Despite a myriad of national space agencies, industrial partners, university laboratories, and policy groups preparing for human exploration of the Martian surface, there remains a need for a single reference mission architecture (RMA) that models and captures the vast design parameter space, and hence the complexities, of a Mars human exploration operation. The available literature often focuses on shorter-term, opposition-class exploration missions of approximately 30 days of surface operations, instead of the more probable, longer-term, conjunction-class exploration missions of approximately 500 days of surface operations. A critical aspect of these longer duration missions is determining the food, medicine, and materials that are necessary to support a crew over the specified lengthy time-period. In the following dissertation I demonstrate the progress towards the development of a biomanufactory-driven RMA. A crewed mission to and from Mars may include an exciting array of enabling biotechnologies that leverage inherent mass, power, and volume advantages over traditional abiotic approaches. I begin this dissertation by articulating the scientific and engineering goals and constraints, along with example systems, that guide the design of a surface biomanufactory. Extending past arguments for exploiting stand-alone elements of biology, I argue for an integrated biomanufacturing plant replete with modules for microbial in situ resource utilization, production, and recycling of food, pharmaceuticals, and biomaterials required for sustaining future intrepid astronauts. Here I also discuss aspirational technology trends in each of these target areas in the context of human and robotic exploration missions. I then formalize the mathematical framework for modeling a biomanufacturing system developing the resources for sustaining a human exploration mission on the surface of Mars by establishing mission goals, extending the Equivalent System Mass framework for comparision of missions, develop the framework for modeling a Martian resource inventory in terms of supplies both produced via ISRU processes and transported as cargo from Earth, and develop the framework required for sustaining a human crew in terms of essential resources. Using this collection of frameworks, I develop a software framework to implement and integrate process models that can be experimentally validated by the collaborations of the Center for the Utilization for Biological Engineering in Space, beginning with the crop cultivation models for food consumption and pharmaceutical development for astronauts. Finally, I presents an argument for how Space Bioprocess Engineering drives sustainability on- and off-World. Although raison d'etre of Space Bioprocess Engineering is the design, realization, and management of biologically-driven technologies for supporting offworld human exploration, it has the potential to offer transformative solutions to the global community in pursuit of the United Nations Sustainable Development Goals. Here we address the growing sentiment that investment in spacefaring enterprises should be redirected towards sustainability programs. In outlining the Earth-benefits of dual-use Space Bioprocess Engineering technologies, we both show that continued investment is justified and offer insight into specific R&D strategies.

Phase separation in multiphase flows in space systems is a challenging task due to the absence of buoyancy. Several phase separation approaches have been presented in the last years given the important role that systems containing multiphase flows will play in future space missions. We present a review of these techniques and a trade-off analysis for their application in life support systems and, in particular, in the MELiSSA (Micro Ecological Life Support System Alternative) photosynthesis bioreactor. The candidate approaches are evaluated, both quantitatively and qualitatively, according to the defined requirements and criteria. The outcome of the trade-off analysis shows passive static and acoustic techniques as the most recommended methods to carry out phase separation in the considered bioreactor.

The cited materials show the use of oxygen, hydrogen, liquefied natural gases (methane) and fluorine as components of the fuel for liquid-propellant rocket engines (LRE). The reasons for the need to use oxygen as an oxidizing agent are indicated. The advantages and disadvantages are disclosed from the point of view of using the listed components as fuel elements for liquid-propellant rocket engines. The issues of ecology when using the considered fuels are reviewed. Shown not only the use of cryogenic components as fuel for LRE, but also in life support systems in manned spacecraft in space research.

This paper is aimed at demonstrating the possibility of growing a tomato ideotype, fortified in anti-oxidant content (derived from Micro-Tom, a model cultivar for tomato research overexpressing anthocyanins) and specifically developed for spatial environment, in a seed-to-seed cycle (70-90 days) on a CubeSat. To reach this goal, a dedicated micro satellite equivalent to 12 U will be developed to be sent into low-orbit. Growing plants in space is a prerequisite to sustain long-term human exploration of the solar system. Plants can increase the independence of a space mission providing astronauts with food, oxygen, waste recycling, water purification increasing quality of life. Preliminary experimental results to simulate the low orbit conditions are here described, together with all the devices used during the test activities. © 2018 IEEE.

Space exploration programmes, currently under discussion in the US and in Europe, foresee human missions to Mars to happen within the first half of this century. In this context, the European Space Agency (ESA) has conducted a study on the human responses, limits and needs for such exploratory missions, the so-called HUMEX study (ESA SP-1264). Based on a critical assessment of the limiting factors for human health and performance and the definition of the life science and life support requirements performed in the frame of the HUMEX study, the following major critical items have been identified: (i) radiation health risks, mainly occurring during the interplanetary transfer phases and severely augmented in case of an eruption of a solar particle event; (ii) health risks caused by extended periods in microgravity with an unacceptable risk of bone fracture as a consequence of bone demineralisation; (iii) psychological risks as a consequence of long-term isolation and confinement in an environment so far not experienced by humans; (iv) the requirement of bioregenerative life support systems complementary to physico-chemical systems, and of in situ resource utilisation to reach a closure of the life support system to the highest degree possible. Considering these constraints, it has been concluded that substantial research and development activities are required in order to provide the basic information for appropriate integrated risk managements, including efficient countermeasures and tailored life support. Methodological approaches should include research on the ISS, on robotic precursors missions to Mars, in ground-based simulation facilities as well as in analogue natural environments on Earth.

The designation of a simple, non-invasive, and highly precise method to monitor the thermal status of astronauts is important to enhance safety during extravehicular activities (EVA) and onboard emergencies. Finger temperature (Tfing), finger heat flux, and indices of core temperature (Tc) [rectal (Tre), ear canal (Tec)] were assessed in 3 studies involving different patterns of heat removal/insertion from/to the body by a multi-compartment liquid cooling/warming garment (LCWG). Under both uniform and nonuniform temperature conditions on the body surface, Tfing and finger heat flux were highly correlated with garment heat flux, and also highly correlated with each other. Tc responses did not adequately reflect changes in thermal balance during the ongoing process of heat insertion/removal from the body. Overall, Tfing/finger heat flux adequately reflected the initial destabilization of thermal balance, and therefore appears to have significant potential as a useful index for monitoring and maintaining thermal balance and comfort in extreme conditions in space as well as on Earth.

The Sasakawa International Center for Space Architecture (SICSA) is undertaking a multi-year research, planning and design study that is exploring near- and long-term commercial space development opportunities. The central goal of this activity is to conceptualize a scenario of sequential, integrated private enterprise initiatives that can carry humankind forward to Mars. Each development stage is planned as a building block to provide the economic foundation, technology advancements and operational infrastructure to support others that follow. This report presents fundamental issues and requirements associated with planning human Mars initiatives that can transfer crews, habitats and equipment from Earth to Mars orbit, deliver them to the planet's surface, and return people and samples safely back to Earth. The study builds in part upon previous studies which are summarized in SICSA's: Commercial Space Development Plan and the Artificial Gravity Science and Excursion Vehicle reports. Information and conclusions produced in this study provide assumptions and a conceptual foundation for a subsequent report titled The First Mars Outpost: Planning and Concepts.

