of an effective conservation plan that involves locals and wildlife officials. Estuarine food webs are generally considered to be supported by marine pelagic and benthic primary producers and by the import of dead organic matter from the open sea. Although estuaries receive considerable amounts of freshwater phytoplankton and organic compounds from adjacent rivers, the potential contribution of these living and dead matter to estuarine food webs is often assumed to be negligible and, therefore, not examined. Based on stable isotope analyses, we report the importance of freshwater suspended particulate organic matter (FW-SPOM) for fuelling estuarine food webs in comparison to estuarine SPOM and microphytobenthos. This previously neglected food source contributed 50–60% (annual average) of food intake of suspension-feeding bivalves such as cockles (Cerastoderma edule), mussels (Mytilus edulis) and Pacific oysters (Magallana gigas) at the Balgzand tidal flats, an estuarine site in the western Wadden Sea (12–32 psu). For these species, this proportion was particularly high in autumn during strong run-off of SPOM-rich freshwater, whilst estuarine SPOM (20%-25%) and microphytobenthos (15%-30%) were relatively important in summer when the freshwater run-off was very low. In cancer tumors, lactate accumulation was initially attributed to high glucose consumption associated with the Warburg Effect. Now it is evident that lactate can also serve as an energy source in cancer cell metabolism. Additionally, lactate has been shown to promote metastasis, generate gene expression patterns in cancer cells consistent with “cancer stem cell” phenotypes, and result in treatment resistant tumors. Therefore, the goal of this work was to quantify the impact of lactate on metabolism in three breast cell lines (one normal and two breast cancer cell lines—MCF 10A, MCF7, and MDA-MB-231), in order to better understand the role lactate may have in different disease cell types. Parallel labeling metabolic flux analysis (13C-MFA) was used to quantify the intracellular fluxes under normal and high extracellular lactate culture conditions. Additionally, high extracellular lactate cultures were labelled in parallel with [U-13C] lactate, which provided qualitative information regarding the lactate uptake and metabolism. The 13C-MFA model, which incorporated the measured extracellular fluxes and the parallel labeling mass isotopomer distributions (MIDs) for five glycolysis, four tricarboxylic acid cycle (TCA), and three intracellular amino acid metabolites, predicted lower glycolysis fluxes in the high lactate cultures. All three cell lines experienced reductive carboxylation of glutamine to citrate in the TCA cycle as a result of high extracellular lactate. Reductive carboxylation previously has been observed under hypoxia and other mitochondrial stresses, whereas these cultures were grown aerobically. In addition, this is the first study to investigate the intracellular metabolic responses of different stages of breast cancer progression to high lactate exposure. These results provide insight into the role lactate accumulation has on metabolic reaction distributions in the different disease cell types while the cells are still proliferating in lactate concentrations that do not significantly decrease exponential growth rates. Anthropogenic pressures, such as contaminant exposure, may affect stable isotope ratios in biota. These changes are driven by alterations in the nutrient allocation and metabolic pathways induced by specific stressors. In a controlled microcosm study with the amphipod Gammarus spp., we studied effects of the β-blocker propranolol on stable isotope signatures (δ15N and δ13C), elemental composition (%C and %N), and growth (protein content and body size) as well as biomarkers of oxidative status (antioxidant capacity, ORAC; lipid peroxidation, TBARS) and neurological activity (acetylcholinesterase, AChE). Based on the known effects of propranolol exposure on cellular functions, i.e., its mode of action (MOA), we expected to observe a lower scope for growth, accompanied by a decrease in protein deposition, oxidative processes and AChE inhibition, with a resulting increase in the isotopic signatures. The observed responses in growth, biochemical and elemental variables supported most of these predictions. In particular, an increase in %N was observed in the propranolol exposures, whereas both protein allocation and body size declined. Moreover, both ORAC and TBARS levels decreased with increasing propranolol concentration, with the decrease being more pronounced for TBARS, which indicates the prevalence of the antioxidative processes. These changes resulted in a significant increase of the δ15N and δ13C values in the propranolol-exposed animals compared to the control. These findings suggest that MOA of β-blockers may be used to predict sublethal effects in non-target species, including inhibited AChE activity, improved oxidative balance, and elevated stable isotope ratios. The latter also indicates that metabolism-driven responses to environmental contaminants can alter stable isotope signatures, which should be taken into account when interpreting trophic interactions in the food webs. The abundance of ungulate populations may fluctuate in response to several limiting factors, including climate, diseases, and predation. In the northern Richardson Mountains, Canada, Dall sheep (Ovis dalli dalli) have undergone a major decline in the past decades and predation by grizzly bears (Ursus arctos) and wolves (Canis lupus) was suspected as a leading cause. To better understand the relationship between these three species located in this rugged and remote ecosystem, we relied on a combination of indirect methods. We investigated the apparent role of predation on the Dall sheep population using spatial ecology and stable isotopes. We examined seasonal variation in predation risk, focusing on how it may affect Dall sheep habitat use and sexual segregation, and we evaluated the proportion of Dall sheep in the diet of both predators using stable isotopes. The movements of the three species were monitored by satellite telemetry. Dall sheep habitat use patterns were analyzed using topographical features, greenness index, land cover, and apparent predation risk. The diets of grizzly bears and wolves were examined using a Bayesian mixing model for carbon and nitrogen stable isotopes. We found that Dall sheep habitat use varied seasonally, with different patterns for ewes and rams. Exposure to grizzly bear risk was higher for rams during summer, while ewes were further exposed to wolf apparent predation risk during winter. The importance of safe habitats for ewes was reflected in space use patterns. Stable isotopes analyses suggested that the diet of grizzly bears was largely from animal sources, with mountain mammals comprising about one quarter. Wolves mostly fed on both aquatic browsers and mountain mammals. Diet variation between individual predators suggested that some individuals specialized on mountain mammals, likely including Dall sheep. We conclude that grizzly bear and wolf apparent predation risk are important in driving Dall sheep habitat use and play a role in sexual segregation. The radiological consequences of a nuclear power plant (NPP) accident, resulting