Objective: BYSL, which encodes the Bystin protein in humans, is upregulated in reactive astrocytes following brain damage and/or inflammation. We aimed to determine the role and mechanism of BYSL in glioma cell growth and survival. Methods: BYSL expression in glioma tissues was measured by quantitative real-time PCR, Western blot, and immunohistochemistry. In vitro assays were performed to assess the role of BYSL in cell proliferation and apoptosis. Protein interactions and co-localization were determined by co-immunoprecipitation and double immunofluorescence. The expression and activity of the AKT/mTOR signaling molecules were determined by Western blot analysis, and the role of BYSL in glioma growth was confirmed in an orthotopic xenograft model. Results: The BYSL mRNA and protein levels were elevated in glioma tissues. Silencing BYSL inhibited glioma cell proliferation, impeded cell cycle progression, and induced apoptosis, whereas overexpressing BYSL protein led to the opposite effects. We identified a complex consisting of BYSL, RIOK2, and mTOR, and observed co-localization and positive correlations between BYSL and RIOK2 in glioma cells and tissues. Overexpressing BYSL or RIOK2 increased the expression and activity of AKT/mTOR signaling molecules, whereas downregulation of BYSL or RIOK2 decreased the activity of AKT/mTOR signaling molecules. Silencing BYSL or RIOK2 decreased the growth of the tumors and prolonged the lifespan of the animals in an orthotopic xenograft model. Conclusions: High expression of BYSL in gliomas promoted tumor cell growth and survival both in vitro and in vivo. These effects could be attributed to the association of BYSL with RIOK2 and mTOR, and the subsequent activation of AKT signaling.

There has been a constant requirement from the clinic to develop biomedical titanium (Ti) implants with high osteogenic ability. In this study, we clarified a novel mechanism of how MAO (microarc oxidation) coating of Ti implants facilitates osteogenic differentiation of human bone marrow mesenchymal stem cells (hB-MSCs) by activating ERK1/2-miR-1827-Osterix signaling pathway in vitro. MAO surface of titanium implant was more favorable to promote osteogenic differentiation than SLA and AOS coating. Besides, titanium implants regulated hB-MSCs osteogenesis through the p38 MAPK pathway and ERK1/2 might be the most efficient target. Furthermore, MAO coating induced osteogenic differentiation though ERK1/2-miR-1827 pathway. Finally, we verified miR-1827 regulated osteogenic differentiation partially through Osterix. Our study reveals novel insights that MAO surface of titanium implant is a prior choice for biomedical trial and for its use in periprosthetic osteolysis (PIO) treatment in an evidence-based rationale.

BYSL, which encodes the human bystin protein, is a sensitive marker for astrocyte proliferation during brain damage and inflammation. Previous studies have revealed that BYSL has important roles in embryo implantation and prostate cancer infiltration. However, the role and mechanism of BYSL in glioblastoma (GBM) cell migration and invasion remain unknown. We found that knockdown of BYSL inhibited cell migration and invasion, downregulated the expression of mesenchymal markers (e.g., β-catenin and N-cadherin), and upregulated the expression of epithelial marker E-cadherin in GBM cell lines. Overexpression of BYSL promoted GBM cell migration, invasion, and epithelial-mesenchymal transition (EMT). In addition, the role of BYSL in promoting EMT was further confirmed in a glioma stem cell line derived from a GBM patient. Mechanistically, overexpression of BYSL increased the phosphorylation of GSK-3β and the nuclear distribution of β-catenin. Inhibition of GSK-3β by 1-Azakenpaullone could partially reverse the effects of BYSL downregulation on the transcriptional activity of β-catenin, the expression of EMT markers, and GBM cell migration/invasion. Moreover, immunohistochemical analysis showed strong expression of BYSL in GBM tissues, which was positively correlated with markers of mesenchymal GBM. These results suggest that BYSL promotes GBM cell migration, invasion, and EMT through the GSK-3β/β-catenin signaling pathway.

Marine organisms have received mounting attention in antiaging activity due to their exclusive chemicals. This review aims at revealing and discussing prospective antiaging substance from marine macroalgae, micaroalgae, invertebrate and vertebrate. The activity and mechanism of the carbohydrate, protein, pigment, flavonoids, fatty acids, phenols from marine organisms were revealed through a variety of antiaging experimental models such as rats, Drosophila melanogaster and Caenorhabditis elegans. And meanwhile, the problems and prospects aspects were discussed for future research in this field. It was suggested that the antiaging functional ingredients from these marine organisms are alternative sources for synthetic ingredients that can contribute to consumer's well-being, as part of nutraceuticals, functional foods and cosmetics.

