Spontaneously arisen hereditary diseases in domestic animals provide an excellent opportunity to study the physiological functions of the altered genes. We investigated two 4-month-old sibling domestic short haired kittens with dry dark debris around the eyes, nose, and ears, dark crusting on the legs and a thin poor hair coat. Skin biopsies revealed abnormal sebaceous gland morphology with lack of normal sebocyte arrangement and differentiation. Hair follicles had a distorted silhouette, interpreted as a change secondary to the observed sebaceous gland dysplasia. Whole genome sequencing on both affected kittens and 65 genetically diverse feline genomes was performed. Filtering for variants that were present in both kittens but absent from the control genomes revealed a homozygous missense variant in SOAT1, encoding sterol O-acyltransferase 1. The protein is localized in the endoplasmic reticulum and catalyzes the formation of cholesteryl esters, an essential component of sebum and meibum. The identified SOAT1:c.1531G > A variant is predicted to change a highly conserved glycine residue within the last transmembrane domain of SOAT1, p.Gly511Arg. In mice, variants in Soat1 or complete knockout of the gene lead to the “hair interior defect” (hid) or abnormal Meibomian glands, respectively. SOAT1:c.1531G > A represents a plausible candidate variant for the observed sebaceous gland dysplasia in both kittens of this study. The variant was not present in 10 additional cats with a similar clinical and histopathological phenotype suggesting genetic heterogeneity. SOAT1 variants should be considered as potential cause in hereditary sebaceous gland dysplasias of humans and domestic animals.

Investigations of hereditary phenotypes in spontaneous mutants may help to better understand the physiological functions of the altered genes. We investigated two unrelated domestic shorthair cats with bulbous swellings of the hair shafts. The clinical, histopathological, and ultrastructural features were similar to those in mice with lanceolate hair phenotype caused by loss-of-function variants in Dsg4 encoding desmoglein 4. We sequenced the genomes from both affected cats and compared the data of each affected cat to 61 control genomes. A search for private homozygous variants in the DSG4 candidate gene revealed independent frameshift variants in each case, c.76del or p.Ile26fsLeu*4 in case no. 1 and c.1777del or p.His593Thrfs*23 in case no. 2. DSG4 is a transmembrane glycoprotein located primarily in the extracellular part of desmosomes, a complex of adhesion molecules responsible for connecting the keratin intermediate filaments of neighbouring epithelial cells. Desmosomes are essential for normal hair shaft formation. Both identified DSG4 variants in the affected cats lead to premature stop codons and truncate major parts of the open-reading frame. We assume that this leads to a complete loss of DSG4 function, resulting in an incorrect formation of the desmosomes and causing the development of defective hair shafts. Together with the knowledge on the effects of DSG4 variants in other species, our data suggest that the identified DSG4 variants cause the hair shaft dystrophy. To the best of our knowledge, this study represents the first report of pathogenic DSG4 variants in domestic animals.

The leishmaniases are a group of diseases caused by the protozoan parasite belonging to the genus Leishmania. In the New World, although dogs are considered the main parasite reservoir, in the last two decades, several studies have confirmed the role of cats (Felis catus) in the epidemiology of the disease and feline leishmaniasis (FeL) is now considered to be an emerging disease. The present review summarizes the current knowledge about FeL, focusing on important immunopathological aspects, epidemiology, and diagnostic methods applied for felines in Brazil. Cats are infected with the same species of Leishmania found in dogs (i.e., Leishmania infantum). Like dogs, skin lesions are the most common in cats with clinical FeL, mainly affecting the cephalic region and less frequently the legs which may be accompanied by generalized signs or visceral involvement. Information on the immune response of cats to Leishmania infection is scarce; however, efficient infection control is seen in most cases. For diagnosis, generally, the same methods as those in dogs are used, mainly serological tools. But there is a lack of studies focusing the performance of these methods for diagnosing FeL. The estimated overall prevalence of FeL in Brazil is 8%, with L. infantum being the most prevalent species. However, infections with Leishmania braziliensis and Leishmania amazonensis have also been reported. In conclusion, although there has been an increase in the publication related to FeL in Brazil in recent years, there is a lack of research relating immune response and diagnosis of these animals. Cats have been shown to be competent hosts for Leishmania parasites, and their role in the epidemiology of the disease cannot be underestimated, especially in areas of Brazil where the disease is historically endemic.

