Biocomposites can be a solution for environmental pollution and sustainability by acting as an alternative to petroleum-based products used in the field of packaging and biomedical applications. Herein, an attempt to valorize the potential of Prosopis juliflora plant to fabricate Polylactic acid (PLA) based biocomposites. The biocomposites comprised of Prosopis juliflora bark powder (PJBP) and PLA were fabricated by solution casting method with various weight percentages (Wt%) of PJBP. PJBP/PLA films show uniform functional group interactions through FTIR analysis. The 25 % PJBP/ PLA biocomposite shows the highest crystalline size of 8.9 nm. The 25 wt% included PJBP increases the thermal stability by 6.9 %, surface roughness by 212 %, and tensile strength by 45.5 % compared to PLA film. The 25 % PJBP film shows increment in the water absorption and water vapor permeability test. In addition, the antibacterial behavior of 25 % PJBP bio composite shows higher inhibition zones against the gram-positive and gram-negative pathogens. Biocomposite films with PJBP content show 40 % degradation in soil for degradation analysis. Rheumatoid arthritis, a chronic autoimmune disorder characterized by joint inflammation, is thought to be exacerbated by bacterial infections, notably Proteus mirabilis. This study explores the combined effects of quercetin, a potent antioxidant and anti-inflammatory flavonoid, and chondroitin sulfate, known for its cartilage-protective properties, as a potential therapeutic approach. Molecular docking analyses revealed favourable interactions between these compounds and key pro-inflammatory cytokines IL-6 and TNF-α, suggesting their potential to disrupt inflammation-related signaling pathways. In vitro assays demonstrated that the quercetin-chondroitin sulfate combination (1:1 ratio) significantly inhibited oxidative stress and hemolysis, highlighting its enhanced anti-inflammatory and membrane-protective effects. The free radical scavenging assays further confirmed the antioxidant potential of this combination, which demonstrated strong radical scavenging activity. Antimicrobial assays showed notable antibacterial effects, with an increased inhibition zone against P. mirabilis when quercetin and chondroitin sulfate were combined, suggesting a synergistic antimicrobial action. In vivo, zebrafish subjected to bacterial stress showed improved survival rates with the quercetin and chondroitin sulfate combination treatment, along with enhanced mineralization and significant modulation of alkaline phosphatase (ALP) and tartrate-resistant acid phosphatase (TRAP) activities, indicating its protective role in maintaining joint health. Furthermore, gene expression analysis revealed a substantial reduction in pro-inflammatory markers, including TNF-α and IL-6, demonstrating the quercetin and chondroitin sulfate combination's ability to mitigate inflammation. Epinecidin-1 (epi-1), an antimicrobial peptide first identified in marine grouper fish, has multifunctional bioactivities. The present study aims to improve its therapeutic potential via structural modifications that could enhance its antimicrobial activity and stability. To achieve it, we replaced glycine and the first histidine in the parent epi-1 with lysine, which resulted in a peptide with a repeating KXXK motif and improved physiochemical properties related to antimicrobial activity. This modified peptide, referred to as glycine-to-lysine replaced-epi-1, also gained stability and a twofold increase in helical propensity. To produce the active peptide, overlap extension PCR was employed to generate the gene of GK-epi-1 via site-directed mutagenesis, which was then cloned into the pET-32a vector and expressed as a recombinant fusion protein in Escherichia coli C43 (DE3) strain. The recombinant protein was purified and digested with enterokinase to release the active peptide fragment, which was then evaluated for antimicrobial activity and stability. Antimicrobial resistance (AMR) is a growing public health threat caused by the widespread overuse of antibiotics. Bacteria with antibiotic resistance may acquire resistance genes from soil or water. Endogenous hydrogen sulfide (H 2 S) production in bacteria confers antibiotic tolerance in many, suggesting a universal defense mechanism against antibiotics. In this study, we isolated and identified soil-based antibiotic-resistant bacteria collected from contaminated areas. An antibiotic-resistant bacterium was identified as non-endogenous-H 2 S-producing, allowing us to examine the effect of exogenous H 2 S on its resistance mechanism. Therefore, we demonstrated that different classes of antibiotic resistance can be reverted by employing H 2 S with antibiotics like ampicillin and gentamicin. Methods like Kirby-Bauer Disk-Diffusion, Scanning Electron Microscopy, and Flow Cytometer analysis were performed to assess the antibacterial activity of H 2 S with ampicillin and gentamicin. The antioxidative efficiency of H 2 S was evaluated using the DCFH-DA (ROS) test, as well as lipid peroxidation, and LDH activity. These were further confirmed with enzymatic and non-enzymatic (SOD, CAT, GST, and GSH) antioxidant studies. These findings support H 2 S as an antibiotic-potentiator, causing bacterial membrane damage, oxidative stress, and disrupting DNA and proteins. The present study aimed to determine the presence of Klebsiella pneumoniae and Escherichia coli with extended-spectrum β-lactamase (ESBL)s property from treated wastewater effluents. Treated effluent samples were collected from two major water treatment plants which located at Avaniyapuram and Sakkimangalam, Madurai, Tamil Nadu, India. Among the 51 isolates, 56.86 % represented E. coli (18 from Avaniyapuram and 11 from Sakkimangalam) and 43.14 % were K. pneumoniae (7 from Avaniyapuram and 15 from Sakkimangalam). Based on the ESBL propensity, E. coli was overrepresented in the present study. All the isolates turned positive for ESBL, while 5.88 % of K. pneumoniae and 7.84 % of E. coli were positive for carbapenemases. Further, K. pneumoniae isolates from both sites showed 100 % resistance to beta-lactams, with resistance to other antibiotics such as tetracycline and meropenem. E. coli isolates were 100 % resistant to ceftazidime and cefuroxime, and 88.9 % were resistant to amoxicillin/clavulanate and ceftriaxone. The MAR indices observed in the present study for E. coli and K. pneumoniae were above the threshold value of 0.2 suggested a high risk of environmental contamination. Biopolymers infused with bimetallic nanoparticles exhibit a wide range of functionalities necessary for efficiently eliminating diverse water contaminants. However, the protracted production process requires further exploration. As such, present study seeks to optimize microwave-assisted technique for the facile synthesis of cross-linked chitosan (CTS) supported bimetallic-oxide nanoparticles, specifically zinc oxide (ZnO) and iron-oxide (Fe 3 O 4 ), denoted as CTS-TTP/Zn-Fe. The primary objective is to investigate the efficacy of these beads in the removal of Paracetamol (PCM) from single and complex water matrices while also assessing their antibacterial properties. Characterization includes chemical composition, surface structures, thermal stability, and magnetic properties. The experimental results demonstrated that CTS-TPP/Zn-Fe beads achieved a remarkable PCM removal efficiency of ~99 % (q m  = 4.98 mg g -1 ), with a Zn:Fe mole ratio of 1:1. The experimental data showed good applicability with Freundlich isotherm and chemisorption-supported rate models (R 2  > 0.9). To evaluate the long-term viability and practicality of these beads, three crucial field applicability tests were conducted. These encompassed competition studies with other pharmaceuticals, desorption investigations for repeated use, and efficiency evaluations in an ionic solution. Foodborne pathogens continue to challenge public health due to their ability to cause severe illness and their increasing resistance to current antimicrobial treatments. Listeria monocytogenes is a resilient foodborne pathogen that poses significant risks to vulnerable populations, leading to severe infections and high hospitalization rates. The