Abstract: This study investigates how simulation of condensed winter light conditions effects the growth of wild caught Arctic charr (Salvelinus alpinus L.) held in culture. After capturing from Altevatn, Northern Norway, juvenile Arctic charr were cultivated and subsequently individually divided into two groups. The fish were held under identical conditions from May to October including continuous light. Starting from week 41 after capture (October) one group was subjected to a winter light regime of 6 h light and 18 h dark per day (Winter Light group—WL), while light regimes in the second group remained continuous (Light group). After an eight weeks treatment period the WL-group was reverted to the continuous light regime until the experiment ended in May 2007. Results revealed no pre-treatment differences in growth between the two groups. However, starting from two months after the short day treatment, fishes from the WL group had significant higher growth rates compared to L group fish. At the end of the experiment, e.g. five months after the light treatment, fishes from WL group had increased their average body weight by 353 g as compared to 252 g in the L group. Consequently, results show that simulation of winter light conditions can increase the productivity of land-based Arctic charr (Salvelinus alpinus L.) cultivation by 25 to 30%. Thus, management of wild caught charr farming, great care should be taken as to what light regimes the fish are exposed to. [Copyright &y& Elsevier]

Abstract: Parr–smolt transformation and growth were studied in captive offspring of anadromous Arctic charr (Salvelinus alpinus) from the Hals watercourse in northern Norway (70°N), held either at a natural temperature (<1 °C until May) or at a temperature elevated to 6 °C in late March. In mid-May, 5 weeks after the increase in photoperiod from 8:16 h light:dark to continuous light, gill Na+, K+-ATPase activity started to increase in both temperature groups, concurrent with the final development of full seawater tolerance. Temperature had no effect on the development of gill Na+, K+-ATPase activity, or on hypoosmoregulatory ability. The fish in both treatments resumed growth in mid-May, but from then on growth was faster in the elevated than in the ambient temperature group. In the former group, fish mass doubled in 6 weeks (from 65 to 137 g), and growth ceased at the time when the fish were about to complete their parr–smolt transformation. These findings show that an early vernal temperature increase advances the seasonal growth cycle, but not the parr–smolt transformation, in anadromous Arctic charr. [Copyright &y& Elsevier]

The article presents information on a study which determined the morphological variability of the Arctic charr representing anadromous, landlocked and stationary fluviatile populations occurring in the region of the Hornsund fjord in SW Spitsbergen, Norway in relation to their trophic preferences. It estimates the food preferences of the examined fish based on the contents of their alimentary tracts. It reveals the ersults of single-factor ANOVA testing differences in fork length of the Arctic charr from the region.

The article presents information on a study which examined the summer dynamics of density and biomass of the Arctic charr, Salvelinus alpinus, in a polar stream in Spitsbergen, Norway. It determines the basic population parameters of the freshwater fish species and their changes in the summer period. It describes the changes in the fish size structure of the Salvelinus alpinus population from the Ariebekken stream in the successive electrofishings in summer 2004. It also reveals the results of one-way ANOVA and t-test testing the difference in fish length in different seazones in 2004 and 2005.

The growth in captivity of a wild Welsh strain (Llyn Cowlyd) of Arctic charr Salvelinus alpinus was compared to that of a commercial Scottish strain over the course of a 12 month experiment. At the conclusion of the study, the Welsh strain had a lower mass and condition factor than the commercial fish, but a similar length. Aspects of the growth and population structure of the Welsh strain imply that it could be a subject for aquaculture development, but such practises will be dependant on further work on selection and broodstock development. Arctic charr, a highly plastic salmonid that inhabits the circumpolar region, colonized its current environment after the last glaciation. Recent colonization limits the capacity of many techniques to define and characterize constituent populations. As a novel approach, we used the major histocompatibility (MH) class IIα gene polymorphism as a marker that would characterize the genetic divergence of global Arctic charr populations caused by drift and by local adaptation to pathogens. We were able to detect significant isolation of all the lineages previously defined by mitochondrial DNA sequencing and also isolation of some populations within those groups. We found that most of the polymorphism of the class IIα gene was distributed globally, which indicates ancestral selection; however, in most cases, distinctive allele frequencies and specific haplotypes distinguished each population suggesting that recent selection has also occurred. Although all studied populations showed similar MH class IIα polymorphisms, we also found variation in which particular amino acid positions were polymorphic and which were constant in the different populations studied. This variation provides a greater adaptive capacity for the MH class IIα receptors in Arctic charr and is yet another illustration of the extraordinary plasticity of the species. Although photoperiod is considered as a major environmental cue for timing of seasonal events in fish, little is known about the photic information perceived by fish in different aquatic environments. The strongly seasonal Arctic charr, Salvelinus alpinus, reside in lakes covered by thick ice and snow throughout the dark winter in the north. In the present study, we have measured diel changes in their plasma melatonin concentrations from September to June in Lake Storvatnet (70°N), northern Norway. In addition, we have measured the in vitro melatonin production of Arctic charr pineal glands held at experimental light conditions. From September to April a diel profile in plasma melatonin was seen in the charr in Lake Storvatn, with highest concentrations at night. This profile reflected the prevailing above-surface photoperiod, even in February when there were minimal changes in sub-surface irradiance between day and night. In June, plasma melatonin was low throughout the 24-hr cycle, despite there being a marked sub-surface difference in irradiance between night and day. At this time the irradiance in night probably remained above the threshold for suppression of melatonin production. The in vitro experiments revealed no endogenous rhythm in the pineal melatonin secretion, supporting the conclusion that the diel profile seen in the Arctic charr in their natural habitat was driven by ambient photoperiod. In conclusion, the Arctic charr appear to keep track of time even under the extreme conditions of high latitudes during winter, when lakes have thick ice and snow cover. [ABSTRACT FROM AUTHOR]

We investigated how Arctic charr young respond behaviourally to olfactory cues from skin-damaged conspecifics, charr-fed pikeperch, and the combination of food-deprived pikeperch and skin-damaged conspecific cues in a two-channel Y-maze fluviarium test arena. Significant antipredator responses were found to skin-damaged conspecifics in three behavioural traits, to charr-fed pikeperch in two traits and to the combination of pikeperch and skin-damaged conspecific cues in all six behaviours investigated. The combination of predator and conspecific cues significantly increased spatial avoidance and cautiousness to approach the source of scent compared with odours from charr-fed predators. The results suggest that damage-released alarm cues exist in charr, and they strengthen the antipredator responses in conspecifics. As the charr used different behaviours and levels of response in the presence of different cues, they seem to have sophisticated skills to distinguish between different odour combinations and be able to adjust their behaviour according to the current predation risk level. The article presents an experimental study which examines the positive effects of exposure to increased water velocity on the growth of newly hatched Arctic charr also known as Salvelinus alpinus L. The experiment used 1224 newly hatched fish with the aid of water velocity treatment which has an effect on growth in terms of length and weight. It was found out the use of a higher water velocity immediately after hatching is associated with a significant increase in growth. The study also suggests that environmental growth advantages achieved early in the life history of cod.