Anellovirus infections are highly prevalent in mammals, however, prior to this study only a handful of anellovirus genomes had been identified in members of the Felidae family. Here we characterise anelloviruses in pumas (Puma concolor), bobcats (Lynx rufus), Canada lynx (Lynx canadensis), caracals (Caracal caracal) and domestic cats (Felis catus). The complete anellovirus genomes (n = 220) recovered from 149 individuals were diverse. ORF1 protein sequence similarity network analysis coupled with phylogenetic analysis, revealed two distinct clusters that are populated by felid-derived anellovirus sequences, a pattern mirroring that observed for the porcine anelloviruses. Of the two-felid dominant anellovirus groups, one includes sequences from bobcats, pumas, domestic cats and an ocelot, and the other includes sequences from caracals, Canada lynx, domestic cats and pumas. Coinfections of diverse anelloviruses appear to be common among the felids. Human DNA methylation profiles have been used successfully to develop highly accurate biomarkers of aging (“epigenetic clocks”). Although these human epigenetic clocks are not immediately applicable to all species of the animal kingdom, the principles underpinning them appear to be conserved even in animals that are evolutionarily far removed from humans. This is exemplified by recent development of epigenetic clocks for mice and other mammalian species. Here, we describe epigenetic clocks for the domestic cat (Felis catus), based on methylation profiles of CpGs with flanking DNA sequences that are highly conserved between multiple mammalian species. Methylation levels of these CpGs are measured using a custom-designed Infinium array (HorvathMammalMethylChip40). From these, we present 3 epigenetic clocks for cats; of which, one applies only to blood samples from cats, while the remaining two dual-species human-cat clocks apply both to cats and humans. We demonstrate that these domestic cat clocks also lead to high age correlations in cheetahs, tigers, and lions. It is expected that these epigenetic clocks for cats possess the potential to be further developed for monitoring feline health as well as being used for identifying and validating anti-aging interventions. Feral cats (Felis catus) pose a significant threat to wildlife, agriculture, and human health through predation, disease transmission, and competition with native animals. Controlling feral cats and their impacts, however, is challenging. New and emerging 1080-based feral cat baits have shown promising results in western and central Australia; however, the safety of these new baits for nontarget species in eastern Australia, where many native animals are more sensitive to compound 1080 (sodium fluoroacetate) than their western conspecifics, has not been assessed. We investigated the uptake of 499 toxic Eradicat® baits by nontarget animals across five different eastern Australian environs and the uptake of nontoxic Eradicat and Hisstory® baits at an additional two sites. Using field-based observations of species eating or removing baits, we determined that 13 nontarget species (eight mammals, four birds, and one reptile) were at high risk of individual mortality, with individuals of 11 of those 13 species (seven mammals, four birds) observed consuming enough toxic Eradicat in a single visit to ingest a lethal dose of 1080. Feral cats (the target species) consumed only 3.1% of monitored baits, which was only 52% of the 31 baits they encountered. We recommend undertaking targeted population monitoring of species identified at high risk of individual mortality, to determine whether Eradicat baits present a population-level risk to these species. Our findings suggest that the small-sized Eradicat baits present a greater risk to nontarget species in eastern Australia than the larger traditional 1080-based meat baits used for the control of wild dogs and foxes. Our study highlights the importance of performing risk assessments for different bait types, even when the same toxin is used, and of performing site-specific nontarget risk assessments of new baits such as Eradicat to assist developing guidelines for their safe and effective use in different environs. Hypertrophic cardiomyopathy (HCM) is a common and potentially fatal heart disease in many cat breeds. An intronic variant in TNNT2, c.95-108G>A, was recently reported as the cause of HCM in the Maine Coon. The aim of this study was to determine this variant's allele frequency in different populations and its possible association with HCM. Based on 160 Maine Coon samples collected in Belgium, Italy, Sweden and the USA, the variant's allele frequency was estimated to be 0.32. Analysis of the 99 Lives feline whole genome sequencing database showed that the TNNT2 variant also occurs in other breeds, as well as mixed-breed cats. Comparison of 31 affected and 58 healthy cats did not reveal significantly increased odds for HCM in homozygotes. Based on the combined evidence and in agreement with the standards and guidelines for the interpretation of sequence variants, this variant is currently classified as a variant of unknown significance and should not be used for breeding decisions regarding HCM.

Contrafreeloading is the willingness of animals to work for food when equivalent food is freely available. This behavior is observed in laboratory, domesticated, and captive animals. However, previous research found that six laboratory cats failed to contrafreeload. We hypothesized that cats would contrafreeload in the home environment when given a choice between a food puzzle and a tray of similar size and shape. We also hypothesized that more active cats would be more likely to contrafreeload. We assessed the behavior of 17 neutered, indoor domestic cats (Felis catus) when presented with both a food puzzle and a tray across ten 30-min trials. Each cat wore an activity tracker, and all sessions were video recorded. Cats ate more food from the free feed tray than the puzzle (t (16) = 6.77, p N = 4) ate most food offered in the puzzle or showed weak contrafreeloading tendencies (N = 5). Eight cats did not contrafreeload. Cats who consumed more food from the puzzle, consumed more food in general, suggesting a relationship between hunger and effort. Further research is required to understand why domestic cats, unlike other tested species, do not show a strong preference to work for food.