Use of halophytes (salt-tolerant vegetation), in a particular vegetable Salicornia europaea plants which are capable of utilizing NaCl in rather high concentrations, is one of possible means of NaCl incorporation into mass exchange of bioregenerative life support systems. In preliminary experiments it was shown that S. europaea plants, basically, could grow on urine pretreated with physicochemical processing and urease-enzyme decomposing of urea with the subsequent ammonia distillation. But at the same time inhibition of the growth process of the plants was observed. The purpose of the given work was to find out the influence of excessive quantities of some mineral elements contained in products of physicochemical processing of urine on the production process and NaCl accumulation by S. europaea plants. As the content of mineral salts in the human liquid wastes (urine) changed within certain limits, two variants of experimental solutions were examined. In the first variant, the concentration of mineral salts was equivalent to the minimum salt content in the urine and was: K – 1.5 g/l, P – 0.5 g/l, S – 0.5 g/l, Mg – 0.07 g/l, Ca – 0.2 g/l. In the second experimental variant, the content of mineral salts corresponded to the maximum salt content in urine and was the following: K – 3.0 g/l, P – 0.7 g/l, S – 1.2 g/l, Mg – 0.2 g/l, Ca – 0.97 g/l. As the control, the Tokarev nutrient solution containing nitrogen in the form of a urea, and the Knop nutrient solution with nitrogen in the nitrate form were used. N quantity in all four variants made up 177 mg/l. Air temperature was 24 °C, illumination was continuous. Light intensity was 690 μmol/m 2 s of photosynthetically active radiation. NaCl concentration in solutions was 1%. Our researches showed that the dry aboveground biomass of an average plant of the first variant practically did not differ from the control and totaled 11 g. In the second variant, S. europaea productivity decreased and the dry aboveground biomass of an average plant totaled 8 g. The increase of K quantity in the experimental solutions resulted in an elevated content of the element in the plants. The increase of K uptake in the second experimental variant was accompanied by a 30–50% decrease of Na content in comparison with the other variants. Comparative Na content in the other variants was practically identical. N, Mg and P content in the control and experimental variants was also practically identical. The increase of S quantity in the second experimental variant also increased S uptake by the plants. But Ca quantity, accumulated in aboveground plants biomass in the experimental variants was lower than in the control. NaCl uptake by plants, depending on the concentration of mineral salts in the experimental solutions, ranged from 8 g (maximum salt content) up to 15 g (minimum salt content) on a plant growth area that totaled 0.032 m 2 . Thus, high concentrations of mineral salts simulating the content of mineral salts contained in urine did not result in a significant decrease of S. europaea productivity. The present work also considers the influence of higher light intensity concentrations on productivity and NaCl accumulation by S. europaea plants grown on experimental solutions with high salt content.

Microalgae culture is likely to play an important role in aquatic food production modules in bioregenerative systems for producing feeds for fish, converting CO2 to O2 and remedying water quality as well as aquatic higher plants. In the present study, the effects of culture conditions on the cellular multiplication of microalgae, Euglena gracilis, was investigated as a fundamental study to determine the optimum culture conditions for microalgae production in aquatic food production modules including both microalgae culture and fish culture systems. E. gracilis was cultured under conditions with five levels of temperatures (25-33 degrees C), three levels of CO2 concentrations (2-6%), five levels of O2 concentrations (10-30%), and six levels of photosynthetic photon flux (20-200 micromoles m-2 s-1). The number of Euglena cells in a certain volume of solution was monitored with a microscope under each environmental condition. The multiplication rate of the cells was highest at temperatures of 27-31 degrees C, CO2 concentration of 4%, O2 concentration of 20% and photosynthetic photon flux of about 100 micromoles m-2 s-1. The results demonstrate that E. gracilis could efficiently produce biomass and convert CO2 to O2 under relatively low light intensities in aquatic food production modules.

This report summarizes a trade study of different options of a bioregenerative Life Support System (LSS) and a subsequent conceptual design of a hybrid LSS. The evaluation was based mainly on the terrestrial testbed projects MELISSA (ESA) and BIOS (Russia). In addition, some methods suggested by the Advanced Life Support Project (NASA) were considered. Computer models, including mass flows were established for each of the systems with the goal of closing system loops to the extent possible. In order to cope with the differences in the supported crew size and provided nutrition, all systems were scaled for supporting a crew of six for a 780 day Mars mission (180 days transport to Mars; 600 days surface period) as given in the NASA Design Reference Mission Scenario [Hoffman, S.J., Kaplan, D.L. Human exploration of Mars: the Reference Mission of the NASA Mars Exploratory Study, 1997]. All models were scaled to provide the same daily allowances, as of calories, to the crew. Equivalent System Mass (ESM) analysis was used to compare the investigated system models against each other. Following the comparison of the terrestrial systems, the system specific subsystem options for Food Supply, Solid Waste Processing, Water Management and Atmosphere Revitalization were evaluated in a separate trade study. The best subsystem technologies from the trade study were integrated into an overall design solution based on mass flow relationships. The optimized LSS is mainly a bioregenerative system, complemented by a few physico-chemical elements, with a total ESM of 18,088 kg, which is about 4 times higher than that of a pure physico-chemical LSS, as designed in an earlier study.

Using biotic turnover of substances in trophic chains, natural and artificial ecosystems are similar in functioning, but different in structure. It is necessary to have quantitative criteria to evaluate the efficiency of artificial ecosystems (AES). These criteria are dependent on the specific objectives for which the AES are designed. For example, if AES is considered for use in space, important criteria are efficiency in use of mass, power, volume (size) and human labor and reliability. Another task involves the determination of quantitative criteria for the functioning of natural ecosystems. To solve the problem, it is fruitful to use a hierarchical approach suitable for both individual links and the ecosystem as a whole. Energy flux criteria (principles) were developed to estimate the functional activities of biosystems at the population, community and ecosystem levels. A major feature of ecosystems as a whole is their biotic turnover of matter the rate of which is restricted by the lack of limiting substances. Obviously, the most generalized criterion is to take into account the energy flux used by the biosystem and the quantity of limiting substance included in its turnover. The use of energy flux by ecosystem, E USED – is determined from the photoassimilation of CO 2 by plants (per time unit). It can be approximately estimated as the net primary production of photosynthesis (NPP). So, the ratio of CO 2 photoassimilation rate (sometimes, measured as NPP) to the total mass of limiting substrate can serve as a main universal criterion (MUC). This MUC characterizes the specific cycling rate of limiting chemical elements in the system and effectiveness of every ecosystem including the global Biosphere. Comparative analysis and elaboration of quantitative criteria for estimation of natural and artificial ecosystems activities is of high importance both for theoretical considerations and for real applications.

The problem of interaction between man and microorganisms in closed habitats is an inextricable part of the whole problem of co-existence between macro- and microorganisms. Concerning the support of human life in closed habitat, we can, conventionally, divide microorganisms, acting in life support system (LSS) into three groups: useful, neutral and harmful. The tasks, for human beings for optimal coexistence with microhabitants seem to be trivial: (1) to increase the activity of useful forms, (2) decrease the activity harmful forms, (3) not allow the neutral forms to become the harmful ones and even to help them to gain useful activity. The task of efficient management and control of microbial population's development in LSS highly depends on mission duration. As for short-term missions without recycling, the proper hygienic procedures are developed. For longer missions, the probability of transformation of the neutral forms into the harmful ones is becoming more dangerous. The LSS for long-term missions are to use cycling-recycling systems, including system with biological recycling. In these systems, microbial populations as regenerative link should be useful and active agents. Some problems of microbial populations control and management are discussed in the paper.

