This study aims to determine the anti-aging effects of Achillea phrygia, an endemic plant, by evaluating its sun protection factor (SPF) level, antioxidant activity, total phenolic content, extracellular matrix-degrading enzymes (ECM) inhibition, genotoxic/anti-genotoxic, and cytotoxic activities. The SPF level was assessed using an in vitro quantitative method, while antioxidant capacity was determined through DPPH, β-carotene, and hydroxyl-radical (H2O2) scavenging assays. The total phenolic content was quantitatively conducted using the Folin Ciocalteu reagent. The inhibition of ECM-degrading enzymes was determined using matrix metalloproteinase-1 (MMP-1), hyaluronidase, and elastase enzymes. Genotoxic/anti-genotoxic properties were assessed using the AMES Salmonella/microsome assay, and cytotoxicity effects were assessed through the MTT assay. The results indicated that A. phrygia showed moderate SPF activity (SPF = 4.013) and exhibited IC50 values of 0.183 ± 0.03, 0.079 ± 0.51, and 1.18 ± 0.35 mg/mL for DPPH, β-carotene, and hydroxyl-radicals, respectively. The total phenolic content was measured to be 23.56 ± 1.42 mg GAE/g dry extract. Furthermore, the extract demonstrated inhibition of MMP-1 (47.98%) and elastase (39.2%) activities. Importantly, it did not induce DNA damage and showed antigenotoxic activity ranging from 10% to 65.6%. The cytotoxicity assay revealed an IC50 value of 42.41±4.05 µg/mL. These findings suggest that A. phrygia could be utilized as a cosmetic ingredient in skincare products due to its ability to protect against UV radiation, exhibit antioxidant properties, prevent extracellular matrix degradation, and inhibit DNA damage. Plants have been widely used as a pharmaceutical to treat diseases in many cultures for centuries. In vitro biological activities of some plants consumed as food and used in traditional medicine in Southwestern Anatolia have been reported. The present study focused on the antimicrobial, antibiofilm, antioxidant, and quorum sensing (QS) inhibitory activities of aqueous and ethanolic extracts of aerial parts of Scandix pectenveneris L., Artedia squamata L., Oenanthe pimpinelloides L. The antimicrobial activity was evaluated against fifteen bacteria and one yeast using disc diffusion and microdilution methods. The aqueous and ethanol extracts of A. squamata showed the highest activity against Staphylococcus aureus MU 47 with 20 and 23 mm zone diameters, respectively. The ethanol extract of S. pecten-veneris inhibited Staphylococcus aureus ATCC 25923 at a 5 mg/mL MIC value. The aqueous extract of O. pimpinelloides inhibited biofilm formation of S. aureus MU 40, P. aeruginosa ATCC 27853, and C. albicans ATCC 10239 at MIC value. All extracts displayed violacein inhibition against Chromobacterium violaceum CV12472 in the concentration range of 1.25 mg/mL to 25 mg/mL. The aqueous extract of A. squamata showed valuable anti-quorum sensing (anti-QS) activity with a pigment inhibition zone of 17 mm on Chromobacterium violaceum CV026. The aqueous extract of S. pecten-veneris inhibited the swarming motility of Pseudomonas aeruginosa PA01 by 47.6%. The aqueous and ethanol extracts of A. squamata indicated the best antioxidant activity. It was proved that these edible plant extracts could be used as natural alternative inhibitors to control microbial pathogenesis and diseases. [ABSTRACT FROM AUTHOR]

Background/aim: This study investigated the antioxidant and antimutagenic properties of Ankaferd blood stopper (ABS), a plant-based topical hemostatic agent used in Turkey to treat external hemorrhages and bleeding during dental surgery. While previous studies have examined the antimicrobial, antiinflammatory, and anticarcinogenic properties of ABS, to our knowledge, this is the first study to report on the antioxidant and antimutagenic activities of this drug. Materials and methods: Antioxidant activity was evaluated using DPPH radical-scavenging and β-carotene-linoleic acid tests. Antimutagenic activity was assessed using the Ames Salmonella/microsome mutagenicity test with the bacterial mutant strains Salmonella typhimurium TA98 and TA100. Results: Although ABS demonstrated no free-radical-scavenging activity in DPPH assays at the tested concentrations, β-carotenelinoleic acid testing found ABS to have a total antioxidant activity rate of 47.06 ± 4.41%. Antimutagenic effects were observed on TA100 at plate concentrations of 5%, 0.5%, and 0.05%, and on TA98 only at a plate concentration of 5%. Conclusion: ABS was shown to possess antioxidant and antimutagenic properties that could be of potential value in the fields of medicine and dentistry. [ABSTRACT FROM AUTHOR]