Members of the family Felidae suffer from a wide range of dental, oral and maxillofacial conditions that can cause significant morbidity and mortality. Although many dental, oral and maxillofacial anomalies of the domestic cat (Felis catus) also occur in wild felines, we could find no investigations of these conditions in servals (Leptailurus serval). The objective of this study was to describe the dental, oral and maxillofacial pathology of a wild serval population in South Africa. Detailed extraoral and intraoral examinations and full-mouth dental radiographs on 30 wild servals revealed 14 different dental conditions but no other oral or maxillofacial pathology.

This study shows evidence of a domestic cat (Felis catus) being able to successfully learn to reproduce human-demonstrated actions based on the Do as I Do paradigm. The subject was trained to reproduce a small set of familiar actions on command “Do it!” before the study began. To test feature–contingent behavioural similarity and control for stimulus enhancement, our test consisted of a modified version of the two-action procedure, combined with the Do as I Do paradigm. Instead of showing two different actions on an object to different subjects, we applied a within-subject design and showed the two actions to the same subject in separate trials. We show evidence that a well-socialized companion cat was able to reproduce actions demonstrated by a human model by reproducing two different actions that were demonstrated on the same object. Our experiment provides the first evidence that the Do as I Do paradigm can be applied to cats, suggesting that the ability to recognize behavioural similarity may fall within the range of the socio-cognitive skills of this species. The ability of reproducing the actions of a heterospecific human model in well-socialized cats may pave the way for future studies addressing cats’ imitative skills. In a litter of Turkish Van cats, three out of six kittens developed severe signs of skin disease, diarrhea, and systemic signs of stunted growth at 6 weeks of age. Massive secondary infections of the skin lesions evolved. Histopathological examinations showed a mild to moderate hyperplastic epidermis, covered by a thick layer of laminar to compact, mostly parakeratotic keratin. The dermis was infiltrated with moderate amounts of lymphocytes and plasma cells. Due to the severity of the clinical signs, one affected kitten died and the other two had to be euthanized. We sequenced the genome of one affected kitten and compared the data to 54 control genomes. A search for private variants in the two candidate genes for the observed phenotype, MKLN1 and SLC39A4, revealed a single protein-changing variant, SLC39A4:c.1057G&gt, C or p.Gly353Arg. The solute carrier family 39 member 4 gene (SLC39A4) encodes an intestinal zinc transporter required for the uptake of dietary zinc. The variant is predicted to change a highly conserved glycine residue within the first transmembrane domain, which most likely leads to a loss of function. The genotypes of the index family showed the expected co-segregation with the phenotype and the mutant allele was absent from 173 unrelated control cats. Together with the knowledge on the effects of SLC39A4 variants in other species, these data suggest SLC39A4:c.1057G&gt, C as candidate causative genetic variant for the phenotype in the investigated kittens. In line with the human phenotype, we propose to designate this disease acrodermatitis enteropathica (AE).

We examined whether cats have a cross-modal representation of humans, using a cross-modal expectancy violation paradigm originally used with dogs by Adachi et al. (Anim Cogn 10:17-21, 2007). We compared cats living in houses and in cat cafés to assess the potential effect of postnatal experience. Cats were presented with the face of either their owner or a stranger on a laptop monitor after playing back the voice of one of two people calling the subject's name. In half of the trials the voice and face were of the same person (congruent condition) whereas in the other half of trials the stimuli did not match (incongruent condition). The café cats paid attention to the monitor longer in incongruent than congruent conditions, showing an expectancy violation. By contrast, house cats showed no similar tendency. These results show that at least café cats can predict their owner's face upon hearing the owner's voice, suggesting possession of cross-modal representation of at least one human. There may be a minimal kind or amount of postnatal experiences that lead to formation of a cross-modal representation of a specific person.

Dismembered cats ( Felis catus) have been found in North American schoolyards, parks, walkways, or lawns and sometimes result in local media attention. When a member of the public encounters these cats, they commonly report finding either the cranial or caudal half of a cat in a prominent location. Such findings cause public consternation and pose difficulties to investigators in identifying whether animal abuse has occurred and whether to concentrate resources on the investigation. This report describes necropsy results from 53 cats involved in such instances in the cities of Edmonton and St. Albert, Canada, from 2007 to 2016. We evaluated these results in relation to 2 hypotheses: that the dismembered cats were the result of human activity, or predation and scavenging by coyotes ( Canis latrans). The main postmortem features were canine tooth wounds in the neck accompanied by tearing of the subcutaneous structures, skin avulsion, broken claws, and removal of internal organs with the colon and intestine attached to and trailing from the carcass. Based on the nature of the lesions, along with other circumstances of the deaths of the cats in this study, we concluded that these dismembered cat remains resulted from coyote predation on living cats and scavenging of the bodies of cats that died of other causes. We offer additional information to assist veterinarians, veterinary pathologists, and civic officials in identifying the probable cause of death for cat carcasses provided by members of the public.

