In the present work, an amphiphilic and magnetically recyclable graphene oxide (MR-GO) demulsifier was devised and synthesized by graft of magnetic nanoparticles (Fe3O4@SiO2-APTES) and ethylenediamine on the GO surface. The wettability and surface charges of MR-GO under various pH conditions can be regulated via adjusting the contents and species of surface functional groups (such as amino, carboxyl, and hydroxyl). In the demulsificaition test, MR-GO displayed favorable demulsification performance for crude oil-in-water (O/W) emulsion under pH of 2.0-10.0, thusly greatly improving the application scope of common demulsifier. The optimal dosage of MR-GO was 200 mg/L and the demulsification efficiency attained a maximum value of 99.7% for crude O/W emulsion with pH of 6.0. What's more, owing to its magnetic response performance, the MR-GO can be reused and the demulsification efficiency remained above 91.0% after six cycles. Based on the strong interfacial activity, MR-GO can arrive to the crude oil-water interface. With the synergy effects of interfacial adsorption (π-π/n-π) interactions and electrostatic attraction of demulsifier and interfacial films, and the aid of external mechanical forces, the interfacial films stabilized the emulsion were disrupted. Therefore, the oil droplets coated on the water droplets were gathered rapidly to form oily flocs and then migrated to the water surface to accomplish the demulsification of crude O/W emulsion.

Van der Waals solids with tunable band gaps and interfacial properties have been regarded as a class of promising active materials for electrocatalytic hydrogen evolution reaction (HER). However, due to the anisotropic features, their basal planes are usually electrochemically inert, only a few unsaturated edge atoms could serve as active centers to actuate H2 generation. Hence, material utilization and productivity efficiency are insufficient for practical applications. Recently, diverse defects have been confirmed to enable tailoring atomic configurations and electronic properties of van der Waals solids, thus triggering their superior catalytic activity of in-plane atoms while introducing high amount of new active sites. In this minireview, we summarize the state-of-the-art progress of defect engineering in van der Waals solids for HER, focusing in particular on their advantages in material modification and corresponding catalytic mechanisms. We also propose the challenges and perspectives of these catalytic materials in terms of both experimental synthesis and fundamental understanding of the defect structures.

The steady increase in the prevalence of multidrug-resistant Staphylococcus aureus has made the search for novel antibiotics to combat this clinically important pathogen an urgent matter. In an effort to discover antibacterials with new chemical structures and mechanisms, we performed a growth inhibition screen of a synthetic library against S. aureus and discovered a promising scaffold with a 1,3,5-oxadiazin-2-one core. These compounds are potent against both methicillin-sensitive and methicillin-resistant S. aureus strains. Isolation of compound-resistant strains followed by whole genome sequencing revealed its cellular target as FabH, a key enzyme in bacterial fatty acid synthesis. Detailed mechanism of action studies suggested the compounds inhibit FabH activity by covalently modifying its active site cysteine residue with high selectivity. A crystal structure of FabH protein modified by a selected compound Oxa1 further confirmed covalency and suggested a possible mechanism for reaction. Moreover, the structural snapshot provided an explanation for compound selectivity. On the basis of the structure, we designed and synthesized Oxa1 derivatives and evaluated their antibacterial activity. The structure-activity relationship supports the hypothesis that noncovalent recognition between compounds and FabH is critical for the activity of these covalent inhibitors. We believe further optimization of the current scaffold could lead to an antibacterial with potential to treat drug-resistant bacteria in the clinic.

Separation of dyes and proteins from water remains a significant challenge. Common absorbents without recyclability might bring secondary pollutions, which may be avoided by developing stimuli-responsive smart absorbents. Here in this work, a CO2-responsive polymer poly(2-(dimethylamino) ethyl methacrylate) was grafted from cellulose fibers (Cell-g-PDEA). The chemical structure and surface morphology were confirmed with nuclear magnetic resonance spectroscopy, Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, X-ray powder diffraction and scanning electron microscope. The CO2-sensitivity was demonstrated by monitoring the pH, surface Zeta potential and dispersed appearance under stimulus of CO2. Furthermore, the modified cellulose fibers were packed into a syringe to construct a smart separation column that reversibly captured and released anionic dyes and proteins from water under gas stimulus. Comparing with traditional pH or temperature responsive materials, the CO2-responsive cellulose fibers take the advantages on energy-saving, good reversibility and moderate running conditions, thus might find applications in smart separations, clinical diagnose and therapies.

A continuous increase in ambient temperature caused by global warming has been considered a worldwide threat. As sessile organisms, plants have evolved sophisticated heat shock response (HSR) to respond to elevated temperatures and other abiotic stresses, thereby minimizing damage and ensuring the protection of cellular homeostasis. In particular, for perennial trees, HSR is crucial for their long life cycle and development. HSR is a cell stress response that increases the number of chaperones including heat shock proteins (HSPs) to counter the negative effects on proteins caused by heat and other stresses. There are a large number of HSPs in plants, and their expression is directly regulated by a series of heat shock transcription factors (HSFs). Therefore, understanding the detailed molecular mechanisms of woody plants in response to extreme temperature is critical for exploring how woody species will be affected by climate changes. In this review article, we summarize the latest findings of the role of HSFs and HSPs in the HSR of woody species and discuss their regulatory networks and cross talk in HSR. In addition, strategies and programs for future research studies on the functions of HSFs and HSPs in the HSR of woody species are also proposed.

Crassulacean acid metabolism (CAM) is an important photosynthetic pathway in diverse lineages of plants featuring high water-use efficiency and drought tolerance. A big challenge facing the CAM research community is to understand the function of the annotated genes in CAM plant genomes. Recently, a new genome editing technology using CRISPR/Cas9 has become a more precise and powerful tool than traditional approaches for functional genomics research in C3 and C4 plants. In this study, we explore the potential of CRISPR/Cas9 to characterize the function of CAM-related genes in the model CAM species Kalanchoë fedtschenkoi. We demonstrate that CRISPR/Cas9 is effective in creating biallelic indel mutagenesis to reveal previously unknown roles of blue light receptor phototropin 2 (KfePHOT2) in the CAM pathway. Knocking out KfePHOT2 reduced stomatal conductance and CO2 fixation in late afternoon and increased stomatal conductance and CO2 fixation during the night, indicating that blue light signaling plays an important role in the CAM pathway. Plants with crassulacean acid metabolism (CAM) generally utilize water 20%–80% more efficiently than non-CAM plants. The whole genomes of several CAM plants have been sequenced or are being sequenced. For effective genome characterization and genome editing of CAM plants, an efficient transformation system is essential. In this study, we developed an Agrobacterium-mediated transformation protocol for Kalanchoe laxiflora, an obligate CAM plant,by optimizing several factors affecting the transformation efficiency. Agrobacterium strains AGL1, C58, EHA105, and GV3101 were all suitable for K. laxiflora transformation. Fifty-nine percent of the leaf explants yielded kanamycin-resistant and GUS-positive shoots. Polymerase chain reaction and quantitative real-time PCR (qRT-PCR) using gusA-, gusPlus-, nptII- and hpt-specific primers confirmed that the transgenes were integrated into K. laxiflora genome and expressed. This efficient transformation system will allow effective functional characterization of genes through over- or down-expression, knockout, or genome editing. Cellulose paper derived from plants has served as a carrier of cultural inheritance since it was invented 2000 years ago. It attracted more and more attentions in recent years working as a versatile platform to develop functional materials because of its good flexibility, sustainability, biodegradability, low cost and great capability to be modified. Once be functionalized, such as modified with stimuli-responsive polymers, small molecules or inorganic particles, papers may be sensitive to external stimuli from environments and consequently find applications in wide ranges including protein separation, controlled drug release, switchable surfaces, sensoring devices and smart substrates with various colors, etc. However, there are no reviews summarized the progress of such exciting research field. Here in this mini review, we discussed the advantages of cellulose paper as stimuli-responsive substrate and summarized the fabrication techniques, properties and applications of various stimuli-responsive papers reported to date which are triggered by temperature, pH, humidity, ions, light, magnetic field, solvent, gas and biomolecules. Additionally, challenges and outlooks of the future development of such materials were also given in the end.

