Conservation aquaculture has prevented extirpation of Upper Columbia River white sturgeon Acipenser transmontanus, but uncertainty remains on how hatchery-origin fish move between habitats located within Canada and the United States. We describe how core and home ranges of sturgeon varied by environmental (country, season) and biological (age, sex, size) factors and determined residence at important habitats. Core range represented areas where individuals spent the majority of their time, while home range represented most areas occupied. Fidelity to specific habitats was high, with home ranges averaging 10 km2 and 50% of individuals occupied the same core range across their entire detection period. Only 12% of individuals had home ranges encompassing both countries. Core and home ranges were larger during the summer compared to winter and larger within the United States compared to Canada. Larger (>120 cm) sturgeon had increased core and home ranges. We documented spawning capable hatchery-origin males and confirmed their residence at spawning sites when spawning was detected. Green sturgeon (Acipenser medirostris) and white sturgeon (A. transmontanus) are closely related, sympatric species that inhabit the San Francisco estuary. Green sturgeon have a more marine life history but both species spawn in the Sacramento River and reside for some duration in San Francisco Bay. These sturgeons are of conservation concern, yet little is known about their dietary competition when they overlap in space and time. To examine evidence of dietary differentiation, we collected whole blood and blood plasma from 26 green sturgeon and 35 white sturgeon in San Francisco Bay. Using carbon and nitrogen stable isotope analyses, we compared their relative trophic levels and foraging locations along the freshwater to marine gradient. Sampling blood plasma and whole blood allowed comparison of dietary integration over shorter and longer time scales, respectively. Plasma and whole blood δ13C values confirmed green sturgeon had more marine dietary sources than white sturgeon. Plasma δ15N values revealed white sturgeon fed at lower trophic levels than green sturgeon recently, however, whole blood δ15N values demonstrated the two species fed at the same trophic level over longer time scales. Larger individuals of both species had higher δ13C values than smaller individuals, reflecting more marine food sources in adulthood. Length did not affect δ15N values of either species. Isotope analyses supported the more marine life history of green than white sturgeon and potentially highlight a temporary trophic differentiation of diet between species during and preceding the overlapping life stage in San Francisco Bay. Five metal mixture dose–response models were used to predict the toxicity of porewater to young sturgeon at areas of interest in the Upper Columbia River (WA, USA/BC, Canada) and to evaluate these models as tools for risk assessments. Dose components of metal mixture models included exposure to free metal ion activities or metal accumulation by biotic ligands or humic acid, and links of dose to response used logistic equations, independent joint action equations, or additive toxicity functions. Laboratory bioassay studies of single metal exposures to juvenile sturgeon, porewater collected in situ in the fast‐flowing Upper Columbia River, and metal mixture models were used to evaluate toxicity. The five metal mixture models were very similar in their predictions of adverse response of juvenile sturgeon and in identifying copper (Cu) as the metal responsible for the most toxic conditions. Although the modes of toxic action and the 20% effective concentration values were different among the dose models, predictions of adverse response were consistent among models because all doses were tied to the same biological responses. All models indicated that 56% ± 5% of 122 porewater samples were predicted to have <20% adverse response, 25% ± 5% of samples were predicted to have 20% to 80% adverse response, and 20% ± 4% were predicted to have >80% adverse response in juvenile sturgeon. The approach of combining bioassay toxicity data, compositions of field porewater, and metal mixture models to predict lack of growth and survival of aquatic organisms due to metal toxicity is an important tool that can be integrated with other information (e.g., survey studies of organism populations, life cycle and behavior characteristics, sediment geochemistry, and food sources) to assess risks to aquatic organisms in metal‐enriched ecosystems. Traditional methods for estimating abundance of fish populations are not feasible in some systems due to complex population structure and constraints on sampling effort. Lincoln's estimator provides a technique that uses harvest and harvest rate to estimate abundance. Using angler catch data allows assumptions of the estimator to be addressed without relying on methods that could be prohibitively field-intensive or costly. Historic estimates of White Sturgeon Acipenser transmontanus abundance in the Sacramento–San Joaquin River basin have been obtained using mark–recapture methods; however, White Sturgeon population characteristics often cause violations of model assumptions, such as population closure and independent capture probabilities. We developed a version of Lincoln's estimator using a joint likelihood, estimated abundance of White Sturgeon in the Sacramento–San Joaquin River basin in 2015 using this method and empirical data and assessed accuracy and precision of estimates in a simulation study. Estimating abundance using harvest and harvest rate, as represented by our model framework, has the potential to be precise and accurate. The joint likelihood–based approach fitted using Bayesian methods is advantageous because it includes all sources of variation in a single model. Precision of abundance estimates was low with application of the model to White Sturgeon in the Sacramento–San Joaquin River basin and to similar conditions in a simulated dataset. Using simulation, precision and accuracy increased with increases in the number of high-reward and standard tags released, tag reporting rate, tag retention rate, and harvest rate. Results demonstrate potential sources of error when using this approach and suggest that increasing the number of tagged fish and tag reporting rate are potential actions to improve precision and accuracy of abundance estimates of the model. Predation of fish released as part of conservation aquaculture programs may undermine conservation goals. Identification of factors influencing predation, therefore, can inform management decisions that minimize predation losses. For the