Background: As a result of the world population and climate change impact increases (especially in arid environments), there is a critical need for high-yield, drought-tolerant wheat. Synthetic hexaploid wheat derived lines (SHW-DL), were created artificially by crossing different durum wheat cultivars (AABB) with accessions of Aegilops tauschii (DD), a beneficial source of new genes for common bread wheat (Triticum aestivum L). Here, we studied the response of a panel of 91 SHW-DL for drought tolerance based on physiological, antioxidant enzyme activities, and drought tolerance indices., Results: A wide range of variation and high values of heritability observed for grain yield, physiological and antioxidant traits indicating that the SHW-DL panel constitutes a valuable gene source for drought tolerance improvement of wheat. Despite decreases in grain yield (YLD), leaf area index (LAI), and relative water content (RWC) an increase in the content of malondialdehyde (MDA) was observed. Moreover, drought streass increased the antioxidant enzyme activities of ascorbate peroxidase (APX), catalase (CAT) and peroxidase (POD), and also photosynthetic pigments, proline (Pro), and MDA content. With higher values of grain yield, physiological and biochemical traits such as photosynthetic pigments, and RWC, and lower content of MDA, and peroxidase (H 2 O 2 ) activity, SHW-DL performed better compared to common wheat lines under water stress conditions., Conclusions: Different responses to water stress within the germplasm and between synthetic and common wheat suggest that selection for adaptive and suitable genotypes is possible for drought tolerance in synthetic wheat germplasm. In the aquaculture industry, silica nanoparticles (SiNPs) have great significance, mainly for confronting diseases. Therefore, the present study aims to assess the antibacterial efficiency of SiNPs as a versatile trial against Aeromonas veronii infection in African catfish (Clarias gariepinus). Further, we investigated the influence of SiNPs in palliating the immune-antioxidant stress biochemical, ethological, and histopathological alterations induced by A. veronii. The experiment was conducted for 10 days, and about 120 fish were distributed into four groups at random, with 30 fish each. The first group is a control that was neither exposed to infection nor SiNPs. The second group (SiNPs) was vulnerable to SiNPs at a concentration of 20 mg/L in water. The third group was experimentally infected with A. veronii at a concentration of 1.5 × 10 7 CFU/mL. The fourth group (A. veronii + SiNPs) was exposed to SiNPs and infected with A. veronii. Results outlined that A. veronii infection induced behavioral alterations and suppression of immune-antioxidant responses that appeared as a clear decline in protein profile indices, complement 3, lysozyme activity, glutathione peroxidase, and total antioxidant capacity. The kidney and liver function biomarkers (creatinine, urea, alkaline phosphatase, and alanine aminotransferase) and lipid peroxide (malondialdehyde) were substantially increased in the A. veronii group, with marked histopathological changes and immunohistochemical alterations in these tissues. Interestingly, the exposure to SiNPs resulted in a clear improvement in all measured biomarkers and a noticeable regeneration of the histopathological changes. Amid climate change and shifts in precipitation patterns, drought conditions are expanding worldwide. Drought stress severely threatens plant growth in arid and semi-arid regions, wherein shrubs play a crucial role in maintaining ecological stability. Despite its ecological significance, studies are lacking on how Nitraria sibirica adapts to long-term drought stress. Therefore, in this study, to elucidate the mechanism of drought stress adaptation in N. sibirica, we analysed morphological, physiological, and transcriptional characteristics of plants in two soil habitats: riparian (moist) and desert (arid). The results showed that in desert soils, as soil water content decreased, leaf thickness increased, while plant height and leaf area decreased. Physiologically, photosynthesis decreased; soluble sugar, starch, proline, and hydrogen peroxide content increased significantly; while soluble proteins decreased significantly. Additionally, membrane lipid peroxidation products and antioxidant enzyme activities significantly increased under drought stress. Then, Kyoto Encyclopaedia of Genes and Genomes (KEGG) enrichment analysis identified 313 key genes, which were considered the most significantly enriched in the photosynthesis and photosynthetic antenna protein pathways. Further, we found that the proteins encoding photosystem II (PsbP, PsbQ, PsbR, PsbY, and Psb27), photosystem I (PsaD, PsaF, PsaG, PsaH, PsaK, and PsaO), photosynthetic electron transport (PetF), and light-trapping antenna proteins were significantly downregulated under drought stress. Taken together, these results suggest that N. sibirica adapts to long-term drought conditions by suppressing photosynthesis, activating antioxidant systems, and recruiting osmoregulators. Background: The liver was identified as a primary target organ for the chemo-radiological effects of uranyl acetate (UA). Although the anti-oxidant and anti-apoptotic properties of gallic acid (GA) make it a promising phytochemical to resist its hazards, there is no available data in this area of research., Methods: To address this issue, eighteen rats were randomly and equally divided into three groups. One group was received carboxymethyl cellulose (vehicle of GA) and kept as a control. The UA group was injected intraperitoneally with UA at a single dose of 5 mg/kg body weight. The third group (GA + UA group) was treated with GA orally at a dose of 100 mg/kg body weight for 14 days before UA exposure. UA was injected on the 15th day of the experiment in either the UA group or the GA + UA group. The biochemical, histological, and immunohistochemical findings in the GA + UA group were compared to both control and UA groups., Results: The results showed that UA exposure led to a range of adverse effects. These included elevated plasma levels of aspartate aminotransferase, lactate dehydrogenase, total protein, globulin, glucose, total cholesterol, triglycerides, low-density lipoprotein cholesterol, and very-low-density lipoprotein and decreased plasma levels of high-density lipoprotein cholesterol. The exposure also disrupted the redox balance, evident through decreased plasma total antioxidant capacity and hepatic nitric oxide, superoxide dismutase, reduced glutathione, glutathione-S-transferase, glutathione reductase, and glutathione peroxidase and increased hepatic oxidized glutathione and malondialdehyde. Plasma levels of albumin and alanine aminotransferase did not significantly change in all groups. Histopathological analysis revealed damage to liver tissue, characterized by deteriorations in tissue structure, excessive collagen accumulation, and depletion of glycogen. Furthermore, UA exposure up-regulated the immuno-expression of cleaved caspase-3 and down-regulated the immuno-expression of nuclear factor-erythroid-2-related factor 2 in hepatic tissues, indicating an induction of apoptosis and oxidative stress response. However, the pre-treatment with GA proved to be effective in mitigating these negative effects induced by UA exposure, except for the disturbances in the lipid profile., Conclusions: The study suggests that GA has the potential to act as a protective agent against the adverse effects of UA exposure on the liver. Drought stress memory occurring in some plants plays a crucial role in their adaptation to unfavorable conditions. However, in open-pollinated plants, this phenomenon is assumed to be affected by population plasticity resulting from kind and level of diversity and inbreeding depression. Physiological perspectives of drought stress memory in four synthetic poly-crossed populations (groups) of orchard grass (Dactylis glomerata) constructed from parental genotypes with contrasting levels (narrow and wide) of molecular and morphological genetic variation were assessed. Populations of two generations (Syn1 and Syn2) were developed and were subjected to three moisture treatments, including normal irrigation (C), primary mild stress-secondary intense stress (D1D2), and secondary intense stress (D2). Pre-exposure to drought significantly improved the mean values of leaf water, chlorophyll, proline, and ascorbate peroxidase compared to intense stress, leading to more effective memory responses. Superiority of groups with high levels of molecular diversity for most traits, suggesting that the molecular genetic distance among parents is an effective predictor of progeny performance. The results indicated that the fitness of progenies of the four polycross groups declines significantly from Syn1 to Syn2, however the magnitude of observed inbreeding depends on the level of diversity and moisture conditions. We propose a hypothesis that underscores the interplay between genetic diversity among parents and drought stress memory providing valuable insights for developing new synthetic varieties in open-pollinated grasses. Despite the potential for temporally dependent relationships between trait values and fitness (e.g. as juveniles approach life-stage transitions such as fledging), how developmental stage affects canalization (a measure of robustness to environmental variation) of morphological and physiological traits is rarely considered. To test the sensitivity of morphological and physiological traits to environmental variation in two developmental stages, we manipulated brood size at hatch in European starlings (Sturnus vulgaris) and cross-fostered chicks between enlarged and reduced broods approaching fledging. We measured body size (mass, tarsus, wing length) and physiological state (aerobic capacity, oxidative status) at asymptotic mass on day 15, then cross-fostered chicks between 'high' and 'low' quality environments and assessed the same traits again on day 20, after 5 days of pre-fledging mass recession. Chicks in reduced broods were heavier at asymptotic mass and had lower reactive oxygen metabolites than enlarged broods, whereas structural size, aerobic capacity and antioxidant capacity were unaffected by experimental brood size. The observed canalization of structural and physiological traits during early development was maintained after cross-fostering, during late development. However, in contrast to early development, antioxidant capacity approaching fledging appeared sensitive to environmental conditions, as trajectories varied by cross-fostering treatment. Elevated reactive oxygen metabolites observed after early development in enlarged brood chicks were maintained after cross-fostering, suggesting that canalized development in low-quality environments could produce oxidative costs that carry over between life stages, even when conditions improve. Background: The land snail, Theba pisana, is one of the most important threats facing agriculture around the globe. Boric acid (BOA) is currently used as a safe alternative molluscicide to control land snails in sustainable agriculture, but the mechanisms of toxicity have not yet been investigated. The present study characterizes the lethal and sub-lethal (0.5 and 1 mg g -1 ) toxic effects of BOA-contaminated food for 14 days by examining physiological, biochemical and histopathological indicators in T. pisana to understand the mechanisms underlying its toxic action., Results: BOA was found to be lethal against T. pisana with LC 50 values of 24.7 and 8.05 mg g -1 after 3 and 7 days of exposure, respectively. BOA sublethal concentrations led to a significant reduction in food consumption and growth of snails after 14 days of exposure. BOA also caused a significant increase in testosterone levels, whereas an opposite effect was observed in estradiol levels. An increase in progesterone levels in snails in the 0.5 mg g -1 BOA group and a decrease in the 1 mg g -1 BOA group were observed after all exposure times. Moreover, the lipid peroxidation level and catalase activity were elevated, whereas acetylcholinesterase activity was inhibited in the treated snails. Alteration in glutathione-S-transferase activity was noticed after exposure to both sublethal concentrations. High ambient temperature has emerged as a major constraint for the future development of the poultry industry, especially in the tropics and subtropics. The scarcity of resources coupled with harsh environmental conditions is the most crucial predicaments in the way to rationalize optimum production of broiler. Heat stress disturbs the physiological biochemistry of the broiler which ultimately reduces feed intake and feed efficiency which ultimately results in reduced performance and productivity. Under hot environmental conditions, feed utilization is disturbed by the deposition of fat and oxidative stress. In addition, changes in blood cells, acid-base balance, immune response, liver health, and antioxidant status are some of the major dynamics altered by heat stress. The broilers have a narrow range of temperatures to withstand heat stress. In this review, we have discussed the various physicochemical changes during heat stress, their possible mechanisms, and mitigation strategies to reduce heat stress.

