Anthocyanins are the main plant pigments responsible for the color of flowers, fruits, and vegetative organs of many plants, and are applied also as safe food colorants. They are efficient antioxidants. In this review, the reactivity of anthocyanins and their aglycones, anthocyanidins, in the main antioxidant assays, and their reactions with reactive oxygen and nitrogen species, effects of interactions with other compounds and metal ions on the antioxidant activity of anthocyanins and the electrochemical properties of anthocyanins are presented. Numerous cases of attenuation of oxidative stress at the cellular and organismal levels by anthocyanins are cited. The direct and indirect antioxidant action of anthocyanins, the question of the specificity of anthocyanin action in complex extracts, as well as limitations of cellular in vitro assays and biomarkers used for the detection of antioxidant effects of anthocyanins, are critically discussed.

Antioxidants remain interesting molecules of choice for suppression of the toxic effects of free radicals in foods and human systems. The current practice involves the use of mainly synthetic molecules as potent antioxidant agents. However, due to the potential negative impact on human health, there is an intensive effort within the research community to develop natural alternatives with similar antioxidant efficacy but without the negative side effects of synthetic molecules. Still, the successful development of new molecules depends on the use of reliable chemical or cell culture assays to screen antioxidant properties. Chemical antioxidant assays include the determination of scavenging ability against free radicals such as DPPH, superoxide anion radicals, hydroxyl radicals, hydrogen peroxide, and nitric oxide. Other antioxidant tests include the ability of compounds to bind and sequester prooxidant metal cations, reduce ferric iron, and attenuate the rate of lipid oxidation. Ex vivo tests utilize cell cultures to confirm entry of the molecules into cells and the ability to quench synthetic intracellular free radicals or to stimulate the increased biosynthesis of endogenous antioxidants. In order to assist researchers in their choice of antioxidant evaluation methods, this review presents background scientific information on some of the most commonly used antioxidant assays with a comparative discussion of the relevance of published literature data to food science and human nutrition applications.

Sinapine is the main secondary metabolite present in rapeseed pomace (RSP) with its concentration being dependent on rapeseed processing, growing conditions, extraction parameters and the country of origin. Here we report, the concentration of sinapine from an extract of defatted RSP harvested in the North East of Scotland. Using liquid chromatography tandem mass spectrometry, the most abundant phenolic compound in the RSP extract was, as expected, sinapine (109.1 mg/g RSP extract). Additionally, sinapic, caffeic, ferulic and syringic acids were identified (0.159-3.91 mg/g RSP extract). Sinapine together with the phenolics at the concentration present in the RSP extract, exhibited ≥50% activity relative to the extract in antioxidant assays. Furthermore, sinapine provided plasmid DNA (pBR322) protection, from 2,2'-azobis(2-amidinopropane) dihydrochloride and inhibited acetylcholinesterase activity by 85%. Molecular docking was utilised to explain the inhibitory activity. The antioxidant capacities of Kyoho skin, seed, and flesh extracts were determined using DPPH and ABTS assays and a suitable statistical program was tested for the prediction of EC 50 values of Kyoho skin, seed, and flesh extracts obtained by DPPH and ABTS assays. Statistical programs: OriginPro® version 8, GraphPad Prism® version 7, GraphPad Prism® version 7 five-parameter (5P) logistic model, SigmaPlot® version 14.0, Microsoft Excel® version 2016, interpolation log, and direct interpolation methods were employed to estimate the EC 50 values. Linear regression and direct interpolation results showed deviations for non-linear models in the prediction of EC 50 values of grape extracts. The correlation results of grape flesh extracts demonstrated a positive correlation (r = ≥ 0.95) between EC 50 values of antioxidant assays. Therefore, non-linear methods were appropriate for EC 50 calculations. Interest in the content of natural antioxidants in plant-based foods can be from the human health perspective, in terms of how these compounds might help promote one's health and wellness, or from the storage point-of-view, as the endogenous antioxidant constituents aid to extend a foodstuff's shelf-life. This chapter reports essential information about the mechanism of antioxidant action and methods employed for determination of their activity, classes of phenolic compounds (phenolic acids, flavonoids, lignans, stilbenes, tannins), sources of plant antioxidants (oil seeds, cereals, legumes, plants of the Lamiaceae family, tea and coffee, tree nuts, fruits, and berries), extraction strategies of phenolic compounds from plant material, and the influence of processing and storage on the content of natural antioxidants in foods and their antioxidant activity. Thermal processing, if not releasing bound phenolics from the structural matrices of the food, tends to decrease the antioxidant potential or, in the best case scenario, has no significant negative impact. Gentler sterilization processes such as high-pressure processing tend to better retain the antioxidant potential of a foodstuff than thermal treatments such as steaming, boiling, or frying. Probiotics prove useful in correcting and preventing numerous health conditions, including those having severe impact on society, e.g., obesity and cancer. Notably, these capabilities of probiotics appear to be associated with their antioxidant properties. The mechanisms of antioxidant action of probiotics range from immediate biochemical scavenging of reactive substances to induction of signaling events leading to increased capacity of the host's cytoprotective systems. Since the antioxidant effects of probiotics significantly vary in types and details, a broad selection of methods of assessment of these properties is required in order to identify, characterize, and develop novel probiotics for medical purposes, as well as to explain the mechanisms of action of probiotics already in use in healthcare. This review revises the versatile toolbox, which can be used to assess the antioxidant properties of probiotics.