in the release of radionuclides to the environment, will depend largely on the mitigating actions instigated shortly after the accident. It is therefore important to make predictions of the radiation dose to the affected population, from external as well as internal exposure, soon after an accident, despite the fact that data are scarce. The aim of this study was to develop a model for the prediction of the cumulative effective dose up to 84 years of age based on the ground deposition of 137Cs that is determined soon after fallout. The model accounts for different assumptions regarding external and internal dose contributions, and the model parameters in this study were chosen to reflect various mitigating actions. Furthermore, the relative importance of these parameters was determined by sensitivity analysis. To the best of our knowledge, this model is unique as it allows quantification of both the external and the internal effective dose using only a fallout map of 137Cs after a nuclear power plant accident. The cumulative effective dose over a period of 50 years following the accident per unit 137Cs deposited was found to range from 0.14 mSv/kBq m-2 to 1.5 mSv/kBq m-2, depending on the mitigating actions undertaken. According to the sensitivity analysis, the most important parameters governing the cumulative effective dose to various adult populations during 50 years after the fallout appear to be: the correlation factor between the local areal deposition of 137Cs and the maximum initial ambient dose rate; the maximum transfer from regional average fallout on the ground to body burden; the local areal deposition of 137Cs; and the regional average 137Cs deposition. Therefore, it is important that mapping of local 137Cs deposition is carried out immediately after fallout from a nuclear power plant accident, followed by calculations of radiation doses for different scenarios using well-known parameters, in order to identify the most efficient mitigation strategies. Proton conductivity is important in many natural phenomena including oxidative phosphorylation in mitochondria and archaea, uncoupling membrane potentials by the antibiotic Gramicidin, and proton actuated bioluminescence in dinoflagellate. In all of these phenomena, the conduction of protons occurs along chains of hydrogen bonds between water and hydrophilic residues. These chains of hydrogen bonds are also present in many hydrated biopolymers and macromolecule including collagen, keratin, chitosan, and various proteins such as reflectin. All of these materials are also proton conductors. Recently, our group has discovered that the jelly found in the Ampullae of Lorenzini- shark’s electro-sensing organs- is the highest naturally occurring proton conducting substance. The jelly has a complex composition, but we proposed that the conductivity is due to the glycosaminoglycan keratan sulfate (KS). Here we measure the proton conductivity of hydrated keratan sulfate purified from Bovine Cornea. PdHx contacts at 0.50 ± 0.11 mS cm -1, which is consistent to that of Ampullae of Lorenzini jelly at 2 ± 1 mS cm -1. Proton conductivity, albeit with lower values, is also shared by other glycosaminoglycans with similar chemical structures including dermatan sulfate, chondroitin sulfate A, heparan sulfate, and hyaluronic acid. Catastrophic events can significantly impact the demographic processes that shape natural populations of organisms. However, linking the outcomes of such events to specific demographic parameters is often challenging due to a lack of detailed pre-event data. The eruption of the Puyehue-Cordon Caulle volcanic complex on 4 June 2011 had profound consequences for the biota of southwestern Argentina. Our long-term behavioral, ecological, and demographic studies of two species of tuco-tucos (Ctenomys sociabilis and C. haigi) that occur in the region most heavily impacted by ash fall from the eruption provided an unusual opportunity to assess the effects of this event on natural populations of mammals. The post-eruption density of the study population for each species was markedly reduced compared to pre-eruption values, with the relative magnitude of this reduction being greater for the group-living C. sociabilis. The more extensive data set for this species indicated that ash fall from the eruption altered the food resources available to these animals; differences in pre- and post-eruption stable isotope signatures for fur samples from C. sociabilis were consistent with observed changes in vegetation. Per capita female reproductive success was also reduced in this species during the first breeding season following the eruption. Based on our detailed demographic records for C. sociabilis, neither survival of yearling females from 2010 to 2011 nor the percentage of unmarked females in the study population in 2011 differed from pre-eruption values. Instead, the post-eruption decrease in population density for C. sociabilis appeared to reflect reduced within-population recruitment of juvenile females to the 2011 breeding population. Although the eruption did not result in the local extinction of either study population, the demographic consequences detected are likely to have impacted the effective sizes of these populations, creating important opportunities to link specific demographic parameters to previously reported decreases in genetic variability detected after this significant natural event. In Florida, resource use patterns by Armases cinereum (Armases), a highly abundant crab in coastal habitats, may serve as important indicators of habitat condition. Here we investigated feeding patterns of Armases in coastal palm scrub forest to intertidal mangrove forest transition zones (transitions) as well as the relationship between habitat disturbance and Armases’ trophic position across three pairs of geographically separated populations in Tampa FL, USA. Each pair of sites represented an unmodified “natural” location as well as a “disturbed” location lacking upland terrestrial palm scrub forested habitat. Laboratory experiments established a baseline understanding of feeding preference of Armases offered strictly mangrove material as well as sources abundant at the transition. In-situ feeding behavior was examined using MixSIAR mixing models with δ13C and δ15N stable isotope tracers. Armases showed a strong preference for consuming partially-decomposed mangrove material from Avicennia germinans and an equally strong preference for Iva frutescens. Armases also displayed predatory behavior under laboratory conditions, confirming omnivory in the presence of mangrove material. Stable isotopes revealed a pattern of elevated trophic position of Armases in disturbed habitats over paired natural locations. Diet reconstruction provided coarse resolution of in-situ feeding and results show high spatial variation: in natural habitats, Armases appears to rely heavily upon upland plant material compared to disturbed habitats where it may consume more animal prey. Agricultural intensification and shifts in precipitation regimes due to global climate change are expected to increase nutrient concentrations in aquatic ecosystems. However, the direct effects of nutrients widely present in wastewaters, such as nitrate, are poorly studied. Here, we use multiple indicators of fish health to