Tetraploids are usually more tolerant to environmental stresses than diploids. Citrus plants face numerous abiotic stresses, including salinity, which negatively affect growth and yield. Double diploid citrus rootstocks have been shown to be more tolerant to abiotic stresses than their diploid relatives. In this study, we evaluated the antioxidative and osmotic adjustment mechanisms of diploid (2x) and double diploid (4x) volkamer lemon (Citrus volkameriana Tan. and Pasq.) rootstocks, which act against salt stress (75 and 150 mM). Results indicated that, under salt stress, all physiological variables (photosynthesis, stomatal conductance, transpiration rate, and leaf greenness) decreased, and these decreases were more noticeable in 2x plants than in 4x plants. On the other hand, accumulation of oxidative markers (malondialdehyde and hydrogen peroxide) was greater in the leaves and roots of 2x seedlings than in 4x seedlings. Similarly, the activities of antioxidative enzymes (peroxidase, ascorbate peroxidase, glutathione reductase, and catalase) were higher in the leaves and roots of 4x plants than in 2x plants. However, superoxide dismutase activity was higher in the roots of 2x seedlings than 4x seedlings. Double diploid plants affected by salt stress accumulated more osmolytes (i.e. proline and glycine betaine) in their leaves and roots than that by 2x plants. Total protein content, antioxidant capacity, and total phenolic content were also higher in 4x plants than 2x plants under salinity. At 150 mM, both 2x and 4x plants showed more symptoms of stress than those at 75 mM. Sodium content was the highest in the roots of 2x plants and in the leaves of 4x plants, while chloride content peaked in the leaves of 2x plants and in the roots of 4x plants. Overall, our results demonstrate that the active antioxidative defence mechanisms of 4x plants increase their tolerance to salinity compared to their corresponding 2x relatives. L-Glutamate (Glu) has traditionally not been considered as a nutrient needed in diets for humans and other animals (including swine) due to the unsubstantiated assumption that animals can synthesize sufficient amounts of Glu to meet their needs. The lack of knowledge about Glu nutrition has contributed to suboptimal efficiency of global livestock production. Over the past 25 years, there has been growing interest in Glu metabolism in the pig, which is an agriculturally important species and also a useful model for studying human biology. Because of analytical advances in its analysis, Glu is now known to be a highly abundant free amino acid in milk and intracellular fluid, a major constituent of food and tissue proteins, and a key regulator of gene expression, cell signaling, and anti-oxidative reactions. Emerging evidence shows that dietary supplementation with 2% Glu maintains gut health and prevents intestinal dysfunction in weanling piglets, while enhancing their growth performance and survival. In addition, the inclusion of 2% Glu is required for dietary arginine to maximize the growth performance and feed efficiency in growing pigs, whereas dietary supplementation with 2% Glu reduces the loss of skeletal muscle mass in endotoxin-challenged pigs. Furthermore, supplementing 2% Glu to a corn- and soybean-meal-based diet promotes milk production by lactating sows. Thus, an adequate amount of dietary Glu as a quantitatively major nutrient is necessary to support maximum growth, development, and production performance of swine. These results also have important implications for improving the nutrition and health of humans and other animals.

The effects of shearing on behavioral patterns, antioxidants, and inflammatory and stress biomarkers was investigated in Ossimi sheep. Clinical parameters and behavioral patterns were recorded, and serum samples were collected pre-shearing and 1, 5, 10 and 15 days post-shearing from 60 Ossimi sheep. The results revealed that grooming and standing idle frequencies were significantly (P<0.01) increased post-shearing. There were significant (P<0.01) increases in IL-6, cortisol, and MDA detected from the 1st to the 10th day post-shearing compared to pre-shearing values. IL-2 and TNF-α significantly (P<0.01) increased from the 1st until the 5th day post-shearing compared to pre-shearing values, while significant (P<0.01) decreases in the values of catalase from the 1st until the 5th day post-shearing compared to pre-shearing values were recorded. Regarding to glutathione reductase, there was a significant (P<0.01) decrease from the 1st until the 10th day post-shearing compared to pre-shearing values. Shearing leads to significant changes in antioxidants, inflammatory, stress biomarkers, and some behaviors in sheep.

