Cordyceps militaris and Lentinula edodes are known for their high extracellular proteolytic, amylolytic, and laccase activity, which is important for bakery production. This article describes the effect of enzymes obtained from mycelial biomass of C. militaris and L. edodes on such properties of flour mixes as amylolytic and proteolytic enzymic activity during baking, technological costs, and bread quality. The research featured strains of C. militaris SRG4 and L. edodes 3790 fungi; mycelial biomass powder of these fungi on a sterile grain substrate (rice, wheat); experimental flour mixes of wheat bread and first-grade baking flour with mycelial biomass powder; bread made from the experimental flour mixes. The experimental part included standard methods used in the bakery industry. The mycelial biomass of C. militaris and L. edodes fungi proved to be a promising baking additive because it possessed hydrolytic enzymes and was rich in protein substances (32.2 ± 1.5 and 26.4 ± 2.0%, respectively), specific mushroom polysaccharides (36.7 ± 0.8 and 52.2 ± 1.2%, respectively), and carotenoids (1600 ± 40 mcg/g of C. militaris biomass). The active amylases and proteinases in the mycelial biomass powder improved the sugar-forming ability and reduced the falling number. In addition, they raised the amount of washed gluten with a slight decrease in elasticity, as well as provided an acceptable structure and viscosity of the dough at 1–4% of mycelial biomass powder. This dose resulted in an optimal bread formulation of standard quality. A bigger amount caused the crumb to darken and increased its humidity while reducing the specific volume. The bread samples with C. militaris were too acid. Mycelial biomass of C. militaris and L. edodes proved to be a good source of enzymes to be used with grain substrates in bakery production. Further research is needed to define the optimal dose and processing modes.

Molecular techniques for studying soil microorganisms allow to quantify the presence of specific functional groups from the total soil DNA and thus to assess their dynamic in response to the land use. Using this approach, a study was carried out to assess the increase the soil organic carbon (C) of arable land in the central Po Valley with three woody species (white willow, black locust, hybrid poplar) in short rotation forestry. Being fungi the most involved microorganisms in the C-cycle and representing the larger part of microbial biomass in volume, total fungi e the two most widespread main fungal degraders in the upper soil layer (Ascomycetes and Basidiomycetes) were used as indicators of the effectiveness of land use in increasing soil organic C. An innovative molecular technique (SYBR® Green I dye on the QuantStudio™ 3D digital PCR system) which allows to overcome the critical issues associated with the relatively low concentration of fungal DNA compared to that of bacteria was used. Compared to the ploughed soil, total organic carbon (TOC), microbial biomass and enzyme activities increased significantly in the woody plantations. However, TOC increased to different degrees among the woody species. White willow and black locust showed a greater increase in soil TOC than arable (+30 and 20% respectively), followed by hybrid poplar with a smaller increase of +12%. The increases in soil TOC in the three plantations were not correlated with the total woody mass, assessed 15 years after abandonment. Total fungi and Ascomycetes were highly correlated with microbial biomass and TOC, while Basidiomycetes were not. Ascomycetes, which commonly prevail in agricultural soils, were still the most abundant even after a short rotation forest cycle of 15 years and can be taken as good indicator of C sequestration activities into the soil. Soil tillage strongly reduced the content of Basidiomycetes and short rotation forestry managed to increase it at least 4-5 times over a 15-year cycle of short rotation forestry, suggesting that it can act as a qualitative indicator of C sequestration. Since Basidiomycetes have different biological and morphological characteristics than Ascomycetes, combining them (i.e., as Ascomicetes:Basidiomycetes ratio) seems to be a good option for choosing the most suitable woody species for carbon accumulation in agricultural soils.

China has a huge area of diverse landscapes and is believed to conceive incredibly high fungal diversity. To systematically and promptly report Chinese fungal species, we initiate the series of Catalogue of fungi in China here. In the first paper of this series, we focus on plant-inhabiting fungi. A total of 33 new taxa are described all over China. These taxa include two new genera, viz. Cremeoefibula and Nothopucciniastrum, 18 new species, viz. Annulohypoxylon lancangensis, Ascotaiwania coffeae, Clitocella neofallax, Coleopuccinia yunnanensis, Cremeoefibula hengduanensis, Crepidotus furcaticystidiosus, C. tomentellus, Diachea macroverrucosa, Helicogloea hangzhouensis, Hyalopsora caprearum, Nemania polymorpha, Phanerochaetella austrosinensis, Physalacria tianzhongshanensis, Setophaeosphaeria panlongensis, Subulicystidium boreale, Trechispora subaraneosa, Vikalpa dujuanhuensis, and Xylaria pteridicola, and 13 new combinations, viz. Nothopucciniastrum actinidiae, N. boehmeriae, N. coriariae, N. corni, N. coryli, N. fagi, N. kusanoi, N. hikosanense, N. hydrangeae-petiolaris, N. miyabeanum, N. styracinum, N. tiliae, and N. yoshinagae. The morphological characteristics and phylogenetic evidence are used to support the establishment of these new taxa and the accuracy of their taxonomic placements. It is controversial how useful bioassays are for identifying the in vivo toxicity of hazardous environmental exposures. In this study, fruiting bodies of forest mushrooms (n = 46), indoor mold colonies (n = 412), fungal secondary metabolites (n = 18), xenobiotic chemicals such as biocides and detergents (n = 6), and methanol extracts of indoor dusts from urban buildings (n = 26) were screened with two different bioactivity assays: boar sperm motility inhibition (BSMI) and inhibition of cell proliferation (ICP) tests. For the forest mushrooms, the toxicity testing result was positive for 100% of poisonous-classified species, 69% of non-edible-classified species, and 18% of edible-classified species. Colonies of 21 isolates of Ascomycota mold fungal species previously isolated from water-damaged buildings proved to be toxic in the tests. Out of the fungal metabolites and xenobiotic chemicals, 94% and 100% were toxic, respectively. Out of the indoor dusts from moldy-classified houses (n = 12) and from dry, mold-free houses (n = 14), 50% and 57% were toxic, respectively. The bioassay tests, however, could not differentiate the samples from indoor dusts of moldy-classified buildings from those from the mold-free buildings. Xenobiotic chemicals and indoor dusts were more toxic in the BSMI assay than in the ICP assay, whereas the opposite results were obtained with the Ascomycota mold colonies and fungal secondary metabolites. The tests recognized unknown methanol-soluble thermoresistant substances in indoor settled dusts. Toxic indoor dusts may indicate a harmful exposure, regardless of whether the toxicity is due to xenobiotic chemicals or microbial metabolites. [ABSTRACT FROM AUTHOR]