experimentally test the effects of three ecologically relevant nitrate concentrations (<10, 50 and 250 mg NO3-/l) on wild-collected mosquitofish (Gambusia holbrooki), a species widely introduced for mosquito biocontrol in often eutrophic waters. Overall, biomarkers (histopathology, feeding assays, growth and caloric content and stable isotopes as indicators of energy content) did not detect overt signs of serious disease in juveniles, males or females of mosquitofish. However, males reduced food intake at the highest nitrate concentration compared to the controls and females. Similarly, juveniles reduced energy reserves without significant changes in growth or food intake. Calorimetry was positively associated with the number of perivisceral fat cells in juveniles, and the growth rate of females was negatively associated with δ15N signature in muscle. This study shows that females are more tolerant to nitrate than males and juveniles and illustrates the advantages of combing short- and long-term biomarkers in environmental risk assessment, including when testing for the adequacy of legal thresholds for pollutants. Archaeological research on the Canadian island of Newfoundland increasingly demonstrates that the island’s subarctic climate and paucity of terrestrial food resources did not restrict past Pre-Inuit (Dorset) and Native American (Beothuk) hunter-gatherer populations to a single subsistence pattern. This study first sought to characterize hunter-gatherer diets over the past 1500 years; and second, to assess the impact of European colonization on Beothuk lifeways by comparing the bone chemistry of Beothuk skeletal remains before and after the intensification of European settlement in the early 18th century. We employed radiocarbon dating and stable carbon and nitrogen isotope ratio analysis of bulk bone collagen from both Dorset (n = 9) and Beothuk (n = 13) cultures, including a naturally mummified 17th century Beothuk individual. Carbon and nitrogen isotope analysis of 108 faunal samples from Dorset and Beothuk archaeological sites around the island were used as a dietary baseline for the humans. We combined our results with previously published isotope data and radiocarbon dates from Dorset (n = 12) and Beothuk (n = 18) individuals and conducted a palaeodietary analysis using Bayesian modelling, cluster analysis and comparative statistical tests. Dorset diets featured more marine protein than those of the Beothuk, and the diets of Beothuk after the 18th century featured less high trophic level marine protein than those of individuals predating the 18th century. Despite inhabiting the same island, Dorset and Beothuk cultures employed markedly different dietary strategies, consistent with interpretations of other archaeological data. Significantly, European colonization had a profound effect on Beothuk lifeways, as in response to the increasing European presence on the coast, the Beothuk relied more extensively on the limited resources of the island’s boreal forests and rivers. Nitrate (NO3-) concentrations and their dual isotopic compositions (δ15N-NO3- and δ18O-NO3-) were measured to constrain N sources and their cyclic processes in summer using samples from the water column of the northern South China Sea (NSCS). Our data revealed that higher NO3- concentrations and δ15N-NO3- values were observed in the upper waters of the coastal areas near the Pearl River Estuary (PRE). The Bayesian stable isotope mixing model was used to calculated the proportion of nitrate sources, the results indicated that the nitrate in the upper waters of the coastal areas near PRE were mainly influenced by manure and sewage (63%), atmospheric deposition (19%), soil organic nitrogen (12%) and reduced N fertilizer (6%). For the upper waters of the outer areas, low NO3- concentrations and δ15N-NO3- values, but high δ18O-NO3- values, reflected that NO3- was mainly influenced by Kuroshio water intrusion (60%), atmospheric deposition (32%) and nitrogen fixation/nitrification (8%). Complex processes were found in bottom waters. Nitrification and phytoplankton assimilation may be responsible for the higher nitrate concentrations and δ15N-NO3- values. The availability of accurate techniques to discriminate between marked laboratory-reared flies and unmarked wild flies captured in monitoring traps is essential for programs that integrate the Sterile Insect Technique (SIT) to manage fruit flies. In this study, the feasibility of using a stable isotope marking technique for the South American fruit fly, Anastrepha fraterculus (Wiedemann), was assessed. Wild flies were collected from apple orchards, which are a target of a SIT project in southern Brazil. To verify if adult flies could be labelled by the stable isotopes from larval diets, larvae were reared on two different C4-based diets and fruits in laboratory. To evaluate the influence of the two most common attractants applied to capture A. fraterculus (grape juice and CeraTrapTM) and the most common preservation method in fruit fly collections (ethanol), laboratory-reared flies were immersed in McPhail traps containing the respective treatments for two periods of time. Samples were analyzed in an elemental analyzer coupled to a Continuous Flow Isotope Ratio Mass Spectrometer (CF-IRMS) at CENA/USP. The δ13C signatures of flies reared on artificial diets differed significantly from the δ13C of flies whose larvae were reared on fruits and from wild flies. In contrast, the δ15N values were less conclusive and the technique could not rely solely on them. In all cases considered, the δ13C and δ15N signatures from males did not differ from females. Despite the alterations caused by the attractants tested and ethanol, laboratory-flies could be distinguished from the wild ones based on δ13C signatures. This is the first comprehensive study to demonstrate that it is possible to distinguish wild A. fraterculus from flies reared on larval diets containing C4 sugar. The first experimentally derived trophic discrimination factors were also obtained for this species. Thus, intrinsic isotope labelling can serve as a backup to conventional dye marking. Rabies is an ancient and neglected zoonotic disease caused by the rabies virus, a neurotropic RNA virus that belongs to the Rhabdoviridae family, genus Lyssavirus. It remains an important public health problem as there are cost and health concerns imposed by the current human post exposure prophylaxis therapy. The use of monoclonal antibodies (mAbs) is therefore an attractive alternative. Rabies mostly affects people that reside in resource-limited areas where there are occasional failures in the cold-chain. These environmental changes may upset the stability of the mAbs. This study focused on mAbs 62-71-3 and E559; their structures, responses to freeze/thaw (F/T) and exposure to reactive oxygen species were therefore studied with the aid of a wide range of biophysical and in silico techniques in order to elucidate their stability and identify aggregation prone regions. E559 was found to be less stable than 62-71-3. The complementarity determining regions (CDR) contributed the most to its instability, more specifically: peptides and found in CDR3, Trp33 found in CDR1 and the oxidised Met34. The constant region “” was also flagged by the special aggregation propensity (SAP) tool