We present novel techniques for realising reliable low overhead logic functions and more complex systems based on the switching characteristics of memristors. Firstly, we show that memristive circuits have inherent properties for realising multiple valued MIN-MAX operations over the post algebra. We then present an efficient hybrid 1T-4M logic architecture for dual XOR/AND and XNOR/OR functionality, which can be seamlessly integrated with the existing CMOS technology. Although memristors are usually considered to operate at lower frequencies, however, recent advances in technology show their potentiality at high frequencies. To this end, we also explore the effects of high frequencies on their performance and thereby propose reliable high frequency design techniques based on our 1T-4M architectures. Experimental results, based on the design of full adders and multipliers over GF, show that the proposed designs require significantly lower power and overhead while maintaining reliable performance at low as well as at high frequencies compared to the existing techniques.

Plant drought stress causes systematic changes to photosynthesis, metabolism, growth, and potentially the phytobiome. Additionally, drought affects plants in both a species-specific and water-deficit-driven manner, causing the response to drought to be dependent both on how drought is being experienced and on any adaptation to prior drought exposure. Thus, understanding the effect of drought on plants requires assessing drought response in multiple conditions, such as progressive acute drought and recurrent cyclic drought, and at different levels of severity. In this study, we have utilized RNA sequencing to identify changes to the plant transcriptome and the phytobiome during both acute progressive drought and cyclic drought at multiple severities. Co-analysis of the plant and phytobiome, utilizing the same RNAseq data, allows for the identification of novel associations that would not be possible otherwise. We have identified that the drought response ranges from increased transcripts related to photosynthesis and metabolic activity in mild acute drought to decreased transcripts related to photosynthesis and metabolic impairment in severe drought. Moreover, while water deficit is a main driver of transcriptional responses in severe drought, there are increases in reactive oxygen species (ROS) metabolism and photosynthetic transcripts in cyclic severe drought compared with acute severe drought, independent of water deficit. The phytobiome exhibits alternate responses to drought when compared with the transcriptome. Specifically, the phytobiome is affected more by the cyclic or acute nature of the drought rather than the severity of the drought, with the phytobiome having an increase in taxa under cyclic drought that are often reported to have beneficial effects on the plants. Lastly, we have identified associations between taxa in the phytobiome with expression of disease response, ROS metabolism, and photosynthesis transcripts suggesting interplay between the host plant and its phytobiome in response to drought.

Background and Aims The use of woody crops for Quad-level (approx. 1 × 1018 J) energy production will require marginal agricultural lands that experience recurrent periods of water stress. Populus species have the capacity to increase dehydration tolerance by lowering osmotic potential via osmotic adjustment. The aim of this study was to investigate how the inherent genetic potential of a Populus clone to respond to drought interacts with the nature of the drought to determine the degree of biochemical response. Methods A greenhouse drought stress study was conducted on Populus deltoides ‘WV94’ and the resulting metabolite profiles of leaves were determined by gas chromatography–mass spectrometry following trimethylsilylation for plants subjected to cyclic mild (–0.5 MPa pre-dawn leaf water potential) drought vs. cyclic severe (–1.26 MPa) drought in contrast to well-watered controls (–0.1 MPa) after two or four drought cycles, and in contrast to plants subjected to acute drought, where plants were desiccated for up to 8 d. Key Results The nature of drought (cyclic vs. acute), frequency of drought (number of cycles) and the severity of drought (mild vs. severe) all dictated the degree of osmotic adjustment and the nature of the organic solutes that accumulated. Whereas cyclic drought induced the largest responses in primary metabolism (soluble sugars, organic acids and amino acids), acute onset of prolonged drought induced the greatest osmotic adjustment and largest responses in secondary metabolism, especially populosides (hydroxycinnamic acid conjugates of salicin). Conclusions The differential adaptive metabolite responses in cyclic vs. acute drought suggest that stress acclimation occurs via primary metabolism in response to cyclic drought, whereas expanded metabolic plasticity occurs via secondary metabolism following severe, acute drought. The shift in carbon partitioning to aromatic metabolism with the production of a diverse suite of higher order salicylates lowers osmotic potential and increases the probability of post-stress recovery.

All-inorganic metal halide perovskites have attracted great interest in recent years due to their good device performance with higher thermal stability than that of their organic–inorganic perovskite counterparts. However, the all-inorganic perovskite polycrystalline films prepared by the conventional spin-coating method possess many pinholes, nonuniform surface with many small crystals, and irregular agglomerates, limiting their device performance. Herein, we introduced a monolayer nano-polystyrene (PS) sphere confined growth method for obtaining CsPbBr3 nanonet films (NFs) with ordered nanostructures grown in the preferred (110) orientation, which is beneficial for the charge carrier transport and the light-harvesting efficiency. The (110) peak intensity of CsPbBr3 NFs increased with the increase of the diameter of the monolayer sphere, while the (001) peak intensity was suppressed greatly, indicating the more preferred (110) oriented growth. The PDs based on (110)-orientation-preferred CsPbBr3 NFs prepared by using 850 nm PS spheres showed the best performance. The best performing device displayed the biggest linear dynamic range of up to 120 dB. In addition, a responsivity of 2.84 A W−1 and a detectivity of 5.47 × 1012 Jones were also achieved.

