Antarctica is an untapped reservoir of bacterial communities, which are able to adapt to a huge variety of strategies to cope with extreme conditions and, therefore, are capable of producing potentially valuable compounds for biotechnological applications. In this study, 31 UV-resistant bacteria collected from different Antarctic aquatic environments (surface sea waters/ice and shallow lake sediments) were isolated by UV-C assay and subsequently identified. A phylogenetic analysis based on 16S rRNA gene sequence similarities showed that the isolates were affiliated with Proteobacteria, Actinobacteria and Firmicutes phyla, and they were clustered into 15 bacterial genera, 5 of which were Gram negative (Brevundimonas, Qipengyuania, Sphingorhabdus, Sphingobium, and Psychrobacter) and 10 of which were Gram positive (Staphylococcus, Bacillus, Mesobacillus, Kocuria, Gordonia, Rhodococcus, Micrococcus, Arthrobacter, Agrococcus, and Salinibacterium). Strains belonging to Proteobacteria and Actinobacteria phyla were the most abundant species in all environments. The genus Psychrobacter was dominant in all collection sites, whereas bacteria belonging to Actinobacteria appeared to be the most diverse and rich in terms of species among the investigated sites. Many of these isolates (20 of 31 isolates) were pigmented. Bacterial pigments, which are generally carotenoid-type compounds, are often involved in the protection of cells against the negative effects of UV radiation. For this reason, these pigments may help bacteria to successfully tolerate Antarctic extreme conditions of low temperature and harmful levels of UV radiation. [ABSTRACT FROM AUTHOR]

Abstract: The Antarctic marine environment is one of the most extreme on Earth due to its stably low temperature and high oxygen content. Here we discuss various aspects of the molecular adaptations evolved by Antarctic fish and marine microorganisms living in this environment. This review will in particular focus on: (i) the genetic/genomic bases of adaptation in Antarctic notothenioid fish; (ii) the role of neuroglobin recently identified in the brain of Antarctic icefish; (iii) the structural and functional features of globins of the Antarctic marine bacterium Pseudoalteromonas haloplanktis TAC125. The dominant perciform suborder Notothenioidei is an excellent study group for assessing the evolution and functional importance of biochemical adaptations to temperature. The availability of notothenioid taxa in a wide range of latitudes (Antarctic and non-Antarctic) provides a tool to enable identification of physiological and biochemical characteristics gained and lost during evolutionary history. Non-Antarctic notothenioids belonging to the most basal families are a crucial source for understanding the evolution of hemoglobin in high-Antarctic cold-adapted fish. This paper focuses on the structure, function and evolution of the oxygen-transport system of Cottoperca gobio, a sub-Antarctic notothenioid fish of the family Bovichtidae, probably derived from ancestral species that evolved in the Antarctic region and later migrated to lower latitudes. Unlike most high-Antarctic notothenioids, but similar to many other acanthomorph teleosts, C. gobio has two major hemoglobins having the β chain in common. The oxygen-binding equilibria and kinetics of the two hemoglobins have been measured. Hb1 and Hb2 show strong modulation of oxygen-binding equilibria and kinetics by heterotropic effectors, with marked Bohr and Root effects. In Hb1 and Hb2, oxygen affinity and subunit cooperativity are slightly higher than in most high-Antarctic notothenioid hemoglobins. Hb1 and Hb2 show similar rebinding rates, but also show significant dynamic differences that are likely to have functional consequences. Molecular dynamic simulations of C. gobio Hb1 were performed on the dimeric protein in order to obtain a better understanding of the molecular basis of structure/function relationships. Abstract: The genome sequence of the Antarctic Gram-negative marine eubacterium Pseudoalteromonas haloplanktis TAC125 is a potential source of useful data on proteins from a cold-adapted microorganism. Identifying the bases of protein adaptation to higher or lower temperatures is important to understand the relationship between structure/function and life history on the Earth. The P. haloplanktis TAC125 genome contains three genes in distinct positions on chromosome I, named PSHAa0030, PSHAa2217 and PSHAa0458. These genes encode three truncated hemoglobins. The amino-acid identity between the three hemoglobins is less than 25% suggesting that these proteins may have different function(s) in bacterial cellular metabolism. The hemoglobin encoded by the PSHAa0030 gene has been cloned, expressed in Escherichia coli, purified and structurally characterised. This truncated hemoglobin is monomeric; circular dichroism shows high temperature resistance. The optical spectra of oxygenated and CO forms are similar to those of other truncated hemoglobins. Phylogenetic analyses show that two truncated globins encoded by the PSHAa0030 and PSHAa2217 genes belong to group II, and the third one encoded by PSHAa0458 to group I. [Copyright &y& Elsevier]