and F/T experiments to be highly prone to aggregation. The E559 peptides “ from the heavy chain and from the light chain, were also highly affected by F/T. These residues may serve as good candidates for mutation, in the aim to bring forward more stable therapeutic antibodies, thus paving a way to a more safe and efficacious antibody-based cocktail treatment against rabies. Seepage of methane (CH4) on land and in the sea may significantly affect Earth’s biogeochemical cycles. However processes of CH4 generation and consumption, both abiotic and microbial, are not always clear. We provide new geochemical and isotope data to evaluate if a recently discovered CH4 seepage from the shallow seafloor close to the Island of Elba (Tuscany) and two small islands nearby are derived from abiogenic or biogenic sources and whether carbonate encrusted vents are the result of microbial or abiotic processes. Emission of gas bubbles (predominantly CH4) from unlithified sands was observed at seven spots in an area of 100 m2 at Pomonte (Island of Elba), with a total rate of 234 ml m-2 d-1. The measured carbon isotope values of CH4 of around -18‰ (VPDB) in combination with the measured δ2H value of -120‰ (VSMOW) and the inverse correlation of δ13C-value with carbon number of hydrocarbon gases are characteristic for sites of CH4 formation through abiogenic processes, specifically abiogenic formation of CH4 via reduction of CO2 by H2. The H2 for methanogenesis likely derives from ophiolitic host rock within the Ligurian accretionary prism. The lack of hydrothermal activity allows CH4 gas to become decoupled from the stagnant aqueous phase. Hence no hyperalkaline fluid is currently released at the vent sites. Within the seep area a decrease in porewater sulphate concentrations by ca. 5 mmol/l relative to seawater and a concomitant increase in sulphide and dissolved inorganic carbon (DIC) indicate substantial activity of sulphate-dependent anaerobic oxidation of methane (AOM). In absence of any other dissimilatory pathway, the δ13C-values between -17 and -5‰ in dissolved inorganic carbon and aragonite cements suggest that the inorganic carbon is largely derived from CH4. We present the first comprehensive multi-isotopic data on human and animal remains from the Final Neolithic Corded Ware culture (ca. 2900–2300 cal. BC) in south-eastern Poland. The study focused on communities of two settlement areas located in the Małopolska Upland and in the Subcarpathian region. Carbon and nitrogen isotopes of bone collagen were investigated to obtain insights into human dietary preferences, whereas the strontium isotope composition of human tooth enamel was used to trace the mobility and provenance of individuals. Sr isotope data point to a non-local origin of at least one-quarter of the investigated individuals in the Subcarpathian region, consistent with associated allochthonous grave inventories of eastern or western origins. In contrast, all investigated individuals in the Małopolska Upland were of local origin. Furthermore, our study shows an example that the use of fauna for the assessment of the local 87Sr/86Sr range of an archaeological site can lead to incorrect conclusions and suggests that a detailed Sr isotopic survey of the geological background and its hydrologic elements is necessary to provide conclusive constraints for the identification of local and non-local individuals in prehistoric communities. Carbon and nitrogen isotope composition of bone collagen indicate an omnivorous diet that included C3-based terrestrial plant and animal resources, in which plant food dominated. Deep-sea hydrothermal vents in the western Pacific are increasingly explored for potential mineral extraction. The study of the composition of the food web plays an important guiding role in the ecological protection and restoration of potential mining areas. The general picture of the nutritional sources of species should be established to assess the potential impacts of future mining activities on the biological composition and food sources. To provide basic information, we analyzed the carbon and nitrogen stable isotope ratios of the dominant macrofauna (mussels, commensal scale worms, crustaceans, gastropods, and vestimentiferans) at three different sites in the PACManus hydrothermal area and the Desmos caldera. The δ13C ratio was significantly different between species: mussels and commensal scale worms showed lighter δ13C ratios, whereas crustaceans showed heavier ratios. In terms of δ15N, mussels had the lowest values and the crustaceans had the highest values. By taking into account these stable isotope signatures, we were able to develop inferences of the food sources for vent community organisms. We found that the food web was based on various species of chemoautotrophic bacteria. Mussels appeared to rely primarily on sulfur-based endosymbionts, which use the Calvin–Benson–Bassham (CBB) cycle and RuBisCO form I as the CO2-fixing enzyme. Commensal polychaetes mostly obtained their nutrition from their hosts. Crustacean species were omnivorous, feeding on chemosynthetic bacteria, sedimentary debris, or even animals according to the local environment. In contrast, gastropods relied mainly on symbiotic bacteria with some supplementary consumption of detritus. Vestimentiferans obtained food from symbiotic bacteria using the RuBisCO form II enzyme in the CBB cycle and may have several symbionts using different fixation pathways. Although most macrofauna relied on symbiotic chemoautotrophic bacteria, our study suggested a closer trophic relationship between animals. Therefore, to evaluate the potential impacts of deep sea mining, it is necessary to study the cascade effects on the food web of the whole ecosystem. Two kinds of phenol- and N- containing borate ester, BTEB and BMEB have good hydrolysis stability due to the B-N coordination bond. The PB value improved by 60.7% and 67.6% respectively at 0.5wt% BTEB, BMEB in rapeseed oil. Their antiwear effect increases with the increase of adding content, and BMEB is better than BTEB. The friction-reducing effect of BTEB is better than BMEB. All additives formed a protective film which containing BOx, FeOx and other organic nitrogen compounds. The better capacities of BMEB may due to the complex boundary lubricating film which contain ferrous sulfate, ferrous sulfide. All additives possessed good antioxidation effect, and it increased the oxidation activation energy than rapeseed oil by 51.15% and 78.82% respectively at 0.25wt%. [ABSTRACT FROM AUTHOR]