Our group previously identified that the NOTCH1 Abruptex domain contains the most mutations in Chinese OSCC patients, including a hotspot mutation (C1133Y). FBXW7 is an E3 ubiquitin ligase that regulates a network of proteins, including NOTCH1, via degradation. In this study, we first described the co-localization of isoform specific FBXW7-FBXW7β and NOTCH1C1133Y mutation in the same cytoplasmic sites. Gain- and loss-of-function assays were performed to examine the tumor suppressor role of FBXW7β in the proliferation and invasion of OSCC cells. The co-expression of NOTCH1C1133Y and FBXW7β significantly attenuated tumor growth. Meanwhile, FBXW7β reversed the oncogenic phenotype and the activation of the AKT/ERK/NFκB pathway induced by NOTCH1C1133Y mutation. FBXW7β downregulated the stability of NOTCH1C1133Y protein and promoted protein ubiquitination. This was the first time that we selected a NOTCH1 hotspot mutation detected in clinical samples and identified the function of FBXW7β that mediated NOTCH1 mutation degradation in OSCC. The newly identified interaction between FBXW7β and NOTCH1C1133Y protein provides new insights into the progression of OSCC, especially regarding Abruptex domain mutations, and represents a valuable target for OSCC therapy.

Arachidonic acid (ARA) is involved in the growth and regulation of immunity in fish. Little research has focused on ARA in crustaceans. This study was an 8-week feeding experiment investigating the effects of dietary ARA on growth, antioxidative indices, and immune characteristics of the juvenile oriental river prawn (Macrobrachium nipponense). An ARA-enriched oil was supplemented into the basal diet to formulate six isonitrogenous and isoenergetic diets containing 0.02% (the control group), 0.28%, 0.54%, 0.86%, 1.22%, and 1.54% ARA dry weight. Diets were fed to juvenile prawns (mean weight: 0.104 ± 0.002 g) twice daily to apparent satiation in three replicates. Weight gain and specific growth rate of M. nipponense significantly increased with increasing dietary levels of ARA from 0.02% to 0.86% and thereafter declined. Hepatopancreas super oxide dismutase and catalase activities significantly increased with supplementation with ARA, particularly in prawns fed 1.22% and 1.54% ARA. The malondialdehyde level in prawns fed 1.54% ARA also significantly increased, and was higher than that of prawns fed ARA from 0.02% to 0.86%. Serum lysozyme activity was significantly greater in prawns fed 0.86% ARA compared with the other groups. mRNA expression of toll-like receptor signaling pathway-related genes was promoted by a dietary ARA level from 0.54% to 0.86% but inhibited with the higher ARA levels (1.22–1.54%). The mortality percentage increased after challenge with live Aeromonas hydrophila. The mortality percentage of prawns fed 1.54% ARA was significantly higher than that of prawns fed 0.28–1.22% ARA. The results suggest that the optimal dietary ARA requirement for juvenile M. nipponense is 0.82% (dry weight) based on second-order regression analysis of weight gain vs. dietary ARA level, and that 0.86% ARA could significantly enhance non-specific immunity of M. nipponense; however, too high a level of ARA will affect the health status of this prawn.

Feeding frequency is important for the improvement of growth performance and immunity of aquatic animals. In this study, the effect of feeding frequency on growth, body composition, antioxidant status and mRNA expression of immunodependent genes before or after ammonia-N stress was examined in Macrobrachium nipponense. Prawns were randomly assigned to one of five feeding frequencies (1, 2, 3, 4 and 6 times/day) following the same ration size over an 8-week growth trial. After the feeding trial, prawns were challenged by ammonia-N. The weight gain of prawns fed with 3-6 times/day was significantly higher than that of prawns fed with 1 time/day. The best feed conversion ratio was obtained from prawns fed with 3-6 times/day. Body crude lipid with feeding frequency of 3, 4 or 6 times/day was quite lower than that with 1 time/day. High feeding frequency (6 times/day) induced significantly elevated hepatopancreas super oxide dismutase and catalase activities. The malondialdehyde level in prawns fed with 6 times/day was also significantly increased, which was higher than that of prawns fed with other feeding frequency. mRNA expression of toll like receptor 3 and myeloid differentiation primary response protein MyD88 was promoted by feeding frequency from 3 to 4 time/day but inhibited by high or low feeding frequency. Similar mRNA expression variation trends of the two genes were observed in prawns after ammonia-N stress. After ammonia-N challenge, the highest cumulative mortality was observed in prawns fed with 6 times/day, which was significantly higher than that of prawns fed with 2-4 times/day. These findings demonstrate that (1) too high feeding frequency induced oxidative stress and malondialdehyde accumulation, negatively affecting the health status of prawns and reduced its resistance to ammonia-N stress; (2) the optimal feeding frequency to improve growth and immune response of this species at juvenile stage is 3-4 times/day; (3) considering costs of labour, a feeding frequency of 3 times/day is recommended for this prawn.