The association of genetic markers linked to the sex-determining locus (SEX) was investigated in five Arctic charr Salvelinus alpinus full sib families, all originating from the Fraser River strain, Labrador, Canada. Two distinct sex-linkage classes were identified: type I (two families), with previously reported markers on linkage group 4 (AC-4) linked with SEX; and type II (three families), with two unlinked segments of the AC-4 linkage group, but with markers in only one cluster associated with SEX. Large differences in recombination rates, pseudolinkage assembly or various chromosomal rearrangements may explain these findings. [ABSTRACT FROM AUTHOR]

Summer habitat use by sympatric Arctic charr Salvelinus alpinus, young Atlantic salmon Salmo salar and brown trout Salmo trutta was studied by two methods, direct underwater observation and electrofishing, across a range of habitats in two sub-arctic rivers. More Arctic charr and fewer Atlantic salmon parr were observed by electrofishing in comparison to direct underwater observation, perhaps suggesting a more cryptic behaviour by Arctic charr. The three species segregated in habitat use. Arctic charr, as found by direct underwater observation, most frequently used slow (mean ±s.d. water velocity 7·2 ± 16·6 cm s−1) or often stillwater and deep habitats (mean ±s.d. depth 170·1 ± 72·1 cm). The most frequently used mesohabitat type was a pool. Young Atlantic salmon favoured the faster flowing areas (mean ±s.d. water velocity 44·0 ± 16·8 cm s−1 and depth 57·1 ± 19·0 cm), while brown trout occupied intermediate habitats (mean ±s.d. water velocity 33·1 ± 18·6 cm s−1 and depth 50·2 ± 18·0 cm). Niche overlap was considerable. The Arctic charr observed were on average larger (total length) than Atlantic salmon and brown trout (mean ±s.d. 21·9 ± 8·0, 10·2 ± 3·1 and 13·4 ± 4·5 cm). Similar habitat segregation between Atlantic salmon and brown trout was found by electrofishing, but more fishes were observed in shallower habitats. Electrofishing suggested that Arctic charr occupied habitats similar to brown trout. These results, however, are biased because electrofishing was inefficient in the slow-deep habitat favoured by Arctic charr. Habitat use changed between day and night in a similar way for all three species. At night, fishes held positions closer to the bottom than in the day and were more often observed in shallower stream areas mostly with lower water velocities and finer substrata. Whole-genome duplication in the ancient ray-finned fish and subsequent tetraploidization in the ancestor to the salmonids have complicated genomic and candidate gene studies in these organisms as many genes with multiple copies are present throughout their genomes. In an attempt to identify genes with a potential influence on growth and development, we investigated the genomic positions of insulin-like growth factors 1 and 2 ( IGF1, IGF2), myogenic factors 5 and 6 ( MYF5, MYF6) and growth hormone-releasing factor/pituitary adenylate cyclase-activating polypeptide ( GRF/ PACAP) in three salmonid species: rainbow trout ( Oncorhynchus mykiss), Atlantic salmon ( Salmo salar) and Arctic charr ( Salvelinus alpinus). Our results suggest a tight association between the IGF1, MYF5 and MYF6 genes in all three species. We further localized the duplicated copies of IGF1 to the homeologous linkage groups RT-7/15 in rainbow trout and AC-3/24 in Arctic charr, and the two copies of MYF6 to homeologous linkage groups AS-22/24 in Atlantic salmon. Localization of GRF/ PACAP to RT-7, AS-31 and AC-27 and IGF2 to RT-27, AS-2 and AC-4 in rainbow trout, Atlantic salmon and Arctic charr respectively is consistent with previously reported homologies among these chromosomal segments identified using other genetic markers. However, localization of the second copy of GRF/ PACAP to RT-19 and AC-14 and the duplicated copy of IGF2 to AC-19 suggest a possible new homology/homeology between these chromosomes. Embryo somatic tissues, non-somatic yolk-sac materials, and whole, individual fingerlings (age 0+) of Arctic charr, Salvelinus alpinus (L.), as well as a commercial trout diet, were analysed for a wide spectrum of amino acids. Analytical material consisted of prefeeding swim-up fry that were separated into discrete yolk sac and somatic embryo tissue samples. Amino acid concentrations in fry somatic tissue and whole fingerlings were generally very similar to each other, but were lower than those measured in yolk materials. Higher correlations were observed between the majority of specific amino acid concentrations in the trout diet when compared with fingerling data ( r2 = 0.91) and fry somatic tissue data ( r2 = 0.89), than when correlated with fry yolk sac material ( r2 = 0.76). These results indicate that the essential amino acid profiles of fry somatic tissue and whole fingerlings are closer to that of a commercial feed than they are to the endogenous profiles found in the embryonic yolk sac material itself. The dietary ratios of individual essential amino acids were also compared with the total essential amino acid concentrations (A/E ratios) in whole fingerling tissues, and these ratios could be used to accurately estimate the apparent essential amino acid requirements of Arctic charr. To evaluate whether paternal effects occur on offspring traits, eggs from Arctic charr Salvelinus alpinus were fertilized with similar amount of sperm from size-matched dominant and subordinate males, in a nested design. Eggs fertilized by subordinate fathers resulted in more offspring produced than eggs fertilized by the same amount of sperm from dominant fathers. The Arctic charr Salvelinus alpinus (L.), or Torgoch (‘red belly’) in Welsh, is found in seven lakes in North Wales comprising three native populations (Bodlyn, Cwellyn and Padarn) and four translocated populations (Cowlyd, Diwaunedd, Dulyn and Ffynnon Llugwy). The Welsh charr populations have not been studied much, but the available data are summarised in this short review. Analysis of stomach contents of the native charr populations shows that they are planktivores feeding mainly on cladocerans and copepods. Adult fish attain a size of 14–24 cm (fork length) and fecundity is low (100–800 eggs per female). The available growth data reveal that Welsh charr grow at a similar rate and attain a size similar to