Imaging was conducted using an electron tracking-Compton camera (ETCC), which measures γ-rays with energies in the range of 200–900 keV from 95mTc. 95mTc was produced by the 95Mo(p, n)95mTc reaction on a 95Mo-enriched target. A method for recycling 95Mo-enriched molybdenum trioxide was employed, and the recycled yield of 95Mo was 70%-90%. Images were obtained with the gate of three energies. The results showed that the spatial resolution increases with increasing γ-ray energy, and suggested that the ETCC with high-energy γ-ray emitters such as 95mTc is useful for the medical imaging of deep tissue and organs in the human body. Schwertmannite is an Fe(III)-oxyhydroxysulfate which is common in acid mine drainage (AMD) and acid sulfate soil (ASS) environments. Natural schwertmannite is often enriched in Cr(III), yet the effects of Cr(III) substitution on schwertmannite transformation to more stable Fe(III) minerals has not been addressed. Here we examine, for the first time, the effects of Cr(III) substitution on the Fe(II)-accelerated transformation of schwertmannite. X-ray diffraction (XRD) and Fe K-edge extended X-ray absorption fine structure (EXAFS) spectroscopy shows that Cr(III) substitution inhibits schwertmannite transformation. Substitution at a Cr(III):Fe(III) ratio of 0.025 decreased schwertmannite transformation (at pH 6.5) by 18–49% (depending on Fe(II) concentrations) relative to that of Cr(III)-free schwertmannite. Formation of crystalline secondary phases (predominantly goethite) caused associated decreases in solid-phase Fe and Cr extractability by 1 M HCl. The extractability of Cr was consistently greater than that of Fe, suggesting some accumulation of Cr(III) at the residual schwertmannite surface–a phenomenon which passivates the surface against Fe(II)/Fe(III) electron transfer and atom exchange required for the Fe(II)-accelerated transformation process. The finding that Cr(III)-substitution inhibits schwertmannite transformation implies that it may also significantly impact associated Fe, S and trace metal(loid) behaviour. [ABSTRACT FROM AUTHOR]

Quality assessment is essential for the computational prediction and design of RNA tertiary structures. To date, several knowledge-based statistical potentials have been proposed and proved to be effective in identifying native and near-native RNA structures. All these potentials are based on the inverse Boltzmann formula, while differing in the choice of the geometrical descriptor, reference state, and training dataset. Via an approach that diverges completely from the conventional statistical potentials, our work explored the power of a 3D convolutional neural network (CNN)-based approach as a quality evaluator for RNA 3D structures, which used a 3D grid representation of the structure as input without extracting features manually. The RNA structures were evaluated by examining each nucleotide, so our method can also provide local quality assessment. Two sets of training samples were built. The first one included 1 million samples generated by high-temperature molecular dynamics (MD) simulations and the second one included 1 million samples generated by Monte Carlo (MC) structure prediction. Both MD and MC procedures were performed for a non-redundant set of 414 RNAs. For two training datasets (one including only MD training samples and the other including both MD and MC training samples), we trained two neural networks, named RNA3DCNN_MD and RNA3DCNN_MDMC, respectively. The former is suitable for assessing near-native structures, while the latter is suitable for assessing structures covering large structural space. We tested the performance of our method and made comparisons with four other traditional scoring functions. On two of three test datasets, our method performed similarly to the state-of-the-art traditional scoring function, and on the third test dataset, our method was far superior to other scoring functions. Mineral-associated organic matter is associated with a suite of soil minerals that can confer stability, resulting in the potential for long-term storage of carbon (C). Not all interactions impart the same level of protection, however; evidence is suggesting that C in certain mineral pools is dynamic and vulnerable to disturbance in the decades following harvesting. The objective of this research was to describe and characterize organic matter-mineral interactions through depth in horizons of soils of contrasting stand age. Sequential selective dissolutions representing increasingly stable mineral-associated organic matter pools from water soluble minerals (deionized water), organo-metal complexes (Na-pyrophosphate), poorly-crystalline minerals (HCl hydroxylamine), and crystalline secondary minerals (Na-dithionite HCl)) were carried out for Ae, Bf and BC horizons sampled from a Young and Mature forest site (35 and 110 years post-harvest) in Mooseland, Nova Scotia, Canada. Sequential selective dissolution extracts were analyzed for C, δ13C, iron (Fe) and aluminum (Al). Organo-metal complexes (OMC) were the largest mineral-associated OM pool in all horizons. This pool dominated the C distribution in B horizons (~60–70% of Bf bulk C), with a minor contribution from poorly-crystalline (PCrys), crystalline (Crys) minerals and water soluble (WS) associations. C in OMC and PCrys pools explained the variation in bulk C in horizons through depth at both sites. Twice as much C in OMC pools was measured at the Mature site compared to the Young site in the Bf horizons, supported by higher C:(Fe+Al) ratios. Isotopic analysis indicated that this extraction procedure isolated distinct mineral-associated OM pools. δ13C signatures of pyrophosphate-extracted OMC pools ranged from -27‰ to -28‰, similar to δ13C of bulk C and to plant-derived humic acids and associated biomass. The water soluble phase (mean δ13C = -29 ‰) was up to 2 ‰ more depleted, whereas the δ13C of Crys pools were more enriched in 13C (-13‰ to -16 ‰) compared to bulk soil. The results from this study suggest that association with minerals does not necessarily confer stability: organo-metal pools dominate in podzol horizons through depth, and contribute most to C storage, but are potentially susceptible to destabilization following the physical changes resulting from forest harvesting disturbance. Environmental heterogeneity can foster opportunistic foraging by mobile species, resulting in generalized resource and habitat use. Determining species’ food web roles is important to fully understand how ecosystems function, and stable isotopes can provide insight into the foraging ecology of bird assemblages. We investigated flexibility of food choice in mangrove bird assemblages of northeast Australia by determining whether species’ carbon and nitrogen isotopic values corresponded to foraging group classification described in the literature, such as groups of species that are omnivorous or insectivorous. Subsequently, we evaluated foraging group isotopic niche size, overlap, degree of individual specialisation, and the probable proportions of coastal resources that contribute to their collective diets. We found that mangrove birds are more opportunistic when foraging than expected from previous diet studies. Importantly, relationships between the dietary diversity of species within a foraging group and isotopic niche size are spatially inconsistent, making inferences regarding foraging strategies difficult. However, quantifying individual specialisation and determining the probable relative contributions of coastal resources to the collective diet of isotope-based foraging groups can help to differentiate between specialised and generalised foraging strategies. We suggest that flexibility in mangrove bird foraging strategy occurs in response to environmental heterogeneity. A complementary approach that combines isotopic analysis with other dietary information (collated from previous diet studies using visual observation or gut content analyses) has provided useful insight to how bird assemblages partition resources in spatiotemporally heterogeneous environments. Two chemotypes were examined in vitro