Arctic species encounter multiple stressors including climate change and environmental contaminants. Some contaminants may disrupt hormones that govern the behavioural responses of wildlife to climatic variation, and thus the capacity of species to respond to climate change. We investigated correlative interactions between legacy and emerging persistent organic pollutants (POPs), mercury (Hg), hormones and behaviours, in thick-billed murres (Uria lomvia) (N = 163) breeding in northern Hudson Bay (2016-2018). The blood profile of the murres was dominated by methylmercury (MeHg), followed by much lower levels of sum (∑) 35 polychlorinated biphenyls (PCBs), hexachlorobenzene (HCB) and p,p'-dichlorodiphenyltrichloroethylene (DDE), polybrominated diphenyl ethers (PBDEs) BDE-47, -99 and BDE-100; all other measured organochlorine pesticides and replacement brominated flame retardants had low concentrations if detected. Inter-annual variations occurred in MeHg, circulating triiodothyronine (T3), thyroxine (T4), and the foraging behaviours of the murres, identified using GPS-accelerometers. Compared to the 50-year mean date (1971-2021) for 50% of sea-ice coverage in Hudson Bay, sea-ice breakup was 1-2 weeks earlier (2016, 2017) or comparable (2018). Indeed, 2017 was the earliest year on record. Consistent with relationships identified individually between MeHg and total T3, and T3 and foraging behaviour, a direct interaction between these three parameters was evident when all possible interactions among measured chemical pollutants, hormones, and behaviours of the murres were considered collectively (path analysis). When murres were likely already stressed due to early sea-ice breakup (2016, 2017), blood MeHg influenced circulating T3 that in turn reduced foraging time underwater. We conclude that when sea-ice breaks up early in the breeding season, Hg may interfere with the ability of murres to adjust their foraging behaviour via T3 in relation to variation in sea-ice.

Avian populations must mount effective immune responses upon exposure to environmental stressors such as avian influenza and xenobiotics. Although multiple immune assays have been tested and applied to various avian species, antibody-mediated immune responses in non-model avian species are not commonly reported due to the lack of commercially available species-specific antibodies. The objectives of the present study were to advance methods for studying wild bird immune responses and to apply these to the evaluation of cytological responses after exposure of American kestrels, Falco sparverius, to a commercial flame retardant mixture containing isopropylated triarylphosphate isomers (ITP). Hatchlings were gavaged daily with safflower oil or 1.5 ug/g bw/day of ITP suspended in safflower oil, then bled on days 9, 17, and 21. The ITP treatment group (n = 18) and a subset of controls (Poly I:C treatment group; n = 10) were injected on days 9 and 15 with a synthetic analog of viral double-stranded RNA, polyinosinic:polycytidylic acid (Poly I:C), a toll-like receptor ligand and synthetic viral mimic, and responses compared to a sham injected control group (n = 8). The hypotheses tested whether kestrels showed immunological differences among treatment groups, genetic sex, and/or white blood cell (WBC) subpopulation type over time. A flow cytometry (FCM) gating strategy categorized heterophils (H), lymphocytes (L), and monocytes (M) and their proportions, and measured relative fluorescence in response to anti-chicken CD4 binding. Fluorescent cell surfaces and some granular/vacuolar inclusions were visualized by epifluorescence microscopy. A fourth subpopulation with higher levels of granularity than M but less than H became increasingly apparent with time and was gated along with the H subpopulation; its frequency of occurrence was lowest in the ITP group (P = 0.0023). The percentages of cells differed among treatment groups, days, and sexes (P = 0.0001). For both sexes, percentages of H and L were higher than M in control and Poly I:C. In the ITP group, L percentages were higher than H and M (P = 0.0457), and H and L were higher than M on days 9 and 21 (P = 0.0001). The ratios of H:L and H:WBC, indicators of robust immunity, were also higher on days 9 and 21 than on 17 (P = 0.0079). For each sex, the highest levels of activity measured by FCM geometric means (GEO) of fluorescence (indicative of antibody binding) were observed on day 9 (P = 0.0001 female, and P = 0.0011 male) in H over both L and M (P

The toxicokinetics of triphenyl phosphate (TPHP) in vivo including the uptake, deposition, and biotransformation into the metabolite diphenyl phosphate (DPHP) is presently reported in embryonated eggs and chicks of Japanese quail. Quail were dosed with TPHP at 3 concentrations by air cell egg injection on embryonic day 0, followed by daily oral dosing after chicks hatched (5 d). Vehicle-only exposed controls were also used. In dosed eggs, only 33% of the TPHP remained 2 d after injection (no hepatic development); after 10 d (post-hepatogenesis), only 2% remained. The estimated TPHP half-lives in the eggs ranged from 1.1 to 1.8 d for the 3 dosed groups. In all exposed eggs and chicks, DPHP significantly increased with dose (0.001 < p < 0.044). It appears that DPHP is an important metabolite in quail, making up 41 to 74% of all metabolites formed in embryonated eggs. In chicks, at medium and high doses, DPHP concentrations significantly exceeded those of TPHP (p ≤ 0.007), making up 67 and 76% of the total burden, respectively. Our findings suggest that rapid TPHP metabolism occurred in chicks and embryonated quail eggs but that this may vary with the age of the embryonated egg and the stage of embryo development, which should be considered when evaluating concentrations of TPHP and DPHP measured in eggs of wild birds. Tetrabromobisphenol A bis(2,3-dibromopropyl) ether (TBBPA-BDBPE) is an additive flame retardant used in polyolefins and polymers. It has been detected in biota, including in avian eggs, yet little is known of its effects. We assessed the pattern of TBBPA-BDBPE concentrations in songbird eggs over the incubation period, and the effects of embryonic exposure to TBBPA-BDBPE in a model songbird species, the zebra finch (Taeniopygia guttata). To assess concentrations during embryo development, eggs were injected on the day they were laid with the vehicle control (safflower oil) or 100 ng TBBPA-BDBPE/g egg, and whole egg contents were collected throughout embryonic development on day 0 (unincubated), 5, 10 and 13. To evaluate effects of embryonic exposure to TBBPA-BDBPE, eggs were injected at Hamburger-Hamilton stage 18 (∼80 h after initiation of incubation) with safflower oil only, 10, 50 or 100 ng TBBPA-BDBPE/g egg (albumin injection volume 1 μl/g). Eggs were monitored for hatching success, and nestlings were monitored for growth and survival. At 15 days post-hatch, tissues were collected to assess physiological effects. TBBPA-BDBPE was incorporated into the egg as the embryo developed, and concentrations started declining in late incubation, suggesting biotransformation by the embryo. There were no effects on hatching success, nestling survival, growth, organ somatic indices, or thyroid hormone homeostasis; however, there was evidence that body condition declined in a dose-dependent manner towards the end of the rapid nestling growth phase. This decreased body condition could be a delayed effect of early developmental exposure, or it may be the result of increased exposure to biotransformation products of TBBPA-BDBPE produced over the nestling period, which are predicted to be more bioaccumulative and toxic than the parent compound.

Global climate change has led to profound alterations of the Arctic environment and ecosystems, with potential secondary effects on mercury (Hg) within Arctic biota. This review presents the current scientific evidence for impacts of direct physical climate change and indirect ecosystem change on Hg exposure and accumulation in Arctic terrestrial, freshwater, and marine organisms. As the marine environment is elevated in Hg compared to the terrestrial environment, terrestrial herbivores that now exploit coastal/marine foods when terrestrial plants are iced over may be exposed to higher Hg concentrations. Conversely, certain populations of predators, including Arctic foxes and polar bears, have shown lower Hg concentrations related to reduced sea ice-based foraging and increased land-based foraging. How climate change influences Hg in Arctic freshwater fishes is not clear, but for lacustrine populations it may depend on lake-specific conditions, including interrelated alterations in lake ice duration, turbidity, food web length and energy sources (benthic to pelagic), and growth dilution. In several marine mammal and seabird species, tissue Hg concentrations have shown correlations with climate and weather variables, including climate oscillation indices and sea ice trends; these findings suggest that wind, precipitation, and cryosphere changes that alter Hg transport and deposition are impacting Hg concentrations in Arctic marine organisms. Ecological changes, including northward range shifts of sub-Arctic species and altered body condition, have also been shown to affect Hg levels in some populations of Arctic marine species. Given the limited number of populations and species studied to date, especially within Arctic terrestrial and freshwater systems, further research is needed on climate-driven processes influencing Hg concentrations in Arctic ecosystems and their net effects. Long-term pan-Arctic monitoring programs should consider ancillary datasets on climate, weather, organism ecology and physiology to improve interpretation of spatial variation and time trends of Hg in Arctic biota.

Arctic marine ecosystems are experiencing rapid change, such as ocean warming and enhanced pollutants. Perfluoroalkyl acids (PFAAs) arriving via long-range transport have been detected in Arctic wildlife, including seabirds which are considered sentinels of marine ecosystem health. There is evidence that PFAA exposure leads to the disruption of thyroid hormones (THs), such as thyroxine (T4) and triiodothyronine (T3), which play important roles in metabolism, incubation, and thermoregulation in seabirds. Here, we investigated relationships between PFAAs and THs [total T4 (TT4), free T4 (FT4), total T3 (TT3) and free T3 (FT3)] in blood plasma collected from 63 thick-billed murres (Uria lomvia) at a colony located in northern Hudson Bay (2016-2018). We then tested if PFAAs and TH levels were related to fitness-associated reproductive traits, such as body mass and hatch dates. PFUdA, PFOS, and PFTrDA were the dominant PFAAs in murre blood, accounting for approximately 77% of ∑PFAA. Females had higher PFAAs than males, possibly due to higher trophic feeding. While FT3 increased with PFOS, PFNA, PFDA, PFDoA, PFTeDA, ∑PFCA7, and ∑PFAA in murres, TT3 decreased with PFOS, PFDoA, and PFTeDA in males, but not females, suggesting thyroid disruption. TT3 increased with body mass, whereas several long-chain PFAAs were negatively correlated with body mass. Negative relationships between PFNA, PFDoA, PFTrDA, PFTeDA, and ∑PFAA with hatch dates may be the result of a disruption in incubation behaviour, resulting in earlier hatch dates. Consequently, TT3 concentrations were highest in males and females in 2018, a year in which PFAAs were lowest and hatch dates were delayed relative to 2017. As an Arctic seabird experiencing several indirect effects of climate change, the interaction of PFAAs on thyroid activity may cause additional stress to murres.