most other planktivorous charr populations that have been studied in the UK. In this study, we synthesised the views of 34 participants in a workshop to consider the status and future conservation and management of the Arctic charr ( Salvelinus alpinus L.). These are expressed as a series of resolutions deriving from a conference on Arctic charr conservation held in Perth, Scotland in June 2004. Agreed resolutions from the conference were as follows. (i) The unique diversity of Arctic charr should be recognised for the contribution it makes to biodiversity of northern aquatic communities. (ii) The recognition by the public, nongovernmental organisations and national governments of the importance of Arctic charr in northern ecosystems is an important step to the management and protection that should be pursued. (iii) The taxonomic status of highly variable charr populations requires to be reviewed. (iv) There should be a separate system, complementary to the taxonomic one, which systematically catalogues the biological diversity of S. alpinus. (v) The Arctic charr provides a unique natural resource to study the process of evolution and this requires the highest level of protection from anthropogenic effects. (vi) The status of conservation policy for Arctic charr needs to be urgently reviewed to take account of its unique position in the fauna. [ABSTRACT FROM AUTHOR]

Hydroacoustic surveys are an important tool for assessing the abundance of Arctic charr ( Salvelinus alpinus (L.)) in lakes, the requirement for which is increasing because of new environmental legislation. Consequently, survey demand is now such that assessments must be undertaken with the minimum of effort compatible with scientific validity. One major complication is seasonal variability in abundance, with which surveyors have attempted to deal by defining somewhat arbitrary seasonal sampling windows. The definition of more objective windows was pursued by analysing variations in the seasonal abundance of Arctic charr in Windermere, UK, recorded using day and night hydroacoustic surveys at monthly intervals from 1991 to 2001. Two alternative sampling windows for night surveys free of significant internal seasonal effects were revealed, i.e., December to September and September to December. From both a modern and a historical perspective there is little doubt that the Arctic charr, Salvelinus alpinus, in Britain and Ireland (as well as elsewhere) is a ‘difficult’ species. Historically 15 separate species have been recognised from populations in Britain and Ireland and there have been recent attempts to reassert these specific names. Here we review the evidence for the status of these ‘species’. We conclude that the evidence for these 15 being afforded full species status is poor. However, both historical and contemporary data show that Salvelinus alpinus (Linnaeus 1758) in Britain and Ireland exhibits levels of variability in form that is much greater than in other species. We argue that a comprehensive genotypic and phenotypic survey of charr populations in Britain and Ireland is required to determine the full extent of variability and the status of populations with a view of providing suitable protection. The effects of formalin and ethanol preservation on the δ13C and δ15N isotope signatures of Arctic charr Salvelinus alpinus muscle tissue were examined. The lipid content of the tissue samples studied ranged from 3·6 to 6·1% and was not correlated with the magnitude of observed isotopic shifts in preserved samples. Ethanol and formalin significantly depleted and enriched, respectively, the δ13C isotope signatures of preserved tissues when compared to control samples. Ethanol did not significantly enrich δ15N signatures in comparison to controls, whereas formalin did. A meta-analysis of multiple species effects further demonstrated significant preservation effects in fish tissue. Statistical analysis of data obtained by correcting preserved tissue isotope signatures with literature, bootstrapped or meta-analysis derived correction factors demonstrated significant differences between corrected and control sample isotope signatures or failure to produce a unity slope when the data sets were regressed against one another. Species-specific, bootstrapped linear correction models resulted in no such errors. Results suggest that species-specific correction methods should be used for fishes because of the known wide variation in fish tissue lipid content and composition. Accordingly, the use of pilot studies will be required to develop correction factors that properly adjust for preservation effects when interpreting temporal patterns in historic analyses of food webs. Abstract: Arctic charr (Salvelinus alpinus L.) from Lake Thingvallavatn, Iceland occur as four distinct morphs: large benthivorous (LB), dwarf benthivorous (DB), piscivorous (PI) and planktonivorous (PL). The morphs differ with respect to body size, head morphology, growth rate, and life history. The aim of this study was to investigate the paired box protein 7 (Pax7) gene as a candidate for such polymorphisms due to its importance in cranio-facial, skeletal muscle, and central nervous system development. No variation in coding and intronic sequences was found between morphs. We identified 10 alternate Pax7 isoforms with insertions/deletions: a four-residue (GNRT) deletion, a GEASS insertion truncated by the first serine residue (GEAS), and a thirteen-residue insertion (GQYA/TGPEYVYCGT). The latter insertion with a threonine (T) contains a putative casein kinase II (CK-2) phosphorylation site. Pax7 spatial expression patterns were identical in embryos of DB-, LB-, and PL-morphs, and were similar to those described for zebrafish Pax7c, but a difference in temporal expression for segmentation was observed between DB and LB morphs. At the end of segmentation, novel expression was observed in the mandibular region as two bilateral domains. The potential role of multiple alternative splicing of the Pax7 gene for the generation of different Arctic charr morphs is briefly discussed. [Copyright &y& Elsevier]