Cell wall recalcitrance is the major obstacle for plant biomass conversion to biofuels. In this study, we functionally characterized a serine hydroxymethyltransferase (SHMT) from Populus and evaluated its potential for developing lignocellulosic feedstocks. SHMT is an enzyme that plays an important role in cellular one-carbon pathways. However, little is known about its function in plant cell wall-related processes. Among nine SHMT genes in the Populus genome, PtSHMT2 was highly expressed in the developing xylem and was co-expressed with secondary cell wall biosynthetic genes. In Populus transgenic plants overexpressing PdSHMT2, the biomass yield and sugar (glucose and xylose) release were increased whereas the lignin content was decreased. Transcriptomics and metabolomics analyses revealed that genes and metabolites related to secondary cell wall biosynthesis were affected by PdSHMT2 overexpression. Based on the transcription factor binding sites of differentially expressed genes in PdSHMT2 overexpression lines, a total of 27 hub transcription factors were identified. We proposed a regulatory model of action of PdSHMT2 with transcriptional master switches of secondary cell wall biosynthesis. Collectively, these results suggest that PdSHMT2 is a promising candidate for genetic engineering to improve feedstock characteristics to enhance biofuel conversion and reduce the cost of lignocellulosic biofuel production.

It has been challenging to simultaneously improve photosynthesis and stress tolerance in plants. Crassulacean acid metabolism (CAM) is a CO2-concentrating mechanism that facilitates plant adaptation to water-limited environments. We hypothesized that the ectopic expression of a CAM-specific phosphoenolpyruvate carboxylase (PEPC), an enzyme that catalyzes primary CO2 fixation in CAM plants, would enhance both photosynthesis and abiotic stress tolerance. To test this hypothesis, we engineered a CAM-specific PEPC gene (named AaPEPC1) from Agave americana into tobacco. In comparison with wild-type and empty vector controls, transgenic tobacco plants constitutively expressing AaPEPC1 showed a higher photosynthetic rate and biomass production under normal conditions, along with significant carbon metabolism changes in malate accumulation, the carbon isotope ratio δ13C, and the expression of multiple orthologs of CAM-related genes. Furthermore, AaPEPC1 overexpression enhanced proline biosynthesis, and improved salt and drought tolerance in the transgenic plants. Under salt and drought stress conditions, the dry weight of transgenic tobacco plants overexpressing AaPEPC1 was increased by up to 81.8% and 37.2%, respectively, in comparison with wild-type plants. Our findings open a new door to the simultaneous improvement of photosynthesis and stress tolerance in plants.

Coal burst continues to be one of the most catastrophic safety hazards faced by future mining as the stress environment will be more complicated with the increase of mining depth. Many chief coal mining countries including Poland, Czech Republic, the U.S., China, and Australia have experienced fatal accidents caused by coal burst and conducted comprehensive research on the driving forces and solving technologies related to coal burst. In this chapter, the research outcomes of the mechanism, risk evaluation, risk monitoring, and prevention of coal burst are reviewed, which is helpful for mining researchers and engineers to understand and control the safety hazards caused by coal burst, and, hence, to achieve sustainable and safe mining.

A grand challenge facing society is climate change caused mainly by rising CO 2 concentration in Earth’s atmosphere. Terrestrial plants are linchpins in global carbon cycling, with a unique capability of capturing CO 2 via photosynthesis and translocating captured carbon to stems, roots, and soils for long-term storage. However, many researchers postulate that existing land plants cannot meet the ambitious requirement for CO 2 removal to mitigate climate change in the future due to low photosynthetic efficiency, limited carbon allocation for long-term storage, and low suitability for the bioeconomy. To address these limitations, there is an urgent need for genetic improvement of existing plants or construction of novel plant systems through biosystems design (or biodesign). Here, we summarize validated biological parts (e.g., protein-encoding genes and noncoding RNAs) for biological engineering of carbon dioxide removal (CDR) traits in terrestrial plants to accelerate land-based decarbonization in bioenergy plantations and agricultural settings and promote a vibrant bioeconomy. Specifically, we first summarize the framework of plant-based CDR (e.g., CO 2 capture, translocation, storage, and conversion to value-added products). Then, we highlight some representative biological parts, with experimental evidence, in this framework. Finally, we discuss challenges and strategies for the identification and curation of biological parts for CDR engineering in plants.

Summary Lysine crotonylation of proteins is a recently identified post‐translational modification (PTM) in plants. However, the function of lysine‐crotonylated proteins in response to abiotic stress in plants has not been reported. In this study, we identified a temperature‐induced lipocalin‐1‐like gene (DgTIL1) from chrysanthemum and showed that it was notably induced in response to cold stress. Overexpression of DgTIL1 enhanced cold tolerance in transgenic chrysanthemum. Ubiquitin membrane yeast two‐hybrid (MYTH) system and bimolecular fluorescence complementation (BIFC) assays showed that DgTIL1 interacts with a nonspecific lipid transfer protein (DgnsLTP), which can promote peroxidase (POD) gene expression and POD activity to reduce the accumulation of reactive oxygen species (ROS) and improve resistance to cold stress in DgnsLTP transgenic chrysanthemum. In addition, we found that DgTIL1 was lysine crotonylated at K72 in response to low temperature in chrysanthemum. Moreover, lysine crotonylation of DgTIL1 prevented DgnsLTP protein degradation in tobacco and chrysanthemum. Inhibition of DgnsLTP degradation by lysine crotonylation of DgTIL1 further enhanced POD expression and POD activity, reduced the accumulation of ROS under cold stress in DgTIL1 transgenic chrysanthemum, thus promoting the cold resistance of chrysanthemum.

Sulfamethoxazole (SMX) is the most widely distributed sulfonamide antibiotics detected in decentralized poultry wastewater in rural communities. As an economically-feasible and eco-friendly technology for decentralized wastewater treatment in rural areas, vertical-flow multi-soil-layering (MSL) system was promising to mitigate the ecological and human health risks from SMX in such areas. The treatment of SMX-contained poultry wastewater by using MSL systems was investigated for the first time, and the main and interactive effects of related multiple variables on system performance were explored through factorial analysis, including material of permeable layer, concentration of SMX, and pH of influent. Results indicated that SMX concentration and pH of influent showed significantly negative effects on SMX removal. Medical stone used in MSL systems with larger surface area could intensify the SMX removal compared to anthracite. MSL systems showed stable performances on SMX removal with the best SMX removal efficiency more than 91%. A novel stepwise-cluster inference (SCI) model was developed for the first time to map the multivariate numeric relationships between state variables and SMX removal under discrete and nonlinear complexities. It was demonstrated that the effect of SMX in wastewater with high concentration was significant on the differentiation of soil bacteria composition in MSL systems based on microbial diversity analysis. These results can help better understand the mechanism of SMX removal in MSL systems from perspectives of factorial analysis, numeric modeling, and microbiological change.