A “producer–consumer” (Chlorella vulgaris–Paramecium caudatum) closed aquatic system has been investigated experimentally and theoretically. It has been found that there is a direct relationship between the growth of the paramecia population and their release of ammonia nitrogen, which is the best form of nitrogen for Chlorella growth. The theoretical study of a model of a “producer–consumer” aquatic biotic cycle with spatially separated compartments has confirmed the contribution of paramecia to nitrogen cycling. It has been shown that an increase in the concentration of nitrogen released as metabolites of paramecia is accompanied by an increase in the productivity of microalgae.

In this paper, the influence of the extent of openness of ecosystem that is defined by the dilution rate, which characterizes the extent of flowage of the pond, on the intensity of the biotic circulation in ecosystems with different regulation types, number of trophic links and extent of closing has been investigated. We considered open systems, we took into account the return of the limiting substances, such as nitrogen and phosphorous, into the cycle by degradation of detritus and products of vital functions of consumers. It was shown by the numerical calculations that the increase of the dilution rate in without recycle ecosystems leads to increase of the net primary production up to the maximum value corresponding to the two-link trophic chain (biogenic substance and producer) and then, to gradually decrease. The residual concentration of biogenic limiting substances monotone increases. Net primary production and residual concentration of biogenic limiting substances in systems with recycle with even number of links behaves similarly to that in without recycle ecosystems. In the systems with recycle with the odd number of links that values lies on the stable level. We showed that in wide range of the dilution rate the recycling of the ecosystem can highly increase the net primary production and reduce residual concentration of biogenic limiting substances. The influence of the dilution rate on numbers of links that may exist in the system was analyzed.

Studying material transformations and biotic cycling in artificial ecosystems (AES), we need to know the principles of biological adaptation of active organisms to change in the environment. Microorganisms in AES for water purification are the most active transforming organisms and consumers of the organic substances contained in wastes. Utilization of organic substances is directly connected with the energy fluxes used by AES. According to energy criteria, the energy fluxes used by a biological system tend to reach maximum values under stable conditions. Unutilized substrate concentration decreases as a result of biological adaptations. After a dramatic change in environmental factors, for example, after a sharp increase in the flow rate of organic substances, the biological system is not able to react quickly. The concentration of unutilized substrate increases and the energy flux used by the biological system decreases. The structure of the microbial community also changes, with a decrease in biological diversity. The efficiency of energy use by simple terrestrial ecosystems depends on the energetic intensity and interactions between plants and rhizospheric microorganisms.

In wheat (Triticum aestivum L.) plants, deficiency of an essential element may drastically affect growth, appearance, and most importantly yield. Wheat, the focus of this study, is one of the crops studied in the CELSS program. Information about nutrient deficiencies in crops grown in controlled environment is essential to optimize food productivity. The main objective of this study was to determine whether deficiency of Nitrogen (N), Phosphorus (P), Potassium (K), Calcium (Ca) and Magnesium (M) alters spectral reflectance properties of wheat leaves. Plants were grown in the greenhouse and growth chamber, in a modified Hoagland's nutrient solution. Spectral reflectance of fully expanded wheat leaves from 280 to 1100 nm, nutrient concentrations (N, P, K, and Ca) and chlorophyll (Chl) were determined when deficiency symptoms were first evident (approximately 6-7 weeks). Chlorophyll content and fresh and dry weight, were used to assess the severity of the nutrient stress. All nutrient deficiencies affected chlorophyll content and generally increased reflectance in the visible (VIS) 400-700 nm and infrared (IR) 700-1100 nm ranges. Magnesium and nitrogen deficiencies had the most pronounced effect on chlorophyll concentration height, and reflectance. All macronutrient deficiencies tested reduced chlorophyll concentration, increase reflectance in the visible range and caused a shift in the position of the red edge (the point of maximum slope on the reflectance spectrum of vegetation between red and near-infrared wavelengths) toward shorter or longer wavelengths; depending upon the element. In the greenhouse, N and Mg induced the greatest increase in reflectance of 33% and 25% in the VI range and 86% and 53% in the IR range, respectively. However, in the growth chamber, an increase of 97% and 25% occurred in the VI range, and 20% and 33% in the IR range, respectively. In the IR range in the growth chamber, P, K, and Ca deficiency caused a reduction in reflectance (412-770 nm). This research indicates that mineral deficiencies and reflectance are not specific to one environment and could have important implications for the design of CELSS in space, and perhaps the future of terrestrial agriculture.

During the years 2002 and 2003, three closed system experiments were carried out in the "Laboratory Biosphere" facility located in Santa Fe, New Mexico. The program involved experimentation of "Hoyt" Soy Beans, (experiment #1) USU Apogee Wheat (experiment #2) and TU-82-155 sweet potato (experiment #3) using a 5.37 m2 soil planting bed which was 30 cm deep. The soil texture, 40% clay, 31% sand and 28% silt (a clay loam), was collected from an organic farm in New Mexico to avoid chemical residues. Soil management practices involved minimal tillage, mulching, returning crop residues to the soil after each experiment and increasing soil biota by introducing worms, soil bacteria and mycorrhizae fungi. High soil pH of the original soil appeared to be a factor affecting the first two experiments. Hence, between experiments #2 and #3, the top 15 cm of the soil was amended using a mix of peat moss, green sand, humates and pumice to improve soil texture, lower soil pH and increase nutrient availability. This resulted in lowering the initial pH of 8.0-6.7 at the start of experiment #3. At the end of the experiment, the pH was 7.6. Soil nitrogen and phosphorus has been adequate, but some chlorosis was evident in the first two experiments. Aphid infestation was the only crop pest problem during the three experiments and was handled using an introduction of Hyppodamia convergens. Experimentation showed there were environmental differences even in this 1200 cubic foot ecological system facility, such as temperature and humidity gradients because of ventilation and airflow patterns which resulted in consequent variations in plant growth and yield. Additional humidifiers were added to counteract low humidity and helped optimize conditions for the sweet potato experiment. The experience and information gained from these experiments are being applied to the future design of the Mars On Earth(R) facility (Silverstone et al., Development and research program for a soil-based bioregenerative agriculture system to feed a four person crew at a Mars base, Advances in Space Research 31(1) (2003) 69-75; Allen and Alling, The design approach for Mars On Earth(R), a biospheric closed system testing facility for long-term space habitation, American Institute of Aeronautics and Astronautics Inc., IAC-02-IAA.8.2.02, 2002).