Exposure to certain perfluoroalkyl acids (PFAAs) can have considerable effects on the endocrine and immune systems, although such effects remain largely uncharacterized inwildlife. Using an apex avian predator, we investigated possible relationships of thyroid hormones (THs), specifically free (F) and total (T) thyroxine (FT4; TT4) and triiodothyronine (FT3; TT3), and the expression of an immune-related microRNA biomarker (i.e., miR-155), with the concentrations of 11 PFAAs in nestling peregrine falcons (Falco peregrinus). Nestling peregrines (n = 56; usually two chicks of each sex per nest) were blood sampled when 23 +/- 4 days old in urban and rural regions of the Laurentian Great Lakes Basin (Ontario, Canada) in 2016 and 2018. The circulating concentrations of several PFAAswere significantly associatedwith THs and estimated thyroid gland activity (TT3:TT4; FT3: FT4), including PFHxS (FT3; FT3:FT4), PFDS (TT3; TT3:TT4), PFOA (TT4; FT3:FT4), PFTeDA (TT4; FT3:FT4), PFHxDA (TT4; TT3:TT4) and SPFCAs (TT4). Our novel evaluation ofmiR-155 in peregrine nestlings identified significantly negative relationships of plasmamiR-155 counts with PFHxS and PFOA concentrations, indicating potential down-regulation of miR-155 expression and impaired immunity. Several PFAA homologues significantly predicted the variation in THs and miR-155 in conjunction with year (e.g., inter-annual differences in weather, ambient temperature, rainfall), region (urban/rural), nestling age, and/or diet (trophic position; delta N-15), which suggests thatmultiple environmental and biological stressors, including PFAA exposure, influenced thyroid activity and immune function in these nestlings. Further research is warranted to identify the mechanisms and additional impacts of PFAA-related thyroid and immune disruption on the growth, development, and health risks in developing birds. Following the ban of many historically-used flame retardants (FRs), numerous replacement chemicals have been produced and used in products, with some being identified as environmental contaminants. One of these replacement flame retardants is 1,2-dibromo-4-(1,2-dibromoethyl)-cyclohexane (DBE-DBCH; formerly abbreviated as TBECH), which to date has not been identified for risk assessment and potential regulation. DBE-DBCH technical mixtures consist largely of α- and β-diastereomers with trace amounts of γ- and δ-DBE-DBCH. The α- and β-isomers are known contaminants in various environmental media. While current global use and production volumes of DBE-DBCH are unknown, recent studies identified that DBE-DBCH concentrations were among the highest of the measured bromine-based FRs in indoor and urban air in Europe. Yet our mass balance fugacity model and modeling of the physical-chemical properties of DBE-DBCH estimated only 1% partitioning to air with a half-life of 2.2 d atmospherically. In contrast, our modeling characterized DBE-DBCH adsorbing strongly to suspended particulates in the water column (~12%), settling onto sediment (2.5%) with minimal volatilization, but with most partitioning and adsorbing strongly to soil (~85%) with negligible volatilization and slow biodegradation. Our modeling further predicted that organisms would be exposed to DBE-DBCH through partitioning from the dissolved aquatic phase, soil, and by diet, and given its estimated logKow (5.24) and a half-life of 1.7 d in fish, DBE-DBCH is expected to bioaccumulate into lipophilic tissues. Low concentrations of DBE-DBCH are commonly measured in biota and humans, possibly because evidence suggests rapid metabolism. Yet toxicological effects are evident at low exposure concentrations: DBE-DBCH is a proven endocrine disruptor of sex and thyroid hormone pathways, with in vivo toxic effects on reproductive, metabolic, and other endpoints. The objectives of this review are to identify the current state of knowledge concerning DBE-DBCH through an evaluation of its persistence, potential for bioaccumulation, and characterization of its toxicity, while identifying areas for future research.

In the Athabasca Oil Sands (OS) Region, the exposure (by air, water, diet), uptake and deposition of polycyclic aromatic compounds (PACs), including parent and alkylated hydrocarbons (PAHs) and dibenzothiophenes (DBTs), was assessed in nestling tree swallows (Tachycineta bicolor) at mining-related (OS1, OS2) and reference (REF) sites. The OS sites did not receive oil-sands processed waters (OSPW) and were ≥ 60 km from the reference sites. Most of the 42 PACs (≤ 98%) were detected in all matrices. Swallows at the OS sites were exposed to higher air and water concentrations of individual PAC congeners, ΣPACs, Σparent-PAHs, Σalkyl-PAHs and ΣDBTs. Compared to reference nestlings (ΣPACs: 13–27 ng/g wet weight (ww)), PACs were significantly higher in OS nestlings (31–106 ng/g ww) that also accumulated higher concentrations of major PAHs (i.e., naphthalene, C1-naphthalene, C2-naphthalene, C1-fluorenes, C2-fluorenes, C1-phenanthrenes) measured in 60% of nestlings. Uptake and deposition of PAHs in the birds' muscle was related to diet (δ15N: C1-naphthalenes, C2-naphthalenes, C1-fluorenes), water (C1-phenanthrenes), and air through inhalation and feather preening (C1-fluorenes), but fecal concentrations were not well explained by diet or environmental concentrations. While PAH concentrations were much higher in muscle than feces, they were highly correlated (p ≤ 0.001 for all). Thus feces may represent a non-lethal method for characterizing PAH exposure of birds, with muscle characterizing accumulation and sources of PAH exposure. Tree swallows in the Athabasca OS Region are exposed to many PACs, accumulating higher concentrations when developing in close proximity to mining activity through diet, aerial deposition and mining-impacted freshwater sources (e.g., wetlands).

Limited research has been conducted on microplastics in terrestrial ecosystems and biota, despite being some of the most ubiquitous environmental pollutants. We investigated the presence of microplastics (over 125 μm) in tree swallow (Tachicyneta bicolor) chicks (10 d. o.), an aerial insectivore whose diet involves terrestrial and/or freshwater sources. Swallows nested immediately downstream (300 m) of the discharge pipe of a large, urban wastewater treatment plant (WWTP) or at a rural conservation area (40 km apart). Anthropogenic microparticles (including microplastics) were identified in nearly all WWTP chicks (90%; N = 20) and reference chicks (83%; N = 20). All microparticles were fibers (100%) in the gastro-intestinal (GI) tracts of WWTP nestlings, whereas unexpectedly, they were more diverse in the GI tracts of reference chicks, with ~15% characterized as pre-production plastic pellets. The fecal sacs of most nestlings (90%) contained microparticles, and all were characterized as fibers suggesting their excretion by tree swallows. Compared to WWTP chicks, the reference chicks had more microparticles in their fecal sacs and larger particles (length, width) in their GI tracts, likely reflecting the more aquatic-based diet of the reference chicks fed insects caught adjacent to the nearby dam, compared to the more terrestrial-based diet of the WWTP chicks. The numbers of microparticles were not correlated between GI tracts and fecal sacs, nor with the chicks' condition or size (weight, organs, feathers). We recommend sampling macroinvertebrate prey to permit stronger conclusions regarding WWTPs as possible sources of microplastics for swallows, and to determine if such macroinvertebrates may be a non-lethal method to characterize microparticle diversity ingested by birds as presently identified in chicks' GI tracts. We conclude that sampling fecal sacs only, while not indicative of the diversity of microplastics ingested by terrestrial passerines (e.g., tree swallows), is useful for determining their exposure to microparticles.

Flame retardants (FRs) are a diverse group of chemicals, many of which persist in the environment and bioaccumulate in biota. Although some FRs have been withdrawn from manufacturing and commerce (e.g., legacy FRs), many continue to be detected in the environment; moreover, their replacements and/or other novel FRs are also detected in biota. Here, we review and summarize the literature on the toxic effects of various FRs on birds. Birds integrate chemical information (exposure, effects) across space and time, making them ideal sentinels of environmental contamination. Following an adverse outcome pathway (AOP) approach, we synthesized information on 8 of the most commonly reported endpoints in avian FR toxicity research: molecular measures, thyroid-related measures, steroids, retinol, brain anatomy, behaviour, growth and development, and reproduction. We then identified which of these endpoints appear more/most sensitive to FR exposure, as determined by the frequency of significant effects across avian studies. The avian thyroid system, largely characterized by inconsistent changes in circulating thyroid hormones that were the only measure in many such studies, appears to be moderately sensitive to FR exposure relative to the other endpoints; circulating thyroid hormones, after reproductive measures, being the most frequently examined endpoint. A more comprehensive examination with concurrent measurements of multiple thyroid endpoints (e.g., thyroid gland, deiodinase enzymes) is recommended for future studies to more fully understand potential avian thyroid toxicity of FRs. More research is required to determine the effects of various FRs on avian retinol concentrations, inconsistently sensitive across species, and to concurrently assess multiple steroid hormones. Behaviour related to courtship and reproduction was the most sensitive of all selected endpoints, with significant effects recorded in every study. Among domesticated species (Galliformes), raptors (Accipitriformes and Falconiformes), songbirds (Passeriformes), and other species of birds (e.g. gulls), raptors seem to be the most sensitive to FR exposure across these measurements. We recommend that future avian research connect biochemical disruptions and changes in the brain to ecologically relevant endpoints, such as behaviour and reproduction. Moreover, connecting in vivo endpoints with molecular endpoints for non-domesticated avian species is also highly important, and essential to linking FR exposure with reduced fitness and population-level effects.

Perfluoroalkyl substances (PFAS) are a large, diverse group of chemicals and several perfluoroalkyl acids (PFAAs) are known environmental contaminants. Wildlife exposure to PFAAs and precursors has been shown, but less is known regarding replacements such as shorter-chain PFAS. In the present study, exposure to a suite of PFAAs and associations with dietary, biological and ecological factors were investigated in populations of a sentinel apex species – the peregrine falcon (Falco peregrinus). Nestling blood (n = 57) and sibling eggs (n = 9) were sampled in 2016 and 2018 from nests in rural and urban regions across the Laurentian Great Lakes Basin, Canada. PFSAs (perfluorinated sulfonic acids) including PFHxS, PFOS, and PFDS were detected in most egg and plasma samples, whereas 11 PFCAs (perfluorinated carboxylic acids; C5–C14, C16) compared to eight PFCAs (C8–C14, C16) were detected in most eggs and plasma, respectively. Shorter-chain C8–C10 PFCAs were more dominant in plasma and longer-chain C12–C14 PFCAs in eggs, but profiles were similar for PFOS, PFDS, PFUdA and PFHxDA. The exposure to PFAAs in peregrine falcons is likely mediated by dietary factors such as foraging location (δ13C and δ34S) and trophic position (δ15N) given the associations observed in eggs and nestling plasma, respectively. Moreover, significant relationships were observed for circulating ΣPFCAs and region (rural/urban), and nestling body condition after adjusting for sampling year and dietary tracers, suggesting that compared to rural nestlings, urban nestlings may be more exposed to ΣPFCAs and prone to their potential physiological impacts. Our findings highlight the importance of integrating dietary, biological and ecological factors when studying PFAS exposure in birds.