A review of selected studies on fungal endophytes confirms the paucity of Basidiomycota and basal fungi, with almost 90% attributed to Ascomycota. Reasons for the low number of Basidiomycota and basal fungi, including the Chytridiomycota, Mucoromycota, and Mortierellomycota, are advanced, including isolation procedure and media, incubation period and the slow growth of basidiomycetes, the identification of non-sporulating isolates, endophyte competition, and fungus–host interactions. We compare the detection of endophytes through culture-dependent methods and culture-independent methods, the role of fungi on senescence of the host plant, and next-generation studies. El hongo Rhizopus stolonifer causa pudrición de frutas y verduras, por lo que es importante la búsqueda de alternativas de control biológico. El objetivo fue realizar antagonismo y evaluar la actividad antifúngica de H. coffeata contra R. stolonifer. Se evaluó el porcentaje de reducción del crecimiento micelial (PRCM) y el tipo de antagonismo de R. stolonifer al crecerlo junto con las cepas antagonistas y se evaluó la actividad antifúngica del caldo de cultivo de H. coffeata. El PRCM fue mayor para H. coffeata y Daldinia sp., presentaron antagonismo de clase 1, inhibición por contacto, ya que el micelio de H. coffeata enrolló las hifas de R. stolonifer. Los tres caldos de cultivo de H. coffeata presentaron efecto sobre el crecimiento de R. stolonifer, pero, fue más evidente en el cocultivo, por lo que mediante este sistema se puede obtener moléculas para utilizarse en el bio-control de R. stolonifer.

Agasthyamala Biosphere Reserve is a part of Western Ghats (India), has diverse ecosystems and constitutes an important biogeographical 'hotspot' which is well known for its species richness and endemism. Since limited information was available on the mycoflora in this area, a survey was conducted to evaluate the macrofungal diversity in the wet evergreen forests of the Agasthyamala Biosphere Reserve. The survey was carried out during the monsoon and post-monsoon seasons of 2021-2022 and revealed the existence of 62 macrofungal species belonging to 43 genera, 24 families, and eight orders. Out of the eight orders, seven orders belong to the division Basidiomycota and the other order Xylariales belongs to Ascomycota. The family Polyporaceae was identified as the dominant family. The survey also noted the presence of saprotrophic and mycorrhizal fungi. Among the identified species, the maximum density was of Panellus pusillus (6.08) followed by Microporus xanthopus (5.38). Microporus xanthopus (82.14%) exhibited the maximum frequency of occurrence and was identified as the most common species. Coprinellus disseminatus was the most abundant species among macrofungi. The assessment of macrofungal diversity using the Shannon biodiversity index resulted in a value of 2.99, indicating a rich and diverse fungal population within the forest. The mesophytic mountain forest is one of the most biodiverse ecosystems in Mexico, in which there is a diverse mycobiota of which many aspects of its biology and properties are unknown. Pseudomarasmius nidus-avis is a recently discovered and described fungus that forms rhizomorphs used in the nesting habits of birds of the family Tyrannidae (Mionectes oleagineus and Tolmomyias sulfurescens). In this work, the bioactive properties of P. nidus-avis, seven basidiomycetes and two wild ascomycetes from the mesophytic mountain forest of central Veracruz state, Mexico, were evaluated in terms of their effect against phytopathogenic fungal (PF) species such as Fusarium oxysporum (ATCC-417 and CBF-185), Sclerotium cepivorum (CBF414) and Leptosphaerulina australis (CBF-433), which generate important economic losses in several crops. In vitro confrontation tests of mycelium isolated from the macrofungi against the PFs were developed, where 50 % of the wild fungi, including P. nidus-avis, showed effect on the phytopathogens. Furthermore, the crude extract obtained from the mycelium and supernatant of P. nidus-avis was evaluated against PF, showing an inhibition of 16.08 % against F. oxysporum (ATCC-417). These results show that P. nidus-avis has bioactive properties with potential for use in the control of some PF diseases of agricultural and forestry importance. [ABSTRACT FROM AUTHOR]

Gene fusion is a process through which two or more distinct genes are fused into a single chimeric gene. Unlike most harmful fusion genes in cancer cells, in this study, we first found that spermidine synthetase- (SPDS, catalyst of spermidine biosynthesis) and saccharopine reductase- (SR, catalyst of the penultimate step of lysine biosynthesis) encoding genes form a natural chimeric gene, FfSpdsSr, in Flammulina filiformis. Through the cloning of full-length ORFs in different strains and the analysis of alternative splicing in developmental stages, FfSpdsSr has only one copy and unique transcript encoding chimeric SPDS-SR in F. filiformis. By an orthologous gene search of SpdsSr in more than 80 fungi, we found that the chimeric SpdsSr exists in basidiomycetes, while the two separate Spds and Sr independently exist in ascomycetes, chytridiomycetes, and oomycetes. Further, the transcript level of FfSpdsSr was investigated in different developmental stages and under some common environmental factors and stresses by RT-qPCR. The results showed that FfSpdsSr mainly up-regulated in the elongation stage and pileus development of F. filiformis, as well as under blue light, high temperature, H2O2, and MeJA treatments. Moreover, a total of 15 sets of RNA-Seq data, including 218 samples of Neurospora crassa, were downloaded from the GEO database