Antioxidants have been the subject of intense research interest mainly due to their beneficial properties associated with human health and wellbeing. Phenolic molecules, such as naturally occurring Resveratrol and Vanillin, are well known for their anti-oxidant properties, providing a starting point for the development of new antioxidants. Here we report, for the first time, the synthesis of a number of new vanillin through the reductive amination reaction between vanillin and a selection of amines. All the compounds synthesised, exhibited strong antioxidant properties in DPPH, FRAP and ORAC assays, with compounds 1b and 2c being the most active. The latter also demonstrated the ability to protect plasmid DNA from oxidative damage in the presence of the radical initiator AAPH. At cellular level, neuroblastoma SH-SY5Y cells were protected from oxidative damage (H 2 O 2 , 400 μM) with both 1b and 2c. The presence of a tertiary amino group, along with the number of vanillin moieties in the molecule contribute for the antioxidant activity. Furthermore, the delocalization of the electron pair of the nitrogen and the presence of an electron donating substituent to enhance the antioxidant properties of this new class of compounds. Most of the fast in vitro assays proposed to determine the antioxidant capacity of a compound/extract lack either biological context or employ complex protocols. Therefore, the present work proposes the improvement of an agarose gel DNA electrophoresis in order to allow for a quantitative estimation of the antioxidant capacity of pure phenolic compounds as well as of a phenolic rich extract, while also considering their possible pro-oxidant effects. The result obtained demonstrated that the proposed method allowed for the evaluation of the protection of DNA oxidation [in the presence of hydrogen peroxide (H 2 O 2 ) and an H 2 O 2 /iron (III) chloride (FeCl 3 ) systems] as well as for the observation of pro-oxidant activities, with the measurements registering interclass correlation coefficients above 0.9. Background: The secondary metabolites of the Artemisia genus are well known for their important therapeutic properties. This genus is one of the valuable sources of flavonoids and other polyphenols, but due to the low contents of these important metabolites, there is a need to either enhance their concentration in the original plant or seek alternative sources for them. The aim of the current study was to detect and enhance the yield of antioxidant compounds of Artemisia carvifolia Buch. HPLC analysis was performed to detect the antioxidants. With the aim of increasing flavonoid content, Rol gene transgenics of A. carvifolia were established. Two genes of the flavonoid biosynthetic pathway, phenylalanine ammonia-lyase and chalcone synthase, were studied by real time qPCR. Antioxidant potential was determined by performing different antioxidant assays., Results: HPLC analysis of wild-type A. carvifolia revealed the presence of flavonoids such as caffeic acid (30 μg/g DW), quercetin (10 μg/g DW), isoquercetin (400 μg/g DW) and rutin (300 μg/g DW). Compared to the untransformed plants, flavonoid levels increased 1.9-6-fold and 1.6-4-fold in rol B and rol C transgenics, respectively. RT qPCR analysis showed a variable expression of the flavonoid biosynthetic genes, including those encoding phenylalanine ammonia-lyase and chalcone synthase, which were found to be relatively more expressed in transformed than wild-type plants, thus correlating with the metabolite concentration. Methanolic extracts of transgenics showed higher antioxidant capacity, reducing power, and protection against free radical-induced DNA damage. Among the transgenic plants, those harboring rol B were slightly more active than the rol C-transformants., Conclusion: As well as demonstrating the effectiveness of rol genes in inducing plant secondary metabolism, this study provides insight into the molecular dynamics of the flavonoid accumulation pattern, which correlated with the expression of biosynthetic genes.

