With respect to salmonid aquaculture, one of the most important bacterial pathogens due to high mortality and antibiotic usage is the causative agent of typical furunculosis, Aeromonas salmonicida spp. salmonicida (Asal). In Atlantic salmon, Salmo salar , the host response during infections with Asal is well-documented, with furunculosis outbreaks resulting in significant mortality in commercial settings. However, less is known about the host-pathogen interactions in the emerging aquaculture species, Arctic charr Salvelinus alpinus. Furthermore, there is no data on the efficacy or response of this species after vaccination with commonly administered vaccines against furunculosis. To this end, we examined the immunological response of S. alpinus during infection with Asal , with or without administration of vaccines (Forte Micro®, Forte Micro® + Renogen®, Elanco Animal Health). Artic charr (vaccinated or unvaccinated) were i.p.-injected with a virulent strain of Asal (106 CFUs/mL) and tissues were collected pre-infection/post-vaccination, 8, and 29 days post-infection. Unvaccinated Arctic charr were susceptible to Asal with 72% mortalities observed after 31 days. However, there was 72–82% protection in fish vaccinated with either the single or dual-vaccine, respectively. Protection in vaccinated fish was concordant with significantly higher serum IgM concentrations, and following RNA sequencing and transcriptome assembly, differential expression analysis revealed several patterns and pathways associated with the improved survival of vaccinated fish. Most striking was the dramatically higher basal expression of complement/coagulation factors, acute phase-proteins, and iron hemostasis proteins in pre-challenged, vaccinated fish. Remarkably, following Asal infection, this response was abrogated and instead the transcriptome was characterized by a lack of immune-stimulation compared to that of unvaccinated fish. Furthermore, where pathways of actin assembly and FcγR-mediated phagocytosis were significantly differentially regulated in unvaccinated fish, vaccinated fish showed either the opposite regulation (ForteMicro®), or no impact at all (ForteMicro®Renogen®). The present data indicates that vaccine-induced protection against Asal relies on the pre-activation and immediate control of humoral immune parameters that is coincident with reduced activation of apoptotic (e.g., NF-κB) and actin-associated pathways. The Swedish breeding programme for Arctic charr aims to improve traits that are important for farming. Selection has been ongoing since the 1980s, and the aim of this study was to re-estimate the genetic parameters in the selected population. In addition, the genetics of skin pigmentation was studied, as previous work indicated a relationship between pigmentation and the ability to cope with stress. One hundred twenty-five full sib families of Arctic charr were hatched and reared separately until they were individually tagged (n = 12,266). In connection with the tagging, three replicates were formed, which were subsequently reared at different sites. The genetic relationships were known from a seven-generation pedigree. Body weight and length were at one site measured at six occasions until 27 months old, and these measurements were used to estimate the DGC (daily growth coefficient) and condition factor. At a second site fish were measured at 21 and 27 months age and at a third site at 27 months age. Skin pigmentation was measured using image analysis. The final weight at 27 months of age varied widely between rearing sites, ranging from 529 g to 1443 g. Weight heritability differed among the sites, at 0.45, 0.11, and 0.07. At one of the sites, where growth was followed closely, weight heritability varied among growth periods, ranging from 0.01 to 0.25, and similar results were obtained for the DGC. In contrast, condition factor heritability did not show such pronounced variations within and among sites. The magnitude of the differences in heritability of weight and growth among different sites that was observed here was not observed previously with this charr population. In particular, the low heritability among the fastest growing fish is unprecedented. Skin pigmentation showed a high heritability of 0.76, indicating a strong additive genetic component. At most ages, pigmentation showed a weak positive genetic correlation with growth. The genotype environment interaction (GEI) was estimated using genetic correlations among sites. For the final weight, genetic correlations were high, indicating weak GEI, although the presence of GEI was suggested from the DGC data. The evaluation of new diet composition is commonly achieved by performing time-consuming growth trials, which may negatively impact the welfare of a large number of fish if the feed is not accepted. Instead, the fish's behavioural responses to a new diet composition can be used as a first step in the evaluation of new diet composition. The taste acceptance of a new diet by Arctic charr ( Salvelinus alpinus ) was evaluated over 16 days based on the self-selection of a test diet and a control diet. The test diet contained ingredients from the