with CYPs 3A4 and 2C19 by molecular docking, metabolic profiles, and intrinsic clearance deuterium isotope effects with specifically deuterated form to assess the potential for enhancement of pharmacokinetic parameters. The results show the complexity of deuteration as an approach for pharmacokinetic enhancement when CYP enzymes are involved in metabolic clearance. With CYP3A4 the rate limiting step was chemotype-dependent. With one chemotype no intrinsic clearance deuterium isotope effect was observed with any deuterated form, whereas with the other chemotype the rate limiting step was isotopically sensitive, and the magnitude of the intrinsic clearance isotope effect was dependent on the position(s) and extent of deuteration. Molecular docking and metabolic profiles aided in identifying sites for deuteration and predicted the possibility for metabolic switching. However, the potential for an isotope effect on the intrinsic clearance cannot be predicted and must be established by examining select deuterated versions of the chemotypes. The results show how in a deuteration strategy molecular docking, in-vitro metabolic profiles, and intrinsic clearance assessments with select deuterated versions of new chemical entities can be applied to determine the potential for pharmacokinetic enhancement in a discovery setting. Global change processes affect seasonal dynamics of salt marshes and thereby their plant and animal communities. However, these changes have been little investigated for microarthropod communities. We studied the effect of seasonality and changes in sea level on oribatid mites in the natural salt marsh and on artificial islands in the back-barrier environment of the island Spiekeroog (Wadden Sea, Germany). Three zones of the artificial islands were filled with transplanted sods from the lower salt marsh zone and thereby exposed to three different inundation frequencies. We hypothesized that oribatid mite communities will differ along the natural salt marsh vegetation zones [upper salt marsh (USM), lower salt marsh (LSM), pioneer zone (PZ)], which are influenced by different tidal regimes. Accordingly, total oribatid mite densities declined from the USM and LSM to the PZ. Similarly, oribatid mite species compositions changed along the salt marsh transect and also responded to variations in inundation frequency in LSM on artificial islands with typical species of the USM, LSM and PZ being Multioppia neglecta (USM), Hermannia pulchella (LSM), Zachvatkinibates quadrivertex (LSM, PZ) and Ameronothrus schneideri (LSM, PZ). Oribatid mite density in the salt marsh and on the artificial islands was at a maximum in winter and spring; this was due in part to high density of juveniles, pointing to two reproductive periods. We hypothesized that oribatid mite trophic structure changes due to variations in abiotic (e.g., tidal dynamics, temperature) and biotic conditions (e.g., resource availability). Stable isotope (15N, 13C) and neutral lipid fatty acid analyses indicated that oribatid mite species have different diets with e.g., Z. quadrivertex feeding on macroalgae and fungi, A. schneideri feeding on microalgae and bacteria, and Scheloribates laevigatus and M. neglecta feeding on dead organic matter, bacteria and fungi. Overall, the results indicate that oribatid mite species in salt marshes are affected by changes in environmental factors such as inundation intensity, with the effects being most pronounced in species with narrow trophic niches and limited niche plasticity. This study first publishes lead isotope data of bronze vessels from Central China in Western Han Dynasty and attempts to find out the lead ores circulation of this time by taking bronzes from the Gejiagou site (Nanyang City, Henan Province) as an example. The elemental concentrations suggest the lead should be introduced on purpose and indicate the provenance information of lead ores. All the lead isotope ratios conform to the characteristics of common lead and most of them are similar to Nanyang local lead ores. The lead of another two bronzes, NY9 and NY13, should be imported from southern China. Combined with the historical background of early Western Han Dynasty, the wider range of the lead ore circulation may be an indicator for, as the loose policy, economic prosperity and transportation improvement. Atmospheric methane is rapidly lost when it enters humid subterranean critical and vadose zones (e.g., air in soils and caves). Because methane is a source of carbon and energy, it can be consumed by methanotrophic methane-oxidizing bacteria. As an additional subterranean sink, it has been hypothesized that methane is oxidized by natural radioactivity-induced radiolysis that produces energetic ions and radicals, which then trigger abiotic oxidation and consumption of methane within a few hours. Using controlled laboratory experiments, we tested whether radiolysis could rapidly oxidize methane in sealed air with different relative humidities while being exposed to elevated levels of radiation (more than 535 kBq m-3) from radon isotopes 222Rn and 220Rn (i.e., thoron). We found no evidence that radiolysis contributed to methane oxidation. In contrast, we observed the rapid loss of methane when moist soil was added to the same apparatus in the absence of elevated radon abundance. Together, our findings are consistent with the view that methane oxidizing bacteria are responsible for the widespread observations of methane depletion in subterranean environments. Further studies are needed on the ability of microbes to consume trace amounts of methane in poorly ventilated caves, even though the trophic and energetic benefits become marginal at very low partial pressures of methane. All Canadian lake sturgeon (Acipenser fulvescens) populations are listed under the Committee on the Status of Endangered Wildlife in Canada (COSEWIC) due to their complex life history, loss of habitat and negative interactions with anthropomorphic activities. Lake sturgeon diets vary considerably across their range and depend on the local benthic invertebrate fauna, substrata, and competition with congener species. Hence, determining population-specific prey assemblages is a vital contribution to effective conservation efforts. We used carbon and nitrogen stable isotopes to identify lake sturgeon prey preferences for juvenile (<100 cm) and adult (>100 cm) age classes and their trophic niche in the Saskatchewan River, SK, Canada. In this system, lake sturgeon were the top predator within the benthic food web with no direct competition for preferred prey items from congener species. Their diet was dominated by crayfish (49.1± 6.4%) and snails (36.3 ± 5.5%), with no significant differences between age classes. Furthermore, lake sturgeon favoured deep rocky pools throughout the river; a habitat where crayfish and snails are typically found. Therefore, conservation efforts should be directed at preserving these habitats for the residing benthic invertebrate community, and the lake sturgeon’s ability to access them. Accurate determination of in vivo circulating concentrations of extracellular adenosine in blood samples is challenging due to the rapid formation and rapid clearance of adenosine in blood. A blood collection protocol was developed based on direct sampling of venous blood into, and