The aim of the present study was to assess the concentrations of lead (Pb) and cadmium (Cd) in the liver and kidney from stray dogs (Canis lupus familiaris) and cats (Felis catus) found dead between 2014 and 2017 in the city of Naples and its surrounding areas. These organs from 290 dogs and 88 cats were collected after ordinary necropsy of stray animals. Heavy metal concentrations were determined by using atomic absorption spectrophotometry (AAS). Concentrations of Pb (up to 5.93 mg/kg) and Cd (ranging from 0.005 to 6.13 mg/kg) were detected in both livers and kidneys analyzed. Differences in concentration were found based on age class, gender, and kind of tissue for both elements, with a trend similar to those already reported in the literature for comparable studies from different countries. Kidney diseases are the most common illness for cats with a prevalence seven times higher than in dogs. Metanephros is the last of three renal systems to be formed during the embryonic period, which then becomes the permanent kidney. The current work aimed to analyse the morphology and to quantify the structures present in the development of metanephros from domestic cat (Felis catus) embryos and foetuses. For this purpose, the evaluation of the biometric parameters of metanephros from cat embryos and foetuses was performed in addition to the quantification of renal corpuscles and volume of cortical and medullary layers by stereological analysis. The evaluated biometric parameters were weight, width, height, thickness and volume. The values of the measured biometric parameters increased throughout the gestational stages. The quantity of renal corpuscles gradually increased following the embryo-foetal development, mainly during the middle of the gestational stage. It was during this phase that morphologically, a complete corticomedullary division was observed. Although the difference in the quantity of renal corpuscles between the middle and the end of the gestational stages was not statistically significant, there was an increase in the volume of the medullary layer and a decrease in the volume of the cortical layer between these two stages. These findings suggest that the metanephros presents a progressive growth with the renal corpuscles following this development until the middle of the gestational stage. Starting from this phase, the differentiation of the corticomedullary layers can be seen with a significant increase in the medullary layer.

Domestication is generally assumed to have resulted in enhanced communication abilities between non-primate mammals and humans, although the number of species studied is very limited (e.g. cats, Felis catus ; dogs, Canis familiaris ; wolves, Canis lupus ; goats, Capra hircus ; horses, Equus caballus ). In species without hands for pointing, gazing at humans when dealing with inaccessible food during an unsolvable task, and in particular gaze alternations between a human and the unsolvable task (considered forms of showing), are often interpreted as attempts at referential intentional communication. We report that kangaroos, marsupial mammals that have never been domesticated, actively gazed at an experimenter during an unsolvable problem task (10/11 kangaroos tested), thus challenging the notion that this behaviour results from domestication. Nine of the 10 kangaroos additionally showed gaze alternations between the unsolvable task and experimenter. We propose that the potential occurrence of these behaviours displayed towards humans has been underestimated, owing to a narrow focus on domestic animals, as well as a more general eutherian research bias.

Toxoplasma gondii is a worldwide-distributed protozoon that infects warm-blooded animals. We determined T. gondii exposure in one marine otter (Lontra felina) and four domestic cats (Felis catus) along the northern and central coast of Chile, indicating a low seroprevalence of 5% (1/19) and 8% (4/50) respectively, likely related to arid environment.

Cottontail rabbits (Sylvilagus spp.) are frequently admitted to wildlife rehabilitation facilities due to predation by domestic cats (Felis catus). Our retrospective study (2015-19) of three species (Sylvilagus audubonii, Sylvilagus floridanus, and Sylvilagus nuttallii) indicated that once juveniles reached a weight over 220 g, they were unlikely to present due to domestic cat interactions. This information should be incorporated into release criteria for these species.

Two domestic cats from the Patagonia rural area in Argentina were found to be naturally infected with Echinococcus granulosus sensu stricto/G1 genotype; so far, the only species/genotype of E. granulosus sensu lato complex described to infect domestic cats. The felines developed abdominal disseminated larval disease; the diagnosis was performed by ultrasound, exploratory laparotomy, and molecular techniques. These results indicate that cystic echinococcosis must be considered for differential diagnosis of felines with abdominal distension and/or observation of vesicles through ultrasound, from endemic areas. Even though cats and dogs are carnivores, differences in digestive physiology and immunological characteristics between them could allow the development of larval or adult worm parasites. Domestic cats with cystic echinococcosis show to be environmentally infected with E. granulosus s. s./G1 eggs.