endangered Nechako white sturgeon (Acipenser transmontanus Richardson, 1836), the survival of individuals released as part of a conservation aquaculture program is threatened by river otter (Lontra canadensis (Schreber, 1777)) predation. Through the recovery of sturgeon passive integrated transponder (PIT) tags from otter latrines, we set out to quantify the extent of predation, identify spatial patterns in predation including identifying predation hotspots, and understand the timing of predation. The recovery of 3.9% of PIT tags from the hatchery-released sturgeon represents a minimum predation estimate based on several identified factors. The distribution of predation hotspots varied by release cohort, with some consistent hotspots among cohorts. An apparent decrease in predation in recent years was seen in conjunction with fewer live captures and lower release numbers, suggesting some level of density-dependent predation. Results from this study can be used to contribute to management decisions related to Nechako white sturgeon recovery efforts, inform further research needs, and contribute to a growing knowledge of sturgeon predation. Improving the status of endangered species can be challenging because the efficacy of conservation actions is often uncertain. Conservation aquaculture has been the main recovery action for endangered white sturgeon (Acipenser transmontanus) in the Transboundary Reach of the upper Columbia River. Using long-term mark-recapture data (2002–2018), we predicted variation in growth rates due to genetic, environmental, and competition effects to evaluate the efficacy of the aquaculture program. Environmental conditions (by season and country) and competition had the greatest effects on growth. Growth, length-at-age, weight-at-age, and condition factor were higher for fish residing in reservoir habitats (US) compared to those in riverine habitat (Canada). Growth declined over the study period but growth in length for larger fish remained higher in the US as fish > 100 cm fork length in Canada were not growing. Small differences in growth among families indicate that differences in genetics among parents spawned in the hatchery had negligible effects on growth in the wild. White sturgeon inhabit large rivers and estuaries along the Pacific coast of North America and play important ecological and cultural roles. As with other poikilotherms, the expected rise in global temperatures will create physiological challenges for sturgeon, including direct effects on physiological rates, and may also cause changes in behaviour that impact survival. In this study, we investigated 'carryover effects' in white sturgeon (Acipenser transmontanus) by incubating fish at one of three embryological temperatures (12 °C, 15 °C, or 18 °C) from fertilization to hatch, and then holding them at 15 °C for the following 30 days post-hatch (dph). We investigated the effect of these incubation temperatures on subsequent behaviours, including locomotion and anxiety-like behaviour, at 18, 24 and 30 dph. The open field test was used to quantify distance moved by the fish and time spent near the outer wall of the arena (to quantify thigmotaxis, an indicator of anxiety-like behaviour) and recorded with motion-tracking software. This test was also used to compare behaviour during the day versus night (at 21 dph) and hourly at night (at 28 dph). Additionally, a light/dark test commonly used in rodents and other fish species was performed for the first time on larval sturgeon at 24 dph. We found a significant family effect; the impact of embryo rearing temperature on locomotion and thigmotaxis varied depending on the family lineage of the fish. Furthermore, we provide evidence that larval sturgeon exhibit greater locomotion at 24 and 30 dph compared to 18 dph, and have a preference for the light zone of the light/dark test. Management objectives for long-lived species are difficult to define because many taxa have delayed maturity and variable recruitment. White Sturgeon Acipenser transmontanus is an example of a species with a complex life history that complicates long-term status monitoring and establishment of management objectives. Historically, White Sturgeon in the Sacramento–San Joaquin River basin have been monitored by tracking the abundance of age-15 individuals as outlined by the Central Valley Project Improvement Act. However, infrequent recruitment complicates progress toward Central Valley Project Improvement Act management objectives because abundance of a single cohort fails to represent overall population trends. By using a Leslie population matrix, we demonstrate that the probability of reaching the Central Valley Project Improvement Act objective of 11,000 age-15 White Sturgeon is highly unlikely. We propose an alternative metric of 155,000 adults, which better represents overall population trends of White Sturgeon in the Sacramento–San Joaquin River basin, can be efficiently monitored, and can support both the goal of the Central Valley Project Improvement Act and management objectives. Background: A new technology for a self-powered acoustic tag (SPT) was developed for active tracking of juvenile fish, intended to avoid the typical battery life constraints associated with active telemetry technology. We performed a laboratory study to evaluate a subdermal tagging technique for the SPT and effects of the tag on survival, tag retention, and growth in juvenile white sturgeon (Acipenser transmontanus). Results: Survival was associated with tag retention. White sturgeon implanted with the SPT (n = 30) had 93% survival and tag retention by day 28, 67% by day 101, and 38% by day 595 post-tagging. Sturgeon implanted with a passive integrated transponder (PIT) tag only (control group) had 96% survival and tag retention by day 28, and through day 101 post-tagging. Fish in the PIT group were repurposed after day 101, so no comparisons with this group were made at day 595 post-tagging. Specific growth rate (SGR) for fork length was a median of 0.25% day−1 by day 28 for the SPT group, which was significantly lower than the PIT group (median: 0.42% day−1; n = 27). The SPT and PIT groups had similar SGR fork length by day 101 post-tagging (0.22 and 0.25% day−1, respectively). SGR weight was also lower for the SPT group compared to the PIT group on day 28 (1.39 and 2.11% day−1, respectively), but the difference again dissipated by day 101 (0.79 and 0.88% day−1, respectively). Conclusion: The tagging technique and placement of the SPT allowed the tag to remain upright along the flank of the sturgeon to ensure maximum battery output of the SPT; however, retention rates of the SPT were not ideal. We provided suggestions