The excessive accumulation of reactive oxygen species (ROS)/free radicals can lead to abnormal oxidation of biomolecules such as proteins, lipids, fats, carbohydrates and nucleic acids in human organisms. Accordingly, endogenous oxidative stress induces the progressive development of various chronic diseases like rheumatoid arthritis, cancers, cardiovascular risks, diabetes, digestive ulcers, hypertension, obesity, neurological disorders, and age-related complications. Therefore, anti-oxidant defense mechanisms are needed to control/prevent the unbalanced molecular oxidative damage. Indeed, the oxidative stress arises from both endogenous and exogenous factors such as smoking, alcohol, medications, air pollution, sunlight, lifestyle disorders, and metabolic processes. Therefore, consumption of fruits, vegetables, grains, beverages, and leafy vegetables rich in antioxidants may inhibit or treat oxidative damage accompanying diseases. From this aspect, dietary foods are rich in various antioxidant metabolites such as flavonoids, vitamin A, C, E, phenolic acids, curcumin, stilbenes, anthocyanins, etc., which promote healthy life and nutritional benefits. Additionally, various studies have also proven that foods rich in antioxidants interact with reactive species to prevent cell damage(s) or therapeutic pathways for diseases. A series of novel α-methyl-l-DOPA urea derivatives viz., 3-(3,4-dihydroxyphenyl)-2-methyl-2-(3-halo/trifluoromethyl substituted phenyl ureido)propanoic acids (6a-e) have been synthesized from the reaction of α-methyl-l-DOPA (3) with various aryl isocyanates (4a-e) by using triethylamine (5, TEA) as a base catalyst in THF at reflux conditions. The synthesized compounds are structurally characterized by spectral (IR, 1 H & 13 C NMR and MASS) and elemental analysis studies and screened for their in-vitro antioxidant activity against DPPH, NO and H 2 O 2 free radical scavenging assays and identified compounds 6c &6d as potential antioxidants. The acquired in vitro results were correlated with the results of molecular docking, ADMET, QSAR and bioactivity studies performed for them and predicted that the recorded in silico binding affinities are in good correlation with the in vitro antioxidant activity results. The molecular docking analysis has comprehended the strong hydrogen bonding interactions of 6a-e with 1CB4, 1N8Q, 3MNG, 1OG5, 1DNU, 3NRZ, 2CDU, 1HD2 and 2HCK proteins of their respective SOD, LO, PRXS5, CP450, MP, XO, NO, PRY5 and HCK enzymes. This has sustained the effective binding of 6a-e and resulted in functional inhibition of selective aminoacid residues to be pronounced as multiple molecular targets mediated antioxidant potent compounds. In addition, the evaluated toxicology risks of 6a-e are identified with in the potential limits of drug candidates. An efficient and convenient Meglumine catalyzed procedure for the synthesis of bis(indolyl) methanes at ambient temperature under aqueous conditions in high yields. The catalytic reaction proceeds very smoothly. Clean reaction, ease of product isolation/purification, easily available reactants, metal free and environmentally friendly reaction conditions are the notable advantages of the present methodology. All the entitled compounds were characterized by IR, 1 H, 13 C NMR, mass spectra and evaluated for their antioxidant (DPPH, H 2 O 2 and NO scavenging methods). They exhibited potent in vitro antioxidant activity dose-dependently. The review discusses recent achievements in the development of more ecofriendly and economically viable processes for the synthesis of biologically potent α-aminophosphonates via Kabachnik-Fields reaction by three-component coupling of carbonyl, amine and hydrophosphoryl compounds. These α-aminophosphonates exhibited promising antioxidant, antimicrobial and anticancer activity. Some recent developments on the synthesis of biologically active α-aminophosphonates in the presence of various catalysts, in catalytic solvent medium, in catalyst-free solvent medium, under solvent-free conditions, and reactions in solution are discussed. In the present study, different extracts of stem bark of Shorea tumbuggaia and roots of Syzygium alternifolium were screened for total phenolic content (TPC), total flavonoid content (TFC) and in vitro DPPH radical scavenging activity to isolate the natural antioxidants. The bioassay-guided fraction of stem bark of Shorea tumbuggaia yielded three compounds, piceatannol (ST-1), resveratrol-12- C - β - d -glucopyranoside (ST-2) and hopeaphenol (ST-3). Similarly, a systematic phytochemical examination of extracts of Syzygium alternifolium roots had resulted five compounds, 5,7,8,5′-tetramethoxy-3′,4′-methylenedioxyflavone (SA-1), 5-hydroxy-4′,7-dimethoxy-6,8-di- C -methylflavone (SA-2), quercetin 7-methylether (SA-3), kaempferol 7,4′-dimethylether 3- O - β - d -glucopyranoside (SA-4) and taxifolin 3-O- α - l -rhamnopyranoside (SA-5). Structures of the isolates were established using UV, IR, Mass, 1H and 13C NMR spectral studies. Density Functional Theory (DFT) calculations, Molecular docking and Lipinski rule of five were conducted to explored the antioxidant activity of isolated compounds. SA-3 (37.52 µg/mL) and ST-1 (42.43 µg/mL) showed highest IC 50 values compared to the Ascorbic acid (45.97 µg/mL) and have highest electron affinities (EA eV) along with smallest HOMO-LUMO energy gap. Molecular docking and binding affinity studies with NADPH and SPSB2 proteins, revealed the prominent antioxidant activity of SA-3 and ST-1 with molecular interaction besides promising solvation energies. A simple method was employed for the synthesis of dihydropyrano[3, 2‐b]chromenedione derivatives (4a‐o) in high yields by condensation of 5, 5‐dimethylcyclohexane‐1, 3‐dione(1), different aromatic aldehydes (2a‐o), and 5‐hydroxy‐2‐(hydroxymethyl)‐4H‐pyran‐4‐one(3), using meglumine as a stable and reusable catalyst. Meglumine, an amino sugar, was employed as an environmentally benign catalyst, due to its splendid properties such as being inexpensive, recyclable, and biodegradable. The accomplished protocol employs low catalyst loading and easy work‐up for the synthesis of 5‐hydroxy‐2‐(hydroxymethyl)‐4H‐pyran‐4‐one derivatives. A great asset is that without any significant loss, the catalyst could be recovered and reused for extended synthetic steps. This offer huge advantage to overcome recyclability issues. Our synthesized compounds were analyzed by IR, 1H, 13C NMR, mass spectra and evaluated for their antioxidant properties by 1, 1‐diphenyl‐2‐picryl hydrazyl radical (DPPH), hydrogen peroxide(H2O2), and nitric oxide (NO) scavenging methods. The correlation in exhibition of antioxidant activity was effective at all doses. The binding interactions and molecular docking studies for entitled compounds were studied against 3MNG protein; 4k exhibited marked binding affinity with excellent docking score of −7.6 Kcal/mol and emerged as a lead compound. A series of N‐substituted maleimide derivatives have been developed via acetic acid‐mediated microwave reaction pathway, which was identified as the incomparable method for this maleimide compounds. All the synthesized compounds were tested for antioxidant activity by DPPH and H2O2 methods. Compounds 5h and 5m were displayed with higher antioxidant activity in two methods. The structure–activity relationship demonstrated that the compounds having electron releasing substitutions 5h and 5m generally show beneficial activity than electron capture substitution cores.