Summary Predation by domestic cats Felis catus can be a threat to biodiversity conservation,1–3 but its mitigation is controversial.4 Confinement and collar-mounted devices can impede cat hunting success and reduce numbers of animals killed,5 but some owners do not wish to inhibit what they see as natural behavior, perceive safety risks associated with collars, or are concerned about device loss and ineffectiveness.6,7 In a controlled and replicated trial, we tested novel, non-invasive interventions that aim to make positive contributions to cat husbandry, alongside existing devices that impede hunting. Households where a high meat protein, grain-free food was provided, and households where 5–10 min of daily object play was introduced, recorded decreases of 36% and 25%, respectively, in numbers of animals captured and brought home by cats, relative to controls and the pre-treatment period. Introduction of puzzle feeders increased numbers by 33%. Fitting Birdsbesafe collar covers reduced the numbers of birds captured and brought home by 42% but had no discernible effect on mammals. Cat bells had no discernible effect. Reductions in predation can be made by non-invasive, positive contributions to cat nutrition and behavior that reduce their tendency to hunt, rather than impede their hunting. These measures are likely to find support among cat owners who are concerned about the welfare implications of other interventions.

The development of assisted reproduction techniques (ART) specifically for felids has been propagated for two main reasons: (i) most felids are threatened and faced with extinction in all or part of their native habitats (IUCN Red List of Threatened Species, www.catsg.org), and (ii) the domestic cat (Felis catus) can serve as a research model for the implementation of advanced assisted reproductive techniques (ART) to be applied in exotic cats. Domestic cat ovaries can be freshly obtained from veterinary clinics and are frequently used for research on preservation of genetic resources in feline species. The presented review will summarize recent advances and obstacles in biobanking of female genetic resources and discuss alternative approaches which are under investigation.

The ovaries from 23 queens were collected after elective ovariohysterectomy at different stages of the estrous cycle and were classified according to the season upon collection (spring/summer vs. fall/winter). Based on the findings, we concluded that queens in tropical regions experience no seasonal influence on oocyte quality, but there is a minimal effect on ovarian morphology, with an increase in ovarian length during proestrus/estrus in the spring/summer period. A variety of cat breeds have been developed via novelty selection on aesthetic, dermatological traits, such as coat colors and fur types. A recently developed breed, the lykoi (a.k.a. werewolf cat), was bred from cats with a sparse hair coat with roaning, implying full color and all white hairs. The lykoi phenotype is a form of hypotrichia, presenting as a significant reduction in the average numbers of follicles per hair follicle group as compared to domestic shorthair cats, a mild to severe perifollicular to mural lymphocytic infiltration in 77% of observed hair follicle groups, and the follicles are often miniaturized, dilated, and dysplastic. Whole genome sequencing was conducted on a single lykoi cat that was a cross between two independently ascertained lineages. Comparison to the 99 Lives dataset of 194 non‐lykoi cats suggested two variants in the cat homolog for Hairless (HR) (HR lysine demethylase and nuclear receptor corepressor) as candidate causal gene variants. The lykoi cat was a compound heterozygote for two loss of function variants in HR, an exon 3 c.1255_1256dupGT (chrB1:36040783), which should produce a stop codon at amino acid 420 (p.Gln420Serfs*100) and, an exon 18 c.3389insGACA (chrB1:36051555), which should produce a stop codon at amino acid position 1130 (p.Ser1130Argfs*29). Ascertainment of 14 additional cats from founder lineages from Canada, France and different areas of the USA identified four additional loss of function HR variants likely causing the highly similar phenotypic hair coat across the diverse cats. The novel variants in HR for cat hypotrichia can now be established between minor differences in the phenotypic presentations.

Reducing the impacts of invasive predators is a key objective for conservation managers, livestock producers and human health agencies globally. The efficacy of invasive predator control programs, however, is highly variable. To improve control efficacy, managers require a fundamental understanding of the factors that contribute to the success or failure of a control program. Using a predator baiting program as a case study, we measured the efficacy of baiting as a control tool to significantly reduce feral cat (Felis catus) populations. We used camera traps and cat-borne GPS collars to monitor changes in feral cat populations at a baited site and an unbaited site, using a Before-After, Control-Impact (BACI) design. We also identified five key elements required for a successful baiting program (bait encounter rate, availability, attractiveness, palatability and lethality) and simultaneously measured these to identify areas for potential improvement. Baiting was ineffective at reducing feral cat populations; collared cat mortality was only 11% (1/9), with camera traps revealing negligible reductions in the number of cat detection events (9%), naive occupancy (15%), and no significant change in the relative abundance of feral cats (F1,54 = 0.8641, P = 0.357). Several factors contributed to the poor control efficacy. Bait encounter rates were low, with cats active along tracks (where baits were laid) 5 kg) feral cat. Our findings suggest that altering bait deployment patterns, increasing bait densities and improving bait palatability could potentially improve the efficacy of baiting programs to reduce feral cat populations. Our study provides a framework to measure and evaluate the key elements that contribute to efficacy of pest control programs, and to identify opportunities for improving outcomes of future control programs.