Summary Prefoldin (PFD) is a group II chaperonin that is ubiquitously present in the eukaryotic kingdom. Six subunits (PFD1‐6) form a jellyfish‐like heterohexameric PFD complex and function in protein folding and cytoskeleton organization. However, little is known about its function in plant cell wall‐related processes. Here, we report the functional characterization of a PFD gene from Populus deltoides, designated as PdPFD2.2. There are two copies of PFD2 in Populus, and PdPFD2.2 was ubiquitously expressed with high transcript abundance in the cambial region. PdPFD2.2 can physically interact with DELLA protein RGA1_8g, and its subcellular localization is affected by the interaction. In P. deltoides transgenic plants overexpressing PdPFD2.2, the lignin syringyl/guaiacyl ratio was increased, but cellulose content and crystallinity index were unchanged. In addition, the total released sugar (glucose and xylose) amounts were increased by 7.6% and 6.1%, respectively, in two transgenic lines. Transcriptomic and metabolomic analyses revealed that secondary metabolic pathways, including lignin and flavonoid biosynthesis, were affected by overexpressing PdPFD2.2. A total of eight hub transcription factors (TFs) were identified based on TF binding sites of differentially expressed genes in Populus transgenic plants overexpressing PdPFD2.2. In addition, several known cell wall‐related TFs, such as MYB3, MYB4, MYB7, TT8 and XND1, were affected by overexpression of PdPFD2.2. These results suggest that overexpression of PdPFD2.2 can reduce biomass recalcitrance and PdPFD2.2 is a promising target for genetic engineering to improve feedstock characteristics to enhance biofuel conversion and reduce the cost of lignocellulosic biofuel production.

The effects of hydraulic retention time (HRT) and influent nitrate-N concentration on nitrogen removal and the microbial community composition of an aerobic denitrification reactor treating recirculating marine aquaculture system effluent were evaluated. Results showed that over 98% of nitrogen was removed and ammonia-N and nitrite-N levels were below 1 mg/L when influent nitrate-N was below 150 mg/L and HRT over 5 h. The maximum nitrogen removal efficiency and nitrogen removal rate were observed at HRT of 6 or 7 h when influent nitrate-N was 150 mg/L. High-throughput DNA sequencing analysis revealed that the microbial phyla Proteobacteria and Bacteroidetes were predominant in the reactor, with an average relative total abundance above 70%. The relative abundance of denitrifying bacteria of genera Halomonas and Denitratisoma within the reactor decreased with increasing influent nitrate-N concentrations. Our results show the presence of an aerobically denitrifying microbial consortium with both expected and unexpected members, many of them relatively new to science. Our findings provide insights into the biological workings and inform the design and operation of denitrifying reactors for marine aquaculture systems. Published version

Global demand for food and bioenergy production has increased rapidly, while the area of arable land has been declining for decades due to damage caused by erosion, pollution, sea level rise, urban development, soil salinization, and water scarcity driven by global climate change. In order to overcome this conflict, there is an urgent need to adapt conventional agriculture to water-limited and hotter conditions with plant crop systems that display higher water-use efficiency (WUE). Crassulacean acid metabolism (CAM) species have substantially higher WUE than species performing C 3 or C 4 photosynthesis. CAM plants are derived from C 3 photosynthesis ancestors. However, it is extremely unlikely that the C 3 or C 4 crop plants would evolve rapidly into CAM photosynthesis without human intervention. Currently, there is growing interest in improving WUE through transferring CAM into C 3 crops. However, engineering a major metabolic plant pathway, like CAM, is challenging and requires a comprehensive deep understanding of the enzymatic reactions and regulatory networks in both C 3 and CAM photosynthesis, as well as overcoming physiometabolic limitations such as diurnal stomatal regulation. Recent advances in CAM evolutionary genomics research, genome editing, and synthetic biology have increased the likelihood of successful acceleration of C 3 -to-CAM progression. Here, we first summarize the systems biology-level understanding of the molecular processes in the CAM pathway. Then, we review the principles of CAM engineering in an evolutionary context. Lastly, we discuss the technical approaches to accelerate the C 3 -to-CAM transition in plants using synthetic biology toolboxes.

The long atmospheric residence time of CO 2 creates an urgent need to add atmospheric carbon drawdown to CO 2 regulatory strategies. Synthetic and systems biology (SSB), which enables manipulation of cellular phenotypes, offers a powerful approach to amplifying and adding new possibilities to current land management practices aimed at reducing atmospheric carbon. The participants (in attendance: Christina Agapakis, George Annas, Adam Arkin, George Church, Robert Cook-Deegan, Charles DeLisi, Dan Drell, Sheldon Glashow, Steve Hamburg, Henry Jacoby, Henry Kelly, Mark Kon, Todd Kuiken, Mary Lidstrom, Mike MacCracken, June Medford, Jerry Melillo, Ron Milo, Pilar Ossorio, Ari Patrinos, Keith Paustian, Kristala Jones Prather, Kent Redford, David Resnik, John Reilly, Richard J. Roberts, Daniel Segre, Susan Solomon, Elizabeth Strychalski, Chris Voigt, Dominic Woolf, Stan Wullschleger, and Xiaohan Yang) identified a range of possibilities by which SSB might help reduce greenhouse gas concentrations and which might also contribute to environmental sustainability and adaptation. These include, among other possibilities, engineering plants to convert CO 2 produced by respiration into a stable carbonate, designing plants with an increased root-to-shoot ratio, and creating plants with the ability to self-fertilize. A number of serious ecological and societal challenges must, however, be confronted and resolved before any such application can be fully assessed, realized, and deployed.

Our society faces multiple daunting challenges including finding sustainable solutions towards climate change mitigation; efficient production of food, biofuels, and biomaterials; maximizing land-use efficiency; and enabling a sustainable bioeconomy. Plants can provide environmentally and economically sustainable solutions to these challenges due to their inherent capabilities for photosynthetic capture of atmospheric CO 2 , allocation of carbon to various organs and partitioning into various chemical forms, including contributions to total soil carbon. In order to enhance crop productivity and optimize chemistry simultaneously in the above- and belowground plant tissues, transformative biosystems design strategies are needed. Concerted research efforts will be required for accelerating the development of plant cultivars, genotypes, or varieties that are cooptimized in the contexts of biomass-derived fuels and/or materials aboveground and enhanced carbon sequestration belowground. Here, we briefly discuss significant knowledge gaps in our process understanding and the potential of synthetic biology in enabling advancements along the fundamental to applied research arc. Ultimately, a convergence of perspectives from academic, industrial, government, and consumer sectors will be needed to realize the potential merits of plant biosystems design for a carbon neutral bioeconomy.