and used to analyze the expression correlation of NcSpds and NcSr. The results showed that the separate NcSpds and NcSr shared highly similar co-expression patterns in the samples with different strains and different nutritional and environmental condition treatments. The chimeric SpdsSr in basidiomycetes and the co-expression pattern of the Spds and Sr in N. crassa indicate the special link of spermidine and lysine in fungi, which may play an important role in the growth and development of fruiting body and in response to the multiple environmental factors and abiotic stresses. Competition between 31 macromycete species and two harmful micromycetes Aspergillus niger and Penicillium polonicum was evaluated using dual-culture plate assay. All investigated fungi, except for Inonotus obliquus and Lepista luscina, possessed different levels of antagonistic activity against the tested micromycetes. Hypsizygus marmoreus and Lyophyllum shimeji were inactive against A. niger. Morchella esculenta and Oxyporus obducens were passive in the case of co-growth with P. polonicum. The study of fungal interactions showed variability in types of reactions and level of their visualisation. Co-cultivation of the studied species resulted in the following reactions: deadlock after mycelial contact and at a distance, partial or complete replacement after initial deadlock on contact. In general, the studied macromycetes showed moderate activity against the two micromycetes, as 11 species showed active and 7 species moderate antagonism. Results revealed higher antagonistic activity of macromycetes against P. polonicum than in A. niger co-cultures. Wood-decaying species Ganoderma lucidum and Trametes versicolor were the most active fungi against the two tested micromycetes with a maximal antagonism index. These findings provide valuable insights which can be further explored by means of in vivo assays to find a suitable agent for the biocontrol of diseases or spoilage caused by A. niger and P. polonicum. Fungi contain species with a plethora of ways of adapting to life in nature. Consequently, they produce large amounts of diverse biomolecules that can be generated on a large scale and in an affordable manner. This makes fungi an attractive alternative for many biotechnological processes. Ascomycetes and basidiomycetes are the most commonly used fungi for synthesis of metal-containing nanoparticles (NPs). The advantages of NPs created by fungi include the use of non-toxic fungus-produced biochemicals, energy efficiency, ambient temperature, pressure conditions, and the ability to control and tune the crystallinity, shape, and size of the NPs. Furthermore, the presence of biomolecules might serve a dual function as agents in NP formation and also capping that can tailor the (bio)activity of subsequent NPs. This review summarizes and reviews the synthesis of different metal, metal oxide, metal sulfide, and other metal-based NPs mediated by reactive media derived from various species. The phyla ascomycetes and basidiomycetes are presented separately. Moreover, the practical application of NP mycosynthesis, particularly in the fields of biomedicine, catalysis, biosensing, mosquito control, and precision agriculture as nanofertilizers and nanopesticides, has been studied so far. Finally, an outlook is provided, and future recommendations are proposed with an emphasis on the areas where mycosynthesized NPs have greater potential than NPs synthesized using physicochemical approaches. A deeper investigation of the mechanisms of NP formation in fungi-based media is needed, as is a focus on the transfer of NP mycosynthesis from the laboratory to large-scale production and application. Peatlands are complex wetland-like ecosystems that harbor diverse microbial communities. In this study, the microbial communities (fungal and actinobacterial) associated with an unimpacted peatland (Vankervelsvlei; VV), an impacted peatland (Goukou River system; GK), and a developing peatland (Nuwejaars River system; NR) were determined through ITS and 16S rRNA metataxonomic analyses. Unidentified Acidimicrobiales dominated in GK and NR, unidentified Intrasporangiaceae and Solirubobacterales in NR, and Corynebacterium, Propionibacterium, and Streptomyces species in VV. The fungal phyla, Ascomycota and Basidiomycota, dominated all three sites, and harbored unique fungal taxa belonging to a wide range of fungal guilds. Physicochemical properties of the peat collected from the three sites were analyzed in association with microbial community structures in order to determine which parameters acted as the main drivers for microbial diversity. BEST analysis (linking microbial diversity patterns to environmental variables) showed that nitrogen (N), aluminum (Al), phosphorus (P), and potassium (K) were the most significant physicochemical drivers of actinobacterial community structure, while iron (Fe) and humification were the environmental parameters that affected the fungal communities the most. In conclusion, this study has provided some insight into the fungal and actinobacterial communities associated with three South African peatlands and the main environmental drivers that influence these communities. The Philippines is known for its ecological diversity due to its climatic condition and geographical location. However, despite having a wide range of organisms, the macrofungal species in the country remain poorly documented. Recent mycological research in the tropics states that more species have yet to be discovered in the Philippines. Despite this, a mycological study still focuses on the ethnomycological survey and species listing. In this study, a survey of publications in macrofungi from 1906 to the present revealed that the Philippines have currently 376 validated species names of macrofungi classified under 66 families and 130 genera. Macrofungal species reported in the country belong to class Ascomycota and Basidiomycota with high species diversity under family Agaricaceae, Lycoperdaceae, Marasmiaceae, and Polyporaceae. Most studies were published internationally and were concentrated in Luzon Island, Philippines. Looking into these, we suggest a biodiversity analysis of macrofungi in other regions of the country. A shift into applying molecular systematics and biotechnology would further develop Philippine macrofungal research, given that the historical trend has focused on morphological identification and classification. We hope that this