The leaves of Linderae aggregate (LAL) has been used as a type of tea in China and other Southeast Asian countries. In this study, 11 polyphenols in LAL were clarified for the first time using multiple high-performance liquid chromatographic techniques. An optimal extracting method was developed through the comparison of the amount of quercetin-3-O-α-L-rhamnoside using a uniform design method. From the fingerprint liquid chromatography data, 11 common peaks in the 8 samples collected from April to November were semi-determined. The antioxidant capacities were examined using the 2,2-diphenyl-1-picrylhydrazyl free radical scavenging assay and the ferric reducing/antioxidant power assay. All 8 samples contained the same 11 polyphenols in similar ratios. Three samples, S2, S5 and S6 contained higher amount of quercetin-3-O-α-L-rhamnoside and were demonstrated to have stronger antioxidant capacities in both antioxidant assays. Antioxidant potential of four methanol extracts from three selected plant species, namely Salvia nubicola (Lamiaceae), Acer oblongifolium (Aceraceae) and Hedera nepalensis (Araliaceae) was measured using assays in aqueous and lipid systems. Antioxidant activities were investigated in aqueous systems by using DPPH radical-scavenging assay, ABTS radical-scavenging assay and DNA protection assay, while antioxidant activity in a lipid system was determined by using the thiobarbituric acid-reactive substances (TBARS) assay. Additionally, the Folin-Ciocalteu method was used to measure total phenolic content. Methanol extracts of leaves and flowers of S. nubicola showed the highest Trolox equivalent (TE) values in the case of the DPPH assay, 2484±4.9mmol TE/g extract, as well as total phenolic content, 139±0.2mg gallic acid equivalents/g extract. Three fractions (A-C) of the methanol extract of S. nubicola leaves and flowers were produced by semi-preparative HPLC. Fraction B was found to be the most active in the DPPH radical-scavenging assay and had the highest total phenol content. HPLC-DAD and LC-MS revealed rosmarinic acid in S. nubicola extracts and chlorogenic acid and rutin in H. nepalensis extracts as the main phenolic antioxidants., (Copyright © 2011 Elsevier Ltd. Our group has seen great promise in using substituted diaryl-hydrazones to alleviate oxidative stress in preeclampsia. Specifically, fluorinated diaryl-hydrazones have shown great efficacy, confirmed via antioxidant assays and animal trials using pregnant mice. In addition to efficient antioxidant properties, these diaryl-hydrazones are also considered non-toxic. While the synthesis of these compounds is relatively simple, it commonly utilizes undesirable solvents and glacial acetic acid as the catalyst; additional solvents are needed for the isolation of the desired products, which negatively affects the green synthesis of the hydrazones. To combat this possible industrial roadblock, we have begun incorporating the use of hydrostatic high pressure (HHP) in the synthesis. The use of HHP allowed us to synthesize substituted diaryl-hydrazones in a 1:1 molar ratio without the need for solvents or acid catalysts. The optimized procedure can produce nearly quantitative yields, leading to an easier isolation of the products. Different HHP methodologies, such as constant high-pressure treatment and cycling (with different number of cycles, holding and decompression times) were applied and cycling was observed to be the most efficient activation for the majority of the reactions. Stability experiments were also conducted with one of the products and observed that although the solid-state storage does not alter the hydrazone, storing it in various solvents may significantly decrease the concentration of the active component which should be considered when performing the biochemical/biological assays. The process of developing of new drugs is greatly hampered by their inadequate physicochemical, pharmacokinetic, and intrinsic characteristics. In this regard, the selected chloro indolinone, (Z)-6-chloro-3-(2-chlorobenzylidene)indolin-2- one (C1), and nitro indolinone, (Z)-6-chloro-3-(2-nitrobenzylidene)indolin-2- one (C2), were subjected to SwissADME and density function theory (DFT) analysis. For compounds C1 and C2, the BOILED-Egg pharmacokinetic model predicted intestinal absorption, blood-brain barrier (BBB) penetration, and p-glycoprotein interaction. According to the physicochemical analysis, C1 has exceptional drug-like characteristics suitable for oral absorption. Despite only being substrates for some of the major CYP 450 isoforms, compounds C1 and C2 were anticipated to have strong plasma protein binding and efficient distribution and block these isoforms. The DFT study using the B3LYP/6-311G(d,p) approach with implicit water effects was performed to assess the structural features, electronic properties, and global reactivity parameters (GRP) of C1 and C2. The DFT results provided further support for other studies, implying that C2 is more water-soluble than C1 and that both compounds can form hydrogen bonds and (weak) dispersion interactions with other molecules, such as solvents and biomolecules. Furthermore, the GRP study suggested that C1 should be more stable and less reactive than C2. A concentration-dependent 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'- azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity was shown by both C1 and C2. Nutrition has crucial effects and a significant role in disease prevention. Recently, nutraceuticals have attracted much attention in scientific research due to their pleiotropic effects and relatively non-toxic behavior. Among the biological effects displayed by plants belonging to the Lamiaceae family, such as