Discussions of future human expeditions into the solar system generally focus on whether the next explorers ought to go to the Moon or to Mars. The only mission scenario developed in any detail within NASA is an expedition to Mars with a 500-day stay at the surface. The technological capabilities and the operational experience base required for such a mission do not now exist nor has any self-consistent program plan been proposed to acquire them. In particular, the lack of an Abort-to-Earth capability implies that critical mission systems must perform reliably for 3 years or must be maintainable and repairable by the crew. As has been previously argued, a well-planned program of human exploration of the Moon would provide a context within which to develop the appropriate technologies because a lunar expedition incorporates many of the operational elements of a Mars expedition. Initial lunar expeditions can be carried out at scales consistent with the current experience base but can be expanded in any or all operational phases to produce an experience base necessary to successfully and safely conduct human exploration of Mars.

The top priority in America's manned space flight program is the assurance of crew and vehicle safety. This priority gained greater focus during and after the Space Shuttle return-to-flight mission (STS-26). One of the interesting challenges has been to assure crew safety and adequate protection of the Space Shuttle, as a national resource, from increasingly diverse cargoes and operations. The control of hazards associated with the deployment of complex payloads and cargoes has involved many international participants. These challenges are examined in some detail along with examples of how crew safety has evolved in the manned space program and how the international partners have addressed various scenarios involving control and mitigation of potential hazards to crew and vehicle safety.

In order to evaluate the effects of gravity on growing plants, we conducted ground based long-term experiments with dwarf wheat, cultivar Apogee and Chinese cabbage, cultivar Khibinskaya. The test crops had been grown in overhead position with HPS lamp below root module so gravity and light intensity gradients had been in opposite direction. Plants of the control crop grew in normal position under the same lamp. Both crops were grown on porous metallic membranes with stable −1 kPa matric potential on their surface. Results from these and other studies allowed us to examine the differences in growth and development of the plants as well as the root systems in relation to the value of the gravity force influence. Dry weight of the roots from test group was decreased in 2.5 times for wheat and in 6 times – at the Chinese cabbage, but shoot dry biomass was practically same for both test and control versions. A harvest index of the test plants increased substantially. The data shows, that development of the plants was essentially changed in microgravity. The experiments in the space greenhouse Svet aboard the Mir space station proved that it is possible to compensate the effects of weightlessness on higher plants by manipulating gradients of environmental parameters (i.e. photon flux, matric potential in the root zone, etc.). However, the average productivity of Svet concerning salad crops even in ground studies did not provide more than 14 g fresh biomass per day. This does not provide a sufficient level of supplemental nutrients to the crew of the ISS. A cylindrical design of a space plant growth chamber (SPGC) allows for maximal productivity in presence of very tight energy and volume limitations onboard the ISS and provides a number of operational advantages. Productivity from this type of SPGF with a 0.5 kW energy utilization when salad growing would provide approximately 100 g of edible biomass per day, which would almost satisfy requirements for a crew of two in vitamin C and carotene and partly vitamin B group as well as rough fiber.

Throughout the evolution process, Earth's magnetic field (MF, about 50 microT) was a natural component of the environment for living organisms. Biological objects, flying on planned long-term interplanetary missions, would experience much weaker magnetic fields, since galactic MF is known to be 0.1-1 nT. However, the role of weak magnetic fields and their influence on functioning of biological organisms are still insufficiently understood, and is actively studied. Numerous experiments with seedlings of different plant species placed in weak magnetic field have shown that the growth of their primary roots is inhibited during early germination stages in comparison with control. The proliferative activity and cell reproduction in meristem of plant roots are reduced in weak magnetic field. Cell reproductive cycle slows down due to the expansion of G1 phase in many plant species (and of G2 phase in flax and lentil roots), while other phases of cell cycle remain relatively stable. In plant cells exposed to weak magnetic field, the functional activity of genome at early pre-replicate period is shown to decrease. Weak magnetic field causes intensification of protein synthesis and disintegration in plant roots. At ultrastructural level, changes in distribution of condensed chromatin and nucleolus compactization in nuclei, noticeable accumulation of lipid bodies, development of a lytic compartment (vacuoles, cytosegresomes and paramural bodies), and reduction of phytoferritin in plastids in meristem cells were observed in pea roots exposed to weak magnetic field. Mitochondria were found to be very sensitive to weak magnetic field: their size and relative volume in cells increase, matrix becomes electron-transparent, and cristae reduce. Cytochemical studies indicate that cells of plant roots exposed to weak magnetic field show Ca2+ over-saturation in all organelles and in cytoplasm unlike the control ones. The data presented suggest that prolonged exposures of plants to weak magnetic field may cause different biological effects at the cellular, tissue and organ levels. They may be functionally related to systems that regulate plant metabolism including the intracellular Ca2+ homeostasis. However, our understanding of very complex fundamental mechanisms and sites of interactions between weak magnetic fields and biological systems is still incomplete and still deserve strong research efforts.

Laboratory Biosphere is a 40 m3 closed life system that commenced operation in May 2002. Light is from 12,000 W of high pressure sodium lamps over planting beds with 5.37 m2 of soil. Water is 100% recycled by collecting condensate from the temperature and humidity control system and mixing with leachate collected from under the planting beds. Atmospheric leakage was estimated during the first closure experiment to be 0.5-1% per day in general plus about 1% for each usage of the airlock door. The first trial run of 94 days was with a soybean crop grown from seeds (May 17, 2002) to harvest (August 14, 2002) plus 5 days of post-harvest closure. The focus of this initial trial was system testing to confirm functionality and identify any necessary modifications or improvements. This paper describes the organizational and physical features of the Laboratory Biosphere.

This article studies the efficiency of ejecting waste generated by the life support system (LSS) of a manned spacecraft to reduce initial mass on low earth orbit. The spacecraft is used for a long-duration interplanetary mission and is equipped with either a chemical or a nuclear-thermal propulsion system. For this study we simulate an optimal control problem for a given spacecraft maneuver. An impulsive approximation of the optimal interplanetary spacecraft trajectory is assumed, which allows us to reduce the general optimal control problem to hierarchic structure of 'outer' and 'inner' subproblems. This structure is analyzed using the Pontryagin's Maximum principle. Numerical results, illustrating the efficiency of waste ejection are shown for typical Earth-Mars transfer trajectories. This results confirm in theory that using a waste ejection system makes an early manned Mars mission possible without having to design and build new, advanced biological LSS.

System-level analyses for Advanced Life Support require mathematical models for various processes, such as for biomass production and waste management, which would ideally be integrated into overall system models. Explanatory models (also referred to as mechanistic or process models) would provide the basis for a more robust system model, as these would be based on an understanding of specific processes. However, implementing such models at the system level may not always be practicable because of their complexity. For the area of biomass production, explanatory models were used to generate parameters and multivariable polynomial equations for basic models that are suitable for estimating the direction and magnitude of daily changes in canopy gas-exchange, harvest index, and production scheduling for both nominal and off-nominal growing conditions. c2004 COSPAR. The different advances in the Micro Ecological Life Support System Alternative project (MELISSA), fostered and coordinated by the European Space Agency, as well as in other associated technologies, are integrated and demonstrated in the MELISSA Pilot Plant laboratory. During the first period of operation, the definition of the different compartments at an individual basis has been achieved, and the complete facility is being re-designed to face a new period of integration of all these compartments. The final objective is to demonstrate the potentiality of biological systems such as MELISSA as life support systems. The facility will also serve as a test bed to study the robustness and stability of the continuous operation of a complex biological system. This includes testing of the associated instrumentation and control for a safe operation, characterization of the chemical and microbial safety of the system, as well as tracking the genetic stability of the microbial strains used. The new period is envisaged as a contribution to the further development of more complete biological life support systems for long-term manned missions, that should be better defined from the knowledge to be gained from this integration phase. The cost of keeping people alive in space is assessed from a theoretical viewpoint and using two actual designs for plant growth systems. While life support is theoretically not very demanding, our ability to implement life support is well below theoretical limits. A theoretical limit has been calculated from requirements and the state of the art for plant growth has been calculated using data from the BIO-Plex PDR and from the Cornell CEA prototype system. The very low efficiency of our current approaches results in a high mission impact, though we can still see how to get a significant reduction in cost of food when compared to supplying it from Earth. Seeing the distribution of costs should allow us to improve our current designs.