paper will be a call to further fill the gaps in what is known about the macrofungi in the Philippines, given the country's potential for further discoveries. Lichens are well-known examples of complex symbiotic associations between organisms from different Kingdoms. Microfungi in particular, establish diverse associations with the hosting lichen thallus, as species-specific parasites or transient co-inhabitants. The whole community of lichen-associated fungi constitute the 'lichen mycobiome' comprising both ascomycetes and basidiomycetes, including filamentous and yeast taxa. Metabarcoding results and microscopy analyses show that in some thalli, basidiomycetes are frequent lichen-associated fungi but still only a few species could be axenically isolated and morphologically characterized. Within a broad project aiming at characterizing the mycobiome diversity by culture-dependent and independent approaches in two lichen species selected as reference models – Rhizoplaca melanophthalma and Tephromela atra , we succeed in isolating and culturing 76 new strains of basidiomycetous yeasts. The lichen thalli were collected in different mountain regions worldwide and at relatively high elevation. The yeast strains were isolated on different growth media and were studied for their morphological and genetic diversity. Nuclear internal transcribed spacer (ITS) and ribosomal large subunit (LSU) sequence analyses identified them to belong to ten families within the orders Agaricostilbomycetes, Cystobasidiomycetes, Microbotryomycetes, Tremellomycetes and Ustilaginomycetes. The yeasts here detected showed patterns of host-preference in a few cases and they are potentially related to the ecological conditions. [ABSTRACT FROM AUTHOR]

While most lichenicolous fungi belong to the Ascomycota, c. 5% of them are members of the Basidiomycota. Among these, the poorly known genus Crittendenia in the Pucciniomycotina has recently been described for lichenicolous fungi with minuscule needle-like synnematous basidiomata. Although only two species were hitherto known, the wide observed host-spectrum suggested a larger number of mainly host-specific species. A classical revision using morphological characters alone proved to be virtually impossible, because of the large variability of the material from each host genus, and the scant morphological differences between hypothetically distinct species from different hosts. We studied over sixty specimens and made a large number of measurements of the available morphological characters. We additionally generated 18 ITS and 21 nuLSU rDNA sequences and conducted maximum likelihood and Bayesian analyses. We also performed one species delimitation analysis (bPTP-ML). By combining sometimes subtle morphological differences, host choice and phylogenetic results, we were able to accept eighteen morphologically and/or genetically distinct species, all confined to a single host genus or to several closely related host genera. Two further putative species are left unnamed, as richer material is needed and molecular data are missing. Sixteen new species are described: Crittendenia absistentis (on Bacidia absistens), C. bacidinae (on Bacidina apiahica), C. bryostigmatis (on Bryostigma muscigenum), C. byssolomatis (on Byssoloma maderense), C. crassitunicata (on Melanohalea ushuaiensis), C. heterodermiae (on Heterodermia comosa), C. hypotrachynae (on Hypotrachyna), C. kakouettae (on 'Byssoloma' kakouettae), C. lecanorae (on Lecanora), C. lecidellae (on Lecidella elaeochroma), C. lopadii (on Lopadium disciforme), C. parvispora (on Bacidia), C. physciiphila (on Phaeophyscia, Physcia and Physciella), C. physconiae (on Physconia distorta), C. stictae (on Sticta fuliginosa) and C. teloschistis (on Teloschistes). Further, a ML analysis of the Agaricostilbomycetes using ITS and nuLSU sequences suggested that Crittendenia cannot be included in any known family, and the new family Crittendeniaceae is therefore described for the genus. Abstract Background Microbial oils, generated from lignocellulosic material, have great potential as renewable and sustainable alternatives to fossil-based fuels and chemicals. By unravelling the diversity of lipid accumulation physiology in different oleaginous yeasts grown on the various carbon sources present in lignocellulose hydrolysate (LH), new targets for optimisation of lipid accumulation can be identified. Monitoring lipid formation over time is essential for understanding lipid accumulation physiology. This study investigated lipid accumulation in a variety of oleaginous ascomycetous and basidiomycetous strains grown in glucose and xylose and followed lipid formation kinetics of selected strains in wheat straw hydrolysate (WSH). Results Twenty-nine oleaginous yeast strains were tested for their ability to utilise glucose and xylose, the main sugars present in WSH. Evaluation of sugar consumption and lipid accumulation revealed marked differences in xylose utilisation capacity between the yeast strains, even between those belonging to the same species. Five different promising strains, belonging to the species Lipomyces starkeyi, Rhodotorula glutinis, Rhodotorula babjevae and Rhodotorula toruloides, were grown on undiluted wheat straw hydrolysate and lipid accumulation was followed over time, using Fourier transform-infrared (FTIR) spectroscopy. All five strains were able to grow on undiluted WSH and to accumulate lipids, but to different extents and with different productivities. R. babjevae DVBPG 8058 was the best-performing strain, accumulating 64.8% of cell dry weight (CDW) as lipids. It reached a culture density of 28 g/L CDW in batch cultivation, resulting in a lipid content of 18.1 g/L and yield of 0.24 g lipids per g carbon source. This strain formed lipids from the major carbon sources in hydrolysate, glucose, acetate and xylose. R. glutinis CBS 2367 also consumed these carbon sources, but when assimilating xylose it consumed intracellular lipids simultaneously. Rhodotorula strains contained a higher proportion of polyunsaturated fatty acids than the two tested Lipomyces starkeyi strains. Conclusions There is considerable metabolic diversity among oleaginous yeasts, even between closely related species and strains, especially when converting xylose to biomass and lipids. Monitoring the kinetics of lipid accumulation and identifying the molecular basis of this diversity are keys to selecting suitable strains for high lipid production from lignocellulose.