antibacterial, anticancer, anti-inflammatory, and anticholinesterase, sage is well known for its antioxidant properties and is a rich source of numerous compounds that are biologically active, amongst them polyphenols, with more than 160 types identified. In this review we summarized some of the significant studies published in the last decade reporting the most employed extraction methods and the different assays that are useful for establishing the antioxidant properties of some sage species. Even though the scientific literature contains plenty of data regarding the antioxidant properties of many sage species, further studies are needed in order to gain a deeper understanding of the mechanism of action and the compounds responsible for their antioxidant activity. Finally, it should be taken into account that the data on the antioxidant properties of sage extracts are often difficult to compare with each other, since a series of variables in the extraction procedures, the type of assay used, and standardization may affect the final result. The following article discusses the antioxidant properties of mandelic acid and its hydroxy and methoxy derivatives. The antioxidant capacity of these compounds is determined by DPPH, FRAP, CUPRAC and ABTS. The mechanisms underlying the antioxidant properties are described by BDE, IP, PDE, ETE and PA calculation method values and referenced to experimental data. Thermochemistry, HOMO/LUMO energies, dipole moments, charge distribution, IR, RAMAN, NMR frequencies, binding lengths and angles were calculated using the B3LYP method and the 6-311++G(d,p) basis set. The present research work depicts a cinch and economical synthesis of silver nanoparticles (AgNPs) phytofabricated utilizing leaf extract of Simarouba glauca and aerial extract of Celastrus paniculatus. Fourier transform infrared spectroscopy (FTIR) analysis displayed the synthesized AgNPs capped with bio-constituents existent in these medicinal plants marked by their functional groups. The morphological features of prepared NPs were characterized by employing X-ray Diffraction (XRD), Scanning Electron Microscope with Energy Dispersive spectra (SEM–EDX) and Transmission Electron Microscope (TEM) analysis illustrated their spherical shape and ~ 40 nm particle size. Green synthesized AgNPs were assessed for their antioxidant potential by free radical scavenging assays; and antibacterial efficacy. The phytofabricated AgNPs were found to induce cell growth arrest in MCF-7 and HT-29 cell lines with an IC50 value of 70.84 ± 0.67 and 158.24 ± 0.89 µg/mL for AgNPs prepared using S. glauca (Ag-SG) while 207.19 ± 0.64 and 221.22 ± 0.57 µg/mL for AgNPs synthesized using C. paniculatus (Ag-CP) in dose-proportional relation. The same samples could not effectively prohibit the growth of immortalized normal human breast epithelial cell lines (MCF-10A). The cytotoxicity induced by apoptosis was further confirmed by fluorescent images. The synthesized nanoparticles also demonstrated less hemolysis efficiency and are evidenced by SEM images. The analyses indicate the potent antioxidant and antiproliferative capacity of stable green synthesized AgNPs derived from the mentioned exorbitant curative herbs. [ABSTRACT FROM AUTHOR]

In this research, supercritical CO2 extraction is applied to extract bioactive compounds from Lamium album (white dead nettle, Lamiaceae). Extraction was performed at various temperatures (40, 50, and 60 °C) using methanol as co-solvent at a constant flow rate of CO2, methanol, and pressure. The collected extracts were characterized in terms of antioxidant capacity by using DPPH, ABTS and FRAP in vitro antioxidant activity assays, whereas the Folin–Ciocalteu procedure was employed to estimate the total phenols content (TPC). On the other hand, phenolic compounds in the extracts were quantitated by liquid chromatography coupled with a photodiode array detector (UPLC-PDA) and confirmed with a mass detector (TQD). The extracts have shown high TPC ranged between 234.17 to 650.17 mg GAE/g extract. DPPH scavenging of the extracts was estimated and obtained EC50 values ranged from 0.12 to 0.37 mg/mL of solution. The ABTS radical scavenging activity ranged from 43.20 to 44.53 µg TE/g. The FRAP value was found within the range of 19.48 to 44.74 µmol TE/g of extract. Differences between extraction conditions were observed. In this research, 50 °C/250 bar was efficient for the TPC, DPPH, ABTS, and FRAP assays; moreover, statistically, TPCs and FRAP assay showed significant differences between the conditions at α = 0.05. The identification of phenolic compounds in the obtained extract of Lamium album flowers, using UPLC/PDA, revealed that chrysin, pinostrobin, myricetin, and trans-3-hydroxycinnamic acid were the significant molecules present, which may be responsible for the high content of polyphenols and antioxidant activity. The results obtained indicated that SC-CO2 could be considered an alternative method for extracting bioactive compounds of Lamium album. High antioxidant activity and the presence of various bioactive compounds indicate the potential of this plant from the Lamiaceae family and the possibility of its application in various industries, including agriculture, food technology, or pharmacy. Interest in nanotechnology is increasing day by day. Silver and iron nanoparticles of the chloroform extract of Psephellus pyrrhoblepharus were prepared for this study by green synthesis method. The characterisation of the nanoparticles was carried out with some methods like the Fourier Transform Infrared Spectroscopy (FTIR), Transmission Electron Microscopy (TEM), Scanning Electron Microscopy (SEM), X-ray Diffraction Analysis (XRD), zeta potential and particle size. The antioxidant activities and total