In designing innovative space plant growth facilities (SPGF) for long duration space flight, various limitations must be addressed including onboard resources: volume, energy consumption, heat transfer and crew labor expenditure. The required accuracy in evaluating onboard resources by using the equivalent mass methodology and applying it to the design of such facilities is not precise. This is due to the uncertainty of the structure and not completely understanding the properties of all associated hardware, including the technology in these systems. We present a simple criteria of optimization for horticultural regimes in SPGF: Q max = max [ M · (EBI) 2 /( V · E · T ) ], where M is the crop harvest in terms of total dry biomass in the plant growth system; EBI is the edible biomass index (harvest index), V is volume occupied by the crop; E is the crop light energy supply during growth; T is the crop growth duration. The criterion reflects directly on the consumption of onboard resources for crop production.

A major challenge of designing a bioregenerative life support system for Mars is the reduction of the mass, volume, power, thermal and crew-time requirements. Structural mass of the greenhouse could be saved by operating the greenhouse at low atmospheric pressure. This paper investigates the feasibility of this concept. The method of equivalent system mass is used to compare greenhouses operated at high atmospheric pressure to greenhouses operated at low pressure for three different lighting methods: natural, artificial and hybrid lighting.

The paper considers problems of biosynthesis of higher plants' biomass and “biological incineration” of plant wastes in a working physical model of biological LSS. The plant wastes are “biologically incinerated” in a special heterotrophic block involving Californian worms, mushrooms and straw. The block processes plant wastes (straw, haulms) to produce soil-like substrate (SLS) on which plants (wheat, radish) are grown. Gas exchange in such a system consists of respiratory gas exchange of SLS and photosynthesis and respiration of plants. Specifics of gas exchange dynamics of high plants — SLS complex has been considered. Relationship between such a gas exchange and PAR irradiance and age of plants has been established. Nitrogen and iron were found to the first to limit plants' growth on SLS when process conditions are deranged. The SLS microflora has been found to have different kinds of ammonifying and denitrifying bacteria which is indicative of intensive transformation of nitrogen-containing compounds. The number of physiological groups of microorganisms in SLS was, on the whole, steady. As a result, organic substances — products of exchange of plants and microorganisms were not accumulated in the medium, but mineralized and assimilated by the biocenosis. Experiments showed that the developed model of a man-made ecosystem realized complete utilization of plant wastes and involved them into the intrasystem turnover.

The European Space Agency has recently initiated a study of the human responses, limits and needs with regard to the stress environments of interplanetary and planetary missions. Emphasis has been laid on human health and performance care as well as advanced life support developments including bioregenerative life support systems and environmental monitoring. The overall study goals were as follows: (i) to define reference scenarios for a European participation in human exploration and to estimate their influence on the life sciences and life support requirements; (ii) for selected mission scenarios, to critically assess the limiting factors for human health, wellbeing, and performance and to recommend relevant countermeasures; (iii) for selected mission scenarios, to critically assess the potential of advanced life support developments and to propose a European strategy including terrestrial applications; (iv) to critically assess the feasibility of existing facilities and technologies on ground and in space as testbeds in preparation for human exploratory missions and to develop a test plan for ground and space campaigns; (v) to develop a roadmap for a future European strategy towards human exploratory missions, including preparatory activities and terrestrial applications and benefits. This paper covers the part of the HUMEX study dealing with lunar missions. A lunar base at the south pole where long-time sunlight and potential water ice deposits could be assumed was selected as the Moon reference scenario. The impact on human health, performance and well being has been investigated from the view point of the effects of microgravity (during space travel), reduced gravity (on the Moon) and abrupt gravity changes (during launch and landing), of the effects of cosmic radiation including solar particle events, of psychological issues as well as general health care. Countermeasures as well as necessary research using ground-based test beds and/or the International Space Station have been defined. Likewise advanced life support systems with a high degree of autonomy and regenerative capacity and synergy effects were considered where bioregenerative life support systems and biodiagnostic systems become essential. Finally, a European strategy leading to a potential European participation in future human exploratory missions has been recommended.

The unprecedented challenges of creating Biosphere 2, the world's first laboratory for biospherics, the study of global ecology and long-term closed ecological system dynamics, led to breakthrough developments in many fields, and a deeper understanding of the opportunities and difficulties of material closure. This paper will review accomplishments and challenges, citing some of the key research findings and publications that have resulted from the experiments in Biosphere 2. Engineering accomplishments included development of a technique for variable volume to deal with pressure differences between the facility and outside environment, developing methods of atmospheric leak detection and sealing, while achieving new standards of closure, with an annual atmospheric leakrate of less than 10%, or less than 300 ppm per day. This degree of closure permitted detailed tracking of carbon dioxide, oxygen, and trice gases such as nitrous oxide and ethylene over the seasonal variability of two years. Full closure also necessitated developing new approaches and technologies for complete air, water, and wastewater recycle and reuse within the facility. The development of a soil-based highly productive agricultural system was a first in closed ecological systems, and much was learned about managing a wide variety of crops using non-chemical means of pest and disease control. Closed ecological systems have different temporal biogeochemical cycling and ranges of atmospheric components because of their smaller reservoirs of air, water and soil, and higher concentration of biomass, and Biosphere 2 provided detailed examination and modeling of these accelerated cycles over a period of closure which measured in years. Medical research inside Biosphere 2 included the effects on humans of lowered oxygen: the discovery that human productivity can be maintained with good health with lowered atmospheric oxygen levels could lead to major economies on the design of space stations and planetary/lunar settlements. The improved health resulting from the calorie-restricted but nutrient dense Biosphere 2 diet was the first such scientifically controlled experiment with humans. The success of Biosphere 2 in creating a diversity of terrestrial and marine environments, from rainforest to coral reef, allowed detailed studies with comprehensive measurements such that the dynamics of these complex biomic systems are now better understood. The coral reef ecosystem, the largest artificial reef ever built, catalyzed methods of study now being applied to planetary coral reef systems. Restoration ecology advanced through the creation and study of the dynamics of adaptation and self-organization of the biomes in Biosphere 2. The international interest that Biosphere 2 generated has given new impetus to the public recognition of the sciences of biospheres (biospherics), biomes and closed ecological life systems. The facility, although no longer a materially-closed ecological system, is being used as an educational facility by Columbia University as an introduction to the study of the biosphere and complex system ecology and for carbon dioxide impacts utilizing the complex ecosystems created in Biosphere '.The many lessons learned from Biosphere 2 are being used by its key team of creators in their design and operation of a laboratory-sized closed ecological system, the Laboratory Biosphere, in operation as of March 2002, and for the design of a Mars on Earth ™ prototype life support system for manned missions to Mars and Mars surface habitats. Biosphere 2 is an important foundation for future advances in biospherics and closed ecological system research.