Xylan is the most common hemicellulose in plant cell walls, though the structure of xylan polymers differs between plant species. Here, to gain a better understanding of fungal xylan degradation systems, which can enhance enzymatic saccharification of plant cell walls in industrial processes, we conducted a comparative study of two glycoside hydrolase family 3 (GH3) β-xylosidases (Bxls), one from the basidiomycete Phanerochaete chrysosporium (PcBxl3), and the other from the ascomycete Trichoderma reesei (TrXyl3A). A comparison of the crystal structures of the two enzymes, both with saccharide bound at the catalytic center, provided insight into the basis of substrate binding at each subsite. PcBxl3 has a substratebinding pocket at subsite -1, while TrXyl3A has an extra loop that contains additional binding subsites. Furthermore, kinetic experiments revealed that PcBxl3 degraded xylooligosaccharides faster than TrXyl3A, while the KM values of TrXyl3A were lower than those of PcBxl3. The relationship between substrate specificity and degree of polymerization of substrates suggested that PcBxl3 preferentially degrades xylobiose (X2), while TrXyl3A degrades longer xylooligosaccharides. Moreover, docking simulation supported the existence of extended positive subsites of TrXyl3A in the extra loop located at the N-terminus of the protein. Finally, phylogenetic analysis suggests that wood-decaying basidiomycetes use Bxls such as PcBxl3 that act efficiently on xylan structures from woody plants, whereas molds use instead Bxls that efficiently degrade xylan from grass. Abstract The fungus–substrate interaction on the antioxidant activity of a solid state fermentation system (SSFS) was investigated employing two basidiomycete fungi: Pleurotus columbinus and P. floridanus and two ascomycetes: Aspergillus fumigatus and Paecilomyces variotii on powdered peels of banana, pomegranate and orange, empty pea pods and rice straw. The oven-dried substrates were moistened at 90% water holding capacity, inoculated with the test fungi and incubated at 25 °C for appropriate time. Culture extracts were tested for the enzymatic and non-enzymatic antioxidant activity. The effect of substrate on the antioxidant activity of the SSFS was stronger than that of the fungal species. Peroxidase (POX) activity was higher in the basidiomycetes than ascomycetes and achieved its maximum in P. floridanus versus complete absence in A. fumigatus. By contrast, catalase (CAT) activity was higher in the ascomycetes, particularly P. variotii on banana peel, than in the basidiomycetes. Phenolics and flavonoids were highest in pomegranate peels but lowest in banana peels and rice straw, and they were subjected to severe consumption by the basidiomycetes versus mild production by the ascomycetes. The reducing power (RP) and DPPH scavenging activity were higher in the peels of pomegranate, orange and banana relative to rice straw and empty pea pods, and the fungal effect was limited and independent of the fungal taxonomic group. Orange peel is the appropriate substrate for production of fungal CAT and POX, which activities were mutually exclusive; but pomegranate peel is more suitable for production of phenolics and flavonoids.

Peatlands are complex wetland-like ecosystems that harbor diverse microbial communities. In this study, the microbial communities (fungal and actinobacterial) associated with an unimpacted peatland (Vankervelsvlei; VV), an impacted peatland (Goukou River system; GK), and a developing peatland (Nuwejaars River system; NR) were determined through ITS and 16S rRNA metataxonomic analyses. Unidentified Acidimicrobiales dominated in GK and NR, unidentified Intrasporangiaceae and Solirubobacterales in NR, and Corynebacterium, Propionibacterium, and Streptomyces species in VV. The fungal phyla, Ascomycota and Basidiomycota, dominated all three sites, and harbored unique fungal taxa belonging to a wide range of fungal guilds. Physicochemical properties of the peat collected from the three sites were analyzed in association with microbial community structures in order to determine which parameters acted as the main drivers for microbial diversity. BEST analysis (linking microbial diversity patterns to environmental variables) showed that nitrogen (N), aluminum (Al), phosphorus (P), and potassium (K) were the most significant physicochemical drivers of actinobacterial community structure, while iron (Fe) and humification were the environmental parameters that affected the fungal communities the most. In conclusion, this study has provided some insight into the fungal and actinobacterial communities associated with three South African peatlands and the main environmental drivers that influence these communities.