phenolic capacities of nanoparticles and three different extracts of P. pyrrhoblepharus (chloroform, methanol:water (1:1) and n-hexane) were compared. Silver nanoparticles were the most active ones by antioxidant activities and total phenolic capacity. Although the iron nanoparticles showed activity, they did not show a successful activity as silver nanoparticles. In this study, four new mixed-ligand saccharin complexes with 2-[[(2-Aminophenyl)imino]methyl]-6-methoxy phenol Schiff base and metal saccharinates (nickel, copper, zinc and cadmium) have been prepared and characterized with various methods including elemental analysis, FT-IR spectroscopy, UV-visible absorption spectroscopy, NMR spectroscopy, QTOF-LC/MS spectroscopy. All methods have confirmed the inclusion of the Schiff base obtained by condensation of o-phenylendiamine and o-vanillin into the metal saccharinates. Besides, DPPH scavenging activities and metal chelating abilities of all complexes have been determined and compared to well-known antioxidant standards (Butylated hydroxyanisole, Butylated hydroxytoluene and α-Tocopherol) along with starting metal saccharinates. According to the results, nickel complex has exhibited the best DPPH scavenging activity, while copper complex has the highest ferrous ion chelating ability. [ABSTRACT FROM AUTHOR]

Objective: To explore the new sources of novel bioactive compounds having pharmaceutical and agricultural interest and to search the endophytic actinobacteria from medicinal plants. Methods: NAF-1 an endophyte actinobacteria was isolated from leaves of medicinal plant Aloe vera collected in Marrakesh, Morocco using Bennett agar as selective medium. NAF-1 was tested for its antimicrobial activity against five pathogenic bacteria such as Staphylococcus aureus PIC 53156, Micrococcus luteus ATCC381, Bacillus subtilis ATCC 14579, Pseudomonas aeruginosa DSM 50090 and Escherichia coli ATCC 8739 and four human clinic fungi belonging to the Candida, Aspergillus and Microsporum genera. Several antioxidant activities were studied such as DPPH free radical scavenging, β -carotene and linoleic acid and reducing power assays. The total of phenol and flavonoid was also calculated. Using Artemia salina shrimp assay, the cytotoxicity of NAF-1 crude extract was determined. Results: The results revealed that the actinobacteria showed a high activity (≥20 mm) against only Gram positive bacteria but it had a moderate activity (between 13 and 15 mm) against Human clinic fungi. The isolate also exhibited a LD50 of 14.20 μg/mL in the cytotoxicity assay. The result showed that the crude extract presented an interesting free radical-scavenging activity with IC50 value of (5.58 ± 0.26) μg/mL and a high value of phenolic and flavonoid compounds with (15.41 ± 0.18) μg GAE/mg extract and (11.41± 0.06) μg QE/mg extract respectively. Moreover, the taxonomic position of our endophyte actinobacteria using the morphological and physiological criteria and using 16S rRNA gene sequence (polyphasic approach) showed that the NAF-1 isolate was similar to Streptomyces hydrogenans which was never described as an endophyte actinobacteria. Conclusions: This isolated strain appears promising resources of bioactive agents and can be exploited to produce therapeutic agents active against pathogenic disease. [ABSTRACT FROM AUTHOR]

Advancements in nanotechnology have led to the development of the nanomedicine, which involves nanodevices for diagnostic and therapeutic purposes. A key requirement for the successful use of the nanoparticles (NPs) in biomedical applications is their good dispensability, colloidal stability in biological media, internalization efficiency, and low toxicity. Therefore, toxicological profiling is necessary to understand the mechanism of NPs and microparticles (MPs). MgO NPs have attracted wide scientific interest due to ease of synthesis, chemical stability and unique properties. However, their toxic effects on humans should also be of concern with the increased applications of nano MgO. The present study was aimed to assess the toxicological potential of MgO NPs in comparison to their micron counterparts in female Wistar rats. Toxicity was evaluated using genotoxicity, histological, biochemical, antioxidant and biodistribution parameters post administration of MgO particles to rats through oral route. The results obtained from the investigation revealed that the acute exposure to the high doses of MgO NPs produced significant (p < 0.01) DNA damage and biochemical alterations. Antioxidant assays revealed prominent oxidative stress at the high dose level for both the particles. Toxicokinetic analysis showed significant levels of Mg accumulation in the liver and kidney tissues apart from urine and feces. Further, mechanistic investigational reports are warranted to document safe exposure levels and health implications post exposure to high levels of NPs. [ABSTRACT FROM AUTHOR]