instant mixing with, a STOP solution developed to conserve in vivo adenosine concentrations by completely preventing both its formation and clearance in collected blood. Stable isotope labeled AMP and adenosine spiked into blood ex vivo were used in combination with mass spectrometry to evaluate conservation of adenosine and prevention of its formation. A number of approved drugs, including the P2Y12 antagonist ticagrelor, have been described to increase extracellular adenosine. This may contribute to its clinical profile, highlighting the importance of accurate measurement of in vivo adenosine concentrations.A high sensitive ultra performance liquid chromatography–tandem- mass spectrometry (UPLC-tandem-MS) analytical method for plasma adenosine was developed and validated with a lower limit of quantification of 2 nmol/L. The method demonstrated plasma adenosine stability during sample processing and analytical method performance relevant to human blood samples. The final STOP solution proved able to conserve exogenous adenosine and to prevent adenosine formation from exogenous AMP added in vitro to human blood over 15 minutes. The mean endogenous adenosine concentration in plasma prepared from venous blood collected from 10 healthy volunteers was 13 ± 7 nmol/L. Finally, the method was used to demonstrate the previously described concentration-dependent ability of ticagrelor to conserve extracellular adenosine at clinically relevant exposures. In conclusion, we report an optimized sampling protocol and a validated analytical method for accurate measurement of in vivo circulating adenosine concentrations in human blood, suitable for use in clinical trials. Stable carbon and nitrogen isotopes are often used to make inferences of past environments and social patterns. We analyze δ 13C and δ 15N values in contemporary kukui (Aleurites moluccanus) endocarp to examine the effects of site environment. Results from across environmental transects on Hawai‘i Island show strong patterns for both stable isotopes. For δ 13C a robust linear relationship with elevation is exhibited, strengthened by the inclusion of rainfall. This relationship breaks down at a minimum threshold of annual rainfall, possible relating to physiological responses to drought. For δ 15N, the only significant relationship observed pertains to substrate age. The endocarp from kukui is one of the most readily identified plant remains in the Pacific archaeological records and is often targeted for radiocarbon dating. Introduction: Childhood obesity is currently a serious public health challenge in developing countries. Therefore, an accurate assessment of adiposity is required. The objective of this study was to validate BIA prediction equations for the assessment of total body water and adiposity or percentage of body fat for the first time in Senegalese school-aged children. Methods: One-hundred-fifty-one (151) pupils who were 8–11 years old were randomly selected from four public schools in Dakar. The body composition measured by deuterium dilution method (DDM) was used as the reference method and compared to that predicted by BIA using a multi-frequency analyser. Stepwise backward multiple linear regression was performed to calculate TBW and %BF in a subsample, which were then validated in the rest of the sample. The Bland and Altman approach was used to assess the agreement between the two methods (bias and limits of agreement). Results: FFM was higher in boys (24.6±6.9 kg) compared to girls (21.2±3.3 kg; P<0.001), and FM was lower in boys: 3.7 kg [0.9–26.4] compared to girls: 4.5 kg [1.7–22.7]. Overall, 11.3% of children presented excess adiposity (%BF >25% in boys, and >30% in girls) and 2.0% were obese according to WHO cut points for obesity (BMI z-score >+2.0). The equations developed were as follows: TBW = 0.376(Height2/Z50)-0.470 (sex) +0.076(weight) +0.065(height)-2.28. %BF = -1.10(height2/Z50) +3.14(sex)+1.57(weight)-4.347. These specific equations showed good precision and a low and non-significant mean bias (0.11 kg, P = 0.279; and 0.19 kg, P = 0.764) for TBW and %BF, respectively. Pinniped vibrissae provide information on changes in diet at seasonal and annual scales; however, species-specific growth patterns must first be determined in order to interpret these data. In this study, a simple linear model was used to estimate the growth rate of vibrissae from adult female California sea lions (Zalophus californianus) from San Esteban Island in the Gulf of California, Mexico. The δ15N and δ13C values do not display a marked oscillatory pattern that would permit direct determination of the time period contained in each vibrissa; thus, time (age) was calculated in two ways: 1) based on the correlation between the observed number of peaks (Fourier series) in the δ15N profile and the length of each vibrissa, and 2) through direct comparison with the observed number of peaks in the δ15N profile. Cross-correlation confirmed that the two peaks in the δ15N profile reflected the two peaks in the chlorophyll-a concentration recorded annually around the island. The mean growth rate obtained from the correlation was 0.08 ± 0.01 mm d-1, while that calculated based on the observed number of peaks was 0.10 ± 0.05 mm d-1. Both are consistent with the rates reported for adult females of other otariid species (0.07 to 0.11 mm d-1). Vibrissa growth rates vary by individual, age, sex, and species; moreover, small differences in the growth rate can result in significant differences over the time periods represented by the isotopic signal. Objectives and study: This study aimed at measuring the effect in normal to restricted protein diets with specific 15N natural isotopic abundance (NIA) given during gestation and/or lactation on the 15N NIA of fur, liver and muscle in dams and their offspring from birth to adulthood. The secondary aim was to study the effect of growth on the same parameters. Methods: Female Balb/c mice were fed normal protein diet containing 22% protein or isocaloric low protein diet containing 10% protein throughout gestation. Dam’s diets were either maintained or switched to the other diet until weaning at 30 days. All animals were fed standard chow thereafter. Offspring were sacrificed at 1, 11, 30, 60, 480 days and a group of dams at d1. Growth was modeled as an exponential function on the group followed up until 480 days. Fur, liver and muscle were sampled at sacrifice and analyzed for bulk 15N NIA. Fixed effects and interactions between fixed effects and random elements were tested by three-way ANOVA. Results: Higher 15N NIA in the diet resulted in higher organ 15N NIA. Switching from one diet to another changed 15N NIA in each organ. Although dam and offspring shared the same isotopic environment during gestation, 15N NIA at day 1 was higher in dams. Growth rate did not differ between groups after 10 days and decreased between 1 and 5 months. 15N NIA differed between organs and was affected by growth and gestation/lactation. Conclusion: Dietary 15N NIA is a major determinant of the 15N NIA of organs. 