Arctic charr, Salvelinus alpinus (L.), is one of the several northern fishes that show resource polymorphisms in postglacial lakes. Two reproductively isolated morphs of Arctic charr coexist in distinct ecological niches in the subarctic lake Fjellfrøsvatn, North Norway. Offspring of the two morphs (termed littoral charr and profundal charr) were reared separately but under identical conditions. Their feeding behaviour was compared experimentally using different kinds of live prey. The fishes had no experience with natural prey before the experiments. The littoral morph were more effective in eating live plankton ( Daphnia) and littoral benthos ( Gammarus), and had a higher attack rate against pleuston (surface prey, Gerris) compared with the profundal morph. The two morphs behaved in accordance with expectations from their in situ niche utilisation towards the three prey types. This indicates a case of incipient ecological speciation where divergence in resource utilisation in contrasting niches has evolved adaptations in feeding behaviour by natural selection. The primary aim of the present study was to evaluate the population level of adherent (autochthonous) aerobic and facultative anaerobic bacteria in the hindgut of healthy Arctic charr ( Salvelinus alpinus L.) fed dextrin or inulin. This was assessed by the dilution plate technique, and visualized using both transmission and scanning electron microscopy. A population level of 4.8 × 105 adherent bacteria per gram wet mass was found in the hindgut of fish fed a casein-based diet supplemented with 15% dextrin. However, substituting dextrin with 15% inulin reduced the bacterial population level in the hindgut (3.56 × 104). A total of 217 bacterial isolates were identified by key phenotypical and biochemical characteristics. In addition, 22 strains were also identified by partial sequencing of the 16S rRNA gene. The composition of bacteria colonizing the hindgut of Arctic charr fed dextrin was dominated by the genera Staphylococcus, Pseudomonas, Micrococcus, Psychrobacter glacincola and Streptococcus. However, bacteria colonizing the hindgut of fish fed inulin were dominated by Gram-positive bacteria of the genera Staphylococcus, Streptococcus, Carnobacterium and Bacillus. While Carnobacterium divergens-like strains were isolated from charr fed dextrin, Carnobacterium maltaromicus-like strains were isolated from the hindgut of fish fed inulin. Electron microscopical analysis of hindgut regions confirmed traditional culture-based microbial analysis as fewer bacterial cells were observed between microvilli and associated with the surfaces of enterocytes of fish fed inulin rather than dextrin. Abstract: This study aimed to test the hypothesis that diets containing relatively high amounts of the Δ6 desaturated fatty acids stearidonic acid (STA, 18:4n −3) and γ-linolenic acid (GLA, 18:3n −6), may be beneficial in salmonid culture. The rationale being that STA and GLA would be better substrates for highly unsaturated fatty acid (HUFA) synthesis as their conversion does not require the activity of the reputed rate-limiting enzyme, fatty acid Δ6 desaturase. Duplicate groups of two Arctic charr (Salvelinus alpinus L.) populations with different feeding habits, that had been reported previously to show differences in HUFA biosynthetic capacity, were fed for 16 weeks on two fish meal based diets containing 47% protein and 21% lipid differing only in the added lipid component, which was either fish oil (FO) or echium oil (EO). Dietary EO had no detrimental effect on growth performance and feed efficiency, mortalities, or liver and flesh lipid contents in either population. The proportions of 18:2n −6, 18:3n −3, 18:3n −6, 18:4n −3, 20:3n −6 and 20:4n −3 in total lipid in both liver and flesh were increased by dietary EO in both populations. However, the percentages of 20:5n −3 and 22:6n −3 were reduced by EO in both liver and flesh in both strains, whereas 20:4n −6 was only significantly reduced in flesh. In fish fed FO, HUFA synthesis from both [1-14C]18:3n −3 and [1-14C]20:5n −3 was significantly higher in the planktonivorous Coulin charr compared to the demersal, piscivorous Rannoch charr morph. However, HUFA synthesis was increased by EO in Rannoch charr, but not in Coulin charr. In conclusion, dietary EO had differential effects in the two populations of charr, with HUFA synthesis only stimulated by EO in the piscivorous Rannoch morph, which showed lower activities in fish fed FO. However, the hypothesis was not proved as, irrespective of the activity of the HUFA synthesis pathway in either population, feeding EO resulted in decreased tissue levels of n −3HUFA and 20:4n −6. This has been observed previously in salmonids fed vegetable oils, and thus the increased levels of Δ6 desaturated fatty acids in EO did not effectively compensate for the lack of dietary HUFA. A hatchery experiment was organised to find out if the high loss rate of coded wire tags, noticed in connection with an Arctic charr Salvelinus alpinus (L.) restocking programme, was size-dependent and also to test head moulds for future taggings. Six moulds made by Northwest Marine Technology Inc. and five moulds made at the Finnish Game and Fisheries Research Institute were used. A total of 100 charr were tagged with standard-length coded wire tags using each mould. One-summer old charr at tagging were held for 534 days and two-summer old charr for 320 days. The tag loss rate varied between 0% and 54% and was negatively related to the size of the charr at tagging. The high loss rate was connected with the poor alignment of some moulds, such that a large proportion of the tags were located in the nasal cavity instead of the nasal cartilage. Testing of the moulds and careful grading of the fish for tagging is emphasised, especially if Arctic charr smaller than 20 g in weight are to be tagged. [ABSTRACT FROM AUTHOR]