The principal components of essential oils, obtained by steam-hydrodistillation from the fresh leaves of five species of the genus Lippia, namely Lippia gracilis AV, Lippia sidoides Mart, Lippia alba carvoneifera, Lippia alba citralifera and Lippia alba myrceneifera and ethanol extracts, were evaluated. The greater antioxidant capacity (IC50 = 980 µg/mL; p < 0.05), assessed by the HPLC-based hypoxanthine/xanthine oxidase assay, was determined in the essential oil obtained from Lippia alba carvoneifera, while the remainder were comparatively inactive (IC50 > 2000 µg/mL). Major compounds in the oils were, thymol (Lippia sidoides Mart, 13.1 g/k), carvone (Lippia alba carvoneifera, 9.6 g/kg), geranial (Lippia alba citraleifera, 4.61 g/kg; Lippia alba myrceneifera, 1.6 g/kg), neral (Lippia alba citraleifera, 3.4 g/kg; Lippia alba myrceneifera, 1.16 g/k), carvacrol (Lippia gracilis AV, 5.36 g/kg) and limonene (Lippia alba carvoneifera, 5.14 g/kg). Of these, carvone (IC50 = 330 µM) along with thymol (1.88 units), and carvacrol (1.74) units, were highly active in the hypoxanthine/xanthine oxidase, and ORAC assays respectively. The following compounds namely 1: calceolarioside E, 2: acteoside, 3: isoacteoside, 4: luteolin, 5: 5,7,3',4'-tetrahydroxy-3,6-dimethoxy flavone (spinacetin), 6: naringenin, 7: apigenin, 8: 6-methoxy apigenin (hispidulin), 9: 5,7,3'-trihydroxy-3,6,4'- trimethoxy flavone, 10: 5,7,4'-trihydroxy-3,6-dimethoxy flavone, 11: naringenin-4'-methyl ether, 12: 5,7- dihydroxy-3,6,4'-trimethoxy flavone (santin), 13: 5,7-dihydroxy-6,4'-dimethoxy-flavone (pectolinaringenin) and 14: 5-hydroxy-3,7,4'-trimethoxy flavone were identified in the ethanol extracts. Of the leaf ethanol extracts, strong antioxidant capacity was only evident in that of Lippia alba carvoneifera (IC50 = 1.23 mg/mL). The aim of this study was to model the extraction of phenolic antioxidants and identify the principal individual phenolic compounds of white tea by LC-DAD-MS/MS. A Box-Behnken design was applied to evaluate the effects of time, temperature and ethanol concentration in the extraction of phenolics and in vitro antioxidant activity. All mathematical models proposed by multiple regression analysis were significant (P < 0.001) and could explain up to more than 85% of the variance (R 2 adj > 0.80). A simultaneous optimization was performed using DPPH, ABTS, FRAP, (−)-epigallocatechin gallate, and (−)-epicatechin gallate to maximize the phenolic extraction and suggested optimum conditions of 10 min, 66 °C and 30% ethanol solution, with absolute error lower than 7%. The suggested optimum conditions were confirmed by external validation. The principal individual compounds identified by mass spectrometry were gallic acid, 5-galloylquinic acid, caffeine, theobromine, gallocatechin, epigallocatechin, epicatechin, epigallocatechin gallate, and epicatechin gallate. The nutritional and antioxidant activity of two species of leafy vegetable Basella were investigated. The moisture content of Basella alba and Basella rubra leaves was 928 and 930 g/kg, respectively. The dietary fibre content constitutes 32 g, 10 g and 22 g total, soluble and insoluble dietary fibre, respectively, in 100 g of dry B. alba leaves. The leaves of Basella spp. are very low in calories (95–110/100 g) and fats, but holds a good amount of vitamins, minerals, and antioxidants. The betalains content in B. rubra was highest in 20 weeks old leaves (80 mg) and vitamin C content of B. alba was more (74 mg) than B. rubra (67 mg) in 100 g fresh leaves. The EC 50 values were found high in BAW extract (3.4 mg/mL) for DPPH radical scavenging activity. BAW extract (1.04) at 50 mg/mL showed high ferric reducing antioxidant power and least in BAM extract. Various extracts from leaves with different constituents were shown dose dependent antioxidant and high scavenging abilities which play a key role in combating the reactive oxygen species. In the present work we synthesized a selected series of hydroxylated 3-phenylcoumarins 5-8, with the aim of evaluating in detail their antioxidant properties. From an in depth study of the antioxidant capacity data (ORAC-FL, ESR, CV and ROS inhibition) it was concluded that these derivatives are very good antioxidants, with very interesting profiles in all the performed assays. The study of the effect of the number and position of the hydroxyl groups on the antioxidant activity was the principal aim of this study. In particular, 7-hydroxy-3-(3'-hydroxy)phenylcoumarin (8) proved to be the most active and effective antioxidant of the selected series in four of the performed assays (ORAC-FL = 11.8, capacity of scavenging hydroxyl radicals = 54%, Trolox index = 2.33 and AI30 index = 0.18). However, the presence of two hydroxyl groups on this molecule did not increase greatly the activity profile. Theoretical evaluation of ADME properties of all the derivatives was also carried out. All the compounds can act as potential candidates for preventing or minimizing the free radical overproduction in oxidative-stress related diseases. Five antioxidant and two oxidative stress assays were applied to serum samples of 43 healthy males. The antioxidant tests showed different inter-assay correlations. A very good correlation of 0.807 was observed between the ferric reducing ability of plasma (FRAP) and total antioxidant status (TAS) assay and also a fair correlation of 0.501 between the biological antioxidant potential (BAP) and TAS assay. There was no statistically significant correlation between the BAP and FRAP assay. The anti-oxidant assays have a high correlation with uric acid, especially the TAS (0.922) and FRAP assay (0.869). The BAP assay has a much lower and no statistically significant correlation with uric acid (0.302), which makes BAP more suitable for the antioxidant status. The total thiol assay showed no statistically significant correlation with uric acid (0.114). The total thiol assay, which is based on a completely different principle, showed a good and statistically significant correlation with the BAP assay (0.510) and also to the TAS assay, but to a lower and not significant extent (0.279) and not with the FRAP assay (-0.008). The oxy-adsorbent test (OXY) assay has no correlation with any of the other assays tested. The oxidative stress assays, reactive oxygen metabolites (ROM) and total oxidant status (TOS), based on a different principle, do not show a statistically significant correlation with the serum samples in this study. Both assays showed a negative, but not significant, correlation with the