Triphenyl phosphate (TPHP) is an organophosphate ester (OPE) used as a flame retardant (FR) and plasticizer. TPHP has previously been shown to disrupt behaviour in fish and mammals, but to our knowledge, this is the first study on the behavioural effects of TPHP in birds. Early life stage Japanese quail (Coturnix japonica) were exposed to nominal doses of 0 ng/g (vehicle-control), 5 ng/g (low dose), 50 ng/g (mid dose), and 100 ng/g (high dose) TPHP, both as embryos (via air cell injection prior to incubation) and as chicks (via daily gavage until 5 days post-hatch). The low dose reflects TPHP levels recorded in wild avian eggs, but actual environmental exposure levels may be higher given that TPHP is known to be rapidly metabolized in birds. We previously reported that the chicks exposed to TPHP in this study experienced reduced growth and resting metabolic rate, and sex-specific changes in thyroid function. The current study focuses on behavioural endpoints. We found that high-TPHP chicks exhibited less neophobia than vehicle-controls, and low-TPHP chicks exhibited more aggression towards conspecifics. No differences were observed in the responses of Japanese quail chicks to activity or tonic immobility (fear response) tests. These data add weight of evidence to previous findings suggesting that TPHP, among other OPEs, can disrupt ecologically-relevant behaviours in exposed vertebrates.

This study quantifies and compares concentrations and profiles of legacy and alternative (alt-) brominated flame retardants (BFRs) in the eggs of three gull (Laridae) species of international/UK conservation concern – great black-backed gulls (Larus marinus; n = 7), European herring gulls (L. argentatus; n = 16) and lesser black-backed gulls (L. fuscus; n = 11) in relation to their foraging ecology and behaviour in order to investigate potential exposure pathways at a remote landfill in western Scotland, UK. Egg concentrations of sum (∑) polybrominated diphenyl ethers (∑8PBDEs) in all three species exceeded those for most reported avian species using landfill, except for those in North America. Despite relatively high detection frequencies of ∑hexabromocyclododecanes (∑3HBCDDs) (94–100%), concentrations of ∑8PBDEs exceeded ∑3HBCDDs and ∑5alt-BFRs, with ∑8PBDE levels similar in all three species. Egg carbon isotopic (δ13C) values highlighted a greater marine dietary input in great black-backed gulls that was consistent with their higher BDE-47 levels; otherwise, dietary tracers were minimally correlated with measured BFRs. ∑3HBCDD egg concentrations of herring gulls markedly exceeded those reported elsewhere in Europe. Decabromodiphenylethane (DBDPE) was the only alt-BFR detected (6–14% detection rate), in a single egg of each species. The great black-backed gull egg contained the highest concentration of DBDPE measured in biota to date globally and provides strong evidence for its emerging environmental presence as a BDE-209 replacement in UK wildlife. Correlations between δ13C (dietary source) and some measured BFRs in eggs suggest multiple routes of BFR exposure for gulls frequenting landfill through their diet, behaviour, preening, dermal exposure and likely inhalation. The frequent use of landfill by herring gulls and their increased egg BFR burdens suggest that this species may be an important bioindicator of BFR emissions from such sites.

Fertile eggs from a captive population of kestrels were injected on embryonic day 5 (ED5) with a vehicle control or one of three doses within the range of concentrations that have been detected in biota (nominal concentrations of 0, 10, 50 or 100 ng/g egg; measured concentrations 0, 3.0, 13.7 or 33.5 ng TBBPA-BDBPE/g egg and 0, 5.3, 26.8 or 58.1 ng BTBPE/g egg). Eggs were artificially incubated until hatching (ED28), at which point blood and tissues were collected to measure morphological and physiological endpoints, including organ somatic indices, circulating and glandular thyroid hormone concentrations, thyroid gland histology, hepatic deiodinase activity, and markers of oxidative stress. Neither compound had any effects on embryo survival through 90% of the incubation period or on hatching success, body mass, organ size, or oxidative stress of hatchlings. There was evidence of sex-specific effects in the thyroid system responses to the BTBPE exposures, with type 2 deiodinase (D2) activity decreasing at higher doses in female, but not in male hatchlings, suggesting that females may be more sensitive to BTBPE. However, there were no effects of TBBPA-BDBPE on the thyroid system in kestrels. For the BTPBE study, a subset of high-dose eggs was collected throughout the incubation period to measure changes in BTBPE concentrations. There was no decrease in BTBPE over the incubation period, suggesting that BTBPE is slowly metabolized by kestrel embryos throughout their ∼28-d development. These two compounds, therefore, do not appear to be particularly toxic to embryos of the American kestrel.

The current-use brominated flame retardant, 1,2-dibromo-4-(1,2-dibromoethyl)cyclohexane (DBE-DBCH), is capable of perturbing sex steroid and thyroid hormone pathways in vitro and in vivo. Chemicals with this capability may also disrupt metabolic processes and are candidate obesogens, but this potential has not yet been determined for DBE-DBCH. Our objective was to examine gross biomarkers of metabolic disruption in captive American kestrels. Birds were exposed by diet to the β isomer at the environmentally relevant dose of 0.239 ng β-DBE-DBCH/g kestrel/day, from 30 days (d) prior to pairing through until chicks hatched (82 d) (n = 30 breeding pairs) or for 28 d (n = 16 pre-breeding pairs), and were compared with vehicle-only exposed controls. Body mass was assessed throughout the breeding season at biologically relevant time points, flight and feeding behavior was measured in 5-min samples daily, and plasma triglycerides and cholesterol were assessed at d10 of brood rearing. Treated males were heavier than controls at pairing (p = 0.051), the final week of courtship (p = 0.061), and at d10 (p = 0.012) and d20 of brood rearing (p = 0.051); β-DBE-DBCH-exposed breeding females were similar in weight to control females. Treated birds tended to have higher plasma triglycerides (p = 0.078), which for females, was positively associated with body mass (p = 0.019). Heavier breeding males had higher plasma concentrations of testosterone and total thyroxine (p ≤ 0.046). Overall, both sexes exposed to β-DBE-DBCH demonstrated reduced flight behavior and increased feeding behavior during courtship. In the pre-breeding pairs, treated male and female kestrels had a higher percentage of body fat than respective controls (p = 0.045). These results demonstrate that β-DBE-DBCH elicited inappropriate fat and weight gain in adult American kestrels, consistent with their increased feeding, reduced flight activity and endocrine changes, and suggests that DBE-DBCH may be an obesogen warranting further research to test this hypothesis.

Bis(2-ethylhexyl)-2,3,4,5-tetrabromophthalate (BEH-TEBP) and 2-ethylhexyl-2,3,4,5-tetrabromobenzoate (EH-TBB), flame retardant components of FireMaster 550® and 600® have been detected in tissues of wild birds. To address the paucity of information regarding potential impacts of flame retardants on the brain, brain volume regions of hatchling American kestrels (Falco sparverius) were evaluated following in ovo injection at embryonic day 5 with safflower oil or to 1 of 3 doses of either BEH-TEBP (12, 60, or 107 ng/g egg) or EH-TBB (11, 55, or 137 ng/g egg). The doses for both chemicals reflected concentrations reported in wild birds. The volumes of the hippocampus and telencephalon and volumetric differences between left and right hemispheres were measured in hatchlings (embryonic day 28). A sex-specific effect of BEH-TEBP on relative hippocampus volume was evident: the hippocampus was significantly enlarged in high-dose females compared to control females but smaller in low-dose females than the other females. There was no significant effect of EH-TBB on hippocampus volume in female kestrel hatchlings or of either chemical in male hatchlings and no effects of these concentrations of EH-TBB or BEH-TEBP on telencephalon volume or the level of symmetry between the hemispheres of the brain. In sum, embryonic exposure of female kestrels to these BEH-TEBP concentrations altered hippocampus volume, having the potential to affect spatial memory relating to ecologically relevant behavior such as prey capture, predator avoidance, and migration. Changes in environmental and wildlife health from contaminants in tailings water on the Canadian oil sands have been well-studied; however, effects of air contaminants on wildlife health have not. A field study was conducted to assess biological costs of natural exposure to oil sands-related air emissions on birds. Nest boxes for tree swallows (Tachycineta bicolor) were erected at two sites; within 5 km of active oil sands mining and extraction, and ≥ 60 km south, at one reference site. Passive air monitors were deployed at the nest boxes to measure nitrogen dioxide, sulfur dioxide, ozone, volatile organic compounds, and polycyclic aromatic hydrocarbons (PAHs). Nestlings were examined at day 9 post hatching to assess T cell function and morphometry. At day 14 post hatching, a subset of nestlings was euthanized to measure detoxification enzymes, endocrine changes, and histological alterations of immune organs. Except for ozone, all air contaminants were higher at the two oil sands sites than the reference site (up to 5-fold). Adult birds had similar reproductive performance among sites (p0.05). Nestlings from industrial sites showed higher hepatic ethoxyresorufin O-dealkylase (EROD) induction (p0.0001) with lower relative hepatic mass (p=0.0001), a smaller T cell response to the phytohemagglutinin skin test (p=0.007), and smaller bursae of Fabricius (p0.02); a low sample size for one site indicating lower body condition scores (p=0.01) at day 14 warrants cautious interpretation. There were no differences among nestlings for feather corticosterone (p0.6), and no histological alterations in the spleen or bursa of Fabricius (p0.05). This is the first report examining toxicological responses in wild birds exposed to air contaminants from industrial activity in the oil sands. It is also the first time that small, individual air contaminant monitors have been used to determine local contaminant levels in ambient air around nest boxes of wild birds.