Haemoglobin carries oxygen from the environment to tissues; in vertebrates, it is contained in specialized cells, called erythrocytes. Over the last century, the study of the chemical properties of this haemoprotein has provided a wealth of information. One of its most important and ancient physiological features is the Root effect, found in many teleost fish (and some amphibians). The Root effect corresponds to an extreme pH sensitivity and can be described as an exaggerated Bohr effect: it dictates to what extent the oxygen tension can be raised in acid-producing tissues. It is likely that the eye choroid rete represents the most ancient anatomical structure associated with the presence of Root effect haemoglobins. This review describes our overall understanding of the molecular properties, biological occurrence, physiological role and evolutionary origin of Root effect haemoglobins. The current knowledge of the structural properties of Root effect haemoglobins is discussed in the light of recent results obtained on the haemoglobins of the cold-adapted notothenioids Trematomus newnesi and T. bernacchii. Sponges are known to produce a series of compounds with bioactivities useful for human health. This study was conducted on four sponges collected in the framework of the XXXIV Italian National Antarctic Research Program (PNRA) in November-December 2018, i.e., Mycale (Oxymycale) acerata, Haliclona (Rhizoniera) dancoi, Hemimycale topsenti, and Hemigellius pilosus. Sponge extracts were fractioned and tested against hepatocellular carcinoma (HepG2), lung carcinoma (A549), and melanoma cells (A2058), in order to screen for antiproliferative or cytotoxic activity. Two different chemical classes of compounds, belonging to mycalols and suberitenones, were identified in the active fractions. Mycalols were the most active compounds, and their mechanism of action was also investigated at the gene and protein levels in HepG2 cells. Of the differentially expressed genes, ULK1 and GALNT5 were the most down-regulated genes, while MAPK8 was one of the most up-regulated genes. These genes were previously associated with ferroptosis, a programmed cell death triggered by iron-dependent lipid peroxidation, confirmed at the protein level by the down-regulation of GPX4, a key regulator of ferroptosis, and the up-regulation of NCOA4, involved in iron homeostasis. Marine sponges commonly host a repertoire of bacterial-associated organisms, which significantly contribute to their health and survival by producing several anti-predatory molecules. Many of these compounds are produced by sponge-associated bacteria and represent an incredible source of novel bioactive metabolites with biotechnological relevance. Although most investigations are focused on tropical and temperate species, to date, few studies have described the composition of microbiota hosted by Antarctic sponges and the secondary metabolites that they produce. The investigation was conducted on four sponges collected from two different sites in the framework of the XXXIV Italian National Antarctic Research Program (PNRA) in November–December 2018. Collected species were characterized as Mycale (Oxymycale) acerata, Haliclona (Rhizoniera) dancoi, Hemigellius pilosus and Microxina sarai by morphological analysis of spicules and amplification of four molecular markers. Metataxonomic analysis of these four Antarctic sponges revealed a considerable abundance of Amplicon Sequence Variants (ASVs) belonging to the phyla Proteobacteria, Bacteroidetes, Actinobacteria and Verrucomicrobia. In particular, M. (Oxymycale) acerata, displayed several genera of great interest, such as Endozoicomonas, Rubritalea, Ulvibacter, Fulvivirga and Colwellia. On the other hand, the sponges H. pilosus and H. (Rhizoniera) dancoi hosted bacteria belonging to the genera Pseudhongella, Roseobacter and Bdellovibrio, whereas M. sarai was the sole species showing some strains affiliated to the genus Polaribacter. Considering that most of the bacteria identified in the present study are known to produce valuable secondary metabolites, the four Antarctic sponges could be proposed as potential tools for the discovery of novel pharmacologically active compounds. Abstract: The recognition of the important role of the polar habitats in global climate changes has awakened great interest in the evolutionary biology of polar organisms. They are exposed to strong environmental constraints, and it is important to understand how they have adapted to cope with these challenges and to what extent adaptations may be upset by current climate changes. We present an introductory overview of the evolution of the Antarctic fish fauna with emphasis on the dominant perciform sub-order Notothenioidei, as well as some specific comments on the biogeography of the three phyletically basal notothenioid families. The wealth of information on the ecology and biodiversity of the species inhabiting high-Antarctic and sub-Antarctic regions provides a necessary framework for better understanding the origin, evolution and adaptation of this unique group of fish. Notothenioidei offer opportunities for identification of the biochemical characters or the physiological traits responsible for thermal adaptation. The availability of phylogenetically related taxa in a wide range of latitudes has allowed to look into the molecular bases of environmentally driven phenotypic gain and loss of function. In the process of cold adaptation, the evolutionary trend of notothenioids has produced unique specialisations, including modification of hematological characteristics, e.g. decreased amounts and multiplicity of hemoglobins. The Antarctic family Channichthyidae (the notothenioid crown group) is devoid of hemoglobin. This loss is related to a single deletional event removing all globin genes with the exception of the inactive 3′ end of adult α-globin. In reviewing hemoglobin structure, function and phylogeny, the evolution of the fish Root effect is analysed in detail. Adaptation of the oxygen-transport system in notothenioids seems to be based on evolutionary changes involving levels of biological organisation higher than the structure of hemoglobin.