antioxidant assays. In conclusion, the ROM, TOS, BAP and TTP assays are based on different principles and will have an additional value when a combination of these assays will be applied in large-scale population studies. Background: Sida cordata, a member of Family Malvaceae is used in folk medicine for various ailments including liver diseases. In this study we investigated, its flavonoid constituents, in vitro antioxidant potential against different free radicals and hepatoprotection against carbon tetrachloride (CCI4)-induced liver damage in rat. Methods: Dried powder of S. cordata whole plant was extracted with methanol and the resultant (SCME) obtained was fractionated with escalating polarity to obtain n-hexane fraction (SCHE), ethyl acetate fraction (SCEE), n-butanol fraction (SCBE) and the remaining soluble portion as aqueous fraction (SCAE). Diverse in vitro antioxidants assays such as DPPH, H2O2, •OH, ABTS, β-carotene bleaching assay, superoxide radical, lipid peroxidation, reducing power, and total antioxidant capacity were studied to assess scavenging potential of methanol extract and its derived fractions. On account of marked scavenging activity SCEE was selected to investigate the hepatoprotective potential against CCl4 induced toxicity in Sprague–Dawley male rats by assessing the level of serum markers (alkaline phosphatase, alanine transaminase, aspartate transaminase, lactate dehydrogenase, bilirubin, and γ- glutamyltransferase) and of liver antioxidant enzymes such as catalase (CAT), superoxide dismutase (SOD), peroxidase (POD), glutathione-S-transfers (GST), glutathione reductase (GSR), glutathione peroxidase (GSH-Px), and reduced glutathione (GSH) and lipid peroxidation (TBARS). Histology of the liver was performed to study alteration in histoarchitecture. Existence of active flavonoids was established by thin layer chromatographic studies. Results: Considerable amount of flavonoid and phenolic contents were recorded in the methanol extract and its derived fractions. Although the extract and all its derived fractions exhibited good antioxidant activities however, the most distinguished scavenging potential was observed for SCEE. Treatment of SCEE decreased the elevated level of serum marker enzymes induced with CCl4 administration whereas increased the activity of hepatic antioxidant enzymes (CAT, SOD, POD, GST, GSR and GSH-Px). Hepatic concentration of GSH was increased while lipid peroxidation was decreased with SCEE administration in CCl4 intoxicated rats. Presence of apigenin with some unknown compounds was observed in SCEE by using thin layer chromatography. Conclusions: These results revealed the presence of some bioactive compound in the ethyl acetate fraction, confirming the utility of S. cordata against liver diseases in folk medicine. Endogenous production and ultraviolet-generated free radicals in the skin can lead to photoaging and even skin cancer. Topical antioxidants have been found to provide benefits against ultraviolet damage and these ingredients have been incorporated into various cosmetic products and claimed to have substantial effects. Currently, there is a lack in a standardized rating system to measure the concentration and activity levels of antioxidants in these products. As a result, it is difficult for consumers and clinicians to evaluate and select commercial products based on readily accessible evidence. In this review, we will describe four assays which have been used to measure antioxidants in various products, and the strengths and weaknesses of each test will be detailed. Abstract: The antioxidant profiles of various espresso coffees were established using HPLC with UV-absorbance detection and two rapid, simultaneous, on-line chemical assays that enabled the relative reactivity of sample components to be screened. The assays were based on (i) the colour change associated with reduction of the 2,2′-diphenyl-1-picrylhydrazyl radical (DPPHi) the emission of light (chemiluminescence) upon reaction with acidic potassium permanganate. Results from the two approaches were similar and reflected the complex array of antioxidant species present in the samples. However, some differences in selectivity were observed. Chromatograms generated with the chemiluminescence assay contained more peaks, which was ascribed to the greater sensitivity of the reagent towards minor, readily oxidisable sample components. The three coffee samples produced closely related profiles, signifying their fundamentally similar chemical compositions and origin. Nevertheless, the overall intensity and complexity of the samples in both UV absorption and antioxidant assay chromatograms were aligned with the manufacturers description of flavour intensity and character. This study was planned to evaluate the antioxidant activity of ethyl acetate extract/fractions of Acacia nilotica Willd. Ex. Del extracted with different solvents of increasing and decreasing order of solvent polarity. The antioxidative activities, including the 1′-1′ diphenylpicryl-hydrazyl (DPPH) radical-scavenging effects, hydroxyl radical scavenging potential, chelating ability, reducing power and lipid peroxidation inhibition in rat tissue homogenate were studied in vitro. It was found that the antioxidative effect provided by extract/fractions was strongly concentration dependent and increased on fractionating the extract into water and ethyl acetate fractions. In general, the antioxidative activity increased with increasing extract/fractions concentration to a certain extent, and then leveled off with further increase in antioxidant activity. From a comparison of the antioxidant potential and IC50 values for different antioxidative reactions, it seemed that extract/fractions were more