The relationship between parasite infection and breeding ornamentation in an Arctic charr Salvelinus alpinus population was examined. Interrelationships between the intensities of parasites with different dietary transmission routes suggested that hosts were segregated by habitat and diet preferences. That is, intensities of parasites transmitted through intermediate hosts that share habitat were, in both sexes, positively interrelated. Negative interrelationships in the intensities of parasites transmitted through different hosts were found only in females. The intensity of breeding colouration was positively correlated with intensities of amphipod-transmitted parasites in both sexes. Intensities of parasites transmitted through copepods showed no relationship to morphological or ornamental characters. Seen in the context of morphology and ornaments of previously described sympatric populations of Arctic charr, these relationships suggested that individual variation in niche background may provide a source for the initial variation in morphology and secondary sexual traits leading to sympatric speciation through sexual selection. Abstract: Domestication and selection to enhance growth of animals is likely to affect social behaviour. Aggression may increase or decrease. A 4-generation selection programme on the salmonid species Arctic charr (Salvelinus alpinus L.) based on a well-defined population origin has made it possible to compare the social behaviour of its progeny with progeny from wild parents of the same population, thus excluding the effects of population differences. During 5 months, the within-group growth variation and specific, defined behaviour parameters were observed and analysed under experimental conditions where dominance hierarchies are promoted. In such conditions, which are opposite from farming conditions created to reduce dominance hierarchies, there was no difference in mean growth rate during the observation period and no difference in the social structure (number of dominant, subdominant and subordinate individuals) in the groups. Also, the level of within group aggression did not appear significantly different between level of domestication when observed between or during feeding periods. However, there was a significantly larger growth variation in the progeny of wild parents compared to the selected progeny. Also, the level of aggression increased significantly only in the groups with a selected origin during feeding events compared to periods in-between feeding. Individuals in the wild origin groups showed a significantly higher level of a passive low-profile behaviour compared to the selected origin groups. These differences suggest that selection for fast growth in farming conditions reduces the frequency of subordinate individual behaviour resulting in a smaller intra specific difference in growth in farm strains. Abstract: This study investigated the influence of nutritional status on the growth hormone (GH)/insulin-like growth factor-I (IGF-I) axis in Arctic charr (Salvelinus alpinus). The objectives were to study the regulation of GH secretion in vitro by somatostatin-14 (SRIF) and hIGF-I, and to determine whether pituitary sensitivity to these factors is dependent upon nutritional status. Arctic charr were fed at three different ration levels (0, 0.35, and 0.70% BWd−1), and pituitary glands were harvested at 1, 2, and 5 weeks for in vitro study. Both SRIF and hIGF-I inhibited GH secretion from Arctic charr pituitary tissue in long-term (18h) static hemipituitary culture, as well as after acute exposure in a pituitary fragment perifusion system. This response appeared to be dose-dependent for SRIF in static culture over the range of 0.01–1nM, but not for hIGF-I. The acute inhibitory action of hIGF-I on GH release in the perifusion system suggests an action that is initially independent of any effects on GH gene expression or protein synthesis. Nutritional status did not affect the sensitivity of Arctic charr pituitary tissue to either SRIF or hIGF-I in vitro, indicating that changes in abundance of pituitary SRIF or IGF-I receptors may not explain the alterations in plasma GH levels found during dietary restriction. Numbers of studies in externally fertilizing fish species provide evidence for an effect of ovarian fluid on sperm motility characteristics such as duration of forward mobility, velocity or percent motile sperm cells. Yet, because of variations among females in the quality of their ovarian fluid, such effects might differ between individuals. Additionally, ovarian fluid from different females could also be expected to affect each ejaculate differently, resulting in cryptic female choice. In this study on Artic charr (Salvelnius alpinus), sperm velocity from several males was measured in the diluted ovarian fluid of several females according to a fully balanced crossing design. This design allowed us to estimate variations among females in the effect of their ovarian fluid on the velocity of sperm from different males, and to detect variations among males in the ability of their sperm to swim in ovarian fluid. Sperm velocity was estimated by computer-assisted sperm analysis. Average velocity was found to vary among females, with some females having constantly higher velocity measurements in their ovarian fluid, and among males, indicating that some males had overall faster sperm in ovarian fluid than others. Moreover, variation in sperm velocity was shown to depend on individual female-male interactions. Our results document that females vary in the effect of their ovarian fluid on sperm velocity and that their ovarian fluid may stimulate sperm velocity according to individual characteristics of males. To examine the immunological impacts of polychlorinated biphenyls (PCBs) in an environmentally relevant way, we orally contaminated Arctic charr (Salvelinus alpinus) with Aroclor 1254. After contamination, fish were either fed (0 and 100 mg Aroclor 1254 kg-1 fish wt) or fasted (0, 1, 10, and 100 mg kg-1) to mimic cycles of feeding--fasting experienced by Arctic animals. After four months, PCB concentrations in muscle were the same in fasted and fed fish; however, PCBs in kidneys of fed fish were 33 to 50% of those in fasted fish. Arctic charr were exposed to Aeromonas salmonicida, the bacteria responsible for furunculosis, by cohabitation with infected conspecifics. Fasted fish had a significant trend toward lower survival with higher dose of PCBs--from 68% in controls to 48% in treatment involving 100 mg kg-1. Independent of PCB contamination, fed fish had the lowest survival; we attribute this to stress associated with establishing and maintaining feeding hierarchies. A significant decrease in the activity of lysozyme was observed in skin mucus, as was hemagglutination ability of a putative rhamnose lectin in fasted, but not in fed, PCB-treated fish. These results demonstrate the immunosuppressive effects of PCBs on Arctic charr, and they illustrate the importance of