The equivocal role of reactive species and redox signaling in exercise responses and adaptations is an example clearly showing the inadequacy of current redox biology research to shed light on fundamental biological processes in vivo. Part of the answer probably relies on the extreme complexity of the in vivo redox biology and the limitations of the currently applied methodological and experimental tools. We propose six fundamental principles that should be considered in future studies to mechanistically link reactive species production to exercise responses or adaptations: 1) identify and quantify the reactive species, 2) determine the potential signaling properties of the reactive species, 3) detect the sources of reactive species, 4) locate the domain modified and verify the (ir)reversibility of post-translational modifications, 5) establish causality between redox and physiological measurements, 6) use selective and targeted antioxidants. Fulfilling these principles requires an idealized human experimental setting, which is certainly a utopia. A hydroponic experiment was conducted to investigate perchlorate (ClO4 (-) ) phytotoxicity in different rice varieties. Considerable variations were observed when 24 rice varieties were treated with ClO4 (-) . The shoot height, root length, and biomass of most varieties were significantly reduced by ClO4 (-) . The roots were more sensitive than the shoots. Hierarchical clustering analysis demonstrated primarily 4 groups: ClO4 (-) -sensitive, medium ClO4 (-) -sensitive, medium ClO4 (-) tolerant, and ClO4 (-) -tolerant. Gannuoxiang (a ClO4 (-) -tolerant variety) and IR65598-112-2 (a ClO4 (-) -sensitive variety) were chosen to explore their antioxidant response when exposed to 0.2 mmol/L, 2.0 mmol/L, and 4.0 mmol/L ClO4 (-) . The results showed that the activities of superoxide dismutase and catalase increased in the shoots and roots of gannuoxiang with increasing doses of ClO4 (-) , but both of them decreased at higher concentrations of ClO4 (-) in IR65598-112-2. The addition of ClO4 (-) led to a significant increase in peroxidase activities for both of the varieties, whereas the increase was more pronounced in gannuoxiang than in IR65598-112-2. No significant difference was found in malondialdehyde (MDA) contents in gannuoxiang, whereas the addition of ClO4 (-) increased the MDA level significantly in IR65598-112-2. The results indicated that gannuoxiang has higher activities of antioxidant enzymes than IR65598-112-2 to cope with oxidative damage caused by ClO4 (-) stress, which may be the main cause of its high tolerance., (© 2015 SETAC.)

The aim of this study was to investigate whether different phases of training affect oxidative stress and antioxidant defences in professional cyclists. Ten professional cyclists, aged 21.8 ± 2.5 years, were enrolled in the study. They were classified into two groups of five athletes each one with similar nutritional intake excepting for the overload of vitamin C (1000 mg day(-1)) and E (400 mg day(-1)) supplementation in one of them. The cyclists of both groups performed the same exercise design, consisting of hard, tapering and recovery training periods. Total antioxidant capacity (TAC) of the diet, plasma oxygen radical absorption capacity (ORAC), lipid peroxidation (LPO), DNA damage (8-OHdG) and erythrocyte glutathione disulfide/glutathione ratio (GSSG:GSH(-1)) were measured. During the intense exercise trainings, the cyclists without vitamin supplements had the TAC of diet significantly lower than the supplemented group. Plasma ORAC, LPO and 8-OHdG were similar in both groups of athletes. Athletes with supplements had a basal LPO:ORAC(-1) ratio lower than that without supplements, but this ratio converged to the same level at the end of the training in both groups of cyclists. Both groups of cyclists showed similar changes in GSSG:GSH(-1) ratio and in GSSG and GSH levels along the study. The data suggest that well-trained athletes with suitable ultra-endurance training volume and intensity do not require antioxidant vitamin supplements to adapt their endogenous antioxidant defenses to exercise-induced ROS.

Burn injury causes a major systemic catabolic response that is associated with mitochondrial dysfunction in skeletal muscle. We investigated the effects of the mitochondria-targeted peptide antioxidant Szeto-Schiller 31 (SS-31) on skeletal muscle in a mouse burn model using in vivo phosphorus-31 nuclear magnetic resonance ((31)P NMR) spectroscopy to noninvasively measure high-energy phosphate levels; mitochondrial aconitase activity measurements that directly correlate with TCA cycle flux, as measured by gas chromatography mass spectrometry (GC-MS); and electron paramagnetic resonance (EPR) to assess oxidative stress. At 6 h postburn, the oxidative ATP synthesis rate was increased 5-fold in burned mice given a single dose of SS-31 relative to untreated burned mice (P=0.002). Furthermore, SS-31 administration in burned animals decreased mitochondrial aconitase activity back to control levels. EPR revealed a recovery in redox status of the SS-31-treated burn group compared to the untreated burn group (P<0.05). Our multidisciplinary convergent results suggest that SS-31 promotes recovery of mitochondrial function after burn injury by increasing ATP synthesis rate, improving mitochondrial redox status, and restoring mitochondrial coupling. These findings suggest use of noninvasive in vivo NMR and complementary EPR offers an approach to monitor the effectiveness of mitochondrial protective agents in alleviating burn injury symptoms.

Literature on serum copper (Cu) and its antioxidant protein (ceruloplasmin) in the African newborn is infrequent, and more reports are evident from developed or affluent societies. We, therefore, studied longitudinally our newborns to delineate their Cu and ceruloplasmin (CLP) status. All infants were born between July 1st, 1991 and June 30th, 1992 at the University of Maiduguri Teaching Hospital, Maiduguri, Nigeria. The preterm infants (PI) (gestational age < or = 36 weeks) were divided into 2 cohorts; A, sick and B, stable; commencing with n = 30 in each group. The groups were matched in respect to gender, gestational age, birthweight, Apgar scores and socioeconomic class. Cu levels were contemporaneously also determined in 30 (M:F, 17:13) stable full-term infants (appropriate-for-dates). Cu determination was made with atomic absorption spectrophotometry and CLP according to colorimetric P-phenylenediamine assay. Sick PI (cohort A) had significantly lower mean (SD) Cu and CLP levels at birth; 0.1 (0.2) micromol/ml and 0.5 (0.8) micromol/dl vs 0.6 (0.2) micromol/ml and 12.5 (1.2) micromol/dl in stable PI (cohort B), respectively; P < 0.05. Cu level was significantly increased by 4 weeks in cohort A; P < 0.001 (n = 25) and approached levels of the stable PI (0.7 (0.3) vs 0.8 (0.2) micromol/ml). Concerning CLP, however, catch-up of levels was delayed till 8 weeks, and a triphasic pattern of linear rise in Cu (both cohorts, but more prominent in A) and CLP (cohort A) was discernible by 24 weeks. The sick PI had mean (SD) serum CLP levels of 0.5 (0.8) micromol/dl, 5.9 (1.4), 15.2 (2.6), 17.3 (2.9), 21.2 (3.8), 25.1 (4.7) and 23.7 (3.8) micromol/dl at birth, 4, 8, 12, 20, and 24 weeks, respectively and were similar from 8 weeks in cohort B. Generally, CLP paralleled serum Cu levels. Cu levels in the full-term infant (FI) were higher at birth and became similar to PI's from 12 weeks, but were overtaken by levels in PI (both cohorts) at 24 weeks. The FI's Cu was significantly elevated by 8 weeks; mean (SD), 0.81 (0.16) micromol/ml vs 1.25 (0.19) micromol/ml; P < 0.01, paired t-test. Decreased growth rate, nonpitting pedal edema, exaggerated physiological anaemia and chronic lung disease were few morbidities noted in association with very low Cu and CLP levels (n = 15). Newborns with serum Cu and CLP>0.2 micromol/ml and >2.3 micromol/dl, respectively, did not have a poor outcome. It is tempting to suggest that absent serum CLP activity may portend a poor prognosis. Our findings (number albeit small) could be taken as a preliminary normative data for further comparisons.