Flame retardants (FR) are industrial chemicals and some are proven environmental contaminants that accumulate in predatory birds. Few studies have examined the influence of diet on FR profiles in nestling raptors and the possible physiological implications of such FR exposure. The objectives of this research were (1) to determine spatial patterns of ≤ 48 polybrominated diphenyl ether (PBDE) congeners and ≤ 26 non-PBDE FRs, including organophosphate esters (OPEs), in nestling peregrine falcons (Falco peregrinus) across the Canadian Great Lakes-St. Lawrence River Basin (GL-SLR; 2010) and in the eastern Canadian Arctic (2007); (2) to identify temporal changes in FR concentrations from the mid-2000s to 2010 in GL-SLR peregrine nestlings; (3) to investigate the role of diet using stable isotopes on exposure patterns of quantifiable FRs; and (4) to assess possible associations between circulating FRs and total (T) thyroxine (TT4) and triiodothyronine (TT3), tocopherol, retinol and oxidative status (isoprostanes). The summed concentrations of the top 5 PBDEs (Σ5) (BDE-47, -99, -100, -154, -153) were significantly higher in rural nestlings than urban nestlings in the GL-SLR, followed by the eastern Arctic nestlings. The PBDE congener profile of rural nestlings was dominated by BDE-99 (34‰), whereas BDE-209 (31‰) became dominant in the 2010 urban PBDE profile marking a shift since the mid-2000s. Low (ppb) concentrations of 25 novel non-PBDE FRs (e.g., 1,2-bis-(2,4,6-tribromophenoxy)ethane (BTBPE), decabromodiphenylethane (DBDPE)) were measured in the nestlings in at least one region, with the first report in peregrines of 15 novel non-PBDE FRs (e.g., 2-ethyl-1-hyxyl 2,3,4,5-tetrabromobenzoate (EHTBB), pentabromo allyl ether (PBPAE), tetrabromoethylcyclohexane (α-, β-DBE-DBCH)) as well as of tris (2-butoxyethyl) phosphate (TBOEP) (0–7.5 ng/g ww) > tris(2-chloroisopropyl) phosphate (TCIPP) (0.1–5.5 ng/g ww) > tris(2-chloroethyl) phosphate (TCEP) (0.02–2.0 ng/g ww) > tris(1,3-dichloro-2-propyl) phosphate (TDCIPP) (0–1.0 ng/g ww). Within the GL-SLR, the urban nestlings’ diet had significantly more terrestrial sources (greater δ13C values) than the broader, more aquatic-based diet of rural peregrines. Dietary source (δ13C) was significantly associated with concentrations of Σ5PBDE, BDE-209, EHTBB, and 2,2–4,4′,5,5′-hexabromobiphenyl (BB-153), with trophic level (δ15N) also positively associated with BDE-209 levels. Compared to urban nestlings, the rural nestlings had significantly lower circulating concentrations of thyroxine (TT4), triiodothyronine (TT3), a greater proportion of TT3 relative to TT4 (TT3:TT4), tocopherol and oxidative status (isoprostanes), but higher retinol levels; the most recalcitrant PBDE congener, BDE-153, in combination with low concentrations of some novel FRs, particularly octabromotrimethylphenyllindane (OBIND), may influence circulating thyroid hormones, especially TT4, and retinol levels of peregrine falcon nestlings. These associations of FR-endocrine-biochemical measures suggest possible exposure-related changes in these birds and further study is warranted.

In this paper, we have compiled and reviewed the most recent literature, published in print or online from January 2010 to December 2012, relating to the human exposure, environmental distribution, behaviour, fate and concentration time trends of polybrominated diphenyl ether (PBDE) and hexabromocyclododecane (HBCD) flame retardants, in order to establish their current trends and priorities for future study. More data are now becoming available for remote areas not previously studied, Indian Ocean islands, for example. Decreasing time trends for penta-mix PBDE congeners were seen for soils in northern Europe, sewage sludge in Sweden and the USA, carp from a US river, trout from three of the Great Lakes and in Arctic and UK marine mammals and many birds, but increasing time trends continue in polar bears and some birds at high trophic levels in northern Europe. This may be partially a result of the time delay inherent in long-range atmospheric transport processes. In general, concentrations of BDE209 (the major component of the deca-mix PBDE product) are continuing to increase. Of major concern is the possible/likely debromination of the large reservoir of BDE209 in soils and sediments worldwide, to yield lower brominated congeners which are both more mobile and more toxic, and we have compiled the most recent evidence for the occurrence of this degradation process. Numerous studies reported here reinforce the importance of this future concern. Time trends for HBCDs are mixed, with both increases and decreases evident in different matrices and locations and, notably, with increasing occurrence in birds of prey. Temporal trends for both PBDEs and HBCD in Asia are unclear currently. A knowledge gap has been noted in relation to metabolism and/or debromination of BDE209 and HBCD in birds. Further monitoring of human exposure and environmental contamination in areas of e-waste recycling, particularly in Asia and Africa, is warranted. More data on temporal trends of BDE and HBCD concentrations in a variety of matrices and locations are needed before the current status of these compounds can be fully assessed, and the impact of regulation and changing usage patterns among different flame retardants determined. Accumulation and evidence of debromination of the flame retardant 2,2′,3,3′,4,4′,5,5′,6,6′-decabromodiphenyl ether (BDE-209) have been reported for biota, including raptorial birds, based on PBDE congener residues in tissues and eggs. However, in vivo studies with BDE-209-exposed birds are rare and unknown for a raptorial species. In the present study, males (n = 22) of raptorial American kestrels (Falco sparverius) were exposed to 116,000 ng of BDE-209 (high purity, >98%; in safflower oil) per day for 21 days (~2,436,000 ng total BDE-209 exposure over this uptake period), followed by a 25-day depuration period. Control males (n = 11) received the safflower vehicle only. In the exposed birds, BDE-209 was quantifiable in all plasma (end of uptake and depuration period) as well as liver and fat (end of depuration only) samples. The mean (±SE) BDE-209 level in plasma was 1474 ± 1145 ng/g wet weight (ww) at the end of the uptake period, and was significantly (p p > 0.001) higher in the exposed relative to the control bird samples (except for BDE-154 and -153 in fat). Additional PBDE congeners found in plasma included nona-BDEs (208, 207 and 206), followed by octa-BDEs (197, 196, 201 and 203), and in liver and/or fat, the hepta-BDEs 180 and 183 and BDE-153. Higher hepatic EROD activity (cytochrome P450 1A1 monooxygenase-mediation) in the exposed birds compared to control birds was strongly suggested to be PBDE-induced, and was consistent with BDE-209 and congener metabolism in the exposed kestrels. The mean EROD activity rate was 36.1 pmol/min/mg protein relative to the (n = 4) control birds whose activity was just above the detection limit (10.3 pmol/min/mg protein). Overall, the results demonstrated that following diet exposure of kestrels to high purity BDE-209, uptake occurred as well as BDE-209 degradation via debromination to lower brominated PBDE congeners. Polybrominated diphenyl ethers (PBDE) are persistent environmental pollutants that have been detected in wildlife globally. American kestrels exposed to the commercial PBDE mixture DE-71 have previously demonstrated reduced reproductive success and behaviors during courtship and brood rearing; however, it remains unknown whether DE-71 affects incubation. During breeding, captive kestrels were exposed to the DE-71 mixture dissolved in safflower oil at two environmentally relevant concentrations (low: 283.5 ± 48.2, high: 1104.8 ± 124.5 ng/g wet weight [ww]) via diet for an average of 75 d. Unexpected low in ovo concentrations of hexabromocyclododecane (HBCD) were also detected (low: 3 ± 1 ng/g ww, high: 16 ± 3 ng/g ww). All comparisons are made to control pairs. Kestrel pairs in the low- and high-exposure groups experienced longer incubation periods with increasing exposure to ΣPBDE and some individual congeners. As incubation progressed, pairs exposed to DE-71 had significantly lower nest temperatures, which were on average 19% lower in low-exposure nests and 35% lower in high-exposure nests during late incubation. The DE-71 exposed pairs (low and high) also demonstrated significantly reduced incubation constancy (defined as percent of temperature readings above the maximum daily ambient temperature) during early incubation compared to controls. Nest temperatures (all pairs) and incubation constancy (high pairs) during early incubation (d 1-3) were significantly and positively associated with the proportion of eggs that hatched per pair. Higher incubation constancy and incubating nest temperatures in the low-exposure group were associated with markedly less egg weight loss by mid-incubation. These findings demonstrate that exposure to PBDE significantly affected kestrels during incubation, a critical period for embryonic development.

Feathers are useful for monitoring contaminants in wild birds and are increasingly used to determine persistent organic pollutants. However, few studies have been conducted on birds with known exposure levels. We aimed to determine how well nestling feather concentrations reflect in ovo exposure to hexabromocyclododecane (α-, β- and γ-HBCDD), and to determine if feather concentrations are related to physiological biomarkers. Captive kestrels (n = 11) were exposed in ovo to maternally transferred HBCDD-isomers at concentrations of 127, 12 and 2 ng/g wet weight of α-, β- and γ-HBCDD (measured in sibling eggs), respectively, and compared to controls (n = 6). Nestling growth was monitored at 5 d intervals and circulating thyroid hormone concentrations assessed at d 20. Tail feathers were collected prior to the first molt and analyzed for HBCDD isomers. The mean ΣHBCDD concentration in feathers was 2405 pg/g dry weight (in exposed birds) and α-, β- and γ-HBCDD made up 32%, 13%, and 55%, respectively of the ΣHBCDD concentrations. This isomer distribution deviated from the typical dominance of α-HBCDD reported in vertebrate samples. Exposed chicks had significantly higher feather concentrations of β- and γ-HBCDD compared with controls (p = 0.007 and p = 0.001 respectively), while α-HBCDD concentrations did not differ between the two groups. Feather concentrations of α-HBCDD were best explained by egg concentrations of β- or γ-HBCDD concentrations (wi = 0.50, 0.30 respectively), while feather concentrations of β- and γ-HBCDD were influenced by growth parameters (rectrix length: wi = 0.61; tibiotarsus length: wi = 0.28). These results suggest that feather α-HBCDD concentrations may reflect internal body burdens, whereas β- and γ-HBCDD may be subject to selective uptake. The α-HBCDD concentrations in the feathers were negatively associated with the ratio of plasma free triiodothyronine to free thyroxine (T3:T4; p = 0.020), demonstrating for the first time that feather concentrations may be used to model the effect of body burdens on physiological endpoints.