considering environmentally relevant nutritional status in ecotoxicological studies. This paper introduces a new approach to incorporating time- dependent overdispersion for Poisson-related regression models. To handle the added flexibility in conditional heteroskedasiicity in time series count data models, some well-known estimators are adapted. and a generalized method of moments (GMM) estimator is suggested. Abstract: A specific sex steroid-binding protein (SBP) is believed to be involved in regulation of circulating sex steroids, steroid delivery to target cells and intracellular signalling in sex steroid-sensitive tissues. In the present work, interactions between xenoestrogens and the plasma SBP in Arctic charr (Salvelinus alpinus L.) were determined using ligand–protein binding studies. The test compounds were all able to displace tritiated 17β-estradiol (E2) from the Arctic charr SBP (acSBP) in a competitive and dose-dependent manner. The acSBP affinities for the xenoestrogens ranged over several orders of magnitude (17β-estradiol≫ethynylestradiol (EE2)>zearalenone (ZEA)>diethylstilbestrol (DES)>genistein (GEN)>bisphenol A (BPA), 4-t-octylphenol (OP)≫o,p′-DDT, and dieldrin (DIN)), but were consistently lower than that of 17β-estradiol (about 4×102–106-fold less potent). The relative binding affinity (RBA) for selected chemicals were independent of both gender, age and maturation status, as well as variations of acSBP binding affinity. The affinity of endogenous steroids and estrogen mimics for the acSBP shows a high correlation to the affinity for the rainbow trout SBP, thus suggesting a phylogenetically conserved ligand-binding site between closely related species. Furthermore, it is argued that interaction with the acSBP- and SBP-mediated processes may introduce novel pathways for endocrine disruption, which may work in concert with the classical receptor-mediated effects. Differences in gonadal development and time of spawning were found in two coexisting morphotypes of Arctic charr Salvelinus alpinus in Kalandsvatnet, Norway. The annual variability in growth and life history traits of brown trout (Salmotrutta L.) and Arctic charr (Salvelinus alpinus (L.)) in Lake Atnsjøen, a Norwegian subalpine lake, was studied over a period of 13 years (1985–1997). The extent to which life-history characteristics recorded on one occasion can be regarded as representative for the population was explored. We found inter-cohort variation in growth for both species; estimates of asymptotic length (L∞) in ten cohorts ranged between 225–305 mm (CV = 10.5%) for brown trout and 273–301 mm (CV = 4.1%) for Arctic charr. However, this variation was much lower than inter-population variation for brown trout based on single samples from 169 populations (CV = 24.6%). In Lake Atnsjøen, annual growth increment correlated highly with the number of days warmer than 7 °C (R2=0.60–0.89) for brown trout, and days warmer than 10 °C (R2=0.40–0.58) for Arctic charr. Females of Arctic charr were younger at sexual maturity than males, while no such difference was found in brown trout. Generally speaking, early maturing individuals of both species grew faster, particularly from age-2 and onwards, than immature individuals. Early maturing individuals, however, were smaller at maturity than those maturing one year older. The impacts of polychlorinated biphenyl (PCB) exposure on smoltification and subsequent seawater performance were investigated in hatchery-reared, anadromous Arctic charr (Salvelinus alpinus). The fish were subjected to a 2-month summer seawater residence, after which they were orally dosed with 0 (Control, C), 1 (Low Dose, LD) or 100 mg Aroclor 1254 kg−1 body mass (High Dose, HD) in November. They were then held in fresh water, without being fed (to mimic their natural overwintering in freshwater), until they had smolted in June the next year. The smolts were then transferred to seawater and fed to mimic their summer feeding residence in seawater, followed by a period without food in freshwater from August until maturation in October. Compared with C and LD charr, the HD charr had either a transient or a permanent reduction in plasma growth hormone, insulin-like growth factor-1, and thyroxin and triiodothyronine titers during the period of smoltification. These hormonal alterations in the HD charr corresponded with impaired hyposmoregulatory ability in May and June, as well as reduced growth rate and survival after transference to seawater. Consequently, fewer fish in the HD group matured in October compared to the other two treatments. The HD fish had a liver PCB concentration ranging between 14 and 42 mg kg−1 wet mass, whereas there were similar, and very low, liver PCB concentrations in LD and C fish throughout the smolting period. The effects of induced water level fluctuations and introduction of the mysid Mysis relicta Lovén on population structure of brown trout, Salmo trutta L., and Arctic charr, Salvelinus alpinus (L.), were studied during 1953–1995 in Limingen hydroelectric reservoir, Norway. The main response was a marked reduction in catch-per-unit-effort (CPUE) for trout and charr, probably caused by reduced recruitment following increased variation in water level. For both species, mean length decreased until 1967 and increased thereafter, whereas mean mass-at-length increased for the whole period. Both length and mass-at-length were negatively correlated with CPUE. The increases in mean length and mass-at-length were probably because of reduced competition following the reduced recruitment. Mysis relicta has become an important food item for charr but not for brown trout, but the increases in mean length and mass-at-length of charr started prior to the appearance of M. relicta in the charr diet. No difference in attraction was observed in sibling Artic charr Salvelinus alpinus between water scented by dominant or subordinate major histocompability complex (MHC) identical fish observed in a two-choice fluviarium. In a second experiment, MHC identical sibling donors were given different types of food pellets before the preference test. The test fish showed a significant attraction to the sibling given the same kind of food as the test fish itself during the first 6 h of the fluviarium tests. Habitat use, growth and food composition of native and stocked Arctic