Plant growth regulators are an appealing and fast-track method for overcoming the drastic effects of drought. Paclobutrazol (PBZ), a member of the triazole family, is a versatile chemical compound that acts as a growth regulator and retardant. This study assessed the effect of two concentrations of PBZ (30 and 60 mg L–1) on strawberry plants under two drought stress levels [40% and 80% field capacity (FC)] and 100% FC set as the control. The findings revealed that drought significantly affected strawberry plants, elevating the levels of reactive oxygen species (ROS) like hydrogen peroxide (H2O2), hydroxide ion (OH–), and superoxide ion (O2–), and reactive carbonyl species (RCS) like malondialdehyde (MDA), methylglyoxal (MG), 4-hydroxy-(E)-2-hexenal (HEE), acetone, propionaldehyde (PD), acetaldehyde (ACD), and formaldehyde (FMD), due to decreased antioxidant activities. The study also found that glutathione (GSH) is crucial in ROS and RCS scavenging. In addition, the presence of thiol groups (–SH compounds) in strawberry plants under moderate and severe drought stress was a noteworthy finding. Due to excess thiol production, drought stress significantly decreased MDA, MG, HEE, PD, ACD, FMD, and acetone levels. However, PBZ application decreased the total thiol content in strawberry leaves. Furthermore, PBZ improved plant growth and development by modulating the production of endogenous growth hormones such as abscisic acid, indole-3-acetic acid, ethylene, gibberellic acid, strigolactones, and brassinosteroids, while increasing cytokinin levels. These changes led to enhanced enzymatic and non-enzymatic antioxidant activities, reduced stomatal conductance, increased relative water content in leaves, improved chlorophyll content, elevated photosynthetic rate, and ultimately enhanced increased fruit yield. Moreover, drought stress enhanced metabolic activity (α-tocopherol, β-carotene, total phenolic content, anthocyanin, and proline) in strawberry plants, indicating that metabolites also function as stress scavengers/protectors. Background: Chromium (Cr) toxicity significantly threatens agricultural ecosystems worldwide, adversely affecting plant growth and development and reducing crop productivity. Trehalose, a non-reducing sugar has been identified as a mitigator of toxic effects induced by abiotic stressors such as drought, salinity, and heavy metals. The primary objective of this study was to investigate the influence of exogenously applied trehalose on maize plants exposed to Cr stress. Results: Two maize varieties, FH-1046 and FH-1453, were subjected to two different Cr concentrations (0.3 mM, and 0.5 mM). The results revealed significant variations in growth and biochemical parameters for both maize varieties under Cr-induced stress conditions as compared to the control group. Foliar application of trehalose at a concentration of 30 mM was administered to both maize varieties, leading to a noteworthy reduction in the detrimental effects of Cr stress. Notably, the Cr (0.5 mM) stress more adversely affected the shoot length more than 0.3mM of Cr stress. Cr stress (0.5 mM) significantly reduced the shoot length by 12.4% in FH-1046 and 24.5% in FH-1453 while Trehalose increased shoot length by 30.19% and 4.75% in FH-1046 and FH-1453 respectively. Cr stress significantly constrained growth and biochemical processes, whereas trehalose notably improved plant growth by reducing Cr uptake and minimizing oxidative stress caused by Cr. This reduction in oxidative stress was evidenced by decreased production of proline, SOD, POD, MDA, H2O2, catalase, and APX. Trehalose also enhanced photosynthetic activities under Cr stress, as indicated by increased values of chlorophyll a, b, and carotenoids. Furthermore, the ameliorative potential of trehalose was demonstrated by increased contents of proteins and carbohydrates and a decrease in Cr uptake. Conclusions: The study demonstrates that trehalose application substantially improved growth and enhanced photosynthetic activities in both maize varieties. Abstract Background Drought is a major determinant for growth and productivity of all crops, including cereals, and the drought-induced detrimental effects are anticipated to jeopardize world food security under the ongoing global warming scenario. Biostimulants such as humic acid (HA) can improve drought tolerance in many cereals, including maize and sorghum. These two plant species are genetically related; however, maize is more susceptible to drought than sorghum. The physiological and biochemical mechanisms underlying such differential responses to water shortage in the absence and presence of HA, particularly under field conditions, are not fully understood. Results Herein, the effects of priming maize and sorghum seeds in 100 mg L−1 HA on their vegetative growth and physiological responses under increased levels of drought (100%, 80%, and 60% field capacity) were simultaneously monitored in the field. In the absence of HA, drought caused 37.0 and 58.7% reductions in biomass accumulation in maize compared to 21.2 and 32.3% in sorghum under low and high drought levels, respectively. These responses were associated with differential retardation in overall growth, relative water content (RWC), photosynthetic pigments and CO2 assimilation in both plants. In contrast, drought increased root traits as well as H2O2, malondialdehyde, and electrolyte leakage in both species. HA treatment significantly improved the growth of both plant species under well-watered and drought conditions, with maize being more responsive than sorghum. HA induced a 29.2% increase in the photosynthetic assimilation rate in maize compared to 15.0% in sorghum under high drought level. The HA-promotive effects were also associated with higher total chlorophyll, stomatal conductance, RWC, sucrose, total soluble sugars, total carbohydrates, proline, and total soluble proteins. HA also reduced the drought-induced oxidative stress via induction of non-enzymic and enzymic antioxidants at significantly different extents in maize and sorghum. Conclusion The current results identify significant quantitative differences in a set of critical physiological biomarkers underlying the differential responses of field-grown maize and sorghum plants against drought. They also reveal the potential of HA priming as a drought-alleviating biostimulant and as an effective approach for sustainable maize and sorghum production and possibly other crops in drought-affected lands.

Background: Kale, a versatile cruciferous crop, valued for its pro-health benefits, stress resistance, and potential applications in forage and cosmetics, holds promise for further enhancement of its bioactive compounds through in vitro cultivation methods. Micropropagation techniques use cytokinins (CKs) which are characterized by various proliferative efficiency. Despite the extensive knowledge regarding CKs, there remains a gap in understanding their role in the physiological mechanisms. That is why, here we investigated the effects of three CKs – kinetin (Kin), 6-benzylaminopurine (BAP), and 2-isopentenyladenine (2iP) – on kale physiology, antioxidant status, steroidal metabolism, and membrane integrity under in vitro cultivation. Results: Our study revealed that while BAP and 2iP stimulated shoot proliferation, they concurrently diminished pigment levels and photosynthetic efficiency. Heightened metabolic activity in response to all CKs was reflected by increased respiratory rate. Despite the differential burst of ROS, the antioxidant properties of kale were associated with the upregulation of guaiacol peroxidase and the scavenging properties of ascorbate rather than glutathione. Notably, CKs fostered the synthesis of sterols, particularly sitosterol, pivotal for cell proliferation and structure of membranes which are strongly disrupted under the action of BAP and 2iP possibly via pathway related to phospholipase D and lipoxygenase which were upregulated. Intriguingly, both CKs treatment spurred the accumulation of sitostenone, known for its ROS scavenging and therapeutic potential. The differential effects of CKs on brassicasterol levels and brassinosteroid (BRs) receptor suggest potential interactions between CKs and BRs. Conclusion: Based on the presented results we conclude that the effect evoked by BAP and 2iP in vitro can improve the industrial significance of kale because this treatment makes possible to control proliferation and/or biosynthesis routes of valuable beneficial compounds. Our work offers significant insights into the nuanced effects of CKs on kale physiology and metabolism, illuminating potential avenues for their application in plant biotechnology and medicinal research. This study investigates the impact of anthocyanin treatment on rice plants under drought stress, focusing on phenotypic, molecular, and biochemical responses. Anthocyanin were treated to one month old plants one week before the droughtexposure. Drought stress was imposed by using 10% polyethylene glycol (PEG 6000). Anthocyanin-treated plants exhibited significant enhancements in various traits, including growth parameters and reproductive characteristics, under normal conditions. When subjected to drought stress, these plants displayed resilience, maintaining or improving essential morphological and physiological features compared to non-treated counterparts. Notably, anthocyanin application mitigated drought-induced oxidative stress, as evidenced by reduced levels of reactive oxygen species (ROS) and lipid membrane peroxidation. The study also elucidates the regulatory role of anthocyanins in the expression of flavonoid biosynthetic genes, leading to increased levels of key secondary metabolites. Incorporating Curcumin into animal diets holds significant promise for enhancing both animal health and productivity, with demonstrated positive impacts on antioxidant activity, anti-microbial responses. Therefore, this study aimed to determine whether adding Curcumin to the diet of dairy calves would influence ruminal fermentation, hematologic, immunological, oxidative, and metabolism variables. Fourteen Jersey calves were divided into a control group (GCON) and a treatment group (GTRA). The animals in the GTRA received a diet containing 65.1 mg/kg of dry matter (DM) Curcumin (74% purity) for an experimental period of 90 days. Blood samples were collected on days 0, 15, 45, and 90. Serum levels of total protein and globulins were higher in the GTRA group (P < 0.05) than the GCON group. In the GTRA group, there was a reduction in pro-inflammatory cytokines (IL-1ß and IL-6) (P < 0.05) and an increase in IL-10 (which acts on anti-inflammatory responses) (P < 0.05) when compared to the GCON. There was a significantly higher (P < 0.05) concentration of immunoglobulin A (IgA) in the serum of the GTRA than the GCON. A Treatment × Day interaction was observed for haptoglobin levels, which were higher on day 90 in animals that consumed Curcumin than the GCON (P < 0.05). The catalase and superoxide dismutase activities were significantly higher (P < 0.05) in GTRA, reducing lipid peroxidation when compared to the GCONT. Hematologic variables did not differ significantly between groups. Among the metabolic variables, only urea was higher in the GTRA group when compared to the GCON. Body weight and feed efficiency did not differ between groups (meaning the percentage of apparent digestibility of dry matter, crude protein, and acid detergent fiber (ADF) and neutral detergent fiber (NDF). There was a tendency (P = 0.09) for treatment effect and a treatment x day interaction (P = 0.05) for levels of short-chain fatty acids in rumen fluid, being lower in animals that consumed curcumin. There was a treatment vs. day interaction (P < 0.05) for the concentration of acetate in the rumen fluid (i.e., on day 45, had a reduction in acetate; on day 90, values were higher in the GTRA group when compared to the GCON). We conclude that there was no evidence in the results from this preliminary trial that Curcumin in the diet of dairy calves interfered with feed digestibility. We investigated environmentally correlated abiotic stressor desiccation (D), heat (H), and starvation (S) in the generation of reactive oxygen and nitrogen species (RONS) using Drosophila melanogaster larvae as an experimental model, subjected to either individual stressors or exposed to a combinatorial form of stressors (D + H, H + S, and D + S). The study was also extended to find synergistic endpoints where the impacts of all three stressors (D + H + S) were exerted simultaneously. We estimated the lethal time (LT20) at specific doses using regression and probit analyses based on the larval survival. LT20 values were used as the base-level parameter for further oxidative stress experimental analysis work. First, all stressors led to the activation of a typical common oxidative stress-mediated response irrespective of the mode of exposure. As envisaged, D. melanogaster larvae exhibited a homeostatic stress tolerance mechanism, triggering an antioxidant defense mechanism, indicated by an elevated level of total antioxidant capacity and enhanced activities of superoxide dismutase, catalase, glutathione reductase, and glutathione peroxidase. In all types of stress-exposed regimes, we found a negative impact of stressors on the activity of mitochondrial enzyme aconitase. Elevated levels of other oxidative stress markers, viz., lipid peroxidation, protein carbonyl content, and advanced oxidative protein products, were obvious although the increment was treatment-specific. Desiccation stress proved to be the most dominant stressor compared to heat and starvation. Among the combination of stressors, rather than a single stressor, D + H impacted more than other binary stress exposures. Focusing on the impact of singular versus combinatorial stress exposure on RONS generation, we observed an increase in the RONS level in both singular and combinatorial forms of stress exposure although the magnitude of the increment varied with the nature of stressors and their combinations. The present study indicated an “additive” effect when all three stressors (D + H + S) operate simultaneously, rather than a “synergistic” effect.