This paper is devoted to the consideration of possible viewpoint on CELSS development and design. If the aim to create practically applicable CELSS is accepted then the task to optimize the process of CELSS research and development in terms of minimum cost, hours, maximum applicability, scientific contribution, etc. becomes actual. Requirements of applicability and scientific significance are synergetic since understanding of general properties of CELSS gives an ability to create CELSS for different applications. To accomplish the task three main groups of parameters have to be optimized: i) configuration and operating parameters of developing CELSS itself; ii) organizational management of research and development of CELSS; iii) features of an area where CELSS is planned to be used (space missions, terrestrial applications, or biosphere investigation) and where requirements to CELSS characteristic come from. Given paper is a brief review presented some attempts to arrange mentioned above into some set of formalized and interacting criteria, and some progression of research stages derived from these criteria.

The C.E.B.A.S.-Minimodule (Closed Equilibrated Biological Aquatic System) is a space qualified aquatic microcosm of 8.6 liters volume of water. Several aquatic species can be reared in parallel. Based on its characteristics (closed system, highly standardized, testruns longer than 4 weeks are possible, organisms of different trophic levels can be investigated) an improved module (C.B.R.U. = Closed Biological Research Unit) is under development for scientific and commercial use in ecotoxicology. In a two year project named AToxMss (Aquatic EcoToxicology in a Multispecies Stem) this goal can be reached. AToxMss is a R&D project of an industry team and two teams of the University of Bremen, funded by industry (OHB-System AG) and the state of Bremen, Germany. Three project phases are already completed: The parameter determination to indicate potential impact of chemicals on biological systems, the selection of test substances, as well as the manufacturing of 3 functional modules, each verified for use in ecotoxicological research. The next phase starts with a series of test runs calibrating the system by using well known toxic substances and chemicals.

Regenerative life support systems potentially offer a level of self-sufficiency and a decrease in logistics and associated costs in support of space exploration and habitation missions. Current state-of-the-art in plant-based, regenerative life support requires resources in excess of allocation proposed for candidate mission scenarios. Feasibility thresholds have been identified for candidate exploration missions. The goal of this paper is to review recent advances in performance achieved in the CELSS Antarctic Analog Project (CAAP) in light of the likely resource constraints. A prototype CAAP crop production chamber has been constructed and operated at the Ames Research Center. The chamber includes a number of unique hardware and software components focused on attempts to increase production efficiency, increase energy efficiency, and control the flow of energy and mass through the system. Both single crop, batch production and continuous cultivation of mixed crops production studies have been completed. The crop productivity as well as engineering performance of the chamber are described. For each scenario, energy required and partitioned for lighting, cooling, pumping, fans, etc. is quantified. Crop production and the resulting lighting efficiency and energy conversion efficiencies are presented. In the mixed-crop scenario, with 27 different crops under cultivation, 17 m2 of crop area provided a mean of 515 g edible biomass per day (85% of the approximate 620 g required for one person). Enhanced engineering and crop production performance achieved with the CAAP chamber, compared with current state-of-the-art, places plant-based life support systems at the threshold of feasibility. c2002 Published by Elsevier Science Ltd on behalf of COSPAR.

The C.E.B.A.S. MINI-MODULE is the miniaturized space flight version of the Closed Equilibrated Biological Aquatic System (C.E.B.A.S.). It fits into a large middeck locker tray and is scheduled to be flown in the STS 85 and in the NEUROLAB missions. Its volume is about 9 liters and it consists of two animal tanks, a plant cultivator, and a bacteria filter in a monolithic design. An external sensor unit is connected to a data acquisition/control unit. The system integrates its own biological life support. The CO 2 exhaled by the consumers (fishes, snails, microorganisms) is assimilated by water plants ( Ceratophyllum demersum ) which provide them with oxygen. The products of biomass degradation and excretion (mainly ammonia ions) are converted by bacteria into nitrite and nitrate. The latter is taken up by the plants as a nitrogen source together with other ions like phosphate. The plants convert light energy into chemical energy and their illumination is regulated via the oxygen concentration in the water by the control unit. In ground laboratory tests the system exhibited biological stability up to three month. The buffer capacity of the biological filter system is high enough to eliminate the degradation products of about one half of the dead animal biomass as shown in a “crash test”. A test series using the laboratory model of the flight hardware demonstrated the biological stability and technical reliability with mission-identical loading and test duration. A comprehensive biological research program is established for the C.E.B.A.S. MINI-MODULE in which five German and three U.S.-American universities as well as the Russian Academy of Sciences are involved.

The colonization of space will depend on our ability to routinely provide for the metabolic needs (oxygen, water, and food) of a crew with minimal re-supply from Earth. On Earth, these functions are facilitated by the cultivation of plant crops, thus it is important to develop plant-based food production systems to sustain the presence of mankind in space. Farming practices on earth have evolved for thousands of years to meet both the demands of an ever-increasing population and the availability of scarce resources, and now these practices must adapt to accommodate the effects of global warming. Similar challenges are expected when earth-based agricultural practices are adapted for space-based agriculture. A key variable in space is gravity; planets (e.g. Mars, 1/3 g) and moons (e.g. Earth's moon, 1/6 g) differ from spacecraft orbiting the Earth (e.g. Space stations) or orbital transfer vehicles that are subject to microgravity. The movement of heat, water vapor, CO2 and O2 between plant surfaces and their environment is also affected by gravity. In microgravity, these processes may also be affected by reduced mass transport and thicker boundary layers around plant organs caused by the absence of buoyancy dependent convective transport. Future space farmers will have to adapt their practices to accommodate microgravity, high and low extremes in ambient temperatures, reduced atmospheric pressures, atmospheres containing high volatile organic carbon contents, and elevated to super-elevated CO2 concentrations. Farming in space must also be carried out within power-, volume-, and mass-limited life support systems and must share resources with manned crews. Improved lighting and sensor technologies will have to be developed and tested for use in space. These developments should also help make crop production in terrestrial controlled environments (plant growth chambers and greenhouses) more efficient and, therefore, make these alternative agricultural systems more economically feasible food production systems.

Waste technologies for Mars missions have been analyzed, considering equivalent system mass and interface loads. Storage or dumping seems most appropriate for early missions with low food closure. Composting or other treatment of inedible biomass in a bioreactor seems most attractive for moderate food closure (50 – 75%). Some form of physicochemical oxidation of the composted residue might be needed for increased food closure, but oxidation of all waste does not seem appropriate due to excess of production of carbon dioxide over demand. More comprehensive analysis considering interfaces with other mission systems is needed. In particular, in-situ resource utilization is not considered, and might provide resources more cheaply than waste processing.