emergence of antimicrobial-resistant (AMR) strains of L. monocytogenes underscores the need for novel therapeutic strategies. In this study, we investigated the antimicrobial efficacy of the (2E)-3-(3,5-dibromo-2-hydroxylphenyl)-1-(5-methylfuran-2-yl) prop-2-en-1-one (DK06) against multidrug-resistant L. monocytogenes. DK06 exhibited a significant dose-dependent inhibition of L. monocytogenes growth, achieving a maximum inhibition of 92.9 % at 320 μM. Molecular docking and dynamics simulations revealed high binding affinities for key virulence proteins PlcB and ArgA, with stable protein-ligand interactions. DK06 also disrupted biofilm formation at sub-MIC levels, reducing extracellular polymeric substances (EPS) and biofilm mass, as observed by scanning electron microscopy (SEM) analysis. Furthermore, DK06 downregulated the expression of virulence genes (plcB, argA, and hly) and decreased hemolytic activity. In vivo zebrafish studies confirmed the safety of DK06 up to 80 μM, demonstrating its efficacy in reducing mortality and oxidative stress associated with L. monocytogenes infection. DK06 also attenuated inflammation by downregulating key inflammatory markers (tnfa, il1b, il6, and nfkb). Background: Wound healing pivots on a finely orchestrated inflammatory cascade, critical for tissue repair. Chronic wounds, compounded by persistent inflammation and susceptibility to infection, pose formidable clinical challenges. Nanofiber dressings offer promising avenues for wound care, yet their interaction with inflammation and infection remains elusive. We aim to delineate the inflammatory cascade preceding wound closure and assess Cu@Bbc nanofibers' therapeutic efficacy in mitigating inflammation and combating infection. Their unique attributes suggest promise in modulating inflammation, fostering tissue regeneration, and preventing microbial colonization. Investigating the intricate interplay between nanofiber scaffolds, inflammation, and infection may unveil mechanisms of enhanced wound healing. Our findings could stimulate the development of tailored dressings, urgently needed for effective wound management amidst immune dysregulation, infection, and inflammation., Methods: In this investigation, we synthesized Cu@Bbc nanofibers, incorporating curcumin and berberine chloride, for wound healing applications. We evaluated their individual and combined antibacterial, anti-biofilm, and antioxidant activities, alongside binding affinity with pro-inflammatory cytokines through molecular docking. Morphological characterization was conducted via SEM, FTIR assessed functional groups, and wettability contact angle measured hydrophobic properties. The physical properties, including tensile strength, swelling behavior, and thermal stability, were evaluated using tensile testing, saline immersion method and thermogravimetric analysis. Biodegradability of the nanofibers was assessed through a soil burial test. Biocompatibility was determined via MTT assay, while wound healing efficacy was assessed with in vitro scratch assays. Controlled drug release and antibacterial activity against MRSA were examined, with in vivo assessment in a zebrafish model elucidating inflammatory responses and tissue remodeling., Results: In this study, the synergistic action of curcumin and berberine chloride exhibited potent antibacterial efficacy against MRSA, with significant anti-mature biofilm disruption. Additionally, the combination demonstrated heightened antioxidant potential. Molecular docking studies revealed strong binding affinity with pro-inflammatory cytokines, suggesting a role in expediting the inflammatory response crucial for wound healing. Morphological analysis confirmed nanofiber quality, with drug presence verified via FTIR spectroscopy. Cu@Bbc demonstrated higher tensile strength, optimal swelling behavior, and robust thermal stability as evaluated through tensile testing and thermogravimetric analysis. Additionally, the Cu@Bbc nanofiber showed enhanced biodegradability, as confirmed by the soil burial test. Biocompatibility assessments showed favorable compatibility, while in vitro studies demonstrated potent antibacterial activity. High levels of antibiotic accumulation and the difficulty of degradation can have serious consequences for the environment and, therefore, require urgent attention. To solve this problem, a synergistic Er and Cd ion-codoped Bi 4 O 5 Br 2 photocatalyst was proposed. The degradation rate of sulfamethoxazole (SMX) by Er/Cd-Bi 4 O 5 Br 2 was eight times higher than that of pure Bi 4 O 5 Br 2 , exceeding that of single Er-doped or Cd-doped Bi 4 O 5 Br 2 , which was attributed to the ability of Er/Cd-Bi 4 O 5 Br 2 to generate a variety of free radicals. Experimental results and theoretical calculations suggested a possible mechanism for the improved photocatalytic degradation rate. The reduction of the band gap can facilitate the production of electron-hole pairs, which play a significant role in the production of reactive radicals. Furthermore, an optimal stabilized structure of the ErCd-Bi 4 O 5 Br 2 dopant system was identified based on the formation energy formulas of different ligand configurations. These findings offer promising potential for the degradation of broad-spectrum antibiotics and provide valuable insights for the design and modification of photocatalytic materials.

Scenedesmus strains have been reported to have the potential to tolerate and bioremediate antibiotic pollutants through bioadsorption, bioaccumulation, and biodegradation mechanism from the wastewater medium. Hormesis effects have been observed in the Scenedesmus strains when exposed to different concentrations of antibiotic pollutants. Lower concentrations of antibiotic pollutants are known to trigger growth-stimulating effects by triggering adaptive responses such as increased metabolic activity and activating detoxifying mechanisms leading to the biotransformation pathway. The present review examines the existing body of information pertaining to biotransformation pathways tolerance, hormesis effects, and efficiency of Scenedesmus strains in removing various antibiotic pollutants. The focus of current advances in nanotechnology has shifted significantly towards environmentally conscious methods that use harmless ingredients and moderated reaction circumstances to promote equitable development. Zinc oxide nanoparticles (NPs) currently grabbed attention of multiple medical fields owing to their unique ability to safeguard against cellular damage and alleviate serious human diseases via processes related to metabolism. This work focused on the generation of ZnO NPs using the peel of Cucumis melo fruit. The NPs were then analyzed and characterized using UV-Vis spectroscopy. The results indicated that at a wavelength of 352 nm, it was proven that the biosynthesis of ZnO NPs had occurred. The XRD pattern indicated the presence of dense crystal structures. The field emission scanning electron microscope (FE-SEM) picture confirmed the existence of polygonal-shaped ZnO NPs. The findings indicate that the produced ZnO NPs possess tough antibacterial properties against Gram-positive and Gram-negative microorganisms. When the ZnO NPs were exposed to direct sunshine for 80 min, they showed an 89% dye breakdown efficiency. This research specifically focused on the decomposition of reactivity dyes, with methylene blue dye being used as the target dye. In this study, mycosynthesized zinc oxide nanoparticles (ZnONPs) are fabricated via Pleurotus sajor-caju mushroom extract, and their potential medical and environmental applications are demonstrated. The biosynthesized ZnONPs were assessed for their antibacterial, anticancer, and biodecolorization potential efficiency. They were also characterized and morphologically analyzed by UV-visible spectroscopy, XRD, FT-IR, FE-SEM, EDX, HR-TEM, Zeta potential, and GC-MS analysis. The UV visible spectrum analysis of synthesized ZnONPs analyzed outcome 354 nm was the SPR peak that the nanoparticles displayed. The characteristic Zn-O bond was indicated by a strong peak in the FT-IR study at 432.05 cm -1 . Based on XRD analysis, P. sajor-caju mediated ZnONPs were crystalline nature, with an average nano particle size of 