A variety of cat breeds have been developed via novelty selection on aesthetic, dermatological traits, such as coat colors and fur types. A recently developed breed, the lykoi (a.k.a. werewolf cat), was bred from cats with a sparse hair coat with roaning, implying full color and all white hairs. The lykoi phenotype is a form of hypotrichia, presenting as a significant reduction in the average numbers of follicles per hair follicle group as compared to domestic shorthair cats, a mild to severe perifollicular to mural lymphocytic infiltration in 77% of observed hair follicle groups, and the follicles are often miniaturized, dilated, and dysplastic. Whole genome sequencing was conducted on a single lykoi cat that was a cross between two independently ascertained lineages. Comparison to the 99 Lives dataset of 194 non-lykoi cats suggested two variants in the cat homolog for Hairless (HR) (HR lysine demethylase and nuclear receptor corepressor) as candidate causal gene variants. The lykoi cat was a compound heterozygote for two loss of function variants in HR, an exon 3 c.1255_1256dupGT (chrB1:36040783), which should produce a stop codon at amino acid 420 (p.Gln420Serfs*100) and, an exon 18 c.3389insGACA (chrB1:36051555), which should produce a stop codon at amino acid position 1130 (p.Ser1130Argfs*29). Ascertainment of 14 additional cats from founder lineages from Canada, France and different areas of the USA identified four additional loss of function HR variants likely causing the highly similar phenotypic hair coat across the diverse cats. The novel variants in HR for cat hypotrichia can now be established between minor differences in the phenotypic presentations.

International audience; In the feline Donskoy breed, a phenotype that breeders call "pink‐eye," with associated light‐brown skin, yellow irises and red‐eye effect, has been described. Genealogical data indicated an autosomal recessive inheritance pattern. A single candidate region was identified by genome‐wide association study and SNP‐based homozygosity mapping. Within that region, we further identified HPS5 (HPS5 Biogenesis Of Lysosomal Organelles Complex 2 Subunit 2 ) as a strong candidate gene, since HPS5 variants have been identified in humans and animals with Hermansky–Pudlak syndrome 5 or oculocutaneous albinism. A homozygous c.2571‐1G>A acceptor splice‐site variant located in intron 16 of HPS5 was identified in pink‐eye cats. Segregation of the variant was 100% consistent with the inheritance pattern. Genotyping of 170 cats from 19 breeds failed to identify a single carrier in non‐Donskoy cats. The c.2571‐1G>A variant leads to HPS5 exon‐16 splicing that is predicted to produce a 52 amino acids in‐frame deletion in the protein. These results support an association of the pink‐eye phenotype with the c.2571‐1G>A variant. The pink‐eye Donskoy cat extends the panel of reported HPS5 variants and offers an opportunity for in‐depth exploration of the phenotypic consequences of a new HPS5 variant.

The aim of this study was to evaluate the applicability of the Cryotech technique for the vitrification of domestic cat (Felis catus) oocytes, as a model for other feline species threatened with extinction. This technique, in which oocytes are stored in a minimal volume of medium, is already widely used in human assisted reproductive technology. In the first part of this study, a viability test (EtBr/FDA) was used to evaluate the toxicity of the vitrification media (solutions). After IVM, oocytes were placed in vitrification and warming solutions according to the manufacturer's procedure, with or without exposure to liquid nitrogen. The solutions and the vitrification procedure each caused a reduction in oocyte viability, with survival rates of 71.4% in oocytes exposed to the Cryotech media (without cooling in liquid nitrogen), and 62% in oocytes that were vitrified. In the second part of the experiment, parthenogenetic activation was used to evaluate the developmental potential of oocytes previously vitrified using the Cryotech method. After warming, the oocytes were activated using a combination of 0.7 µM ionomycin in TCM 199 medium (5 min) followed by 2 mM 6-DMAP in TCM 199 supplemented with 10% FBS (3 hr), then cultured and evaluated every 24 hr for parthenogenetic cleavage. In the experimental group, 23/50 (46%) cleaved embryos were obtained. Domestic cat oocytes, vitrified by the Cryotech method, are characterized by high survival rates. However, it is necessary to improve the technique to increase the developmental competence of embryos obtained from vitrified oocytes.

