Schistosomiasis is a public health problem, in many developing: countries including Egypt, Determination of the antigenic relationship between S mansoni and its intermediate snail host (IMH) Biomphalaria alexandrina can open a new field for diagnosis and control of the dis- ease. In the present study infected and non-infected B. alexandrina foot and visceral hump tissue as well as S. mansion crude Ag (SWAg) were fractionated using SDS-PAGE. It's specific and cross reacted protein fractions were determine using EITB versus experimentally prepared mice hyper immune sera (HIS) versus each antigen. After treatment of fractionated S.mansoni crude worm antigens (SWAg) versus HIS produced after vaccination of mice by the same Ag, 8 kda protein fractions ranged from 35-140 kda were reacted specifically. Treatment of fractionated B.alexandrina infected and non-infected foot and visceral hump Ag versus previous HIS revealed presence of common polypeptides bands between SWAg and non-infected snail antigens. The fraction at 135 kda, 68 kda, were detected in all cases, while that at 40-42 kda and that at 35 kda was diagnosed in SWAg and that of infected snails only. The fraction at 68 kda was reacted specifically between SWAg and all tested fractionated snail antigens either that of foot or visceral hump when they treated separately by HIS of mice vaccinated by each snail A eparately. The fraction at 135 kda was common between SWAg and snail (infected and non-infected) visceral hump antigen. The fraction at the level of 110 kda was diagnosed inSWAg, in non-infected foot antigen and visceral hump Ag. The fraction at the level of 46-48 (da are common between SWAg and snail foot and visceral hump Ag after treated by HIS of mice vaccinated by foot Ag, Presence of common antigenic fractions between snail tissues and Schistosoma species can prefer an easily source of antigen valuable for diaguosis or vaccination as well as can be considered as new tool for determination to the snail IMH of new discovered trematode Darasites.

It can be safely said that up till now, no method of vaccination (including recent genes encoding vaccines) has yet proved to be totally effective since they gave partial and low levels of protection against S. mansoni infection. The objective of this work is to try testing the immunogenic effect of two purified non infected B. alexandrina hepatopancreas through histopathological changes in liver of Swiss albino mice (15-20 gm). Gel filtration chromatography was used to fractionate the crude antigen into five fractions followed by re-fractionation and determination of their molecular weights by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). Four dilutions of Fiv (20000-29000 daltons) and Fv (20000-24000 daltons) were injected in two groups of mice (33 each) at weekly intervals and another control group was injected with phosphate buffer saline (PBS) in the same manner. Sacrification was done seven weeks from infection with 100 S. mansoni cercariae through immersion method. The results revealed that there is marked histopathological changes in liver of the control group in comparison to the two vaccinated groups which appear more or less normal with slight inflammatory infiltrate.

Current stratigies for the control of schistosomiasis are based primarily on chemotherapy but successful vaccination against infection has been also demonstrated in several host parasite models. In this study, the immunogenic effect of two purified antigens (Fiv 20-29 000 daltons & Fv 20-24 000 daltons) extracted from non infected hepatopancreas of B. alexandrina as measured by worm load, state of copulation and viability of ova in tissues. The antigens were prepared using gel filtration chromatography and their molecular weights were estimated through sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). Four dilutions from each antigen were prepared and injected in two groups (33 each) of Swiss albino mice at weekly intervals. A control group was injected with phosphate buffer saline (PBS) in the same manner. All mice were injected with 100 cercariae using immersion method. Sacrification was done regularly after 7 weeks of injection. On the basis of worm load, Fiv gave protection of 44% while Fv gave only 36%. The number of worms in copula was significantly reduced and they became delicate, fragile, stunted and malformed. Both antigens gave a significant reduction in total viable eggs in tissues.

Rabbit antisera, raised against whole, homogenized hepatopancreas from uninfected Biomphalaria glabrata, were used to screen a cDNA library prepared from adult Schistosoma mansoni. Of 1.8 x 10(5) recombinants screened, 34 clones were specifically immunoreactive with the antisera. Twenty of the clones were subcloned for further analysis, and 13 of the 20 were demonstrated to hybridize to a cDNA probe encoding schistosome tropomyosin I (pSMTM) at high stringency (Xu et al. Experimental Parasitology 69, 373-392, 1989). One partial cDNA clone (pSM4), which failed to hybridize to pSMTM, was used to rescreen the adult worm cDNA library and a 1381-bp cDNA clone, SmTMII, was identified and characterized. SmTMII contained a single 284-amino acid open reading frame and was shown to be 65.8% homologous to SMTM. Computer-assisted analysis of the predicted protein structure depicted a hydrophilic molecule with the extensive alpha helical secondary structure characteristic of tropomyosins. SmTMII was expressed in Escherichia coli and the resulting 45-kDa fusion protein was recognized by both a polyclonal antiserum specific for SMTM and the antiserum of mice chronically infected with S. mansoni. Northern analysis showed the SmTMII mRNA in the adult stage to be about 1.6 kb in size. Analysis of the SmTMII gene by Southern blot revealed a complex hybridization pattern suggesting the presence of introns or multiple gene copies. The 650-bp EcoRI fragment of SmTMII was used to screen a cDNA library prepared from uninfected Biomphalaria glabrata and two clones, each encoding a novel snail tropomyosin, BgTMII, were isolated and characterized. The two clones, consisting of 2816 and 2314 bp, respectively, differed from each other only in the polyadenylation signal used and each contained a single and identical 284-amino acid open reading frame having 94.7% homology with the previously reported snail tropomyosin, BG39 (Dissous et al. Molecular and Biochemical Parasitology 43, 245-256, 1990). BgTMII was shown to have a higher amino acid homology with SMTM (68.3%) than with BG39 (65.1%) and a higher amino acid homology with SMTM than the two schistosome tropomyosins (SMTM and SmTMII) have with each other (65.8%). Implications of these observations in relation to the concept of host-parasite antigen mimicry are discussed.

