The formation and mitigation of furan in pumpkin puree (PP) were studied during the complete process of producing PP. The content of furan was determined using headspace solid-phase microextraction combined by GC-MS analysis. Various PP samples added with precursors (glucose, fructose, linoleic acid, linolenic acid, β-carotene, ascorbic acid, glutamic acid, alanine, and serine) showed increased furan formation (30.81 ∼ 94.45 µg/kg) compared with the control (30.81 µg/kg), with β-carotene resulting in the formation of the largest amount of furan. The effects of antioxidants, such as caffeic acid, chlorogenic acid, quercetin, and butylated hydroxytoluene, on the reduction of furan in PP containing β-carotene were also investigated. All antioxidants showed significant reduction of furan. During sterilizing, the content of furan was considerably affected by temperature but not heating time. In this study, we used response surface methodology (RSM) to optimize the extraction conditions for recovering lutein from paprika leaves using accelerated solvent extraction (ASE). The lutein content was quantitatively analyzed using a UPLC equipped with a BEH C18 column. A central composite design (CCD) was employed for experimental design to obtain the optimized combination of extraction temperature (°C), static time (min), and solvent (EtOH, %). The experimental data obtained from a twenty sample set were fitted to a second-order polynomial equation using multiple regression analysis. The adjusted coefficient of determination (R(2)) for the lutein extraction model was 0.9518, and the probability value (p=0.0000) demonstrated a high significance for the regression model. The optimum extraction conditions for lutein were temperature: 93.26°C, static time: 5 min, and solvent: 79.63% EtOH. This study investigated total carotenoid content, comparative carotenoid composition, vitamin C content, and total antioxidant capacity of three citrus varieties which are Yuza (Citrus junos Sieb ex Tabaka), Kjool (Citrus unshiu Marcow), and Dangyooja (Citrus grandis Osbeck). Seven carotenoids were identified, with β-cryptoxanthin, astaxanthin, and zeaxanthin being predominant in citrus varieties. Ripening increased the total carotenoid in three citrus varieties. Individual carotenoid of canthaxanthin, astaxanthin, and α-carotene in citrus varieties decreased with maturation, whereas the others increased with ripening. Yuza exhibited the highest total antioxidant capacity in 2,2'-azino-bis-3-ethylbenzthiazoline-6-sulfonic acid (ABTS) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) assays, with VCEAC values of 582.9 mg/100 g and 451.5 mg/100g, respectively. The relative VCEAC values were vitamin C (1.00)>lycopene (0.375), α-carotene (0.304), β-carotene (0.289), β-cryptoxanthin (0.242), and zeaxanthin (0.099). Methanol extract subfractions of the edible white jelly mushroom (Tremella fuciformis), were assessed for the following antioxidant properties: ABTS(+) radical scavenging activity, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, and inhibitory activity of human low-density lipoprotein (LDL) oxidation. Among the subfractions tested, the chloroform subfraction exhibited the strongest antioxidant activity, with the highest total phenolic content (66.31 μg CAE/mg extract) and flavonoids content (5.12 μg QE/mg extract). The ABTS(+) radical scavenging activity of the chloroform subfraction was 7.89 μmol trolox/mg extract, which was the highest among all subfractions. This subfraction also showed the highest DPPH radical scavenging activity and inhibitory activity of LDL oxidation. In addition, the chloroform subfraction demonstrated anti-inflammatory activity through inhibition of nitric oxide production and inducible nitric oxide synthase expression in RAW 264.7 cells. The phytochemical composition of carotenoids, tocopherols, free sugars, organic acids, L-ascorbic acid, capsaicinoids, and flavonoids in green and red paprika (GP and RP), and paprika leaves (PL) cultivated in Korea were analyzed. The ethanolic extracts of GP, RP, and PL were obtained with 80% ethanol, and their antioxidative activities were determined by measuring their ABTS and DPPH radical scavenging activities. RP showed the highest contents of capsanthin (58.33 ± 3.91 mg/100 g dry weight) and L-ascorbic acid (1987.25 ± 19.64 mg/100 g dry weight), and main compounds of PL were lutein, chlorophyll, and γ-tocopherol (96.91 ± 14.58, 2136.71 ± 21.11, and 723.49 ± 54.10 mg/100 g dry weight, respectively). RP showed the strongest antioxidant activity (IC(50) = 55.23 ± 6.77 μg/mL in a 2, 2'-azino-di-[3-ethylbenzthiazoline sulphonate] assay and 150.40 ± 8.07 μg/mL in a 2, 2-diphenyl-2-picrylhydrazyl assay), and the antioxidant activity of PL was higher than β-carotene but lower than RP. The results indicate that the amounts of capsanthin and L-ascorbic acid in RP correlate well with antioxidant activity. PL, which has various phytochemicals such as lutein, chlorophyll, and γ-tocopherol, might be used in nutraceuticals and pharmaceuticals for improving human health.