1,2-dibromo-4-(1,2-dibromoethyl)cyclohexane (DBE-DBCH - formerly TBECH) is an emerging brominated flame retardant (BFR) pollutant with androgen potentiating ability and other endocrine disrupting effects in birds and fish. The objectives of this study were to determine the effects of exposure to environmentally-relevant levels of DBE-DBCH on circulating levels of thyroid and sex steroid hormones in American kestrels, and if hormonal concentrations were related to previously reported changes in reproductive success and courtship behaviors. Sixteen kestrel pairs were exposed to 0.239ng β-DBE-DBCH/g kestrel/day by diet, based on concentrations in wild bird eggs, from 4 weeks before pairing until the chicks hatched (mean 82 d), and were compared with vehicle-only-exposed control pairs (n=15). As previously reported, DBE-DBCH concentrations were not detected in tissue or eggs of these birds, nor were any potential metabolites, despite the low method limits of detection (≤0.4ng/g wet weight), suggesting it may be rapidly metabolized and/or eliminated by the kestrels. Nevertheless, exposed kestrels demonstrated changes in reproduction and behavior, indicating an effect from exposure. During early breeding, males were sampled at multiple time points at pairing and during courtship and incubation; females were blood sampled at pairing only; both sexes were sampled at the end of the season. All comparisons are made to control males or control females, and the relative differences in hormone concentrations between treatment and control birds, calculated separately for each sex, are presented for each time point. Males exposed to β-DBE-DBCH demonstrated significantly (p=0.05) lower concentrations of total thyroxine (TT4) overall, that were 11-28% lower than those of control males at the individual sampling points, yet significantly higher (p=0.03) concentrations of free thyroxine (FT4), that were 5-13% higher than those of control males at the individual sampling points; females had similar concentrations of TT4 and FT4 at the time of pairing, and T4 was similar in both sexes at the end of the breeding season. Testosterone (T) concentrations in the treatment males were significantly higher during early (85%) and mid-courtship (30%) (time*treatment p=0.001), whereas females demonstrated a reduction in T at the time of pairing (17%, p=0.05). In the treatment females, concentrations of 17β-estradiol (E2) showed a non-significant decrease (20%) and were positively correlated with T concentrations (p=0.03); E2 concentrations were below quantification limits in males. Hexabromocyclododecane (HBCD) is a high production volume brominated flame retardant that has been detected in the environment and wildlife at increasing concentrations. This study was designed to determine potential effects of dietary exposure to environmentally relevant levels of HBCD on behavior during reproduction in captive American kestrels. Twenty kestrel pairs were exposed to 0.51 μg technical HBCD g −1 kestrel d −1 from 4 weeks prior to pairing until chicks hatched (∼75 d). Ten pairs of controls received the safflower oil vehicle only and were used for comparison. During the courtship period the chitter-calls were reduced in both sexes ( p = 0.038) and females performed fewer bonding displays ( p = 0.053). Both sexes showed a propensity to be less active than controls during courtship. The reduction in male courtship behavior was correlated with reduced courtship behaviors of females ( p ⩽ 0.008) as well as reduced egg mass ( p = 0.019). During incubation, nest temperatures of treatment pairs were lower at mid-incubation ( p = 0.038). HBCD-exposed males performed fewer key parental behaviors when rearing nestlings, including entering the nest-box, pair-bonding displays and food-retrievals. HBCD-exposed females appeared to compensate for the reduced parental behavior of their mates by performing these same behaviors more frequently than controls ( p = 0.004, p = 0.027, p = 0.025, respectively). This study demonstrates that HBCD affects breeding behavior in American kestrels throughout the reproductive season and behavioral alterations were linked to reproductive changes (egg size). This is the first study to report HBCD effects on reproductive behavior in any animal model.

Hexabromocyclododecane (HBCD) is a high-production-volume, brominated flame-retardant that is used in items such as polystyrene foams. HBCD has been detected in the environment, wildlife tissues and in humans globally with some of the highest recorded levels in predatory birds. This study examined the effects of exposure to environmentally relevant levels of HBCD on the reproductive physiology of captive male American kestrels (Falco sparverius), a predatory bird. Two sets of males were used: one group not housed with females (unpaired: nc=12, nHBCD=10) and the second group housed with females (breeding: nc=10, nHBCD=20). All treatment birds were exposed to 0.51 μg HBCD/g kestrel/day technical HBCD, and controls to safflower oil only, injected into their food during seasonal testicular development. Unpaired males were exposed for 3 weeks and euthanized for testicular analysis. Breeding males were exposed for 3 weeks prior to pairing and throughout the courtship period. The HBCD-exposed unpaired males had heavier testes (p≤0.017) and a trend towards more seminiferous tubules containing elongated spermatids (p=0.052). There was also a moderate increase in plasma testosterone concentrations (p=0.056) compared to controls. In breeding males, testosterone levels increased during courtship to culminate in higher levels than controls by the time the first egg was laid (p=0.010) and circulating free and total T4 was reduced throughout. The number of sperm cells reaching the perivitelline layer of the first egg for breeding males did not differ between the two groups. This study is the first report that HBCD exposure at environmentally relevant levels alters reproductive physiology in male birds and suggests that birds may be more sensitive to HBCD than mammals.

Microsatellites mutate frequently by replication slippage. Empirical evidence shows that the probability of such slippage mutations may increase with the length of the repeat region as well as exposure to environmental mutagens, but the mutation rate can also differ between the male and female germline. It has been hypothesized that more intense sexual selection or sperm competition can also lead to elevated mutation rates, but the empirical evidence is inconclusive. Here, we analyzed the occurrence of germline slippage mutations in the hypervariable pentanucleotide microsatellite locus HrU10 across six species of swallow (Aves: Hirundinidae). These species exhibit marked differences in the length range of the microsatellite, as well as differences in the intensity of sperm competition. We found a strong effect of microsatellite length on the probability of mutation, but no residual effect of species or their level of sperm competition when the length effect was accounted for. Neither could we detect any difference in mutation rate between tree swallows (Tachycineta bicolor) breeding in Hamilton Harbour, Ontario, an industrial site with previous documentation of elevated mutation rates for minisatellite DNA, and a rural reference population. However, our cross-species analysis revealed two significant patterns of sex differences in HrU10 germline mutations: (1) mutations in longer alleles occurred typically in the male germline, those in shorter alleles in the female germline, and (2) male germline mutations were more often expansions than contractions, whereas no directional bias was evident in the female germline. These results indicate some fundamental differences in male and female gametogenesis affecting the probability of slippage mutations. Our study also reflects the value of a comparative, multi-species approach for locus-specific mutation analyses, through which a wider range of influential factors can be assessed than in single-species studies.

Temporal and spatial trends in contaminant concentrations were assessed in Lake Erie watersnakes, a threatened (USA)/endangered (Canada) species restricted to western Lake Erie. Temporal changes in plasma contaminant levels were determined in 1990 and 2003, and spatial patterns in 2003 at 12 sites, throughout the species' range. During this period, the watersnakes' diet changed from fish (75%) and amphibians (25%) that avoid zebra mussels, to round gobies (95%) that feed extensively on zebra mussels. Temporal trends indicate that watersnakes on Pelee and North Bass Islands showed a marginal increase in hexachlorobenzene levels, and a significant decline in dieldrin, oxychlordane, and heptachlor epoxide, likely reflecting declines in aerial deposition and clearing of local vineyards. The contaminants with the greatest burdens, sum PCBs and p,p′-DDE, remained stable in the snakes, consistent with trends in other local biota, suggesting that although the dietary switch to round gobies meant consumption of a more contaminated diet, their diet remained at the same trophic position. We suggest that the watersnakes' PCB and p,p′-DDE temporal patterns reflect the lack of change in sediment concentrations with minimal influence from their dietary switch. Similar to top avian predators, PCBs, p,p′-DDE, and technical chlordane, are most prevalent in watersnakes; this ranking remains unchanged. In 2003, the watersnakes demonstrated significant spatial differences in concentrations of p,p′-DDE, dieldrin, technical chlordane and its metabolites. Their 2003 concentrations of p,p′-DDE, and to a lesser extent PCBs, exceed the recommended interim no-observable effects levels on watersnake embryonic survival. Further investigations are required to determine if these higher levels of PCBs, p,p′-DDE, and technical chlordane, affect reproductive and physiological parameters of the Lake Erie watersnake. Until concentrations of sediment contaminants decline in western Lake Erie, these endangered/threatened watersnakes will continue to be exposed to higher concentrations of persistent organic pollutants. Polybrominated diphenyl ethers (PBDEs) are increasing in biota. Here, captive adult American kestrels were exposed daily by diet to safflower oil (controls), or one of two levels of a pentaBDE technical formulation, DE-71 (0.3 or 1.6 ppm), for approximately 75 days, commencing 21 days before breeding. This exposure resulted in eggs having PBDE concentrations similar (low exposure) or within the same order of magnitude (high exposure) reported for wild American kestrels and gulls in the Great Lakes. Compared to controls, kestrels in both exposure groups copulated less, spent less time in their nest boxes, and participated in fewer pair-bonding behaviors. Furthermore, the timing of these behaviors, which is important to creating and maintaining the pair-bond, also differed significantly from the controls. The females in the low-exposure group made fewer compatible trilling calls and ate less frequently. These behavioral changes were compounded by increasing exposure to DE-71 during the 9-day courtship period immediately preceding egg laying, a standard measure of the kestrel courtship period. The birds in the high-exposure group made more food transfers, excited "klee" calls, and copulations, the latter only when compared to the low-exposure birds, whereas the low-exposure males performed fewer pair-bonding behaviors. This study demonstrates that the exposure of kestrels to environmentally relevant levels of DE-71 modifies the quality of the pair-bond, affects the reproductive behavior of both sexes, and occurs when birds are exposed for a short period as adults. In addition, these behavioral effects are consistent with the observed reproductive changes in these birds.