Mice and rabbits were immunized with antigens derived from Bge cells, Biomphalaria glabrata hemolymph, or Schistosoma mansoni. Antisera from mice given molluscan antigens did not form immunoprecipitates with soluble antigen from adult worms, but their binding to surfaces of sporocysts, cercariae, and schistosomules suggests the presence of cross-reacting determinants. In vitro, cell-mediated immune responses to Bge antigens were not demonstrable in infected nor in immunized mice. Mice immunized with Bge cell-line antigens and challenged with S. mansoni cercariae showed no reduction in worm burden when compared with control mice.

An antigen was isolated and partially characterized from adult Schistosoma mansoni which immunologically cross-reacted with Fasciola hepatica and Biomphalaria glabrata, a schistosome intermediate snail host. The antigen was isolated from a solubilized freeze-thaw preparation containing 0.5% Triton X-100 by passage through a CNBr-activated Sepharose 4B column coupled with rabbit IgG prepared against a homogenate of B. glabrata hepatopancreas. The eluted antigen (designated as SMw-53P) stained with Coomassie Brilliant Blue R250, but not with Nile Blue A or periodic acid-Schiff's stains. The antigen did not bind to a Concanavalin A affinity column. SMw-53P was determined to have a molecular weight of 53,000 daltons by SDS-polyacrylamide gel electrophoresis. Western blotting, using sera from mice infected with S. mansoni, revealed the presence of the antigen in whole worm preparations of both S. mansoni and F. hepatica. The serodiagnostic potential of the antigen was evaluated by ELISA utilizing sera from S. mansoni-infected mice, or rabbits injected with homogenates of F. hepatica or S. mansoni whole worm, or B. glabrata hepatopancreas. SMw-53P was shown to strongly react with all antisera, but not normal mouse or rabbit sera. These data suggest a limited value for the antigen for the specific immunodiagnosis of schistosomiasis mansoni, but do suggest a possible potential as a general screening tool for detecting trematode infections. Further studies regarding the antigen's potential as a vaccine are indicated.

Serum from humans infected with Schistosoma mansoni, when reacted with a Biomphalaria glabrata soluble hepatopancreas antigen extract (BgSHA) yields 2 lines of precipitation by gel diffusion and 1 by immunoelectrophoresis. The IgG from the serum of a human infected with S. mansoni was coupled to CNBr-activated Sepharose 4B. BgSHA (8.0 mg) was then filtered through the gel and the bound antigens, denoted BgSm, eluted with HCl-glycine, pH 2.6. These bound antigens comprised 2.8% of the total BgSHA. BgSm was then applied to an anti-Fasciola hepatica column as above. The drop through in PBS (38% yield) and containing the BgSm antigens depleted of cross-reactivity with F. hepatica was then tested by the ELISA to evaluate its serodiagnostic potential. These antigens detected a primary S. mansoni infection by 4 wk but were less sensitive than SmSEA in the detection of a primary infection with S. mansoni. However, the BgSm-specific antigens were more specific than SmSEA and showed less cross-reactivity with the serum of mice infected with F. hepatica. At least 16 peptides were seen by silver staining following SDS-PAGE with 5-20% gradient gels. The 2 more prominent bands obtained were estimated to have molecular weights of 62 and 66 kd. Nitrocellulose strips blotted with BgSHA were incubated with the serum of mice infected with S. mansoni for 12 wk and developed 6 bands with molecular weights of 66, 57, 55, 50, 48 and 32 kd.(ABSTRACT TRUNCATED AT 250 WORDS)

Mice were immunized against hepatopancreas homogenate of Biomphalaria alexandrina, using Al(OH)3 as adjuvant, and challenged by subcutaneous (s.c.) injection of Schistosoma mansoni cercariae, percutaneous invasion of cercariae and intravenous (i.v.) injection of mechanically prepared schistosomula of different ages. Significantly reduced (about 50%) numbers of adult S. mansoni were found 42 days after challenge in immunized animals infected by s.c. injection of cercariae. Protective effects were detectable as early as 4 days after infection when the numbers of schistosomules in the lungs were reduced in immunized animals. However, immunized animals lacked protective immunity against S. mansoni when infected via the percutaneous route with cercariae or by i.v. injection of schistosomules of different ages. Experiments show that immunization of hosts against intermediate host antigens does not protect against S. mansoni infection via the natural route.
