Society's interest in natural and clean products in many areas, such as food and cosmetics, has increased considerably. It has led to the development of new techniques in the packaging of products so that the wastes from the preferred products can be recycled. In this context, Pinus brutia bark was preferred within the scope of the study to transform natural wastes into functional components and use them as packaging material. P. brutia bark (PBB) samples were collected from Bursa, Turkey. PBB samples were ultrasonically extracted using various solvents (acetone, butanol, ethanol, ethyl acetate, hexane, methanol, petroleum ether, and water) and a solvent-acidic hydrolysis system. The phenolic content profile of PBB samples was determined using high-performance liquid chromatography with diode-array detection, and total flavonoid content, antioxidant capacity, and total phenolic content were determined. Chitosan-polyvinyl alcohol (CS-PVA) films loaded with polysaccharides and containing methanolic extract were developed. The physical, chemical, and mechanical properties of the films were characterized. It is known that the thickness of the films determines the mechanical properties required to maintain the integrity of the packaging during storage and transport. From the results of the study, it was concluded that the elongation at break value was higher in CS-PVA-PBB-M films (111.08% ± 10.46%), Young's modulus (31.74 ± 21.37 N/mm 2 ), and tensile strength (3.01 ± 0.50 N/mm 2 ) values were higher in CS-PVA films. In this study, the bark of the Aquilaria agallocha was extracted, and the phenolic content of A. agallocha extract was determined using high-performance liquid chromatography-diode array detector. The A. agallocha extract-chitosan edible films were prepared by using a different amount of A. agallocha extract (0, 1, 4, and 8 mL) and chitosan solution. Some physical properties of A. agallocha extract-chitosan edible films, water vapor permeability, solubility, swelling ratio, humidity ratio, thickness, scanning electron microscopy, and Fourier transform infrared spectroscopy analysis were investigated. The antibacterial activities, total phenolic content, and antioxidant capacity analysis of the A. agallocha extract-chitosan edible films were performed. The total phenolic content (0, 1, 4, and 8 mL of A. agallocha extract-chitosan edible films 0.92 ± 0.09, 1.34 ± 0.04, 2.94 ± 0.10, and 4.62 ± 0.10 mg gallic acid equivalent (GAE)/g film, respectively) and antioxidant capacity (0, 1, 4, and 8 mL of A. agallocha extract-chitosan edible films 52.61 ± 2.85, 104.28 ± 4.78, 304.30 ± 18.23, and 592.11 ± 0.67 mg Trolox equivalent (TE)/g film, respectively) of A. agallocha extract-chitosan edible films increased with the increase in the amount of extract. At the same time, the increase in the amount of antioxidant capacity improved the physical features of the films. The results of the antibacterial activity studies showed that all of the A. agallocha extract-chitosan edible films prevented bacterial growth from Escherichia coli and Staphylococcus aureus considering controlling group. PRACTICAL APPLICATIONS: To investigate the activity of antioxidant extract-biodegradable film, the A. agallocha extract-chitosan edible film had prepared. Introduction: Food industry generates large amounts of waste by-products rich in natural antioxidants. On the other hand, application of advanced processes for the recovery of these fine chemicals is another popular topic of recent years., Objective: The purpose of this study is to propose a green extraction method by application of deep eutectic solvent-based automated solvent extraction (AMSE) from lemon peels., Methods: The primary polyphenols (hesperidin, naringin, and p-coumaric acid) and the total polyphenols of the lemon peel extract were quantified and used as response for the optimisation of the AMSE conditions. The Box-Behnken design type of the response surface method (RSM) was chosen for optimisation study. Scavenging activity of the lemon peel extract against 2,2-diphenyl-1-picrylhydrazil (DPPH) free radical was also measured in vitro., Results: The optimum conditions for the highest total phenolic (7.47 mg-gallic acid equivalent [GAE]/g-lemon peel [LP]), naringin (5.05 mg/g-LP), p-coumaric acid (3.27 mg/g-LP), and hesperidin (0.07 mg/g-LP) yields were obtained by 1.5 h of extraction time, 46% water (v/v), and 5 g of peel. The antioxidant activity changed between 37.31% and 94.10% in the peels., Conclusions: Extraction time was the most effective process factor for the total phenolic and p-coumaric acid yields, while water addition was statistically very important (p < 0.0001) for the naringin and hesperidin yields in the current AMSE system. The second-order models generated for the selected systems represent the data satisfyingly based on the high coefficients of determination (> 0.99), statistically significant p-values (<0.0001), coefficient of variation values (< 10%), and non-significant lack-of-fit values (p > 0.05)., (© 2022 John Wiley & Sons, Ltd.)