14.21 nm and a polydispersity directory of 0.29. The nanoparticles exhibit modest constancy, as shown by their zeta potential value of - 33.2 mV. The presence of oxygen and zinc was verified by EDX analysis. The ZnONPs were found to be spherical in shape and crystalline nature structure, with smooth surface morphology and a mean particle size of 10 nm using HR-TEM and SAED analysis. The significant antibacterial activity against S. aureus (6.2 ± 0.1), S. mutans (5.4 ± 0.4), and B. subtilis (5.2 ± 0.1 mm) was demonstrated by the synthesized ZnONPs made using mushroom extract. It was discovered that when the concentration of mushroom extract was increased together with synthesized ZnONPs, the bactericidal activity increased considerably. A higher concentration of ZnONPs demonstrated superior antibacterial activity across the ZnONPs ratio tests. The in vitro cytotoxicity assay showed that ZnONPs, even at low doses, had a substantial number of cytotoxic effects on liver cancer cells (LC 50 values 47.42 µg/mL). The effectiveness test revealed that acid blue 129 was degraded. The best decolorization of acid blue 129 at 72.57% after 3 h of soaking serves as evidence for the theory that myco-synthesized ZnONPs by P. sajor-caju mushroom can function as catalysts in reducing the dye. The mycosynthesized ZnONPs from P. sajor-caju extract, and its potential for antibacterial, anticancer, and decolorization are in this investigation. The direct-linked coumarin-benzimidazole hybrids, featuring aryl and n-butyl substituents at the N1-position of benzimidazole were synthesized through a Knoevenagel condensation reaction. This reaction involved the condensation of 1,2-diaminobenzene derivatives with coumarin-3-carboxylic acids in the presence of polyphosphoric acid (PPA) at 154 °C. The in vitro antibacterial potency of the hybrid molecules against different gram-positive and gram-negative bacterial strains led to the identification of the hybrids 6m and 6p with a MIC value of 6.25 μg/mL against a gram-negative bacterium, Klebsiella pneumonia ATCC 27736. Cell viability studies on THP-1 cells demonstrated that the compounds 6m and 6p were non-toxic at a concentration of 50 µM. Furthermore, in vivo efficacy studies using a murine neutropenic thigh infection model revealed that both compounds significantly reduced bacterial (Klebsiella pneumonia ATCC 27736) counts (more than 2 log) compared to the control group. Additionally, both compounds exhibited favorable physicochemical properties and drug-likeness characteristics. Oxytetracycline Hydrochloride (OTC), a common antibiotic used to treat specific illnesses in humans and animals, is characterized by poor absorption into cells, low volatility, and high hydrophilicity. It is a potent contaminant that poses a serious threat to the ecosystem, particularly the aquatic sources. Adsorption onto natural adsorbents is one of the most successful, economical, and ecologically friendly ways to remove antibiotics from waste water. The present work focuses on the adsorption of OTC utilizing alginate biochar beads (AlBCB) and biochar powder (BC) derived from bagasse. The influence of several factors were studies and optimized through batch studies employing BC and AlBCB. After 50 min BC displayed a removal of 97%, at an initial concentration of 10 ppm. The experimental data was discovered to follow PFO kinetics and fit with the Freundlich isotherm adsorption model. AlBCB, after a contact time of 40 min, indicated a maximum percentage removal of 86% for initial concentration of 10 ppm OTC. Al-biochar beads showed the maximum percentage removal at pH 10. 0.5 g of adsorbent was used to carry out all batch experiments at room temperature. Arctium lappa (Burdock) root is used in various culinary applications especially in Asian Cuisine. Arctigenin (ARC) is a polyphenolic compound abundant in the roots of the burdock plant from which it derives its name. The emergence of bacterial resistance is a growing global worry, specifically due to the declining availability of new antibiotics. Screening for the antibacterial candidates among the safe natural products is a promising approach. The present study was aimed to assess the antibacterial activity of ARC against Pseudomonas aeruginosa exploring its effect on the bacterial cell membrane. Furthermore, the anti-virulence activities and anti-quorum sensing (QS) activities of ARC were in vitro, in vivo and in silico assessed against P. aeruginosa . The current results showed the ARC antibacterial activity was owed to its disruption effect of the cell membrane. ARC at sub-MIC significantly decreased the formation of biofilm, motility, production of extracellular enzymes and in vivo protected mice against P. aeruginosa . These anti-virulence activities of ARC are owed to its interference with bacterial QS and its expression. Furthermore, ARC showed mild effect on mammalian erythrocytes, low probability to induce resistance and synergistically combined with antibiotics. In summary, the promising anti-virulence properties of ARC indicate its potential as an effective supplement to conventional antibiotics for treating severe P. aeruginosa infections.

Shigella flexneri is a gram-negative bacterium responsible for shigellosis and bacterial dysentery. Despite using various synthetic antimicrobial agents and antibiotics, their efficacy is limited, prompting concerns over antibiotic resistance and associated health risks. This study investigated eugenol, a polyphenol with inherent antioxidant and antibacterial properties, as a potential alternative treatment. We aimed to evaluate eugenol's antibacterial effects and mechanisms of action against S. flexneri and its impact on biofilm formation. We observed significant growth suppression of S. flexneri with eugenol concentrations of 8-10 mM (98.29%). Quantitative analysis using the Crystal Violet assay demonstrated a marked reduction in biofilm formation at 10 mM (97.01 %). Assessment of Cell Viability and morphology via Fluorescence-Activated Cell Sorting and Scanning Electron Microscopy confirmed these findings. Additionally, qPCR analysis revealed the downregulation of key genes responsible for adhesion (yebL), quorum sensing (rcsC, sdiA), and EPS production (s0482) associated with bacterial growth and biofilm formation. Bone tissue engineering offers a promising alternative to stimulate the regeneration of damaged tissue, overcoming the limitations of conventional autografts and allografts. Recently, titanium alloy (Ti) implants have garnered significant attention for treating critical-sized bone defects, especially with the advancement of 3D printing technology. Although Ti alloys have impressive versatility, their lack of cellular adhesion, osteogenic and antibacterial properties are significant factors that contribute to their failure. Hence, to overcome these obstacles, this study aimed to incorporate osteoinductive and antibacterial cue-loaded hydrogels into 3D-printed Ti (3D-Ti) scaffolds. 3D-Ti scaffolds were synthesized using the direct metal laser sintering method and loaded with a gelatin (Gel) hydrogel containing strontium-doped silver nanoparticles (Sr-Ag NPs). Compared with Ag NPs, Sr-doped Ag NPs increased the expression of Runx2 mRNA, which is a key bone transcription factor. We subjected the bioactive 3D-hybrid scaffolds (3D-Ti/Gel/Sr-Ag NPs) to physicochemical and material characterization, followed by cytocompatibility and osteogenic evaluation. The microporous and macroporous topographies of the scaffolds with Sr-Ag NPs showed increased Runx2 expression and matrix mineralization, with potent antibacterial properties. This study focuses on the optimization of Hydrastis canadensis-based nanocarriers in environmental and microbial applications like antibacterial and dye degradation. Hydrastis canadensis (H. canadensis) is loaded into the nanocarrier using a gelation method. Characterization involves pH analysis, UV-VIS spectrophotometry, scanning electron microscopy, Fourier-transform infrared spectroscopy, dynamic light scattering, high-performance liquid chromatography, encapsulation efficiency. Further antimicrobial activity against Staphylococcus aureus and Escherichia coli were tested. Dye degradation was evaluated