Abstract

International audience; Novel genosensors (Brett DNA) on nanostructured ultrafine coverslips are reported for the identification of Brettanomyces bruxellensis yeast that is well-known to create complex and difficult problems in aged red wine all over the word. Surface Enhanced Raman Scattering (SERS) and Localize Surface Plasmon Resonance (LSPR) are used as detection methods of target-ssDNA yeast. A specific detection is obtained in the presence of thiol-DNA probe. SERS technique made possible the detection of Brett-DNA with a limit of detection (LD) of 0,1 ng/μL((57,2 nM) and with a wide dynamic range (DR), while LSPR revealed a DL of 0,5 ng/μL real DNA extracted from Brettanomyces bruxellensis DKA in the laboratory and a limited DR (0.5 ng/ μL to 5 ng/ μL). In this work, 14 yeasts that can be found in wine-growing environments were SERS tested to confirm the selectivity of the proposed Brett-DNA sensing schemes and used in the construction of the first SERS phylogenetic tree.

Abstract

International audience; Phthalocyanines are frequently used as probing molecules in the field of single-molecule surface-enhanced Raman spectroscopy (SERS) and tip-enhanced Raman spectroscopy (TERS). In this work, we systematically compare the SERS and TERS spectra from a thin cobalt phthalocyanine (CoPc) film that is deposited on a Au film. The contributions from electromagnetic (EM), resonance, and charge-transfer enhancements are discussed. Radially and azimuthally polarized vector beams are used to investigate the influences of molecular orientation and the localized surface plasmon resonance (SPR). Furthermore, two different excitation wavelengths (636 and 532 nm) are used to study the resonant excitation effect as well as the involvement of the charge-transfer processes between CoPc and the Au substrate. It is shown that the Raman peaks of CoPc are mostly enhanced by 636 nm excitation through a combination of resonant excitation, high EM enhancement, and chemical enhancement via charge transfer from the metal to the molecule. At 532 nm excitation, however, the SERS and TERS spectra are dominated by photoluminescence, which originates from a photo-induced charge-transfer process from the optically excited molecule to the metal. The contributions of the different enhancement mechanisms explain the optical contrasts seen in the TERS images of Au nanodisks covered by the CoPc film. The insight achieved in this work will help to understand the optical contrast in sub- or single-molecule TERS imaging and apply SERS or TERS in the field of photocatalysis.

Abstract

International audience; The active control of the quantum dots (QD) photoluminescence (PL) is a challenge for many applications, including the development of new commercial LEDs or new single photon sources in the emerging field of quantum nanooptics. A way to modulate the QD’s luminescence is to combine them with chromophores. Indeed, by a Förster resonance energy transfer (FRET) process, a donor fluorophore can transfer its energy to an acceptor chromophore, without the involvement of photons. Interesting dyes are those, which exhibit photochromism properties. Indeed, due to their ability to optically switch between two molecular states with different absorption properties, the photochromic molecules (PM) have been used to actively control the QD’s PL1. As this modulation is based on an energy transfer from the QD, the PL can never reach a higher intensity than the one emitted by the QD alone. However, it has been proven that the PL intensity can be enhanced by coupling the QDs to plasmonic nanoparticles (NPs), like metallic nanoparticles 2. In this talk, we report on the optical control of the de-excitation pathways of a coupled system between CdSe/ZnS QDs and silver nanocylinders of varying diameters using the conformational change of photochromic spiropyran molecules 3. We use Fluorescence Lifetime Imaging Microscopy (FLIM) to record quantum dots (QDs) photoluminescence lifetime and intensity before and after the photochromic transition.

Abstract

International audience; Surface-enhanced Raman spectroscopy (SERS) is a powerful method to probe the changes that occur in a molecule in contact with a metal. However, the molecules interacting with a plasmonic substrate make a complicated picture of the vibrational spectra. Understanding the different contributions requires a multi-dimensional approach involving the spectroscopic investigation under various experimental parameters such as plasmonic nanostructure material, density and kind of hotspots, substrate temperature, laser energy, and molecular layer density (from bulk crystals to ultrathin 1 and 2 nm thick layers). In this contribution, we report our findings deduced from a systematic investigation of all these parameters. The new insights deduced from this analysis include the equivalency between Cu and Au nanostructures. For these two materials, we found that it is not the chemistry what makes the largest changes but the density and geometry of hotspots and electric field amplification. The chemical contribution is evidenced in Ag and bimetallic nanostructures under resonance and non-resonance excitations. The temperature-dependent investigations indicate that the temperature coefficients and minima in intensity vs. T curves observed for some molecule-plasmonic nanostructures can be correlated to perturbations in the electronic structure of the molecule. These results show that controlling internal and external parameters in plasmon-enhanced spectroscopy allows deducing novel insights on the chemical and electromagnetic effects in these hybrid organic/inorganic systems.

Abstract

International audience; Metallic nanoparticles (MNP) may sustain Localized Surface Plasmon (LSP) resonances and the coupling between MNP and quantum dots (QD) is known to lead either to the enhancement or to the quenching of the QD photoluminescence [1]. The distance between QD and MNP is one of the switch parameters between both regimes. The goal of this study is to control the coupling distance (different from the physical distance) between QD and MNP by changing the refractive index of the surrounding medium using photochromic molecules (PM). These molecules are optical switches, which move from a transparent state to a colored one by absorbing UV light. The main drawback concerns the spectral overlap and the lifetime of each optical phenomenon, since the photochromic molecules can couple to LSP to induce strong coupling [2] or couple to QDs to quench the photoluminescence [3]. In this study, we measured photoluminescence spectra, images and lifetimes of QDs deposited on a glass substrate, with or without the presence of MNPs and PM.

Abstract

International audience; In this paper gold nanorings (NRs) are applied as particularly well-suited sensing elements for mapping the radially symmetric electric fields in the high numerical aperture focus of cylindrical vector beams. The optical properties of gold nanorings are analyzed by a combination of extinction and single particle dark field spectroscopy as well as confocal photoluminescence (PL) imaging. The results are compared to numerical calculations. The in-plane components in the focus of the cylindrical vector beams are estimated through the PL intensity distributions of the NRs. The optimum overlap between the structure and excitation is visualized by a narrow centre spot in the far-field PL scan.

Abstract

International audience; The optical characterization of a single metallic nanostructure has a strong interest in the scientific community owing to its localized surface plasmon resonances. For a single nano-object, the simplest and the accepted optical characterization technique is dark-field spectroscopy, even if there are many drawbacks and a certain complexity to operate it. We propose here using extinction spectroscopy of nanoparticles ensembles to characterize optically a single nanostructure. The scattering spectrum of a single gold nanocylinder and the extinction spectrum of a well-chosen array show similar results. We perform an experimental and numerical comparative study to draw parallels between both techniques.

Abstract

International audience; Metallic nanoparticles (MNP) may sustain Localized Surface Plasmon (LSP) resonances and the coupling between MNP and quantum dots (QD) is known to lead either to the enhancement or to the quenching of the QD photoluminescence. The distance between QD and MNP is one of the switch parameters between both regimes. The goal of this study is to control the coupling distance (different from the physical distance) between QD and MNP by changing the refractive index of the surrounding medium using photochromic molecules (PM). These molecules are optical switches, which move from a transparent state to a colored one by absorbing UV light. The main drawback concerns the spectral overlap and the lifetime of each optical phenomenon, since the photochromic molecules can couple to LSP to induce strong coupling or couple to QDs to quench the photoluminescence. In this study, we measured photoluminescence spectra, images and lifetimes of QDs deposited on a glass substrate, with or without the presence of MNPs and PM.