Background: Arsenic (As) is a highly toxic and carcinogenic pollutant commonly found in soil and water, posing significant risks to human health and plant growth. Objective: The objectives of this study to evaluate morphological, biochemical, and physiological markers, as well as ion homeostasis, to alleviate the toxic effects of As in sunflowers through the exogenous application of salicylic acid (SA), γ-aminobutyric acid (GABA), and their combination. Methods: A pot experiment was conducted using two sunflower genotypes, FH-779 and FH-773, subjected to As stress (60 mg kg−1) to evaluate the effects of SA at 100 mg L−1, GABA at 200 mg L−1, and their combination on growth and related physiological and biochemical attributes under As stress. Results: The study revealed that As toxicity had a detrimental effect on various growth parameters, chlorophyll pigments, relative water content, total proteins, and nutrient uptake in sunflower plants. It also led to increased oxidative stress, as indicated by higher levels of malondialdehyde (MDA) and hydrogen peroxide (H2O2), along with As accumulation in the roots and leaves. However, the application of SA and GABA protected against As-induced damage by enhancing the enzymatic antioxidant defense system. This was achieved through the activation of superoxide dismutase (SOD), catalase (CAT), and peroxidase (POD) activities, as well as an increase in osmolytes. They also improved nutrient acquisition and plant growth under As toxicity. Conclusions: We investigated the regulatory roles of SA and GABA in mitigating arsenic-induced phytotoxic effects on sunflower. Our results revealed a significant interaction between SA and GABA in regulating growth, photosynthesis, metabolites, antioxidant defense systems, and nutrient uptake in sunflower under As stress. These findings provide valuable insights into plant defense mechanisms and strategies to enhance stress tolerance in contaminated environments. This study aimed to examine the effects of inoculating Fritillaria taipaiensis P.Y.Li leaves with different strains of potassium-solubilizing bacteria (KSB), or combinations thereof, focusing on aspects of photosynthesis and physiological and biochemical characteristics. At present, some studies have only studied the rhizosphere microbial community characteristics of F. taipaiensis and have not discussed the effects of different microbial species on the growth promotion of F. taipaiensis. This paper will start from the perspective of potassium-solubilizing bacteria to conduct an in-depth study. Seed cultivation commenced at the base with three different KSBs in early October 2022. The growth of F. taipaiensis leaves was observed after different treatments. Both single-plant and compound inoculations were executed. A total of eight treatment groups were established, with aseptic fertilizer and sterilized soil functioning as the control group. The results reveal that intercellular CO2 concentration (Ci), stomatal conductance (Gs), and transpiration rate (Tr) were at their apex in the S7 group. Most treatment groups exhibited an increase in leaf area, photosynthetic pigment content, soluble sugar, soluble protein, Superoxide Dismutase (SOD), Peroxidase (POD), Catalase (CAT) activities, and proline content. The expression levels of POD, SOD, and CAT genes were evaluated, following inoculation with different KSB. The highest was the S7 group. The inoculation with various KSB, or combinations thereof, appears to bolster the growth and development of F. taipaiensis. The composite inoculation group S7, comprising Bacillus cereus, Burkholderia cepacia, and Bacillus subtilis, manifested the most favorable impact on the diverse indices of F. taipaiensis, thereby furnishing valuable insights for the selection of bacterial fertilizer in the artificial cultivation of F. taipaiensis. Herein we attempt to shed light on the potential improving effect of Eruca sativa seeds (ESS) on the reproductive aspects of male Japanese quails. To accomplish this objective, two groups of quails were supplemented with ESS powder at doses of 5 and 10 g/kg feed from 7 days to 140 days of age, in addition to the control group, which did not receive treatment. Forty males were reared singly in cages to evaluate sperm characters and 32 males were raised with 64 females to evaluate fertility and sperm penetrability. Sixty‐six phytochemical compounds were found according to gas chromatography–mass spectrometry analysis of ESS. The most plentiful ones are 13‐docosenoic acid methyl ester, 9‐octadecenoic acid methyl ester, and linoleic acid methyl ester. Both 5 g/kg and 10 g/kg doses of ESS showed similar effectiveness in enhancing various reproductive parameters, including gonadal index, sperm characteristics, fertility, libido, and cloacal gland attributes. However, some aspects like sperm concentration and testosterone levels exhibited a dose‐dependent response. There is no significant change in mortality rate of supplemented groups compared to the control one. ESS also caused a reduction in feed intake and an enhancement in feed conversion ratio without affecting final body weight and body weight gain. This suggests potential nutritional benefits beyond reproductive health. The low‐dose‐fed group showed a significant reduction in total cholesterol and malondialdehyde compared to the high‐dose‐fed and unfed groups. The higher dose notably increased total antioxidant capacity compared to the lower dose and control group. Despite the positive effects on male reproductive parameters, there wasn't a significant impact on hatchability percentage, indicating that while male fertility improved, it might not have directly affected the viability of the eggs. Overall, the study suggests that ESS could be a safe and promising addition to the diet of male Japanese quails to enhance their reproductive capabilities without adverse effects. Many studies have found that tea has an antioxidant, anti-inflammatory, anti-cancer, anti-obesogenic and anti-diabetic effect, mostly associated with the content of anti-oxidant compounds. Polyphenols, being the main secondary metabolites in tea, are often considered the physiological markers determining a tea's quality. Apart from the tea production process and tea components, brewing conditions can also influence the levels of antioxidants in tea. This study aimed to verify whether the brewing time of various tea types (5, 10 and 15 min) affects the level of extraction of antioxidant compounds into infusions and their antioxidant activity. We examined 11 types of tea: green leaf tea, green tea bags, white tea bags, black tea bags, red tea bags, black leaf tea, yerba mate, raspberry tea bags, butterfly pea flower (Clitoria ternatea) tea, white lychee plum tea and hibiscus flower tea. Total polyphenol (TPC), flavonoids and anthocyanins content, as well as determination of antiradical and antioxidant capacity with DPPH radical and ABTS radical cation, were determined using spectrophotometric assays. Due to the antioxidant activity of tea infusions, the optimum brewing time for green tea (leaf and bags), black tea (leaf and bags), butterfly pea flower tea, white tea, white lychee plum tea, raspberry tea and yerba mate is 15 min. Red tea brewing time should be ten minutes, and for hibiscus flower tea it should be five minutes. The results refer to the brewing temperature recommended by tea manufacturers. Elevated sodium level (Na+) poses significant threat to crop plant physio-biochemical processes, leading to impaired growth followed by decline in productivity. Addressing this challenge, requires an eco-friendly and cost-effective strategy that enhances plant salt stress tolerance capacity. In this context, the exogenous source of plant growth regulators (PGRs) proved to be an efficient approach. Of various PGRs, salicylic acid (SA) is an emerging signaling molecule that boosts plant stress endurance mechanism. This study investigates SA-mediated salt stress tolerance in maize (Zea mays L.) seedlings, by examining morpho-physiological and biochemical traits. Maize seedlings were subjected to varying levels of salt stress (0, 25, 50, 75, 100, and 150 mM NaCl) for a period of 10-days. The results revealed that, a substantial decline in germination percentage, shoot and root length, plant biomass, vigour index, and various other physiological parameters under salt stress causing concentrations. Conversely, salt stress increased oxidative stress indicators, including hydrogen peroxide (H2O2) and malondialdehyde (MDA), osmolytes and elemental concentrations as well as antioxident enzymes (SOD, CAT, POX, APX, GR, AsA). However, the exogenous supplementation of SA at 0.1 mM significantly restored most morpho-physiological attributes in maize under salt stress conditions. This suggests that SA actively triggers the ascorbate-glutathione (AsA-GSH) pathway and other key enzymes, leading to sodium extrusion and improving antioxidant defense in maize seedlings. This finding provides valuable insights for maize farmers that employing SA could lead to improved maize production in saline soils.