at concentrations of 1 % of high molecular (HM) and 1.5 % of low molecular (LM) chitosan nanoparticles with both 3C and 1000C concentrations of the drug. The obtained results confirm the presence of chitosan nanocarrier alongside the pure drug in 1 % HM and 1.5 % LM chitosan particles with a notable encapsulation efficiency activity in both 3C and 1000C concentrations. Antimicrobial studies were carried out using the agar well diffusion method and revealed a significant zone of inhibition of 20 mm and 25 mm for E. coli and S. aureus, respectively in chitosan nanocarrier-loaded samples compared to pure drug and chitosan nanocarriers samples. The dye degradation studies of four dyes methylene blue, methylene orange, methylene red, and safranin using both pure drugs and chitosan nanocarrier-loaded drugs showed the highest percentage of degradation (76 %) against methylene blue in the chitosan nanocarrier-drug loaded formulation. These findings cumulatively underscore chitosan nanoparticles can be used as an effective carrier for Hydrastis Canadensis, with enhanced antimicrobial and dye degradation capabilities. Varied concentrations and molecular weights highlight the versatility of the ionotropic gelation method in optimizing drug delivery. Enhanced efficacy of the nanocarrier was evident in the observed zone of inhibition in antimicrobial testing. The substantial degradation percentage in methylene blue emphasizes the formulation's applicability in environmental dye removal processes, with potential avenues for improvement explored through interactions between the chitosan nanocarrier and H. canadensis characteristics. Developing high-performance and durable catalysts presents a significant challenge for oxidizing toxic inorganic and pharmaceutical compounds in wastewater. Recently, there has been a surge in the development of new heterogeneous catalysts for degrading pharmaceutical compounds, driven by advancements in electrocatalysts and photoelectrocatalysts. In this study, a plasmonic Ag nanoparticles decorated CoFe 2 O 4 @TiO 2 heteronanostructures have been successfully designed to fabricate a high-performing photoelectrode for the oxidation of pharmaceutical compounds. The developed Ag-CoFe 2 O 4 @TiO 2 possessed a higher electrochemical stability and effectively harvested the UV to visible and NIR radiation in sunlight which generates the enormous photochemical reactive species that involved in the oxidation of ibuprofen in wastewater. Under direct sunlight irradiation, Ag-CoFe 2 O 4 @TiO 2 achieved complete oxidation of ibuprofen in wastewater at 0.8 V vs RHE. This indicates that metallic Ag nanoparticles are involved in the charge separation and transport of charge carriers from the photoactive sites of CoFe 2 O 4 @TiO 2 , promoting the generation of abundant hydroxy, oxy, and superoxide radicals that actively break the bonds of ibuprofen. Additionally, oxidation agents such as urea and H 2 O 2 were utilized to enhance the formation of superoxide ions and hydroxyl radicals, which rapidly participate in the oxidation of ibuprofen. Significantly, testing for recyclability confirmed the stability of the Ag-CoFe 2 O 4 @TiO 2 photoanode, ensuring its suitability for prolonged use in photoelectrochemical advanced oxidation processes. Urinary tract infections (UTIs) by Uropathogenic Escherichia coli (UPEC) are a significant health concern, especially due to the increasing prevalence of antibiotic resistance. This study focuses on isolating and characterizing bacteriophages specific to UPEC strains isolated from UTI samples. The isolated phages were assessed for their ability to target and lyse UPEC in vitro, focusing on their efficacy in disrupting biofilms, a key virulence factor contributing to UTI recurrence and antibiotic resistance. The morphological structure observed by TEM belongs to Myoviridae, the phage exhibited icosahedral symmetry with a long non-constricting tail, the approximate measurement of the phage head was 39 nm in diameter, and the phage tail was 105.317 nm in length. One-step growth experiments showed that the latent period was approximately 20 min, followed by a rise period of 40 min, and a growth plateau was reached within 20 min and the burst size observed was 26 phages/infected bacterial cells. These phages were capable of killing cells within the biofilms, leading to a reduction in living cell counts after a single treatment. This study highlights the potential of phages to play a significant role in disrupting, inactivating, and destroying Uropathogenic Escherichia coli (UPEC) biofilms. Such findings could be instrumental in developing treatment strategies that complement antibiotics and disinfectants. A higher prevalence of Acinetobacter baumannii infections and mortality rate has been reported recently in hospital-acquired infections (HAI). The biofilm-forming capability of A. baumannii makes it an extremely dangerous pathogen, especially in device-associated hospital-acquired infections (DA-HAI), thereby it resists the penetration of antibiotics. Further, the transmission of the SARS-CoV-2 virus was exacerbated in DA-HAI during the epidemic. This review specifically examines the complex interconnections between several components and genes that play a role in the biofilm formation and the development of infections. The current review provides insights into innovative treatments and therapeutic approaches to combat A. baumannii biofilm-related infections, thereby ultimately improving patient outcomes and reducing the burden of HAI.

Andrographis echioides has been extensively utilized in traditional Indian folk medicines for several skin disorders and other biological actions such as diuretic, antimicrobial, anthelmintic, anti-ulcer, and hepatoprotective properties. Different crude extracts were extracted from A. echioides leaves using various solvents such as methanol and water. The prepared crude extracts were utilized to formulate different herbal ointments. Further, the prepared ointments were examined against wounds and bacterial pathogens. The wound healing ability of the prepared formulations was observed for F 1 , F 2 , and F 3 , to be (89.84%, 95.11%, and 95.75%) respectively. Moreover, wound healing capabilities were compared with standard Betadine which exhibits 98.12%, those results indicating that the prepared herbal ointment also has a promising wound healing ability. The F 2 formulations outperform the other two formulations (F 1 and F 2 ) in terms of their antibacterial ability to combat Staphylococcus aureus, Klebsiella pneumoniae Bacillus subtilis, and Escherichia coli. Moreover, there are two compounds were successfully isolated and identified from methanolic extract, which are 2-(3,4-dihydroxyphenyl)-3,4-dihydro-2H-chromene-3,5,7-triol and 3-(3,4-Dihydroxyphenyl)-2-propenoic acid. Meanwhile, the molecular docking investigation exposed high binding energy Staphylococcus aureus TyrRS (-8.9 kcal/mol), Isoleucyl-tRNA synthetase (-7.5 kcal/mol), Penicillin-binding protein 2a (-8.0 kcal/mol), S. aureus DNA Gyrase (-7.2 kcal/mol), GSK-3beta (Glycogen synthase kinase-3 beta) (-8.3 kcal/mol) and TGF - Beta Receptor Type 1 Kinase Domain (-8.7 kcal/mol) indicating high degree of interaction between Compound-1 2-(3,4-dihydroxyphenyl)-3,4-dihydro-2H-chromene-3,5,7-triol (DHPDHC) and 7 clinically important skin infective pathogen Staphylococcus aureus proteins at the active sites. Alternate antibiotics developed through the involvement of nanomaterials are gaining interest due to their economical and lower toxicity concerns. A newly developed biopolymer-based polyvinylpyrrolidone/zinc oxide (PVP/ZnO) nanocomposite (NCs) was efficiently synthesized by an environment-friendly approach, utilizing onion and garlic peel extract as a bio-surfactant, zinc acetate as the source, PVP as the stabilizing agent, and sodium hydroxide as the precipitant. Fourier transform infrared spectroscopy (FT-IR) and X-ray diffraction (XRD) investigations verified the crystalline properties of ZnO, PVP, and PVP/ZnO-based NCs. The structure of the biopolymer-linked ZnO particles interpolated inside the PVP array was seen to have a layered and flaky structure, as validated by field emission scanning electron microscopy (FE-SEM) analysis, which revealed its occurrence in the nanometer range. The XRD examination verified that the surface topographical image of PVP/ZnO NCs had an average thickness of 21 nm. The PVP/ZnO nanocrystals demonstrated exceptional photocatalytic efficacy, with a breakdown rate of 88% and almost 92% for the methylene blue dye. Therefore, the PVP/ZnO matrix exhibits superior antibacterial activity compared to other extracts, resulting in greater microbial suppression. The results above indicate that the ZnO-intercalated PVP array has a stronger reinforcing effect than other components. Catheter related atrial thrombus (CRAT) is a devastating complication associated with tunneled hemodialysis catheter. Abiotrophia defectiva is a rare fastidious pathogen implicated predominantly in culture negative infective endocarditis. Here we report three cases of CRAT in maintenance hemodialysis patients with variable clinical presentation caused by Abiotrophia defectiva. Video assisted thoracoscopic retrieval of atrial thrombus is a novel technique which is scarcely reported in medical literature for surgical management of large atrial thrombus. Our cases were managed by timely administration of antibiotics and anticoagulants followed by surgical retrieval of atrial thrombus with removal of tunneled dialysis catheter. Multifunctional nanoparticles (NPs) production from phytochemicals is a sustainable process and an eco-friendly method, and this technique has a variety of uses. To accomplish this, we developed zinc oxide nanoparticles (ZnONPs) using the medicinal plant Tinospora cordifolia (TC). Instruments such as UV-Vis, XRD, FTIR, FE-SEM with EDX, and high-resolution TEM were applied to characterize the biosynthesized TC-ZnONPs. According to the UV-vis spectra, the synthesized TC-ZnONPs absorb at a wavelength centered at 374 nm, which corresponds to a 3.2 eV band gap. HRTEM was used to observe the morphology of the particle surface and the actual size of the nanostructures. TC-ZnONPs mostly exhibit the shapes of rectangles and triangles with a median size of 21 nm. The XRD data of the synthesized ZnONPs exhibited a number of peaks in the 2θ range, implying their crystalline nature. TC-ZnONPs proved remarkable free radical scavenging capacity on DPPH (2,2-Diphenyl-1-picrylhydrazyl), ABTS (2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid), and NO (Nitric Oxide). TC-ZnONPs exhibited dynamic anti-bacterial activity through the formation of inhibition zones against Pseudomonas aeruginosa (18 ± 1.5 mm), Escherichia coli (18 ± 1.0 mm), Bacillus cereus (19 ± 0.5 mm), and Staphylococcus aureus (13 ± 1.1 mm). Additionally, when exposed to sunlight, TC-ZnONPs show excellent photocatalytic ability towards the degradation of methylene blue (MB) dye. Parmotrema perlatum, a lichen belonging to the family Parmeliaceae, is well known for its culinary benefits and aroma used as a condiment in Indian homes is also known as the "black stone flower" or "kalpasi" in India. This research intends to analyze the antioxidant power of the crude extracts using four pH-based buffers solubilized proteins/peptides and RP-HPLC fractions of P. perlatum obtained by purification. The proteins that were extracted from the four different buffers were examined using LC-MS/MS-based peptide mass fingerprinting. When compared to the other buffers, the 0.1 M of Tris-HCl buffer pH 8.0 solubilized proteins/peptides had the strongest antioxidant capacity. The sequential purification of the peptide was carried out by using a 3-kDa cut-off membrane filter and semipreparative RP-HPLC. Additionally, the purified fractions of the peptide's antioxidant activity were assessed, and effects were compared with those of the crude and 3 kDa cut--off membrane filtrates. The peptide fractions were sequenced by LC-MS/MS, which reveals that fraction 2 from RP-HPLC with the sequence LSWFMVVAP has shown the highest antioxidant potential in comparison with other fractions which can serve as the potential natural antioxidant drug. This study was conducted to determine whether the macroalgae Ulva papenfussi and Ulva nematoidea could be alternatives for preventing Litopenaeus vannamei vibriosis caused by the bacterium Vibrio parahaemolyticus . Phytochemical screening was performed on methanolic extracts to qualitatively determine the main groups of bioactive compounds, previous to an in vitro antibacterial test against V. parahaemolitycus . Phenols, polyphenols, flavonoids, and the high presence of carbohydrates were found in both macroalgae. U. papenfussi showed more presence of lipids and alkaloids than U. nematoidea . Macroalgae extracts prepared (v:v) with a 1:1 methanol: dichloromethane solvent was used for the in vitro test using the disc diffusion method (MDD). Filter paper discs impregnated with 1.0, 1.5, 2.0, 3.0, and 4.0 mg of the extracts showed antibacterial activity against V. Parahaemolitycus in a dose-dependent manner in both macroalgae. The inhibition zone varied significantly ( p  < 0.05) from 8.33 ± 0.12 to 11.41 ± 0.73 mm for 1 to 3 mg of extract levels, respectively. In conclusion, both macroalgae have antibacterial activity in their crude extracts against this bacteria. It is suggested to evaluate it as a feed additive for L. vannamei . The design and preparation of dual-functional photocatalysts for simultaneously realizing photocatalytic wastewater purification and hydrogen energy generation pose significant challenges. This article presents the engineering of a binary heterostructured photocatalyst by combining TiO 2 (nanorods) and MoS 2 nanosphere using a straightforward solvothermal method and the assessment of the phase structures, morphologies, and optical properties of the resulting nanocomposites using diverse analytical techniques. The TiO 2 (Rod)/MoS 2 composite exhibits remarkable efficacy in degrading ciprofloxacin, achieving 93% removal rate within 1 h, which is four times higher than that of bare TiO 2 . Moreover, the optimized TiO 2 (Rod)/MoS 2 presents an outstanding hydrogen production rate of 7415 μmol g -1 , which is ∼24 times higher than that of pristine TiO 2 . Under UV-visible light irradiation, the TiO 2 (Rod)/MoS 2 heterojunction displays an exceptional photocatalytic performance in terms of both photodegradation and hydrogen production, surpassing the performance of TiO 2 particle/MoS 2 . The study findings demonstrate that TiO 2 (Rod)/MoS 2 nanocomposites exhibit considerably improved photocatalytic degradation and hydrogen generation activities. Antimicrobial resistance has emerged as a global threat to the treatment of infectious diseases. Antibacterial photodynamic therapy (aPDT) is a promising alternative approach and is highly suitable for the treatment of cutaneous bacterial infections through topical applications. aPDT relies on light-responsive compounds called photosensitizer (PS) dyes, which generate reactive oxygen species (ROS) when induced by light, thereby killing bacterial cells. Despite several previous studies in this area, the molecular details of targeting and cell death mediated by PS dyes are poorly understood. In this study, we further investigate the antibacterial properties of two water-soluble Sn(IV) tetrapyridylporphyrins that were quaternized with methyl and hexyl groups ( 1 and 2 ). In this follow-up study, we demonstrate that Sn(IV)-porphyrins can be photoexcited by blue light (a 427 nm LED) and exhibit various levels of bactericidal activity against both Gram-(+) and Gram-(-) strains of bacteria. Using localization studies through fluorescence microscopy, we show that 2 targets the bacterial membrane more effectively than 1 and exhibits comparatively higher aPDT activity. Using multiple fluorescence reporters, we demonstrate that photoactivation of 1 and 2 results in extensive collateral damage to the bacterial cells including DNA cleavage, membrane damage, and delocalization of central systems necessary for bacterial growth and division. In summary, this investigation provides deep insights into the mechanism of bacterial killing mediated by the Sn(IV)-porphyrins. Moreover, our approach offers a new method for evaluating the activity of PS, which may inspire the discovery of new PS with enhanced aPDT activity.