High concentrations of polybrominated diphenyl ethers (PBDEs) accumulate in predatory birds. Several PBDE congeners are considered thyroid disruptors; however, avian studies are limited. The authors examined circulating thyroid hormones and thyroid gland function of nestling American kestrels (Falco sparverius) at 17 d to 20 d of age, following embryonic exposure by maternal transfer only to environmentally relevant levels of PBDEs (DE-71 technical mixture). Nestlings were exposed to in ovo sum (Σ) PBDE concentrations of 11 301 ± 95 ng/g wet weight (high exposure), 289 ± 33 ng/g wet weight (low exposure), or 3.0 ± 0.5 ng/g wet weight (controls, background exposure). Statistical comparisons are made to controls of the respective sexes and account for the relatedness of siblings within broods. Circulating concentrations of plasma total thyroxine (TT4 ) and total triiodothyronine (TT3 ) in female nestlings were significantly influenced overall by the exposure to DE-71. Following intramuscular administration of thyroid-stimulating hormone, the temporal response of the thyroid gland in producing and/or releasing TT4 was also significantly affected by the females' exposure to DE-71. The altered availability of T4 for conversion to T3 outside of the gland and/or changes in thyroid-related enzymatic activity may explain the lower TT3 concentrations (baseline, overall) and moderately altered temporal TT3 patterns (p = 0.06) of the treatment females. Controlling for the significant effect on TT3 levels of the delayed hatching of treatment females, baseline TT3 levels were significantly and positively correlated with body mass (10 d, 15 d, 20 d), with PBDE-exposed females generally being smaller and having lower TT3 concentrations. Given that exposure concentrations were environmentally relevant, similar thyroidal changes and associated thyroid-mediated processes relating to growth may also occur in wild female nestlings. Polybrominated diphenyl ethers (PBDEs), a class of additive flame retardants, are temporally increasing in wildlife tissues and capable of disrupting normal endocrine function. We determined whether in ovo and post-hatch exposure of captive American kestrels (Falco sparverius) to environmentally relevant PBDEs alter thyroid, retinol, and oxidative stress measures. Control eggs were injected with safflower oil and subsequent nestlings fed the same vehicle; dosed eggs received PBDE congeners (BDE-47, -99, -100, -153), which mainly comprise the Penta-BDE commercial mixture, dissolved in safflower oil at concentrations (1500 ng/g total [Sigma] PBDEs) approximating those in Great Lakes gull eggs. Nestlings hatching from dosed eggs were orally exposed for 29 days to variable SigmaPBDE concentrations that are similar to levels reported in tissues of Great Lakes trout (100 ng/g). Treatment kestrels had lower plasma thyroxine (T(4)), plasma retinol, and hepatic retinol and retinyl palmitate concentrations, but unaltered triiodothyronine (T(3)) concentrations and thyroid glandular structure. BDE-47, -100, and -99 were negatively associated with plasma T(4), plasma retinol (BDE-100, -99) and hepatic retinol (BDE-47). Despite an antioxidant-rich diet, PBDE exposure induced hepatic oxidative stress, particularly in females, with an increased hepatic GSSG:GSH ratio, a marginal increase in lipid peroxidation, and increased oxidized glutathione. Positive associations were found between concentrations of BDE-183 and thiols and, in males, between BDE-99 and reduced GSH, but a negative association occurred between BDE-99 and TBARS. Subsequently, concentrations of PBDE congeners in wild birds may alter thyroid hormone and vitamin A concentrations, glutathione metabolism and oxidative stress.

Electrical power lines are ubiquitous in the developed world and in urban areas of the developing world. All electrical currents, including those running through power lines, generate electric and magnetic fields (EMFs). Electrical power lines, towers,and distribution poles are used by birds for perching, hunting, and nesting. Therefore, many bird species, like humans, are exposed to EMFs throughout their lives. EMFs have been implicated in adversely affecting multiple facets of human health,including increasing the risks of life-threatening illnesses such as leukemia, brain cancer, amyotrophic lateral sclerosis, clinical depression, suicide, and Alzheimer's disease. A great deal of research and controversy exists as to whether or not exposure to EMFs affects the cellular, endocrine, immune, and reproductive systems of vertebrates. Laboratory work has used mice, rats, and chickens as models for this EMF research in an effort to understand better the possible implications of EMF exposure for humans. However, EMF exposure of wild birds may also provide insight into the impacts of EMFs on human health. This review focuses on research examining the effects of EMFs on birds; most studies indicate that EMF exposure of birds generally changes, but not always consistently in effect or in direction, their behavior, reproductive success, growth and development, physiology and endocrinology, and oxidative stress under EMF conditions. Some of this work has involved birds under aviary conditions, while other research has focused on free-ranging birds exposed to EMFs. Finally, a number of future research directions are discussed that may help to provide a better understanding of EMF effects on vertebrate health and conservation.

Hexabromocyclododecane (HBCDD) is a flame retardant and a global contaminant, yet the toxicokinetics of HBCDD diastereoisomers remains unknown in wildlife species. The present study examined in captive American kestrels (Falco sparverius) (diastereo) isomer-specific HBCDD uptake, depletion, tissue distribution, and transfer to eggs in a dietary dosing study with an HBCDD technical mixture (HBCDD-TM). Adult tissue and plasma collections were from separate cohorts of unpaired individual males (n = 10) and females (n = 10) exposed for 21 d to 800 ng/g wet weight of HBCDD-TM (in safflower oil and injected into their cockerel [brain] diet), followed by a 25-d depuration period. A separate cohort of 12 males only was used for control adult tissue and plasma collections. For egg collections, separate cohorts of 11 control pairs (n = 22 birds) and 20 HBCDD-exposed pairs (n = 40 birds) were allowed to breed, and their eggs were collected (n = 19 exposed eggs and n = 10 control eggs). The sum (Σ) HBCDD concentrations were near or below detection (0.01-0.1 ng/g wet wt) in all control samples but quantifiable in all samples from exposed birds (no differences [p 0.05] between males and females). Arithmetic mean ΣHBCDD concentrations were highest in fateggsliverplasma. The mean ΣHBCDD depletion rate in plasma between the uptake and depuration periods was estimated to be 0.22 ng/g/d with a half-life of approximately 15 d. The γ-HBCDD diastereoisomer was60% of the ΣHBCDD in plasma after the uptake period and similar to the HBCDD-TM (∼80%). After the depuration period, α-HBCDD was70% of the HBCDD in plasma, fat, liver, and eggs; and this α-HBCDD domination indicated isomer-specific accumulation as a result of selective metabolism, uptake, protein binding, and/or in ovo transport.

In the oil sands of Alberta, Canada, toxicology research has largely neglected the effects of air contaminants on biota. Captive Japanese quail (Coturnix c. japonica) and American kestrels (Falco sparverius) were exposed to mixtures of volatile organic compounds and oxidizing agents (benzene, toluene, NO2 and SO2) in a whole-body inhalation chamber, to test for toxicological responses. Hepatic biotransformation measured through 7-ethoxyresorufin-O-dealkylase (EROD) tended to be increased in exposed kestrels (p=0.06) but not in quail (p=0.15). Plasma corticosterone was increased in the low dose group for quail on the final day of exposure (p=0.0001), and midway through the exposure period in exposed kestrels (p=0.04). For both species, there was no alteration of T and B-cell responses, immune organ mass, or histology of immune organs (p>0.05). This study provides baseline information valuable to complement toxicology studies and provides a better understanding of potential health effects on wild avifauna.

Reproductive success of birds is influenced by maternal factors, ambient temperatures, predation, food supply, and/or exposure to environmental contaminants e.g., flame retardants (FRs). Reproduction of tree swallows (Tachycineta bicolor) was compared among waste water treatment plants (WWTPs) and a reference reservoir in Ontario, Canada (2007-2010), to determine the importance of exposure to polybrominated diphenyl ether (PBDEs) FRs within a complex contaminant cocktail, relative to natural and biological factors known to influence avian reproduction. The birds primarily consumed insects emerging from the reference reservoir and WWTP outflows, where effluent mixed with receiving waters. FR egg concentrations were dominated by 5 PBDE congeners (∑5PBDEs): 2,2'.4,4',5-pentabromodiphenyl ether (BDE-99), 2,2',4,4'-tetrabromodiphenyl ether (BDE-47), 2,2'4,4',6-pentabromodiphenyl ether (BDE-100), 2,2',4,4',5,5'-hexabromodiphenyl ether (BDE-153), and 2,2',4,4',5,6'-hexabromodiphenyl ether (BDE-154), with much lower concentrations of decabromodiphenylether (BDE-209), hexabromocyclododecane (HBCDD), and novel FRs. Although higher than ∑5PBDEs, polychlorinated biphenyls (PCBs) egg concentrations were unlikely to affect the swallows' reproduction. Clutch size and timing, fledging, breeding success, and predation, varied significantly among sites, generally being poorer at WWTP1 and better at WWTP2. The early reproductive stages were sensitive to some FRs at measured concentrations. The ∑5PBDEs, maternal age, and minimum ambient temperatures predicted onset of egg laying in the most parsimonious statistical model, and there were positive relationships between egg size and HBCDD or BDE-209 concentrations. However, there were no significant correlations with any reproductive measures, individual BDE congeners or low concentrations of novel FRs, in this first such report for novel FRs and wild birds. Tree swallows are passerines, and passerines may differ from birds of prey in their reproductive sensitivity to PBDE exposure: lay date, minimum temperatures, and predation, but not PBDE exposure, predicted reproductive productivity. Overall, there was some influence of the PBDEs available in these WWTP outflows on early reproductive parameters, but not reproductive output, of these passerines.

Captive American kestrels (Falco sparverius) were exposed via diet during reproduction to an environmentally relevant concentration of β-1,2-dibromo-4-(1,2-dibromoethyl)cyclohexane (β-TBECH). The β-TBECH isomer was injected into the food source at a daily dosing concentration of 0.239 ng/g kestrel/day (22 pairs); control birds were exposed via diet to the safflower oil vehicle only (24 pairs). Eight pairs in each group were exposed for four weeks and sacrificed for tissue analysis; the remaining pairs completed their breeding cycle, with exposure ceasing at the end of incubation (82 days). α- and β-TBECH appeared to be rapidly metabolized and/or eliminated from fat, liver, and plasma; both isomers and potential hydroxylated metabolites of β-TBECH (plasma) were undetected. Notwithstanding, compared to controls, pairs exposed to β-TBECH laid fewer eggs (p = 0.019) and laid lighter eggs (successful eggs: p = 0.009). Exposed pairs also demonstrated poorer egg fertility (p = 0.035) although testis mass and histology were similar among males. Reductions in egg production and fertility resulted in decreased hatchling success (p = 0.023). The β-TBECH-exposed pairs also produced fewer males overall (p = 0.009), which occurred concurrently with increased estradiols maternally deposited in eggs (p = 0.039). These findings demonstrate that β-TBECH may be detrimental for breeding in wild birds receiving similar exposure levels.