charr, Salvelinus alpinus (L.), were studied in the subarctic Lake Muddusjärvi, northern Finland, to investigate reasons for poor stocking success. Samples were collected with pelagic and epibenthic gill nets. Stocked and native charr occurred in similar epibethic habitats, whereas pelagic habitat was avoided. Native charr grew fast after shifting to piscivory. Growth rate of stocked charr was slow because only a small proportion of stocked fish became piscivorous during the first year after stocking. During the first lake year, stocked charr divided into slow-growing planktivores and fast-growing piscivores. Piscivorous stocked and native charr consumed only whitefish, Coregonus lavaretus (L.), as their prey. Small-sized (<10 cm) whitefish were preferred when shifting to piscivory. Populations of heterotrophic bacteria present in the hindgut region of Arctic charr Salvelinus alpinus L. fed dietary soybean, linseed and marine oils before challenge with Aeromonas salmonicida ssp. salmonicida and marine oil after challenge were estimated using the dilution plate technique. There were differences in bacterial composition between the rearing groups before and after challenge, as well as interindividual variations. For example, carnobacteria were only isolated from the hindgut region of fish fed soybean oil and linseed oil before challenge, whereas Carnobacterium spp. and Carnobacterium funditum-like species were isolated from fish fed the same oils after challenge. Three non-motile Aeromonas spp. were isolated from infected fish fed marine oil. One of these isolates was identified as identical to A. salmonicida ssp. salmonicida used in&the challenge test by microbial fingerprinting (amplified fragment length polymorphism). Electron microscopic examinations of hindgut regions demonstrated substantial numbers of bacterial cells associated with enterocytes, but bacterial colonization of the enterocyte surface varied between different rearing groups. The potential of bacteria found associated with the hindgut region to inhibit the fish pathogens A. salmonicida, Vibrio salmonicida and Vibrio anguillarum differed between rearing groups. Like the spermatozoa of most other fish species spawning in fresh water, Arctic charr Salvelinus alpinus sperm were short-lived (mean 42 s) after activation and their swimming speed declined rapidly during this period, e.g. from a mean speed of 106 μm s−1 at 10 s after activation in fresh water to 21 μm s−1 only 20 s later. Ovarian fluid significantly influenced sperm longevity (duration of forward mobility), per cent motility, swimming speed and the linearity of sperm movement. All of these variables generally increased as the concentration of ovarian fluid increased from 0 to 50%, even though ovarian fluid is more than three times as viscous as fresh water. It is concluded that ovarian fluid enhances sperm movement in this species and thus has the potential to influence both fertilization success and the outcome of sperm competition. Arctic charr, Salvelinus alpinus, and brook charr, Salvelinus fontinalis, inhabiting three lakes in the de la Trinité River and adjacent watershed, north shore, Gulf of St. Lawrence, were sampled in 1998. Arctic charr growth differed among lakes with the smallest fish coming from the largest lake. Arctic charr weight–length equation exponents were almost identical at all sample sites. Brook charr growth was also similar in all lakes. July stomach samples from Arctic charr consisted almost entirely of cladocerans in the largest lake, less so in the intermediate sized lake and were mostly aquatic insects in the smallest lake. Brook charr stomach contents were more varied and included fish. Carbon, nitrogen and sulphur stable isotope analyses (SIA) were used to provide a spatially and temporally integrated image of charr diets. SIA corroborated observed among-lake differences and similarities in species diets and suggested lake morphometry may influence measured results. The δ 15N signature in brook charr muscle increased with fork-length, as a result of a shift towards piscivory with size. The δ 15N signature in Arctic charr muscle tissue showed a significant negative correlation with fork-length in two of the studied lakes that appears related to dietary niche shifts. A long-term field study of a perturbed host–helminth system provides indirect evidence that a long-lived swimbladder nematode, Cystidicola farionis, induces mortality of Arctic charr, Salvelinus alpinus. The prevalence and abundance of this parasite has changed little over the period from 1987 to 1999. The cumulative numbers of L3-stage larvae steadily increased with increasing host age, indicating a continuous exposure to infection throughout the life of the target fish host. Indirect methods, which used data pooled over years and long-term cohort analyses, indicate that parasite-induced host mortality (PIHM) occurs in hosts older than 10 years. Furthermore, using a short-term cohort method adjusted for worm recruitment, we found indications of PIHM occurrence even in younger age groups. These patterns do not seem to be caused by high parasite mortality rates since dead worms are rarely observed inside the swimbladder. Age-related changes in infection rates or in resistance to infection seem to play only a minor role as there were only slight changes in the preference of charr for feeding on amphipods (which are intermediate hosts) and in the acquisition rate of L3 larvae in older hosts. The serotonin1A (5-HT1A) receptor agonist 8-hydroxy-2-(di-N-propylamino)tetralin (8-OH-DPAT) was administrated to juvenile Arctic charr either by intraperitoneal (i.p.) injections or through a permanent i.p. implanted catheter. 