N-acetyl- d -neuraminic acid (Neu5Ac) has been considerably focused due to its promising potential applications in pharmaceuticals and dairy products. A whole-cell biocatalyst process is an important tool for synthesis of pharmaceutical intermediates and fine chemicals. In this study, a whole cell process using engineered Escherichia coli strain was developed and stepwise optimization was employed for Neu5Ac production. N-acetyl- D -glucosamine 2-epimerase and Neu5Ac aldolase were overexpressed in E. coli individually and the activity ratio was optimized by varying recombinant amounts of cell biomass for synthesis of Neu5Ac. Moreover, substrate concentrations and ratio of pyruvate and N-acetyl- D -glucosamine (GlcNAc) and detergent concentrations were optimized to increase product synthesis. The resulting process generated 237.4 mM Neu5Ac with a yield of 40.0% mol/mol GlcNAc. Furthermore, transporter pathways involved in Neu5Ac and GlcNAc were engineered and their impact on the Neu5Ac synthesis was evaluated. Using a stepwise optimization, an overall whole-cell biocatalytic process was developed and a maximum titer of 260.0 mM Neu5Ac (80.4 g/L) with a conversion yield of 43.3% from GlcNAc was achieved. The process can be used for industrial large-scale production of Neu5Ac in terms of efficiency and economy.

Long QT (LQT) type 2 (LQT2) is caused by HERG mutation. L539fs/47 encodes a truncated protein, and its mechanisms in HERG mutation are unknown. HERG mutation plasmids were overexpressed in HEK293T cells, respectively, followed by analyzing lysates with Western blot. Transfected HEK293T cells were treated with or without N-acetyl-l-leucyl-l-leucyl-l-norleucinal (ALLN) and Propranolol (Prop) at 24 or 48 h. HERG-WT, HERG-A561V, WT/A561V, HERG-L539fs/47, WT/L539fs/47, and Calnexin (CNX)/Calreticulin (CRT) protein expression and their interactions were detected by Western blot and immunoprecipitation. Each group with HERG currents (Ikr) were detected by Patch-clamp technique. Treated with ALLN, the expression of mature HERG protein and the CNX/CRT protein increased. The interaction of HERG-A561V and WT/A561V protein with the chaperone CNX/CRT increased significantly. The maximum peak currents and tail currents density increased by 70% and 73%, respectively, while maximal peak currents density (24%) and tail currents density (19%) were slight increased in WT-HERG cells. Treated with Prop, the expression and interaction of mature HERG and chaperones CNX/CRT had no difference in each group. The maximal currents and tail currents density were slight increased. CNX/CRT might play a crucial role in the trafficking-deficient process and degradation of HERG-A561V mutant protein, however they had no effect on L539fs/47 HERG due to protein transport deletion. ALLN can restore HERG-A561V mutant protein trafficking process and rescue the dominant negative suppression of WT-HERG.

Background Serum liver enzymes, which catalyze relevant catabolic pathways, have been indicated to be diagnostic and prognostic tools for several malignant tumors. The correlation between serum liver enzymes levels and survival in patients with oral and oropharynx squamous cell carcinomas (OSCC) is still absent. Here, we conducted a study focusing on predictive value of serum liver enzymes in terms of prognosis in the patients. Methods A retrospective study including 134 OSCC patients from years 2009 to 2014 was performed to investigate the association between levels of pre-treatment serum liver enzymes, various clinical parameters and prognostic outcomes, which are overall survival (OS) and disease-free survival (DFS). Log-rank tests with Kaplan-Meier method were used to detect potential prognostic biomarkers. Multivariate analyses by Cox proportional hazards model were used to identify significant predictors of prognosis. Results Serum adenosine deaminase (ADA) level was associated with patients' OS and DFS by univariate analyses (P = 0.006 and P = 0.024, respectively). Multivariate analyses showed that higher serum ADA (>17.2 U/L) (P = 0.019) as well as positive lymph node status (P = 0.035) independently predicted worse OS of patients with OSCC. In addition, older age (≥60 years) (P = 0.043) and positive lymph node status (P = 0.027) were independently prognostic parameters for poorer DFS. Conclusions Pre-operative serum ADA levels may serve as a reliable independent prognostic predictor for OS in OSCC patients.

N-acetyl- d -neuraminic acid (Neu5Ac) is a valuable resource that has seen increasing demand in both medicine and biotechnology. Although enzymatic systems and whole-cell biocatalysts have been developed for the synthesis of Neu5Ac, low yield and productivity still hamper the use of these methods on larger scales. We report the creation of an Escherichia coli biocatalyst for the efficient synthesis of Neu5Ac using a metabolic and protein engineering strategy. Expression of the two enzymes, N-acetyl-D-glucosamine 2-epimerase (AGE) and Neu5Ac lyase (NAL), was balanced using promoter engineering. Genes encoding competing pathways and GlcNAc catabolism were deleted, and then a structure-guided process was used to identify a more efficient NAL and an AGE mutant with a higher rate of Neu5Ac synthesis. The resulting biocatalyst produced 351.8 mM Neu5Ac with a yield of 58.6% from GlcNAc. This work exemplifies the use of rational design and protein engineering to construct a complex bacterial biocatalyst that can serve as a platform for the large-scale synthesis of a useful biological material.