Metal nanoparticles play an increasingly pivotal role in promoting efficient wound healing and preventing secondary bacterial infections. Their small size and high surface area facilitate enhanced biological interaction and penetration at the wound site. Specifically designed for topical drug delivery, these nanoparticles enable the sustained release of therapeutic molecules, such as growth factors and antibiotics. This targeted approach ensures optimal cell-to-cell interactions, proliferation, and vascularization, fostering effective and controlled wound healing. Nanoscale scaffolds have significant attention due to their attractive properties, including delivery capacity, high porosity and high surface area. The aim of this work is to prepare ternary blended polyvinyl alcohol (PVA)-urea hydrogels containing Ormocarpum cochinchinense, Cinnamomum zeylanicum and antibiotic cephalexin by freezing-thawing method in order to assess the wound healing qualities. In addition to being a synthetic polymer, PVA is a recyclable and biocompatible artificial polymer blend that has attracted a lot of interest in biological applications. The freezing-thawing process with PVA-urea blend is used to make hydrogel film. Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD), scanning electron microscopy (SEM) and swelling investigations were carried out for the composite membranes. Biological studies involving antibacterial, antifungal, cytotoxicity and wound healing activities were also carried out for the composite membranes. The composite membrane developed has a lot of potential for wound dressing and other applications.

Aim: Acinetobacter baumannii (AB) is a clinically important bacterial pathogen responsible for nosocomial infections. The biofilm-forming capability of these pathogens reduces the antibiotic penetration and its efficacy, thereby complicating the treatment. The current work aims to isolate the most potent biofilm-forming Acinetobacter species from clinical isolates of the patient samples and to evaluate the efficacy of the amikacin-humic acid combination against it. Methods: The combination effect of Amikacin-Humic (AMK-HUM) acid against the highest biofilm-producing A. baumannii SLMK001 was studied via in - vitro (microscopic analysis) and in - silico (Network Pharmacology) analysis. Results: The amikacin-humic acid combination significantly inhibited both the biofilm formation and cell viability of A. baumannii SLMK001. The images observed via Scanning Electron Microscope (SEM) showed a significant decrease in the biofilm matrix. Confocal Laser Scanning Microscope (CLSM) confirmed a reduction of the Z value of its three-dimensional structure. Further, the Network Pharmacology approach supported these experimental findings by identifying the key targets of the amikacin-humic acid combination against the biofilm pathways of A. baumannii . Conclusion: The in-vitro results aligned with the in-silico findings, indicating that the AMK-HUM combination is a promising treatment that significantly activates the key proteins against A. baumannii biofilm formation and pathogenesis.

The human gut microbiota plays a crucial role in maintaining overall health. However, the widespread use of antibiotics has raised concerns about its impact on the microbial ecosystem. This review explores the multifaceted relationship between antibiotics and gut dysbiosis, highlighting the mechanisms underlying these interactions and their implications for human health. Antibiotics, while invaluable in treating infections, disrupt the gut microbiota by indiscriminately targeting both harmful and beneficial microorganisms. This disturbance leads to a reduction in microbial diversity, altered metabolite production, and compromised immune responses, resulting in a state referred to as dysbiosis. Broad-spectrum antibiotics tend to induce more severe dysbiosis compared to narrow-spectrum agents. Antibiotic-induced dysbiosis has been linked to the onset and progression of these disorders, emphasizing the far-reaching consequences of microbial imbalance. The review highlights various strategies to mitigate the adverse effects of antibiotics on gut health, like probiotics, fecal microbiota transplantation (FMT), and phage therapy, as promising approaches to restore and maintain a balanced gut microbiota. Introduction: The health-related information-seeking through internet sources has drastically increased throughout the world. This study aimed to analyze the Global public interest on "antibiotics" and "antibiotic resistance"., Methods: The worldwide internet trend for the search terms "antibiotics" and "antibiotic resistance" from February 2017 to February 2022 was obtained using Google trends. The seasonal variation of interest was analyzed using the Seasonal Decomposition of Time Series by Loess., Results: The mean interest for the search term "antibiotics" and "antibiotic resistance" is found to be 78.02 ± 7.5 and 2.3 ± 0.8, which will increase at 2.56 % and 16 % per year. It was observed that there was a significant relationship between antibiotic consumption, the number of physicians, and individuals using the internet in the countries with the search term "antibiotics". The study also indicates that there is a peak in search volume for the term during the COVID-19 Pandemic., Conclusion: Our study observed, that antibiotics related search questions on google by the public indicate the chances of antibiotic misuse. Fluoroquinolones (FQs) are the class of Antibiotics (ABs) that have been extensively used worldwide for the treatment of diseases caused by bacterial infections. In India, most of these untreated ABs and their unused metabolites present in treated and untreated wastewater end up in agricultural land and water bodies. This can accelerate the problem of antimicrobial resistance in the community. Hence, we aim to develop a cost-effective sensor to detect and monitor the presence of such drugs in water bodies. We have prepared a chemically reduced graphene oxide (rGO) integrated luminescent cerium metal-organic framework (MOF) that specifically targets and recognizes ciprofloxacin (CPFX), norfloxacin (NFX), and ofloxacin (OFX) achieving excellent sensing activity. A remarkable quenching of the fluorescence of MOF composite was observed upon interaction with CPFX, NFX, and OFX with 57.9, 46.3, and 51.6 ppb as limits of detection, respectively, through a Forster resonance energy transfer from the Ce-MOF to the analytes. The applicability of the sensor was also examined with real-time samples collected from the rivers of Chennai city and the MOF probe exhibited an appreciable recovery of results. Antimicrobial resistance (AMR) is a growing public health concern worldwide, and it poses a significant threat to human, animal, and environmental health. The overuse and misuse of antibiotics have contributed significantly and others factors including gene mutation, bacteria living in biofilms, and enzymatic degradation/hydrolyses help in the emergence and spread of AMR, which may lead to significant economic consequences such as reduced productivity and increased health care costs. Nanotechnology offers a promising platform for addressing this challenge. Nanoparticles have unique properties that make them highly effective in combating bacterial infections by inhibiting the growth and survival of multi-drug-resistant bacteria in three areas of health: human, animal, and environmental. To conduct an economic evaluation of surveillance in this context, it is crucial to obtain an understanding of the connections to be addressed by several nations by implementing national action policies based on the One Health strategy. This review provides an overview of the progress made thus far and presents potential future directions to optimize the impact of nanobiotics on AMR.