Many applications in plant biology requires editing genomes accurately including correcting point mutations, incorporation of single-nucleotide polymorphisms (SNPs), and introduction of multinucleotide insertion/deletions (indels) into a predetermined position in the genome. These types of modifications are possible using existing genome-editing technologies such as the CRISPR-Cas systems, which require induction of double-stranded breaks in the target DNA site and the supply of a donor DNA molecule that contains the desired edit sequence. However, low frequency of homologous recombination in plants and difficulty of delivering the donor DNA molecules make this process extremely inefficient. Another kind of technology known as base editing can perform precise editing; however, only certain types of modifications can be obtained, e.g., C/G-to-T/A and A/T-to-G/C. Recently, a new type of genome-editing technology, referred to as “prime editing,” has been developed, which can achieve various types of editing such as any base-to-base conversion, including both transitions (C→T, G→A, A→G, and T→C) and transversion mutations (C→A, C→G, G→C, G→T, A→C, A→T, T→A, and T→G), as well as small indels without the requirement for inducing double-stranded break in the DNA. Because prime editing has wide flexibility to achieve different types of edits in the genome, it holds a great potential for developing superior crops for various purposes, such as increasing yield, providing resistance to various abiotic and biotic stresses, and improving quality of plant product. In this review, we describe the prime editing technology and discuss its limitations and potential applications in plant biology research.

Memristors are finding applications in memory, logic, neuromorphic systems, and data security. To this end, we leverage the non-linear behaviour of memristors to devise a low overhead physical unclonable function using a memristive chaos circuit in conjunction with a non-linear memristive encoder. We demonstrate the effectiveness of this architecture in Challenge-Response-Pair based authentication, and for its physical uncloneability. This architecture is highly versatile and can be implemented with a single encoder or a number of encoders running in parallel, each one with its own merit, for extending the sizes of CRPs. To demonstrate its effectiveness, we subject the architecture to machine learning based modelling attacks e.g. Logistic Regression, Support Vector Machines, Random Forest, as well as Artificial Neural Network classifiers. We found out that the proposed PUF architecture provides better resistance to such attacks, even for smaller bit sizes and at reduced overheads.

Human life intimately depends on plants for food, biomaterials, health, energy, and a sustainable environment. Various plants have been genetically improved mostly through breeding, along with limited modification via genetic engineering, yet they are still not able to meet the ever-increasing needs, in terms of both quantity and quality, resulting from the rapid increase in world population and expected standards of living. A step change that may address these challenges would be to expand the potential of plants using biosystems design approaches. This represents a shift in plant science research from relatively simple trial-and-error approaches to innovative strategies based on predictive models of biological systems. Plant biosystems design seeks to accelerate plant genetic improvement using genome editing and genetic circuit engineering or create novel plant systems through de novo synthesis of plant genomes. From this perspective, we present a comprehensive roadmap of plant biosystems design covering theories, principles, and technical methods, along with potential applications in basic and applied plant biology research. We highlight current challenges, future opportunities, and research priorities, along with a framework for international collaboration, towards rapid advancement of this emerging interdisciplinary area of research. Finally, we discuss the importance of social responsibility in utilizing plant biosystems design and suggest strategies for improving public perception, trust, and acceptance.

For decades, plants have been the subject of genetic engineering to synthesize novel, value-added compounds. Polyhydroxyalkanoates (PHAs), a large class of biodegradable biopolymers naturally synthesized in eubacteria, are among the novel products that have been introduced to make use of plant acetyl-CoA metabolic pathways. It was hoped that renewable PHA production would help address environmental issues associated with the accumulation of nondegradable plastic wastes. However, after three decades of effort synthesizing PHAs, and in particular the simplest form polyhydroxybutyrate (PHB), and seeking to improve their production in plants, it has proven very difficult to reach a commercially profitable rate in a normally growing plant. This seems to be due to the growth defects associated with PHA production and accumulation in plant cells. Here, we review major breakthroughs that have been made in plant-based PHA synthesis using traditional genetic engineering approaches and discuss challenges that have been encountered. Then, from the point of view of plant synthetic biology, we provide perspectives on reprograming plant acetyl-CoA pathways for PHA production, with the goal of maximizing PHA yield while minimizing growth inhibition. Specifically, we suggest genetic elements that can be considered in genetic circuit design, approaches for nuclear genome and plastome modification, and the use of multiomics and mathematical modeling in understanding and restructuring plant metabolic pathways.

Coal burst is the violent failure of overstressed coal, and it is often accompanied by sound, coal ejection and seismic events. It is subsequently recognized as a serious safety risk of Australia after double fatalities coal burst happened at Austar Coal Mine. Considering the increasing trend of coal burst severity and frequency with mining depth, it is an urgent task to develop the coal burst risk assessment methods for Australia underground coal mines. Coal burst propensity index method is a widely used method of burst risk evaluation of coal as it is summed up from the coal burst research and practice of many countries. This paper presents the experimental and theoretical research of coal burst propensity index method for coal burst risk assessment in Australia. The definition of four indexes including elastic strain energy index (WET), bursting energy index (KE), dynamic failure time (DT) and uniaxial compression strength (RC) is introduced in the first part. Then, the standard laboratory test process and test parameter of coal burst propensity index is presented. DT test is conducted with 0.3 mm/min displacement control loading rate while other test is with 0.5 mm/min. Besides, modified data processing and risk classification method of test are proposed. Differentiate analysis of stress-strain curve is adopted in the data processing of DT and KE index. A four level risk classification form of burst risk is recommended for Australian underground coal mines. Finally, two likely improvement methods of WET test, including volumetric strain indicator method and theoretical calculation method, are discussed. To investigate the bedding influence on coal mechanical behaviour in underground environments such as coal or rock burst, simulations of dynamic SHPB tests of pre-stressed coal specimens with different bedding angles were carried out using a particle flow code 2-dimensional (PFC2D). Three impact velocities of 4, 8 and 12 m/s were selected to study dynamic behaviours of coal containing bedding planes under different dynamic loads. The simulation results showed that the existence of bedding planes leads to the degradation of the mechanical properties and their weakening effect significantly depends on the angle θ between the bedding planes and load direction. With θ increaseing from 0° to 90°, the strength first decreased and subsequently increased and specimens became most vulnerable when θ was 30° or 45°. Five failure modes were observed in the specimens in the context of macro-cracks. Furthermore, energy characteristics combined with ultimate failure patterns revealed that maximum accumulated energy and failure intensity have a positive relation with the strength of specimen. When bedding planes were parallel or perpendicular to loading direction, specimens absorbed more energy and experienced more violent failure with increased number of cracks. In contrast, bedding planes with θ of 30° or 45° reduced the specimens’ ability of storing strain energy to the lowest with fewer cracks observed after failure. Owing to their low trap-state density, high carrier mobility, and high thermal stability, CsPbBr3 perovskite microcrystals (MCs) have attracted significant attention for applications as photodetectors (PDs). However, solution synthesis processes lead to MC films with high void density, seriously limiting the performance of the PDs. Here, a pressure-assisted annealing strategy is introduced to significantly reduce the void density and decrease the surface roughness. The resulting self-powered all-inorganic CsPbBr3 perovskite MC thick-film PDs show improved performance characteristics, with responsivities and detectivities of up to 0.206 A W–1 and 7.23 × 1012 Jones, respectively. Moreover, the on/off ratios of the devices are up to 106, and the highest linear dynamic range reaches 123.5 dB. These improved results indicate that the pressure-assisted annealing method is an effective strategy to enhance the performance of solution-synthesized perovskite MC PDs.