Anthropogenic changes to the environment expose wildlife to many pollutants. Among these, tropospheric ozone is of global concern and a highly potent pro-oxidant. In addition, human activities include several other implications for wildlife, e.g., changed food availability and changed distribution of pathogens in cities. These co-occurring habitat changes may interact, thereby modulating the physiological responses and costs related to anthropogenic change. For instance, many food items associated with humans (e.g., food waste and feeders for wild birds) contain relatively more ω6-than ω3-polyunsaturated fatty acids (PUFAs). Metabolites derived from ω6-PUFAs can enhance inflammation and oxidative stress towards a stimulus, whereas the opposite response is linked to ω3-derived metabolites. Hence, we hypothesized that differential intake of ω6-and ω3-PUFAs modulates the oxidative stress state of birds and thereby affects the responses towards pro-oxidants. To test this, we manipulated dietary ω6:ω3 ratios and ozone levels in a full-factorial experiment using captive zebra finches (Taeniopygia guttata). Additionally, we simulated an infection, thereby also triggering the immune system’s adaptive pro-oxidant release (i.e., oxidative burst), by injecting lipopolysaccharide. Under normal air conditions, the ω3-diet birds had a lower antioxidant ratio (GSH/GSSG ratio) compared to the ω6-diet birds. When exposed to ozone, however, the diet effect disappeared. Instead, ozone exposure overall reduced the total concentration of the key antioxidant glutathione (tGSH). Moreover, the birds on the ω6-rich diet had an overall higher antioxidant capacity (OXY) compared to birds fed a ω3-rich diet. Interestingly, only the immune challenge increased oxidative damage, suggesting the oxidative burst of the immune system overrides the other pro-oxidative processes, including diet. Taken together, our results show that ozone, dietary PUFAs, and infection all affect the redox-system, but in different ways, suggesting that the underlying responses are decoupled despite that they all increase pro-oxidant exposure or generation. Despite lack of apparent cumulative effect in the independent biomarkers, the combined single effects could together reduce overall cellular functioning and efficiency over time in wild birds exposed to pathogens, ozone, and anthropogenic food sources.

This is a narrative review of the evidence of α-tocopherol importance in human health, especially with regards to its vitamin role. α-Tocopherol is a potent peroxyl radical scavenger, and this role is prominent in its efficacy in maintaining the metabolic health of tissues. Vitamin E deficiency is discussed as a tool to understand the impact of α-tocopherol’s absence promoting increased lipid peroxidation and polyunsaturated fatty acid depletion. Downstream deficiency consequences include impacts on choline and one-carbon metabolism, glucose and energy metabolism, and their interactions with critical thiols, such as glutathione. Importantly, human vitamin E deficiency, caused by genetic defects in the α-tocopherol transfer protein (α-TTP), provides important clues for the necessity of α-tocopherol for the peripheral nervous system. Moreover, α-TTP expression in the liver, brain, eyes, and placenta illustrates that these tissues are especially vulnerable and require this specific α-tocopherol delivery mechanism for their protection. Although clinical trial evidence is limited and equivocal about the health benefits of vitamin E supplements, there is epidemiologic evidence of the long-term benefits of increased α-tocopherol intakes in ’healthy’ diets (high in vegetables and fruits, fish, nuts, and seeds, as well as fiber). Significance statement The elaborate regulation of α-tocopherol concentrations by the human body suggests that the consistent consumption of the recommended amounts of dietary α-tocopherol (15 mg) over a lifetime are protective of the at-risk tissues, as well as providing protection from chronic diseases.

