Aneuploidy has previously been observed in the Pacific oyster, Crassostrea gigas, and shown to be negatively correlated with growth. Moreover, a significant impact of atrazine exposure has been described in C. gigas, and persistence of that effect has been observed between generations. Evidence of differential chromosome loss has been demonstrated in aneuploid karyotypes of C. gigas using the G-banding technique. Pairs 1, 5, 9, and 10 are characterized by the loss of 1 chromosome. As restriction enzyme (RE) digestion chromosome banding allows a better identification of chromosome pairs, we used this technique to identify which chromosomes are affected when aneuploidy is increased by exposure to atrazine. The progeny of oysters contaminated by atrazine were analysed using the restriction enzyme HaeIII. The study of 26 RE-banded aneuploid karyotypes showed that the same chromosome pairs (1, 5, 9, and 10) were affected by the loss of 1 chromosome (61%, 15%, 42%, and 42%, respectively). Further investigation is required to enable a better understanding of aneuploidy in oysters, especially with respect to why some chromosomes are more easily lost than others, and why cells tolerate the loss of these chromosomes.

Aneuploidy has previously been described and studied in the Pacific oyster, Crassostrea gigas, and has been shown to be negatively correlated with growth. The present study investigated the effect of atrazine on the level of aneuploidy in this species. Crassostrea gigas adults and juveniles were subjected to different concentrations of atrazine representing a peak value found in a polluted environment (46.5 nM) and a value 10 times higher (465 nM). Although atrazine did not show any effect on the oyster mortality, significant differences in aneuploidy level were observed between the different treatments (9% for the control, 16% for 46.5 nM and 20% for 465 nM atrazine). Moreover, the same levels of aneuploidy were observed at adult and juvenile stages. This is the first reported evidence for an environmental effect on aneuploidy in C. gigas. These results will be useful for the oyster aquaculture industry and management of resources. The lowest atrazine level in the current study represents realistic potential exposure, and the results suggest that studies should be made on other aquatic species at risk of exposure to atrazine in the wild. This widely used compound may be an important factor causing damage to genetic material.

The Qatari mangroves of Al-Khor are being increasingly exposed to a wide variety of anthropogenic pollutants due to land reclamation and urban expansion. In this study, we evaluated the lethal and genotoxic effects of methylmercury, trace metals, and organotins, assessing mortality and aneuploidy levels (abnormal number of chromosomes) in the endemic shrimp Palaemon khori under laboratory conditions. In the experimental design, two different concentrations were used for each family of contaminant (single or combined): an environmental concentration equivalent to the maximum value reported in the environment and a value ten times higher, for a period of eight weeks. Survival decreased significantly when pollutants were administrated in combination, even at environmental concentrations (as shown by Cox proportional hazards ratios): similar levels of mortality would be reached by individual type of pollutants only at ten times the environmental concentration. This critical result, under controlled lab conditions, highlights the importance of monitoring mixtures of contaminant types over single ones in the marine environment. Aneuploidy was reported in all treatments and control ranging from 5% to 19% at week four and from 7% to 21% at week eight. All treatments presented significantly higher aneuploidy levels when compared to the control. However, no significant difference was observed between the two time periods, even though 30% of the treatments could not be assessed at week eight, as not enough animals were still alive. In conclusion, the use of endemic species should be considered a valuable tool to determine local perturbations, representing a regional bioindicator of multiple environmental stressors from the initial stages of contamination. Cytogenetic abnormalities associated with viral infections, including from viruses of the Herpesvirales order, have been reported in vertebrate species. Ostreid herpesvirus 1 (Os HV-1) has been detected worldwide during mortality outbreaks of the Pacific oyster Crassostrea gigas. On the other hand, a high proportion of aneuploid cells in somatic tissues have been observed in C. gigas. In this study, we analysed the putative association between aneuploidy levels and the detection of Os HV-1 in gills of C. gigas, the Portuguese oyster C. angulata and their F1 hybrids cultured in Ria Formosa (Portugal). Os HV-1 was detected by PCR in 5.4% of the total of oysters analysed ( n = 111) namely in 11.1%, 8.0% and 1.7% of C. gigas, C. angulata and F1 hybrid respectively. Sequencing analysis of a viral fragment amplified with the C2/C6 primer pair revealed a high similarity with the Os HV-1 reference type. Moreover, in situ hybridization confirmed the presence of Os HV-1 in gill tissue. Oysters where Os HV-1 was detected had a significantly higher mean percentage of aneuploid cells (25%) than the ones where the virus was not detected (18%). However, the overall low percentage of positive samples contrasted with the high mean percentage of aneuploidy observed, with 50% of the oysters analysed showing a percentage of aneuploid cells between 20% and 30%. We hypothesize that somatic aneuploidy may adversely affect oysters making them more prone to Os HV-1 infection, but the virus is unlikely to be the cause of somatic aneuploidy. Abstract: The Portuguese oyster, Crassostrea angulata, is taxonomically close to the Pacific oyster, C. gigas, but there are clear genetic and phenotypic differences between these taxa. Among those differences, the faster growth of C. gigas compared with C. angulata has often been observed in the field. Crosses between C. angulata and C. gigas were performed to investigate the relationship between growth variation and somatic aneuploidy at the individual level in the two taxa and their reciprocal hybrids. The different progenies were reared in Ria Formosa (Portugal) under standard farming conditions. Growth rate and survival were significantly higher in C. gigas than in C. angulata, and the hybrids showed intermediate performances. Significant differences were also observed in the proportion of aneuploid cells (PAC) and of missing chromosomes (PMC) between the two taxa, C. angulata showing the highest values. Intermediate values of PAC and PMC were observed in the hybrids, supporting additive genetic bases of these parameters. Our results also confirm the negative correlation between somatic aneuploidy and growth rate at the individual level, as previously reported in C. gigas. [Copyright &y& Elsevier]