Research and design of subsurface flow wetland wastewater treatment systems for a ground-based experimental prototype Mars Base facility has been carried out, using a subsurface flow approach. These systems have distinct advantages in planetary exploration scenarios: they are odorless, relatively low-labor and low-energy, assist in purification of water and recycling of atmospheric CO2, and will support some food crops. An area of 6-8 m2 may be sufficient for integration of wetland wastewater treatment with a prototype Mars Base supporting 4-5 people. Discharge water from the wetland system will be used as irrigation water for the agricultural crop area, thus ensuring complete recycling and utilization of nutrients. Since the primary requirements for wetland treatment systems are warm temperatures and lighting, such bioregenerative systems may be integrated into early Mars base habitats, since waste heat from the lights may be used for temperature maintenance in the human living environment. "Wastewater gardens (TM)" can be modified for space habitats to lower space and mass requirements. Many of its construction requirements can eventually be met with use of in-situ materials, such as gravel from the Mars surface. Because the technology requires little machinery and no chemicals, and relies more on natural ecological mechanisms (microbial and plant metabolism), maintenance requirements are minimized, and systems can be expected to have long operating lifetimes. Research needs include suitability of Martian soil and gravel for wetland systems, system sealing and liner options in a Mars Base, and wetland water quality efficiency under varying temperature and light regimes.

Life support approaches for Mars missions are evaluated using an equivalent system mass (ESM) approach, in which all significant costs are converted into mass units. The best approach, as defined by the lowest mission ESM, depends on several mission parameters, notably duration, environment and consequent infrastructure costs, and crew size, as well as the characteristics of the technologies which are available. Generally, for the missions under consideration, physicochemical regeneration is most cost effective. However, bioregeneration is likely to be of use for producing salad crops for any mission, for producing staple crops for medium duration missions, and for most food, air and water regeneration for long missions (durations of a decade). Potential applications of in situ resource utilization need to be considered further.

For systematic human Mars exploration, meeting crew safety requirements, it seems perspective to assemble into a spacecraft: an electrical rocket, a well-shielded long-term life support system, and a manipulator-robots operating in combined “presence effect” and “master-slave” mode. The electrical spacecraft would carry humans to the orbit of Mars, providing short distance (and low signal time delay) between operator and robot-manipulators, which are landed on the surface of the planet. Long-term hybrid biological and physical/chemical LSS could provide environment supporting human health and well being. Robot-manipulators operating in “presence effect” and “master-slave” mode exclude necessity of human landing on Martian surface decreasing the level of risk for crew. Since crewmen would not have direct contact with the Martian environment then the problem of mutual biological protection is essentially reduced. Lightweight robot-manipulators, without heavy life support systems and without the necessity of returning to the mother vessel, could be sent as scouts to different places on the planet surface, scanning the most interesting for exobiological research site. Some approximate estimations of electric spacecraft, long-term hybrid LSS, radiation protection and mission parameters are conducted and discussed.

The degradation of an anionic surfactant (Igepon TC-42) was investigated as part of an integrated study of direct recycling of human hygiene water through hydroponic plant growth systems. Several chemical approaches were developed to characterize the degradation of Igepon and to measure the accumulation of intermediates such as fatty acids and methyl taurine. Igepon was rapidly degraded as indicated by the reduction of methylene blue active substances (MBAS) and component fatty acids. The Igepon degradation rate continued to increase over a period of several weeks following repeated daily exposure to 18 micrograms/l Igepon. The accumulation of free fatty acids and methyl taurine was also observed during decomposition of Igepon. The concentration of methyl taurine was below detection limit (0.2 nmol/ml) during the slow phase of Igepon degradation, and increased to 1-2 nmol/ml during the phase of rapid degradation. These findings support a degradation pathway involving initial hydrolysis of amide to release fatty acids and methyl taurine, and subsequent degradation of these intermediates. Ground-based experiments at RF SSC-IBMP RAS (State Science Center of Russian Federation--Institute of Biomedical Problems of Russian Academia of Science) were aimed at overall studies of a human-unicellular algae-mineralization LSS (life support system) model. The system was 15 m3 in volume. It contained 45 L of algal suspension with a dry substance density of 10-12 g per liter; water volume, including the algal suspension, was 59 L. More sophisticated model systems with partial substitution of unicellular algae with higher plates (crop area of 15 m2) were tested in three experiments from 1.5 to 2 months in duration. The experiments demonstrated that LSS employing the unicellular algae play not only a macrofunction (regeneration of atmosphere and water) but also carry some other functions (purification of atmosphere, formation of microbial cenosis etc.) providing an adequate human environment. It is also important that functional reliability of the algal regenerative subsystem is secured by a huge number of cells able, in the event of death of a part of population, to recover in the shortest possible time the size of population and, hence, functionality of the LSS autotrophic component. For a long period of time a Martian crew will be detached from Earth's biosphere and for this reason LSS of their vehicle must be highly reliable, robust and redundant. One of the approaches to LSS redundancy is installation of two systems with different but equally efficient regeneration technologies, i.e. physical-chemical and biological. At best, these two systems should operate in parallel sharing the function of regeneration of the human environment. In case of failure or a sharp deterioration in performance of one system the other will, by way of redundancy, increase its throughput to make up for the loss. This LSS design will enable simultaneous handling of a number of critical problems including adequate satisfaction of human environmental needs.

Sustainability is one of the most important criteria in the creation and evaluation of human life support systems intended for use during long space flights. The common feature of biological and physicochemical life support systems is that basically they are both catalytic. But there are two fundamental properties distinguishing biological systems: 1) they are auto-catalytic: their catalysts--enzymes of protein nature--are continuously reproduced when the system functions; 2) the program of every process performed by enzymes and the program of their reproduction are inherent in the biological system itself--in the totality of genomes of the species involved in the functioning of the ecosystem. Actually, one cell with the genome capable of the phenotypic realization is enough for the self-restoration of the function performed by the cells of this species in the ecosystem. The continuous microalgal culture of Chlorella vulgaris was taken to investigate quantitatively the process of self-restoration in unicellular algae population. Based on the data obtained, we proposed a mathematical model of the restoration process in a cell population that has suffered an acute radiation damage.

The effect of elevated temperatures of 35 and 45 degrees C (at the intensities of photosynthetically active radiation 322, 690 and 1104 micromoles m-2 s-1) on the photosynthesis, respiration, and qualitative and quantitative composition of the volatiles emitted by wheat (Triticum aestuvi L., cultivar 232) crops was investigated in growth chambers. Identification and quantification of more than 20 volatile compounds (terpenoids--alpha-pinene, delta 3 carene, limonene, benzene, alpha- and trans-caryophyllene, alpha- and gamma-terpinene, their derivatives, aromatic hydrocarbons, etc.) were conducted by gas chromatograph/mass spectrometry. Under light intensity of 1104 micromoles m-2 s-1 heat resistance of photosynthesis and respiration increased at 35 degrees C and decreased at 45 degrees C. The action of elevated temperatures brought about variations in the rate and direction of the synthesis of volatile metabolites. The emission of volatile compounds was the greatest under a reduced irradiation of 322 micromoles m-2 s-1 and the smallest under 1104 micromoles m-2 s-1 at 35 degrees C. During the repair period, the contents and proportions of volatile compounds were different from their initial values, too. The degree of disruption and the following recovery of the functional state depended on the light intensity during the exposure to elevated temperatures. The investigation of the atmosphere of the growth chamber without plants has revealed the substances that were definitely technogenic in origin: tetramethylurea, dimethylsulfide, dibutylsulfide, dibutylphthalate, and a number of components of furan and silane nature.