As a result of environmental difficulties and water scarcity, maize production has been challenged in arid and semi - arid regions. A management strategy for stabilizing corn production under water shortage conditions involves the use of drought - tolerant hybrids and suitable irrigation regimes. The study was conducted in the Prof. Dr. Abdulkadir AKÇIN trial area of "Selcuk University, Agriculture Faculty, Crop Science Department, Konya, TÜRKİYE" during 2019 and 2020 growing seasons. The aim of the current work is determining the effects of deficit irrigation and grain colour factors on water shortage tolerance of maize in terms of bio - active compounds, quality traits and grain yield. The factors of the trial were irrigations [50% (I1), 75% (I2) and 100% (I3) of evaporation from Class A evaporation PAN] and genotypes [DKC 5783, red corn and Sakarya]. It was noted that DKC 5783 had the highest values at two (thousand grain weight, grain yield), Sakarya had at two (total phenolic compounds and grain crude oil) and red corn had at four (total anthocyanin content, total antioxidant activity, grain crude protein and starch) properties among eight under I1 during 2019 as well as red corn had at two (total anthocyanin content and total phenolic compounds), Sakarya had at two (grain crude protein and grain crude oil) and DKC 5783 had highest values at four (thousand grain weight, total antioxidant activity, starch and grain yield) features among eight under I1 during 2020. Thousand grain weight values of I3 were 6.71% and 0.57% more than I2 while starch was 9.19% and 3.96% more than I2 during 2019 - 2020. Red corn had better contents of bio - active compounds than other two varieties during both years of the trial. The mean yield of I3 was 6.87% more than I2 in 2019 and 9.05% more than I2 in 2020 which revealed that regulated deficit irrigation might help growers to cope with decline in water availability also 25% water restriction caused tolerable decreases in grain yield and some yield compounds of the current work. Enhanced anthropogenic activities affect agricultural production in many ways. An integrated study assessing the combined impact of increased tropospheric ozone and carbon dioxide on agriculture is still elusive. An investigation was carried out to study the impacts of elevated levels of CO2 (eCO2) and O3 (eO3) and their interaction on a C4 plant, maize cultivar DHM117, based on growth, physiological, and yield responses using open top chambers. Plant growth parameters remained unaffected due to eCO2 exposure. However, the toxic impact of O3 was alleviated to some extent under eCO2 concentration, as revealed by the increment in total biomass under combined exposure. CO2 fertilization increased yield by 13.8%, while a reverse trend was observed under eO3 treatment. Lowering of stomatal conductance under eCO2 partially protected the plants against O3 uptake and resulting phytotoxicity. Stimulation of total phenolic and phenylalanine ammonia lyase (PAL) activity confirmed the activation of defence mechanism to counter the oxidative stress under high O3 dose. Likewise, a significant rise in the activities of antioxidative enzymes [peroxidase (POX) and ascorbate peroxidase (APX)] involved in defence mechanisms was observed under ECO2 + EO3, leading to a reduced accumulation of H2O2 content in the test plant. This observation was further affirmed by the redundancy analysis, which revealed the significant role of enzymatic antioxidants (explained variance = 15.5%) in compensating for the damaging effect of reactive oxygen species (ROS), translating to significant changes in yield attributes under combined exposure of CO2 and O3. Sweet basil is a perishable green herb that has a short shelf life after harvest. Techniques to extend its shelf life and prolong its storability may be commercially beneficial. In this study, the impact of different ultrasound powers on the postharvest quality and bioactive compounds of sweet basil during storage was investigated. The sampled basil leaves were immersed in an ultrasonic bath (40 kHz) at 200 or 400 W for 3 min at ambient temperature (28 ℃). The control samples were treated with tap water. All of the treatments were packed in a basket covered with a polyethylene sheet and kept at 13 ℃. Changes in the physical and chemical properties were recorded every 3 h to determine the impact of the ultrasound treatment on prolonging the postharvest quality and maintaining the bioactive compounds of the sweet basil leaves. The results showed that ultrasound at 400 W significantly reduced the weight loss and respiration rate of the sweet basil leaves. Additionally, ultrasound at 400 W increased the total phenolics, total flavonoids, antioxidant activity, and vitamin C content, which were related to the activity of phenylalanine ammonium lyase and ascorbate peroxidase, compared with the control. Abstract The large conductance, calcium, and voltage‐active potassium channels (BKCa) were originally discovered in Drosophila melanogaster as slowpoke (slo). They are extensively characterized in fly models as ion channels for their roles in neurological and muscular function, as well as aging. BKCa is known to modulate cardiac rhythm and is localized to the mitochondria. Activation of mitochondrial BKCa causes cardioprotection from ischemia–reperfusion injury, possibly via modulating mitochondrial function in adult animal models. However, the role of BKCa in cardiac function is not well‐characterized, partially due to its localization to the plasma membrane as well as intracellular membranes and the wide array of cells present in mammalian hearts. Here we demonstrate for the first time a direct role for BKCa in cardiac function and cardioprotection from IR injury using the Drosophila model system. We have also discovered that the BKCa channel plays a role in the functioning of aging hearts. Our study establishes the presence of BKCa in the fly heart and ascertains its role in aging heart function.

Ultrasonic seed (US) treatment could alter seed germination mechanism, however, US induced alterations in morph-physiological attributes and yield of fragrant rice were rarely reported. In the present study, the seeds of three fragrant rice cultivars viz., Xiangyaxiangzhan, Meixiangzhan 2, Ruanhuayou 6100 and one non-fragrant rice viz., Wufengyou 615 were exposed to ultrasonic waves at 20–40 kHz for 1.5 min (T) whereas the seeds without exposure were taken as control (CK). Results showed that US treatment caused minor cracks on seed surface while improved seed germination rate (1.79 %-11.09 %) and 3-indoleacetic acid (IAA) (3.36 %-46.91 %). Furthermore, peroxidase (POD) activity and methionine sulfoxide reductase activity was increased by 29.15 %-74.13 % and 11.26 %-20.87 %, respectively; however, methionine sulfoxide reductase related protein repairing gene MSRA4 was down-regulated by 17.93 % −41.04 % under T, compared to CK. Besides, US treatment also improved soluble protein in flag leaf (0.92 %-40.79 %), photosynthesis (3.37 %-16.46 %), biomass (5.17 %-31.87 %), as well as 2-acetyl-1-pyrroline content (4.77 %-15.48 %) in rice grains. In addition, multivariate analysis showed that the dry weight at the maturity stage were significantly related to the POD, glutathione reductase (GR) activity, IAA, and abscisic acid (ABA) content while germination rate was positively related to the GR activity, ABA content, and yield, but which were negatively related to the IAA and gibberellic acid content.

Drought stress is an ever‐present threat to wheat growth, development, and productivity, especially in arid and semi-arid areas where rainfall is an essential aid to agriculture. The application of vermicompost has been proven to be an efficient approach to combat the drought-induced growth and developmental limitation of wheat plants and to promote cost-effective and sustainable crop production. A wire-house pot experiment was conducted to examine the effects of cow manure vermicompost on morpho-physiological and biochemical attributes of wheat seedlings under different deficit water levels. The treatments were included: three drought levels; control (70 % of field capacity (FC, D0)), mild drought (45 % of FC, D1), and severe drought (30 % of FC, D1), four vermicompost application rates; control, 4, 6 and 8 t ha−1 (designated as VT0, VT1, VT2 and VT3, respectively), and two contrasting wheat cultivars; Faisalabad-08 (drought tolerant) and Galaxy-13 (drought sensitive). Results showed that vermicompost application improved crop performance under control and drought stress conditions where VT2 depicted significantly higher values in both cultivars, particularly under drought stress. Under severe drought, VT2 increased root fresh weight by 6.13 and 10.63 %, shoot fresh weight by 15.62 % and 23.58 %, root dry weight by 40.81 % and 50 % and shoot dry weight by 20.68 and 22.22 % in Faisalabad-08 and Galaxy-13, respectively. Also, VT2-treated seedlings exhibited significantly higher gas exchange attributes, chlorophyll pigments and antioxidative enzyme activities under drought conditions. Among cultivars, Faisalabad-08 was found comparatively more drought tolerant than Galaxy-13. Our findings demonstrated that drought severely affects morphological, physiological, and biochemical attributes of wheat. However, soil applied vermicompost proved beneficial for improved morphophysiological and biochemical traits under the regime of drought stress in wheat.

Numerous neurodegenerative diseases result from altered ion channel function and mutations. The intracellular redox status can significantly alter the gating characteristics of ion channels. Abundant neurodegenerative diseases associated with oxidative stress have been documented, including Parkinson’s, Alzheimer’s, spinocerebellar ataxia, amyotrophic lateral sclerosis, and Huntington’s disease. Reactive oxygen and nitrogen species compounds trigger posttranslational alterations that target specific sites within the subunits responsible for channel assembly. These alterations include the adjustment of cysteine residues through redox reactions induced by reactive oxygen species (ROS), nitration, and S-nitrosylation assisted by nitric oxide of tyrosine residues through peroxynitrite. Several ion channels have been directly investigated for their functional responses to oxidizing agents and oxidative stress. This review primarily explores the relationship and potential links between oxidative stress and ion channels in neurodegenerative conditions, such as cerebellar ataxias and Parkinson’s disease. The potential correlation between oxidative stress and ion channels could hold promise for developing innovative therapies for common neurodegenerative diseases.

Purpose: To investigate the relationship between the Oxidative Balance Score (OBS) and kidney stone risk using NHANES 2007-2018 data, and to explore potential mechanisms and population-specific effects.Materials and methods: Data from the NHANES 2007-2018 were analyzed. OBS was calculated based on 16 dietary components and 4 lifestyle components. Multivariate logistic regression was employed to investigate the relationship between OBS and kidney stone. Further stratified analyses were conducted to examine the associations across different subgroups.Results: A total of 19,799 participants were included in the study. There was a consistent inverse association between OBS and the risk of kidney stones (OR = 0.97; 95% CI: 0.96–0.99). After dividing the participants into quartiles based on OBS, compared to the lowest quartile of OBS, the risk of kidney stones in the highest quartile of OBS was reduced by 33% (95% CI 0.50–0.89; p = 0.002). This association was consistent across both dietary and lifestyle OBS scores. The protective effect of OBS was notably pronounced among Non-Hispanic white and Other race groups, and among individuals with a higher level of education. However, the association was not significant among individuals with diabetes.Conclusion: A higher OBS, indicating a balance skewed towards antioxidants, is associated with a reduced risk of kidney stones, especially among specific population subgroups. These findings underscore the potential role of oxidative balance in kidney stone pathogenesis and highlight the importance of considering individual and population-specific factors in future research and preventive strategies.