Utilization of economical and environmental methods as an alternative process to recover the industrial crops and food products into high-added value compounds is of great significant. In the current study, microwave assisted extraction (MAE) was optimized by response surface methodology (RSM) in order to evaluate the waste of sour cherry as a source of phenolic compounds rich in anthocyanins. The process parameters (microwave power, irradiation time and ethanol solvent concentration) of MAE method were optimized by face centered composite design of RSM. Responses such as total phenolic componds (TPC), total anthocyanin (TA) contents and antioxidant capacity (DPPH) of extracts were measured spectrophotometrically after extractions of samples. The optimized result of MAE was 500 W of microwave power, 90 s of irradation time and 80% ethanol solvent concentration. Antioxidant capacity was tested using by 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical. Chromatographic analysis (HPLC) was also used to measure the concentration of major anthocyanin (cyanidin-3-glucoside) of the samples. Maximum predicted TPC, TA and DPPH yields on optimized conditions were 44.15 mg-GAE/g-FM (mg- gallic acid equivalent per g- fresh matter), 12.47 mg-cyanidin-3-glucoside/g-FM and 69.90 (%, inhibition), respectively. A stability assay under different conditions (light, dark, ambient condition, refrigerator and deep freezer) has been also performed in order to display the stability of bioactivity profile. All of the process parameters were significant at the level of p  < 0.0001.

Chestnut bee pollen has potential nutritional and medicinal effects and is an important natural bee product. This study focused on the investigation of the antioxidant capacity and DNA damage inhibition ability of chestnut bee pollen (CBP) from Bursa (Turkey). The phenolic compounds (rosmarinic acid, vitexin, hyperoside, pinocembrin, trans-chalcone, apigenin, protocatechuic, and galangin) and carotenoids in CBPE were determined by HPLC-DAD (high-performance liquid chromatography-diode array detection). Additionally, the protective ability of CBPE against DNA damage by oxidation was investigated. In this study, it was determined that CBPE has a high total phenolic compound content, and the antioxidant capacity of CBPE inhibits DNA oxidation (34% reduction of DNA damage in Fenton reaction media). This study could reveal new information regarding the use of CBPE as a protective agent for DNA in the future. PRACTICAL APPLICATIONS: Phenolic compounds and carotenoids prevent some diseases because of their important biological activities. One of the potential food sources chestnut bee pollen contains sugar, carbohydrates, amino acids, proteins, lipids, vitamins, hormones, enzymes, and flavonoids. Chestnut bee pollen, which has protective activity against DNA oxidation, could be an excellent potential source of a protective agent against some degenerative diseases through future applications., (© 2019 Wiley Periodicals, Inc.)

Sideritis montana L. endemic of Turkey was screened for its polyphenols content and antioxidant activity. Factor analysis and experimental design have been applied to understand the structure of the separation process, to determine the effective parameters, and to accomplish the performance improvement. Face-centred composite design (FCD) of response surface methodology (RSM) was applied to evaluate the influences of solvent concentration, solvent amount, extraction time, and stirring speed of homogenizer-assisted extraction (HAE) as well as to model and to optimize the HAE. Quadratic models were highly significant (p < 0.0001) for the responses studied with high coefficients of determination (R 2 ) of 0.9440, 0.9415 and 0.9521. The result of the study suggests that 15.02 mL of 22.69% EtOH solution (v/v), 70.16 s, and 9524.52 rpm of mixing speed are the optimal conditions to obtain the highest yield of total polyphenols (TPC) and flavonoids (TFC), and the best antioxidant activity (AA). Response surface methodology (RSM) and artificial neural networks (ANN) were evaluated and compared in order to decide which method was the most appropriate to predict and optimize total phenolic content (TPC) and oleuropein yields in olive tree leaf ( Olea europaea ) extracts, obtained after solvent-free microwave-assisted extraction (SFMAE). The SFMAE processing conditions were: microwave irradiation power 250-350 W, extraction time 2-3 min, and the amount of sample 5-10 g. Furthermore, the antioxidant and antimicrobial activities of the olive leaf extracts, obtained under optimal extraction conditions, were assessed by several in vitro assays. ANN had better prediction performance for TPC and oleuropein yields compared to RSM. The optimum extraction conditions to recover both TPC and oleuropein were: irradiation power 250 W, extraction time 2 min, and amount of sample 5 g, independent of the method used for prediction. Under these conditions, the maximal yield of oleuropein (0.060 ± 0.012 ppm) was obtained and the amount of TPC was 2.480 ± 0.060 ppm. Moreover, olive leaf extracts obtained under optimum SFMAE conditions showed antibacterial activity against S. aureus and S. epidermidis , with a minimum inhibitory concentration (MIC) value of 1.25 mg/mL.

A chromium reducing antioxidant capacity (CHROMAC) assay was presented to measure antioxidant capacity of selected plants and fruits and compared its performance with other commonly used antioxidant capacity methods of 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and cupric reducing antioxidant capacity (CUPRAC). The assay is based on the spectrophotometric measurement of colored a chelate complex of Cr(III) and diphenylcarbazone formed by the reaction of Cr(VI) and 1,5-diphenylcarbazid in acidic medium. Phenolic compounds react with excessive amounts of Cr(VI) at low pH values, causing reduction of Cr(VI) to Cr(III) and conversion of phenols to oxidized products. The assay comprises of the antioxidant with a chromium(VI) solution, a 1,5-diphenylcarbazid in acidic medium and subsequent measurement of the developed absorbance at 540 nm after 50 min. The color development is stable for phenolic compounds in plant and fruit. The selectivity of the assay for phenolic compounds was improved by adjusting pH to 2.8 and reduction potential between 0.2 and 0.9 V. The developed assay was successfully applied to the measurement of antioxidant capacity of three plants and one fruit (Prunus divaricata Ledeb.subsp. divaricata) samples and comparable results were obtained by ABTS and CUPRAC assays., (Copyright © 2013 Elsevier B.V.