Proline-specific peptidases (PSP) play a crucial role in the processing of fungal toxins, pheromones, and intracellular signaling. They are of particular interest to biotechnology, as they are able to hydrolyze proline-rich oligopeptides that give a bitter taste to food and can also cause an autoimmune celiac disease. We performed in silico analysis of PSP homologs in the genomes of 42 species of higher fungi which showed the presence of PSP homologs characteristic of various kingdoms of living organisms and belonging to different families of peptidases, including homologs of dipeptidyl peptidase 4 (DPP4) and prolyl aminopeptidase 1 found in almost all the studied fungal species. Homologs of proliniminopeptidases from the S33 family absent in humans were also found. Several studied homologs are characteristic of certain taxonomic groups of fungi. Phylogenetic analysis suggests a duplication of ancestral DPP4 into transmembrane and secreted versions, which predate the split of ascomycete and basidiomycete lineages. Comparative biochemical analysis of DPP4 in alkaliphilic and alkali-tolerant strains of fungi showed that, notwithstanding some individual features of these enzymes, in both cases, the studied DPP4 are active and stable under alkaline conditions and at high salt concentrations, which makes them viable candidates for biotechnology and bioengineering [ABSTRACT FROM AUTHOR]

A preliminary checklist of macromycetes has been previously published with reference to six governorates located in northern Tunisia. This paper provides an updated version of the inventory of macromycetes in Tunisia, which includes 268 species, of which seven are worldwide recognized as rare. The investigation was carried out from September 2015 to December 2018 in 13 governorates of the Tunisian territory. This updated checklist reports 145 new national species records (139 basidiomycetes and six ascomycetes) belonging to 57 genera and 13 families. In nature, it is often surmised that lignin is degraded almost exclusively by basidiomycetes, with minor contributions from bacteria and even fewer from ascomycetes. Yet, numerous studies underscore the ability of many ascomycetes from a wide range of taxonomic classes to degrade newly-formed and ancient lignin, sometime more efficiently than basidiomycetes. Few genomes from ascomycetes have been analyzed in relation with their potential ability to breakdown lignin; yet, these also point out a rich repertoire of enzymes with auxiliary activities suspected to participate in lignin lysis. Not surprisingly, the molecular mechanisms used by ascomycetes to break down lignin are largely unknown. Here, we review the literature on this topic. We also present potential mechanisms by which Podospora anserina, a model ascomycete well-suited to decipher how ascomycetes breakdown lignin, may do so. Owing to their huge biomass, which dominates in soils, and despite the fact that they usually are less efficient than basidiomycetes, it is most likely that ascomycetes actually play important roles in ensuring the recycling of lignin in nature. Colonies of piscivorous birds nesting in forests bring in loads of allochthonous material in amounts significantly exceeding atmospheric depositions, eventually causing major ecosystem changes. We studied the effect of increasing ornithogenic impact on ectomycorrhizal (EMF) and other root-associated fungi in a Scots pine forest affected by a colony of great cormorants. We evaluated quantitative data of ectomycorrhiza (numbers of root tips and morphotypes) and identified fungal species from pine root tips and from sporocarps collected in three designated study zones: D (zone of active nesting), E (colony margin), and G (pristine forest). Species-bearing sporocarps were absent in zone D, and their number steadily decreased in zone E and was significantly lower than in zone G. Species communities and occurrence frequencies of individual species differed significantly between the zones. Environmental factors, both directly (nutrient addition) and indirectly (changes in vegetation cover), associated with bird activity have significantly influenced fungal communities. We hypothesize that the first signs of increased avian impact on root-associated fungi are the reduced diversity of sporocarps and the low presence and diversity of boletoid fungi in root tips. Background. In the Republic of Bashkortostan, macromycete fungi are insufficiently studied. The paper presents the results of a study of the mycobiota of the Gafuriysky district of the Republic of Bashkortostan. The territory of the Gafuri district was studied during one growing season (May – November 2020), and therefore it is impossible to say that the list of fungi presented in this work is the final version. Materials and methods. Material – macromycetes fungi. Methods – practical (collection and definition), descriptive, computer-statistical. Results. The species composition of macromycetes fungi is presented in an annotated list of 44 species. A taxonomic analysis of mycobiota consisting of two divisions (Ascomycota and Basidiomycota), two classes (Pezizomycetes and Agaricomycetes), 8 orders, 25 families, and 33 genera is given. The trophic structure was analyzed. Conclusions. The predominant groups are saprotrophs on destroyed wood – Crepidotus mollis, Pleurotus ostreatus, Coprinus micaceus, etc. (27.3 %) and mycorrhizal agents – Cortinarius triumfans, Russula cyanoxantha, Russula vesca, etc. (15.9 %).

