For parasites that sequester themselves within a vacuole, new rules governing antigen presentation are coming into focus. Components of the host's autophagy machinery and the parasite's membranous nanotubular network within the parasitophorous vacuole play a major role in determining antigenicity of Toxoplasma proteins. As such, both parasite and vaccinologist may exploit these pathways to regulate the ever important CD8 T cell response to apicomplexan parasites., (Copyright © 2015. Published by Elsevier Ltd.)

The indirect fluorescent antibody test was applied for detection of circulating antibodies in sheep as a result of Theileria hirci infections. A schizont antigen was prepared from an in vitro culture suspension of lymphoid cells infected with T. hirci macroschizonts. The peak antibody titre of 1/8, 192 was reached 24 days after the initial antibody rise in the sheep experimentally infected by means of Hyalomma anatolicum anatolicum ticks.

The method of counterimmunoelectrophoresis was used to detect rapidly soluble antigens and antibodies in the sera of chickens infected with Leucocytozoon caulleryi. This method retained the specificity revealed by the Ouchterlony gel-diffusion technique, and induced no false positive reactions. Therefore, it was applicable to the serological diagnosis of chicken leucocytozoonosis, as well as the Ouchterlony technique which is used in Japan at present.

Sera from Theileria parva infected, recovered and rechallenged cattle were tested in complement-dependent cytotoxicity, membrane immunofluorescence and antibody-dependent cellular cytotoxicity assays for the presence of antibodies against cell membrane antigens of T. parva transformed cell lines. In the complement-dependent antibody-mediated cytotoxicity assay, sera from lethally infected animals were negative. Some recovered cattle showed a positive reaction, but such reactions were also observed when an eland cell line infected with T. taurotragi, and bovine lymphoblastoid cells were used as targets. Reaction was less against Ig-negative peripheral blood lymphocytes. Evidence is presented that these reactions could be evoked by attachment of immune complexes to Fc-receptors. It is concluded that cattle exposed to T. parva infection do not develop antibodies against specific T. parva (or T. parva-induced) cell surface antigens.

Bovine mononuclear cells in the presence of bovine anti-chicken erythrocyte sera at high dilutions induce release of chromium-51 from labeled chicken erythrocytes. Bovine effector cells are capable of recognizing both bovine immunoglobulin G(1) and bovine immunoglobulin G(2); in contrast, human effector cells only recognize immunoglobulin G(1). Effector cell activity of bovine mononuclear cells is equally distributed between peripheral blood and spleen. As in other species, thymus and lymph node cells exert no antibody-dependent effect, although some direct cytotoxicity by lymph node cells may be observed. Antibody-dependent cell-mediated cytotoxicity against a bovine cell line can also be detected. By using a tannic acid technique, it was found that chicken erythrocytes coated with Theileria parva piroplasm antigen or with Trypanosoma rhodesiense variant-specific coat antigen form suitable targets for bovine antibody-dependent cell-mediated cytotoxicity assays. By using such targets, a moderate degree of direct cytotoxicity by bovine mononuclear cells, in the absence of antibody, is always observed; this may be reduced by choosing optimal conditions of tannic acid treatment and antigen sensitization and by the use of short incubation periods for the cytotoxicity assay. Observations have been made on the variant specificity, time course of appearance, and association with immunoglobulin G(1) of the antibody activity responsible for cell-dependent cytotoxicity against chicken erythrocytes coated with T. rhodesiense antigens. The potential usefulness of this technique in the analysis of protective immune responses against protozoal infections is discussed.

A schizont antigen for the indirect fluorescent antibody test against Theileria parva was prepared from a T parva-infected bovine lymphoblastoid cell line by fixing the cells in suspension with a mixture of acetone and formaldehyde. The antigen was stored in suspension in phosphate buffered saline for one and a half years at -60 degrees C without loss of activity; the antigen could also be lyophilised. The fluorescence of the intracellular schizonts was bright and specific with T parva positive bovine control serum and absent with negative bovine control serum and Theileria mutans positive bovine control serum. Fluorescence of the lymphoblastoid cell itself was observed with Trypanosoma brucei positive control serum and some bovine test sera: this fluorescence, which masked the intracellular schizonts, was eliminated by absorbing the sera in the supernatant of sonicated lymphocytes obtained from bovine lymph nodes. The antigen was evaluated with sera from cattle experimentally infected with T parva. In an epizootiological study on East Coast fever in the Coast Province of Kenya, there was good correlation between the serological responses of cattle to T parva schizont antigen and the distribution of Rhipicephalus appendiculatus ticks.

The proteins of purified macroschizonts from Theileria parva, T. lawrencei, and T. taurotragi were analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis. The major proteins of all species had molecular weights of 120,000, 70,000, 65,000, 62,000, 55,000, 44,000, and 35,000. All further experiments were carried out with T. parva. Using 125I surface labelling it was established that proteins with molecular weights of 70,000, 50,000, and 44,000 were membrane constituents. Staphylococcus aureus protein A-mediated immune precipitation studies with 125I-labelled lysates of macroschizonts and a rabbit anti-macroschizont serum specifically recognized proteins with molecular weights of 120,000, 91,000, 70,000, 62,000, and 35,000. A small proportion of sera recovered from Theileria immune cattle specifically recognized proteins with molecular weights of 180,000 and 70,000 in macroschizont-lysates.

Proteosomes are hydrophobic, membranous, multimolecular preparations of meningococcal outer membrane proteins that are also B cell mitogens. These characteristics suggested that proteosomes may serve as carrier proteins and adjuvants to enhance peptide immunogenicity. Although high titers of malaria circumsporozoite (CS) antibodies protect against malaria, vaccines thus far tested in humans have been insufficiently immunogenic to be clinically useful. Here it is shown that synthetic CS peptides hydrophobically complexed to proteosomes by way of lauroyl-cysteine become highly immunogenic in mice without other adjuvants. The high titers of antibodies produced and the safety of proteosomes in humans suggest that this novel system is widely applicable for the development of peptide vaccines to protect against many diseases.