Background Munchkin cats were founded on a naturally occurring mutation segregating into long-legged and short-legged types. Short-legged cats showed disproportionate dwarfism (chondrodysplasia) in which all four legs are short and are referred as standard Munchkin cats. Long-legged animals are referred as non-standard Munchkin cats. A previous study using genome-wide single nucleotide polymorphisms (SNPs) for genome-wide association analysis identified a significantly associated region at 168–184 Mb on feline chromosome (FCA) B1. Results In this study, we validated the critical region on FCA B1 using a case-control study with 89 cats and 14 FCA B1-SNPs. A structural variant within UGDH (NC_018726.2:g.173294289_173297592delins108, Felis catus 8.0, equivalent to NC_018726.3:g.174882895_174886198delins108, Felis catus 9.0) on FCA B1 was perfectly associated with the phenotype of short-legged standard Munchkin cats. Conclusion This UGDH structural variant very likely causes the chondrodysplastic (standard) phenotype in Munchkin cats. The lack of homozygous mutant phenotypes and reduced litter sizes in standard Munchkin cats suggest an autosomal recessive lethal trait in the homozygote state. We propose an autosomal dominant mode of inheritance for the chondrodysplastic condition in Munchkin cats.

After complete spinal cord injury, mammals, including mice, rats and cats, recover hindlimb locomotion with treadmill training. The premise is that sensory cues consistent with locomotion reorganize spinal sensorimotor circuits. Here, we show that hindlimb standing and locomotion recover after spinal transection in cats without task-specific training. Spinal-transected cats recovered full weight bearing standing and locomotion after five weeks of rhythmic manual stimulation of triceps surae muscles (non-specific training) and without any intervention. Moreover, cats modulated locomotor speed and performed split-belt locomotion six weeks after spinal transection, functions that were not trained or tested in the weeks prior. This indicates that spinal networks controlling standing and locomotion and their interactions with sensory feedback from the limbs remain largely intact after complete spinal cord injury. We conclude that standing and locomotor recovery is due to the return of neuronal excitability within spinal sensorimotor circuits that do not require task-specific activity-dependent plasticity.

In many mammals, gender and skull shape are related to the bone morphology of the entire body; however, this has not been well established in the domestic cat (Felis catus). This study aims to find a relationship between cervical, thoracic and lumbar vertebrae morphometrics with regard to the sex and skull shape of domestic cats. A total of 79 samples obtained from 92 dried bones of domestic cats were used to determine morphometric measurements for a total of 29 parameters. Hierarchical clustering was used to cluster the vertebral bones found in three groups: C3-T1, T2-T11 and T12-L7. The skull shape identification process employed discriminative analysis and revealed the highest training data accuracy rate at up to 86.20% in T4 followed by L1 (86.04%) Axis (85.71%) and C5 (85.18%). Sex identification employed discriminative analysis and displayed the highest training data accuracy rate at up to 75.58% in L1 followed by, T7 (71.87%) and C6 (71.79%). Moreover, we found that 14% of the samples had one vertebra missing (T13 or L1). In conclusion, domestic cat vertebral morphometrics were found to be more related to skull shape than gender. In addition, bone clustering employed morphometric data and yielded a result that was similar to that of traditional cluster analysis involving body regions.

The domestic cat (Felis catus) was used as a sentinel of exposure to polychlorobiphenyls (PCBs) in indoor urban environments y. Sera from 120 cats were pooled to form 30 different groups selected by age ( 2 ≤ 8 years; > 8 years), sex, municipality (Bologna and Turin) and environment (indoor vs. outdoor). Test portions of 1 mL were analyzed by means of gas chromatography coupled to high-resolution mass spectrometry (GC-HRMS) for six selected indicators non-dioxin-like PCBs (∑6 PCBs: congeners #28, #52, #101, #132, #153 and #180) and the results were computed in the upper-bound mode. The internal dose of PCBs attributable to the cats' alimentary lipid intake ranged from 32.4 to 1,446 ng/g (P50 165; mean 258). The Wilcoxon test revealed significantly lower PCB burden in “outdoor” groups than in “indoor” groups. Age correlated well with the heptachlorinated and most bio-accumulative congener, PCB #180, and slightly with hexachlorinated PCBs #138 and #153. Contamination attributable to house dust collected in 15 living-rooms ranged from 10.0 to 279 ng/g dry weight (P50 97.4; mean 94.4). Exposure estimates indicated a 0.6–16 ng/kg bw range of daily ∑6 PCB intake from a default value of 200 mg/cat of dust ingestion. The intake of PCBs due to dust ingestion fell within the same order of magnitude as that computed from a 60 g daily intake of commercial dry pet foods, while inhalation accounted for 0.21–8.2 ng/kg bw/day, on setting the nominal ∑6 PCB contamination in outdoor and indoor air at 0.37 and 15 ng/m3, respectively. Italian indoor cats could be exposed to higher levels of ∑6 PCBs than the Reference Dose (RfD) of 10 ng/kg/bw/day; this supports the World Health Organization's statement that the quality of the indoor environment is a major determinant of health.