A zinc-catalyzed intermolecular alkyne-carbonyl metathesis reaction of ynamides with isatins followed by an amide to ester conversion has been developed, which produces the indolone derivatives with a fully substituted alkene species in good to high yields. The salient features of this reaction include the following: mild reaction conditions, an inexpensive zinc catalyst, a broad substrate scope, the excellent regiocontrol and stereoselectivity, and amenable to the gram scale.

Background Plant secondary cell wall is a renewable feedstock for biofuels and biomaterials production. Arabidopsis VASCULAR-RELATED NAC DOMAIN (VND) has been demonstrated to be a key transcription factor regulating secondary cell wall biosynthesis. However, less is known about its role in the woody species. Results Here we report the functional characterization of Populus deltoides WOOD-ASSOCIATED NAC DOMAIN protein 3 (PdWND3A), a sequence homolog of Arabidopsis VND4 and VND5 that are members of transcription factor networks regulating secondary cell wall biosynthesis. PdWND3A was expressed at higher level in the xylem than in other tissues. The stem tissues of transgenic P. deltoides overexpressing PdWND3A (OXPdWND3A) contained more vessel cells than that of wild-type plants. Furthermore, lignin content and lignin monomer syringyl and guaiacyl (S/G) ratio were higher in OXPdWND3A transgenic plants than in wild-type plants. Consistent with these observations, the expression of FERULATE 5-HYDROXYLASE1 (F5H1), encoding an enzyme involved in the biosynthesis of sinapyl alcohol (S unit monolignol), was elevated in OXPdWND3A transgenic plants. Saccharification analysis indicated that the rate of sugar release was reduced in the transgenic plants. In addition, OXPdWND3A transgenic plants produced lower amounts of biomass than wild-type plants. Conclusions PdWND3A affects lignin biosynthesis and composition and negatively impacts sugar release and biomass production.

Cell walls in crops and trees have been engineered for production of biofuels and commodity chemicals, but engineered varieties often fail multi-year field trials and are not commercialized. We engineered reduced expression of a pectin biosynthesis gene (Galacturonosyltransferase 4, GAUT4) in switchgrass and poplar, and find that this improves biomass yields and sugar release from biomass processing. Both traits were maintained in a 3-year field trial of GAUT4-knockdown switchgrass, with up to sevenfold increased saccharification and ethanol production and sixfold increased biomass yield compared with control plants. We show that GAUT4 is an α-1,4-galacturonosyltransferase that synthesizes homogalacturonan (HG). Downregulation of GAUT4 reduces HG and rhamnogalacturonan II (RGII), reduces wall calcium and boron, and increases extractability of cell wall sugars. Decreased recalcitrance in biomass processing and increased growth are likely due to reduced HG and RGII cross-linking in the cell wall.

Self-powered light-emitting photodetectors (SLEPs) can simultaneously transmit and receive information using light, offering huge potential for applications in interactive communication systems. However, high-performance SLEPs are very hard to achieve due to the seriously contradictory principles of carrier transport in SLEPs. In this study, a novel structure based on a ZnO hollow ball/CsPbBr3 heterojunction, in which ZnO hollow balls function as an ultraviolet (UV) response center and CsPbBr3 is designed to be a green light-emitting material, is reported as a high-performance SLEP. Since the ZnO hollow balls have superior light trapping characteristics for UV light, the device shows a huge UV response with the on/off ratio and detectivity up to 16 527 and 2.4 × 1013 Jones, respectively. Moreover, it displays blue emission with a low threshold voltage because of CsPbBr3. Compared with the traditional ZnO/GaN structure devices, the device showed enhanced dual-functional performance at various wavelengths due to the insertion of CsPbBr3, which resulted in a better contact interface and green-light emission center. Our structure separating the light-responsive and emitting active regions avoids the contradictory principles of carrier transport, which may provide new access to gain high-performance dual-functional devices.

Background The development of fast-growing hardwood trees as a source of lignocellulosic biomass for biofuel and biomaterial production requires a thorough understanding of the plant cell wall structure and function that underlie the inherent recalcitrance properties of woody biomass. Downregulation of GAUT12.1 in Populus deltoides was recently reported to result in improved biomass saccharification, plant growth, and biomass yield. To further understand GAUT12.1 function in biomass recalcitrance and plant growth, here we report the effects of P. trichocarpa GAUT12.1 overexpression in P. deltoides. Results Increasing GAUT12.1 transcript expression by 7–49% in P. deltoides PtGAUT12.1-overexpression (OE) lines resulted in a nearly complete opposite biomass saccharification and plant growth phenotype to that observed previously in PdGAUT12.1-knockdown (KD) lines. This included significantly reduced glucose, xylose, and total sugar release (12–13%), plant height (6–54%), stem diameter (8–40%), and overall total aerial biomass yield (48–61%) in 3-month-old, greenhouse-grown PtGAUT12.1-OE lines compared to controls. Total lignin content was unaffected by the gene overexpression. Importantly, selected PtGAUT12.1-OE lines retained the recalcitrance and growth phenotypes upon growth for 9 months in the greenhouse and 2.8 years in the field. PtGAUT12.1-OE plants had significantly smaller leaves with lower relative water content, and significantly reduced stem wood xylem cell numbers and size. At the cell wall level, xylose and galacturonic acid contents increased markedly in total cell walls as well as in soluble and insoluble cell wall extracts, consistent with increased amounts of xylan and homogalacturonan in the PtGAUT12.1-OE lines. This led to increased cell wall recalcitrance, as manifested by the 9–15% reduced amounts of recovered extractable wall materials and 8–15% greater amounts of final insoluble pellet in the PtGAUT12.1-OE lines compared to controls. Conclusions The combined phenotype and chemotype data from P. deltoides PtGAUT12.1-OE and PdGAUT12.1-KD transgenics clearly establish GAUT12.1 as a recalcitrance- and growth-associated gene in poplar. Overall, the data support the hypothesis that GAUT12.1 synthesizes either an HG-containing primer for xylan synthesis or an HG glycan required for proper xylan deposition, anchoring, and/or architecture in the wall, and the possibility of HG and xylan glycans being connected to each other by a base-sensitive covalent linkage.