Chromium (Cr) is the seventh most toxic heavy metal on the earth's crust. Crop growth, development, and yield are greatly hindered by the massive accumulation of Cr that discharges from various industries and mines in agricultural land. Chromium can mediate phytotoxicity either via direct interaction with multiple plant components and metabolic pathways or by promoting reactive oxygen species (ROS) generation. Thus, this investigation evaluated the effect of Cr on growth, physiological and oxidative stress, and other parameters, including essential oil (EO) constituents of Mentha arvensis L. plants. The soil was treated with various concentrations of Cr (10, 20, 40, 60, and 80 mg kg−1), except the control. In this study, 20 mg kg−1 of Cr concentration was beneficial in increasing plant growth via the enhancement of biomass, photosynthesis, biochemical markers and EO constituents in M. arvensis. The data demonstrated that the morphological and physiological parameters were negatively correlated with high Cr concentrations (above 20 mg kg−1). It has been hypothesized that Cr poisoning leads to oxidative damage, as evidenced by decreases in photosynthetic rate, chlorophyll, and stomatal characteristics, as well as increases in lipid peroxidation and ROS. The accumulation of Cr positively correlates with the enhancement of proline and antioxidant activities of M. arvensis. Thus, we concluded that excess Cr causes redox imbalance by uplifting the level of lipid peroxidation and ROS by destabilizing photosynthetic machinery and other physiological and morphological markers in plants. The study was performed to examine the role of heme oxygenase (HO) in tolerance and accumulation of cadmium in Eruca sativa (var. DPY BKP 98). Seedlings of E. sativa were subjected to different levels of CdCl2 (10 to 150 μM) for 96 h. The result shows that lower dose of Cd (10 to 25 μM) had mild effect on plant biomass and photosynthetic pigments due to which E. sativa can efficiently uptake and translocate Cd, but higher concentration significantly reduces these attributes by increasing membrane damage (MDA and H2O2 content) and downregulating the activities of major antioxidants. However, at 125 μM treatment improved HO activity was recorded that enhances plant growth by increasing plant biomass and chlorophyll content; augmented activities of antioxidants (proline, APX, and CAT); lowered oxidative damage; and increased tolerance ability of plant toward Cd. Interestingly over-expression of HO gene at 125 μM increases Cd accretion and bioconcentration factor (BCF) both in leaves and roots, and enhances the translocation of cadmium from roots to leaves. Thus, our findings for the first time proposed that HO operates dual function in Eruca seedlings by rising phytoremediation efficiency and escalating stress tolerance via upregulating antioxidant defense machinery. This study aimed to investigate the physiological and cellular mechanisms of Sesuvium portulacastrum under heavy metal stress to evaluate possible tolerance and adaptation mechanisms in a metal-polluted environment. The physiological and cellular ultrastructural responses of S. portulacastrum were studied hydroponically under exposure to a range of cadmium (Cd) concentrations (50 µM to 600 µM) for 28 days. The activity of antioxidant enzymes like catalase (CAT), superoxide dismutase (SOD), and peroxidase (POD), changes in chlorophyll, and cellular ultrastructural content were examined. There was no significant difference in chlorophyll content in the leaf under the stress of 300 μM, but 400 μM and 600 μM Cd stress showed significantly decreased chlorophyll content. The SOD activity indicates an increase under the Cd stress of 100 μM for leaves, 300 μM for stems, and 50 μM for roots; after that, the SOD activity gradually decreased with increasing Cd concentrations. But POD activity was considerably increased with increasing Cd stress. CAT activity showed a gradual increase in concentrations until 300 μM of Cd stress and then decreased sharply in roots, stems, and leaf tissues. Cd stress had a considerable impact on the structure of the roots, stems, and leaves cells, such as distorted and thinner cell walls and the deformation of chloroplasts, mitochondria, and other organelles. Therefore, the increased number of nucleolus in the cell nucleus suggests that cells may be able to maintain their protein synthesis in a stressful environment. This study concludes that SOD is the dominant antioxidant enzyme activity during low Cd toxicity (<100 μM), while POD is the dominant enzyme activity during higher Cd toxicity (>100 μM). Low temperature stress significantly threatens crop productivity and economic sustainability. Plants counter this by deploying advanced molecular mechanisms to perceive and respond to cold stress. Transmembrane proteins initiate these responses, triggering a series of events involving secondary messengers such as calcium ions (Ca2+), reactive oxygen species (ROS), and inositol phosphates. Of these, calcium signaling is paramount, activating downstream phosphorylation cascades and the transcription of cold-responsive genes, including coldregulated (COR) genes. This review focuses on how plants manage freezeinduced damage through dual strategies: cold tolerance and cold avoidance. Tolerance mechanisms involve acclimatization to decreasing temperatures, fostering gradual accumulation of cold resistance. In contrast, avoidance mechanisms rely on cryoprotectant molecules like potassium ions (K+), proline, glycerol, and antifreeze proteins (AFPs). Cryoprotectants modulate intracellular solute concentration, lower the freezing point, inhibit ice formation, and preserve plasma membrane fluidity. Additionally, these molecules demonstrate antioxidant activity, scavenging ROS, preventing protein denaturation, and subsequently mitigating cellular damage. By forming extensive hydrogen bonds with water molecules, cryoprotectants also limit intercellular water movement, minimizing extracellular ice crystal formation, and cell dehydration. The deployment of cryoprotectants is a key adaptive strategy that bolsters plant resilience to cold stress and promotes survival in freezing environments. However, the specific physiological and molecular mechanisms underlying these protective effects remain insufficiently understood. Therefore, this review underscores the need for further research to elucidate these mechanisms and assess their potential impact on crop productivity and sustainability, contributing to the progressive discourse in plant biology and environmental science. Biochar can change the availability and morphology of soil Cd. However, the influence of biochar on Cd chemical form and subcellular fraction in rice is poorly understood, particularly under different irrigation methods. A pot experiment of biochar application combined with two irrigation methods (continuous flooding and intermittent irrigation, CF and II) was conducted. The Cd accumulation, chemical form and subcellular fraction in rice organs and the associated physiological responses were examined. Biochar significantly reduced soil available Cd (30.85–47.26% and 32.35–52.35%) under CF and II but increased the Cd content (30.4–63.88% and 13.03–18.59%) in brown rice. Additionally, the Cd content in shoots/grains under II was higher than that under CF. Biochar elevated the Cd soluble fraction in roots while lowered the cell wall fraction under both irrigation methods, whereas the opposite result was observed in leaves. Biochar increased water-, ethanol-, and NaCl-extractable Cd in roots meanwhile increased ethanol-extractable Cd in leaves under both irrigation methods. Moreover, the total amount of water-, ethanol-, and NaCl-extractable Cd in rice roots was higher under II than under CF. Related hormones and antioxidant enzymes may also be involved in biochar-mediated Cd accumulation in rice grains. Thus, changes in Cd chemical form and subcellular fraction in the root and leaf are the main mechanisms of biochar-induced rice grains Cd accumulation. Seasonal soil freezing (F) and freeze–thaw cycles (FTCs) are common natural phenomena in high latitude or altitude areas of the world, and seriously affect plant physiological processes. However, studies on the effect of soil F and FTCs on fine roots are less common, especially in subalpine coniferous forests of western Sichuan, China.We set up a controlled experiment in growth chambers to explore the effects of F and FTCs on low‐order fine roots of Picea asperata and differential responses of first‐order roots and the first three root orders (1st, 2nd and 3rd order roots combined as a unit).Soil F and FTCs resulted in serious damage to cell membranes and root vitality of low‐order fine roots, accompanied by increased MDA content and O2·− production. FTCs had a stronger effect than F treatment. In turn, low‐order fine roots are the unit that responds to cold stress. These roots had increased unsaturated fatty acid contents, antioxidant enzyme activities, osmolytes and plant hormones contents when acclimation to cold stress. The first‐order roots were more sensitive to cold stress than the combined first three root orders for several processes (e.g. antioxidant enzymes, osmolytes and hormones) because of their specific structure and physiological activity.This study explains physiological differences in responses of fine roots of different root orders to seasonal soil freezing, which will improve the understanding of fine root heterogeneity and support agriculture and forest management.