8-OH-DPAT induced an elevation of plasma cortisol in non-stressed fish (receiving the drug through a permanent i.p. catheter), whereas 8-OH-DPAT had the opposite effect when administrated through a standard i.p. injection, dampening the rise in cortisol induced by handling and injection, an effect which was most pronounced at low doses of 8-OH-DPAT. In both cases, plasma levels of adrenocorticotropic hormone followed the same general pattern as plasma cortisol. The results suggests that 5-HT1A receptors are involved in the regulation of hypothalamic–pituitary–interrenal axis activity in Arctic charr, but the effect of 5-HT1A stimulation seems to vary depending on the state of the fish. Planktivorous and benthivorous morphs of wild Arctic charr Salvelinus alpinus from Loch Rannoch, Perthshire, Scotland, reared in the laboratory from artificially fertilized eggs under identical conditions, were morphologically distinct (based on a series of head measurements) from an early age. For some morphometric characters, these differences became more marked with increasing body size, reflecting differences in the allometric growth patterns of the two forms. These data show that the observed phenotypic differences between these two trophic variants were, to some extent at least, inherited. Thus it is concluded that the mechanisms regulating expression of phenotype in the sympatric morphs of Loch Rannoch are significantly different from the environmentally determined body size polymorphisms reported from Norway but are closer to the more completely divergent forms from Thingvalavatn, Iceland. Abstract The effects of temperature and salinity on the reproductive success of Arctic charr, Salvelinus alpinus (L.), were examined by holding broodstock under the following conditions from mid-May until the end of September: fresh water at ambient temperature (NFW; 8–16 °C); salt water (25–30‰) at ambient temperature (NSW; 4–10 °C); fresh water cooled to saltwater temperature (CFW; 4–10 °C); or salt water heated to freshwater temperature (HSW; 8–16 °C). The relative fecundity of females was similar among groups (P > 0.05; 2685 ± 706 eggs), but females reared in NSW produced significantly larger eggs than those raised in NFW. The highest spermatozoa concentrations were found in milt from males reared in SW and the highest milt glucose concentration was from males kept under coldwater conditions (CFW, NSW). Eggs from NSW and HSW females contained more proteins than eggs produced by NFW females. Eggs from NSW females also contained 40% more lipids than was observed in the other groups, and total energy content was 27% higher in eggs from NSW females than in eggs from NFW females. When FW was cooled (CFW), females produced eggs with protein contents similar to those in NSW, but the lipid contents remained 30% lower. Finally, the best survival at the eyed stage and at hatch was observed in families produced by NSW broodstock. Intermediate values were observed in families from NFW or CFW while the highest mortality occurred in families from the HSW group. All these results suggest that, under the experimental conditions used in the present study, coastal seawater conditions offered the most favourable summer rearing conditions with respect to the reproductive success of Arctic charr. Summary: An atypical strain of Carnobacterium divergens, strain 6251, was isolated from the small intestine of Arctic charr (Salvelinus alpinus L.), fed high dietary carbohydrate. This strain showed marked growth inhibitory effects in vitro against the fish pathogens Aeromonas salmonicida subsp. salmonicida(furunculosis), Vibrio anguillarum(vibriosis) and Vibrio viscocus(winter ulcer). The strain is a non-motile Gram-positive psychrotrophic rod that lacks both catalase and oxidase, grows at pH 9.1 (CTAS agar), but not on acetate containing media (pH ≤ 5.4), on TCBS or at ≤6% sodium chloride content. Strain 6251 is facultatively anaerobic and utilises tryptone as a sole source of nutrient. Further characterisation showed the most abundant cellular fatty acid of strain 6251 to be oleic acid (18:1) (n-9) (36.0%). Sequencing of a 16S rDNA region of 578 nucleotides and AFLPTM microbial fingerprinting suggested that strain 6251 is not closely related to any carnobacteria known, however, DNA-DNA similarity determinations showed high similarity (96.2%) with the type strain of Carnobacterium divergens. The growth of Arctic charr Salvelinus alpinus in five high altitude lakes differed between lakes during the first 2 years but was similar thereafter. The average temperature during the summer had a direct effect on growth and an indirect effect on the productivity level of the lakes, particularly on the abundance of planktonic crustaceans. Stable isotopes of carbon and nitrogen were used to examine differences in the feeding ecology of sympatric morphotypes of Arctic charr Salvelinus alpinus from Lake Hazen, Ellesmere Island, in the Canadian High Arctic. Large and small morphotypes possessed significantly different carbon and nitrogen signatures with large-form Arctic charr being more depleted in 13C and more enriched in 15N than the small-form. Isotope and stomach content analyses yielded consistent results and indicated short- and long-term reliance on fish as a food for large Arctic charr. Large-form individuals predate on juveniles but do not predate on small-form individuals ≥250 mm. The observed cannibalism by large-form individuals, therefore, does not act to maintain the bimodal length-frequency distribution in Lake Hazen. Bimodality is argued to arise for ecological reasons connected with differing habitat use by the morphotypes and the associated differences in resource consumption opportunities. Arctic charr (Salvelinus alpinus L.) were fed diets based on a commercial recipe supplemented with either linseed, soybean or marine oil prior to cohabitant challenge with Aeromonas salmonicida ssp. salmonicida. Mortality varied significantly between the three dietary groups. Highest mortality (48%) was observed in fish fed the marine oil and the lowest mortality (20%) was in the group fed soybean oil. Transmission electron microscopy (TEM) examination of the digestive tract of uninfected fish demonstrated substantial numbers of bacterial cells between microvilli. However, only a few bacteria were recovered that were associated with the microvilli of infected fish. Immunocytochemical staining/labelling investigations using TEM and an immunogold method were performed on mid-gut segments of fish fed the marine oil diet and showed augmentation of goblet cells and the presence of A. salmonicida ssp. salmonicida in the gastrointestinal tract of diseased fish after challenge with the pathogen. It is suggested that the gastrointestinal tract could be an infection route of A. salmonicida ssp. salmonicida. The greater prevalence of goblet cells supports the suggestion that sloughing off mucus is a protective response against bacterial infections.