In addition to green microalgae, aquatic higher plants are likely to play an important role in aquatic food production modules in bioregenerative systems for producing feed for fish, converting CO2 to O2 and remedying water quality. In the present study, the effects of culture conditions on the net photosynthetic rate of a rootless submerged plant, Ceratophyllum demersum L., was investigated to determine the optimum culture conditions for maximal function of plants in food production modules including both aquatic plant culture and fish culture systems. The net photosynthetic rate in plants was determined by the increase in dissolved O2 concentrations in a closed vessel containing a plantlet and water. The water in the vessel was aerated sufficiently with a gas containing a known concentration of CO2 gas mixed with N2 gas before closing the vessel. The CO2 concentrations in the aerating gas ranged from 0.3 to 10 mmol mol-1. Photosynthetic photon flux density (PPFD) in the vessel ranged from 0 (dark) to 1.0 mmol m-2 s-1, which was controlled with a metal halide lamp. Temperature was kept at 28 degrees C. The net photosynthetic rate increased with increasing PPFD levels and was saturated at 0.2 and 0.5 mmol m-2 s-1 PPFD under CO2 levels of 1.0 and 3.0 mmol mol-1, respectively. The net photosynthetic rate increased with increasing CO2 levels from 0.3 to 3.0 mmol mol-1 showing the maximum value, 75 nmol O2 gDW-1 s-1, at 2-3 mmol mol-1 CO2 and gradually decreased with increasing CO2 levels from 3.0 to 10 mmol mol-1. The results demonstrate that C. demersum could be an efficient CO2 to O2 converter under a 2.0 mmol mol-1 CO2 level and relatively low PPFD levels in aquatic food production modules. c2003 COSPAR. The parallels between the challenges facing bioregenerative life support in artificial closed ecological systems and those in our global biosphere are striking. At the scale of the current global technosphere and expanding human population, it is increasingly obvious that the biosphere can no longer safely buffer and absorb technogenic and anthropogenic pollutants. The loss of biodiversity, reliance on non-renewable natural resources, and conversion of once wild ecosystems for human use with attendant desertification/soil erosion, has led to a shift of consciousness and the widespread call for sustainability of human activities. For researchers working on bioregenerative life support in closed systems, the small volumes and faster cycling times than in the Earth's biosphere make it starkly clear that systems must be designed to ensure renewal of water and atmosphere, nutrient recycling, production of healthy food, and safe environmental methods of maintaining technical systems. The development of technical systems that can be fully integrated and supportive of living systems is a harbinger of new perspectives as well as technologies in the global environment. In addition, closed system bioregenerative life support offers opportunities for public education and consciousness changing of how to live with our global biosphere.

The working efficiency and state-of-mind of a Space vehicle crew on long-term missions is dependent on the suitability of living conditions including food. Our purpose was to establish the feasibility of an in vitro muscle protein production system (MPPS) for the fabrication of surrogate muscle protein constructs as food products for Space travelers. In the experimental treatments, we cultivated the adult dorsal abdominal skeletal muscle mass of Carassius (Gold fish). An ATCC fish fibroblast cell line was used for tissue engineering investigations. No antibiotics were used during any phase of the research. Our four treatments produced these results: a low contamination rate, self-healing, cell proliferation, a tissue engineered construct of non-homologous co-cultured cells with explants, an increase in tissue size in homologous co-cultures of explants with crude cell mixtures, maintenance of explants in media containing fetal bovine serum substitutes, and harvested explants which resembled fresh fish filets. We feel that not only have we pointed the way to an innovative, viable means of supplying safe, healthy, nutritious food to Space voyagers on long journeys, but our research also points the way to means of alleviating food supply and safety problems in both the public and private sectors worldwide.

The started assembly of the International Space Station (ISS) and its further operation will call for a great number of extravehicular activity sorties (EVA) to be performed by ISS crews. Therefore, of great importance is to make use of the EVA experience gained by cosmonauts in the process of 15-year operation of the Mir orbiting station (OS). Over the 15-year period, Mir crewmembers wearing Orlan type semi-rigid space suits have accumulated 158 man/sorties from the orbiting station. Crewmembers used 15 suits in orbit and some of the suits were in operation for more than 3 years. The paper presents principal design features, which provide effective and safe operation of orbit-based suits, and briefly describes procedures for preparation and maintenance of suit systems, which ensure long-term operation of space suit in orbit. The paper gives results of the space suit modifications, presents suit performance characteristics and lists novel or upgraded components of the space suit and its systems. The paper also summarizes improvements in the Orlan type suits described in some earlier publications. They refer, in the first run, to the improvement of space suit operations characteristics and reliability, and the utilization of the Orlan type space suit in the ISS program. The paper analyses the experience gained and drawbacks detected and observations made, and gives statistical data on long-term space suit operations aboard the Mir station. The paper reviews certain problems in the process of EVAs performed from the station, and describes the ways of their solution as applied to the further utilization of the suit within the ISS program.

The closed equilibrated biological aquatic system (C.E.B.A.S) is a man-made aquatic ecosystem which consists of four subcomponents: an aquatic animal habitat, an aquatic plant bioreactor, an ammonia oxidizing bacteria filter and a data acquisition/control unit. It is a precursor for different types of fish and aquatic plant production sites which are disposed for the integration into bioregenerative life-support systems. The results of two successful spaceflights of a miniaturized C.E.B.A.S version (the C.E.B.A.S. MINI MODULE) allow the optimization of aquatic food production systems which are already developed in the ground laboratory and open new aspects for their utilization as aquatic modules in space bioregenerative life support systems. The total disposition offers different stages of complexity of such aquatic modules starting with simple but efficient aquatic plant cultivators which can be implemented into water recycling systems and ending up in combined plant/fish aquaculture in connection with reproduction modules and hydroponics applications for higher land plants. In principle, aquaculture of fishes and/or other aquatic animals edible for humans offers optimal animal protein production under lowered gravity conditions without the tremendous waste management problems connected with tetrapod breeding and maintenance. The paper presents details of conducted experimental work and of future dispositions which demonstrate clearly that aquaculture is an additional possibility to combine efficient and simple food production in space with water recycling utilizing safe and performable biotechnologies. Moreover, it explains how these systems may contribute to more variable diets to fulfill the needs of multicultural crews.

We studied the effects of accumulated nitrate in water on the spawning, hatching and development of medaka using a simple nitrifying filter and a combined filter having both nitrifying and denitrifying capabilities. A nitrate concentration of 100 mg NO3(-)-N/L was clearly of lethal toxicity to fish when they were exposed to nitrate in both adult and the growing phases. A nitrate concentration of 75 mg NO3(-)-N/L reduced the fertilization rate, delayed the hatching time and reduced the hatching rate of the eggs laid by adults and decreased the growth rate of juveniles. In addition, nitrate accumulations as low as 50 mg NO3(-)-N/L remarkably retarded spawning and lowered the number of eggs laid by fish exposed in the juvenile phase. The effects on the reproduction system may be initiated by a low concentration, approximately 30 mg NO3(-)-N/L.