to improve the tagging technique. Suggestions included tagging fish that are > 400 mm FL, moving the incision location to extend the cavity and create a pocket for the placement of the SPT, and performing a quantitative wound-healing evaluation. The migration of larval fish from spawning to rearing habitat in rivers is not well understood. This paper describes a methodology to predict larval drift using a Lagrangian particle-tracking (LPT) model with passive and active behavioural components loosely coupled to a quasi-three-dimensional hydraulic model. In the absence of measured larval drift, a heuristic approach is presented for the larval drift of two species of interest, white sturgeon (Acipenser transmontanus) and burbot (Lota lota), in the Kootenai River, Idaho. Previous studies found that many fish species prefer certain vertical zones within the water column; sturgeon tend to be found near the bottom and burbot close to the water surface. Limiting the vertical movement of larvae is incorporated into the active component of the LPT model. The results illustrate a pattern of drift where secondary flow in meander bends and other zones of flow curvature redistributes particles toward the outside of the bend for surface drifters and toward the inside of the bend for bottom drifters. This pattern periodically reinforces the intersection of drifting larvae with channel margins in meander bends. In the absence of measured larval drift data, the model provides a tool for hypothesis testing and a guide to both field and laboratory experiments to further define the role of active behaviour in drifting larvae. White Sturgeon (Acipenser transmontanus), a species of concern in the San Francisco Estuary, is in relatively low abundance because of a variety of factors. The purpose of our study was to identify the estuarine habitat used by White Sturgeon to aid in the conservation and management of the species locally and across its range. We seasonally sampled subadult and adult White Sturgeon in the central estuary using setlines across a habitat gradient representative of three primary structural elements: shallow wetland channel (mean sample depth = 2 m), shallow open-water shoal (mean sample depth = 2 m), and deep open-water channel (mean sample depth = 7 m). We found that the shallow open-water shoal and deep open-water channel habitats were consistently occupied by White Sturgeon in spring, summer, and fall across highly variable water quality conditions, whereas the shallow wetland channel habitat was essentially unoccupied. We conclude that subadult and adult White Sturgeon inhabit estuaries in at least spring, summer, and fall and that small, shallow wetland channels are relatively unoccupied. [ABSTRACT FROM AUTHOR]

Temperature differentially influences developmental trajectories of traits during early life stages that can affect survival and recruitment. Experiments were conducted to quantify temperature-induced developmental responses of White Sturgeon (Acipenser transmontanus) yolk-sac larvae (YSL) reared at temperatures encountered across the species' range (12.5, 14.0, 15.5, and 17.0 °C). We quantified effects of temperature on timing of transitions between sequential developmental stages from hatch to initiation of exogenous feeding. Rate of development significantly increased at warmer compared to cooler temperatures; no significant difference was observed between 15.5 and 17.0 °C or 12.5 and 14.0 °C. When standardized by relative timing of development (RTi), developmental rate was not significantly different among treatments. Morphological traits (total length; body area; yolk-sac area; head area; gill filament area; mouth area; pectoral fin area) were measured daily, though only data for YSL reared at 12.5 and 17.0 °C was used to quantify phenotypic variation. Morphological traits (excluding yolk-sac area) were generally larger 48+ hours post hatch for YSL reared at 17. 0 °C compared to 12.5 °C. In contrast, these same traits, with the exception of gill filament area, were larger in 12.5 °C reared YSL when considered as a function of developmental stage. These opposing results suggest trade-offs associated with allocating resources to a particular trait depended on rearing temperature. Our results provide the ability to estimate timing of critical early life stages (i.e., hatch, emergence) as a function of temperature which is an important management tool to understand how early life development contributes to recruitment processes and adaptability in thermally altered systems. Outbreaks of an infectious disease affecting cultured white sturgeon (Acipenser transmontanus) were investigated. Clinical signs included erratic swimming, arching of the back and mortality. Necropsy findings included poorly demarcated yellow to dark‐red and friable lesions in the epaxial muscle, ulcerative skin lesions and haemorrhages in the swim bladder and coelomic wall. Histological evaluation revealed areas of necrotizing and heterophilic myositis with aggregates of bacterial cocci. The lumen of blood vessels in the dermis, under ulcerated areas, and in the posterior kidney, was occluded by fibrin thrombi. Aggregates of Gram‐positive cocci were observed in the muscle lesions and within the fibrin thrombi in the dermis and kidney. Genetically homogeneous Streptococcus iniae strains were recovered from affected fish from different outbreaks. The isolates shared high degree of similarity at gene locus (gyrB) with previously characterized S. iniae from cultured fish in California, confirming the emergence of this particular strain of S. iniae in US aquaculture. Veronaea botryosa is the etiological agent of a systemic phaeohyphomycosis known as "fluid belly" in white sturgeon (Acipenser transmontanus). Fluid belly is a critical disease affecting sturgeon aquaculture and the caviar industry for which there are no commercially available vaccines or approved antifungal treatments to manage outbreaks. The primary aim of this study was to investigate the effect of a V. botryosa [conidia], a V. botryosa [mold], and a Saccharomyces cerevisiae [yeast] formalin-killed vaccine on sturgeon immune responses to fungal challenge. Immunization consisted of an initial intracoelomic injection with one of the three treatment preparations, followed by a vaccine booster four weeks later by the same route and dose. Experimental challenge by intramuscular injection with a virulent V. botryosa conidia suspension followed after another four weeks. Non-challenged control fish received injections of PBS. The inactivated vaccines proved safe for white sturgeon fingerlings. Sturgeon immunized with either V. botryosa [mold] or S. cerevisiae [yeast] exhibited