Background: Microbial oils, generated from lignocellulosic material, have great potential as renewable and sustainable alternatives to fossil-based fuels and chemicals. By unravelling the diversity of lipid accumulation physiology in different oleaginous yeasts grown on the various carbon sources present in lignocellulose hydrolysate (LH), new targets for optimisation of lipid accumulation can be identified. Monitoring lipid formation over time is essential for understanding lipid accumulation physiology. This study investigated lipid accumulation in a variety of oleaginous ascomycetous and basidiomycetous strains grown in glucose and xylose and followed lipid formation kinetics of selected strains in wheat straw hydrolysate (WSH). Summary: Genomic regions determining sexual compatibility often display recombination suppression, as occurs in sex chromosomes, plant self‐incompatibility loci and fungal mating‐type loci. Regions lacking recombination can extend beyond the genes determining sexes or mating types, by several successive steps of recombination suppression. Here we review the evidence for recombination suppression around mating‐type loci in fungi, sometimes encompassing vast regions of the mating‐type chromosomes. The suppression of recombination at mating‐type loci in fungi has long been recognized and maintains the multiallelic combinations required for correct compatibility determination. We review more recent evidence for expansions of recombination suppression beyond mating‐type genes in fungi ('evolutionary strata'), which have been little studied and may be more pervasive than commonly thought. We discuss testable hypotheses for the ultimate (evolutionary) and proximate (mechanistic) causes for such expansions of recombination suppression, including (1) antagonistic selection, (2) association of additional functions to mating‐type, such as uniparental mitochondria inheritance, (3) accumulation in the margin of nonrecombining regions of various factors, including deleterious mutations or transposable elements resulting from relaxed selection, or neutral rearrangements resulting from genetic drift. Three ascomycetes (Morchella vulgaris AMRL 36, M. elata AMRL 63, Tuber aestivum AMRL 364) and four basidiomycetes strains (Lentinula edodes AMRL 124 and 126, Agaricus bisporus AMRL 208 and 209) were screened for their ability to grow on liquid static flask cultures of glucose, glycerol, molasses and waste flour-rich hydrolysates with C/N ratio of 20 and produce biomass, exopolysaccharides and lipids. The profile of lipid fatty acids was also assessed. Selected strains were furthermore cultivated in C/N = 50. Results showed that substrate consumption, biomass formation and secondary metabolites production were strain, substrate and C/N ratio dependent. The maximum biomass (X), lipid (L) and exopolysaccharides (EPS) values noted were Xmax = 25.2 g/L (C/N = 20; molasses) and Lmax = 6.51 g/L (C/N = 50; rice cereal hydrolysates) by T. aestivum strain AMRL 364 and EPSmax = 2.41 g/L by M. elata strain AMRL 63 (C/N = 50; molasses), respectively. When C/N ratio of 50 was applied, biomass, lipid production and substrate consumption seem to be negatively affected in most of the trials. The adaptation and capability of the mushroom strains to be cultivated on substrates based on agro-industrial waste streams and infant food of expired shelf date offers the opportunity to set a circular oriented bioprocess. Organisms fossilised in amber include insects, plant remains and (rarely) fungi. One reason for the latter could be that fungal tissue decomposes quickly under the (sub-)tropical conditions where amber-producing trees thrive. Moreover, resin production is highest during drier seasons, while fungi usually occur during wetter periods. One way to improve the knowledge about fungal and similar records in amber may lie in looking for spores which are generally hardier than hyphal tissue. Traditional imaging methods mostly need laborious preparations, such as producing thin sections, without knowing whether microfossils are present in a sample. Here, a novel method to isolate fungal spores and other palynomorphs from amber using solvents is presented. Microscope slides can easily be coated with the suspended particles, embedded and microscopically examined. In this way, fungal remains including fungal spores were recovered from seven out of nine amber samples processed. A total of over 200 palynomorphs of various origins were extracted. They include pollen, moss and fern spores, and various cysts. At the same time, it became clear that treatment with nine different polar and non-polar solvents revealed provenance-specific solubility patterns. This simple 'solvent fingerprinting' may help identify provenances. Of all the provenances, Sumatran amber was the richest in all kinds of palynomorphs, and thus suitable to further develop the method, particularly concerning improved imaging. RNA editing occurs in all kingdoms of life and in various RNA species. The editing of nuclear protein-coding transcripts has long been known in metazoans, but was only recently detected in fungi. In contrast to many metazoan species, fungal editing sites occur mostly in coding regions, and therefore, fungal editing can change protein sequences and lead to modified or new functions of proteins. Indeed, mRNA editing is thought to be generally adaptive on fungi. Although RNA editing has been detected in both, Ascomycota and Basidiomycota, there seem to be considerable differences between these two classes of fungi concerning the types, the timing, and the purpose of editing. This review summarizes the characteristics of RNA editing in fungi and compares them to metazoan species and bacteria. In particular, it will review cellular processes affected by editing and speculate on the purpose of editing for fungal biology with a focus on the filamentous ascomycetes. In this study, five fungal strains (Aspergillus niger SCBM1 – Ni, Aspergillus fumigatus SCBM6 – Fu, Trametes versicolor 561 – Tr, Ganoderma lucidum 601 – Ga and Pleurotus ostreatus PL06 – Pl) were cultivated individually and in consortium for biosynthesis of lignocellulose-degrading enzymes by solid-state fermentation (SSF). The enzyme production was investigated using a 25−1 fractional factorial design, with a total of 16 experiments (F1–F16) using raw sugarcane bagasse and raw wheat bran as substrates. Among the enzymatic extracts produced, Ni (F1) exhibited the highest production of endoglucanase (82.70 U/gds) (units per gram of dry substrate), exoglucanase (80.48 U/gds), β-xylosidase (145.01 U/gds) and manganese peroxidase (3.38 U/gds). For filter paper cellulase, Tr cocktail (F5) was the one that stood out (9.45 U/gds). Among the extracts produced in consortium, Ni + Tr + Pl (F6) presented the highest production of β-glucosidase (171.09 U/gds), β-xylosidase (139.99 U/gds) and manganese peroxidase (3.29 U/gds). For FPase, Ni + Fu + Ga (F12) exhibited the best production (10.46 U/gds). The highest xylanase biosynthesis (2582.38 U/gds) was obtained in Ni + Fu + Pl extract (F4). For laccase, the maximum biosynthesis (25.27 U/gds) was obtained in Tr + Ga + Pl (F13). The cocktails that presented the best enzyme production were: Ni (F1), Ni + Fu + Pl (F9), Ni + Tr + Pl (F6) and Ni + Ga + Pl (F10), demonstrating that the use of microbial consortium can be a promising alternative to obtain enzymatic cocktails with high synergism. • Fungal species were cultivated for biosynthesis of lignocellulose-degrading enzymes. • A total of 16 solid-state fermentations (SSF) were carried out. • Highest productions were obtained in Ni, Ni + Fu + Pl, Ni + Tr + Pl and Ni + Ga + Pl enzymatic cocktails. • The fungal consortia can be a promising alternative to obtain cocktails with high synergism. We studied the changes in the wood-inhabiting fungal communities on dead wood of Scots pine (Pinus sylvestris) under varying duration and intensity of cormorant nesting. The activities of the birds created large amounts of dead wood, changed wood chemistry and altered stand characteristics. Analyses based on records of fruiting structures of fungi showed that fungal communities in the cormorant colony differed from communities in unaffected forest. Ornithogenic factors impacted least on generalist species and promoted the occurrence of fungi uncharacteristic of coniferous wood or wood in general. The rate of ornithogenic change was rapid: a shift in fungal assemblages was evident by the third year of expansion of the bird colony into unaffected forest. It featured the disappearance of tremelloid fungi, a decline of corticioid fungi and an increase of hyphomycetoid fungi. The proportion of coniferous wood specialists decreased and that of generalists increased. It is expected that the changes in fungal communities will persist until the forest is fully regenerated. • The impact of cormorant nesting on wood-inhabiting fungal communities was studied. • Fungal communities under bird impact differed from those in unaffected forest. • Fungi not normally associated with coniferous wood were observed on pine woody debris. • Generalist species and hyphomycetous fungi were more tolerant of bird impact. • A shift in fungal assemblages was recorded three years after the start of bird impact. Interest in the fungi of this region after the Second World War led to the discovery of species then regarded as being new to Science. A scan of the Index of Fungi issued in the period running from 1940-2000 revealed that 240 novel taxa had then been proposed. The recorded novelties were examined following the chronology of their introduction, their distribution in the local fifteen political states and their gross taxonomic characters at the Class level. These new additions were characterised at the rate of 40 units / decade. Most originated from Egypt, Iraq and the Palestine-Israel area and relate to the Classes Mitosporic Fungi, Ascomycetes and Basidiomycetes. All together 145 generic names are reported in this group of novelties; twelve were based on type material collected in Egypt (5 genera), the Palestine-Israel area, Iraq, Kuwait, Lebanon and Sudan. The present group of novelties was also surveyed in relation to the nature of the substrate sustaining the selected holotypes. The relevant Mitosporic fungi (93 taxa) were equally isolated from soil or from living or decaying plant parts. For Ascomycetous novelties (86 taxa), although their soil-borne elements outnumber the plant related ones, several also developed on other types of substrates. In the case of novel Basidiomycetes (37 taxa), the plant-parasitic species encompass those collected on the ground surface. Finally, a limited number of these novelties disclosed notable thermotolerant abilities and some even qualify as thermophiles. The main features of these novel records underlines that in Egypt more attention was awarded to the local Mitosporic fungi and to Ascomycetes inhabiting its soil-borne communities. For the Basidiomycetes (sensu lato) marked interest developed solely in the Palestine-Israel area while in Iraq taxonomic studies focussed on Ascomycetes including those developing on dung substrates. The distribution of the few reported Chytridiomycetes, Zygomycetes and ‘Oomycetes’ also proved to be restricted to the former three states. Present data clearly underlines limited interest has been awarded to the fungi of a region presumed to harbour a specific mycobiota due to its marked arid features. Since 1940, only four novel taxa were thus proposed per annum from a small fraction of the Middle East. Future research should focus on plant related forms of lower (basal clades) and higher (Dikarya) fungi of the area. Conservation measures should also be adopted to ensure an adequate protection of the natural local habitats against the negative pressures generated by the increase in population and the detrimental effects of its activities. Finally, in view of the overwhelming implication of mycology in the fields of biotechnology, significant knowledge of the Middle East fungi is promising.

The macrofungi of Pulau Sibu, Johor were surveyed from 21st to 24th July 2017 in conjunction with Pulau Sibu Scientific Expedition organised by Marine Park Department. Fresh fruit bodies were collected at northern, southern (S) trails of the island, and around Twin Beach resort area. A total of 28 genera from 19 families were collected and documented from these sites. Of these collections, 9 specimens were identified to species level, while 19 specimens were only identified to genus level. Besides that, there were 27 unknown specimens yet to be identified due to limited keys for the identification of tropical macrofungi. Findings in this study reflected that Basidiomycetes are dominant in comparison to Ascomycetes fungi. Wood inhabiting fungal species were dominant in this island as more than 80% of the collections were made from decaying plant materials that include dead wood and leaf litters (twigs, branches, dead leaves).
