Background: A quality marker (Q-marker) for Chinese materia medica (CMM, Zhongyao in Chinese) is a new concept proposed by the academician Changxiao Liu for the quality control of CMM. CMM refers to natural medicines used under the guidance of the theory of traditional Chinese medicine (TCM, Zhongyi in Chinese), and includes Chinese herbal medicines (Zhongyaocai in Chinese), decoction pieces of Chinese medicines (Yinpian in Chinese) and proprietary Chinese medicines (Zhongchengyao in Chinese). The quality of Chinese herbal medicines is a prerequisite for using the decoction pieces of Chinese medicines and proprietary Chinese medicines.Hypothesis/purpose: We believe that the Q-markers for Chinese herbal medicines refer to certain types of substances or characteristics that can represent the quality of Chinese herbal medicines. In a narrow sense, the Q-marker refers to the active components (marker components) or chemicals that can be transferred during the subsequent processing of the proprietary Chinese medicines. Broadly speaking, the Q-marker refers to the morphological, chemical, biological, and ecological characteristics and markers that impact the quality of Chinese herbal medicines. We study on a Q-marker system for Chinese Herbal Medicines using burdock as an example in the paper.Methods: Based on more than 20 years of research by our team, the present paper uses burdock as an example to discuss the establishment of a Q-marker system for Chinese herbal medicines.Results: The Q-marker system for burdock was established from the morphological, chemical and biological aspects of burdock. During cultivation, the ecological environment is an important factor that affects burdock quality and arctiin accumulation.Conclusion: The Q-markers for the quality control of Chinese herbal medicines are the basis for CMM. Background: Isoproterenol is a drug used for the treatment of bradycardia that has many side effects due to its non-selective β-adrenoceptor agonist properties. Aim: In this study, we evaluated an in vivo experimental model for isoproterenol (ISO) cardiac injury using BALB/c mice and protective treatment with a combined optimized dose of Arctiin (AR) and Lactobacillus plantarum (LP) (ARLP; 30 mg/kg b.w. AR and viable 106 CFU/mL LP p.o.) together. Introduction: Isoproterenol causes cardiac injury in many instances. Arctiin is a naturally occurring lignin glycoside present in many herbal medicinal plants that has many beneficial effects for humans. Lactobacillus plantarum is yet another beneficial probiotic that benefits largely to humans. Materials and Methods: Male BALB/c mice in appropriate groups (6 animals each) were treated with ARLP for 7 days with ISO administration (100 mg/kg i.p.) on days 5 and 6. On the 8th day, animals were sacrificed. Serum and heart samples were collected for further processing. CK-MB, cardiac troponin, AST, ALT, LDH, GST, GPx, CAT, SOD, GSH, GSSG, MDA, nuclear factor kappa B (NFκB), TNF-α, and IL-6 by ELISA and gene expressions of nuclear factor erythroid 2-related factor 2 (Nrf2) were estimated in samples. Results: Mice administered with ISO showed a prominent rise in the serum marker enzyme activities and tissue oxidative stress markers (MDA and GSSG). Furthermore, marked reductions in body weight and enzymatic and non-enzymatic antioxidant levels were noticed in ISO toxicity. Alterations in proinflammatory cytokines (TNF-α and IL-6) and Nrf2 levels by RT-PCR were analyzed. Histopathology of the heart also indicated cardiac injury in ISO-intoxicated mice. ARLP pretreatment prevented all these cardiac injuries and exerted significant (p ≤ 0.05) prophylactic protection toward cardiac tissue. Further, we analyzed the possible interactions between AR and LP in vitro, to understand the influence of ARonLP, which showed no significant suppressive/antibacterial activity on LP. Greater burdock (Arctium lappa L.), a biennial herbaceous plant belonging to the Asteraceae family, is a rich source of some bioactive compounds including chlorogenic acid, arctiin, and inulin which are involved in the treatment of a broad range of disorders. In this study, the phytochemical variability and antioxidant activity of nine Iranian A. lappa populations collected from different geographical regions were investigated. The highest content of chlorogenic acid was recorded in Baft (76.48 mg/g DW), Rudsar (75.5 mg/g DW), and Bijar (74.8 mg/g DW) populations, respectively. The content of chlorogenic acid in A. lappa seeds ranged from 23.4 to 75.1 mg/g DW with the lowest content in the Baft population and the highest content in the Savadkooh population. In addition, the highest content of arctiin was found in the seeds of Savadkooh (78.21 mg/g DW) and Baft (77.7 mg/g DW) populations. The mean total carbohydrate content in A.lappa populations was 31.8 mg/g DW. The maximum antioxidant activity was