Bu nedenle, bu makalede, hem bu konuda çalışanları, hem de veteriner hekimleri bilgilendirmek amacıyla F. subrostratushakkında ayrıntılı bilgi verilmiştir., We present this case report to provide information about Felicola subrostratus (Burmeister, 1838) (Phthiraptera: Ischnocera) detected on a2-year-old stray cat (Felis catus) brought for treatment to a veterinary clinic in Kocaeli, Turkey. Macroscopically, severe louse infestation wasdetected on the cat. Some of the lice on the cat were collected using forceps and preserved in 70% ethanol. Then, they were sent to Departmentof Parasitology, Veterinary Faculty, Selçuk University in Konya for identification. Samples were examined under a microscope andidentified as F. subrostratus (Burmeister, 1838). F. subrostratus has been known to be detected on cats in Turkey, but no study is found aboutthis species. For this reason, in this case report, F. subrostratus was studied to further educate veterinarians and parasitologists in the country.

We aimed to determine the profile of IgE reactivity to three major cat allergens, Fel d 1, Fel d 2 and Fel d 4, in cat-allergic patients in the Moscow region in Russia. sIgE levels to recombinant proteins expressed in Escherichia coli (Fel d 1 and Fel d 4) and to Fel d 2 protein purified from cat serum were measured using a microarray method developed in our laboratory. Sera from 174 anonymous subjects with a positive reaction (≥0.35 IU/mL) to cat dander extract (e1, ImmunoCAP) and 56 negative controls were used for IgE testing. Fel d 1 was recognized by 92.5%, Fel d 2 by 29.9% and Fel d 4 by 39.1% of the tested patient sera. The sensitivity to these three proteins was approximately 98% compared to cat dander extract (correlation coefficient to ImmunoCAP is 0.94 with PPV = 0.99 and NPV = 0.95). These predictive values appeared to be even more statistically significant than the divergence between the ISAC IgE test and the extract-based singleplex ImmunoCAP. The combination of the three investigated proteins (Fel d 1, Fel d 2 and Fel d 4) is suitable for in vitro molecular (serological) diagnosis of cat allergy in this region as a complement to cat dander extract. Moreover, with this method, we found distinction between Fel d 2 and other Feline sIgEs formation.

Owners of free-ranging domestic cats (Felis catus) are under increasing pressure to keep their pet contained within their house or yard, in an effort to reduce adverse impacts on cat welfare, ecosystem biodiversity and neighbourhoods. We conducted a randomised online experiment to assess the effectiveness of two persuasive messages to encourage cat owners to contain their pets. A total of 512 Australian cat owners, who currently do not contain their cats, were randomly assigned to view one of three short video messages: one framed to highlight the negative impact of cats' on wildlife and biodiversity ('wildlife protection' frame), one framed to highlight the health and safety benefits of keeping cats contained ('cat benefit' frame), and a control message focused on general information about cats ('neutral' frame). We assessed the impact of these video messages on two post-treatment outcome variables: (1) the intention of owners to contain their cat; and (2) the adoption of containment practices, based on a 4-week follow-up survey. Mediation analysis revealed both the 'wildlife protection' and 'cat benefit' messages increased owners' motivation to contain their cat and their beliefs that they could effectively contain their cat to achieve the desired outcomes (response efficacy). In turn, higher levels of motivation and response efficacy predicted increased cat containment intentions and increased adoption of cat containment. In addition, the response efficacy effects of the 'cat benefit' message were strengthened by the cat owner's bond to their pet, suggesting audience segmentation may improve the effectiveness of interventions. Implications for future intervention development are discussed.

The authors analyzed the daily activity rhythms of the domestic cat and of eight of the ten feral felid species that are indigenous to South America. All species showed daily rhythmicity of activity in captivity under a natural light-dark cycle. The robustness of the rhythmicity varied from species to species, but the grand mean of 34% was within the range of robustness previously described for mammalian species ranging in size from mice to cattle. There was not a sharp division between diurnal and nocturnal felids. Instead, what was found was a gradient of diurnality going from the predominantly nocturnal margay (72% of activity counts during the night) to the predominantly diurnal jaguarundi (87% of activity counts during the day) with the remaining species lying in between these two extremes. The ecological implications of temporal niche variations are discussed.
