O abacate é uma fruta tropical muito rica em nutrientes, sua árvore é conhecida como abacateiro, que pertence a família da laureáceas. O Brasil é o terceiro produtor mundial do abacate, possui uma produção de ordem de 500 milhões de fruto por ano. O óleo de abacate é extraído quando os frutos estão maduros, isto é, com consistência mole, que é quando apresentam teores mais elevados de óleo. O principal obstáculo para obtenção do óleo é o alto teor de umidade que afeta o rendimento da extração. O presente trabalho tem por objetivo, otimizar técnicas de extração de óleo da polpa de abacate e assim comparar os resultados com o método recomendado de acordo com AOAC e realizar a determinação de ácidos graxos na polpa de abacate por cromatografia gasosa com detector de ionização de chama. Para tanto, foram utilizadas quatro tipos de extração, a primeira por aparelho Soxhlet variando o solvente; a segunda, por aparelho Soxhlet com adição prévia da enzima celulase; a terceira, por maceração com fricção e a ultima por tratamento em banho de ultra-som. Os rendimentos obtidos foram superiores em até 46,3%(m/m) em comparação com o método recomendado por AOAC, sendo que as extrações que apresentaram melhores resultados foram a maceração por fricção e o tratamento com banho de ultra-som, ambas a partir da amostra com a umidade reduzida em forno microondas. A derivação dos ésteres de ácidos graxos foi feita segundo método de Hartman e Lago. A melhor separação dos ésteres de ácidos graxos estudados foi obtida empregando um sistema de cromatografia gasosa, coluna cromatográfica ZB-5HT - (15m x 0,25mm x 0,25μm) com “Guard Column” de Sílica fundida (5m x 0,52mm)... The oils and the fats have been recognized as essential nutrients in such a way in the animal diets as in the human being ones and they constitute the most known concentrated source of power. The avocado is a very rich tropical fruit in nutrients, its tree is known as avocado tree, that belongs to the laureaceae family. Brazil is the third world-wide producer of avocado, which has a production of about 500 million fruit per year. The avocado oil is extracted when the fruits are mature, that is, with a soft consistency, when they present higher oil contents. The main obstacle for the attainment of the oil is the high content of humidity that affects the income of the extraction. The present project has as an aim, the acid extraction and determination of fatty acid in the pulp of avocado for gaseous chromatography with detector of ionization of flame. For such a way, four types of extraction have been used, the first one for Soxhlet varying the solvent; the second, for Soxhlet with previous addition of the cellulose enzyme; the third, for maceration and the last one for sonication. The amounts of extracted oil have been compared with the gravimetrical method using Soxhlet, recommended in accordance with AOAC. The obtained results have been superior up to 46,3% (m/m), whereas the extractions that presented better results were the maceration and the sonication, both from the dehydrated sample in microwaves. The derivation of esters of fatty acid was made according to the method of Hartman and Lago. The best separation of studied esters of fatty acid was obtained by using the Gaseous Chromatography system, chromatographic column ZB-5HT - (15m x 0,25mm x 0,25μm) with Guard Column of fused silica (5m x 0,52mm) and detector of ionization of flame.