Abstract: The carpet shell, Ruditapes decussatus is a species with a high commercial importance in Portugal and other Southern European countries. However, high growth rate variability is a major problem in the aquacultural production of this species. The objective of this work was then to study the possible differences between fast-, medium- and slow-growing individuals at three times of sampling, of a same cohort of R. decussatus in terms of eventual numerical chromosomal changes, aneuploidy, but also the level of physiological capacity, by assessing the condition index and the biochemical composition: proteins, total lipids, neutral lipids, phospholipids, carbohydrates, free reducing sugars, polysaccharides and total energy. Hypoploid cells of 2n=37, 36, 35 or 34 were observed in all groups of individuals. The aneuploidy values observed ranging from 19% to 79%, were substantially higher than those observed in previous studies for other bivalve species. The differences in the observed aneuploidy between fast- and slow-growing clams were highly significant and a negative correlation was also observed between the level of aneuploidy and length. In general, the synthesis of the evaluated biochemical components showed no significant differences among groups suggesting that this synthesis could be related to extrinsic factors or that the analyzed biochemical components were not specific to detect the lack of certain aminoacids and enzymes involved in growth. The only exception was for total lipids, which were significantly lower in the medium and slow growing groups relative to the fast growing group. The evaluation of the relationships between all the studied parameters was performed through a principal component analysis (PCA). The results of this study demonstrate the existence of a negative correlation between somatic aneuploidy and growth in the carpet shell, as previously reported for other bivalve species. Moreover, for the first time in this study, this negative correlation is confirmed over time in a same cohort of the clam R. decussatus. [Copyright &y& Elsevier]

Abstract: Aneuploidy is a cytogenetic phenomenon known as an abnormal diploid chromosome number. A negative relationship between growth rate, one of the biggest problems faced by bivalve producers, and this phenomenon was already verified for two oyster species, the Japanese oyster Crassostrea gigas, the Portuguese oyster Crassostrea angulata and their interspecific hybrids and, more recently, in the carpet shell clam Ruditapes decussatus. The main objective of this study was to assess whether chromosome losses in aneuploid situations could be explained by differential chromosomal susceptibility, as previously reported in the oyster C. gigas. Thereby, we applied the restriction enzyme (RE) digestion chromosome banding technique to aneuploid karyotypes of R. decussatus and C. angulata, in order to identify the missing chromosomes. The results of this study showed that 4 out of the 19 chromosome pairs (viz. 1, 6, 12, and 19) of R. decussatus and 3 out of the 10 chromosome pairs (viz. 1, 9, and 10) of C. angulata were preferentially affected by the loss of one homologous chromosome. The chromosomal loss in C. angulata was very similar to the one previously observed in C. gigas. These results open a new field for further research in order to have a better understanding of the aneuploidy phenomenon in bivalves and particularly its negative relationship with growth rate.