observed in the roots of Roodsar (IC50 = 78.2 µg/mL) and seeds of Firoozkooh (IC50 = 174.8 µg/mL) populations. In comparison, the antioxidant activity of the roots was the most reliable among the extracts. This information provides a perspective for the plant used in domestication, conservation, and breeding programs and for further commercial exploitation in food industries as a potent natural antioxidant source. The poor oral bioavailability of arctiin caused by its low water solubility is the biggest obstacle in developing it as a drug. In this work, a new water-soluble glucuronide derivative of arctiin (arctigenin-4′-O-glucuronide) was synthesized through 2,2,6,6-tetramethylpiperidine 1-oxyl mediated oxidation reaction. Subsequently, its anti-inflammatory effect was evaluated by mice acute lung injury model in vivo. The results showed that the glucuronide derivative of arctiin not only had better water solubility but also displayed improved anti-inflammatory activity in vivo, thus serving as an innovative compound in the drug development of arctiin. Loss of extracellular matrix (ECM) of cartilage due to oxidative stress injury is one of the main characteristics of osteoarthritis (OA). As a bioactive molecule derived from the traditional Chinese Burdock, arctiin exerts robust antioxidant properties to modulate redox balance. However, the potential therapeutic effects of arctiin on OA and the underlying mechanisms involved are still unknown. Based on the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP) tool, Burdock-extracted small molecule arctiin was identified as a potential anti-arthritic component. In vitro, treatment using arctiin rescued the interleukin (IL)-1β-induced activation of proteinases and promoted the cartilage ECM synthesis in human chondrocytes. In vivo, intraperitoneal injection of arctiin ameliorated cartilage erosion and encountered subchondral bone sclerosis in the post-traumatic OA mice. Transcriptome sequencing uncovered that arctiin-enhanced cartilage matrix deposition was associated with restricted oxidative stress. Mechanistically, inhibition of nuclear factor erythroid 2-related factor 2 (NRF2) abolished arctiin-mediated anti-oxidative and anti-arthritic functions. To further broaden the application prospects, a gellan gum (GG)-based bioactive gel (GG-CD@ARC) encapsulated with arctiin was made to achieve long-term and sustained drug release. Intra-articular injection of GG-CD@ARC counteracted cartilage degeneration in the severe (12 weeks) OA mice model. These findings indicate that arctiin may be a promising anti-arthritic agent. Abstract: Arctiin, an important lignan compound in Fructus Arctii, has been reported to possess various kinds of bioactivities. Previous studies on the pharmacokinetic of arctiin after oral administration showed that it had a rapid absorption phase followed by a sharp but lasting disappearance. To gain deep insight into the action mechanism of arctiin, the excretion and metabolism of arctiin in vivo should be further studied. In this paper, three metabolites were isolated and identified in rat feces as (−)-enterolactone (M-1), (−)-arctigenin (M-2) and [(2R,3R)-2-(3′-hydroxybenzyl)-3-(3″,4″-dimethoxybenzyl)-butyrolactone] (M-3). Based on the structures of three metabolites, possible metabolic pathways of arctiin in rats are proposed. At the same time, the cumulative excretion rate of arctiin and its metabolites in rat urine and feces were determined, indicating that arctiin was excreted 19.84% in urine and 1.80% in feces, respectively, enterolactone, the most main metabolite, was excreted 35.80% in feces. These results provide very important information for understanding the metabolism and excretion of arctiin in vivo and speculating its action mechanism, they can provide useful information and reference for further metabolic investigations on arctiin in humans. The chemical composition of the CHCl fraction of the total extract (95% EtOH) of Alfredia cernua (L.) Cass aerial part included triterpene alcohols (α-amyrin, β-amyrin, moretenol, lupeol) and butyrolignan (arctiin) in addition to various simple phenols and organic acids and their esters. Background: H9N2 avian influenza viruses (AIVs) infect avian and mammalian hosts and provide internal genes for new emerging highly pathogenic avian viruses that cause severe pneumonia with high mortality, for which few medications are available. Arctiin, a bioactive lignan glycoside, has been reported to possess multiple pharmacological properties. However, the effect of arctiin on H9N2 virus infection is unclear. In the current study, we analyzed the effect of arctiin on H9N2 virus infection and the underlying molecular mechanism in vitro. Methods: The antiviral effect against H9N2 virus was determined by plaque reduction assay (PRA) and progeny virus reduction assay. We employed MTT