nutrient-enriched Baltic Sea, and it is hoped that this diet can contribute nutrients to the nutrient-poor waters in northern Sweden in which Arctic charr are farmed. The main ingredients in the test feed were blue mussel meat ( Mytilus edulis ), meat from sprat ( Sprattus sprattus ) and herring ( Clupea harengus ), baker's yeast ( Saccharomyces cerevisiae ), fish oil and regionally produced rapeseed oil. Individual fish ( n = 15) were allowed to choose between abundances of both the test diet and a fishmeal based control diet that mimicked a standard feed for Arctic charr. After only a few days, the fish demonstrated a significant preference for the Baltic Sea test feed. These results may depend on betaine, which was found at concentrations that were four times higher in the test feed than in the control feed. Betaine is a known attractant for many fish species and is abundant in marine animals, such as mussels. Thus, the test diet can be evaluated further without additional taste stimulants before the new feed formula can be used commercially to ensure fish welfare, the product quality and the economic feasibility of the new formula. We investigated whether Fraser strain Arctic charr Salvelinus alpinus L. reared in fresh water ( FW) differed in survival and growth from full siblings transferred to brackish water ( BW) and whether family performance was consistent throughout 1 year of commercial growth. Fish were grown communally ( n = 3600) in either FW or BW from May 2008 to May 2009. Mean family survival in BW was moderately correlated with mean family survival in FW ( r = 0.342; P = 0.032). Fish which survived until the conclusion of the experiment were significantly heavier in body weight at the baseline assessment than their full-sibs which died. Family by treatment interactions were detected for body weight, condition factor and specific growth across all measurement dates (May 2008, October 2008 and May 2009) and growth intervals. Despite the varied response of families to treatment, mean family body weight in FW was correlated phenotypically with BW body weight in October 2008 ( r = 0.633, P < 0.0001) and marginally correlated in May 2009 ( r = 0.289, P = 0.061). These data suggest that body weight in FW and body weight in BW should be analysed as separate, but correlated traits in Arctic charr breeding programs. [ABSTRACT FROM AUTHOR]

Behavioural traits have been shown to have implications in fish welfare and growth performances in aquaculture. If several studies have demonstrated the existence of repeatable and heritable behavioural traits (i.e., animal personality), the methodology to assess personality in fishes is often carried out in solitary context, which appears to somewhat limit their use from a selective breeding perspective because these tests are too time consuming. To address this drawback, group-based tests have been developed. In Nordic country, Arctic charr (Salvelinus alpinus) is widely used in aquaculture, but no selection effort on behavioural traits has yet been carried out. Specifically, in this study we examined if risk-taking behaviour was repeatable and correlated in group and solitary context and if the early influences of physical environment affect the among-individual variation of behavioural trait across time in order to verify whether a group risk-taking test could be used as a selective breeding tool. Here, we found that in both contexts and treatments, the risk-taking behaviour was repeatable across 7 days. However, no cross-context consistency was found between group and solitary, which indicates that individual Arctic charr express different behavioural trait in group and solitary. • Risk-taking behaviour was assessed in the Arctic charr. • Fish were raised in plain versus enriched environments. • Risk-taking were tested in solitary versus group contexts. • Risk-taking behaviour was repeatable across 7 days in both contexts. • No consistency was found between the solitaryand groupcontexts. Fatty acids (FA) of muscle tissue of Salvelinus species and its forms, S. alpinus, S. boganidae, S. drjagini, and S. fontinalis, from six Russian lakes and two aquacultures, were analyzed. Considerable variations in FA compositions and contents were found, including contents of eicosapentaenoic and docosahexaenoic acids (EPA and DHA), which are important indicators of fish nutritive value for humans. As found, contents of EPA+DHA (mg·g−1 wet weight) in muscle tissue of Salvelinus species and forms varied more than tenfold. These differences were supposed to be primarily determined by phylogenetic factors, rather than ecological factors, including food. Two species, S. boganidae and S. drjagini, had the highest EPA+DHA contents in their biomass and thereby could be recommended as promising species for aquaculture to obtain production with especially high nutritive value. Basing on revealed differences in FA composition of wild and farmed fish, levels of 15-17-BFA (branched fatty acids), 18:2NMI (non-methylene interrupted), 20:2NMI, 20:4n-3, and 22:4n-3 fatty acids were recommended for verifying trade label information of fish products on shelves, as the biomarkers to differentiate wild