a significantly different pro-inflammatory response upon challenge with V. botryosa compared to non-immunized fish. Challenged fish developed clinical signs similar to those reported during natural outbreaks of fluid belly. Positive control treatments (those not immunized but challenged with V. botryosa) experienced the highest mortality; however, survival curves were similar amongst all treatments (p < 0.05). Furthermore, the S. cerevisiae [yeast] vaccine resulted in comparatively lower fungal persistence and fewer lesions following histological analysis. Abstract Previous studies have demonstrated reduced performance in triploid fish when reared under suboptimal conditions, which may be the result of a higher susceptibility to stressors when compared to diploids. The goal of this project was to investigate differences in the capacity of diploid (8 N) and triploid (12 N) white sturgeon, Acipenser transmontanus , to respond to both warm acclimation (6-weeks of acclimation to either 18 or 22 °C) and a subsequent acute stress (10-min low water stress). Following the 6-week acclimation, fish were sampled either before or following an acute low water stress. Bioindices of the primary and secondary stress response, hematology and cellular metabolic status were measured. We also sought to determine if time to peak cortisol levels were similar between diploid and triploid sturgeon after exposure to a severe acute stressor (netting stress). While both ploidies had similar primary and secondary responses to acute stress, both with and without warm acclimation, warm acclimation impacted the ability of diploid and triploid white sturgeon to mount a typical stress response to an acute stressor. In response to warm acclimation, triploids exhibited little change in branchial lactate dehydrogenase activity, while diploids increased activity. After exposure to an acute water reduction stress, diploids increased citrate synthase activity, yet triploids showed a decrease in activity. Differences in metabolic enzyme activity in response to warm acclimation and acute stress suggest triploid white sturgeon may have a reduced cellular metabolic capacity under chronic and acute stress, which may impact performance of triploid sturgeon in suboptimal conditions. Freshwater fish such as white sturgeon (Acipenser transmontanus) are particularly vulnerable to the effects of anthropogenically induced global warming. Critical thermal maximum tests (CT max) are often conducted to provide insight into the impacts of changing temperatures; however, little is known about how the rate of temperature increase in these assays affects thermal tolerance. To assess the effect of heating rate (0.3 °C/min, 0.03 °C/min, 0.003 °C/min) we measured thermal tolerance, somatic indices, and gill Hsp mRNA expression. Contrary to what has been observed in most other fish species, white sturgeon thermal tolerance was highest at the slowest heating rate of 0.003 °C/min (34.2 °C, and CTmax of 31.3 and 29.2 °C, for rates 0.03 and 0.3 °C/min, respectively) suggesting an ability to rapidly acclimate to slowly increasing temperatures. Hepatosomatic index decreased in all heating rates relative to control fish, indicative of the metabolic costs of thermal stress. At the transcriptional level, slower heating rates resulted in higher gill mRNA expression of Hsp90a, Hsp90b, and Hsp70. Hsp7 0 mRNA expression was increased in all heating rates relative to controls, whereas expression of Hsp90a and Hsp90b mRNA only increased in the two slower trials. Together these data indicate that white sturgeon have a very plastic thermal response, which is likely energetically costly to induce. Acute temperature changes may be more detrimental to sturgeon as they struggle to acclimate to rapid changes in their environment, however under slower warming rates they demonstrate strong thermal plasticity to warming. • White sturgeon increased thermal tolerance as CT max warming rate decreased. • Sturgeon were able to rapidly acclimate during the slowest CT max trial increasing their CT max. • HSP expression differed between trial length and fish's individual thermal tolerance. • Hepatosomatic indices were affected by CT max suggesting thermal stress has a metabolic cost. Image analysis can predict the fat content of sturgeon ovaries that had been categorized as having a low, medium, and high fat content based upon the caviar yield expressed as a percent of the total ovary weight, and were correlated with the chemical measurement of total fat (R2 = 0.83). The fatty acid composition of eggs was not influenced by ovary fat content. Palmitic acid (16:00) was the most abundant saturated fatty acid and oleic acid (18:1n-9) the most predominant monounsaturated fatty acids in sturgeon eggs regardless of the ovary fat content. No significant differences (P[0.05) were observed in docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) in eggs from fish with different fat ovaries. Fourier transform infrared spectroscopy (FT-IR) coupled with principal component analysis indicated no significant difference in chemical compositions in sturgeon eggs separated from ovaries of different fat contents confirming the fatty acid composition results. White sturgeon, Acipenser transmontanus (Richardson), are at risk of entrainment from dredging, with young-of-the-year fish at greatest risk. To evaluate this entrainment risk, swimming performance trials were conducted in a laboratory swim tunnel with hatchery-reared juvenile white sturgeon with varying experience levels including: naïve (only tested once), tested (re-tested after being kept in no flow) and trained (re-tested after kept in flow for nearly three weeks). Individuals of various sizes (80–100 mm TL) and all experience levels were strongly rheotactic (> 80%), but endurance was highly variable among fish. Small juveniles [< 82 mm total length (TL)] had lower escape speeds (< 40 cm s−1) than medium (82–92 mm TL) and large (> 93 mm TL) naïve fish (42–45 cm s−1), all of which had lower escape speeds than trained fish (72 cm s−1). Behavior was also highly variable among fish. Overall, benthic station-holding behaviors were least frequent in small fish, intermediate in medium and large fish, and most frequent in trained large fish. Probability of entrainment of juvenile white sturgeon can be reduced by maintaining dredge head flow fields at less than 45 cm s−1 for wild-spawned fish or by rearing hatchery fish to > 93 mm TL and exposing the fish to moderate flow velocities (10–12 cm s−1) prior to their release. A total of 35, age 1 juvenile Kootenay River white sturgeon ( Acipenser transmontanus), were fitted with sonic tags in 2005 and released