By employing a conjugated amine-functionalized dicarboxylic ligand (H2L = 2,2'-diamino-4,4'-stilbenedicarboxylic acid, H2SDCA-NH2), we have successfully synthesized and characterized a porous and visible light responsive zirconium metal-organic framework ([Zr6O4(OH)4(L)6]·8DMF, denoted as Zr-SDCA-NH2). This Zr-MOF showed good chemical stability and broad visible light absorption with an absorption edge at about 600 nm. When used as a photocatalyst, Zr-SDCA-NH2 exhibits visible-light activity for CO2 reduction with a formate formation rate of 96.2 μmol h-1 mmolMOF-1, which is higher than the series of reported amine-functionalized Zr-MOFs. Mott-Schottky measurements, photoluminescence study and photocatalytic experiments demonstrated that the Zr6 oxo cluster through the LMCT process and the organic ligand both contributed to the CO2 photoreduction. This study indicates that the combination of amino groups and highly conjugated molecules is a feasible and simple strategy to extend light absorption of the organic ligand, which is beneficial for designing a visible light responsive MOF photocatalyst.

The detergent has wide range of applications and plays a critical role in daily life. However, the existing commercial detergents present the drawbacks of irreversible environmental pollution due to their discharge with wastewater. Herein, halloysite clay nanotube (HNT) with unique hollow structure was explored as a green, non-toxic, multifunctional, and cheap detergents. HNT has large aspect ratio, small size, and high adsorption ability, which provides the possibility for removing various stains from different substrates. The increased contact angle and decreased surface tension of HNT dispersion are achieved by increasing the HNT concentration, and HNT tends to stabilize oil-in-water (O/W) Pickering emulsion. The cleaning capacity of HNT on cotton fabric for dyes is almost twice that of laundry powder, indicating that highly efficient cleaning capability of HNT. HNT detergent is also effective to remove ink, tea, and chili oil from textiles with clean efficacy higher than 88%. As kitchen detergent, HNT can effectively eliminates chili oil from ceramic, stainless steel, glass, and plastic plate. Seed germination and crop growth experiment demonstrate that HNT detergent shows no toxicity towards plants and no contamination towards soil. Furthermore, the residue on the cloth of HNT detergent is negligible. Overall, HNT has great potential as an alternative detergent in the future because of its excellent cleaning efficacy and ecological friendliness.

Kiwifruit tends to develop alcoholic off-flavour after long-term storage, leading to decreased fruit market value. Actinidia eriantha ‘White’ and Actinidia deliciosa ‘Bruno’ are two major cultivated kiwifruits in China. Compared to A. eriantha ‘White’, A. deliciosa ‘Bruno’ is prone to developing alcoholic off-flavour associated with ethanol accumulation during storage at ambient conditions without any stresses. In this study, to better understand the complex mechanisms responsible for developing alcoholic off-flavour in kiwifruit during postharvest, transcriptome analysis was used to compare the difference in metabolic pathways between kiwifruits ‘white’ and ‘Bruno’ during ripening at ambient storage. Our results indicated that the development of alcoholic off-flavour in kiwifruit ‘Bruno’ might be associated with three metabolic pathways: the respiration and ethylene metabolisms, starch and sucrose metabolism and ethanol fermentation metabolism. In kiwifruit ‘Bruno’, the higher respiration rate and ethylene production might increase the energy conversion and internal carbon dioxide (CO2) accumulation. Moreover, the higher expressions of starch phosphorylases (SPs), beta-amylases, UDP-glucose pyrophosphorylases (UGPases), sucrose synthases (SSs) and invertases (INVs) accelerated starch degradation along with soluble sugars accumulation in kiwifruit ‘Bruno’, which could provide adequate substrates for ethanol fermentation. In addition, the higher activity of ethanol fermentation metabolism associated with higher expressions of pyruvate kinases (PKs), NADP-dependent malic enzymes (NADP-MEs), pyruvate decarboxylases (PDCs) and alcohol dehydrogenases (ADHs) might directly increase the ethanol production.

Industrially, the Agave tequilana plant is most commonly associated with beverage production, which requires it to be grown for 8–10 year before harvesting. However, its robust growth characteristics and low water usage requirements suggest that it could also be utilized as a substrate for alternative fermentation processes that do not require such long crop turnover times. Therefore, a study was undertaken to characterize the feasibility of utilizing the leaves from two year-old plants as a microbial fermentation biomass substrate. Quantitative saccharification revealed the carbohydrate concentration of two year-old A. tequilana leaves to be 633 mg g−1 dry weight biomass. HPLC analysis identified the primary carbohydrates as glucans, with minor contributions from xylans, galactans, arabinanas, and mannans. Fructose was observed predominantly as inulin. The direct use of unpretreated A. tequilana leaf biomass as a sole carbon source could support yeast growth, however, dilute acid pretreatment removed sufficient levels of carbohydrates to prevent the growth of wild type or inulinase-expressing yeast strains. Up to 133.5 g kg−1 of soluble carbohydrates and 69.6 g kg−1 of soluble inulin were extracted in the pretreatment soakate and 4.0 g kg−1 of soluble carbohydrates and 1.3 g kg−1 of soluble inulin remained in the biomass post processing. Inulinase gene expression did not significantly improve yeast growth rates, but permitted up to 6.3 g kg−1 of the available soluble inulin from the dilute acid pretreatment soakate to be metabolized towards cellular growth.

Ceramic filters are a point-of-use (POU) technology applied for water purification in developing regions. Nano-CeO2 modified ceramic filter water purifier (CeO2–CFP) is designed to provide clean potable water and address drinking water safety issues in remote areas. To assess the impact of the entire life cycle of CeO2–CFP on the water environment, a life cycle assessment (LCA)-based water footprint framework was established. The context of remote areas was used to exemplify the calculation of the developed model under different technical scenarios. The production of CeO2–CFP in the high-tech scenario exhibited excellent environmental performance and water resource cost-effectiveness was found to be only 1.59. Raw materials (71.41%) and staff consumption (82.54%) represented the largest share of water footprint in the high-tech and low-tech scenarios, respectively. Sensitivity analysis was proceeded to identify the critical factors affecting the water footprint of CeO2–CFP system and the interactions of these significant factors were investigated. A results-based analysis was carried out in consideration of environmental, social, and economic aspects, and some recommendations for reducing water footprint of CeO2–CFP were formulated.

The remediation of water that polluted by organic dyes and nanoparticles (NPs) remains a great challenge. However, second pollution of solid waste may produce if the adsorbents are not degradable or recyclable. The stimuli-responsive materials were regarded to be able to resolve this problem by reversible adsorption-desorption under stimuli. Here in this work, the surface of cellulose fibers was modified with CO2-responsive molecules through a simple esterification followed by an aza-Michael addition reaction. The chemical structure was confirmed with Fourier transform infrared spectra (FT-IR), X-ray photoelectron spectroscopy (XPS), x-ray diffractometer (XRD) and thermal analysis. The gas-responsiveness was demonstrated by monitoring of the pH, conductivity and surface zeta potential. Anionic dyes and negative charged NPs in water can be selectively and reversibly captured by the CO2-responsive surface on cellulose fibers, suggesting the inexpensive, readily available and smart cellulose fibers may find great potential in water remediation.