Three citrus varieties grown in Korea, namely, Yuza (Citrus junos Sieb ex Tanaka), Kjool (Citrus unshiu Marcow), and Dangyooja (Citrus grandis Osbeck), were evaluated for their dietary fiber, total phenolics, total flavonoids, and total carotenoids as well as individual flavonoids composition. The biochemical characteristics of citrus varieties were examined by the antioxidant capacity (731 to 1221 micromol of Trolox equivalent/g), total phenolic (334 to 411 mg of chlorogenic acid equivalent/100 g), total flavonoid contents (214 to 281 mg of catechin equivalent/100 g), and total carotenoid contents (63 to 84 mg/100 g). Six flavonoids, including naringin, naringenin, hesperidin, hesperetin, neohesperidin, and luteolin, were tentatively identified. Naringin, hesperidin, and neohesperidin were the predominant flavonoids in 3 citrus varieties. Among the citrus varieties studied, Yuza showed higher antioxidant activity, total phenolics, total carotenoids, and lower superoxide radical scavenging assay (SRSA) IC(50) values than other varieties. Furthermore, Korean citrus showed higher protective effect on gap-junction intercellular communication (GJIC) as compared to vitamin C and luteolin.

Peanut sprouts are a functional food material rich in phytochemicals, including trans‐resveratrol. This study aimed to optimize the recovery of trans‐resveratrol from peanut sprouts using a combination of peanut varieties and sawdust medium through accelerated solvent extraction (ASE) and the response surface method (RSM). We also aimed to determine the antioxidant activity of this trans‐resveratrol extract. Optimal fermentation periods of sawdust and peanut variety for cultivating peanut sprouts were determined on the basis of trans‐resveratrol content via high‐performance liquid chromatography. The extraction variables temperature, static time, and ethanol concentration were used to create a 20‐sample set fit to a second‐order polynomial equation through multiple regression analysis (R2 = 0.8787, P < 0.01). Trans‐resveratrol content (19.62 ± 2.33 µg/g) peaked in the Palgwang variety cultured in sawdust medium fermented for 45 days. Optimal conditions for ASE were determined regarding the extraction temperature (90.29 °C), static time (3.95 min), and solvent (81.54% EtOH/water), and the predicted trans‐resveratrol content under optimal conditions was 30.23 µg/g. Sawdust medium was more effective in increasing the trans‐resveratrol content than conventional hydroponics, and the optimized process of combining fermented sawdust cultivation for harvesting peanut sprouts with ASE has potential as an efficient method of obtaining mass quantities of trans‐resveratrol from peanut sprouts with improved nutritional and functional properties. Practical Application: This study showed that sawdust medium is more effective than hydroponics in increasing the trans‐resveratrol content in peanut sprouts. The recovery of trans‐resveratrol from peanut sprouts and its antioxidant activity were optimized via accelerated solvent extraction (ASE) and the response surface methodology (RSM). The optimized process of combining fermented sawdust cultivation for harvesting peanut sprouts with ASE potentially provides an efficient method to obtain mass quantities of trans‐resveratrol from peanut sprouts with improved nutritional and functional properties.