as part of larger hatchery release groups at five sites to evaluate dispersal and subsequent movements (seven tags per site). Juvenile sturgeon released at three locations within the deep, low gradient reach (typical gradient of 0.02 m km−1 and velocities of <0.4 m s−1) of the Kootenay River below Bonners Ferry, ID showed substantial dispersal both up and downstream; however, downstream redistribution was more common. White sturgeon from all three release locations overlapped during dispersal, with 9% of tagged fish moving from river release sites into Kootenay Lake. The three hatchery release locations in this low gradient reach produced good dispersal of hatchery progeny into available habitats. Tagged fish released above Bonners Ferry in the shallow, higher gradient reach (typical gradient of 0.6 m km−1, and velocities >0.8 m s−1) at two additional sites all moved downstream of the gradient break at Bonners Ferry, ID into the lower gradient reach within 2 months of release. In total, 93% of these tagged fish relocated to the low gradient section within 25 days of release, with some fish undertaking this movement within 1 day. Abstract  In general little is known about hormones and the ontogeny of the stress response in the early developmental stages of chondrostean fishes and in particular of white sturgeon (Acipenser transmontanus, Richardson 1836). In this study, we measured for the first time cortisol and sex steroids (testosterone and estradiol) in eggs, larvae, post-larvae, and fry of white sturgeon by radioimmunoassay (RIA), to elucidate some endocrine aspects of its development. The cortisol, testosterone, and 17β-estradiol of maternal origin found in unfertilized eggs of white sturgeon probably regulate both growth and development of the embryo. Cortisol decreased after fertilization, whereas testosterone and 17β-estradiol did not significantly change. During the late stages of embryo development and immediately after hatching, endogenous production of cortisol and sexual steroids, respectively, occurred. Sex steroids may be physiological inducers of gonad sex differentiation in sturgeon. All steroids showed an increase 10 days post-hatch (dph), near the transition from an endogenous to an exogenous energy source. Cortisol maintained the same basal levels even after metamorphosis, whereas testosterone and 17β-estradiol declined significantly in post-larvae at 35 and 45 days post-hatch. In addition, to evaluate the ontogeny of a functional hypothalamic-pituitary-interrenal (HPI) axis, larvae and fry were submitted to acute stress. White sturgeon ( Acipenser transmontanus) ≥95 cm TL were monitored using acoustic and radio telemetry at a large hydroelectric dam (the Dalles Dam) on the Columbia River, during March 2004 through November 2005 to determine timing and routes of passage and to characterize general movements. Transmitters were surgically implanted into 148 fish during the study; 90 were released into the tailrace and 58 into the forebay. We documented 26 passage events by 19 tagged fish: eight upstream via fish ladders and 18 downstream, mostly through open spill gates. During the study 17 fish entered the two ladders one or more times; 11 entered only the east ladder, three entered only the north ladder, and three entered both ladders at sometime. Residence time within the ladders by individual fish was variable, ranging from about 1 min to nearly 6 months (median = 7.7 h). Only six fish successfully ascended the east ladder, one fish twice. We could not unequivocally determine which fish ladder one fish used to pass upstream. Differences in construction between the north and east fish ladders may account for the greater success of the east fish ladder in passing sturgeon upstream. Changes to operations at hydroelectric dams to benefit migrating anadromous salmonids may influence upstream or downstream passage by white sturgeon. Altering patterns and timing of spill discharge, altering fish ladder entrance attraction flows, and the use of lights, sound, and partial barriers to direct other species of fish to preferred passage routes have unknown effects on sturgeon passage. Fifty white sturgeon ( Acipenser transmontanus) eleutheroembryos (average size 0.17 g) were placed onto each of four quadrants (0.45 m2 quadrant−1; 200 fish tank−1) of different sized substrates in four circular tanks (approximately 562 L). Each of three quadrants had a different size substrate and the fourth quadrant was left bare. We used one replicate of smaller size substrates (0.5–11.9 mm) and one replicate of larger size substrates (21.7–88 mm). It was found that the white sturgeon eleutheroembryos preferred substrate with an average size (longest diameter) of 12 mm (11.9) in the smaller substrate range and 22 mm (21.7) in the larger substrate range. These data improve our knowledge of white sturgeon early life history, and if confirmed in the wild can be used to protect areas that are crucial for white sturgeon recruitment and survival. Pectoral fin tissue of white sturgeon was investigated as a potential non-lethal sample source for the detection of white sturgeon iridovirus (WSIV) infection. Histopathology and polymerase chain reaction (PCR) results using fin tissue were compared with the standard lethal histopathology sampling method that utilizes head tissue. Tissues for each of the three sampling methods were collected weekly for 8 weeks from individual sturgeon undergoing an experimental cohabitation challenge with fish infected with the Abernathy isolate of WSIV. Non-lethal fin histopathological evaluation did not reveal infection during the first 3 weeks of sampling, while non-lethal PCR and the lethal method were variable. However, all three sampling methods were equally capable of identifying infection from 4 to 8 weeks post-exposure. Of the survivors tested, all were negative by PCR and the lethal method, and only one fish was identified as being positive by non-lethal fin histopathology. In another experiment, all three sampling methods were applied to asymptomatic WSIV carriers in a case study conducted at the Kootenai Tribal Sturgeon Conservation Hatchery. Results showed that both lethal and non-lethal fin histopathology were equally effective in detecting infection, but PCR was unable to identify this strain of WSIV. Depending on the virus isolate, these results suggest that non-lethal sampling of fin tissue (histopathology or PCR) is comparable with the lethal sampling method at identifying WSIV infection once infection is established, and under certain circumstances may provide an alternative to lethal sampling. Concern over passage of sturgeon barriers, has focused attention on fishway design