15N NIA depended on organ and age (i.e. growth) suggesting an effect of metabolism and/or dilution space. Post-natal normal-protein diet of lactating dams could reverse the effect of a protein-restricted diet during gestation on the offspring growth. In aquatic systems, food web linkages are often assessed using diet contents, stable isotope ratios, and, increasingly, fatty acid composition of organisms. Some correlations between different trophic metrics are assumed to be well-supported; for example, particular stable isotope ratios and fatty acids seem to reflect reliance on benthic or pelagic energy pathways. However, understanding whether the assumed correlations between different trophic metrics are coherent and consistent across species represents a key step toward their effective use in food web studies. To assess links among trophic markers, we compared relationships between major diet components, fatty acids, and stable isotope ratios in three fishes: yellow perch (Perca flavescens), round goby (Neogobius melanostomus), and spottail shiner (Notropis hudsonius) collected from nearshore Lake Michigan. Yellow perch and spottail shiner are native in this system, while round goby are a relatively recent invader. We found some evidence for agreement between different trophic metrics, especially between diet components, n-3:n-6 fatty acid ratios, and stable isotope ratios (δ13C and δ15N). However, we also observed significant variation in observed relationships among markers and species, potentially due to taxonomic variation in the specific diet items consumed (e.g., chydorid microcrustaceans and Dreissena mussels) and species-specific biochemical processes. In many of these latter cases, the invasive species differed from the native species. Understanding the effects of taxonomic variation on prey and predator signatures could significantly improve the usefulness of fatty acids in food web studies, whereas diet contents and stable isotopes appear to be reliable indicators of trophic niche in aquatic food webs. The inland area of southwestern Sweden is well known for its well-preserved archaeological animal and human remains dating back to the Mesolithic and Neolithic (10000–4000 and 4000–1700 BC). They allow application of multiple bioarchaeological methods, giving insights into various and complementary aspects of prehistoric human life, as well as economic and social structures. One important aspect concerns human mobility and its relation to social networks and to circulation of objects. Here, strontium isotope analysis plays a crucial role. The present study aims to construct a strontium isotope baseline of southwestern Sweden with considerably greater coverage and higher resolution than previously published data. As the region has been affected by glacial events, the relation between bedrock geology and isotope signals of the bioavailable strontium in such areas is given special attention. We determined strontium isotope ratios for 61 water and five archaeological animal samples, and combined the data with previous measurements of two water and 21 non-domestic faunal samples. The results reveal a complex pattern. Several areas with distinct baseline ranges can be distinguished, although with overlaps between some of them. Overall, the bioavailable strontium isotope signals mirror the basement geology of the region. The highest ratios occur in the geologically oldest eastern parts of the Precambrian terrain, while lower ratios are found in the western part, and the lowest ratios occur in the youngest Paleozoic areas. At the same time, there are minor deviations compared to the underlying bedrock, due to glacial transport, overlying sediments, and local intrusions of younger rocks. The background data set now available allows for more nuanced and detailed interpretations of human and animal mobility in the region, in particular by identification of subregions with differing strontium isotope ratios within the Precambrian province. The feeding ecology of two reef fishes associated with artificial reefs in the northwest Gulf of Mexico (GoM) was examined using gut contents and natural stable isotopes. Reefs were divided into three regions (east, central, west) across an east to west gradient of increasing reef complexity and salinity. Gray triggerfish (Balistes capriscus) primarily consumed reef-associated prey (xanthid crabs, bivalves, barnacles) and pelagic gastropods, while red snapper (Lutjanus campechanus) diets were mainly comprised of non-reef prey (stomatopods, fishes, portunid crabs). Natural stable isotopes of carbon (δ13C), nitrogen (δ15N), and sulfur (δ34S) were measured in consumer muscle tissue as well as potential primary producers. Gray triggerfish occupied a lower trophic position than red snapper, with lower δ13C and δ15N values across all size classes and regions, and generally higher δ34S values. Red snapper had a smaller range of stable isotope values and corrected standard ellipse areas across all size classes and regions, indicating a smaller isotopic niche. Contribution estimates of particulate organic matter (26 to 54%) and benthic microalgae (BMA, 47 to 74%) for both species were similar, with BMA contributions greater across all three size classes (juveniles, sub-adults, adults) of red snapper and all but the juvenile size class for gray triggerfish. Species gut contents and stable isotopes differed by region, with fishes consuming more crabs in the east region and more gastropods in the central and west regions. δ13C and δ15N values generally decreased from east to west, while δ34S increased across this gradient. Results highlight species-specific feeding differences associated with artificial reefs, where gray triggerfish may be more dependent on the reef structure for foraging opportunities. The IsoFishR application is a data reduction and analysis tool for laser-ablation strontium isotope data, following common best practices and providing reliable and reproducible results. Strontium isotope ratios (87Sr/86Sr) are a powerful geochemical tracer commonly applied in a wide range of scientific fields and laser-ablation inductively coupled mass spectrometry is considered the method of choice to obtain spatially resolved 87Sr/86Sr isotope ratios from a variety of sample materials. However, data reduction and analyses methods are variable between different research groups and research communities limiting reproducibility between studies. IsoFishR provides a platform to standardize these methods and can be used for both spot and time-resolved line transects. Furthermore, it provides advanced data analysis tools and filters for outlier removal, noise reduction, and visualization of time resolved data. 1D and 2D NMR investigations as well as computational studies, including static quantum-mechanics calculations, density function theory formalism, and classical molecular dynamics, were applied to determine the protonation sites in the thermolabile protecting group (TPG) containing a 2-pyridynyl moiety within its structure. This protecting group has three possible sites for protonation: an azomethine (pyridinic) atom (N1), 2-aminoethanol residue (N2), and 4-amino substituent (N4). Our investigations showed that the protonation mainly occurs on the N1 atom. Such protonation seems to be a major inhibitory factor in the thermal removal of 2-pyridynyl TPG by the “chemical switch” approach and decreases the aromaticity of the pyridine ring. We also discussed possible participation of N2 nitrogen in irreversible intramolecular cyclization under acidic conditions. [ABSTRACT FROM AUTHOR]