effective in scavenging DPPH and hydroxyl radicals than reducing, chelating heavy metals and lipid peroxidation inhibitory potential. The best health and nutrition results can be achieved not only from the consumption of fruits and vegetables with high antioxidant capacities, but also from medicinal plants and herbs. Therefore, in the present investigation, the bioactive compounds (polyphenols and flavonoids) and the radical scavenging capacities of Prolipid, a mixture of herbs, were studied. Water extracts showed relatively high capacity of about 61.5% inhibition with the β-carotene-linoleic acid assay. In order to support the data obtained with β-carotene-linoleic acid assay, three different antioxidant assays were used: ferric-reducing/antioxidant power, trolox equivalent antioxidant capacity, and 1,1-diphenyl-2-picrylhydrazyl radical with prolonged time of their reactions. It was found that the amounts of polyphenols in water and methanol extracts were 22.849±2.267 and 3.241±0.325 mg gallic acid equivalent/g dry weight, and the antioxidant capacities in same extracts as determined by the 2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) test after 120min were 275.81±27.58 and 62.25±6.11 micromole Trolox equivalents (µMTE) dry weight, respectively. The correlation coefficients between polyphenols and antioxidant capacities of Prolipid with 1,1-diphenyl-2-picrylhydrazyl radical and β-carotene-linoleic acid assay were about 0.97 and 0.98. In conclusion, the bioactivity of Prolipid was high and the total polyphenols were the main contributors to the overall antioxidant capacity. The results of our investigation in vitro are comparable with other medicinal plants and fruits widely used in the treatment of humans without known side effects. In the present work the antioxidant activity of different solvent (100% methanol, 80% methanol, 100% acetone, 80% acetone) extracts of rice bran was evaluated following different antioxidant assays and using sunflower oil as oxidation substrate. The rice bran extracts were evaluated from the estimate of % inhibition of peroxidation in linoleic acid system, total phenolics content (TPC) and loss of β-carotene in a linoleic acid system. Additionally, crude concentrated rice bran extracts were added into the sunflower oil samples and stored under ambient conditions. The extent of oxidative deterioration was followed by the measurement of peroxide-, p-anisidine-, conjugated diene-, and triene- values. The general order of antioxidant efficacy of rice bran extracts as determined by various antioxidant assays was 80% methanolic extract > 100% methanolic extract > 80% acetone extract > 100% acetone extract. The results of the present comprehensive analysis demonstrate that rice bran extracts of the Super Kernel variety indigenous to Pakistan are a viable source of natural antioxidants and might be exploited for functional foods and nutraceutical applications. European beech (Fagus sylvatica L.) is a deciduous tree, widely distributed in Europe and largely appreciated for its wood and nutritive nuts. Beech leaf also enjoys food use as salad, but an understanding of its nutraceutical value is still far from being achieved. Indeed, and also taking into account beech leaf as a consistent biomass residue available beechwood production and use, it needs to be explored as a valuable renewable specialized source of bioactive molecules. In this context, an untargeted ultra-high-performance liquid chromatography hyphenated with high resolution mass spectrometry (UHPLC-HRMS) approach was favorably applied to a beech leaf alcoholic extract, which also was evaluated for its antiradical capability (by means of assays based on 2,2-diphenyl-1-picrylhydrazyl (DPPH) and [2,2'-azinobis-(3-ethylbenzothiazolin-6-sulfonic acid)] (ABTS) radical cation) and its ferric ion reducing power. Redox mitochondrial activity towards Caco-2 cells paved the way to explore the extract's capability to inhibit intracellular Reactive Oxygen Species (ROS) using 2',7'dichlorofluorescin diacetate (DCFH-DA) assay. Hydroxycinnamoyl derivatives, mainly belonging to the chlorogenic acid class, and flavonoids were the main constituents. Uncommon flavanone C-glycosides were also found, together with a plentiful flavonol diversity. Cell-free and cell-based assays highlight its dose-dependent antioxidant efficacy, providing a foundation for further investigation of beech leaf constituents and its valorization and use as a reservoir of bioactive natural products with potential nutraceutical applications. The plant kingdom is a rich source of health-promoting compounds and has always played a fundamental role in the isolation, identification, and modification of compounds able to perform several properties on live organisms. Among them, the so-called "antioxidants" have a major potentiality to increase human wellness. Antioxidants are important components in the signaling and defense mechanisms in some plants, where they are precursors of compounds of greater complexity, the modulator of plant growth, and the defensive system against pathogenic organisms and predators. The extraordinary variety of chemical structure and substitution present in the different plant antioxidants make them an inestimable source of interesting compounds, with the ability to counter reactive oxygen/nitrogen species (ROS/RNS) and to stimulate the activation of signal cascade inside the cells. The mechanisms by which antioxidants detoxify these dangerous compounds are complex and involve either direct or indirect interaction with radicals. Antioxidants inhibit or quench free radical reactions mainly based on their reducing capacity or hydrogen atom-donating capacity, their solubility, and chelating properties.