that accommodates its swimming performance. In order to evaluate swimming performance, regarding fish ladder type partial barriers, wild adult sturgeons, Acipenser transmontanus; 121–76m fork length, were captured in the San Francisco Bay Estuary and Yolo Bypass toe drain. Hydrodynamic forces and kinematic parameters for swimming performance data were collected in a laboratory flume under three flow conditions through barriers and ramp. The experiments were conducted in a 24.4 m long, 2.1 m wide, and 1.62 m deep aluminum channel. Two geometric configurations of the laboratory model were designed based on channel characteristics that have been identified in natural river systems. At a given swimming speed and fish size, the highest guidance efficiencies of successful white sturgeon passage as a function of flow depth, flow velocity, turbulence intensity, Reynolds number, Froude number and shear velocity observed in the steady flow condition, tested with the horizontal ramp structure, occurred at an approach velocity of 0.33 ms-1. The guidance efficiency of successful sturgeon passage increased both with increasing flow velocity and Froude number, and decreased both with the flow depth and the turbulence intensity. This study also provides evidence that tail beat frequency increases significantly with swimming speed, but tail beat frequency decreases with fish total length. Stride length increases both with swimming speed and fish total length. The importance of unsteady forces is expressed by the reduced frequency both with swimming speed and fish total length. Regression analysis indicates that swimming kinematic variables are explained by the swimming speed, the reduced frequency and the fish total length. Abstract: White sturgeon (Acipencer transmontanus) were exposed to 7.2μM (1.0ppm) 14C-labeled p-nitrophenol (PNP) in brackish water for 24h and then allowed to depurate in clean brackish water for another 24h. Absorption, conditional uptake clearance, and conditional elimination rate constants were 0.08±0.04h−1, 8.1±3.6mLg−1 h−1, and 0.46±0.21h−1, respectively. A whole-organism total concentration factor of 18.7±2.6 was determined from equilibrium tissue and water concentrations. Sturgeon depurated 89.4% of absorbed PNP within 24h, of which 53.0±8.3% was unmetabolized parent compound, 9.6±3.6% was p-nitrophenyl-β-d-glucuronide, and 39.1±8.3% was p-nitrophenylsulfate. This study determined the partitioning of total mercury in liver, gonad, and cheek muscle of white sturgeon ( Acipenser transmonatus) in the lower Columbia River. The relationship between tissue mercury concentrations and various physiologic parameters was assessed. White sturgeon were captured in commercial fisheries in the estuary and Bonneville, The Dalles, and John Day Reservoirs. Condition factor (CF), relative weight (Wr), and gonadosomatic index (GSI) were determined for each fish (n = 57). Gonadal tissue was examined histologically to determine sex and stage of maturity. Liver (n = 49), gonad (n = 49), and cheek muscle (n = 57) were analyzed for total mercury using cold-vapor atomic fluorescence spectrophotometry. Tissue protein concentrations were measured by ultraviolet-visible spectroscopy. Plasma was analyzed for testosterone (T), 11-ketotestosterone (KT), and 17ß-estradiol (E2) using radioimmunoassay. Mean tissue mercury concentrations were higher in muscle compared with liver and gonad at all sampling locations, except Bonneville Reservoir where mean liver mercury content was the highest tissue concentration observed in the study. Significant negative correlations between plasma androgens (T and KT) and muscle mercury content and plasma E2 and liver mercury content were found. A significant positive linear relationship between white sturgeon age and liver mercury concentrations was evident. Significant negative correlations between CF and relative weight and gonad and liver mercury content were found. In addition, immature male sturgeon with increased gonad mercury content had decreased GSIs. A simple, inexpensive apparatus (embryo incubation unit [EIU]) was developed and used to assess the relationship between sediment cover (Kootenai River sediments, 97% by weight in the 0.83-mm- to 1.0-mm-diameter range) and survival of white sturgeon Acipenser transmontanus embryos in the laboratory. An apparatus-testing trial assessed the effects of two sediment depths (5 and 20 mm), three EIU ventilation hole sizes (4.8, 6.8, and 9.5 mm) providing three levels of intrasediment flow, and EIU location (upstream or downstream in laboratory troughs) on embryo survival at two above-substrate flow velocities (0.05 and 0.15 m/s). A second trial assessed the effects of sediment cover duration (5-mm sediment cover for 4, 7, 9, 11, or 14 d, with a ventilation hole size of 9.5 mm and a flow velocity of 0.17 m/s) on mean embryo survival and larval length and weight. In the apparatus-testing trial, embryo survival was reduced (P < 0.0001) to 0-5% under sediment covers of either 5 or 20 mm in both the higher-flow and lower-flow troughs; survival in control EIUs without sediments exceeded 80%. Survival was not significantly affected by ventilation hole size but was weakly affected by EIU location. In the second trial, embryo survival was negatively correlated (P = 0.001) with increasing duration of sediment cover and was significantly higher for embryos covered for 4 d (50% survival) or 7 d (30% survival) than for those covered for 9, 11, or 14 d (15-20% survival). Sediment cover also delayed hatch timing (P < 0.0001) and decreased mean larval length (P < 0.0001). This study sought to determine whether wild white sturgeon from the Columbia River (Oregon) were exhibiting signs of reproductive endocrine disruption. Fish were sampled in the free-flowing portion of the river (where the population is experiencing reproductive success) and from three reservoirs behind hydroelectric dams (where fish have reduced reproductive success). All of the 18 pesticides and almost all of the 28 polychlorinated biphenyls (PCBs) that were analyzed in livers and gonads were detected in at least some of the tissue samples. Metabolites of p,p´-dichlorodiphenyltrichloroethane (DDT) [p,p´-dichlorodiphenyldichloroethylene (DDE) and p,p´-1,1-dichloro-2,2-bis(4-chlorophenyl)ethane (DDD)]were consistently found at relatively high levels in fish. Some males and immature females showed elevated plasma vitellogenin; however, concentrations were not correlated with any of the pesticides or PCBs analyzed. Negative correlations were found between a number of physiologic parameters and tissue burdens of toxicants. Plasma triglycerides and condition factor were negatively correlated with