Recently, the ban of hexabromocyclododecane (HBCD), a high-production-volume flame retardant, was announced in Europe and North America. However, the effects of HCBD remain understudied in birds. The objectives of the present comparative effects study were to determine whether exposure to an HBCD technical mixture (HBCD-TM) altered avian reproductive measures at an environmentally relevant concentration. American kestrels were exposed daily by food to HBCD-TM, i.e., 0.51 µg HBCD/g kestrel/d; exposed kestrels laid eggs that had α-HBCD concentrations (163.5 ± 75.1 ng/g wet wt) tenfold greater than β- and γ-HBCD isomers, an isomer profile and concentrations similar to those of eggs of wild peregrine falcons (Falco peregrinus). Concentrations of HBCD were not detected in the control kestrel eggs. In comparison with controls, the kestrels exposed to HBCD began to lay their eggs 6 d earlier and laid larger clutches of smaller eggs. The size of the eggs was inversely correlated with the in ovo α-HBCD concentrations. The smaller eggs of the HBCD exposed kestrels also lost more weight by midincubation, suggesting increased eggshell porosity since eggshell thickness was comparable. Generally birds that lay more eggs and lay earlier in the breeding season gain the advantage of better hatching and fledging success, yet the kestrels exposed to HBCD failed to have better reproductive success than the control birds. These reproductive changes were a function of HBCD exposure, likely through changes in food consumption, with possible impacts on, for example, reproductive behavior and/or alterations in thyroid hormones. Comparisons of brominated flame retardants (BFRs) in the eggs of peregrine falcons (Falco peregrinus) recently collected (2003–2007), are made between Canada (N=12) and Spain (N=13). Overall, concentrations of sum (Σ) polybrominated diphenyl ethers (PBDEs; 16 di-deca-BDE congeners) exceeded Σhexabromocyclododecane (HBCD) and were an order of magnitude higher than 2,2′4,4′,5,5′-hexabromobiphenyl (BB-153)>hexachlorocyclopentenyl-dibromocyclooctane (HCBDCO)>1,2-bis(2,4,6-tribromophenoxy)ethane (BTBPE)>decabromodiphenylethane (DBDPE)>octabromotrimethylphenyllindane (OBIND)>hexabromobenzene (HBB)>bis(2-ethyl-1hexyl)tetrabromophthalate (BEHTBP). This is the first report of detectable HBCDCO and BEHTBP concentrations in biota, and the highest in ovo concentration of ∑HBCD (14,617 ng/g lw; Montreal, Canada) to date. There were significantly greater egg concentrations of BB-153, ΣHBCD, and ΣPBDE including BDE-153, -99, -100 and -183, in Canadian than Spanish peregrines with a terrestrial diet. HBB, BTBPE, and OBIND were detected in eggs from both countries, but only Canadian peregrine eggs had detectable levels of HCDBCO (25%) and DBDPE (N=1). The in ovo PBDE congener profile was dominated by BDE-153>BDE-99>BDE-47>BDE-183>BDE-100>BDE-209, with the isomeric HBCD pattern being α-HBCD>γ-HBCD (β-HBCD undetected). The Canadian peregrine eggs had lower enantiomeric HBCD values consistent with their higher fractions of (−) α-HBCD, suggesting selective enantiomeric enrichment or that the (+) α-isomer is more readily metabolized and so deposited in the egg through maternal transfer. Continental differences in egg burdens of peregrines are discussed relative to BFR usage patterns and exposure of peregrines on their breeding grounds. Polybrominated diphenyl ethers (PBDEs) are additive flame retardants that are environmentally persistent and bioaccumulative. The developmental effects of in ovo exposure to environmentally relevant levels of the PBDE technical mixture, DE-71, on male reproductive physiology in captive American kestrels (Falco sparverius) was determined. Males were exposed in ovo by direct maternal transfer to DE-71 at three mean concentrations of 289 ng/g ww (low exposure), 1131 ng/g ww (high-exposure), or background levels of 3 ng/g ww (control). As adults, males were paired with unexposed females for breeding and, 1 year later, sacrificed for testes evaluation. While breeding, high-exposure males demonstrated a trend of reduced circulating testosterone levels when their female mate commenced egg laying when compared with controls (p = 0.056). No differences in circulating free T₄ or T₃ were detected. Sperm numbers were elevated on the perivitelline layer of the first egg of both high- and low-exposure males when compared with controls (p = 0.021). High-exposure males had a higher gonadosomatic index (p = 0.046) and heavier right testis than controls (p = 0.034) with a similar trend for their left testis (p = 0.055). High-exposure males had more seminiferous tubules containing lumen than controls (p = 0.030), and in proportion to the total number of tubules, low-exposure males had more tubules containing lumen than did controls (p = 0.016). Only high-exposure males had fewer than half of tubules containing final spermatids (43%). The results of the present study demonstrate that embryonic exposure to technical DE-71 affects the reproductive tract of adult male kestrels.

Concentrations of Mirex, Dechlorane (Dec) Plus (DP), Dec 602, Dec 603, and Dec 604 were significantly higher in peregrine falcon (Falco peregrinus) eggs from Canada than Spain, with the former having the only measurable concentrations of the dechlorinated DP products, decachloropentacyclooctadecadiene (aCl10DP) and undecachloropentacyclooctadecadiene (aCl11DP). Large variations also occurred in the in ovo concentrations of the DP monoadduct, DPMA. This is the first study to report the accumulation and metabolism of DP by peregrines, both DP and dechloranes in European biota, as well as dechloranes in a terrestrial organism and one at the top of the food web. The geographical differences in the measured in ovo concentrations reflect local exposure of the adult peregrines on their breeding grounds, likely differences in diet of the adults, the production of DP on the Niagara River, and the greater use of Mirex and dechloranes as flame retardants in North America than Europe.

Polybrominated diphenyl ethers (PBDE) are ubiquitous, lipophilic, and bioaccumulative brominated flame retardants. Plasma retinol concentrations of captive adult American kestrels were assessed at the beginning of the breeding season following 3 wk of daily dietary exposure to vehicle (control), low (0.3 ng/g wet weight [ww]), or high (1.6 ng/g ww) concentrations of DE-71 and in their 25-d-old nestlings following embryonic exposure by maternal deposition to environmentally relevant low (291 ± 48 ng/g ww) or high (1111 ± 160 ng/g ww) sum (Σ) PBDE concentrations. Unexpectedly, low in ovo concentrations of total-α-hexabromocyclododecane (HBCD) were detected. Plasma retinol concentrations of adult males exposed to higher DE-71 concentrations were negatively correlated with in ovo ΣPBDE, BDE-100, and HBCD levels. Maternal (13%) and nestling (11%) retinol levels were lower in the low-exposure group compared to respective controls, and biologically significant since their retinol levels were correlated with hatching success and growth, respectively. Maternal retinol levels were also correlated with BDE-153. The underlying mechanisms may involve (1) PBDE exposure, hydroxylated (OH-) metabolites, and subsequent changes in retinol mobilization; (2) decreased maternal food consumption; and (3) reduced maternal retinol yolk deposits. The apparent lack of retinol changes in the high-exposure kestrel may reflect compensation occurring, either by increased mobilization and transportation of retinol, and/or higher food consumption in these birds. When highly mobile as evidenced during reproduction or development, retinol concentrations of adult and nestling kestrels are sensitive to environmentally relevant PBDE and HBCD levels.

Concentrations and spatial patterns of persistent organic (chlorinated) pollutants (POPs), polybrominated diphenyl ether (PBDE), hexabromocyclododecane (HBCD), other flame retardants, and hydroxylated (OH) PBDE and PCB compounds were determined in nestling peregrine falcons across the Canadian Great Lakes Basin. The highest geometric mean plasma POP concentrations (ng/g ww) were sum (Sigma)PCBs (35.16), SigmaPBDEs (15.38), and SigmaOH-PCB (8.77) with the lowest mean levels in nestlings from urban versus remote nests. PBDE congeners derived from PentaBDE and OctaBDE technical mixtures had the highest concentrations, sometimes exceeding 100 ng/g wet weight (ww); BDE-99, -153, -47, -100, and -183 comprised 92.7% of the Sigma(14)PBDE levels. BDE-209 proportions were minimal (1%). North Shore (Lake Superior) nestlings had the highest Sigma(14)PBDE concentrations, with BDE-99, -153, and -47 dominant. Urban nestlings had higher BDE-99:BDE-153 ratios, higher BDE-183 proportions, and the only detectable HBCD concentrations, suggesting greater and more localized exposure to HBCD and PBDEs derived from OctaBDEs. Spatial patterns reflected differences in diet, local contaminant sources, and/or atmospheric deposition. Metabolism of PCBs and PBDEs likely occurred in these nestlings: OH-PCB metabolites were detected, and 4-OH-CB187 was the most abundant of these metabolites. Low ppb levels of putative OH-PBDE metabolites, dominated by 6'-OH-BDE47, were also detected in the peregrine nestlings.

Polybrominated diphenyl ethers (PBDEs) and total alpha-hexabromocyclododecane (HBCD) are flame-retardant additives that are commonly used in household and commercial applications. PBDE congeners, which are comprised of technical mixtures such as DE-71, are globally persistent and their concentrations are increasing in many species. Captive American kestrels (Falco sparverius) were exposed by diet to vehicle (safflower oil), or one of two environmentally relevant concentrations of DE-71 and unintentionally to HBCD. This exposure resulted in the birds laying eggs that contain PBDE and HBCD concentrations currently found in wild herring gull (Larus argentatus) and peregrine falcon (F. peregrinus) eggs, and compared to control kestrels, resulted in delayed egg laying and smaller eggs being laid, caused thinner eggshells and differential weight loss during embryonic development, and reduced fertility and reproductive success. The thickness of the eggshell declined as the concentrations of all measured PBDE and the total amount of a-HBCD congeners (except BDE-183 and BDE-209) increased; increasing concentrations of BDE-153, BDE-154, BDE-28, BDE-17, delayed egg laying, reduced eggshell mass (plus sigmaPBDEs), and reduced fledging success (BDE-153 and BDE-154 only). BDE-153 is the dominant congener recently found in peregrine eggs. The results of this study are consistent with the PBDE-associated brood reduction in wild European peregrines and may partially explain the decline of kestrels in North America.

