Tri-n-butyl phosphate (TnBP) is a widely used organophosphate ester, but its effects on the regenerative process under damaging circumstances remain unknown. In the present study, zebrafish larvae were exposed to 0, 50, 100, 200 and 1000 μg/L TnBP, and the caudal fins were cut at 72 hours post fertilization (hpf). First, after exposure to TnBP, the number of total neutrophils decreased together with decreased neutrophils in the tail, and TnBP inhibited chemotaxis. Second, reactive oxygen species (ROS) levels in the zebrafish decreased greatly. Following exposure to TnBP, transcription levels of many genes regulating fin regeneration, such as fgf20a, fgfr1a, bmp2a and bmp4, were significantly downregulated, while inflammatory factors such as cxcl8a, cxcl18b, il-6, and tnfa were abnormally upregulated. In addition, TnBP inhibited the regenerative area after caudal fin amputation. The inflammatory state was adverse during the regenerative process. In summary, TnBP exposure is immunotoxic and decreases oxidative stress in injured zebrafish larvae.

In this work, the electrospun polyacrylonitrile/covalent organic frameworks Tp-BD nanofibers (PAN/Tp-BD) were synthesized and applied as an adsorbent for thin film microextraction (TFME) of seven sulfonamides in animal derived food samples. The morphology, structure, porosity, and stability of the prepared nanofibers were investigated. The PAN/Tp-BD nanofibers exhibited good chemical stability, high flexibility, porous fibrous structure, and excellent extraction efficiency. Based on the PAN/Tp-BD nanofibers as the adsorbent, a thin film microextraction-high performance liquid chromatography (TFME-HPLC) method for the determination of seven sulfonamides (SAs) in food samples was developed. In this study, we present a general method to prepare the core-shell magnetic molecularly imprinted polymers (MIPs) nanoparticles (NPs) for sulfamethazine (SMZ). The resulting Fe₃O₄@MIPs NPs possess a highly improved imprinting effect, fast adsorption kinetics and high adsorption capacity, and can be applied to extract sulfonamide in the poultry feed. In this protocol, the magnetite NPs were synthesized by co-precipitating Fe²⁺ and Fe³⁺ in an ammonia solution first. Silica was then coated on the Fe₃O₄ NPs using a sol-gel method to obtain silica shell magnetic NPs. Subsequently, the vinyl groups were grated onto silica-modified Fe₃O₄ surface by 3-methacryloyloxypropyltrimethoxysilane. Finally, the MIPs films were formed on the surface of Fe₃O₄@SiO₂ by the copolymerization of vinyl end groups with functional monomer, methacrylic acid, cross-linking agent, ethylene glycol dimethacrylate, the initiator azo-bis-isobutyronitrile and template molecule, sulfamethazine. The morphology, magnetic, adsorption and recognition properties of Fe₃O₄@MIPs NPs were characterized using transmission electron microscope (TEM), scanning electron microscope (SEM), Fourier transform infrared (FT-IR) spectrometer, vibrating sample magnetometer (VSM) and re-binding experiments. The results showed that the binding sites of Fe₃O₄@MIPs were good accessibility, fast adsorption rate and the maximum adsorption capacity of Fe₃O₄@MIPs to SMZ was 344.8 μg g⁻¹. The selectivity of the obtained Fe₃O₄@MIPs NPs were elucidated by the different rebinding capability of SMZ and structural related sulfonamides in the mixed solution. The results indicated that the Fe₃O₄@MIPs had high imprinting factor 9.5 and significant selectivity. A method was developed for enrichment and determination of SMZ in the poultry feed samples with recoveries of duck and chicken feed ranging from 63.3 to 76.5% and 68.7 to 74.7%, respectively and the relative standard deviations (RSD) (6.7%).

A novel molecularly imprinted polymer monolith was prepared by the room temperature ionic liquid-mediated in situ molecular imprinting technique, using norfloxacin (NOR) as the template, methacrylic acid as the functional monomer, ethylene glycol dimethacrylate as the cross-linker. The optimal synthesis conditions and recognition properties of NOR-imprinted monolithic column were investigated. The results indicated that the imprinted monoliths exhibited good ability of selective recognition against the template and its structural analog. Using the fabricated material as solid-phase extraction sorbent, a sample pre-treatment procedure of molecularly imprinted solid-phase extraction coupling with HPLC was developed for determination of trace quinolone residues in animal tissues samples. The recoveries ranging from 78.16 to 93.50% for eight quinolones antibiotics such as marbofloxacin, NOR, ciprofloxacin, danofloxacin, difloxacin, oxolinic acid, flumequine and enrofloxacin were obtained.

The synthesis of poly-aminophenylboronic acid (APBA) imprinted hybrid silica-based polymers for selective recognition of bovine hemoglobin (BHb) was described, where the mesoporous hybrid silica supporting matrices were prepared by a mild sol-gel process with tetraethoxysilane and 3-aminopropyltriethoxysilane as two precursors. Covalent immobilization of BHb was adopted in order to create homogeneous recognition sites. After removal of the template, the resulting imprinted polymers showed high binding affinity toward BHb and the imprinting factor (alpha) reached 2.12. The specificity of the BHb recognition was evaluated with competitive experiments, indicating the imprinted polymers have a higher selectivity for the template BHb. The easy preparation protocol and good protein recognition properties made the approach an attractive solution to depletion of high-abundance protein from bovine blood.

For reasons of animal welfare and better science, a series of in vitro methods, including assays based isolated eye or cornea, Chorioallantoic membrane, cultured tissue or cell, had been established and evaluated to replace the Draize eye irritation test. This paper provides a brief review of the recent progress in the in vitro alternatives to Draize eye irritation test, and the principles of scientific validation of these alternatives were also discussed.