total DDT (DDD + DDE + DDT), total pesticides (all pesticides detected - total DDT), and PCBs. In males, plasma androgens and gonad size were negatively correlated with total DDT, total pesticides, and PCBs. Fish residing in the reservoir behind the oldest dam had the highest contaminant loads and incidence of gonadal abnormalities, and the lowest triglycerides, condition factor, gonad size, and plasma androgens. These data suggest that endocrine-disrupting chemicals may be accumulating behind dams over time. Overall, results of this study indicate that exposure to environmental contaminants may be affecting both growth and reproductive physiology of sturgeon in some areas of the Columbia River. We conducted laboratory trials to test the vulnerability of young white sturgeon, Acipenser transmontanus, to predation when an alternative prey was available. In trials with two species of predators, we observed two feeding patterns. When equal numbers of white sturgeon and goldfish, Carassius auratus, were available, prickly sculpins, Cottus asper, ingested more white sturgeon. Conversely, northern pikeminnow, Ptychocheilus oregonensis, ate more juvenile coho salmon, Oncorhynchus kisutch, than white sturgeon in three out of four sets of trials, but ate more white sturgeon in one set of trials. White sturgeon size and the availability of cover did not affect the proportions of prey species ingested. Our results indicate that predation may be affecting survival of white sturgeon larvae and juveniles in the wild and could be one factor limiting recruitment of young-of-the-year white sturgeon in some locations. This study uses a genetic individual-based model of white sturgeon (Acipenser transmontanus) populations in a river to examine the genetic and demographic trade-offs associated with operating a conservation hatchery. Simulation experiments evaluated three management practices: (i) setting quotas to equalize family contributions in an effort to prevent genetic swamping, (ii) an adaptive management scheme that interrupts stocking when introgression exceeds a specified threshold, and (iii) alternative broodstock selection strategies that influence domestication. The first set of simulations, designed to evaluate equalizing the genetic contribution of families, did not show the genetic benefits expected. The second set of simulations showed that simulated adaptive management was not successful in controlling introgression over the long term, especially with uncertain feedback. The third set of simulations compared the effects of three alternative broodstock selection strategies on domestication for hypothetical traits controlling early density-dependent survival. Simulated aquaculture selected for a density-tolerant phenotype when broodstock were taken from a genetically connected population. Using broodstock from an isolated population (i.e., above an upstream barrier or in a different watershed) was more effective at preventing domestication than using wild broodstock from a connected population. Ultrasonic telemetry was used to assess habitat features utilized by 36 endangered juvenile white sturgeon,Acipenser transmontanus, in the lower 120 km of the Kootenai River of Idaho, USA and British Columbia, Canada during the summer and early fall of 1999 and 2000. All 36 fish were initially captured in pools using gillnets and released there, but most of the subsequent telemetry contacts were in glides, indicating these fish moved freely between the two macro-habitats. The low electivity indices indicated little preference between glides and pools. Most contacts were in glides, in the outside bend of the river channel (50), and in or near a visually defined thalweg. Contacts were most often associated with sand substrates and no cover. Physical habitat characteristics (nose [bottom] water velocity, depth, substrate, and cover) were recorded at 168 contact locations. The combination of significantly greater velocities and depths at contact sites vs. non-contact sites (p<0.01) indicated these fish actively found and used areas of higher velocity and greater depth within the Kootenai River. There was little cover found for fish in the river other than large sand dunes and depth. In sturgeon aquaculture, it is advantageous to sex fish as early as possible so that males can be used for meat production and females reserved for caviar production. Currently, growers wait 3 to 4 years before fish are sexed by an invasive surgical examination of the gonads. The goal of this study was to determine when sex could be determined by examining levels of plasma steroids. Populations of white sturgeon were held at three locations and water temperatures (Oregon—13±0.1 °C, Idaho—15±0.1 °C and California—18±0.5 °C). Blood was sampled every 3 months and plasma testosterone (T), 11-ketotestosterone (KT) and estradiol (E2) concentrations were measured by radioimmunoassay (RIA). By 18 months of age, males from California had significantly higher levels of T and KT than females. By 21 months, all males could be separated from females based on T and KT content (>2 ng/ml). However, males and females from Idaho and Oregon had similar levels of T and KT up to 36 and 34 months of age, respectively, and sex could not be delineated based on plasma sex steroid content. There were no differences in E2 levels between males and females in all populations, and at all sample periods. Histological analysis of gonads revealed that all but one of the males from California had well differentiated testes with spermatogonia undergoing mitosis in cysts (stage 2), whereas the testes of all fish from Idaho and Oregon had just begun to differentiate (stage 1). Growth rates of fish were highest in California, intermediate in Idaho and lowest in Oregon. It appears that proliferation of spermatogonia is necessary for production of androgens and that growth rate, as influenced by water temperature, is the main contributor to the early onset of this process. Nine tetramer motif (GATA)[sub n] microsatellite systems were developed for use in the white sturgeon, Acipenser transmontanus. We report inheritance patterns for these nine systems, which range from one possible disomic system to tetrasomy and octosomy, with some systems containing null alleles. Because of the complex modes of inheritance underlying these systems and the highly duplicated nature of the genome, we propose each allele be scored as its own dominant marker, similar to AFLPs or RAPDs. The cytoarchitecture of the diencephalon and pretectum of the white sturgeon, Acipenser transmontanus, was studied utilizing cresyl violet stained serial paraffin sections. The identified cell groups were assigned to the