Pharmaceutically active compounds are an emerging water contaminant that resists conventional wastewater treatments. Herein, the sonophotocatalytic degradation of Tetracycline (TC) antibiotics as a model contaminant was carried out over a rod-like g-C 3 N 4 /V 2 O 5 (RCN-VO) nanocomposite. RCN-VO nanocomposite was synthesized via ultrasound-assisted thermal polycondensation method. The results showed that the RCN-VO nanocomposite could completely remove the TC in water within 60 min under simultaneous irradiation of visible light and ultrasound. Moreover, the sonophotocatalytic TC degradation (a synergy index of ∼1.5) was superior to the sum of individual sonocatalytic and photocatalytic degradation using RCN-VO nanocomposite. Besides, the enhanced sonophotocatalytic activity of RCN-VO can be attributed to the 1D/2D nanostructure and the S-scheme heterojunction formation between RCN and VO where the electrons migrated from RCN to VO across the RCN-VO interface. Under irradiation, the built-in electric field, band edge bending and Coulomb interaction can synergistically facilitate the unavailing electron-hole pair recombination. Thereby, the cumulative electron in RCN and holes in VO can actively take part in the redox reaction which generates free radicals and attack the TC molecules. Wound healing is a multi-stage process that is dynamic, interactive, and complicated. However, many nanomaterials are employed to expedite wound healing by demonstrating antibacterial activity or boosting cell proliferation. But only one phase is focused during the wound healing process. As a result, there is a need for optimum wound dressing materials that promotes different wound healing cascades with ideal properties. Herein, Graphene Oxide loaded with vanadium (V) doped titanium dioxide (TiO 2 ) blended with chitosan, and polyvinyl alcohol (CS/PVA/GO/TiO 2 -V) patch was developed for wound healing. XRD, FTIR and FE-SEM analyses were carried out to study the morphology and structural property of the patch. The fabricated patch has a high surface porosity, excellent moisture vapor transfer rate, appropriate swelling behaviour, and oxygen permeability, which results in an excellent moist environment for wound breathing and effective management of wound exudates. The antibacterial test showed significant antibacterial efficacy against wound infections in the presence of light when compared to dark. In-vitro analysis such as hemocompatibility, cytotoxicity, cell adhesion, and scratch assay show the predicted potential wound healing application with high biocompatibility. These results suggest that CS/PVA/GO/TiO 2 -V patch provides a microenvironment favourable to cells' growth and differentiation and positively modulates full-thickness wounds' healing., (Copyright © 2021 Elsevier B.V. The immune system is a dynamic network of cells and cytokines are the major mediators of immune responses which combat pathogens. Based on the cytokine production, effector T cells differentiate into subsets known as Th1, Th2, Th17, or Treg. This system serves as a barrier to intracellular pathogens, bacterial infections and stimulates the production of reactive oxygen species (ROS), reactive nitrogen intermediates, and nitric oxide, which diffuses across membranes and engulfs intracellular pathogens. Oxidative stress occurs when ROS, reactive nitrogen species (RNS) production, and antioxidant defences become imbalanced. Oxidative stress generated by infected cells produces a substantial amount of free radicals which enables the killing of intracellular pathogens. Intracellular pathogens are exposed to endogenous ROS as part of normal aerobic respiration, also exogenous ROS and RNS are generated by the host immune system in response to infection. Nanoparticles which are designed for drug delivery are capable of trapping the desired drug in the particles which protect the drug from enzymatic degradation in a biological system. The subcellular size of nanoparticles enables higher intracellular uptake of the drug which results in the reduction of the concentration of free drugs reducing their toxic effect. Research on the modulation of immune response and oxidative stress using nanoparticles used to encapsulate drugs has yet to be explored fully. Development of antimicrobial drugs against multidrug-resistant (MDR) bacteria is a great focus in recent years. TG12, a short peptide molecule used in this study was screened from tachykinin (Tac) protein of an established teleost Channa striatus (Cs) transcriptome. Tachykinin cDNA has 345 coding sequence, that denotes a protein contained 115 amino acids; in which a short peptide (TG12) was identified at 83-94. Tachykinin mRNA upregulated in C. striatus treated with Aeromonas hydrophila and Escherichia coli lipopolysaccharide (LPS). The mRNA up-regulation was studied using real-time PCR. The up-regulation tachykinin mRNA pattern confirmed the immune involvement of tachykinin in C. striatus during infection. Further, the identified peptide, TG12 was synthesized and its toxicity was demonstrated in hemolytic and cytotoxic assays using human erythrocytes and human dermal fibroblast cells, respectively. The toxicity study exhibited that the toxicity of TG12 was similar to negative control, phosphate buffer saline (PBS). Moreover, the antibiogram of TG12 was active against Klebsiella pneumonia ATCC 27736, a major MDR bacterial pathogen. Further, the antimicrobial activity of TG12 against pathogenic bacteria was screened using minimum inhibitory concentration (MIC) and anti-biofilm assays, altogether TG12 showed potential activity against K. pneumonia. Fluorescence assisted cell sorter flow cytometer analysis (FACS) and field emission scanning electron microscopy (FESEM) was carried on TG12 with K. pneumonia; the results showed that TG12 significantly reduced K. pneumonia viability as well as TG12 disrupt its membrane. In conclusion, TG12 of CsTac is potentially involved in the antibacterial immune mechanisms, which has a prospectus efficiency in pharma industry against MDR strains, especially K. pneumonia., (Copyright © 2021 Elsevier Inc. Evolution of bacterial tolerance to antimicrobials precedes evolution of resistance and may result in cross-tolerance, cross-resistance, or collateral sensitivity to other antibiotics. Transient exposure of gut bacteria to glyphosate, the world's most widely used herbicide, has been linked to the activation of the stress response and changes in susceptibility to antibiotics. In this study, we investigated whether chronic exposure to a glyphosate-based herbicide (GBH) results in resistance, a constitutive activation of the tolerance and stress responses, and cross-tolerance or cross-resistance to antibiotics. Of the 10 farm animal-derived clinical isolates of Salmonella enterica subjected to experimental evolution in increasing concentrations of GBH, three isolates showed stable resistance with mutations associated with the glyphosate target gene aroA and no fitness costs. Global quantitative proteomics analysis demonstrated activation of the cellular tolerance and stress response during the transient exposure to GBH but not constitutively in the resistant mutants. Resistant mutants displayed no cross-resistance or cross-tolerance to antibiotics. These results suggest that while transient exposure to GBH triggers cellular tolerance response in Salmonella enterica , this response does not become genetically fixed after selection for resistance to GBH and does not result in increased cross-tolerance or cross-resistance to clinically important antibiotics under our experimental conditions. IMPORTANCE Glyphosate-based herbicides (GBH) are among the world's most popular, with traces commonly found in food, feed, and the environment. Such high ubiquity means that the herbicide may come into contact with various microorganisms, on which it acts as an antimicrobial, and it may select for resistance and cross-resistance to clinically important antibiotics. Graphitic carbon nitride (g-C 3 N 4 ) is an analog of graphite due to its unique electronic structure. g-C 3 N 4 based materials have been used in photocatalytic applications. However, pure g-C 3 N 4 suffers from major shortcomings which include poor disparity, low surface area and a high recombination rate of photo generated electron-hole pairs that significantly reduce its photocatalytic activity. In this work, self-assembly of g-C 3 N 4 sheet into rod shaped g-C 3 N 4 was developed via a simple polymerisation method. A composite made of g-C 3 N 4 nanorods and rGO (rGO-RCN) was also prepared. The band gap g-C 3 N 4 was shifted from 2.77 to 2.6 eV evidented by UV-DRS data. As a result, rGO-RCN showed a relatively high absorption in the visible region. Moreover, a fast electron transfer rate was observed with rGO-RCN composite as conformed from PL analysis and photocurrent measurement. The formation of nanorod and sheet morphologies was confirmed via TEM analysis. The photocatalytic activities of prepared sheet-g-C 3 N 4 (SCN) , Rod g-C 3 N 4 (RCN), reduced graphene oxide supported sheet-g-C 3 N 4 (rGO-SCN) and reduced graphene