and farmed charr. The 'earthy' and 'muddy' off-flavours in pond-reared fish are due to the presence of geosmin or 2-methylisoborneol in the flesh of the fish. Similar off-flavours have been reported in fish raised in recirculating aquaculture systems (RAS); however, little information is available regarding the cause of these off-flavours. Our hypothesis was that earthy and muddy off-flavour compounds, found previously in pond-raised fish, are also responsible for off-flavours in fish raised in RAS. In this preliminary study, we examined water, biofilms in RAS and fillets from cultured arctic charr known to have off-flavours and requiring depuration using instrumental [solid-phase microextraction procedure and gas chromatograph-mass spectrometry (GC-MS)] and human sensory analyses. Geosmin was present in the samples taken from the biofilter and on the side walls of the tanks. Two-methylisoborneol was only found in low levels in the samples. The GC-MS results indicated the presence of geosmin in the fillets (705 ng kg), but lower levels were found in the water (30.5 ng L). Sensory analyses also detected an earthy flavour (i.e., geosmin presence) in the fillets, and, therefore, it appears that geosmin is the main compound responsible for the off-flavour in RAS. Results and experiences from a selective breeding programme aiming at improving the performance of an Arctic charr strain in aquaculture are presented. The programme, which has been running since 1985, uses traditional quantitative genetic methods based on relatedness and trait measurements. Traits considered included growth, age at sexual maturity, flesh colour, fat content and other features. Estimates in the early phase of the programme showed promising heritabilities for most traits, e.g. growth heritabilities of 0.34–0.52, and suggested that improvement by selection was feasible. Several traits such as a high rate of early maturation and poor flesh colour that were considered problematic by the Arctic charr farming industry at the programme’s start no longer hamper farming. Considerable improvement of growth by selection, estimated at 8% per generation, has contributed to shortening the production cycle in commercial farming. Results from studies of genotype–environment interactions are presented and discussed. Poor survival of fertilised eggs is a major problem in Swedish Arctic charr farming as indicated by presented survival rates. Efforts have consistently been made to avoid inbreeding in the selected strain, and at the present seventh generation the accumulated increase in inbreeding is estimated from pedigree data to be only approximately 5%. We present an overview of 27 years of experience of domestication and farming of Arctic charr ( Salvelinus alpinus) in Sweden. The domestication process included an evaluation of suitable strains for farming, a breeding programme and the study of the biological and behavioural characteristics of the species. Traits of three different Arctic charr populations differing in ecology and appearance were compared during initial 2-year trials under farming conditions. The best-performing population with respect to the growth rate and the lowest frequency of early sexual maturation was a piscivore form and this became the foundation for a breeding programme intended to select for an Arctic charr strain suitable for farming. After 23 years and 7 generations, our selective breeding has resulted in a fast-growing, late-maturing strain much appreciated by farmers. The biological and behavioural characteristics studied included annual and diel locomotor activity, feeding, social and thermal behaviour. Applying our findings in these areas has greatly improved both profits and conditions for the fish. Other investigations have focused on the application and further evaluation of the results from research in practical farming trials, such as evaluation of growth at different farms with different temperature conditions, optimal time and stocking density for start-feeding and evaluation of different feeding schedules. In Sweden, Arctic charr is mainly farmed in net pens situated in nutritionally depleted and extremely unproductive water reservoirs formed by damming rivers to create electric power. Judicious farming of Arctic charr in such reservoirs can restore their nutritional and productivity state to that which existed before regulation. Site selection criteria for Arctic charr farming in such waters have been developed. The development of intensive farming of Arctic charr in Sweden is discussed together with current limitations and future possibilities. [ABSTRACT FROM AUTHOR]

The growth in captivity of a wild Welsh strain (Llyn Cowlyd) of Arctic charr Salvelinus alpinus was compared to that of a commercial Scottish strain over the course of a 12 month experiment. At the conclusion of the study, the Welsh strain had a lower mass and condition factor than the commercial fish, but a similar length. Aspects of the growth and population structure of the Welsh strain imply that it could be a subject for aquaculture development, but such practises will be dependant on further work on selection and broodstock development.