preoptic area, hypothalamus, thalamus and posterior tubercle, epithalamus, synencephalon and pretectum. The outlines of the diencephalic and pretectal nuclei were projected graphically onto a midsagittal section of the brain, thus providing a reconstruction of the relative positions of the major cell groups. This facilitated comparisons with the diencephalic and pretectal nuclei of other ray-finned fishes. Our cytoarchitectural analysis indicates that the diencephalon and pretectum of the white sturgeon is intermediate to that described for cladistians and neopterygians. The preoptic area in Acipenser is relatively conservative compared to other ray-finned fishes but lacks distinct subdivisions in the magnocellular periventricular preoptic nucleus and includes a unique migrated rostral accessory nucleus. The hypothalamic walls are rather thin, and due to the presence of extensive lateral and posterior recesses and the lack of migrated nuclei, they superficially resemble the condition seen in sharks. The dorsal and ventral thalamic nuclei do not exhibit much variation compared to other ray-finned fishes, except for the presence of a small lateral posterior thalamic nucleus, the absence of a distinct ventrolateral thalamic nucleus, and slight differences in the internal organization of the ventromedial thalamic nucleus. The posterior tubercle in Acipenser clearly comprises more migrated cell groups than that of Polypterus, however, these cell groups are considerably less well defined than in neopterygians. As in other nonteleost actinopterygians, the habenular nuclei are highly asymmetrical with the right side larger than the left side. The cytoarchitectonic complexity of the pretectum in Acipenser is intermediate to that observed in Polypterus and neopterygians in that a magnocellular component within the superficial pretectal nucleus is clearly present but cannot be delineated as a distinct magnocellular superficial pretectal nucleus. Also, the posterior pretectal nucleus which is absent in Polypterus but which has been identified as a small nucleus both in Lepisosteus and Amia is represented in Acipenser by a small group of scattered cells. The effect of environmental hypercapnia on respiratory and acid-base variables was studied in white sturgeon, Acipenser transmontanus. Blood PCO2, PO2, pH, hemoglobin concentration, and plasma lactate, glucose, catecholamines and cortisol were measured first under normocapnia (water PCO2 < 0.5 Torr, 1 Torr = 133.32 Pa), then under hypercapnia (25–35 Torr) and a final return to normocapnia at 19 ± 0.5 °C. Acute (≤ 2h) hypercapnia significantly increased arterial PCO2 (8-fold increase), ventilation frequency (2-fold increase), plasma HCO3 − (2.3-fold) and decreased arterial pH (to 7.15 ± 0.02). After 24 h, norepinephrine, epinephrine and cortisol, were significantly increased, and arterial pH reached its nadir (7.10 ± 0.03). During the 72- and 96-h-periods, arterial PCO2 (24 ± 4.4 Torr) and ventilatory frequency (105 ± 5 breaths min−1) stabilized, HCO3 − reached its apparent maximum (23.6 ± 0.0 mmol−1), glucose decreased by 32%, and pH increased significantly to 7.31 + 0.03. The return to normocapnia completely restored arterial PCO2 (2.5 ± 0.14 Torr), HCO3 − (7.4 ± 0.59 mmol · l−1), ventilation frequency (71 ± 7 breaths · min−1 equilibrium curves at 15 °C and 20 °C. Recent studies have shown that white sturgeon (Acipenser transmontanus) are more resistant to cadmium (Cd) compared to rainbow trout (Oncorhynchus mykiss), whereas they are more sensitive than rainbow trout when exposed to copper (Cu). Differences in the subcellular distribution of metals among species could be one of the factors responsible for the differences in the sensitivity to metals. Although, subcellular distribution has been studied extensively in many species with many metals, its direct role in species-specific differences in the sensitivity has not been well studied. The objective of this study was to evaluate the role of subcellular distribution of metals in species-specific differences in the sensitivity to metals between sturgeon and trout. We compared the subcellular distribution of metals Cd and Cu in the cellular debris, heat-stable proteins, heat-denatured fraction, metal-rich granules, and organelles fractions from the gills and liver after exposure of juveniles of both species to 1.25 and 20 μg/L Cd and Cu for 8 days, respectively. Sturgeon diverted a higher amount of Cd towards biologically inactive metal pool (BIM) and a lower amount towards the biologically active metal pool (BAM) compared to trout in both tissues. This explained why sturgeon are able to tolerate a relatively higher exposure level to Cd compared to trout. For Cu, there was no statistically significant species-specific differences in the amounts diverted towards either BAM or BIM; hence, white sturgeon's greater sensitivity to Cu was not explained by its subcellular distribution strategies. White sturgeon are the largest freshwater fish in North America and are the focus of an intense catch-and-release (C&R) fishery; the effects are largely unknown. We assessed the effect of fight and handling time, water temperature, river discharge rate, and fish size on physiological and reflex impairment responses of wild white sturgeon to angling. Sixty of these fish were tagged with acoustic transmitters to assess survival and post-release behaviour. Survival was high (100%). Water temperature and discharge influenced post-capture blood physiology. Specifically, lactate, chloride, and cortisol concentrations were elevated in individuals fought longer, and captured at higher water temperatures and river discharge. Cortisol was affected by fish size, with lower concentrations found in larger individuals. Only lactate and chloride were positively related to reflex impairment scores. Post-release movements were correlated with physiological state, fight characteristics and the environment. Specifically, higher blood lactate and chloride and those with longer fight times moved shorter distances after release. Contrastingly, higher levels of circulating glucose and potassium, as well as larger fish captured during periods of high discharge moved longer distances. Sturgeon tended to move shorter distances and at slower rates when reflex impairment was high, although reflex impairment in general did not explain a significant proportion of the variance in any movement metric. Our results show intriguing variance in the physiological and behavioural response of individual white sturgeon to C&R angling, with some degree of environmental dependence, and highlights the importance of understanding drivers of such variation when managing fisheries.
