International audience; The Morte-Femme peatland (Vosges Mountains, Northeastern France) is a protected area receiving treated wastewater from several textile industries. In spite of the treatment, these effluents still contain non-biodegradable micropollutants related to the textile process. Composed of highly substituted aromatic structures, optical brighteners (OBs) are water-soluble substances with a high affinity for cellulose fibers. OBs are used by the textile industry to mask the yellowing of cotton fabrics, as the blue light (400 – 440 nm range) they emit after UV excitation makes the fabrics appear whiter to the human eye. OBs can be quantified by HPLC-based techniques but this requires an expensive equipment. Furthermore, the exact nature of the OBs is generally unknown. Fluorescence-based methods are preferred for routine monitoring (Cao et al., 2009). For this purpose, yhe presence of OBs in the different watercourses running across the peatland has been assessed by differential synchronous fluorescence (i.e. before and after irradiation by UV (365 nm), with synchronous fluorescence spectra collected with a gap of 50 nm between excitation and emission) (Assaad et al., 2015). In order to discriminate OBs from the humic substances naturally present in the water samples, different irradiation durations have been tested. If a 15min irradiation is sufficient to detect OBs in solution in pure water, the presence of other elements (salts, dissolved organic matter, etc.) seems to hinder the irradiation effect. Therefore the effect of the irradiation duration has been tested on synthetic solutions of several OBs in presence of salts, organic matter and other potential textile additives. The final procedure has then been applied in the framework of the monitoring of the quality of the peatland surface water over two years. The results are discussed with respect to the streams natural discharge rate, which is highly dependent upon the season, and the textile industry activity.

Abstract

International audience; Measuring flow-rate and pollutant discharges through sewer networks is required for legal, technical and management reasons. This paper presents an experimental study combining continuous monitoring of flow and water quality parameters with studies of the velocity and suspended solids spatial distribution in order to understand both the question of temporal and spatial representativeness of continuous measurements. The experimental site has been chosen to investigate the situation in both a uniform and a complex situations, it is formed by the junction of two channels. Three measurement points were selected: upstream, downstream and at the junction. Firstly, the context of this project is reported, secondly the experimental investigations and data validation procedure are detailed and finally the results are presented and discussed. Logical and statistical automated methods were used to validate data. The necessity of the application of data validation procedure was proved for faulty data detection and faulty data correction. The ranges of quantity (heights, velocities and so flow rates) parameters were determined using continuous monitoring. Some trend evolutions and ranges were noted for temperature, conductivity, pH.

International audience; La bonne gestion des réseaux d'assainissement est un élément essentiel pour répondre à des objectifs aussi bien techniques qu'économiques de recouvrement du bon état écologique des milieux aquatiques. Elle nécessite de mieux connaître les charges polluantes déversées aux échelles événementielle et annuelle, d'améliorer le fonctionnement et la gestion des réseaux pour, à terme, minimiser les rejets non traités. Cet article présente une étude de l'influence d'une singularité, en l'occurrence une confluence, sur la répartition spatiale des vitesses et des concentrations afin d'évaluer la représentativité des mesures en continu de débits liquides et des mesures de charges polluantes. Des informations sur le fonctionnement des réseaux seront obtenues ainsi que des données pouvant contribuer à la validation des modélisations numériques. Les résultats contribueront à la finalisation de la méthodologie d'aide à la qualification des sites de mesures, objet du projet national MENTOR.

Abstract

Modern quantitative image analysis has been extensively used to characterize the morphology of microorganisms, especially those of the filamentous type. More recently physiological features have been quantified, making use of classical stains as well as fluorescent dyes. The potential of the technique is illustrated by a detailed analysis of the differentiation of Streptomyces ambofaciens. Three different staining procedures have been used to monitor the thinning and septation of hyphae (with propidium iodide staining), the leakage of cellular components through the membrane (with carbol gentian violet staining) and the respiration (with INT staining) in a batch submerged culture.