assay, qRT-PCR, ELISA, immunofluorescence and Western blotting to better understand the anti-inflammatory effect and corresponding mechanism of arctiin on H9N2 virus-infected cells. Results: The results showed that arctiin had antiviral activity against H9N2 virus. Arctiin treatment reduced H9N2 virus-triggered proinflammatory cytokines, such as IL-6, and TNF-α. Moreover, arctiin significantly suppressed H9N2 virus-mediated expression of COX-2 and PGE2. Furthermore, we found that arctiin inhibited H9N2 virus-mediated activation of RIG-I/JNK MAPK signaling. Interestingly, arctiin treatment obviously reversed H9N2 virus-induced reduction of Nrf2, increased the nuclear translocation of Nrf2, and upregulated Nrf2 signaling target genes (HO-1 and SOD2). Zinc protoporphyrin (Znpp)—an HO-1 inhibitor—weakened the inhibitory effect of arctiin on H9N2 virus-induced RIG-I/JNK MAPK and proinflammatory mediators. Conclusion: Taken together, these results suggested that the anti-inflammatory effects of arctiin on H9N2 virus infection may be due to the activation of Nrf2/HO-1 and blocked RIG-I/JNK MAPK signaling; thus, arctiin may be a promising agent for prevention and treatment of H9N2 virus infections. Human keratinocytes are located in the outermost skin layer and thus particularly vulnerable to ultraviolet B (UVB) radiation exposure. Previous studies have focused on the cellular and molecular perspectives of UVB-induced keratinocyte damage. In the present study, it was demonstrated that pretreatment with the phytochemical arctiin, one of the lignin compounds, protects human HaCaT keratinocytes from UVB-mediated damage. Biochemical assays revealed that UVB-induced cytotoxicity and cell death were significantly reduced in arctiin-pretreated HaCaT cells. In addition, arctiin promoted the wound healing and DNA repair properties of keratinocytes. The photoprotective effects of arctiin were associated with changes in the expression levels of specific microRNAs (miRNAs) in HaCaT cells. A bioinformatics analysis demonstrated that the miRNAs were functionally involved in cancer, cell cycle, and Wnt and mitogen-activated protein kinase signaling pathways. In the present study, the results from the cellular and molecular assays demonstrated a novel role for arctiin in UVB protection in keratinocytes, which is mediated by miRNA responses and the suppression of UVB-induced cell death. The study of the interaction of drugs purified from natural sources and a transport protein, such as β-lactoglobulin (BLG), at the atomic level could be a valuable factor to control their transport to biological sites. In the present study, molecular docking and molecular dynamics simulation methods were used to study the interaction of arctiin and liquiritin as natural drugs and BLG as the transport protein. The molecular docking results indicated that these drugs bind in the internal cavity of BLG and the BLG affinity for binding the liquiritin is greater than arctiin. The docking results also indicated that the hydrogen bond interactions have a dominant role in the BLG-drug complex stability. The analysis of MD simulation trajectories showed that the root mean square deviation (RMSD) of BLG-liquiritin, unliganded BLG, and BLG-arctiin reached equilibrium and fluctuated around the mean value at about 1000, 3500, and 4000 ps, respectively. The time evolution of the radius of gyration and total solvent accessible surface of the protein showed that BLG-arctiin and BLG-liquiritin complexes became stable around 2500 and 5000 ps, respectively. To develop an efficient method for extracting and purifying the active ingredient, arctiin, from Fructus arctii and to investigate the protective effect of arctiin against glucose-induced rat aortic endothelial cell (RAEC) injury was investigated. Using a L9 (34) orthogonal array and two-step column chromatography (with AB-8 macroporous resin) arctiin extraction was optimized using a reflux method with 70 % ethanol. The RAECs were then treated with different concentrations of arctiin (1, 10, or 100 μg/ml). The effects of arctiin on cell viability in a high glucose medium, malondialdehyde (MDA) levels, and lactate dehydrogenase were measured using commercially available assays. After extraction, the purity of arctiin reached 95.7 %. In rats, arctiin was shown to stimulate the proliferation of RAECs in a high glucose medium in a dose-dependent manner. Exposure of RAECs to high glucose resulted in a significant increase in MDA and release of lactate dehydrogenase. This was accompanied by significant increase in nitric oxide release and expression of antiendothelial nitric oxide synthase. This technique resulted in relatively pure arctiin extraction. Furthermore, the results from this study suggest that arctiin could potentially function as a protector against vascular endothelial cell injury and further investigation is warranted.