oxide supported Rod-g-C 3 N 4 (rGO-RCN) were evaluated using a commonly used antibiotic (tetracycline). Among these catalysts, rGO-RCN nanocomposite showed sonophotocatalytic activity 3 times higher compared to pure g-C 3 N 4 . This superior sonophotocatalytic activity could be due to enhanced visible light absorption of the material, active sites generated by ultrasound, and the high electron transport property of rGO., (Copyright © 2020. Penaeid prawns are considered as most demanding fishery resources. The current study aims to purify and characterize lectin from the haemolymph of Penaeus semisulcatus. The semisulcatus-lectin was purified by affinity chromatography using mannose coupled Sepharose CL-4B column and purified lectin exhibited a single band of 66 kDa in SDS-PAGE. The purity and crystalline structure of purified lectin was confirmed by HPLC and X-ray diffraction analysis. Semisulcatus-lectin exhibited yeast agglutination activity against Saccharomyces cerevisiae and agglutinated human erythrocytes. Semisulcatus-lectin was evaluated for phenol oxidase activation and phagocytic activities. It was observed that semisulcatus-lectin had antibacterial activity against Gram-negative Vibrio parahaemolyticus and Aeromonas hydrophila, suggesting a potential therapeutic strategy in aquaculture industry for disease management., (Copyright © 2020 Elsevier Ltd. Soil and sludge are major reservoirs of organic compounds such as fluoroquinolones (FQs) which are broad-spectrum antibacterial agents. Hence, we monitored three major FQs, namely, ciprofloxacin (CIP), norfloxacin (NOR), and ofloxacin (OFL), in surface soil from two major dumpsites and dry and wet sludge from sewage treatment plants in Chennai city. The mean concentration of FQs in soil and sludge samples were 20 μg/g and 26 μg/g, respectively. Nearly 50% of the total FQs in dumpsite soil was contributed by CIP followed by NOR (32%) and OFL (13%). Similarly, CIP was the major contributor in sludge samples followed by NOR and OFL. The concentration of FQs was two folds higher in wet sludge than dry sludge most likely indicating that water solubility of these compounds might play an important role for elevated level of FQs in wet sludge. Solid waste from pharmaceutical industries, households, and sludge from wastewater treatment plants were expected to be the major source of FQs in dumpsite soil. Predicted risk assessment using soil to water migration concentrations via surface run off indicated high risk to aquatic organisms. However, risk quotient (RQ) was found less to earthworm in most of the soil samples. Pathogenic microorganism delays wound-healing process by causing infection. In recent years, researchers have developed various kinds of photo-active nanomaterials with enhanced antibacterial properties. This work focus on the preparation of graphene oxide and TiO 2 nanocomposites (GO/TiO 2 ) as a visible light-induced high efficiency antibacterial material. The hydrothermal method was used for the synthesis of GO/TiO 2 nanocomposites at 180 o C for 3 h with different loading percentages of GO (10, 20, 30, 40 and 50 wt. %). The systematic characterization tools including X-ray diffraction analysis, FT-IR, UV-vis, Raman and TEM which were used to understand the physicochemical properties of the prepared GO/TiO 2 nanocomposites. Furthermore, GO/TiO 2 nanocomposites were used as photocatalytic active materials against wound infection-causing bacteria in the presence of visible light irradiation. The possible antibacterial mechanism under presence and absence of light were depicted. The antibacterial mechanism of the GO/TiO 2 nanocomposite was investigated on wound infection-causing bacteria such as Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli, and Enterococcus faecalis. The high hemocompatibility and the cellular biocompatibility of the nanocomposite aids in using it for wound-healing application. Overall, the results suggest that the GO/TiO 2 nanocomposite could be developed as a photo-active nanomaterial against pathogenic microorganisms that are present in wound.

Seven piperic acid amides along with their lower homologs (12) were synthesized using HATU-DIPEA coupling reagent. All the synthesized derivatives were evaluated for their antibacterial activities against Staphylococcus aureus, Pseudomonas aeruginosa, and vancomycin-resistant P. aeruginosa. They were found to be more active on P. aeruginosa than on S. aureus. However, they did not exhibit potent activity on Vancomycin resistant P. aeruginosa. Among the tested compounds, methylenedioxycinnamic acid amide of anthranilic acid (MDCA-AA, 2a) was found to be most active against S. aureus with MIC of 3.125 μg/ml. The PAS and INH amides of piperic acid were screened against Mycobacterium tuberculosis H37R a strain. Titanium dioxide nanoparticles exhibit good anticancer and antibacterial activities. They are known to be environmentally friendly and stable, less toxic and excellent biocompatibility nature. In this paper we report the biological properties of pure TiO 2 nanoparticles modified with Withania somnifera (Ashwagandha), Eclipta prostrata (Karisalankanni) and Glycyrrhiza glabra (Athimathuram) for biological applications. X-ray diffraction results revealed the anatase nature of the samples. From the TEM analyses, it is observed that there is an increase in the particle size of the bio modified samples. UV results show the red shift for the bio modified samples when compared with the pure samples. The samples are then subjected to MTT assay to determine the cell viability. KB oral cancer cells are used for the determination of anticancer nature of the pure and bio modified nanoparticles. It is observed that Withania somnifera - Eclipta prostrate modified TiO 2 nanoparticles exhibit excellent anticancer activities among other bio modified and pure samples. The samples are then examined for their antibacterial activities against three Gram-negative bacterial strains namely, Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa and two Gram-positive bacterial strains namely, Staphylococcus aureus and Streptococcus mutans. Purpose: The preclinical pharmacokinetic and pharmacodynamic properties of a potent fluoroketolide RBx14255 against Streptococcus pneumoniae and Haemophilus influenzae was compared with telithromycin and human clinical dose was predicted for preclinical development., Methods: The in vitro pharmacokinetic characterization was performed for solubility, Caco-2 permeability, microsomal stability, CYP inhibition and plasma protein binding. In vivo pharmacokinetic studies were performed in Swiss albino mice, Sprague Dawley rats and Beagle dogs. The pharmacodynamic studies were carried out in mouse against S. pneumoniae in systemic infection and against S. pneumoniae and H. influenzae in rat lung infection models., Results: RBx14255 showed superior potency and efficacy in mouse and rat infection models. RBx14255 showed pH dependent solubility (0.41 mg/mL at pH 6.8 and >1 mg/mL at pH 1.2), moderate Caco-2 permeability (A to B: 12 nm/s) with high efflux ratio. It showed high plasma protein binding (>97%) in mouse and low binding (45- 70%) in rat, dog and human. The compound is mainly metabolized through CYP3A4. Pharmacokinetic parameters and absolute bioavailability of both, RBx14255 and telithromycin are similar in mouse. Both the ketolides showed low plasma clearance (18% of the normal hepatic blood flow rate) in mouse, moderate to high clearance in rat and dog. Mean oral bioavailability was high in mouse (≥85%), moderate in rat (RBx14255: 15% and telithromycin: 51%) and high to moderate in dog (RBx14255: 98% and telithromycin: 56%). The predicted efficacious dose for a 70 kg man ranges from 124 mg BID to 226 mg BID., Conclusion: RBx14255 displayed significantly better pharmacodynamics which correlates with the pharmacokinetic properties against S. pneumoniae and H. influenzae as compared to telithromycin. The predicted human efficacious doses are in the range of 124-226 mg, making it amenable to oral dosage form drug in human. This could be a promising clinical candidate for future studies.

Biocompatible polymers are being used in recent times for treating skin injuries and burn wounds. Polymers like Poly Vinyl Alcohol and Chitosan are proven to be biocompatible with least toxic to treat injuries with minimal side-effects. Curcumin, a primary component of turmeric has anti-inflammatory properties and anti-microbial activity but has extremely low bioavailability. Converting Curcumin to its nano form increased its bioavailability exponentially allowing it to play a vital role in the process of wound healing. This PVA/Chi/Cur patch increased cell proliferation as shown by the results of cell line studies and MTT assay. Its anti-bacterial activity against four major bacterial strains commonly found in wound sites and water retainability indicates it to be a perfect material for wound treatment. Results of in-vivo studies conducted on wistar rats by testing the patch's healing ability on a surgically induced wound displayed its superiority over commercial ointment.