The mechanism between food gelation and its digestibility has attracted increasing attention over the past few decades. This study aimed to evaluate the effect of glycation degree on the gelation and digestibility of egg white gel (EWG) using an in vitro model and a multi-scale characterization of gel structure. Results showed that EWG glycated with increasing d -ribose by covalent bonds exhibited better gelling properties and lower in vitro digestibility according to the appearance of soluble proteins and peptides. Besides, glycation preference for ovotransferrin at lysine might be important for regulating gel structure and proteolysis accessibility via the ratio of fibrous and granular aggregates. Moreover, gel structure was predominant over amino acids modification for digestibility. Binding disorder and steric hindrance could ascribe to the lower digestibility of gels. These findings are enlightening for the formulation and production of food matrix with controlled digestibility through glycation in food and related pharmaceutical fields.

Since 2009, the subsidy for large-scale photovoltaic (PV) power plants had been launched, which effectively promoted the development of PV industry. At the same time, negative effects, like serious oversupply of PV industry, were brought about by these large scale governmental subsidies. Although governmental subsidy strongly supports the China PV companies, few of them have competitiveness in the global market. This dramatically conflictive phenomenon attracted many researchers’ attentions in recent years. However, investigations on the best entry and exit occasions of governmental subsidies for the PV industry were rarely reported in previous studies. Therefore, based on the existing division method of enterprise development model, classification of 72 PV companies listed in Shanghai and Shenzhen Stock Exchanges in China was firstly carried out in this paper. This is followed by studying the influence of governmental subsidies on the indexes of different stages of enterprise development. Finally, a conclusion was drawn that the governmental subsidies at Early Exploratory Stage can maximize the social and economic effects, suggesting the best entry occasion, and subsidies at Intermediate Stage and Mature Stage have little effects on its turnover and aggravate the overcapacity of PV supply, suggesting a suitable exit occasion.

The aim of this work was to experimentally examine apparent rheological properties of fluids in microchannels based on pressure drop. Pressure drops of Newtonian and non-Newtonian fluids in microchannels were measured using pressure sensors. The effects of flow rate, Reynolds number, and viscosity of fluids on the pressure drop of single phase flow in microchannels were investigated, and the measurements of pressure drop were compared with prediction models. For Newtonian fluids, the friction factor in the laminar regime is f/2 = 26/Re. The results show that the pressure drop of Newtonian fluid is in good agreement with the prediction formula proposed by Cornish. However, the pressure drop of non-Newtonian fluid deviates from the theoretical prediction. A power-law is proposed for the relationship between the apparent viscosity of non-Newtonian fluid and the characteristic shear rate in a microchannel based on the pressure drop measurement. This relationship between the apparent viscosity and the characteristic shear rate in a specific microchannel is completely different from the rheological curve measured by cone-and-plate rheometer, due to the heterogeneous distribution of shear rates at the radial direction for non-Newtonian fluids flowing in a microchannel.

CO2 uptake and water loss in plants are regulated by microscopic pores on the surface of leaves, called stomata. This enablement of gas exchange by the opening and closing of stomata is one of the most essential processes in plant photosynthesis and transpiration, affecting water-use efficiency (WUE) and thus drought susceptibility. In plant species with crassulacean acid metabolism (CAM) photosynthesis, diel stomatal movement pattern is inverted relative to C3 and C4 photosynthesis species, resulting in much higher WUE and drought tolerance. However, little is known about the molecular basis of stomatal movement in CAM species. The goal of this study is to identify candidate genes that could play a role in stomatal movement in an obligate CAM species, Kalanchoë fedtschenkoi. By way of a text-mining approach, proteins were identified in various plant species, spanning C3, C4, and CAM photosynthetic types, which are orthologous to proteins known to be involved in stomatal movement. A comparative analysis of diel time-course gene expression data was performed between K. fedtschenkoi and two C3 species (i.e., Arabidopsis thaliana and Solanum lycopersicum) to identify differential gene expression between the dusk and dawn phases of the 24-h cycle. A rescheduled catalase gene known to be involved in stomatal movement was identified, suggesting a role for H2O2 in CAM-like stomatal movement. Overall, these results provide new insights into the molecular regulation of stomatal movement in CAM plants, facilitating genetic improvement of drought resistance in agricultural crops through manipulation of stomata-related genes.

Genome-wide association studies (GWAS) have great promise for identifying the loci that contribute to adaptive variation, but the complex genetic architecture of many quantitative traits presents a substantial challenge. We measured 14 morphological and physiological traits and identified single nucleotide polymorphism (SNP)-phenotype associations in a Populus trichocarpa population distributed from California, USA to British Columbia, Canada. We used whole-genome resequencing data of 882 trees with more than 6.78 million SNPs, coupled with multitrait association to detect polymorphisms with potentially pleiotropic effects. Candidate genes were validated with functional data. Broad-sense heritability (H2 ) ranged from 0.30 to 0.56 for morphological traits and 0.08 to 0.36 for physiological traits. In total, 4 and 20 gene models were detected using the single-trait and multitrait association methods, respectively. Several of these associations were corroborated by additional lines of evidence, including co-expression networks, metabolite analyses, and direct confirmation of gene function through RNAi. Multitrait association identified many more significant associations than single-trait association, potentially revealing pleiotropic effects of individual genes. This approach can be particularly useful for challenging physiological traits such as water-use efficiency or complex traits such as leaf morphology, for which we were able to identify credible candidate genes by combining multitrait association with gene co-expression and co-methylation data.

The groundwater with high arsenic concentration is widespread, especially in many remote areas of developing countries. Arsenic existing in drinking water sources has a high risk to human health. In this study, an innovative effort to remove As(V) from water using ceramic filters functionalized with CeO2 nanocomposites (CF-CeO2) was investigated. Considering removal efficiency and flow rate, the suitable coating amount of CeO2 was determined for the production of CF-CeO2. Based on the factorial analysis, influent arsenic concentration and pH were found to be significant factors in As(V) removal. Furthermore, CF-CeO2 exhibited a good removal capability over a wide pH range and was insensitive to the change of background electrolyte concentration. In the treatment of natural water, the existence of medium and low turbidity levels facilitated As(V) removing, while the high turbidity level exhibited the opposite effect. Based on macroscopic experiments and microscopic characterizations, it was revealed that the As(V) removal mechanism by the CF-CeO2 mainly included ion-exchange and electrostatic attraction. The findings in this study provided convincing evidence for the use of CF-CeO2 as a high-efficiency, low-cost, and safe approach for water purification in the remote areas of developing countries.

The importance of increasing crop production while reducing resource inputs and land-use change cannot be overstated especially in light of climate change and a human population growth projected to reach nine billion this century. Mutualistic plant–microbe interactions offer a novel approach to enhance agricultural productivity while reducing environmental costs. In concert with other novel agronomic technologies and management, plant-microbial mutualisms could help increase crop production and reduce yield losses by improving resistance and/or resilience to edaphic, biologic, and climatic variability from both bottom-up and top-down perspectives.