Captive breeding (n = 25 pairs) and nonbreeding (n = 25) American kestrels were exposed to a mixture of polychlorinated biphenyls (PCBs) (Aroclor 1248:1254:1260) through their diet of day-old cockerels. Kestrels ingested approximately 7 mg/kg body weight each day of PCBs, and this dosage resulted in environmentally relevant total PCB residues in eggs (geometric mean of 34.1 microg/g). An equal number of unexposed birds served as controls. Bare areas of skin known as brood patches function during incubation to warm eggs; therefore, brood patch size could potentially influence hatching success, or patches may be a confounding factor in the relationship between poor incubation behavior and hatching failure observed in birds in toxicological studies. Exposure to PCBs altered the size of brood patches in American kestrels. PCB-exposed male and female nonbreeders had two of three brood patches that were larger than those of control nonbreeders. Breeding males exposed to PCBs had smaller patches than controls, whereas PCB-exposed female kestrels had one larger and one smaller patch than controls. Patch sizes were not related to total PCB residue levels in eggs of exposed birds. Brood patches were not related to various incubation behaviors or hatching success in either control or PCB-exposed kestrels.

Polybrominated diphenyl ether (PBDEs) concentrations are increasing exponentially in biota. We studied the growth of American kestrel ( Falco sparverius ) nestlings exposed in ovo and during development to environmentally relevant PBDE congeners and concentrations. Eggs within each clutch, divided between groups by laying sequence, were injected into the air cell at 19 days of incubation with safflower oil or penta-BDE congeners BDE-47, -99, -100, and -153 dissolved in safflower oil (18.7 μg total [Σ] PBDEs/egg), approximating current levels in Great Lakes herring gulls. The measured proportions of BDE congeners found in the dosing oil were 56.4% of BDE-47; 27.2% of BDE-99; 24.8% of BDE-100; and 0.6% of BDE-153. For 29 days, nestlings were orally gavaged daily with the same ΣPBDE mixture (15.6 ± 0.3 ng/g body weight/day). Relative congener abundances in the dosing mixture compared to the carcasses suggest biotransformation of BDE-47; BDE-183 was also detected. PBDE exposure did not affect hatching or fledging success. PBDE-exposed nestlings were larger (weight, bones, feathers) as they gained weight more quickly and ate more food, the latter in association with their ΣPBDE body burdens. BDE-100 was most influential on nestling growth, being positively associated with size, weight gain, and food consumption. Increasing concentrations of BDE-183 and -153 were related to longer bones, and BDE-99 to longer feathers. The larger size of the PBDE-exposed birds may be detrimental to their bone structure and have excessive energetic costs. The repeated relationships with BDE-100 and growth may be important for wild Falconidae , since this is the predominant penta-BDE congener in these raptors. Copyright© Taylor & Francis Group, LLC.

We collected tissues from herring gulls (Larus argentatus) nesting within and outside of the Great Lakes basin. Genotoxin exposure was assessed as fluorescent aromatic compounds (FACs) in bile and SOS Chromotest-inducing activity in muscle extracts. We determined whether these exposures were associated with decreased erythrocyte DNA strand length and/or induction of hepatic ethoxyresorufin-O-deethylase (EROD) activity. FACs were detected in all bile samples. Most muscle extracts produced a positive or marginal SOS response in the presence of S9. SOS induction potentials were strongly associated with dietary trophic level. The median molecular length of DNA isolated from erythrocytes for 14 of 17 adult and 10 of 11 prefledgling collections was reduced compared to the modal class for their respective age group suggesting widespread DNA damage. DNA damage was greatest in gulls from Saginaw Bay, Lake Huron. Median EROD activity in both adults and prefledglings from remote locations was significantly lower than that of gulls from the lower Great Lakes and was not associated with concentrations of benzo[a]pyrene (B[a]P)-like FACs. Our results indicate Great Lakes herring gulls were exposed to genotoxins and Ah-receptor activating agents in biologically significant concentrations in the early 1990s. These agents appear to be persistent bioaccumulative compounds and/or their metabolites.

We investigated whether exposure to environmentally relevant polybrominated diphenyl ethers (PBDEs) causes immunomodulation in captive nestling American kestrels (Falco sparverius). Eggs within each clutch, divided by laying sequence, were injected with safflower oil or penta-BDE congeners-47, -99, -100, and -153 dissolved in safflower oil (18.7 microg sigmaPBDEs/egg) approximating Great Lakes birds. For 29 days, nestlings consumed the same PBDE mixture (15.6+/-0.3 ng/g body weight per day), reaching sigmaPBDE body burden concentrations that were 120x higher in the treatment birds (86.1+/-29.1 ng/g ww) than controls (0.73+/-0.5 ng/g ww). PBDE-exposed birds had a greater PHA response (T-cell-mediated immunity), which was negatively associated with increasing BDE-47 concentrations, but a reduced antibody-mediated response that was positively associated with increasing BDE-183 concentrations. There were also structural changes in the spleen (fewer germinal centers), bursa (reduced apoptosis) and thymus (increased macrophages), and negative associations between the spleen somatic index and sigmaPBDEs, and the bursa somatic index and BDE-47. Immunomodulation from PBDE exposure may be exacerbated in wild birds experiencing greater environmental stresses.

This study found abnormalities in multiple reproductive stages in captive American kestrels (Falco sparverius) when exposed to polychlorinated biphenyls (PCBs) through dietary and in ovo exposure. American kestrels laid eggs with environmentally relevant total PCB levels (34.1 micrograms/g whole egg wet weight) when consuming PCB-spiked (Aroclor 1248:1254:1260) food (5-7 micrograms/g body weight per day) for 100 d only in 1998. In 1999, the same adults laid eggs with estimated total PCBs of 23 micrograms/g. Effects of maternal (only female exposed) and paternal (only male exposed) in ovo PCB exposure were investigated. Maternal F1 eggs contained predicted total PCB concentrations of 0.34 microgram/g. Specific abnormalities occurred more frequently during dietary F0 exposure, particularly aggressive courtship interactions, clutch abandonment, occurrences of cracked eggs, and developmental effects. Multiple developmental effects were more pronounced during than after dietary PCB exposure of adults, and although these effects were limited, nevertheless they occurred in the F1 maternal and F1 paternal pairs. However, the incidence of multiple deformities throughout the breeding season increased dramatically from 1998 (13%) to 1999 (56%) in F0 PCB-exposed pairs. Developmental abnormalities were unlikely to be attributed to the extrinsic factors of disease, genetics, or nutritional (vitamin D3) deficiencies, but rather to adverse changes in parental behavior and intrinsic factors involving altered genetic material and PCB exposure. Readily cleared PCB congeners may induce specific types of behavioral and developmental abnormalities, but persistent congeners and metabolites are likely producing (1) odd laying patterns, (2) odd laying patterns, (2) developmental effects including embryonic underdevelopment and edema, and (3) increased incidences of multiple deformities within a clutch.

The development of second-generation nestling American kestrels (Falco sparverius) was altered by in ovo exposure of only one parent to polychlorinated biphenyls (PCBs). Polychlorinated biphenyls appear to alter nestling development through both maternally and paternally mediated effects. In 1998, F0 parent kestrels consumed approximately 5 to 7 microg total PCBs/g bird/d (Aroclors 1248:1254: 1260) for approximately 100 d prior to eggs hatching; these eggs, containing total PCB concentrations of 34.1 microg/g, produced 13 F1 offspring, which were then paired in 1999 with unexposed kestrels to examine developmental effects of maternal or paternal in ovo PCB exposure. Using a toxicokinetics model, eggs from the maternally exposed group had predicted PCB levels of 0.03 to 0.34 microg/g, with enriched higher chlorinated congeners. Polychlorinated biphenyl concentrations in eggs of all generations have recently been found in eggs and nestlings of free-ranging eagles. Consistent with the first generation, maternally exposed F2 females generally were larger, had altered growth rates, and delayed maximal growth and fledging compared with control females. Maternally exposed F2 males were heavier but had shorter bones, grew more quickly and earlier, and fledged 2 d later than control males. In the maternally exposed group, concentrations of plasma triiodothyronine were elevated in F2 females but suppressed in F2 males. Paternally exposed F2 hatchlings of both sexes were comparable in size to controls with the exception of having longer tarsi bones, but subsequently showed slower, delayed growth (both sexes) and fledging (females) and lower thyroxine concentrations (males). The alterations in thyroid hormones in the F2 generation are discussed in light of the enrichment of higher chlorinated PCB congeners and hydroxylated PCB congeners. The developmental changes in the kestrel nestlings are likely a function of several possible mechanisms involving maternal PCB deposition, parental behavior, and neurobehavioral and endocrine-thyroid function in nestlings.

Given factors such as their persistence and toxicity, legacy brominated flame retardants (BFRs) like polybrominated diphenyl ethers (PBDEs) and hexabromocyclododecane (HBCDD), are designated as persistent organic pollutants (POPs) and are subject to regulation. Waste streams likely represent a substantial reservoir of legacy BFRs given that they were once widely applied to goods which are increasingly likely to be obsolete. Waste streams are also increasingly likely to be a source of emerging flame retardants, in particular, novel BFRs (NBFRs), the halogenated norbornene flame retardant Dechlorane Plus (DDC-CO) and the brominated, chlorinated or non-halogenated organophosphate triester flame retardants (PFRs). Many bird populations rely on landfill and its surrounding land-use for inter alia the opportunities it provides for activities such as foraging and resting. However, studies on captive and wild (free-living) birds have demonstrated deleterious effects of several FRs. Globally, approximately 250 bird species, including many of conservation concern, are reported to use landfill and surrounding habitat (including wastewater treatment operations), thus putting birds potentially at risk of exposure to such chemicals. We synthesise and critically evaluate a total of 18 studies covering eight avian species published between 2008 and 2018 (inclusive) across four continents that report flame retardant (FR) burdens in birds utilising landfill. Several such studies found FRs at among the highest concentrations detected in wild biota to date. We recommend that ongoing research be focused on landfill-associated birds, given that landfill is an important source of FRs and other anthropogenic chemicals, and particularly at sites where species are of conservation concern. We suggest ways in which the comparative power of studies could be enhanced in the future, the reporting of a minimum common suite of key chemicals, and where feasible, standardisation of the tissue compartments (i.e., eggs) to be studied. We conclude by identifying future research directions.
